For a number of years, the company has been experiencing problems which have resulted in the final soda ash product being contaminated and discoloured. The problems experienced at Sua Pan have been reported also to occur in other salt works all over the world. It has been suggested that contamination in many salt works could be possibly be due to the microbial activity by halophilic algae and bacteria that grow in the solar ponds. This study was undertaken to investigate the nature of the contaminating organic compounds present in the brine, to identify the compounds, and to establish how these components vary during the various stages of the soda ash processing. For this study, two sets of brine samples were used; the first set was collected before the summer rains and the second set was collected after the summer rains. Solid bicarbonate and soda ash samples were also used. Extractions, desalting, UV and HPLC analysis and oxidative biotransformations using four enzymes, were used for developing profiles and characterizing the brine components. From these studies, we were able to confirm that the components of the brine are organic in nature. A thorough study of one of the compounds isolated,from solid bicarbonate and soda ash was conducted using UV, HPLC, IR, NMR, HPLC-MS, GC-MS and TLC. The results of these analyses, show that the. isolated compound was benzyl butyl phthalate which is generally regarded to be humic in nature. This compound was found to be present in all the brine samples collected after the summer rains including the well brine, suggesting this compound occurs naturally and is not formed during the processing.

An investigation into the possible neuroprotective role of melatonin in copper-loading

Copper is vital for life, as it is essential for critical enzymes in biological systems. It is free copper in the biological systems that is toxic, as free copper induces free radical generation, which disrupts lipid membranes, interacts with DNA causing mutations, and eventually leads to cell death. Wilson’s disease is a inherited copper disease, which results in hepatolenticular disease. Copper is unable to be excreted, and thus accumulates, eventually spilling over into the bloodstream from the liver, and “poisons” the patient. The Wilson’s disease patient leads a short life, due to neurological and hepatological problems. There is no cure for Wilson’s disease, only chelation therapy using potent chelators such as penicillamine and EDTA. Zinc, in high doses, can be used to compete with copper absorption. This has proved to be the only successful therapy at present. This study investigates the possible use of melatonin as a copper binder/chelator. Melatonin has been shown to interact with copper in vitro. By binding/chelating to copper, melatonin may inhibit copper-induced free radical generation, and thus prevent copper from interacting with DNA to cause mutations and act as a cytotoxin. In vivo studies on copper (2mg/kg) administered for 2-weeks and 6-weeks were carried out on Wistar rats. The potential of melatonin (12mg/kg) to prevent copper-induced cellular damage was investigated. The results indicate that melatonin does not protect the lipid membranes from copper-induced lipid peroxidation. In vitro investigations using 1mM, 5mM and 10mM copper and 5mM melatonin, show that melatonin prevents copper-induced lipid peroxidation at a copper concentration of 1mM (p<0.001). The 5mM and 10mM copper induces less lipid peroxidation, compared to the 1mM copper. It has been reported that metal ions, antioxidants and chelating agents can influence peroxide decomposition during the assay. Melatonin (5mM) administration does not significantly prevent copper-induced lipid peroxidation at 5mM and 10mM copper. It is possible that due to melatonin’s relatively low concentration, it is unable to inhibit lipid peroxidation induced by the copper. The chemical nature of the interaction between melatonin and copper was also investigated, using NMR, IR and electrochemistry techniques. The NMR and IR techniques show that melatonin coordinates with Cu²⁺ and not Cu¹⁺, at the carbonyl group of melatonin. The electrochemistry experiments using cyclic voltammetry and adsorptive stripping voltammetry, show that melatonin forms a strong bond with Cu¹⁺. Cu²⁺ prefers binding to oxygen, and that is clearly seen in the NMR and IR. Cu¹⁺ prefers binding to nitrogen and then oxygen, and this is seen in the electrochemistry, as Cu¹⁺ is forced to bind through one of the nitrogens on the melatonin. Previously, it has been shown that melatonin binds/chelates with Cu²⁺. Histochemical investigations show that copper administration for 2-weeks and 6-weeks, causes extensive mitochondrial damage in liver and kidney’s proximal convoluted tubule epithelium cells. Melatonin (12mg/kg) co-administration with copper for 2-weeks and 6-weeks did not significantly protect the mitochondria from copper-induced damage. Copper-specific stains (rhodanine, silver sulphide and rubeanic acid) were used to stain liver, brain and kidney tissue samples. Rhodanine and silver sulphide were equally sensitive in staining copper in the 2-week samples, but not at all in the 6-week samples. This could not be explained. Rubeanic acid was ineffective in all samples tested. Thus, it appears that specific copper stains cannot be used in making a definitive diagnosis in cases of copper overload, and that specific copper stains do not always correlate with a high concentration of copper present in tissues. Pineal organ culture was used to determine the effect of copper administration on pineal indole synthesis. Exogenous (³H) tryptophan was administered to the pineal organ cultures, and the level of (³H) pineal indoles synthesised, were measured. Pineals from 2-week and 6-week copper/melatonin treated animals exhibited paradoxical 5- methoxytryptophol (ML) levels, as compared to the 2-week and 6-week copper treated animals. The 2-week copper/melatonin administered animals, showed a decrease in the ML level (p<0.01), and the copper/melatonin administered for 6-weeks, showed an increase in the ML levels (p<0.01). This indicates that melatonin interacts with the HIOMT enzyme. Pineals from 6-week copper/melatonin treated animals, as compared to the 6-week copper treated animals, showed an increase in N-acetylserotonin levels. This indicates that melatonin prevents the inhibition of the NAT enzyme. The final experiment was to determine in vitro, the effect of Cu²⁺ and Cu¹⁺ administration, on mitochondrial electron transport chain. Rat liver homogenate was incubated with and solutions of Cu²⁺ (10mM) and Cu¹⁺ (10mM) and melatonin (10mM). Cu²⁺ administration caused an inhibition of the electron transport at t=0 and t=60, whereas Cu¹⁺ administration at t=0 caused an inhibition of electron transport, but at t=60, Cu¹⁺ administration stimulated electron transport. Melatonin administered with Cu²⁺, resulted in an inhibition of the electron transport chain at t=0 and t=60. The findings of this study indicate that melatonin might have a potentially beneficial effect in copper overloading, by binding/chelating copper.

Applicability of a health literacy test from the U.S. in a South African population

- Description: This thesis investigates the suitability and applicability of a health literacy test from the U.S. in a black, Xhosa-speaking, South African population. The concept of literacy is a controversial one which has been much debated, as it is not easy to classifY people as simply either literate or illiterate. As a result there are a number of definitions of literacy that vary with purpose and culture, but the most common one is that a person is literate if he/she can read and write. Estimating literacy from years of schooling is an inexpensive method but is also unreliable, since people generally read 3 to 5 grades below their stated educational level. This method affords little insight into the ability of patients to adequately function in a health care enviromnent, an ability which is referred to as functional health literacy. A number of health literacy tests such as the REALM (Rapid Estimate of Adult Literacy in Medicine) test have been developed to assess this skill. The REALM test is a word recognition test which places people into a relevant grade range estimate according to the number of words pronounced correctly. It appears to assume understanding of the word if the person is able to read that word correctly. In this project 125 black Xhosa-speaking respondents of varying educational levels who were literate in English were interviewed with the aid of an interpreter. Comprehensive demographic data were collected. Respondents were first asked to read all 66 words aloud during which time pronunciation was checked, and thereafter they were asked to explain each word. It was found that the ability to automatically decode and read the words did not necessarily guarantee comprehension of these words. Many of the words proved to be unfamiliar to the majority of the Xhosa respondents who were able to pronounce them correctly, but could not explain them. These tended to be phonetically transparent words which were therefore more accessible to the unfamiliar reader. This research has proven to be of great value in helping identify such words which should be substituted with simpler words for use in health information materials. A number of words could neither be pronounced nor understood by the population majority and, interestingly, a small group of words could not be pronounced but were satisfactorily explained by some respondents. The results showed an extremely poor correlation between the stated educational level and the REALM grade range estimate. This emphasizes the inappropriateness of years of formal schooling as an indicator of functional health literacy. The criteria were established for deciding cases in which the REALM test could be applied (or succeeds) and when it is inapplicable (or fails). It was found to be inapplicable in 41% of cases which clearly indicates that, in its current form, it is not a valid, reliable test to use in determining health literacy in this English second language population. It can, however, be used as a basis fur the development of a more appropriate test. Assembly of full-length cDNA, and heterologous expression, of Nudaurelia B virus RNA

- Description: Nudaurelia beta virus (NβV) is a monopartite genome virus belonging to the family Tetraviridae. Its host range has been found to be limited to a single insect order, the Lepidoptera (moths and butterflies). The single-stranded positive-sense RNA genome consists of 6625 nucleotides containing two open reading frames (ORFs). The 5' proximal ORF of 5778 nucleotides encodes a protein of 215 kDa containing three functional domains characteristic of RNA-dependent RNA polymerase. The 3' proximal ORF, of 1836 nucleotides, encodes the 66 kDa capsid precursor protein and overlaps the replicase gene by more than 99% and is in the +1 reading frame relative to the replicase reading frame. The full-length cDNA construct of the NβV genome was assembled using a homologous overlapping PCR linking method. The starting material consisted of seven overlapping pieces that were constructed for sequencing. Due to the degradation of the full-length RNA obtained from virus extracted from field-collected Nudaurelia cytherea capensis larvae other alternative methods needed to be applied. Sub-cloning using restriction enzyme sites also required an alternative method being used, due to the abundance of restriction sites of the same type in the NβV genome. This led to the use of a method similar to "DNA Shuffling" where overlapping pieces were connected using a modified PCR protocol. After the construction of the NβV genome, the full-length PCR product was cloned and checked for large insertion and deletions that could have resulted from the PCR amplification. The heterologous expression of the NβV capsid protein linked to a fusion protein (Glutathione S-transferase) in E.coli, confirmed the authenticity of the prescribed capsid gene ORF. The expression showed that the virus protein was subjected to protease digestion in DH5α E.coli, suggesting that the protein was insoluble in the cell cytoplasm. The capsid gene expression in a modified E.coli strain, Epicurian Coli BL21-CodonPlus (DE3)-RIL, resulted in high levels of the correct molecular weight protein with minimal degradation. The modified strain was designed for over-expression of eukaryotic protein with lowered protease activity. The above results have opened the way for further research that would yield valuable insight into the molecular biology and replication strategy of the NβV in cell cultures.

- Description: Bandwidth management is a topic which is often discussed, but on which relatively little work has been done with regard to compiling a comprehensive set of techniques and methods for managing traffic on a network. What work has been done has concentrated on higher end networks, rather than the low bandwidth links which are commonly available in South Africa and other areas outside the United States. With more organisations increasingly making use of the Internet on a daily basis, the demand for bandwidth is outstripping the ability of providers to upgrade their infrastructure. This resource is therefore in need of management. In addition, for Internet access to become economically viable for widespread use by schools, NGOs and other academic institutions, the associated costs need to be controlled. Bandwidth management not only impacts on direct cost control, but encompasses the process of engineering a network and network resources in order to ensure the provision of as optimal a service as possible. Included in this is the provision of user education. Software has been developed for the implementation of traffic quotas, dynamic firewalling and visualisation. The research investigates various methods for monitoring and management of IP traffic with particular applicability to low bandwidth links. Several forms of visualisation for the analysis of historical and near-realtime traffic data are also discussed, including the use of three-dimensional landscapes. A number of bandwidth management practices are proposed, and the advantages of their combination, and complementary use are highlighted. By implementing these suggested policies, a holistic approach can be taken to the issue of bandwidth management on Internet links.

A series of mixed ligand Re(V)-oxo complexes using N, N-diethyl-Nʾ-(R' )benzoylthiourea (LR'),N,N-morpholino-N' -(R')benzoylthiourea (morph-LR') and 8-(N-(R')benzoylthiocarbamoyl)-1,4-dioxa-8-azaspiro[4.5]decane ligands (spiro-LR') (where R' = H,CH₃,CI, OCH₃ and N0₂) ((1)(14) have been prepared by the reaction of [ReO₂(py)₄]CI and the ligand in the presence of sodium acetate in methanol. The solution state chemistry of these complexes has shown that complexes(1)-(14) (with the exception of (1b)) undergo dimerization in solution to give complex (15) in the presence of water. Preliminary ¹H NMR kinetics studies of the dimerization of (1), (6) and (7) to (15) have shown that the rate of dimerization decreases in the order (7) > (6) > (1). The rate of dimerization has also been compared for complexes (1), [ReO(morph-L)₂(OMe)] (8) and [ReO(spiro-L)₂(OMe)] (13) and the rate of dimerization was found to be fastest for (13). The crystal structures of (1), [ReO(LN0₂)₂(OMe)] (4), (6) and (15) have been determined. The Re(V)-oxo complexes (1), (4) and (6) have a slightly distorted octahedral geometry with the two acylthiourea ligands binding in a cis arrangement in the equatorial plane of the octahedron. The alkoxy and oxo ligands occupy the axial positions and are situated trans to each other. The crystal and molecular structure of complex (15), consist of two slightly distorted octahedral [ReO(L)₂] moieties bridged by an oxygen atom with a Re-O-Re bond angle of 175.2(2)°. The preliminary studies done in the present study have indicated that the complexation chemistry of technetium(V) with the N,N-diethyl-benzoylthiourea is different to that of rhenium (V). The reaction between [n-BuN₄][TcOCl₄] and N,N-diethyl-N'-benzoylthiourea yielded the square pyramidal cationic complex [TcO(L)₂]Cl. By contrast the octahedral methoxy complex [ReO(L )₂( OMe)] was obtained when the analogous Re(V)-oxo precursor, [n-Bu₄N] [ReOCI₄], was reacted with N,N-diethyl-N'-benzoylthiourea under the same reaction conditions.

Ecology and breeding biology of Lanner Falcons in the Eastern Cape Province, South Africa

- Description: Lanner Falcons Falco biarmicus are the commonest large falcons in Africa and this study in the coastal area of the Eastern Cape Province, South Africa investigated their ecology from 1984 to 2000. Lanners have recently been categorised as near-threatened and this study was initiated to ascertain if the population was stable. Breeding success of a small population, in different habitat types, was monitored intensively from 1997 to 2000, and the factors that inhibited breeding were investigated. The earliest onset of incubation was 17 July and the latest 16 August, with 2.2 young fledged per breeding pair over the four-year period. Incubation lasted 32 days and young fledged after 42-45 days. All nest sites were on cliffs with a mean height of 114 m, a mean vertical face of 53 m; 45% of the nests were in ravens’ nests. The aspect of cliff sites also influenced breeding success. Radio tracking was used to determine home range, habitat use and hunting methods, with prey species identified. Home ranges were between 66 km² and 249 km². Preferred prey was domestic chickens, pigeons and small birds during the breeding season. Foraging ranges were smaller in intensively cultivated areas with seed crops. Data obtained from ringing returns showed that no long distance movements occurred in this region, but two juveniles dispersed 152 km and 127 km. Conservation aspects with possible threats to Lanner Falcons such as poisons, electrocutions and direct persecution are discussed with some recommendations made for future research. Evidence from this study indicates that Lanner breeding success is not limited by nest site availability, but by rainfall timing and prey availability. Lanners foraged more in open areas than areas with tall vegetation, and benefited from intensive agriculture. This population appeared to be healthy and in no danger of declining in the near future.

Effect of feeding regimen, temperature and stocking density on growth and survival of juvenile clownfish (Amphiprion percula)

- Description: In aquaculture, a thorough knowledge of the specific environmental requirements of a species is needed in order to maximize growth rate and survival. There is a paucity of data regarding the fundamental environmental requirements for the ongrowing phase of clownfish juveniles. This prompted the design of three experiments to determine the best feeding regimen, temperature and stocking density that maximize growth and survival of Amphiprion percula. Ration size and feeding frequency are important factors for optimizing fish growth during the juvenile grow-out phase. A factorial growth trial was conducted to determine the effect of feeding frequency and ration size on the growth of juvenile clownfish (Amphiprion percula).Three feeding frequencies (1, 2 and 3 times daily) and six rations (2,4,6,8, 10 and 12 % body weight per day (BW.day⁻¹)) were used to test the growth response over a twelve week period. Non-linear regression analysis on the effect of ration, independent of feeding frequency, on growth resulted in a significant (n = 36; r² = 68.7) parabolic model: In y = -0.0302x² + 0.5159x + -4.4377. Maximum growth corresponded to a ration of 8.5% BW.day⁻¹. Survival as a function of ration was significantly lower at 2% BW.day⁻¹. Data were further examined with Analysis of CoVariance (ANCOVA) to determine the effect of ration on growth at each feeding frequency. The combination revealed a maximum growth rate when the fish were fed a ration of 10% BW.day⁻¹ divided into two equal meals. The required ration per meal to maintain maximum growth was also found to decrease as feeding frequency increased. The determination of the best temperature for growth is of great importance due to the direct relationship between fish metabolism and temperature. The thermal preferendum of A. percula has already been estimated at 26 ± 0.7°C but it is not known whether this closely approximates the temperature for maximum growth. Sixteen tanks were set to different temperatures ranging between 21.5 and 30.2 °C. Ten juvenile A. percula were placed in each tank and growth was measured fortnightly over the course of the 10 week experiment. Non-linear regression analysis resulted in significant models for fish length (y = -0.0005x² + 0.00267x - 0.0338; r² = 56.7, n = 11) and weight (y= -0.00016x² + 0.0084x - 0.1073; r² = 61.6, n = 11). These models predict that maximum growth would be at 27.7 and 27.1 °C for length and weight, respectively. Temperature, over the range tested, had no apparent effect on survival. There was no significant difference between the temperatures for maximum growth and the preferred temperature. The effect of stocking densities ranging from 0.2 fish.L¹ to 4.0 fish.L⁻¹ on growth were used in the third experiment. Enzymology of activated sewage sludge during anaerobic treatment of wastewaters : identification, characterisation, isolation and partial purification of proteases

- Description: During anaerobic digestion bacteria inside the digester require a carbon source for their growth and metabolism, sewage sludge was used as a carbon source in this study. The COD content was used to measure the disappearance of the substrate. COD content was reduced by 48.3% and 49% in the methanogenic and sulphidogenic bioreactors, respectively, while sulphate concentration was reduced by 40%, producing 70mg/L of hydrogen sulphide as the end product over the first 5-7 days. Sulphate (which is used as a terminal electron acceptor of sulphur reducing bacteria) has little or no effect on the sulphidogenic and methanogenic proteases. Sulphite and sulphide (the intermediate and end product of sulphate reduction) increased protease activity by 20% and 40%-80%, respectively. Maximum protease activity occurred on day 21 in the methanogenic reactor and on day 9 in the sulphidogenic reactor. The absorbance, which indicates the level of amino acid increased to 2 and 9 for methanogenic and sulphidogenic bioreactors, respectively. Proteases that were active during anaerobic digestion were associated with the pellet (organic particulate matter) of the sewage. These enzymes have an optimum activity at pH 10 and at temperature of 50°C. The proteases that were active at pH 5 and 7, had optimum temperatures at 30°C and 60°C, respectively. Due to their association with organic particulate matter, these enzymes were stable at their optimum temperatures for at least five hours at their respective pH. Inhibition by PMSF, TPCK and 1.10-phenanthroline suggested that proteases inside the anaerobic digester are a mixture of cysteine, serine and metalloproteases. At pH 5, however, EDTA appeared to enhance protease activity by 368% (three-fold). Acetic acid decreased protease activity by 21%, while both propionic and butyric acid at 200 mg/L cause total inhibition of protease activity while these acids at higher pH (where they exist as their corresponding salts) exerted little effect. Copper, iron and zinc inhibited protease activity by 85% at pH 5 with concentrations ranging between 200 and 600 mg/L. On the other hand, nickel, showed an increase in protease activity of nearly 250%. At pH 7 and 10, copper had no effect on protease activity while iron, nickel and zinc inhibited these enzymes by 20-40%. Proteases at pH 7 were extracted from the pellet by sonication, releasing 50% of the total enzymes into the solution. The enzymes were precipitated by ammonium sulphate precipitation, and further purified by ion exchange chromatography and gel filtration. Ion exchange chromatography revealed that most of the enzymes that hydrolyse proteins are negatively charged while gel filtration showed that their molecular weight is approximately 500 kDa.

The COD content was used to measure the disappearance of the substrate. COD content was reduced by 48.3% and 49% in the methanogenic and sulphidogenic bioreactors, respectively, while sulphate concentration was reduced by 40%, producing 70mg/L of hydrogen sulphide as the end product over the first 5-7 days. Sulphate (which is used as a terminal electron acceptor of sulphur reducing bacteria) has little or no effect on the sulphidogenic and methanogenic proteases. Sulphite and sulphide (the intermediate and end product of sulphate reduction) increased protease activity by 20% and 40%-80%, respectively. Maximum protease activity occurred on day 21 in the methanogenic reactor and on day 9 in the sulphidogenic reactor. The absorbance, which indicates the level of amino acid increased to 2 and 9 for methanogenic and sulphidogenic bioreactors, respectively. Proteases that were active during anaerobic digestion were associated with the pellet (organic particulate matter) of the sewage. These enzymes have an optimum activity at pH 10 and at temperature of 50°C. The proteases that were active at pH 5 and 7, had optimum temperatures at 30°C and 60°C, respectively. Due to their association with organic particulate matter, these enzymes were stable at their optimum temperatures for at least five hours at their respective pH. Inhibition by PMSF, TPCK and 1.10-phenanthroline suggested that proteases inside the anaerobic digester are a mixture of cysteine, serine and metalloproteases. At pH 5, however, EDTA appeared to enhance protease activity by 368% (three-fold). Acetic acid decreased protease activity by 21%, while both propionic and butyric acid at 200 mg/L cause total inhibition of protease activity while these acids at higher pH (where they exist as their corresponding salts) exerted little effect. Copper, iron and zinc inhibited protease activity by 85% at pH 5 with concentrations ranging between 200 and 600 mg/L. On the other hand, nickel, showed an increase in protease activity of nearly 250%. At pH 7 and 10, copper had no effect on protease activity while iron, nickel and zinc inhibited these enzymes by 20-40%. Proteases at pH 7 were extracted from the pellet by sonication, releasing 50% of the total enzymes into the solution. The enzymes were precipitated by ammonium sulphate precipitation, and further purified by ion exchange chromatography and gel filtration. Ion exchange chromatography revealed that most of the enzymes that hydrolyse proteins are negatively charged while gel filtration showed that their molecular weight is approximately 500 kDa.

- Description: The blue duiker, Philantomba monticola and common or grey duiker, Sylvicapra grimmia represent two of the three duiker genera as well as two of the three species occurring in Southern Africa. The two species have not been adequately studied in their habitats within the Eastern Cape Province of South Africa, and this thesis centres around their biology with focus on diet and dental microwear. Faecal analysis was used to study the diet of blue duiker at Salem in the Albany district, and of common duiker on two farms, one a predominantly cattle farm at Kasouga and the other a farm for cultivation of chicory at Grants valley, both in the Bathurst district. The validity of faecal analysis was reviewed, and its applicability to blue and common duiker assessed. The method was appropriate for diet analysis of both species, as the common duiker is shy and secretive and the blue duiker is a rare and protected species which does not habituate readily. Results showed that the blue duiker was mainly folivorous with a seasonally stable diet of 79 percent dicot foliage and only 17 percent fruit. Common duiker diet at both study sites comprised mostly dicot foliage, with only two monocotyledonous plant species. Twenty-seven and nineteen plant species were identified in the diets of common duiker at Kasouga and Grants valley respectively and the annual percentage occurrences of dicot foliage in the diets were about 99 percent at both study sites. The predominantly browsing common duiker, therefore offered negligible competition for food resources to the grazing cattle on Kasouga farm. Both blue and common duiker fed selectively, with approximately one third of the total number of plant species identified in their diets providing at least 50 percent of the food eaten annually. Ehretia rigida was the most important plant species in the diets of both duiker species. Chicory, Chichorium intybus provided more than one third (35.6 percent) of the winter diet and a substantial proportion (14.4 percent) of the spring diet of common duiker at Grants valley, thereby confirming earlier reports of this species feeding on chicory and other cultivated crops. The diet of common duiker at Kasouga did not vary seasonally but that of common duiker at Grants valley did vary and this was attributed to utilisation of chicory. Results from the dental microwear analyses did not show any significant differences in dental microwear between blue and common duiker, but supported and confirmed that the two were browsing species, characterised by many pits and few scratches on their dental surfaces. A high incidence of pits was found on the dental surfaces of both duiker species, and was attributed to utilisation of fruit in the diet.

These dosage forms facilitate patient compliance by simplifying the dosage regimen, and decrease the risk of adverse effects by reducing large fluctuations in the plasma concentration of the drug. The objective of this study was to formulate a repeat-action tablet to provide a sustained release dose of pseudoephedrine sulfate (PSS), and an immediate release dose of both PSS and loratadine. The release profile was compared to that of a commercially available preparation, Clarityne-D®. This formulation developed presents a novel mechanism of sustaining the release of PSS. The prototype tablet consisted of a sustained release core coated with an ethylcellulose dispersion to introduce a lag phase into the release profile and a second outer film coat incorporating PSS and loratadine. The core comprised an ethylcellulose granulation of PSS compressed into a hydroxypropyl methylcellulose matrix. The release of PSS from prototypes was assessed using USP Apparatus 3, as this apparatus was more representative of in vivo conditions and discriminated more effectively between the different tablet compositions produced during development. All dissolution samples were analysed for PSS and loratadine using validated highperformance liquid chromatographic methods. The prototype sustained release cores were found to be more resistant than the reference product to elevated temperature and humidity (40°C/87% RH) with fewer observed changes to the release profiles following storage for up to six months. This study was a feasibility study to obtain proof of concept. The release profile obtained from the prototype tablets was similar (f₂ = 50.0) to that of the reference product. Further development and optimisation of this dosage form is necessary, including evaluation of the choice of hydrophobic polymer, the effect of compression force and tablet geometry and characterisation of the release mechanism from the coated matrix. Assessment of these factors is necessary in order to optimise the formulation with respect to the desired therapeutic objectives.

Ichthyofaunal community structures in different types of Eastern Cape estuaries

- Description: This investigation had three major objectives; firstly to document the estuarine fishes from several poorly studied Eastern Cape estuaries, secondly, to identify similarities and differences in the fish communities from permanently open and intermittently open estuaries, and thirdly, to establish which physico-chemical or environmental variables are responsible for the structuring of these communities. The study area incorporated 10 estuaries along a 70 km long stretch of the Eastern Cape coastline between the towns of Seafield (33° 32' 42" S, 27° 03' 05" E) in the south-east and Hamburg (33° 16' 45" S, 27° 29' 50" E) in the north-west. The systems investigated included two permanently open estuaries and eight intermittently open systems of varying sizes. Overall fish abundance and species richness in the study area revealed no significant seasonal variation. A contrast between open and closed estuaries was evident in the dominance of species from different estuarine dependence categories. Open estuaries comprised mainly category Ib, lla and IIb species, with a high proportion of category IV species. Closed estuaries were dominated by category IIa species, with few category IV species represented. Clear longitudinal trends in relative abundance were demonstrated for individual species. Margalefs species richness index for seine net catches, and a combination of the seine and gill net data, were significantly correlated with catchment size, mean annual runoff, estuarine area and linear length. Fish assemblages in the permanently open and intermittently open estuaries were significantly different (p<0.001) when analysed on a community and density basis. Similarly, the communities in large and small intermittently open estuaries differed significantly on a community (p=0.01) and density basis (p<0.001). These differences where caused by changes in abundance of the dominant estuarine resident and marine migrant species. The environmental variable that was dominant in accounting for these differences was the estuary mouth status. Estuarine resident species had smaller overall body lengths when compared with the marine migrant species. In addition, the estuarine resident species had smaller modal size classes in the closed estuaries when compared with the open estuaries. These differences may be linked to the foraging strategies of the species, as well as food availability and different levels of predation in the different estuary types. The marine migrant species did not show any trends in this respect although the closed systems had the largest individuals for all four dominant marine species. This finding may be linked to the inability of these species to breed in closed estuaries, thus channelling reproductive energy resources into somatic growth. In addition, prolonged periods of mouth closure prevent the emigration of large individuals back to the sea. Otter trawling was conducted in fiye estuaries and captured mostly demersal species. The otter trawl data confirmed the seine net community analysis, with significantly different communities identified in the permanently open and intermittently open estuaries (p=0.02), as well as in the small and large intermittently open estuaries (p=0.03). The environmental variables responsible for these community differences were mouth status and variables associated with estuarine size. The identification of estuary mouth status by this study as the most influential environmental factor is compounded by this "ariable affecting or being indicative of numerous other physico-chemical features. Ibuprofen may be administered in a number of different forms via the oral as well as the topical route. Published evidence suggests that topical, unlike oral, non-steroidal anti-inflammatory drugs are associated with few systemic side effects as plasma concentrations are low compared to oral therapy. In some countries it is particularly difficult to obtain human skin for in vitro experimentation and it is therefore important to have alternate biological or synthetic membranes which mimic human skin for diffusion experiments. Synthetic membranes serve as predictive models for topical drug release and in South Africa, shed snake skin is easily obtainable from the many snake parks present in the country. The FDA guidelines were considered when choosing the apparatus to be used in the comparative diffusion study on proprietary ibuprofen-containing topical preparations from three countries and the verification of the usefulness, or otherwise, of shed snake skin as a biological membrane for the assessment of the permeation of ibuprofen. Two diffusion techniques were considered appropriate for the measurement of the amount of ibuprofen released from a topical formulation during in vitro testing. One was the Franz diffusion cell, as modified by Keshary and Chien (88,169) and the other was the European Pharmacopoeia diffusion cell (187). High performance liquid chromatography was used as the analytical technique for the analysis of ibuprofen in aqueous solution using ultraviolet detection at 222 nm. The validated method was applied to the determination of the diffusion of ibuprofen from topical ibuprofen-containing formulations (gels, creams and mousse) through synthetic silicone membrane and shed snake skin biological membrane from four different species. In a study of fifteen topical ibuprofen-containing formulations (gels, creams and mousse) from three countries (South Africa, United Kingdom and France) it was found that there was a trend of products from two countries consistently exhibiting superior diffusion characteristics as well as products from the same two countries consistently exhibiting the lowest diffusion of ibuprofen. Interpretation of the results of these studies demonstrated the importance of employing a combination of statistical analyses and peak integration values when drawing conclusions regarding comparative diffusion characteristics. Shed snake skin has been described as a 'model' membrane, i.e. a membrane which shows similar permeability to human stratum corneum. The results reported here show clearly that, for ibuprofen, the four species of snake produce shed skin with completely different diffusion characteristics when all other conditions are identical. It may well be that there is one particular species of snake which produces shed skin of identical permeability to human stratum corneum, but to describe shed snake skin in general as a model membrane seems incorrect. It is therefore important that if shed snake skin is used as a membrane, the species, skin site and orientation should be reported. The European Pharmacopoeia diffusion apparatus was judged to be the better of the two diffusion techniques assessed for the measurement of the amount of ibuprofen released from a topical formulation during in vitro testing using silicone membranes and for the measurement of the amount of ibuprofen diffusing across the ventral outside orientation of shed skin during in vitro testing, whereas the Franz diffusion apparatus was judged to be better for the measurement of the amount of ibuprofen diffusing across the dorsal outside orientation of shed skin during in vitro testing. However, the choice of this diffusion apparatus must be weighed against the relatively poor reproducibility as compared with the European Pharmacopoeia diffusion apparatus.

Two diffusion techniques were considered appropriate for the measurement of the amount of ibuprofen released from a topical formulation during in vitro testing. One was the Franz diffusion cell, as modified by Keshary and Chien (88,169) and the other was the European Pharmacopoeia diffusion cell (187). High performance liquid chromatography was used as the analytical technique for the analysis of ibuprofen in aqueous solution using ultraviolet detection at 222 nm. The validated method was applied to the determination of the diffusion of ibuprofen from topical ibuprofen-containing formulations (gels, creams and mousse) through synthetic silicone membrane and shed snake skin biological membrane from four different species. In a study of fifteen topical ibuprofen-containing formulations (gels, creams and mousse) from three countries (South Africa, United Kingdom and France) it was found that there was a trend of products from two countries consistently exhibiting superior diffusion characteristics as well as products from the same two countries consistently exhibiting the lowest diffusion of ibuprofen. Interpretation of the results of these studies demonstrated the importance of employing a combination of statistical analyses and peak integration values when drawing conclusions regarding comparative diffusion characteristics. Shed snake skin has been described as a 'model' membrane, i.e. a membrane which shows similar permeability to human stratum corneum. The results reported here show clearly that, for ibuprofen, the four species of snake produce shed skin with completely different diffusion characteristics when all other conditions are identical. It may well be that there is one particular species of snake which produces shed skin of identical permeability to human stratum corneum, but to describe shed snake skin in general as a model membrane seems incorrect. It is therefore important that if shed snake skin is used as a membrane, the species, skin site and orientation should be reported. The European Pharmacopoeia diffusion apparatus was judged to be the better of the two diffusion techniques assessed for the measurement of the amount of ibuprofen released from a topical formulation during in vitro testing using silicone membranes and for the measurement of the amount of ibuprofen diffusing across the ventral outside orientation of shed skin during in vitro testing, whereas the Franz diffusion apparatus was judged to be better for the measurement of the amount of ibuprofen diffusing across the dorsal outside orientation of shed skin during in vitro testing. However, the choice of this diffusion apparatus must be weighed against the relatively poor reproducibility as compared with the European Pharmacopoeia diffusion apparatus.

Transmissivities from pumping tests are within the range 4.7 m²/d to 13.6 m²/d with an average of 8.9m²/d. The low transmissivities seem to be a major limiting factor in the exploitation of the groundwater resources. Thus the sustainable borehole yields tend to be small, mean values ranging from 33 m²/d to 253 m²/d. Specific yield could not be determined from the pumping tests due to the lack of observation boreholes. Low chemical concentrations render the water suitable for irrigation of all crops, while neither total nor any individual concentrations present health hazards to human or livestock. An average recharge value of 47.7 mm/y was inferred from water table fluctuation method. Chloride mass balance technique in the same area indicates recharge value in the order of 67.4 mm/y. Because the chloride mass balance gives a long-term mean annual recharge, the recharge figure of 67.4 mm/y was adopted for the study area. Based on the abstractable proportion of recharge, the sustainably exploitable volume of groundwater of the order of 2.68 x 10⁷ m³/y was established. This volume is more than 100 times the estimated current demand for groundwater (1.35 x 10⁵ m³/d), implying that there are large volumes of surplus water, which can be utilised for irrigation.

Cardiac responses were measured using heart rate monitoring and perceptual measures assessed using Borg’s (1971) RPE scale. The results of the testing showed significant differences in agonist and antagonist responses at all three testing speeds, the only exception being slow speed trunk values (peak torque). Upper- to lower-extremity ratios highlighted a possible weakness in the elbow flexors group, while correlations between LTs and OSTs highlighted the specificity of strength principle, as poor relationships were observed.

Novel approaches to the synthesis of quinoline derivatives

- Description: The investigation has been concerned with the application of the Baylis-Hillman methodology to the synthesis of quinoline derivatives. An extensive range of novel Baylis-Hillman products has been prepared, typically in moderate to excellent yields, by condensing 2-nitrobenzaldehyde derivatives with various vinyl ketones and acrylic esters in the presence of diazabicyclo[2.2.2]octane (DABCO). Reduction of the nitro group in the Baylis-Hillman products was effected by catalytic hydrogenation in ethanol using a 10% palladium-on-carbon catalyst to afford quinoline, quinoline-N-oxide and quinolone derivatives. In all cases, it is apparent that cyclisation involves exclusive attack of nucleophilic nitrogen at the carbonyl centre, with acrylic ester derivatives affording quinolones and vinyl ketone derivatives affording quinolines and the corresponding quinoline-N-oxides. No products arising from a conjugate addition pathway were observed. The use of stannous chloride as an alternative reagent to effect reductive cyclisation of the Baylis-Hillman products has been explored, and found to favour the formation of 1,2- dihydroquinoline derivatives, with cyclisation occurring via a conjugate addition pathway. Isolation of the products, following work-up of the stannous chloride reactions, however, presented some difficulty. All compounds were characterised by spectroscopic (NMR and IR) and, where appropriate, elemental (high-resolution MS) analysis. Interconversion of the quinoline and quinoline-N-oxide derivatives has been explored and finally achieved in quantitative yields. Reduction of 2,3-dimethylquinoline-N-oxide to the corresponding quinoline was effected using phosphorus tribromide in DMF, and the reverse transformation with meta-chloroperbenzoic acid (MCPBA) in CHCl₃. Photosensitizing properties of non-transition metal porphyrazines towards the generation of singlet oxygen

- Description: Metallophthalocyanine complexes containing non-transition metals are very useful as sensitizers for photodynamic therapy, a cure for cancer that is based on visible light activation of tumour localized photo sensitizers. Excited sensitizers generate singlet oxygen as the main hyperactive species that destroy the tumour. Water soluble sensitizers are sought after for the convenience of delivery into the body. Thus, phthalocyanine (pc), tetrapyridinoporphyrazines (tppa) and tetramethyltetrapyridinoporphyrazines (tmtppa) with non-transition central metal atoms of Ge, Si, Sn and Zn were studied. First was the synthesis of these complexes, followed by their characterisation. The characterisation involved the use of ultraviolet and visible absorption spectroscopy, infrared spectroscopy, nuclear magnetic resonance spectroscopy, electrochemical properties and elemental analysis. Photochemical properties of the complexes were then investigated. Photolysis of these macrocycles showed two processes; -reduction of the dye and photobleaching, which leads to the disintegration of the conjugated chromophore structure of the dye. Photobleaching is the reductive quenching of the excited state of the sensitizers. The intensity of the quenching decreased progressively from tmtppa, tppa to pc metal complexes with photobleaching quantum yields, 6.6 x 10.5⁻¹, 1.8 x 10.5⁻¹ and 5.4 x 10⁻⁶ for Zntmtppa, Zntppa and Znpc, respectively. Efficiency of singlet oxygen sensitization is solvent dependent with very different values obtained for the same compound in different solvents, for example, 0.25 and 0.38 were observed as singlet oxygen quantum yields for Gepc complex in DMSO and DMF respectively. In DMSO the efficiency of ¹O₂ generation decrease considerably from pc to tppa and finally tmtppa. In water Getmtppa exhibits much higher singlet oxygen quantum yield, hence promising to be effective as a sensitizer for photodynamic therapy.

Processes and products in the kimberlitic crater facies of the south lobe, Jwaneng Mine, Botswana

- Description: The Pennian (~ 245 Ma) Jwaneng kimberlite, situated in southern Botswana, comprises three steep-sided pipes that coalesce approximately 100m below the present day surface to fonn a 54ha body. These pipes have been labelled the South, Centre and North Lobes. The kimberlites intruded a thick sequence of Proterozoic shales, dolomites and sandstones and a thin veneer of consolidated to poorly consolidated mudstones and siltstones of the Karoo Supergroup. Although the shapes of these pipes are comparable to other southern African pipes, they are filled predominantly with crater facies volcaniclastic kimberlite. No tuffisitic kimberlite breccia, the characteristic rock type of the diatremes of other southern African pipes, has yet been identified. The Jwaneng kimberlite thus represents an exception to the standard model for southern African kimberlites, implying that different processes need to be invoked to explain its fonnation. The present study involves a detailed volcanological and sedimentological analysis of the volcaniclastic fill of the Jwaneng South Lobe. Two principal and distinct lithofacies have been identified: the quartz-free RVK facies and the quartz-bearing QRVK facies. Both facies include fine to coarse grained, predominantly massive and subordinate chaotically bedded deposits. The volcaniclastic rocks have been classified as resedimented volcaniclastic kimberlite (RVK) , since their deposition is ascribed to mechanisms dominated by mass flow processes. Based on certain characteristics and differences between the two principal facies, and their spatial distribution within the pipe, they are interpreted as being the products of at least two separate eruption episodes. Certain characteristics (e.g. }hape, granularity~ of the juvenile· magma clasts III the volcaniclastic kimberlite suggest complete crystallisation and devolatilisation of the magma at depth prior to explosive fragmentation. A scenario in which this might have occurred, and which led to catastrophic explosive eruption and pipe excavation is proposed. Explosive eruption and associated tuff cone formation is followed by resedimentation of the material back into the pipe by mass flow processes. Mass flow processes are dominated by debris flow, with lesser grain flow, hyperconcentrated flow and subaqueous mud flow and suspension settling of muddy kimberlitic sediments. Geochemical analyses of the latter indicate a high degree of contamination and weathering, and mixing between pristine kimberlite and silicic shale/mud compositions. Failure and collapse of parts of the underlying pipe walls yielded megablocks of poorly consolidated Permian Karoo mudstone in the peripheral zone of the pipe. This source of the megablocks is supported by their bulk chemical composition. Minor phreatic/phreatomagmatic eruptions are suggested by the presence of rare accretionary and armoured lapilli within both the QRVK and RVK facies. Subsidence of the volcaniclastic pipe fill, inferred mainly from the oversteepened dips of the bedded QRVK and RVK facies, may be related to gravity-induced compaction, late-stage phreatomagmatic eruptions or eruption ofthe adjacent Centre Lobe.

Subsidence of the volcaniclastic pipe fill, inferred mainly from the oversteepened dips of the bedded QRVK and RVK facies, may be related to gravity-induced compaction, late-stage phreatomagmatic eruptions or eruption ofthe adjacent Centre Lobe.

Removal of copper and nickel from solution by the non-viable biomass of the water fern Azolla filiculoides in an upscaled fixed-bed column system

- Description: The potential of non-viable Azalia filiculaides for the removal of Cu and Ni from aqueous solutions and the possibility of scaling up existing lab scale Azalia column systems was investigated. The effects of factors such as metal starting concentration, pH and two metals in solution on the removal of Ni and Cu from aqueous solution by dried and crushed Azalia biomass were studied in batch systems. Aqueous solutions of Ni with starting concentrations between 1000 and 2000J.lmolll gave the most efficient Ni removal by Azalla biomass. For Cu the optimum starting concentration for adsorption was 50J.lmol/l. The adsorption capacity of both eu and Ni increased as the starting pH of the sorption media increased. The optimum pH for Ni adsorption was found at pH 7 and for Cu, at pH 5. - Awlla biomass had a higher. maximum binding capacity (qrnax) for Cu than for Ni at pH 5. The removal of both Cu allct Ni showed little or no variation with the presence another metal in solution. Kinetic studies show that both Cu and Ni adsorbed rapidly onto the Azalia biomass. The removal of Cu and Ni from aqueous solutions using non-viable Azalia biomass was investigated in a lab scale fixed-bed column and an upscaled 4L column system. The nonviable Azalla filiculaides biomass when dried and used in a column for adsorption of Cu and Ni showed good physical stability under many different conditions. Preparation of the biomass before it could be used in the columns was very simple and did not involve any significant pretreatment steps. Prolonged exposure to UV light decreases Azalia biomass capacity for Ni and Cu adsorption. Column adsorption of Cu and Ni from aqueous solutions was successfully upscaled approximately 100 times. Relative to the lab scale column, the 4L column performed better for the uptake of Cu and Ni per gram of biomass. The larger column was also able to operate at relatively higher flow rates. The biomass showed good reusability with little change in the amount of Ni adsorbed in 10 consecutive cycles. Electron micrographs showecf little or no change in the physical structure and integrity of the Azolla biomass after exposure to mineral acids, Ni solution and high flow rates over 10 consecutive adsorption and desorption cycles. As much as 80% Ni and 70 % Cu was recovered when desorption profiles were generated using O.lMHCI as a desorption agent. The 4L column system was also tested using a highly concen~rat:~ Ni plating bath solution.(Nicrolyte 1). Only 18 % of the Ni could be removed from the expended Nicrolyte 1 pla~Jng solution after treating only 25L, indicating that Azolla biomass is more suited for removal of metals from more dilute industrial effluents.
