- Description: Tin(IV) complexes of a 4-methylthiophenyl functionalized porphyrin (1-Sn) and its corrole analogue (2-Sn) were synthesized so that their photophysicochemical properties and photodynamic activities against MCF-7 breast cancer cells could be compared. Singlet oxygen luminescence studies revealed that 1-Sn and 2-Sn have comparable ΦΔΦΔ values in DMF of 0.59 and 0.60, respectively, while the IC5050 values after irradiation of MCF-7 cells for 30 min with a Thorlabs 625 nm LED (432 J · cm−2)−2) were determined to be 12.4 and 8.9 μμM. The results demonstrate that the cellular uptake of 2-Sn and its molar absorptivity at the irradiation wavelength play a crucial role during in vitro cytotoxicity studies.

- Description: Phytochemical investigation of the methanolic extract of Monodora minor Engl. and Diels (Annonaceae) stem bark yielded a new indole (E)-4-(1H-indol-5-yl)-but-3-en-2-one (1), a known indole 5-formyl-1H-indole (2) and an ubiquitous steroid sitosterol (3). The investigations of the methanolic extract of Uvaria tanzaniae Verdc. (Annonaceae) root bark yielded two previously reported C-benzylated dihydrochalcones namely uvaretin (4) and diuvaretin (5). Structures of the isolated compounds were elucidated based on NMR spectroscopy and high resolution electron ionization mass spectrometry (HR-EI-MS) data. All compounds were tested against Trypanosoma brucei brucei and Plasmodium falciparum. At a single concentration (20 μM) in the antitrypanosomal and antiplasmodial assays, compound 4 exhibited remarkable activities against T. brucei brucei and P. falciparum with percentage inhibition of 97.3% and 83.0% respectively, whereas compounds 1, 2, 3 and 5 were inactive. In a dose response antiplasmodial assay compound 4 exhibited moderate activity against P. falciparum with an IC50 value of 7.20 μM.

- Description: Among endo-mannanases, glycoside hydrolase (GH) family 26 enzymes have been shown to be more catalytically active than GH5 enzymes on mannans. However, only GH5 endo-mannanases have been used for the formulation of enzyme cocktails. In this study, Bacillus sp.-derived GH5 and GH26 endo-mannanases were comparatively analysed biochemically for their synergistic action with a commercial cellulase blend, CTec2, during pre-treated lignocellulose degradation. Substrate specificity and thermo-stability studies on mannan substrates showed that GH26 endo-mannanase was more catalytically active and stable than GH5. GH26 also exhibited higher binding affinity for mannan than GH5, while GH5 showed more affinity for lignocellulosic substrates than GH26. Applying the endo-mannanases in combination with CTec2 for lignocellulose degradation led to synergism with a 1.3-fold increase in reducing sugar release compared to when CTec2 was used alone. This study showed that using the activity of endo-mannanases displayed with model substrates is a poor predictor of their activity and synergism on complex lignocelluloses.

- Description: Cellulase cocktails formulated to degrade crystalline cellulose generally contain cellobiohydrolases (CBHs), referred to as CBHI (Cel7A) and CBHII (Cel6A), as the major constituents. The combined hydrolytic activities of CBHI and CBHII improve the release of fermentable sugars (β-1,4-cellobiose as the main product) from crystalline cellulose. In this study, a novel cellobiohydrolase (Exg-D) sourced from a metagenome of hindgut bacterial symbionts of a termite was heterologouly expressed, purified, and functionally characterised. Exg-D specific activity was higher on insoluble barley β-glucan (38.94 U/mg protein), soluble wheat flour β-glucan (12.71 U/mg protein) and oat β-glucan (8.89 U/mg protein) compared to cellulosic substrates; Avicel and CMC. We further explored Exg-D activity on the unpretreated or NaOH-pretreated (mercerised) Avicel and compared its activity to commercially available CBHI and CBHII on these celluloses. CBHI displayed the highest activity of 4.74 U/mg protein on mercerised cellulose followed by CBHII (2.14 U/mg protein), while Exg-D activity on untreated and mercerised cellulose was 1.66 and 1.67 U/mg protein, respectively. The high activity of CBHI was supported by binding assays, which revealed that CBHI has a higher binding capacity towards crystalline cellulose compared to Exg-D and CBHII. Only CBHI and CBHII showed synergism during the hydrolysis of mercerised Avicel, showing a degree of synergy (DS) of about 1.299 and yielded about 1.43 μmol/ml of reducing sugars higher than control. In contrast, Exg-D and CBHII displayed synergism during β-glucan degradation, displaying a DS of about 1.22. Thus, we propose that Exg-D should only be used synergistically with other CBHs to degrade mixed linked-β-(1,3)-(1,4)-glucan.

- Description: In this study, solvent dependence of electronic and nonlinear optical properties of a new Schiff base, 2-(((5-aminonaphthalen-1-yl)imino)methyl) phenol (DANOHB), has been investigated. Electronic and optical properties of the Schiff base studied using Uv-visible absorption and fluorescence spectroscopic techniques in solution show that the Schiff base exhibits fluorescence with a quantum yield of 0.21 and a Stokes shift of 39 nm. Quantum chemical calculations were performed on the Schiff base at time-dependent density functional level of theory using Becke-3-Lee-Yang-Parr method with 6-311++G(d,p) basis set. Solvent dependence of the excited state energies (EE), energy gap (ΔE), first-order (β) and second-order (γ) hyperpolarizabilities were studied in gas, cyclohexane and ethanol. The medium perturbed the energy levels which implies that the reactivity, activity and stability of the system are solvent sensitive. The γ is more than thirty times higher than urea (standard NLO material) and this property is further enhanced in less polar medium. Small ΔE and large hyperpolarizabilities obtained for this system suggest its good potential in photonics and nonlinear optical devices.

- Description: This work entails the study of electronic and nonlinear optical (NLO) properties of two synthesized Schiff bases namely 3-(((2-methyl-4-nitrophenyl)imino)methyl)phenol (MMP) and 3-(((2,4-dinitrophenyl)imino)methyl)phenol (DMP). Electronic absorption properties studied using UV–Visible spectrophotometry in ethanol reveal low-energy absorption indicating a low band gap. Time-dependent density functional calculations on their excited state properties with ωB97X-D/6-311++G(d,p) method show comparable transition energies and intensities with the experimental results. Energy gaps (ΔE) and NLO properties were evaluated from the frontier orbitals and hyperpolarizabilities (βtot and γ) in gas, cyclohexane and ethanol. Low ΔE obtained shows the potentials of the systems as organic semiconductor with remarkable band gap stabilization in ethanol. Higher βtot and γ than urea imply that the Schiff bases possess potentials for NLO applications. The solvatochromic shift in transition energies and perturbation of electronic properties reveal the solvent dependence of the electronic and optical properties of MMP and DMP due to solute-solvent interactions.

- Description: Antiretroviral therapy remains the most efective means of managing the human immune defciency virus/acquired immune defciency syndrome (HIV/AIDS). Application of therapeutics has been hampered by factors including poor bioavailability of most anti-retroviral compounds (ARV), side efects and an alarming emergence of drug resistant strains of the virus. Recent developments and use of drug delivery systems (DDS) has shown potential for improving the pharmacological profle of ARV. Amongst these complex DDS, liposomes have been explored for delivery of ARV. In this study, we have aimed at exploring efcient encapsulation of efavirenz (EFV), a potent ARV using diferent mass ratios of crude soybean lecithin and cholesterol. The EFV-loaded liposomes (EFL) were prepared using thin flm hydration and evaluated for particle size, zeta potential (ZP), encapsulation efciency (EE%), morphology and drug release studies. Diferential scanning calorimetry (DSC), X-ray difraction (XRD), energy dispersity spectroscopy (EDS) and Fourier transform infrared (FTIR) spectroscopy were used for comprehensive physicochemical characterization of EFL. EFL exhibited high encapsulation (99%) in 1:1 crude lecithin to cholesterol mass ratio. The average particle size and Zeta Potential of EFL were found to be 411.10±7.40 nm and −53.5.3±0.06 mV, respectively. EFL showed a relatively controlled EFV release behaviour that was similar to the dissolution profle of un-encapsulated EFV. This suggests that EFL represents a promising vehicle for efective EFV delivery while providing the advantages of a nano-scaled delivery system

- Description: Background: Over the past three decades, a range of international stakeholders have highlighted the possibility that genomic research may impact stigma associated with psychiatric disorders. Limited research has been conducted in Africa to investigate this relation. Method In the present study, using focus group discussions, we explored the relation between genetic attribution and stigma among 36 Xhosa people with schizophrenia. We addressed three main questions: (1) What causal beliefs do Xhosa people with schizophrenia use to explain their illness and to what extent do genetic explanations play a role in these beliefs? (2) What are the internalised stigma experiences of Xhosa people with schizophrenia? (3) How do genetic explanations relate to stigma experiences, if at all? Results Most participants were able to define genetics and some linked genetics to disease causation. Despite adequate knowledge of genetics and an emphasis on genetic explanations of schizophrenia in the study, most participants held a multitude of causal explanations including: psychosocial, environmental, and cultural. Moreover, participants rarely mentioned disease cause when describing their stigma experiences. Discussion For this population group, there was no straight-forward relation between a genetic attribution and stigma. Therefore, we did not fnd evidence that genetic attribution may signifcantly increase stigma. Although North American and European literature provides conficting evidence regarding this relation, there is increased consensus that biomedical explanations for psychiatric disorders may reduce blame. This study found evidence supporting that consensus. This study provides an empirical foundation to inform ongoing work on the psychosocial implications of psychiatric genomics research in non-Western contexts.

- Description: Water hyacinth, Pontederia crassipes (Martius) [≡Eichhornia crassip es (Martius) Solms-Laubach] (Pontederiaceae), is native to South America, but has expanded its range to many other regions of the world including South Africa. Megamelus scutellaris Berg (Hemiptera: Delphacidae) was released as a biological control agent and has established in several regions. Recently, the indigenous species Echthrodelphax migratorius Benoit, (Hymenoptera: Dryinidae) was discovered in South Africa parasitising M. scutellaris. This newly discovered relationship might have repercussions for the efficacy of biological control of water hyacinth by the delphacid. The wasp may negatively impact M. scutellaris populations making it difficult for the agent to successfully manage the invasive weed. Contrarily, the parasitoid may be beneficial by keeping the M. scutellaris populations stable, serving as a natural enemy.

- Description: Lycium ferocissimum (African boxthorn) is a Weed of National Significance in Australia. Biological control may have potential to manage this weed, but taxonomic uncertainty needs to be addressed first to facilitate searches for potential agents. We sampled putative L. ferocissimum (i.e. tentatively identified morphologically in the field) across its native range in South Africa and introduced range in Australia. Morphometric and genetic analyses were conducted to confirm the species identity of these samples, and to assess morphological and genetic variation across both ranges. All samples collected in Australia were confirmed as L. ferocissimum, with no evidence of hybridisation with any other Lycium species. Nuclear and chloroplast genetic diversity within L. ferocissimum across both South Africa and Australia was low, with no evidence of genetic structure. One of the two common chloroplast haplotypes found across Australia was found at only two sites in South Africa, both near Cape Town, suggesting that the Australian lineage may have originated from this region. Ten samples from South Africa putatively identified in the field as L. ferocissimum were genetically characterised as different (unidentified) Lycium species. Our morphometric analyses across different Lycium species in South Africa did not identify any leaf or floral characteristics unique to L. ferocissimum, and thus morphological identification of the latter species in its native range may remain problematic. To ensure the correct Lycium species is surveyed for candidate biological control agents we suggest that individuals should be permanently tagged and putative morphological determinations supplemented with genetic analyses to confirm species identity.

- Description: Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidae) is native to South America but has invaded the Afro-Eurasian supercontinent where it is currently the most devastating invasive arthropod pest of tomato. As a part of the first classical biological control programme against T. absoluta in Africa, a larval parasitoid, Dolichogenidea gelechiidivoris Marsh. (Syn.: Apanteles gelechiidivoris Marsh) (Hymenoptera: Braconidae), of T. absoluta was imported from Peru into the quarantine facility of the International Centre of Insect Physiology and Ecology, Kenya. We report on the host larval preference of D. gelechiidivoris and the host suitability, and the parasitoid’s reproductive strategy, including lifetime fecundity and egg maturation dynamics. Dolichogenidea gelechiidivoris females preferentially oviposited in early (1st and 2nd) larval instars of T. absoluta but parasitized and completed development in all four instars of the host. Host instar did not affect D. gelechiidivoris sex-ratio but females reared on the first instar had significantly fewer eggs than when reared in late larval instars (3rd and 4th). Females of the parasitoid emerged with a high mature egg load which peaked 2 d post eclosion. The females of D. gelechiidivoris survived 8.51 ± 0.65 d and produced 103 ± 8 offspring per female at 26 ± 4 °C (range: 24 to 29 °C) and 50–70% relative humidity (RH) with males present and fed honey-water (80% honey). Increasing maternal age decreased the proportion of female offspring. Under the aforementioned laboratory conditions, the Gross and Net reproductive rates were 72 and 39.5 respectively, while the mean generation time was 20 d. The potential intrinsic rate of natural increase was 0.18. This study shows that D. gelechiidivoris is a potential biological control agent of T. absoluta and should be considered for release in Kenya and across Africa following host specificity testing and risk assessments.

- Description: Background. Evidence from high-income countries suggests that childhood trauma is associated with schizophrenia. Studies of childhood trauma and schizophrenia in low and middle income (LMIC) countries are limited. This study examined the prevalence of childhood traumatic experiences among cases and controls and the relationship between specific and cumulative childhood traumatic experiences and schizophrenia in a sample in South Africa. Methods. Data were from the Genomics of Schizophrenia in the South African Xhosa people study. Cases with schizophrenia and matched controls were recruited from provincial hospitals and clinics in the Western and Eastern Cape regions in South Africa. Childhood traumatic experiences were measured using the Childhood Trauma Questionnaire (CTQ). Adjusted logistic regression models estimated associations between individual and cumulative childhood traumatic experiences and schizophrenia. Results. Traumatic experiences were more prevalent among cases than controls. The odds of schizophrenia were 2.44 times higher among those who experienced any trauma than those who reported no traumatic experiences (95% CI 1.77–3.37). The odds of schizophrenia were elevated among those who experienced physical/emotional abuse (OR 1.59, CI 1.28–1.97), neglect (OR 1.39, CI 1.16–1.68), and sexual abuse (OR 1.22, CI 1.03–1.45) compared to those who did not. Cumulative physical/emotional abuse and neglect experiences increased the odds of schizophrenia as a dose–response relationship. Conclusion. Childhood trauma is common in this population. Among many other benefits, interventions to prevent childhood trauma may contribute to a decreasing occurrence of schizophrenia.

- Description: The aim of these studies was to determine the miscibility of different API with lipid excipients to predict drug loading and encapsulation properties for the production of solid lipid nanoparticles and nanostructured lipid carriers. Five API exhibiting different physicochemical characteristics, viz., clarithromycin, efavirenz, minocycline hydrochloride, mometasone furoate, and didanosine were used and six solid lipids in addition to four liquid lipids were investigated. Determination of solid and liquid lipids with the best solubilization potential for each API were performed using a traditional shake-flask method and/or a modification thereof. Hansen solubility parameters of the API and different solid and liquid lipids were estimated from their chemical structure using Hiroshi Yamamoto’s molecular breaking method of Hansen Solubility Parameters in Practice software. Experimental results were in close agreement with solubility parameter predictions for systems with ΔδT larger than 4.0 MPa1/2. A combination of Hansen solubility parameters with experimental drug-lipid miscibility tests can be successfully applied to predict lipids with the best solubilizing potential for different API prior to manufacture of solid lipid nanoparticles and nanostructured lipid carriers.

This work aimed at evaluating the potential of a cost-effective lipid material, crude soybean lecithin (CL), to co-encapsulate RIF and INH for liposomal dual delivery. RIF was encapsulated in CL-liposomes with/without cholesterol using film hydration method, after which INH was incorporated using a freeze–thawing technique. Dynamic light scattering, differential scanning calorimetry, X-ray diffraction and dialysis were used for liposome characterization. Liposomes containing CL alone (CLL) exhibited 90%encapsulation efficiency for RIF and 59%for INH. The mean size and surface charge of CLL were 1114nm and –63mV, respectively. In addition, CLL showed a controlled release profile for the co-encapsulated drugs. CLL would be promising vehicles for macrophage-targeting drug delivery. The present findings demonstrate the feasibility of using CL for preparation of combination products for liposomal delivery.

- Description: This work reports on the synthesis, photophysico-chemical properties and photodynamic therapy activity of novel zinc (1) and indium (2) tetra substituted cinnamic acid phthalocyanine (Pc) complexes linked to amino functionalized magnetic nanoparticles (AMNPs) through an amide bond. Asymmetric ZnPc complex (3) showed better triplet and singlet oxygen quantum yields as compared to its symmetrical analogues (1 and 2). The AMNPs (1-AMNPs and 2-AMNPs) linked conjugates depicted increased triplet quantum yields in comparison to their unlinked Pcs, while 3-AMNPs showed a decrease compared to 3. The complexes showed increased in-vitro photo-cytotoxic effect against MCF-7 cells with an increase in drug concentration. At 80 µg/mL, 2 and 3, 2-AMNPs and 3-AMNPs with higher singlet oxygen quantum yields caused more cytotoxic effect on the cancer cells in the presence of light as compared to 1 and 1-AMNPs respectively.

- Description: A functional hybrid of graphene quantum dots (GQDs) and cobalt pyrene-derivatized phthalocyanine (CoPc) used as a sensitive nanoprobe for the recognition of cyanide ion (CN−) is described in this work. The fluorescence of GQDs was quenched upon non-covalent (π-π stacking) hybrid formation with CoPc via a possible energy transfer pathway. However, in the presence of CN−, the interaction between GQDs and CoPc was perturbed, such that the fluorescence of GQDs initially quenched by CoPc was found to be efficiently recovered in the presence of CN−. Amongst the molecules and anions tested to ascertain their effects on the fluorescence behaviour of the hybrid, only CN− ion induced the tunable “off-on” restoration of the fluorescence of GQDs, which demonstrates the selectivity of the hybrid towards CN−. The restored fluorescence signals of the GQDs were linearly modulated by different concentrations of CN− and were used for the quantitative assay of CN− with high sensitivity coupled with rapid detection time. The detection was in the linear range of 1.0–50.0 nM with limits of detection (LOD) of 0.5 nM. The analysis of spiked samples for the recovery of CN− further demonstrated the applicability of the hybrid for the satisfactory detection of the target analyte.

- Description: As the world continues to face widespread food insecurity, achieving food security for all at all times is increasingly complicated. In South Africa, social grants and the use of wild foods have been reported as some ways to improve household food insecurity and reduce poverty. The study examined if social grants and consumption of wild foods alleviate food insecurity in South Africa. Household surveys and focus group discussions were conducted along the rural-urban continuum in three South African towns situated along an agro-ecological gradient. We explored the differences in household food security indicators, mean monthly food expenditure and wealth index between households receiving social grants, households consuming wild foods, and those who did not. Households receiving social grants were more food insecure with lower mean monthly food expenditure and wealth index than those who did not. Overall all towns, the use of wild foods improved household food security which was not true within towns where wild foods were mostly consumed by low income and more food insecure households. Social grants alone cannot eradicate food insecurity as the money is not enough to cater for all household needs whilst wild foods can potentially alleviate household food insecurity. As food prices continue to increase gradually and worsening household food insecurity, ways to shift ‘income circumstances’ of households and promote the use and consumption of wild foods which may increase dietary diversity and diversifying food access. Focus should be placed on capacity building, employment creation and promoting awareness on own food production which is one way to improve food security of poor households rather than depending on purchases from the grant money alone.

- Description: Antiplasmodial, antitrypanosomal and anti-HIV-1 activities of crude extracts, fractions and some isolated compounds from two Cameroonian medicinal plants: Antrocaryon klaineanum Pierre (Anacardiaceae) and Diospyros conocarpa Gürke ex K. Schum. (Ebenaceae) were assessed. The phytochemical studies led to the isolation of eight compounds (1–8) from Diospyros conocarpa and six compounds (6, 9–13) from Antrocaryon klaineanum. These compounds were identified as mangiferolic acid (1), 3β, 22(S)-dihydroxycycloart-24E-en-26-oic acid (2), lupeol (3), aridanin (4), betulin (5), betulinic acid (6), bergenin (7), D-quercitol(8), entilin C(9), entilin A(10), antrocarine A(11), 7R,20(S)-dihydroxy-4,24(28)-ergostadien-3-one(12) and stigmasterol glucoside (13). The criteria for activity were set as follows: an IC50 value more than 10 μg/mL for crude extracts and more than 1 μg/mL for pure compounds. The hexane/ethyl acetate (1:1) fraction of A.klaineanum root bark (AKERF1) and the hexane/ethyl acetate (1:1) fraction of A.klaineanum trunk bark (AKETF1) presented the strongest antiplasmodial activities with IC50 values of 0.4 and 4.4 μg/mL, respectively. Aridanin (4) and antrocarine A(11), as well as the crude extract of D.conocarpa roots (EDCR), AKERF1 and AKETF1 showed moderate trypanocidal effects. The crude extract of A.klaineanum root bark (AKER) and AKETF1 exhibited attractive activities on HIV-1 integrase with IC50 values of 1.96 and 24.04 μg/mL, respectively. The results provide baseline information on the use of A.klaineanum and D.conocarpa extracts, as well as certain components, as sources of new antiplasmodial, antitrypanosomal and anti-HIV drugs.
