Appraising the appraisal framework: evidence from Grahamstown property advertisements
- Authors: Beangstrom, Tracy
- Date: 2015
- Language: English
- Type: text , Thesis , Masters , MA
- Identifier: http://hdl.handle.net/10962/54744 , vital:26608
- Description: This thesis considers how interpersonal meaning choices in property advertisements are best reflected in a context of constraint in Grahamstown, South Africa, using and appraising Martin & White’s (2005) APPRAISAL Framework. The study is comparative in two senses since the data is analysed using Martin & White’s (2005) APPRAISAL Framework and a revision to it, and property advertisements from two competing estate agencies are analysed: Remax Frontier Properties and Pam Golding Properties. An initial strict application of Martin & White’s (2005) framework is followed by a second, based on analyst difficulties and framework limitations experienced in the first analysis, as well as those experienced by other researchers. Revisions to the original framework include added ‘local’, context-driven features and sub-categories: Exclusivity and Convenience in Attitude, a Scale of Intensity in Graduation, and a category ‘Invite’ in Engagement. These enable a richer, more detailed account of the alignment strategies and interpersonal micro-politics at play in the property advertisements than is possible using the original framework. Findings from the analyses reveal four facts of note. Firstly, that while the original Martin & White (2005) APPRAISAL Framework captures a general level of interpersonal meaning in the data, it does so more fully when it includes contextual and contextually-driven categories that are informed by local knowledge. Secondly, two levels of meaning are expressed in the data. One is aimed at an ‘external’ audience; the other, truer, fuller and more contentious, is aimed at what appears to be the intended audience only. Thirdly, and relatedly, specific contextual and cultural knowledge is needed by the intended audience to access the intended meaning. Fourthly, both estate agencies promote values of high prestige, exclusivity, elitism and wealth as their intended meaning to align a like-minded audience, although Remax Frontier Properties attribute these values to location and features of the home to a greater extent than Pam Golding Properties, who place greater emphasis on the size of the home. The thesis suggests further avenues for research into the discourse of property advertising, as well as into overcoming certain context-specific limitations of the APPRAISAL Framework.

The effects of the composition and structure of quantum dots combined with cobalt phthalocyanine and an aptamer on the electrochemical detection of prostate specific antigen

- Description: The ability to functionalize and modify graphene-based quantum dots has also allowed for the tuning of their activity in different applications. A comparative study is carried out in this work where a prostate specific aptamer (PSA) and cobalt phthalocyanine molecule are combined with graphene quantum dots (GQDs), nitrogen doped graphene quantum dots (NGQDs) and graphitic carbon nitride quantum dots (gCNQDs) in order to determine the effects of quantum dots type (and therefore composition) on the electrocatalytic ability of the sensor to detect the analyte of interest. For the detection of prostate-specific antigen, electrochemical impedance spectroscopy (EIS) is employed to determine detection limits as well as sensitivity and selectivity of the aptasensors. Results showed a good performance of all sensors fabricated with the NGQDs based aptasensor showing the best performance with a detection limit of 1.54 pM in EIS as well as negligible interference in mixed samples. This electrode was further verified in spiked human serum samples and good recovery and low detection limits were determined, putting this aptasensor in good stead as a reliable, efficient PSA aptasensor.

- Description: The Eastern Cape Karoo region of South Africa typically displays low rainfall and high temperatures, which is particularly concerning considering the shale gas exploration/extraction project proposed recently. Almost 90% of the fracking fluid consists of water and with the Karoo region already classified as a water-stressed region, the increased demand for freshwater for fracking could put further pressure on water resources of the region. By understanding the ecological functioning of the lower trophic levels, likely implications for higher trophic levels can be determined, and ultimately, recommendations for mitigation actions can be made. Here, observations aimed at understanding the dynamics of lower trophic levels are presented, that is, the microalgal biomass and assemblage composition within three characteristic surface waterbodies of the region. In addition, the work focuses on the smaller, more dynamic temporary wetland systems typical of this water-scarce region, with the aim of understanding the significance of single species macrophyte cover in relation to the lower trophic levels. Thirty-three waterbodies were sampled (natural depressions = 13, dams = 9, rivers = 11) during November 2014 (austral spring) and April 2015 (austral autumn). Waterbody physico-chemistry, hydro-morphometry, habitat cover and surrounding land-use, as well as microalgal biomass and assemblage composition within each waterbody type were measured. Thirteen natural depression wetlands were sampled during October 2015, December 2015 and March 2016. Once again, waterbody physico-chemistry, microalgal biomass and assemblage composition were measured. However, in addition, Schoenoplectus decipiens macrophyte cover and microalgal (diatom) species composition were also determined. In general, no significant differences in microalgal biomass between the three main waterbody types of the Eastern Cape Karoo region were found; however, the different waterbody types each displayed distinct microalgal assemblage compositions. Also, Schoenoplectus decipiens cover had negligible effects on microalgal biomass and assemblages in the temporary wetlands investigated here. These results suggest that the generalist nature of species occupying these systems allows them to adapt to the ever-changing conditions typical of this region. Ongoing monitoring initiatives considering impending hydraulic fracturing activities should include a comprehensive species-level diversity census, which could further inform management of any impacts associated with this potential disturbance.
