- Description: The pharmaceutical industry is global, is highly regulated and is able to achieve reasonable product quality but at high cost with maximum effort. Numerous challenges face the pharmaceutical industry and include a shrinking research pipeline, less innovation, outsourcing, investments, increasing research and development costs, long approval times, growth of the generic industry, failure to understand or analyze manufacturing failure and wastage as high at fifty percent for some pharmaceutical products. An efficient and flexible pharmaceutical sector should be able to consistently produce high quality pharmaceutical products at a reduced cost with minimal waste. As a result, Food and Drug Administration (FDA) and other agencies such as the International Conference on Harmonization (ICH) have embraced a “Quality by Design” (QbD) paradigm and this has become the “desired state” so as to shift manufacturing from being empirical to a science, engineering, and risk based approach. QbD is a systematic approach for the development of high quality pharmaceutical dosage forms that begins with predefined objectives based on the premise that quality must be built into and not tested into a product. QbD together with the establishment of a design space for dosage forms is a fairly new concept and there is limited published data on QbD concepts that report the entire process of identifying Critical Quality Attributes (CQA), design of a formulation and manufacturing process to meet product CQA, understanding the impact of material attributes and process parameters on product CQA, identification and controlling sources of variability in materials and processes that affect the CQA of a product and finally establishing, evaluating and testing a design space using both in vitro and in vivo approaches to assure that a product of consistent quality can always be produced. The objective of these studies was to implement a QbD approach to establish a design space for the development and manufacture of a safe, effective, stable generic formulation containing 0.05% w/w clobetasol 17-propionate (CP) that had similar in vitro and in vivo characteristics to an innovator product, Dermovate® (Sekpharma® Pty Ltd, Sandton, Gauteng, RSA). Such a product would pose a minimal risk of failure when treating severe skin disorders such as seborrhoeic dermatitis, extreme photodermatitis and/or severe psoriasis in HIV/AIDS patients in Southern Africa.

Despite these important contributions to the economies of countries, a shortage of entrepreneurial activity exists across borders and specifically in developing countries such as South Africa. The purpose of this study was to determine the perception and image of an entrepreneur in the eyes of various stakeholders. The reasoning behind this was that if the image of an entrepreneur could be determined, one could establish whether the image positively or negatively influences entrepreneurial intentions as well as potential future entrepreneurial activity. More specifically, the primary objective was to identify the perception and image that potential entrepreneurs (students) and existing entrepreneurs (small business owners) have of an entrepreneur. In the body of knowledge or general literature on entrepreneurship, the most commonly discussed topics are the nature and importance of entrepreneurship, the attributes (personality traits, characteristics and skills) associated with an entrepreneur, various push and pull factors, various rewards and drawbacks of such a career and the challenges entrepreneurs face. It is these aspects of entrepreneurship that stakeholders will most likely have been exposed to, and that most possibly have influenced their perception and image of an entrepreneur. The aforementioned aspects provided an overview of the theoretical body of knowledge on which the perception and image of an entrepreneur is based. The present study adopted a qualitative research paradigm with a phenomenological approach to achieve the research objectives of the study. Within this context, the study made use of a qualitative method for data collection and a quantitative method for data analysis. As such, a mixed methods approach was adopted. More specifically, a qualitative dominant mixed research method was implemented. A continuous word association test, which is a projective technique, was adopted as the qualitative means of data collection. This test involved asking participants to recall the words that come to mind when presented with the word “entrepreneur”. This method was selected because of its ability to reveal both affective and cognitive associations with the concept “entrepreneur”. A quantitative summative (manifest) content analysis was used as the quantitative research method for analysing the data. The continuous word association test was conducted among three sample groups, namely students prior to commencing, and students after completing a module in entrepreneurship, and small business owners. Student and small business owner participants were asked to write down as many words or phrases as possible that came to mind when they thought of the word “entrepreneur”, which was the stimulus word, within a ten-minute period. These responses were then collated and coded by developing a coding framework based on brand image and entrepreneurship literature. In studies on brand image, the components of image are considered to be tristructured in nature, consisting of cognitive (what the individual knows), affective (how the individual feels) and holistic (overall symbolism, combination of affective and cognitive) evaluations. The words generated by the participants in this study were broadly coded according to these categories and further subcategorised by searching for themes within the broad categories, which was facilitated and guided by an in-depth investigation of the entrepreneurship literature. The findings of this study show that the words generated by all three groups of participants were mostly of a cognitive nature, followed by words of a general or affective nature. As such, the vast majority of words generated by all three groups related to what the participants knew about an entrepreneur (cognitive) versus how they felt about one (affective), and were grounded in the management or entrepreneurship literature. When comparing the top ten words most frequently associated with the term “entrepreneur” by the three groups of participants, the attribute risk-taker was the most frequently recalled word among all three groups. Students prior to undertaking the entrepreneurship module associated an entrepreneur with being creative and a risk-taker, having a business enterprise and being involved in the selling of goods and services. Students after completing the module in entrepreneurship associated an entrepreneur with being profit-orientated, a risk-taker, innovative and original, and being opportunistic. Small business owners, on the other hand, associated an entrepreneur with being a risk-taker, innovative and original, goal- and achievement-orientated and profit-orientated. The findings show that all groups of participants associated an entrepreneur principally with certain attributes rather than with learned skills and competencies, and that all groups had a more positive than negative image of an entrepreneur. It was also found that exposure to entrepreneurship literature has an influence on the perception and image that students have of an entrepreneur. Because the words recalled by students after completing the entrepreneurship module were more in line with those recalled by small business owners, than with those recalled by students before starting the module, it can be suggested that entrepreneurship literature contributes to a more realistic image of an entrepreneur among students. This study has contributed to the field of entrepreneurship research by adopting a qualitative dominant research paradigm in conjunction with quantitative research methods to explore the complexity of the term “entrepreneur”. Furthermore, this study has been able to establish how individuals feel about entrepreneurship, in terms of being either positive or negative, by adding an affective aspect to the cognitive aspect of entrepreneurial decision-making. By conducting a continuous word association test among students prior to beginning and after completing a module in entrepreneurship, the entrepreneurial knowledge of students before being exposed to entrepreneurship literature was established, and subsequently the effectiveness of the entrepreneurship module determined. It is hoped that the findings of this study have added value to the entrepreneurship body of knowledge and can be used in future studies as a tool to address the problem of low entrepreneurial intention and activity among South Africans. Furthermore, it is hoped that by creating a positive image of an entrepreneur, entrepreneurship as a desirable career choice can be promoted and an entrepreneurial culture developed within communities and broader society.

Ethanol production from lignocellulosic sugarcane leaves and tops

- Description: Various methods for the production of bioethanol using different feedstocks have been researched on. In most work on bioethanol synthesis from sugar cane, tops and leaves have been regarded as waste and generally removed and thrown away. In this work, lignocellulosic sugarcane leaves and tops were not discarded but instead used as biomass to evaluate their hydrolyzate content. The leaves and tops were hydrolysed using different methods, namely concentrated acid, dilute acid pre-treatment with subsequent enzyme hydrolysis and compared with a combination of oxidative alkali pretreatment and enzyme hydrolysis. Subsequent fermentation of the hydrolyzates into bioethanol was done using the yeast saccharomyces cerevisae. Acid hydrolysis has the problem of producing inhibitors, which have to be removed and this was done using overliming with calcium hydroxide and compared to sodium hydroxide neutralization. Oxidative alkali pre-treatment with enzyme hydrolysis gave the highest yields of fermentable sugars of 38% (g/g) using 7% (v/v) peroxide pre-treated biomass than 36% (g/g) for 5% (v/v) with the least inhibitors. Concentrated and dilute acid hydrolysis each gave yields of25% (g/g) and 22% (g/g) yields respectively although for acid a neutralization step was necessary and resulted in dilution. Alkaline neutralization of acid hydrolyzates using sodium hydroxide resulted in less dilution and loss of fermentable sugars as compared to overliming. Higher yields of bioethanol, 13.7 (g/l) were obtained from enzyme hydrolyzates than 6.9 (g/l) bioethanol from dilute acid hydrolyzates. There was more bioethanol yield 13.7 (g/l) after 72h of fermentation with the yeast than 7.0 (g/l) bioethanol after 24h. However, the longer fermentation period diminishes the value of the increase in yield by lowering the efficiency of the process.

Drawing from literature and documented deliberations by industry professionals, content-specific analysis protocols were applied and tested using a case study representing topic-specific responses to the Two Oceans Quay 5 product launch. In doing so, a logical observation of the communication in unpaid media placements and relevant discussions distributed in public media channels was completed. It is in this sense that this research enriches the study of public relations with a particular focus on output-driven evaluation. It provides insights into qualitative and quantitative publicity measurement and suggests how these methods can be useful to explicate the impact of media coverage as a public relations element. While it largely focused on discovering improved media content analysis solutions, this study revealed that content-driven analysis can only be useful when its protocols are aligned with the context of the data and if communication practitioners remain aware and transparent of its subjectivity. In this regard, this study helps to generate an understanding of the subjective dynamic of public relations and the importance of in-depth and adaptable publicity assessments to help distinguish public relations as a purposeful branding function next to advertising and marketing.

This research looked to establish an ideal ratio between the recruitment from internal and external employees. To achieve the objectives, a literature review was conducted to determine the various combinations of recruitment. The literature was divided into talent management, internal and external recruitment, as well as succession management. The research took the form of a case study, looking at recruitment practices within the AGSA. A questionnaire was developed to gather information from employees in the AGSA, around their assessment of recruitment and their opinion on internal and external recruitment. The questionnaire was distributed online and various levels of employees within the organisation participated in responding to the questionnaire. The most recent recruitment reports were presented and analysed. Information was gathered from literature on recruitment and the AGSA employee’s by means of an online questionnaire. Recommendations were made to the AGSA taking into consideration the current practices within the organisation and the research undertaken for this dissertation.

The purpose is to look at the factors that affect housing delivery in the area, focusing more especially at the management of the beneficiaries and the process thereof. Buffalo City Metropolitan Municipality is not immune to the housing problem hence the study was initiated. Respondents were selected from among others the housing beneficiary and the predominantly the staff of Buffalo City Metropolitan Municipality to respond to the questionnaires related to the beneficiary management. Permission was sought from the Municipality to conduct the research, after which the questionnaires were distributed to all sampled respondents accordingly in line with the objectives of the research. The researcher distributed the questionnaires and allowed a period of three weeks for the respondents to respond to them, after which the researcher collected all the questionnaires. The study revealed the great understanding of the housing delivery process by the housing recipients. Inclusive of the administrators, i.e. Buffalo City Metropolitan Municipality staff members, are fully aware of the expectations from the recipients and the process. The findings further indicates that there are serious challenges facing housing delivery in the province and the country at large. Hence the beneficiaries could not get their houses on time and the housing projects did not get completed within the scheduled period, this ends up delaying the delivery process.

It has been discovered that the inability to provide effective service delivery is a complex challenge facing many municipalities in South Africa, and although such a situation has been co-determined by many other factors including financial restraints, the root cause of service delivery incapability’s can be traced back to weather the citizens are actually participating in the management of public finances through participatory budgeting or not. More often than not, the failure to accommodate the citizens in local government affairs has been the spark that has been responsible for setting alight strikes and service delivery protest marches in most of South Africa’s Provinces. Evaluating the post-implementation effectiveness of selected household water treatment technologies in rural Kenya

- Description: Water, sanitation and hygiene-related diseases are responsible for 7% of all deaths and 8% of all disability adjusted live years (DALYs), as well as the loss of 320 million days of productivity in developing countries. Though laboratory and field trials have shown that household water treatment (HWT) technologies can quickly improve the microbiological quality of drinking water, questions remain about the effectiveness of these technologies under real-world conditions. Furthermore, the value that rural communities attach to HWT is unknown, and it is not clear why, in spite of the fact that rural African households need household water treatment (HWT) most, they are the least likely to use them. The primary objective of this multi-level study was to assess the post-implementation effectiveness of selected HWT technologies in the Nyanza and Western Provinces of Kenya. The study was carried out in the rainy season between March and May, 2011 using a mixed method approach. Evidence was collected in order to build a case of evidence of HWT effectiveness or ineffectiveness in a post-implementation context. A quasi-experimental design was used first to conduct a Knowledge, Attitudes and Practices (KAP) survey in 474 households in ten intervention and five control villages (Chapter 3). The survey assessed the context in which household water treatment was being used in the study villages to provide real-world information for assessing the effectiveness of the technologies. An interviewer-administered questionnaire elicited information about the water, sanitation and hygiene-related KAP of the study communities. A household water treatment (HWT) survey (Chapter 4) was carried out in the same study households and villages as the KAP study, using a semi-structured questionnaire to gather HWT adoption, compliance and sustained use-related information to provide insight into the perceived value the study households attach to HWT technologies, and their likelihood of adoption of and compliance with these technologies. The drinking water quality of 171 (one quarter of those surveyed during KAP) randomly selected households was determined and tracked from source to the point of use (Chapter 5). This provided insights into HWT effectiveness by highlighting the need for HWT (as indicated by source water quality) and the effect of the study households’ KAP on drinking water quality (as indicated by the stored water quality). Physico-chemical and microbiological water quality of the nineteen improved and unimproved sources used by the study households was determined, according to the World Health Organisation guidelines. The microbiological quality of 291 water samples in six intervention and five control villages was determined from source to the point-of-use (POU) using the WHO and Sphere Drinking Water Quality Guidelines. An observational study design was then used to assess the post-implementation effectiveness of the technologies used in 37 households in five intervention villages (Chapter 6). Three assessments were carried out to determine the changes in the microbiological quality of 107 drinking water samples before treatment (from collection container) and after treatment (from storage container) by the households. The criteria used to assess the performance of the technologies were microbial efficacy, robustness and performance in relation to sector standards. A Quantitative Microbial Risk Assessment (QMRA) was then carried out in the HWT effectiveness study households to assess the technologies’ ability to reduce the users’ exposure to and probability of infection with water-borne pathogens (Chapter 7). The KAP survey showed that the intervention and control communities did not differ significantly in 18 out of 20 socio-economic variables that could potentially be influenced by the structured manner of introducing HWT into the intervention villages. The majority of the intervention group (IG) and the control group (CG) were poor or very poor on the basis of household assets they owned. The predominant level of education for almost two-thirds of the IG and CG respondents was primary school (completed and non-completed). Though very few were unemployed in IG (8.07%) and CG (14.29%), the two groups of respondents were predominantly engaged in subsistence farming — a low income occupation. With regard to practices, both groups had inadequate access to water and sanitation with only one in two of the households in both IG and CG using improved water sources as their main drinking water source in the non-rainy season. One in ten households in both study groups possessed an improved sanitation facility, though the CG was significantly more likely to practice open defecation than the IG. The self-reported use of soap in both study groups was mainly for bathing and not for handwashing after faecal contact with adult or child faeces. Despite the study groups' knowledge about diarrhoea, both groups showed a disconnection between their knowledge about routes of contamination and barriers to contamination. The most frequent reason for not treating water was the perceived safety of rain water in both the IG and CG. , The HWT adoption survey revealed poor storage and water-handling practices in both IG and CG, and that very few respondents knew how to use the HWT technologies correctly: The IG and CG were similar in perceived value attached to household water treatment. All HWT technologies had a lower likelihood of adoption compared to the likelihood of compliance indicators in both IG and CG. The users’ perceptions about efficacy, time taken and ease of use of the HWT technologies lowered the perceived value attached to the technologies. The assessment of the drinking water quality used by the study communities indicated that the improved sources had a lower geometric mean E. coli and total coliform count than the unimproved sources. Both categories of sources were of poor microbiological quality and both exceeded the Sphere Project (2004) and the WHO (2008) guidelines for total coliforms and E. Coli respectively The study communities’ predominant drinking water sources, surface water and rainwater were faecally contaminated (geometric mean E. coli load of 388.1±30.45 and 38.9±22.35 cfu/100 ml respectively) and needed effective HWT. The improved sources were significantly more likely than the unimproved sources to have a higher proportion of samples that complied with the WHO drinking water guidelines at source, highlighting the importance of providing improved water sources. The lowest levels of faecal contamination were observed between the collection and storage points which coincided with the stage at which HWT is normally applied, suggesting an HWT effect on the water quality. All water sources had nitrate and turbidity levels that exceeded the WHO stipulated guidelines, while some of the improved and unimproved sources had higher than permissible levels of lead, manganese and aluminium. The water source category and the mouth type of the storage container were predictive of the stored water quality. The active treater households had a higher percentage of samples that complied with WHO water quality guidelines for E. coli than inactive treater households in both improved and unimproved source categories. In inactive treater households, 65% of storage container water samples from the improved sources complied with the WHO guidelines in comparison to 72% of the stored water samples in the active treater households. However the differences were not statistically significant. The HWT technologies did not attain sector standards of effective performance: in descending order, the mean log10 reduction in E. coli concentrations after treatment of water from unimproved sources was PUR (log₁₀ 2.0), ceramic filters (log₁₀ 1.57), Aquatab (log₁₀ 1.06) and Waterguard (log₁₀ 0.44). The mean log10 reduction in E. coli after treatment of water from improved sources was Aquatab (log₁₀ 2.3), Waterguard (log₁₀ 1.43), PUR (log₁₀ 0.94) and ceramic filters (log₁₀ 0.16). The HWT technologies reduced the user’s daily exposure to water-borne pathogens from both unimproved and improved drinking water sources. The mean difference in exposure after treatment of water from unimproved sources was ceramic filter (log₁₀ 2.1), Aquatab (log₁₀ 1.9), PUR (log₁₀ 1.5) and Waterguard (log₁₀ 0.9), in descending order. The mean probability of infection with water-borne pathogens (using E.coli as indicator) after consumption of treated water from both improved and unimproved sources was reduced in users of all the HWT technologies. The difference in reduction between technologies was not statistically significant. The study concluded that despite the apparent need for HWT, the study households’ inadequate knowledge, poor attitudes and unhygienic practices make it unlikely that they will use the technologies effectively to reduce microbial concentrations to the standards stipulated by accepted drinking water quality guidelines. The structured method of HWT promotion in the intervention villages had not resulted in more hygienic water and sanitation KAP in the IG compared to the CG, or significant differences in likelihood of adoption and compliance with the assessed HWT technologies. Despite attaching a high perceived value to HWT, insufficient knowledge about how to use the HWT technologies and user concerns about factors such as ease of use, accessibility and time to use will impact negatively on adoption and compliance with HWT, notwithstanding their efficacy during field trials. Even though external support had been withdrawn, the assessed HWT technologies were able improve the quality of household drinking water and reduce the exposure and risk of water-borne infections. However, the improvement in water quality and reduction in risk did not attain sector guidelines, highlighting the need to address the attitudes, practices and design criteria identified in this study which limit the adoption, compliance and effective use of these technologies. These findings have implications for HWT interventions, emphasising the need for practice-based behavioural support alongside technical support.

The improved sources were significantly more likely than the unimproved sources to have a higher proportion of samples that complied with the WHO drinking water guidelines at source, highlighting the importance of providing improved water sources. The lowest levels of faecal contamination were observed between the collection and storage points which coincided with the stage at which HWT is normally applied, suggesting an HWT effect on the water quality. All water sources had nitrate and turbidity levels that exceeded the WHO stipulated guidelines, while some of the improved and unimproved sources had higher than permissible levels of lead, manganese and aluminium. The water source category and the mouth type of the storage container were predictive of the stored water quality. The active treater households had a higher percentage of samples that complied with WHO water quality guidelines for E. coli than inactive treater households in both improved and unimproved source categories. In inactive treater households, 65% of storage container water samples from the improved sources complied with the WHO guidelines in comparison to 72% of the stored water samples in the active treater households. However the differences were not statistically significant. The HWT technologies did not attain sector standards of effective performance: in descending order, the mean log10 reduction in E. coli concentrations after treatment of water from unimproved sources was PUR (log₁₀ 2.0), ceramic filters (log₁₀ 1.57), Aquatab (log₁₀ 1.06) and Waterguard (log₁₀ 0.44). The mean log10 reduction in E. coli after treatment of water from improved sources was Aquatab (log₁₀ 2.3), Waterguard (log₁₀ 1.43), PUR (log₁₀ 0.94) and ceramic filters (log₁₀ 0.16). The HWT technologies reduced the user’s daily exposure to water-borne pathogens from both unimproved and improved drinking water sources. The mean difference in exposure after treatment of water from unimproved sources was ceramic filter (log₁₀ 2.1), Aquatab (log₁₀ 1.9), PUR (log₁₀ 1.5) and Waterguard (log₁₀ 0.9), in descending order. The mean probability of infection with water-borne pathogens (using E.coli as indicator) after consumption of treated water from both improved and unimproved sources was reduced in users of all the HWT technologies. The difference in reduction between technologies was not statistically significant. The study concluded that despite the apparent need for HWT, the study households’ inadequate knowledge, poor attitudes and unhygienic practices make it unlikely that they will use the technologies effectively to reduce microbial concentrations to the standards stipulated by accepted drinking water quality guidelines. The structured method of HWT promotion in the intervention villages had not resulted in more hygienic water and sanitation KAP in the IG compared to the CG, or significant differences in likelihood of adoption and compliance with the assessed HWT technologies. Despite attaching a high perceived value to HWT, insufficient knowledge about how to use the HWT technologies and user concerns about factors such as ease of use, accessibility and time to use will impact negatively on adoption and compliance with HWT, notwithstanding their efficacy during field trials. Even though external support had been withdrawn, the assessed HWT technologies were able improve the quality of household drinking water and reduce the exposure and risk of water-borne infections. However, the improvement in water quality and reduction in risk did not attain sector guidelines, highlighting the need to address the attitudes, practices and design criteria identified in this study which limit the adoption, compliance and effective use of these technologies. These findings have implications for HWT interventions, emphasising the need for practice-based behavioural support alongside technical support.

Presumptive bacteria counts ranged as follows: faecal coliforms 0 to 1.6 × 103 CFU/100 ml, E. coli 0 to 1.4 × 103 CFU/100 ml and E. coli O157:H7 0 to 9.6 × 102 CFU/100 ml. Forty eight percent (305/626) of the presumptive E. coli isolates were confirmed using species-specific uidA gene which code for β-glucuronidase enzyme in E. coli. Antibiotic susceptibility profile of the isolate using a panel of 10 antibiotics shows 100% (150/150) resistance to antibiotics rifampicin and penicillin G while 49.3% (74/150) of the isolates and 46.7% (70/150) were susceptible to streptomycin and cefotaxime respectively. Multiple antibiotic resistance phenotypes (MARP) of the isolates showed resistance to two or more test antibiotics while the calculated multiple antibiotic resistance index (MARI) for the tested isolated is 0.49. The detection of potentially pathogenic E. coli in the final effluents suggestspotential danger to the receiving water bodies where the effluents are discharge. The high MARI valued obtained in this study indicates that the isolates are form environment where the tested antibiotics are being used and may further lead to the spread of multiple antibiotics resistance among other pathogens that may be present in the same environment.

- Description: The human immunodeficiency virus (HIV) is one of the most common and dreaded diseases of the 21th century. Today, the disease is still spreading with increasing incidence. “Since the beginning of the epidemic, almost 75 million people have been infected with the HIV virus and about 36 million people have died of HIV. Globally, 35.3 million [32.2–38.8 million] people were living with HIV at the end of 2012” (http://www.who.int/gho/hiv/en/). Several studies have been conducted on herbs under a multitude of ethnobotanical grounds. The use of medicinal plants for the management of HIV has become a common practice especially, in the Eastern Cape Province of South Africa (Wilfred Otang Mbeng, 2013 PhD thesis, UFH). At the beginning of this programme, an ethnomedicinal survey of plants used for the management of HIV infection was carried out in targeted areas of the Province and information on the names of plants, the parts and the methods of preparation were collected. The survey revealed that 18 species representing 12 families were found to be commonly used for the management of HIV, as well as other opportunistic diseases such as tuberculosis, diabetes mellitus, sores, high blood pressure, etc. Carpobrotus edulis was selected for this research because it was the most frequently used in the Province. The foliar micro morphological contents of the plant, its phytochemical and antioxidant activity, in vitro antimicrobial activity, inhibitory effect against HIV-1 protease and reverse transcriptase, mechanisms of action and cytotoxicity were investigated. In terms of the foliar micro morphological contents in plants, an electron microscopy scanning (SEM) was completed. Investigation revealed that both glandular tricomes and calcium oxalate crystals (CaOX) were observed. Consequently, it is hypothesized that the bioactive therapeutic compounds secreted by C. edulis may be produced in the glandular trichomes. An investigation of phytochemical content of the plant extracts (C. edulis) was completed using four solvent extracts (hexane, acetone, ethanol and water). Results of the phytochemical analysis showed that proanthocyanidins (86.9 ± 0.005%) where highest in the aqueous extract with phenolics at 55.7 ± 0.404% in acetone extract, tannin at 48.9 ± 0.28% in ethanol extract, while the hexane extract had the highest levels of flavonoids (0.12 ± 0.05%) and flavonols (0.12 ± 0.05%). Antioxidant studies of the various extracts revealed that aqueous and ethanol extracts were found to be the best solvents for antioxidant activity in C. edulis leaves. GC-MS analysis of the essential oil from C. edulis leaves revealed that the essential oil contained at least 28 compounds. These included, in order of abundance: Oxygenated monoterpenes (36.61%); fatty acids esters (19.25%); oxygenated diterpenes (19.24%); monoterpenes (10.6%); sesquiterpenes (3.58%); and diterpenes (1.43%). Similarly, a GC-MS analysis of the crude hexane, acetone and ethanol extracts from C. edulis leaves identified a total of 59 compounds. Of the 59 compounds, 12 major phyto-metabolites that are active against infectious diseases were identified. To comfirm the potential use of C. edulis to treat infectious disease, antifungal activity of the crude essential oil extract and the four solvent extracts were tested against Candida albicans, Candida krusei, Candida glabrata, Candida rugosa and Cryptococcus neoformans strains. The essential oil extract was found to be the most active against all the fungal strains tested and performed better than the four extracts used various solvents used to extract (hexane, acetone, ethanol and water) the C. edulis leaves were tested for antibacterial and anti HIV-1 reverse transcriptase (RT) activity. The results indicated that both gram-positive and gram-negative isolates were inhibited by the extracts (hexane, acetone and ethanol) but antimicrobial activity was observed for the water extract. The lowest minimum inhibitory concentration values were obtained for the ethanol extract, followed by acetone and hexane extracts. No inhibition of HIV-1 reverse transcriptase was observed for any of the leaf extracts, even up to concentrations of 16 mg/ml. The potential inhibitory activities of the various solvent extracts against HIV-1 protease were evaluated at four different concentrations (16, 1.6, 0.16 and 0.016 mg/ml). Results indicated that the water extract showed almost 100% inhibition of HIV-1 protease activity, with an IC50 of 0.86 mg/ml leaf extract. Other solvent extracts (hexane, acetone and ethanol) however, did not show any inhibition activity above that observed for the DMSO control. The metabolic components in the water extract were subjected to LC-MS/MS analysis, which identified at least 91 compounds present in the water extract. Further studies involving the molecular modelling need to be carried out to confirm the inhibitory potential of these compounds. The cytotoxicity of the water extract of C. edulis leaves was also screened using human Chang liver cells at concentrations ranging bewteen 0.005 mg/ml and 1 mg/ml. Results indicated that the water extracts were not toxic. In conclusion the results from this study support the use of water extracts of C. edulis leaves by traditional healers to treat HIV infections and have identified possible mechanisms of action of the water extracts of C. edulis.

- Description: Certain areas in the world still depend on the receiving water bodies as sources of domestic water and for recreational purposes. The discharge of poor quality effluents from wastewater treatment plants can impact negatively on these water bodies, as they can act as vehicles for pathogens to the environment, posing a threat to humans if such water is used without precaution. Vibrio species are amongst those pathogens that can survive wastewater treatment processes, ending up in the environment, hence the aim of this study was to evaluate the final effluents of some wastewater treatment plants as sources of vibrio pathogens. Five wastewater treatment plants (WWTP) located in Amathole and Chris Hani District Municipalities in the Eastern Cape were used in this study. Samples were collected monthly from September 2012 – August 2013 and analysed using the standard membrane filtration technique. Yellow and green colonies on TCBS agar were enumerated as presumptive Vibrio species and expressed as CFU/100ml for each plant. Colonies were later picked based on their phenotypic characteristics, sub-cultured on fresh TCBS agar to ascertain purity. These presumptive isolates were then subjected to Gram staining and Oxidase test. Gram negative and Oxidase positive isolates were selected for further confirmation using Polymerised Chain Reaction (PCR). PCR was also employed for characterisation of Vibrio into three species viz V. parahaemolyticus, V. fluvialis and V. vulnificus. Antibiogram profile of the characterised species was then determined together with the presence of relevant antibiotic resistance genes Vibrio densities for the twelve month period ranged between 0 - 1.48×104 CFU/100ml with two of the plants located in East bank and Queenstown characterized by extremely high counts and one plant( Reeston) with very low counts.Three hundred presumptive Vibrio isolates were screened for identity confirmation. Of these, the dominating species found was V. fluvialis (28.6%) followed by V. vulnificus (28%) and the least was found to be V. parahaemolyticus (11.6%). The remaining unidentified 31.6% were suspected to belong to other Vibrio species not covered within the scope of this study. All the confirmed isolates i.e., V. parahaemolyticus, V. vulnificus and V. fluvialis were susceptible to imipenem, gentamicin and meropenem and resistant to only tetracycline. Between 60-100% of the V. parahaemolyticus isolates, 7.1% to 100 % V. vulnificus isolates and 2.5 to 100 % V. fluvialis showed resistances to polymixin B, sulfamethazole, erythromycin, penicillin G, chloramphenicol, trimethroprim and trimethroprim & sulfamethazole. Antibiotic Resistance Genes that were assessed included dfRA, SXT, floR and Sul2 varying in proportion with each species showing diversity in the Vibrio community. The dfR A gene was detected in all the V. parahaemolyticus isolates while floR gene was not detected in any of the isolates belonging to the three species. The distribution of sul2 gene cut across the species being 1% (1) in V. fluvialis, 3% (1) in V. parahaemolyticus and 4% (3) in V. vulnificus. The SXT gene was only determined in V. parahaemolyticus. It is clear that the final effluents of the selected plants are reservoirs for Vibrio pathogens as well as antibiotic resistance genes in the environment. The isolation of Vibrio from WWTP shows that this pathogen is in circulation in some pockets of the population. Therefore, wastewater treatment plants need to be properly monitored to ensure that they comply with set guidelines.

The discharge of poor quality effluents from wastewater treatment plants can impact negatively on these water bodies, as they can act as vehicles for pathogens to the environment, posing a threat to humans if such water is used without precaution. Vibrio species are amongst those pathogens that can survive wastewater treatment processes, ending up in the environment, hence the aim of this study was to evaluate the final effluents of some wastewater treatment plants as sources of vibrio pathogens. Five wastewater treatment plants (WWTP) located in Amathole and Chris Hani District Municipalities in the Eastern Cape were used in this study. Samples were collected monthly from September 2012 – August 2013 and analysed using the standard membrane filtration technique. Yellow and green colonies on TCBS agar were enumerated as presumptive Vibrio species and expressed as CFU/100ml for each plant. Colonies were later picked based on their phenotypic characteristics, sub-cultured on fresh TCBS agar to ascertain purity. These presumptive isolates were then subjected to Gram staining and Oxidase test. Gram negative and Oxidase positive isolates were selected for further confirmation using Polymerised Chain Reaction (PCR). PCR was also employed for characterisation of Vibrio into three species viz V. parahaemolyticus, V. fluvialis and V. vulnificus. Antibiogram profile of the characterised species was then determined together with the presence of relevant antibiotic resistance genes Vibrio densities for the twelve month period ranged between 0 - 1.48×104 CFU/100ml with two of the plants located in East bank and Queenstown characterized by extremely high counts and one plant( Reeston) with very low counts.Three hundred presumptive Vibrio isolates were screened for identity confirmation. Of these, the dominating species found was V. fluvialis (28.6%) followed by V. vulnificus (28%) and the least was found to be V. parahaemolyticus (11.6%). The remaining unidentified 31.6% were suspected to belong to other Vibrio species not covered within the scope of this study. All the confirmed isolates i.e., V. parahaemolyticus, V. vulnificus and V. fluvialis were susceptible to imipenem, gentamicin and meropenem and resistant to only tetracycline. Between 60-100% of the V. parahaemolyticus isolates, 7.1% to 100 % V. vulnificus isolates and 2.5 to 100 % V. fluvialis showed resistances to polymixin B, sulfamethazole, erythromycin, penicillin G, chloramphenicol, trimethroprim and trimethroprim & sulfamethazole. Antibiotic Resistance Genes that were assessed included dfRA, SXT, floR and Sul2 varying in proportion with each species showing diversity in the Vibrio community. The dfR A gene was detected in all the V. parahaemolyticus isolates while floR gene was not detected in any of the isolates belonging to the three species. The distribution of sul2 gene cut across the species being 1% (1) in V. fluvialis, 3% (1) in V. parahaemolyticus and 4% (3) in V. vulnificus. The SXT gene was only determined in V. parahaemolyticus. It is clear that the final effluents of the selected plants are reservoirs for Vibrio pathogens as well as antibiotic resistance genes in the environment. The isolation of Vibrio from WWTP shows that this pathogen is in circulation in some pockets of the population. Therefore, wastewater treatment plants need to be properly monitored to ensure that they comply with set guidelines.

Researchers agree that learners have to be exposed to various strategies so that they are able to see that there is a difference between additive reasoning and multiplicative reasoning. In order to classify learners’ conceptual understanding of multiplication an analysis of the data was done and learners were allocated levels according to the Wright, et al. (2006) levels of achievement. For the classification of learner dispositions, the data was analysed in terms of the elements of productive disposition as defined by Kilpatrick, et al. (2001) and Carr and Claxton (2002). The key findings of the study indicate that for conceptual understanding most of the learners depended on using concrete materials in solving multiplication and they also used basic strategies and methods. The findings for productive dispositions were that most of the learners saw themselves as competent in doing multiplication but the aspect of sense making and steady effort was less developed. The findings for the relationship between conceptual understanding and productive disposition were that both strands have a mutual relationship in which one helped the other to develop.

Exchange rate misalignment and economic growth: a case study of South Africa

This study examines the relationship between the exchange rate misalignment and economic growth in South Africa, by specifically looking at the fundamental determinants of the real exchange rate and the impact of the misalignment on economic growth for the period 1980 to 2012. There has been a lot of research conducted on DCB and different terms and phases have been used to describe the behaviour, ie “dysfunctional customer behaviour”, (Daunt and Harris, 2012), “aberrant consumer behaviour” (Fullerton and Punj, 1993) and “consumer misbehaviour” (Fullerton and Punj, 1997). However, the research conducted on how the behaviour affects retailers, particularly grocery retailers has been limited. The primary objective of this study was to examine how DCB affects grocery retailers in Nelson Mandela Bay Municipality. More specifically, the relationship between DCB (dependent variable) and independent variables - impact on profits (RPROF), impact on inventory (INV), impact on grocer security (SECU), impact on employees (EMPL), impact on customers (CUST) and measures to minimize or stop DCB (STOP) was established. Questionnaires were used as a measuring instrument. Out of the 100 questionnaires distributed, 70 were usable. The empirical results showed that there is a significant relationship between DCB and the independent variables. Recommendations on how best the stores can deal with DCB were made and measures to minimize or stop DCB were presented. Recommendations for future studies were also presented.

The excessive costs of health service adopting a 100% staffing model has created the use of agency nursing to help meet fluctuating and unpredictable health care demands in Australia and overseas; agency staff are used to meet the shortfalls in staff-patient ratios (Peerson et al., 2002:505). Hurst & Smith (2011:287) agree that agency nurse staffing form a large part of the health service in the UK and state that this was unlikely to change because of recruitment and retention problems, high absenteeism, and staff wanting to work casually. In their study, they identified three types of agency nurses: A bank nurse – the hospital’s own employees or those supplied by NHS Professionals, who work when required, usually at short notice; agency nurses provided by commercial organisations, who are equally flexible but less familiar with the ward patients and procedures; and permanent ward staff working paid overtime. In South Africa, this also remains a problem in public and private institutions. The World Health Organization (WHO) states that the minimum ratio for nurses to population is 200:100 000 or 500 people per nurse. According to South African Nursing Council statistics of nursing manpower as at the end of 2008, South Africa has 437 nurses for every 100 000 people. This assumed that all nurses registered with SANC are working in South Africa at the time. Therefore, if the assumed number of staff not working in South Africa is subtracted, the ratio can increase to between 600 and 678 people per nurse (WHO, 2006). Joubert (2009:2) shows that a shortage of nurses contributes to deaths in hospitals in South Africa that would otherwise have been avoidable. The use of agency staff is becoming an appropriate means of providing cost-effectiveness and flexibility to staffing needs.

The objectives of the study were to explore and describe experiences of college students, regarding the mentoring role of registered nurses in Public Services of the Eastern Cape, to analyse the results and to make suggestions from findings that were obtained. A qualitative, exploratory, descriptive and contextual design was used for this study to explore and describe the experiences of the college students regarding the mentoring role of registered nurses in the Public Services of the Eastern Cape, South Africa. The researcher studied the population of student nurses in the public nursing college in the Eastern Cape at the East London Campus. Purposive sampling which is referred to as a judgmental approach was used. Data were collected by means of focus group interviews. An interview schedule with an unstructured questionnaire was used to investigate and explore information gained from the participants.
