- Description: This thesis examines selected speculative novels by Margaret Atwood, Ursula K. Le Guin, Doris Lessing and Marge Piercy. It argues that a specifiable ecological ethic can be traced in their work – an ethic which is explored by them through the tensions between utopian and dystopian discourses. The first part of the thesis begins by theorising the concept of an ecological ethic of respect for the Other through current ecological philosophies, such as those developed by Val Plumwood. Thereafter, it contextualises the novels within the broader field of science fiction, and speculative fiction in particular, arguing that the shift from a critical utopian to a critical dystopian style evinces their changing treatment of this ecological ethic within their work. The remainder of the thesis is divided into two parts, each providing close readings of chosen novels in the light of this argument. Part Two provides a reading of Le Guin’s early Hainish novels, The Left Hand of Darkness, The Word for World is Forest and The Dispossessed, followed by an examination of Piercy’s Woman on the Edge of Time, Lessing’s The Marriages Between Zones Three, Four and Five, and Atwood’s The Handmaid’s Tale. The third, and final, part of the thesis consists of individual chapters analysing the later speculative novels of each author. Piercy’s He, She and It, Le Guin’s The Telling, and Atwood’s Oryx and Crake are all scrutinised, as are Lessing’s two recent ‘Ifrik’ novels. This thesis shows, then, that speculative fiction is able to realise through fiction many of the ideals of ecological thinkers. Furthermore, the increasing dystopianism of these novels reflects the greater urgency with which the problem of Othering needs to be addressed in the light of the present global ecological crisis.

- Description: This thesis argues that a prominent concern among contemporary writers of fiction is the recuperation of lost or occluded histories. Increasingly, contemporary writers, especially postcolonial writers, are using the medium of fiction to explore those areas of political and cultural history that have been written over or unwritten by the dominant narrative of “official” History. The act of excavating these past histories is simultaneously both traumatic and liberating – which is not to suggest that liberation itself is without pain and trauma. The retelling of traumatic pasts can lead, as is portrayed in The God of Small Things (1997), to further trauma and pain. Postcolonial writers (and much of the world today can be construed as postcolonial in one way or another) are seeking to bring to the fore stories of the past which break down the rigid binaries upon which colonialism built its various empires, literal and ideological. Such writing has in a sense been enabled by the collapse, in postcolonial and postmodernist discourse, of the Grand Narrative of History, and its fragmentation into a plurality of competing discourses and histories. The associated collapse of the boundary between history and fiction is recognized in the useful generic marker “historiographic metafiction,” coined by Linda Hutcheon. The texts examined in this study are all variants of this emerging contemporary genre. What they also have in common is a concern with the consequences of exile or diaspora. This study thus explores some of the representations of how the exilic experience impinges on the development of identity in the postcolonial world. The identities of “displaced” people must undergo constant change in order to adjust to the new spaces into which they move, both literal and metaphorical, and yet critical to this adjustment is the cultural continuity provided by psychologically satisfying stories about the past. The study shows that what the chosen texts share at bottom is their mutual need to retell the lost pasts of their characters, the trauma that such retelling evokes and the new histories to which they give birth. These texts generate new histories which subvert, enrich, and pre-empt formal closure for the narratives of history which determine the identities of nations.

'The Most Amazing Show': performative interactions with postelection South African society and culture

Specifically, it looks at the relationship between a popular comic variety show The Most Amazing Show (TMAS), and aspects of contemporary South African society and culture, particularly relating to prominent efforts to affect post-election transformation of South African society and culture through the construction of a South African 'rainbow nation'. By analysing TMAS in terms of broader historical, performative and discursive contexts, it engages a relational reading of the performance. The study argues that TMAS both challenges and participates in the manner in which rainbow nation discourse constructs South African society and culture. Firstly, it considers the performance's construction of hybrid South African identities, including white Afrikaans, white English and white masculine identities. It argues that these reconstructions undermine the tendency within rainbow nation discourse to construct cultural hybridity in terms of stereotypically distinct identities. Secondly, it considers TMAS' construction of collective experience and social integration, which subverts the often glamorised and superficial representations of social healing and integration that are constructed within rainbow nation discourse. The analysis makes prominent reference to the notion of 'liminality' in order to describe the manner in which TMAS constructs significance within the tension that it establishes between oppositional, and often contradictory, positions. Furthermore, it attempts to establish a link between this notion of liminality and notions of theatrical syncretism that are prominent in contemporary South African theatre scholarship, and emphasise processes of signification that are constantly shifting and unstable.

- Description: Falling within the ambit of the Department of English Literature but with interdisciplinary scope and method, the research undertaken in this thesis examines Michael Radford’s 2004 film production William Shakespeare’s The Merchant of Venice using the Book History approach to textual study. Previously applied almost exclusively to the study of books, Book History examines the text in terms of both its medium and its content, bringing together bibliographical, literary and historical approaches to the study of books within one theoretical paradigm. My research extends this interdisciplinary approach into the filmic medium by using a modified version of Robert Darnton’s “communication circuit” to examine the process of transmission of this Shakespearean film adaptation from creation to reception. The research is not intended as a complete Book History study and even less as a comprehensive investigation of William Shakespeare’s The Merchant of Venice. Rather, it uses a Shakespearean case study to bring together the two previously discrete fields of Book History and filmic investigation. Drawing on film studies, literary concepts, cultural and media studies, modern management theory as well as reception theories and with the use of both quantitative and qualitative data, I show Book History to be an eminently useful and constructive approach to the study of film.

- Description: The media, which help define what we think and our roles in the society, have a crucial role to project both men and women’s issues so as to change people’s perceptions and stereotypes about the role men and women play in the society. There is need, therefore, to ensure gender equality in the operations of the media so that issues to do with both men and women get adequate and equal coverage. This study on the reportage of the land redistribution exercise in Zimbabwe has, however, exposed the gendered nature of the operations of the media, particularly in the news production process. It provides that, overally, the news discourse is a masculine narrative whose androcentric form is a result of, and is protected by, claims to ‘objectivity,’ ‘professionalism’, ‘impartiality’ and the pursuit of a journalistic routine system that hegemonically prioritises men’s issues over those of women. The situation, as the research shows, has not been helped by journalists’ incapacity to do thematic appreciation of issues and their over-inclination towards a simplistic event-based journalism that fails to question policies as they are enacted and implemented in gender-skewed processes. The lack of gender policies, the operations of patriarchy and the pursuit of a journalistic routine system that sees nothing wrong with the ostracisation of women issues are very fundamental findings that the research uses in its attempts to explain why the gender equality discourse was left out of the news reports about the land reform exercise in Zimbabwe.

- Description: The aims of this study were to determine the potential use of biomarkers at multiple levels of biological organisation together with a fish community bioindicator to assess the estuarine health status of three temporarily open/closed estuaries. The estuaries investigated were the East Kleinemonde (EK), Old Woman's (OW) and Mtana (MTN), all of which are situated in the Eastern Cape Province. Three biomarkers, the acetylcholinesterase (AChE) assay, lipid peroxidation (LPx) assay, liver histopathology and a condition factor were used to determine sub-organism health and one bioindicator, the Estuarine Fish Community Index (EFCI), was used as a bioindicator of community health. The estuarine-dependent marine species Rhabdosargus holubi was selected as an indicator species for the sub-organism level analyses. The results from the community analyses indicated that the EK and OW estuaries were in 'good' condition, while the MTN was found to be in 'moderate' condition. Histological analyses revealed that R. holubi from all three estuaries showed signs of pathological changes to the liver, with the fish from the MTN eliciting the highest occurrence of these changes. The LPx assay found that R. holubi from both the OW and MTN showed signs of oxidative damage in the liver tissue, but those from the EK did not appear to be affected. The AChE assay showed that only the fish from the OW had been affected by anticholinesterase compounds. A laboratory study was undertaken using R. holubi as a positive control for the AChE and LPx assay. The fish were exposed to 3 μg/L chlorpyrifos, a known cholinesterase inhibitor, for six hours and their tissues were examined for changes to LPx levels and AChE activities. AChE activity was significantly inhibited (Mann Whitney U test, Z = 3.65, n = 38, P < 0.001) by the exposure, but LPx levels were not significantly affected. A composite index incorporating the biomarkers at different biological levels of organisation was developed. The index was designed to assist managers and scientists to determine whether the ichthyofauna of a system was being affected by environmental stressors and what management interventions could be undertaken to ameliorate the water quality in an estuary. The index was applied to the three estuaries investigated during the present study and both the OW and MTN were assessed to be in need of immediate management intervention. The fish in the OW were found to be stressed at all the sub-organism levels measured and the reason for this was hypothesised to be as a result of golf course activities in this adjacent estuary. A number of management actions are proposed to reduce the sub-organism stress observed in the fish from the OW. The livers of fish from the MTN were shown to be under stress; however the causative agent of this stress was unknown because there is no formal development in the MTN catchment. However, a possible contaminant source is proposed and management interventions to alleviate the stress on the biota of the MTN are suggested. The EK does not require immediate management intervention, however, continuous routine monitoring is recommended to ensure that conditions do not deteriorate. Shortcomings of the index were outlined and a number of suggestions were made in terms of other measures of biological health which could be incorporated into the index.

A single parasitoid species, A. bishopi, was reared from these larvae. In 2006 the highest parasitism rate, 11.43%, was recorded in May and in 2007, the highest parasitism rate, 13.27%, was in April. Agathis bishopi parasitizes larvae in instars 2 and 3, possibly due to the accessibility of these younger instars to the female parasitoid and possibly due to the length of the life cycle of this koinobiont. Second instar hosts yielded the highest number of parasitoids, and there was no emergence of parasitoids from fifth instar larvae. Females of A. bishopi live for 18.5 days (n = 20; S.E. = 3.1) and males for 8.25 days (n = 20; S.E. = 1.23). Females produce an average of 23 offspring in a lifetime, while female false codling moths produce about 800 eggs each. A high number of parasitoids will be required per hectare to reduce the population of false codling moth. Captive rearing of A. bishopi proved difficult due to viral and fungal contamination. Agathis bishopi has potential for use in an integrated pest management programme once the hurdle of mass-rearing has been overcome.

This case study, therefore, sought to investigate Red Pepper as a media phenomenon in Uganda in the 21st century, specifically between 2001 and 2004. Employing quantitative and qualitative methodologies, it focused on the production process and the text. Although sexualised content dominate its pages, and news about issues such as the environment and education are near-absent, its managers describe the publication as legitimate, normative and consistent with liberal media standards. Accordingly, to interrogate Red Pepper in terms of its journalistic functions, selected debates associated with liberal approaches to news media, media political economy, tabloidisation, pornography and gendered relations were reviewed. The analysis entailed five phases. The first was a denotative or descriptive analysis, which focused on the publication's structure and content focus. This was followed by an interview with management, a broad content analysis to establish the incidence of predefined content categories expected of the tabloid, pornographic and liberal press and, lastly, a theme-based content analysis that sought to establish the potential meanings and framing of the dominant content categories of gossip and sexualised copy. Overall, the study found Red Pepper to be a misogynistic tabloid, having elements said to belong to pornography and homophobia. According to the findings, not only does Red Pepper fall short of a liberal understanding of a newspaper in terms of diversity of topics, provision of information and professional practice, it also does not fit the understanding of an alternative public sphere, mainly because it fails to challenge the patriarchal framing of sex, sexuality and gendered relations. This framing is undertaken deliberately as a means to securing economic rather than journalistic ideals to which the editors pay lip service. Consequently, the gossip and sexualised content are not problematised and as such discourses and power relations therein are not interrogated. Neither are inadequacies in local systems addressed nor corrective action mobilised as expected of some tabloids. All in all, the publication fronts superficial entertainment content that echoes particular gender constructions and patriarchal commonsense and entrenches the (undesirable) status quo which, ironically, it claims to challenge.

- Description: The widespread acceptance of topical formulations intended for local and/or regional activity has prompted renewed interest in developing a model to determine the bioavailability of drugs in order to establish bioequivalence as a means of evaluating formulation performance of multisource products and also for use during formulation development. Current in vivo techniques such as blister suction and skin biopsy amongst others used to determine the bioavailability and/or bioequivalence of topical formulations are either too invasive to generate appropriate concentration-time profiles or require large numbers of study subjects thereby making the study expensive and time-consuming. Moreover, there are currently no sampling techniques that can demonstrate dermal bioavailability and/or bioequivalence of topical formulations intended for local and/or regional activity. Dermal microdialysis is a relatively new application of microdialysis that permits continuous monitoring of endogenous and/or exogenous solutes in the interstitial fluid. The technique is involves the implantation of semi-permeable membranes which are perfused with an isotonic medium at extremely slow flow rates and collection of microlitre sample volumes containing diffused drugs. Tape stripping, a relatively older technique, has been extensively used in comparative bioavailability studies of various topical formulations. However, due to shortcomings arising from reproducibility and inter-subject variation amongst others, the published FDA guidance outlining the initial protocol was subsequently withdrawn. The incorporation of transepidermal water loss with tape stripping has garnered renewed interest and has been used for the determination of drug bioavailability from a number of topical formulations. Hence the primary objective of this research is to develop and evaluate microdialysis sampling and tape stripping techniques, including the incorporation of the determination of transepidermal water loss, to assess the dermal bioavailability of ketoprofen from topical gel formulations and to develop models for bioequivalence assessment. A rapid UPLC-MS/MS method with requisite sensitivity for the analysis of samples generated from dermal microdialysis was developed and validated which accommodated the microlitre sample volumes collected. An HPLC-UV method was developed and validated for the analysis of samples generated from the in vitro microdialysis and in vivo tape stripping studies. The work presented herein contributes to a growing body of scientific knowledge seeking to develop a model for the determination of bioequivalence of pharmaceutically equivalent topical formulations intended for local and/or regional activity in human subjects.

- Description: Capsicum baccatum var. pendulum was first grown in the Makana District in 2005. Extremely little was known about best practices for cultivation or the insects and diseases associated with the crop in this area. The study was conducted during the second year of production, November 2005 and November 2006, in an attempt to identify the composition and phenology of insects occurring on C. baccatum. In the more rural parts of the Eastern Cape, and more particularly in Grahamstown, there are very few industries. With the advent of this new agricultural venture, a processing factory has been opened in Grahamstown creating more than 600 seasonal jobs in the factory and 1000 seasonal jobs on farms for local people. This business enterprise has not only brought about the creation of jobs, but also training and skills development and empowerment, generating much-needed income in this area. An extensive literature review yielded limited information on insect pests associated with Capsicum. Data from a pilot sampling trial undertaken were statistically analyzed to establish the number of plants to be scouted per site and the most effective scouting techniques to use. Based on the data available and insects collected during the pilot sampling trial, a surveillance programme was designed. Five different types of monitoring traps were placed in each of the eight study sites. Collection of trap catches and scouting of fifteen individual plants per site was undertaken on a weekly basis over the 52-week study period. The most commonly occurring potential insect pests were African Bollworm Helicoverpa armigera (Hübner), False Codling Moth Thaumatotibia leucotreta (= Cryptophlebia leucotreta) (Meyrick), Mediterranean Fruit Fly Ceratitis capitata (Wiedemann) and several species of thrips. Population densities of these pests and their phenology on Capsicum were determined. Statistical analyses established the efficacy of the monitoring traps for each pest, tested for differences among and between study sites, calculated an estimate of the number of pods damaged and a measure of plant damage.The results show that the majority of damage caused to the Capsicum baccatum cropping system was due to Mediterranean Fruit Fly populations. It was established that, although African Bollworm and False Codling Moth were present during the study period, their numbers were negligible and only nominal damage was caused by these pests. Damage caused by thrips species was apparent but not quantifiable. Intervention strategies using an Integrated Pest Management approach, are discussed.

With the advent of this new agricultural venture, a processing factory has been opened in Grahamstown creating more than 600 seasonal jobs in the factory and 1000 seasonal jobs on farms for local people. This business enterprise has not only brought about the creation of jobs, but also training and skills development and empowerment, generating much-needed income in this area. An extensive literature review yielded limited information on insect pests associated with Capsicum. Data from a pilot sampling trial undertaken were statistically analyzed to establish the number of plants to be scouted per site and the most effective scouting techniques to use. Based on the data available and insects collected during the pilot sampling trial, a surveillance programme was designed. Five different types of monitoring traps were placed in each of the eight study sites. Collection of trap catches and scouting of fifteen individual plants per site was undertaken on a weekly basis over the 52-week study period. The most commonly occurring potential insect pests were African Bollworm Helicoverpa armigera (Hübner), False Codling Moth Thaumatotibia leucotreta (= Cryptophlebia leucotreta) (Meyrick), Mediterranean Fruit Fly Ceratitis capitata (Wiedemann) and several species of thrips. Population densities of these pests and their phenology on Capsicum were determined. Statistical analyses established the efficacy of the monitoring traps for each pest, tested for differences among and between study sites, calculated an estimate of the number of pods damaged and a measure of plant damage.The results show that the majority of damage caused to the Capsicum baccatum cropping system was due to Mediterranean Fruit Fly populations. It was established that, although African Bollworm and False Codling Moth were present during the study period, their numbers were negligible and only nominal damage was caused by these pests. Damage caused by thrips species was apparent but not quantifiable. Intervention strategies using an Integrated Pest Management approach, are discussed.

This provides an illustration of some of the approaches to narrative form emergent in contemporary South African choreographic practices. Development of a novel in situ CPRG-based biosensor and bioprobe for monitoring coliform β-D-Galactosidase in water polluted by faecal matter

- Description: The ultimate objective of this work was to develop a real-time method for detecting and monitoring β-D-galactosidase as a suitable indicator of the potential presence of total coliform bacteria in water environments. Preliminary comparison of the chromogenic substrate, chlorophenol red β-D-galactopyranoside and the fluorogenic substrate, MuGAL, revealed unreliable results with the fluorogenic technique due to interference from compounds commonly found in environmental water samples. Thus, the chromogenic assay was further explored. Hydrolysis of the chromogenic substrate chlorophenol red β-D-galactopyranoside by β-D-galactosidase to yield chlorophenol red was the basis of this assay. Fundamental studies with chlorophenol red β-Dgalactopyranoside showed that β-D-galactosidase occurs extracellularly and in low concentrations in the polluted water environment. A direct correlation between enzyme activity and an increase in environmental water sample volume, as well as enzyme activity with total coliform colony forming unit counts were observed. Spectrophotometric detection was achieved within a maximum period of 24 h with a limit of detection level of 1 colony forming unit 100 ml[superscript -1]. This enzyme also exhibited physical and kinetic properties different from those of the pure commercially available β-D-galactosidase. Cell permeabilisation was not required for releasing enzymes into the extracellular environment. PEG 20 000 offered the best option for concentrating β-D-galactosidase. The source of β-D-galactosidase in the polluted environmental water samples was confirmed as Escherichia coli through SDS-PAGE, tryptic mapping and MALDI-TOF, thus justifying the further use of this method for detecting and/or monitoring total coliforms. Several compounds and metal ions commonly found in environmental water samples (as well as those used in water treatment processes) did have an effect on β-D-galactosidase. All the divalent cations except Mg [superscript 2+], at the concentrations studied, inhibited the relative activity of β-D-galactosidase in both commercial β-D-galactosidase and environmental samples. Immobilisation of chlorophenol red β-D-galactopyranoside onto a solid support material for the development of a strip bioprobe was unsuccessful, even though the nylon support material yielded some positive results. A monthly (seasonal) variation in β-Dgalactosidase activity from the environmental water samples was observed, with the highest activity coinciding with the highest monthly temperatures. Electro-oxidative detection and/or monitoring of chlorophenol red was possible. Chlorophenol red detection was linear over a wide range of concentrations (0.001-0.01 μg ml[superscript -1]). Interference by chlorophenol red β-D-galactopyranoside in the reduction window affected analysis. A range of phthalocyanine metal complexes were studied in an attempt to reduce fouling and/or increase the sensitivity of the biosensor. The selected phthalocyanine metal complexes were generally sensitive to changes in pH with a reduction in sensitivity from acidic pH to alkaline pH. The tetrasulphonated phthalocyanine metal complex of copper was, however, more stable with a minimum change of sensitivity. The phthalocyanine metal complexes were generally stable to changes in temperature. While only two consecutive scans were possible with the unmodified glassy carbon electrode, 77 consecutive scans were performed successfully with the CuPc-modified glassy carbon electrode. Among the phthalocyanine metal complexes studied, the CuPc-modified glassy carbon electrode therefore provided excellent results for the development of a biosensor. The CuPc modified-glassy carbon electrode detected 1 colony forming unit 100 ml[superscript -1] in 15 minutes, while the plain unmodified glassy carbon electrode required 6 hours to detect the equivalent number of colony forming units. CoPc, ZnPc and CuTSPc required 2, 2.25 and 1.75 h, respectively, to detect the same numbers of colony forming units. The CuPcmodified glassy carbon electrode detected 40 colony forming units 100 ml[superscript -1] instantly. In general, a direct correlation between colony forming units and current generated in the sensor was observed (R2=0.92). A higher correlation coefficient of 0.99 for 0-30 coliform colony forming units 100 ml[superscript -1] was determined. Current was detected in some water samples which did not show any colony forming units on the media, probably due to the phenomenon of viable but non-culturable bacteria, which is the major disadvantage encountered in the use of media for detecting indicator microorganisms. This novel biosensor therefore presents a very robust and sensitive technique for the detection and/or monitoring of coliform bacterial activity in water.

Education in the wetlands and wetlands in the education: a case of contextualizing primary/basic education in Tanzania

- Description: This dissertation describes an action research case study carried out at a sub-village school at Nyamakurukuru, Utete, Rufiji District, Tanzania. The study was a fully independent research activity funded and led by a female Irish environmental and community specialist who has fifteen years experience of working in rural Tanzania, five of which were in Rufiji District. The aim of the action research was to engage a community of villagers, teachers, students and district officers in a participatory process to adapt a module of a school curriculum to the local context, and teach it in order to describe one way in which contextualization, using local and indigenous knowledge and active discovery teaching-learning processes, can be done. The major research question, which I wished to answer for one specific case, was: Does integrating local environmental cultural knowledge into formal schooling contribute to curriculum relevance? If so, in what way? This document describes the background and context of the research, the motivation and the theoretical basis for the work, the methodology and methods, and the action research process itself. The results are interpreted and discussed in the light of current theoretical perspectives on education and environmental education. The main findings within the case are that: Contextualization improved relevance of education and thus its quality by: • breaking through traditional frames/barriers between teachers and students, students and elders and community and teachers, • allowing formal education to take place outside of the school, • necessitating a change in pedagogy1 to more learner-centered, discovery methods, • allowing indigenous knowledge to come into the classroom, • stimulating creativity and increased confidence, and • bringing local socio-political environmental issues into the classroom. This study provides a case example of how education processes, when engaging local cultural knowledge, can improve the relevance, and thus an aspect of the quality of teaching and learning in school-community contexts, while providing a conduit for integrating environmental education into the formal school curriculum. It provides insights into the key issue of relevance which currently faces educators of children in wetlands in Tanzania. Recommendations were made for the case studied and may be useful beyond the boundaries of the case: • Give more explicit government policy and strategic support for community involvement in educational content–epistemologies and pedagogies. • Weaken framing (hierarchical power positions) to encourage greater partnership between school, home and community to improve relevance. • Investigate the provision of education beyond schools. • Provide practical teacher and community training on use of learner-centered, discovery and active pedagogies. • Provide teacher and community education on biodiversity and the environment. • Provide relevant reference texts and research data on the ecology, biodiversity, vegetation, hydrology, agriculture, sociology, history and other relevant subjects. • Officially nurture a culture that learning should be enjoyable. • Allow the curriculum freedom, in these times of increasing risk for rural tropical wetland communities, to make the curriculum fit the local issues rather than vice versa. • Nurture critical analysis of the curriculum in local pedagogic discourse i.e., at the local contextualization level of the home, community and school.

The species include Chrysomya albiceps, C. putoria, C. chloropyga, C. megacephala, C. marginalis, C. inclinata and Calliphora croceipalpis. A robust experimental design for estimating developmental models is outlined and tested. It is recommended that forensic entomologists should involve at least six constant temperatures, starting at about 7°C above the relevant developmental zero (D0) and going to about 10°C above the upper critical temperature, and a temporal sampling interval with a relative precision of about 10%. Using this design, focused experiments consistently provided the most reliable developmental data, while data pooled from different studies yielded inconsistent results. Similarly, developmental data from closely related species differed significantly, and surprisingly so did developmental data from different populations of the same species. Possible explanations for the latter lay in the different methods of data collection but only temporal sampling resolution had a direct influence on the accuracy of developmental data. Consequently, disparities in such data were primarily ascribed to genetic differences and phenotypic plasticity. This case study has shown that farmers select seed to plant for food as a result of various learning interactions they engage in, which include inter-generational knowledge transfer, farmer to farmer extension and external training by extension organisations and NGOs. A communities of practice (COPs) (Wenger, 2007) framework was used to gain an understanding of the learning interactions among farmers and their stakeholders in Nyanga and Marange COPs of small grain farmers in Manicaland Province, Zimbabwe, who have been working with Environment Africa (A regional NGO). A critical realist causal analysis was conducted to unravel the various causal factors influencing choice. A number of underlying structures and causal mechanisms were found to influence learning interactions and choices in these communities of practice, and they include ambivalence, which influences the changing domain and practice. Climate change, drought and risk were found to affect farmer practice, while power relations affect the community, its practice, domain, sponsorship and the learning interactions in the COPs. The political economy was also found to have a profound effect on the domain and practice. A space was found for the influence of capacity and knowledge sharing in participatory frameworks of the communities, implying that extension quality can be enhanced to promote locally adapted and diverse seed varieties for food security improvement. The study shows that a deeper understanding of the mechanisms influencing the context of teaching and learning provides a more refined insight into the learning interactions and choices of farmers. This, coupled with the social processes descriptors provided by Wildemeersch (2007) has given me a more detailed understanding of the nature of learning interactions influencing farmer choices.

- Description: The larval fish assemblage in the shallow, nearshore region of a proposed marine protected area (MPA) in eastern Algoa Bay, temperate South Africa was investigated. Current velocities and direction and the swimming abilities of late-stage larvae were further assessed to determine potential larval movement to and from the MPA. In total, 6045 larval fishes were collected along two depth contours (~5m and ~15m) in the shallow nearshore of eastern Algoa Bay using stepped-oblique bongo net tows, twice per season for two years (March 2005 – January 2007). These larvae represented 32 families and 78 species. The Gobiidae, Cynoglossidae, Clupeidae, Engraulidae and Sparidae were the dominant fish families. Catches varied significantly between seasons peaking in spring with a mean of 64 larvae/100m3. Preflexion stage larvae dominated catches (75%). All developmental stages of Diplodus capensis, Engraulis capensis, Heteromycteris capensis, Sardinops sagax and Pomadasys species were found in the study area. It appears that these species use the shallow nearshore as a nursery area. Analysis of 12 months (May 2006 – May 2007) data from a bottom-moored Acoustic Doppler Current Profiler within the study area showed that offshore south eastward (39%) and onshore north westward currents (33%) dominated. The south westward current (15%) and north eastward current (12%) occurred less frequently. Current velocity decreased with depth in the nearshore, with a mean velocity of ~29 cms⁻¹ recorded at a depth of 4 m and a mean velocity of ~11 cms⁻¹ recorded at a depth of 14 m. Understanding the dispersal and movement of marine fish larvae in coastal habitats requires knowledge of active swimming abilities. The critical speed and endurance swimming of late stage larvae of two common inshore species occurring in the study area, Diplodus capensis and Sarpa salpa (Family Sparidae), were measured in a laboratory swimming chamber. The mean U-crit value for D. capensis (18.6 cms⁻¹) was similar to that of S. salpa (18.0 cms⁻¹), whereas mean endurance (km swum) was greater in S. salpa (8.4 km) than D. capensis (5.9 km). These swimming abilities exceed the average current velocities observed in the shallow nearshore providing larvae with the ability to greatly alter their passive dispersal trajectories and ultimately influence their distribution in the nearshore.

- Description: A great deal of research has demonstrated that Non-Timber Forest Products (NTFPs) play a crucial role in the livelihoods of the rural poor, and are particularly important to the most marginalised people throughout the developing world. However, these livelihood benefits are not without cost to the natural resource base that rural communities depend so heavily upon. The continued dependence on NTFPs as a major livelihood source must be contingent upon the minimisation of the level of disturbance created through this dependency. This study assesses the level of disturbance created through natural resource harvesting in eight study sites around South Africa, and applies a predictive conceptual model created by Shankaar et al. (2004b) to try and ascertain under what conditions the level of disturbance created through natural resource harvesting will be high. It assesses the three key factors that Shankaar et al. (2004b) identified (level of Local Ecological Knowledge (LEK), level of dependency and access to markets) in relation to the level of disturbance found at each of the study sites. It was found that there was a statistically significant relationship between the level of dependency and the level of disturbance, but there was no statistically significant relationship between either access to markets or the level of LEK and disturbance. Regulation of land use is a key issue, with weak local institutions in communal areas making effective resource management difficult. The significance of these findings is discussed, and priorities for future research are identified. This study adds to the body of knowledge related to NTFP harvesting and critically analyses the conflicts between the livelihood gains and the level of disturbance created through NTFP harvesting in an attempt to ascertain how livelihoods can be safeguarded. And in the longer-term, so that management strategies can be identified where resource extraction is not at the cost of undermining the very livelihoods that depend upon the natural resource base.

Plant aphid interactions : effects of diuraphis noxia and rhopalosiphum padi on the structure and function of the transport systems of leaves of wheat and barley

- Description: The infestation of the cultivated grain crops by phloem feeding aphids has generated a great deal of interest over the years, due to the serious damage they cause to the crops and yield losses that result. The mechanism of the interaction between aphids and host plants remains largely unknown in spite of efforts to understand the basis of aphid feeding on grain crops. Greater efforts are required to explain the mechanism(s) of this interaction in order to achieve sustainable agriculture. This thesis focused on an investigation of the mechanism of feeding by the Russian wheat aphid, Diuraphis noxia Mordvilko (RWA) and the bird cherry-oat aphid, Rhopalosiphum padi L. (BCA) on barley and wheat cultivars. These two aphids co-occur naturally, but they inflict very different feeding effects on host plants. Structural and functional approaches were employed to investigate their feeding habits and these were then related to the observed differences in their host plants. Transmission electron microscopy (TEM) techniques were used to study the ultrastructural damage, while fluorescence microscopy techniques – using aniline blue fluorochrome (a specific stain for callose) and 5, 6-CFDA (a phloem-mobile fluorophore) – were employed to investigate the functional response to damage via wound callose formation and phloem transport capacity respectively. RT-PCR and quantitative real-time RT-PCR techniques were used to investigate the regulation of the genes involved in callose synthesis and degradation at the transcriptional level. Morphological observation of the damage caused by the aphids show that infestation by RWA results in extensive leaf chlorosis, necrosis and rolling, while infestation by BCA does not lead to any observable symptoms within the same period. Interestingly, the population study shows that BCA breeds faster than RWA within the two-week experimental period. The ultrastructural study of feeding damage caused by the two aphids on the vascular bundles of susceptible barley cv Clipper, shows a different patterns of damage. Probing the vascular bundles results in the puncturing of vascular parenchyma by both aphids, but severe damage occurs in sieve tubes-companion cell complex during sustained feeding by RWA. In contrast, less damage occurs when BCA feeds on the phloem. Drinking from the xylem by RWA results in deposition of a large quantity of electron-dense watery saliva, which apparently seals the xylem vessels completely, by blocking all the pit membrane fields between the xylem vessels and associated parenchyma cells. In contrast, drinking from xylem by BCA results in deposition of a dense, granular saliva into the xylem vessels only, which does not appear to totally occlude the pit membrane fields. This is the first known report in which ultrastructural evidence of aphids’ drinking in xylem is provided. The comparative effects of RWA feeding on a susceptible Betta and resistant Betta-Dn1 wheat cultivars showed that after two weeks, the Betta cultivar expressed damage symptoms such as chlorosis, necrosis and leaf roll, while few chlorotic patches and necrotic spots occur in resistant Betta-Dn1 cultivars. An ultrastructural investigation of the feeding damage caused to all leaf tissues revealed, for the first time, that RWA is capable of both intra- and inter-cellular probing within mesophyll cells. Probing in the mesophyll cells induces a more severe damage in susceptible Betta than in the resistant Betta-Dn1 counterpart. Similar differences in damage occurred during feeding in the thin-walled sieve tubes of the phloem, with the sieve tubes of the Betta showing more damage than that of the resistant Betta-Dn1. However, drinking from xylem resulted in the characteristic occlusion of metaxylem vessels by copious deposition of saliva by RWA in both Betta and Betta-Dn1 cultivars. In all cases of probing, feeding, and drinking by RWA in both cultivars, all probed cells with evidence of salivary material deposit and those cells adjacent to salivary material deposit, exhibit significant damage in susceptible Betta cultivar, whereas similar cells in Betta-Dn1 cultivars do not show as damage as severe. Investigation of the functional response of the plants to feeding by aphids through the deposition of wound-induced callose shows that formation and deposition of wound callose occurs in both longitudinal and cross veins within 24h of feeding by RWA. This deposition increases through short-term feeding (72h) and prolonged feeding (14d). This is in sharp contrast to the observations with BCA feeding,where little or no callose formation occurs within the same time frame. Callose formation and deposition occurs only when a higher population of BCA feeds on barley leaves. This is the first report of aphid-induced wound callose by BCA. In all cases of callose deposition, aphid stylet tracks were associated with callose and the deposition of callose appears to be a permanent feature, because wound callose remained in the leaf tissues even after 120h of the aphids’ removal. Wound callose signals (defence and anti-defence) are discovered to be transported in the phloem tissues and are dependent on the direction of assimilate flow. Examination of the possible regulation of wound callose genes at the transcriptional level shows that the two expressed glucan synthase gene sequences (GSL – genes involved in callose formation) analysed did not show any significant increase or regulation upon aphid infestation. Contrary to expectation, all three aphid-induced β-1, 3-glucanases (genes which are thought to be involved in callose degradation) showed higher expression in RWA-infested tissue than in BCA-infested tissue. The results of the feeding damage on the transport capacity of the phloem shows that BCA infestation does not lead to a significant reduction in the phloem transport capacity during short-term feeding (72h), while RWA-infested leaves showed considerable reduction in the transport capacity of the phloem within the same period. However, prolonged feeding (14d) by BCA induces a considerable reduction on the transport capacity of the phloem on the infested tissues. In contrast, a marked reduction in the transport capacity of the phloem occurs in RWA-infested leaves and in most cases, complete cessation of transport ensues. In conclusion, these data collectively suggest that RWA is a serious and most destructive phloem feeder in comparison to the BCA. RWA causes severe damage to all cellular tissues of the host plants, which result in apoplasmic and symplasmic isolation of xylem and phloem tissues, while BCA infestation does not result in such isolation within the same time and population levels. Resistance genes appear to function by conferring resistance to cell damage on the resistant cultivars during aphid feeding. Responses by plants to aphid infestation via wound callose deposition are again shown to be species-specific. A quick response results when RWA feeds, even at a very low population level, while a response occurs only at a higher infestation level by BCA, and this response was shown as not regulated at the transcriptional level. Differences in the damage to leaf tissues and wound callose deposition eventually lead to varying degrees of damage to the transport capacity of the phloem. These differences in the damage signatures are hereby suggested to be the cause of the diversity in the observed damage symptoms and the yield losses upon infestation by the two aphid species.

The mechanism of the interaction between aphids and host plants remains largely unknown in spite of efforts to understand the basis of aphid feeding on grain crops. Greater efforts are required to explain the mechanism(s) of this interaction in order to achieve sustainable agriculture. This thesis focused on an investigation of the mechanism of feeding by the Russian wheat aphid, Diuraphis noxia Mordvilko (RWA) and the bird cherry-oat aphid, Rhopalosiphum padi L. (BCA) on barley and wheat cultivars. These two aphids co-occur naturally, but they inflict very different feeding effects on host plants. Structural and functional approaches were employed to investigate their feeding habits and these were then related to the observed differences in their host plants. Transmission electron microscopy (TEM) techniques were used to study the ultrastructural damage, while fluorescence microscopy techniques – using aniline blue fluorochrome (a specific stain for callose) and 5, 6-CFDA (a phloem-mobile fluorophore) – were employed to investigate the functional response to damage via wound callose formation and phloem transport capacity respectively. RT-PCR and quantitative real-time RT-PCR techniques were used to investigate the regulation of the genes involved in callose synthesis and degradation at the transcriptional level. Morphological observation of the damage caused by the aphids show that infestation by RWA results in extensive leaf chlorosis, necrosis and rolling, while infestation by BCA does not lead to any observable symptoms within the same period. Interestingly, the population study shows that BCA breeds faster than RWA within the two-week experimental period. The ultrastructural study of feeding damage caused by the two aphids on the vascular bundles of susceptible barley cv Clipper, shows a different patterns of damage. Probing the vascular bundles results in the puncturing of vascular parenchyma by both aphids, but severe damage occurs in sieve tubes-companion cell complex during sustained feeding by RWA. In contrast, less damage occurs when BCA feeds on the phloem. Drinking from the xylem by RWA results in deposition of a large quantity of electron-dense watery saliva, which apparently seals the xylem vessels completely, by blocking all the pit membrane fields between the xylem vessels and associated parenchyma cells. In contrast, drinking from xylem by BCA results in deposition of a dense, granular saliva into the xylem vessels only, which does not appear to totally occlude the pit membrane fields. This is the first known report in which ultrastructural evidence of aphids’ drinking in xylem is provided. The comparative effects of RWA feeding on a susceptible Betta and resistant Betta-Dn1 wheat cultivars showed that after two weeks, the Betta cultivar expressed damage symptoms such as chlorosis, necrosis and leaf roll, while few chlorotic patches and necrotic spots occur in resistant Betta-Dn1 cultivars. An ultrastructural investigation of the feeding damage caused to all leaf tissues revealed, for the first time, that RWA is capable of both intra- and inter-cellular probing within mesophyll cells. Probing in the mesophyll cells induces a more severe damage in susceptible Betta than in the resistant Betta-Dn1 counterpart. Similar differences in damage occurred during feeding in the thin-walled sieve tubes of the phloem, with the sieve tubes of the Betta showing more damage than that of the resistant Betta-Dn1. However, drinking from xylem resulted in the characteristic occlusion of metaxylem vessels by copious deposition of saliva by RWA in both Betta and Betta-Dn1 cultivars. In all cases of probing, feeding, and drinking by RWA in both cultivars, all probed cells with evidence of salivary material deposit and those cells adjacent to salivary material deposit, exhibit significant damage in susceptible Betta cultivar, whereas similar cells in Betta-Dn1 cultivars do not show as damage as severe. Investigation of the functional response of the plants to feeding by aphids through the deposition of wound-induced callose shows that formation and deposition of wound callose occurs in both longitudinal and cross veins within 24h of feeding by RWA. This deposition increases through short-term feeding (72h) and prolonged feeding (14d). This is in sharp contrast to the observations with BCA feeding,where little or no callose formation occurs within the same time frame. Callose formation and deposition occurs only when a higher population of BCA feeds on barley leaves. This is the first report of aphid-induced wound callose by BCA. In all cases of callose deposition, aphid stylet tracks were associated with callose and the deposition of callose appears to be a permanent feature, because wound callose remained in the leaf tissues even after 120h of the aphids’ removal. Wound callose signals (defence and anti-defence) are discovered to be transported in the phloem tissues and are dependent on the direction of assimilate flow. Examination of the possible regulation of wound callose genes at the transcriptional level shows that the two expressed glucan synthase gene sequences (GSL – genes involved in callose formation) analysed did not show any significant increase or regulation upon aphid infestation. Contrary to expectation, all three aphid-induced β-1, 3-glucanases (genes which are thought to be involved in callose degradation) showed higher expression in RWA-infested tissue than in BCA-infested tissue. The results of the feeding damage on the transport capacity of the phloem shows that BCA infestation does not lead to a significant reduction in the phloem transport capacity during short-term feeding (72h), while RWA-infested leaves showed considerable reduction in the transport capacity of the phloem within the same period. However, prolonged feeding (14d) by BCA induces a considerable reduction on the transport capacity of the phloem on the infested tissues. In contrast, a marked reduction in the transport capacity of the phloem occurs in RWA-infested leaves and in most cases, complete cessation of transport ensues. In conclusion, these data collectively suggest that RWA is a serious and most destructive phloem feeder in comparison to the BCA. RWA causes severe damage to all cellular tissues of the host plants, which result in apoplasmic and symplasmic isolation of xylem and phloem tissues, while BCA infestation does not result in such isolation within the same time and population levels. Resistance genes appear to function by conferring resistance to cell damage on the resistant cultivars during aphid feeding. Responses by plants to aphid infestation via wound callose deposition are again shown to be species-specific. A quick response results when RWA feeds, even at a very low population level, while a response occurs only at a higher infestation level by BCA, and this response was shown as not regulated at the transcriptional level. Differences in the damage to leaf tissues and wound callose deposition eventually lead to varying degrees of damage to the transport capacity of the phloem. These differences in the damage signatures are hereby suggested to be the cause of the diversity in the observed damage symptoms and the yield losses upon infestation by the two aphid species.
