- Description: This research explores the lived experience of being a vertically or prenatally-infected HIV-positive adolescent. It looks specifically at how the participants experience intimacy, sex and sexuality. HIV/AIDS remains a global pandemic and vertically-infected adolescents are a growing new demographic. However, there is a poverty of research, and therefore interventions and support, for this demographic. This qualitative research conducted six individual, in-depth, semi-structured, psychoanalytic research interviews with three participants. The interviews were structured around projective drawings that the participants did in the course of each interview. The interviews were transcribed and analysed using psychodynamic object relations theory and organised through interpretative phenomenological analysis. Every effort was made to ensure that the research was conducted ethically and validly. The analysis found that the participants' experience of intimacy has resulted in a self that is constantly under threat. This in turn has negatively impacted on the participants' experience of sex and sexuality. The implication of this research is that more in-depth research needs to be done into this demographic so that better interventions and support may be offered.

- Description: In this thesis we consider invariant control systems and Hamilton-Poisson systems on the three dimensional semi-Euclidean group SE(1,1). We first classify the left-invariant control affine systems (under detached feedback equivalence). We provide a complete list of normal forms, as well as classifying conditions. As a corollary to this classification, we derive controllability criteria for control affine systems on SE(1,1). Secondly, we consider quadratic Hamilton-Poisson systems on the (minus) Lie-Poisson space se(1,1)*. These systems are classified up to an affine isomorphism. Six normal forms are identified for the homogeneous case, whereas sixteen representatives (including several infinite families) are obtained for the inhomogeneous systems. Thereafter we consider the stability and integration of the normal forms obtained. For all homogeneous systems, and a subclass of inhomogeneous systems, we perform a complete stability analysis and derive explicit expressions for all integral curves. (The extremal controls of a large class of optimal control problems on SE(1,1) are linearly related to these integral curves.) Lastly, we discuss the Riemannian and sub-Riemannian problems. The (left-invariant) Riemannian and sub-Riemannian structures on SE(1,1) are classified, up to isometric group automorphisms and scaling. Explicit expressions for the geodesics of the normalised structures are found.

This research seeks to investigate the experiences of refugees in Port Elizabeth, South Africa, as reported by practitioners assisting refugees in the area. The explorative study aims at examining the issues refugees commonly find challenging, describing positive and negative developments in the experience of refugees and discussing how their experience can be improved. An analysis of basic human needs theory serves as a framework for the study. The researcher conducted face-to-face interviews with the participants. The analysis of the data gathered emphasised the challenges refugees face in seeking to satisfy their needs. The main themes that were identified from the data include: a lack of knowledge and awareness about refugee issues and a misconceptions regarding different groups of migrants; current socioeconomic challenges in South Africa; xenophobia; refugees as a heterogeneous group; and refugee access to services. The challenges faced by refugees in Port Elizabeth were identified as access to justice and correct documentation, access to healthcare, housing and education, as well as xenophobia, police protection and crime. Recommendations were made regarding the improvement of the refugees’ experience, including educating public sector employees and government officials about refugee rights, sensitising communities about refugee issues, offering refugees language courses and providing them with an induction into their rights and employing translators in health clinics to improve access to healthcare.

I use the Vygotskian (1978) socio-cultural perspective where the process of learning takes place within the Zone of Proximal Development (ZPD). Polya's problem solving heuristics (Polya, 1973) and Kilpatrick's "Instructional Triangle" (Kilpatrick, Swafford & Findell, 2001) provided the analytical framework for the study. Seven grade 7 learners from a Ex-Model C school, volunteered to participate in the study. The data gathering process involved an initial problem solving assessment, a written questionnaire, observations and video recordings of the seven learners during a series of after school problem solving sessions and post intervention learner interviews. The study showed that group discussion can have a positive impact on learners' problem solving in several respects: My key findings point to: Mathematical communication does play a role in development of problem solving strategies. A more knowledgeable other, with regards to Vygotsky's (1978) ZPD and Kilpatrick et al's (2001) instructional triangle is a critical factor in the development of problem solving strategies. All five strands of Kilpatrick et al., (2001), strands for mathematical proficiency are required for correct solutions to be calculated. At times Polya's (1973) steps for problem solving move at a rapid pace and are difficult to notice. These steps develop at different speeds for different people.

The purpose of this study was to investigate the strategies to improve reading in an Eastern Cape community. To realise this aim, I first looked for strategies that are currently used by teachers. Then I focussed on additional strategies that could be of use in improving reading levels. This study observed the practices and accessed perceptions of teachers in two primary schools regarding literacy accomplishments in order to come up with relevant strategies to improve reading levels. My study is a qualitative case study focussed on Foundation and Intermediate Phase literacy. The two schools used in this study have different language policies. The one uses English as the language of learning and teaching and the other school uses isiXhosa. The home language of most of the learners in both schools is isiXhosa. I observed and interviewed teachers to get deeper understanding of the problem in the two schools. The Four Resources Model of Freebody and Luke (1990) proved to be a strategy that could be used to support the development of reading from the early years and on into the high school years. This model was found to articulate well with the official school literacy curriculum. In-school reading strategies could be well supplemented by assisting learners to engage with the four roles of Code Breaker, Text Participant, Text User and Text Analyst. In my investigation I found out that there is a problem in our schools in both home and additional language contexts. There is a great need for both in-school and out-of-school strategies to improve the situation. An Asset Mapping strategy revealed the range of existing and potential strategies available to a particular Eastern Cape community to raise reading levels and thereby improve learning.

- Description: Many tertiary institutions are confronted with the challenge of unprepared students. This epidemic is largely due to factors in and out of individuals‟ control. These factors include academic proficiency, course-student match, support and integration, financial stability, preparation, personal circumstances and engagement. The main objective of this study is therefore threefold: Firstly, to describe what student readiness entails; secondly to understand why there is a lack of readiness among first-year Human Resource Management students, and thirdly, to identify and improve upon interventions that prepare students for tertiary education. For this study student readiness refers to the ability of students to meet the basic requirements to successfully enrol for a course without remedial assistance and to successfully complete their studies in the recommended timeframe. The target population of this study focussed on students who registered on a full-time basis for the National Diploma in Human Resource Management for the academic year 2014 (n=117). A self-administered questionnaire was distributed to the students and an open-ended questionnaire additionally to the lecturers who teach first-year Human Resource Management students. The study was subjected to various statistical techniques such as Cronbach‟s alpha, t-tests, Pearson moment product correlation and Cohen‟s d. In doing so an improved insight into student readiness and success was developed. The main findings of the study indicated that there is a relationship between the type of schooling and readiness factors as statistical significance was noted. In addition, students who scored high on readiness factors were more likely to succeed in tertiary education. Moreover the number of interventions students is exposed to positively influence student readiness and success. This study has contributed to the body of knowledge which relates to the broader educational environment in South Africa and provides recommendations to address student readiness and success.

Investigating the English vocabulary needs, exposure, and knowledge of isiXhosa speaking learners for transition from learning to read in the Foundation Phase to reading to learn in the Intermediate Phase : a case study

- Description: The nature of learners' transition from Grade 3 to Grade 4 has serious and far reaching ramifications for their educational success in the subsequent grades. This is true of the South African education system, as it is for several other education contexts. The challenge that complicates the transition is largely a lexical one. It is with that understanding that the present study aimed to do three things namely: to determine the vocabulary needs of Grade 3 learners for transitioning to Grade 4; to establish the extent to which Grade 3 isiXhosa speaking learners are exposed to, and prepared for, the acquisition of that requisite vocabulary; and to establish the extent of Grade 4 learners' knowledge of that vocabulary at the beginning of the year. The vocabulary needs of Grade 3 learners for the transition to Grade 4 were determined from the Grade 4 subject textbook corpus. Word frequency was the criterion used to determine the usefulness and critical importance of a word for the transition. The AntConc concordance software program was used to generate word frequencies. Words with high frequency across the different subject areas, which were confirmed as high frequent in four other known word lists, were considered as constituting the vocabulary needs of learners at the verge of this significant transition. The extent of learners' preparedness for transitioning to Grade 4 in terms of their vocabulary exposure and vocabulary instruction was determined through teacher interviews, classroom observations, analysis of teacher classroom talk, analysis of the exposure and recycling of high frequent vocabulary in Grade 3 reading materials and classroom print. From these diverse sources, findings point to a paucity in both the exposure and recycling of the requisite vocabulary in these sources of classroom language input. Classroom observations and teacher interviews attest to lack of deliberate vocabulary instruction in the Grade 3 English First Additional Language lessons. The only extensive coverage of the requisite vocabulary was in the Grade 3 reading materials which included Big books, Readers and Workbooks. Investigating the provision of nutritional support to critically ill hospitalised patients by registered nurses in East London public and private hospitals in the Eastern Cape

- Description: Critical illness is typically associated with a catabolic stress state in which patients commonly demonstrate a systemic inflammatory response that brings about changes in their body systems. Changes in the body systems make the critically ill dependent on mechanical ventilation and inotropic support for longer periods in order to survive. However, this inflammatory response can be attenuated by the timely introduction of nutritional support to provide energy and nutrients to diminish catabolism and promote anabolism. The result could be a decrease in the morbidity and mortality rates, as well as the financial burden on the patients, institutions and the state. Since registered nurses initiate and utilise feeding protocols to achieve target goals, there is a strong need for nurse-initiated feeding protocols. These protocols should be coupled with a comprehensive nurse-directed nutritional educational intervention that will focus on their safe and effective implementation. This focus on nursing nutrition education represents a major shift away from traditional education which has focused on dietitians and physicians. Evidence suggests that incorporating guideline recommendations into nurse-initiated protocols for starting and advancing enteral feedings is an effective strategy to improve the delivery of nutritional support. The study was aimed at exploring the provision of nutritional support to critically ill hospitalised patients by registered nurses to identify and describe possible gaps in the practice, through determining the potential relationship between the provision of nutritional support and characteristics of its providers. A quantitative, descriptive correlational study was undertaken. Seventy registered nurses working in neonatal/paediatric and adult critical care units in two public and three private hospitals in East London in the Eastern Cape participated in the study. The sample also included public critical care students. The results showed that registered nurses in private hospitals have more knowledge about the importance of nutritional support than their public hospital counterparts and students. The mean score was on the question was 80.3% with the highest score of 91% which was for the private hospital nurses, followed by 77.2% for public and 71.4% for students. Again, the mean score for knowledge on timing of initiating nutritional support was 48%, the highest score being 69.4% for students followed by private hospital nurses with 49.6%. Close to 63% (n = 44) of these nurses were either unsure about the availability of nutritional protocols or clearly attested to their non-availability. This is seen as an issue of concern because a protocol is meant to be a standard document with which all members of the ICU should be familiar. It is meant to guide and facilitate the manner of working in the unit. While facilitation of maintenance of nutritional support to patients is the responsibility of registered nurses, according to Regulation 2598(1984) section 45 (1) (q) of the South African Nursing Council, 68% (n = 48) of the respondents felt that this was in the practising scope of doctors and dietitians. The study concluded that the nurses are knowledgeable about the importance of nutritional support but knowledge gaps have been identified as far as the timing of initiating nutritional support is concerned. Some attested to unavailability of standard guidelines that are tailored into protocols guiding the provision of nutritional support by registered nurses in the critical care units. Nutrition should be prioritised as an important therapy for improving the outcomes of critically ill patients. Nurses need to analyse its provision, identify barriers to nutritional strategies and engage in nutritional education to empower themselves regarding the practice. Most importantly, there is a need for nurse-initiated nutritional protocols that are tailored from the broad nutritional guidelines and aligned with the local context and ways of practising. Nutritional support should be included as a key component of the curriculum in academic programmes that specialise in critical care nursing.

- Description: South Africa is the world's second largest exporter of fresh citrus and is ranked 14th in citrus production. Fungal pathogens such as Phytophthora and Pythium cause economic losses as a result of root rot and brown rot. Mycorrhizal fungi are specialized members of the fungal community forming a mutualistic relationship with plant roots. Mycorrhizal fungal structures are known to associate with other soil microorganisms and these may contribute to improved plant growth. A diverse group of bacteria that interact with the mycorrhizal fungi are known as Mycorrhizal Helper Bacteria (MHB). The aim of this study was to investigate the role of arbuscular mycorrhiza and associated bacteria isolated from spores and determine whether they had any plant growth promoting potential. A total of 19 bacteria were isolated from arbuscular mycorrhizal spores and were molecularly identified as belonging to several Bacillus, Micrococcus, Onchrobactrum and Staphylococcus sp. All bacterial isolates were tested for plant growth promotion abilities. One Bacillus isolate was able to solubilise phosphate. Four isolates Micrococcus sp, Micrococcus leteus, Ochrobacterum sp and Ochrobacterum antropi were able to produce Indole Acetic Acid and three isolates showed potential to reduce growth of Phytophthora nicotianae, P. citrocola and P. citrophthora in in vitro plate cultures. Further tests using culture supernatants of the Bacillus sp, Micrococcus sp and Bacillus cereus confirmed their ability to inhibit or reduce growth of the three Phytophthora species in a 96 well bioassay. Bacillus sp and Bacillus cereus were able to inhibit Phytophthora spp by 95 to 100 % and Micrococcus spp was able to decrease pathogen growth by 60 to 94 %. These bacterial isolates were further evaluated for plant growth promoting abilities on citrus rough lemon seedlings alone or in combination with arbuscular mycorrhizal inoculum. Bacterial and mycorrhizal inoculants influence the increase in shoot and root biomass. Bacillus cereus in combination with mycorrhizal inoculum significantly increased seedling shoot to root ratio while root biomass was significantly increased with mycorrhizal inoculation. Due to the short duration of the trial mycorrhizal colonisation could not be assessed. It is evident that selected combinations of bacteria and mycorrhizal fungi could promote citrus seedling growth and potentially improve seedling health. Further studies under nursery conditions are recommended.

The findings of my study suggested the HIV/AIDS awareness and prevention interventions that have been implemented since the outbreak of the epidemic have not their stated objectives aimed at reducing infections and children’s feeling of stigma towards their HIV/AIDS infected peers in schools. More worrying is the fact that the results suggested that primary school children lacked the appropriate knowledge about HIV/AIDS. The outcome of this lack of knowledge is that many primary school children have adopted risky behaviours towards the disease. The study recommended that all stakeholders must shift prevention interventions from media-based programmes focussed on adolescents to school-based programmes focused on primary school learners. The literature reviewed and my findings suggested school-based prevention awareness campaigns and inclusion of HIV/AIDS knowledge into school curriculum for all learners might produce better results.

- Description: The Internet of Things (IoT) environment involves the interaction of numerous ‘things’. These ‘things’ are embedded with different kinds of technologies such as RFID technology, NFC technology and sensors. This causes IoT to bring into play many security risks apart from the ones that already exist in the current Internet, for example, embedded RFID tags can be easily triggered and send their content which could be private information. IoT also introduces internal attacks such as on-off attack, bad mouthing and white washing attacks into networks that already exist. These internal attacks cannot be solved by hard security mechanisms such as cryptographic algorithms and firewalls because they guarantee total trust. This eliminates uncertainty which should always be available where trust exist. That is, hard security mechanisms enable IoT ‘things’ to either trust another ‘thing’ completely or not and this makes them unsuitable for the IoT environment. When objects in any network are communicating, there is some element of uncertainty. Also, hard security mechanisms such as public key cryptography cause communication overheard in the already resource-constrained IoT devices and these conventional cryptography methods cannot deal with internal attacks. This brings about the need for a middleware that includes functions that will manage trust, privacy and security issues of all data exchange, communications and network connections. For IoT to be successful, the provision of trust, security and privacy measures are essential. Trust management may enhance the adoption and security measures in IoT. Trust helps in identifying trustworthy ‘things’ in the network and give ‘things’ in the network the ability to reason in all aspects concerning trust in the environment. Trust can be administered through a trust management model. This research notes that most of the trust models that have been proposed fail to address scalability challenges and lack suitable computation methods. It is on that premise that this research focuses on developing a suitable trust model for the IoT environment. The research also introduces new ways of creating relationships in IoT. This enables the creation of new cooperation opportunities in the environment. In overall, this research aimed to design and develop a generic trust and authority delegation model for the heterogonous IoT environment that is scalable and generalized to cater for the heterogeneous IoT environment. This research was conducted in three phases. The first phase reviewed literature in order to identify outstanding issues in IoT trust management and also identify the suitable computational method. This provided a critical analysis of different computational methods highlighting their advantages and limitations. In the second phase of the research, the proposed trust model was designed and tested. In the last phase, the feasibility of the proposed model was evaluated. The proposed model is based on fuzzy logic. Fuzzy logic was selected for trust computation because it is able to imitate the process of the human mind through the use of linguistic variables and it can handle uncertainty. The proposed model was tested in a simulated environment. The simulation results showed that the proposed model can identify selfish and malicious entities effectively. The results also showed that the model was able to deal with different types of behaviours of entities. The testing proved that the proposed trust model can support decision making in IoT based on trust. The results from the evaluation show that this research ameliorates the design and development of trust management solutions for the IoT environment.

- Description: The Internet of Things (IoT) environment involves the interaction of numerous ‘things’. These ‘things’ are embedded with different kinds of technologies such as RFID technology, NFC technology and sensors. This causes IoT to bring into play many security risks apart from the ones that already exist in the current Internet, for example, embedded RFID tags can be easily triggered and send their content which could be private information. IoT also introduces internal attacks such as on-off attack, bad mouthing and white washing attacks into networks that already exist. These internal attacks cannot be solved by hard security mechanisms such as cryptographic algorithms and firewalls because they guarantee total trust. This eliminates uncertainty which should always be available where trust exist. That is, hard security mechanisms enable IoT ‘things’ to either trust another ‘thing’ completely or not and this makes them unsuitable for the IoT environment. When objects in any network are communicating, there is some element of uncertainty. Also, hard security mechanisms such as public key cryptography cause communication overheard in the already resource-constrained IoT devices and these conventional cryptography methods cannot deal with internal attacks. This brings about the need for a middleware that includes functions that will manage trust, privacy and security issues of all data exchange, communications and network connections. For IoT to be successful, the provision of trust, security and privacy measures are essential. Trust management may enhance the adoption and security measures in IoT. Trust helps in identifying trustworthy ‘things’ in the network and give ‘things’ in the network the ability to reason in all aspects concerning trust in the environment. Trust can be administered through a trust management model. This research notes that most of the trust models that have been proposed fail to address scalability challenges and lack suitable computation methods. It is on that premise that this research focuses on developing a suitable trust model for the IoT environment. The research also introduces new ways of creating relationships in IoT. This enables the creation of new cooperation opportunities in the environment. In overall, this research aimed to design and develop a generic trust and authority delegation model for the heterogonous IoT environment that is scalable and generalized to cater for the heterogeneous IoT environment. This research was conducted in three phases. The first phase reviewed literature in order to identify outstanding issues in IoT trust management and also identify the suitable computational method. This provided a critical analysis of different computational methods highlighting their advantages and limitations. In the second phase of the research, the proposed trust model was designed and tested. In the last phase, the feasibility of the proposed model was evaluated. The proposed model is based on fuzzy logic. Fuzzy logic was selected for trust computation because it is able to imitate the process of the human mind through the use of linguistic variables and it can handle uncertainty. The proposed model was tested in a simulated environment. The simulation results showed that the proposed model can identify selfish and malicious entities effectively. The results also showed that the model was able to deal with different types of behaviours of entities. The testing proved that the proposed trust model can support decision making in IoT based on trust. The results from the evaluation show that this research ameliorates the design and development of trust management solutions for the IoT environment.

The combination of these two technologies has resulted in the creation of electronic commerce which also enables mobile payments. Mobile payments (mpayments) are enabled by many technologies with Near Field Communication (NFC) being the most recent one. NFC is a wireless technology that enables mobile devices in close proximity to exchange data. The mobile device has already been enthusiastically accepted by the customers and they carry it with them wherever they go and this makes it a good device for providing a payment method alternative. This research looks at contactless mobile payment as a payment method. Customers in marginalized rural areas lack a payment alternative to cash hence in this research we are investigating and proposing the use of a NFC enabled mobile payment application for Marginalized Rural Areas. This research extensively evaluates and assesses the potential of using NFC enabled m-payments in Marginalized Rural Areas in South Africa by carrying out an investigation of the technology and its acceptance by customers. The investigation of the technology included implementation of a prototype application which was used to introduce the technology to the consumers. The customer acceptance of the NFC enabled mobile payments was evaluated using the Technology Acceptance model (TAM). The model was modified to suit the context of this study by adding more constructs. - Description: The purpose of the study has been to assess the question of sustainability of economic growth and human development, particularly using sub-Saharan Africa in context. Sub-Saharan Africa is an interesting case study because, on the one hand, it has been mired in poverty and remains the least developed region in the world, and on the other, it has experienced a revival in economic growth since the mid-1990s. Economists tend to use the term economic development and economic growth interchangeably. However, questions have been raised about whether Africa’s latest growth episode is indeed ‘development’. Although there are many issues at stake, the key question, and the focus of this thesis, is whether sub-Saharan Africa’s revival is sustainable. The paper sets out the debate between the ‘World Bank view’ and the ‘alternative view’. The main debate lies around how genuine development should be achieved. Firstly, the ‘World Bank view’ claims that economic growth is necessary and sufficient condition to achieve development. Economic growth will be generated by ‘orthodox’ policies and this growth will automatically trickle-down and stimulate development. Secondly, the ‘alternative view’ argues that economic growth is necessary but it is not sufficient to stimulate sustainable development. Economic growth without ‘qualitative’ change is not ‘sustainable’. Indeed, human development shortfalls (as well as other, social, political and structural problems), if not addressed through appropriate policy interventions, can undermine economic growth. The ‘alternative view’ appears to be strongly supported by evidence from other developing regions such as Latin America and East Asia. The empirical study conducted in this thesis reinforces doubts about ‘sustainability’. Even though there are signs of convergence in some indicators; this is not the case for all indicators. More importantly the gap between sub-Saharan Africa and other developing regions remains very wide. Sub-Saharan Africa’s development path remains uncertain. The intention in this study is not to be conclusive that sub-Saharan Africa cannot achieve sustainable development. Rather the study attempts to identify potential hindrances to sub-Saharan Africa’s development and to provide a solid foundation for further research in the same direction.

Isolation, Characterisation, and biological activity evaluation of essential Oils of Cymbopogon validus (Stapf) Stapf ex Burtt Davy and Hyparrhenia hirta (L.) Stapf

- Description: Cymbopogon validus and Hyparrhenia hirta belong to the Poaceae botanical family. Both plants are used as thatching material; H. hirta is also used for weaving mats and baskets. In this study, we investigated the anti-inflammatory effects of C. validus and H. hirta essential oils on fresh egg-albumin induced edema on Wistar rats. To fully understand the chemically induced anti-inflammatory properties of these plants, we first analyzed the chemical composition of the essential oils. The essential oils were analyzed using gas chromatography-mass spectrometry (GC-MS). In C. validus, 13 compounds accounted for 74.3% of fresh leaves oil, 14 compounds 71.8% of dried leaves oil and 12 compounds 73.3% of flower oil were identified from the GC-MS Chromatogram. The percentage yields were as follows fresh leaves oil 2.2%, dried leaves oil 2.0% and flower oil 2.4% v/w respectively. Linalool (3.2-29.6%) and northujane (4.4-16.9%) were the dominant compounds found in the 3 oils analyzed. While, α-terpineol 37.5% and verbenone 13.5% was only found in the fresh leaf oil, this was absent in the dried leaves oil and the flowers oil of C. validus. In H. hirta, 25 compounds accounted for 68.1% of fresh leaves oil, 40 compounds 71.9% of dried leaves oil, 23 compounds 77.6% of fresh flowers oil and 18 compounds 80.1% of dried flowers oil were identified from the GC-MS Chromatogram. The percentage yields obtained from the different parts were 3.4% for fresh leaves oil, 2.8% for dried leaves oil, 2.8% for fresh flowers oil and 0.7% for dried flowers oil v/w respectively. Northujane (8.5-30.0%), diisooctyl phthalate (4.4-26.5%), phytone (1.1-10.4%) were the dominant compounds found in the 4 oils analyzed. While, dibutyl phthalate 26.9% was only found in the fresh flowers and was absent in the dried flowers oil. Moreover, caryophyllene oxide (1.7-9.6%) was found in fresh leaves and dried flowers oil of H. hirta. In vivo analysis revealed that the two essential oils displayed significant edema inhibition effect overtime. They displayed strong anti-inflammatory properties when compared to control group. However, the H. hirta essential oil was more effective than that of C. validus. Linalool, α-terpineol, and northujane extracted from C. validus and H. hirta essential oils might have contributed to the anti-inflammatory effects observed in Wistar rats. This study, confirms the anti-inflammatory properties of C. validus and H. hirta suggesting that they may be used in diseases related to anti-inflammation.

Isolation, purification and partial characterisation of cancer procoagulant from placental amnion-chorion membranes and its role in angiogenesis inflammation and metastasis

- Description: Cancer procoagulant (EC 3.4.22.26) is an enzyme that is derived from tumour and foetal tissue, but not normal tissue. It is a direct activator of factor X and has been isolated from amnion-chorion membranes as well as from extracts and cells from human melanoma. The presence of cancer procoagulant has been associated with the malignant phenotype, as well as having a particularly high activity in metastatic cells. Cancer procoagulant activity is elevated in the serum of early stage breast cancer patients and decreased to normal in the advanced stages of the disease. In this study, cancer procoagulant was successfully isolated from amnion-chorion membranes and purified to homogeneity. The molecular weight of cancer procoagulant was determined using SDS-PAGE and was found to be 68 kDa. Cancer procoagulant was delipidated and it was shown that its activity was increased by the presence of lipids in a dose-dependent manner. Recovery of cancer procoagulant after delipidation is poor, consequently, a larger mass of sample is required to obtain sufficient amounts of delipidated material for N-terminal amino acid analysis. The optimum pH of cancer procoagulant was determined to be pH 8 and its optimal temperature was found to be 50°C. Novel synthetic substrates were designed to assay for cancer procoagulant activity. Currently, 2 potential candidates have been identified, namely, PQVR-AMC and AVSQSKP-AMC. Cancer procoagulant-induced expression of cytokines is differently modulated in the less aggressive MCF-7 cell line as compared to the metastatic and more aggressive MDA-MB-231 cell line. There are marked similarities in the inflammatory response produced by cancer procoagulant in hTERT-HDLEC and MDA-MB-231 cells, which are both associated with migratory capacity. Furthermore, cancer procoagulant-induced PDGF-β expression in hTERT-HDLEC and MDA-MB-231 cells could point to involvement of cancer procoagulant in wound healing and metastatic spread, respectively. Cancer procoagulant induced the motility of MDA-MB-231, MCF-7 and hTERT- cells in vitro in a time- and dose-dependent manner. Together, these results suggest that cancer procoagulant plays a role in the migration of breast cancer cells as well as the migration of endothelial cells.

Job engagement and locus of control were the independent variables and organisational citizenship behaviour was the dependent variable. Data was drawn from a sample of 300 participants. A questionnaire was used to collect the data. It consisted of four sections including biographical and occupation data questionnaire. To measure job engagement, the 18-item Rich et al., (2010) job engagement scale, with a 5-point Likert scale was used. To measure locus of control, the 16-item Spector (1988) work locus of control scale was used, with 6-point Likert scale. To measure organisational citizenship behaviour, the Fox & Spector (2011) 20 item organisational citizenship behaviour questionnaire, with a 5-point Likert scale was used. Data was analysed using various statistical techniques including the Pearson Product Moment Correlation Technique and the generalised linear regression model. The results indicated that job engagement has a significant positive correlation with organisational citizenship behaviour and locus of control also has a significant positive correlation with organisational citizenship behaviour. However the results also indicated that when job engagement and locus of control are put together, they do not account for a significantly higher proportion of variance in organisational citizenship behaviour than each of them separately. Furthermore, as far as OCB-P and OCB-O are concerned, the results indicated a partial support of the research hypothesis that job engagement and locus of control together account for a significantly higher proportion of variance in organisational citizenship behaviour than any of the two separately. The study recommends that managers in organisations must focus on improving job engagement and promoting internal locus of control in order to ensure high levels of organisational citizenship behaviour.

The study recommends that managers in organisations must focus on improving job engagement and promoting internal locus of control in order to ensure high levels of organisational citizenship behaviour.

- Description: This research study investigated the effect of job engagement to organisational stress and the employees’ intention to quit among administrative personnel at the University of Fort Hare. The main objectives of this study were determining the effects of organisational stress on employees’ intention to quit, examining the role of job engagement as a moderator of the relationship between organisational stress and employees’ intention to quit, and providing results that would lead to a deeper understanding of the nature and extent of the relationship between organisational stress and intention to quit. A survey method was adopted for this study. A questionnaire comprising of four sections: biographical information, ERI questionnaire for stress, turnover intention, and UWES scale, was administered to non-academic support personnel of the University of Fort Hare in all three campuses namely Alice, Bhisho, and East London. Simple random sampling was used to select the participants. The literature reviewed displayed a great need for managers to understand organisational stress and its causes, and how to manage and cope with stress so as to avoid losing employees, especially highly competent employees. The study yielded results that show that there are interrelationships amongst the three variables, and that job engagement does moderate the relationship between organisational stress and employees’ intention to quit.

Journalism sourcing and credibility: a study of Raia Mwema’s use of anonymous sources

- Description: This study analyses how readers impute credibility to the Tanzanian investigative newspaper Raia Mwema which, as a matter of routine journalistic practice, uses a high number of anonymous sources. Against the backdrop of a strong theoretical position, espoused by media studies theorists in both Western media contexts and in Africa, in which this practice is deemed to diminish the credibility of both journalists and their stories, the study’s main purpose is to examine how readers make sense of this very prevalent practice in a country that has recently opened up to media plurality. It also investigates where they locate the source of credibility for this high-selling newspaper, if not in the traditional way through its named sources of information. The study surveys the frequency of appearance of anonymous sources in this newspaper for the period of one year from January 2011 to December 2011. Then the study considered how this practice is viewed and understood by the wider journalistic community in Tanzania as well as looking into how the journalists and owners of Raia Mwema make choices about attributing their journalism. Lastly, the study engaged with particular readers to understand what sense they make of this practice in the wider landscape of Tanzanian media and the post-repressive political situation. The study makes use of theories of the sociology of news production so as to understand the context in which Raia Mwema has routinised the practice of anonymous attribution and whether the journalistic community and newspaper readers find the practice credible. The study also employs reception analysis in order to understand to what extent Raia Mwema readers negotiate and make sense of the mainly political, and often critical, media messages they get from newspaper. In this way, it introduces the importance of the reader in the production of meaning and of assessment of credibility of journalism. The interviews – ranging from journalists working at the paper, through the wider journalistic community and taking in the readership of the paper – show that theoretical considerations of journalistic credibility must take into account the political, social and media context in which journalism is produced. Pronouncements on the overuse of anonymous sources, without understanding the way readers and journalists negotiate the complexities of an actual situation, do not tell us much about credibility and how readers understand the messages they are given. From this study, it is clear that in African countries embarking on opening media systems, credibility involves more factors than have been discussed in the literature and that readers and journalists are sophisticated consumers and producers of media messages in countries that place a host of obstacles in the way of investigative journalism and open political communication.
