Peripheral vision field fatigue during simulated driving : the effects of time on task and time of day on selected psychophysiological, performance and subjective responses

- Description: Worldwide, motor accidents are responsible for a large number of deaths and disabilities (Connor et al., 2001), and one of the major causes of motor accidents is driver fatigue. Although majority of drivers are aware of the dangers of fatigued driving, accidents related to this continues to contribute to a large percentage of all accidents, between 5 and 50% (Nilsson et al., 1997; Williamson et al., 2011). The purpose of the research was to establish the effect that fatigue renders on an individual’s peripheral visual field and to determine whether a decrement in driving performance occurs at the same rate as a decrement in peripheral visual performance. Fatigue was induced through time of day as well as time on task. Sixteen students from Rhodes University were recruited, subject to no previous sleep disorders, among other criteria. Each participant was required to partake in two conditions, namely a day condition (09h00–11h00) and a night condition (23h00– 01h00). Each condition consisted of a 90 minute dual task; the primary task was a tracking task, in which participants were instructed to track a white line as accurately as possible. A secondary peripheral response task was introduced, in which participants were instructed to respond as quickly as possible to the peripheral stimuli, by pressing one of two clickers located on the steering wheel. The peripheral stimuli were located at 20º, 30º and 40º visual angle. Psychophysiological, performance and subjective measures were obtained before, during and after the main task. The pre- and post-tests included core body temperature, critical flicker fusion frequency threshold, a digit span memory test, Wits Sleepiness Scale and a NASA-TLX questionnaire. The psychophysiological and performance measures of heart rate, heart rate variability, blink frequency, blink duration, lane deviation, number of saccades towards peripheral stimuli, response time to peripheral stimuli and the percentage of missed peripheral responses were all recorded throughout the 90 minute main dual task. The results revealed significant differences (p<0.05) for heart rate variability, number of saccades towards peripheral stimuli and the Wits Sleepiness Scale, with regard to time of day. For time on task, significant effects were established for lane deviation, response time to peripheral stimuli, percentage of missed peripheral responses, heart rate, heart rate variability, blink frequency, blink duration, critical flicker fusion frequency threshold, core body temperature and the Wits Sleepiness Scale. Eccentricity was analysed and found to be significant for response time to peripheral stimuli, as well as for the percentage of missed peripheral responses; there was a significant increase in both measures with an increase in the stimuli eccentricity. No significances were established for time of day or between the pre- and post-tests conducted for the digit span memory performance; however, a significant interactional effect between the two was established. When assessing the percentage rate of decrement of driving performance compared to the percentage rate in the decrement of the missed peripheral responses, it was found that the percentage rate of decrement was equal for both measures. Thus from this research it can be seen that, concurrent with a decrement in driving performance, there are adverse effects on an individuals' peripheral vision, which have great implications for the safety of workers in industry and transport, as well as motorists. It was also established that time on task is possibly a more appropriate variable to consider than time of day, when implementing work schedules and rest breaks in industry, transport and fields alike, as more significant findings were seen for time on task compared to time of day. , Adobe Acrobat 9.53 Paper Capture Plug-in

- Description: This project considered the use of the yeast Saccharomyces cerevisiae as a biosorbent for the removal and recovery of a range of metals from contaminated waters. S. cerevisiae, as a biosorbent, has the potential to provide a cost effective, selective and highly efficient purification system. Initial studies focused on metal accumulation by an immobilized baker's S. cerevisiae biosorbent. The parameters affecting metal uptake were investigated, these included metal concentration, time and solution pH. Metal uptake was rapid. Gold and cobalt reached saturation within 5 min of contact with the biosorbent in batch reactors. Copper, zinc, nickel, cadmium and chromium reached saturation within 30 min of contact. Metal accumulation was pH dependent and was generally unaffected at a solution pH ≥ 4, and was substantially decreased at pH ≤ 2. The exception was gold which was preferentially accumulated at a solution pH of 2. The immobilized baker's yeast accumulated metals with maximum binding capacities in the order of gold > cadmium > cobalt > zinc > copper > chromium > nickel. A rapid method to assess metal recovery was developed. Bioaccumulated metal was efficiently recovered using dilute mineral acids. Copper recovery of ≥ 80 % was achieved by decreasing the solution pH of the reaction mixture to 2 with the addition of nominal quantities of HCl, H₂SO₄ or RNO₃. Adsorption-desorption over 8 cycles had no apparent adverse effect on metal uptake or recovery in batch reactors. Transmission electron microscopy showed no evidence of damage to cells used in copper adsorption-desorption investigations. Biosorption columns were investigated as bioreactors due to their application potential. The metals investigated were effectively removed from solution. At a saturation threshold, metal uptake declined rapidly. Most metals investigated were desorbed from the columns by eluting with 0.1 M HCl. Initially recoveries of copper, cobalt and cadmium were as high as 100%. Desorbed copper, zinc, cadmium, nickel and cobalt were concentrated in 10 to 15 ml of eluent, representing up to a 40 fold decrease in solution volume. Cadmium, nickel and zinc uptake increased with the second application to the columns. Initial accumulation of gold and chromium was 42.2 μmol/g and 28.6 μmol/g, however, due to the low recoveries of these two metals, a second application was not investigated. Copper was applied to a single column for 8 consecutive adsorption-desorption cycles. Uptake increased from an initial 31.3 μmol/g to 47.8 μmol/g at cycle 7. The potential for selective metal recovery was demonstrated using two biosorption columns in series. Copper was accumulated and recovered most efficiently. Zinc, cobalt and cadmium were displaced to the second column. Copper bound preferentially to zinc at a ratio of 6:1. Copper bound preferentially to cobalt at a ratio of 4:1. Cadmium was only displaced at a ratio of 2:1. The successful transfer of the bioremediation technology from the laboratory to an industrial application has yet to be realized. Bioremediation of a Plaatjiesvlei Black Mountain mine effluent, which contained copper, zinc, lead and iron, was investigated in this project. The removal of the metals was most effective at pH 4. A combined strategy of pH adjustment and bioremediation using immobilized S. cerevisiae decreased the copper concentration by 92.5%, lead was decreased by 90% and zinc was decreased by 60%. Iron was mostly precipitated from solution at pH ≥ 4. An ageing pond at the mine with conditions such as; pH, water volume and metal concentration, which were more conducive to biological treatment was subsequently identified. The investigation indicated a possible application of the biomass as a supplement to chemical remediation. The metal removal capability of a waste brewer's yeast was subsequently investigated. A yeast conditioning step increased metal uptake up to 100% and enhanced reproducibility. Metal removal from solution was rapid and pH dependent. The metals were efficiently removed from solution at pH ≥ 4. Uptake was substantially inhibited at pH ≤ 3. The waste brewer's yeast accumulated metals with maximum binding capacities in the order of copper (25.4 μmol/g) > lead (19.4 μmol/g) > iron (15.6 μmol/g) > zinc (12.5 μmol/g). No correlation between cell physiology and metal uptake was observed. Uptake of the four metals was confirmed by energy dispersive X-ray microanalysis. The interference of lead, zinc and iron on copper uptake by the waste brewer's yeast, and the interference of copper on the uptake of lead, zinc and iron was investigated. Maximum copper uptake was not decreased in the presence of lead. The Bmax remained constant at approximately 25 μmol/g. The dissociation constants increased with increasing lead concentrations. Lead bioaccumulation was significantly decreased in the presence of copper. The type of inhibition was dependent on the initial copper concentrations. Zinc had a slight synergistic effect on copper uptake. The copper Bmax increased from 30.8 μmol/g in a single-ion system to 34.5 μmol/g in the presence of 200 μmol/l of zinc. Zinc uptake was severely inhibited in the presence of copper. The maximum uptake and dissociation constant values were decreased in the presence of copper, which suggested an uncompetitive inhibition. The affinity of copper was substantially higher than zinc. The presence of higher levels of copper than zinc in the yeast cells was confirmed by energy dispersive microanalysis. Copper uptake was decreased in the presence of iron, with the copper Bmax being decreased from 25.4 μmol/g in a single-ion system to 20.1 μmol/g in the presence of 200 μmol/l iron. Iron Bmax values remained constant at 16.0 μmol/g. Combined biosorption and EDXA results suggested the iron bound at a higher affinity than copper to the cell wall. Total copper removal was higher as larger quantities of copper were deposited in the cell cytoplasm. Metal removal from the Plaatjiesvlei effluent by free cell suspensions of the waste brewer's yeast was satisfactory. Copper levels were decreased by 96%, iron by 42%, lead 25% and zinc 2%. Waste brewer's yeast is a cheap source of biomass in South Africa, and could potentially provide the basis for the development of an innovative purification system for metal-contaminated waters.

A collated digital, geological map database for the central Namaqua Province using geographical information system technology

- Description: The genlogy of the Namaqua Province is notoriously difficult to map and interpret due to polymetamorphic and multiple deformation events and limlted outcrop. Current maps of the Province reflect diverse interpretations of stratigraphy as a consequence of these difficulties. A Geographic Information System is essentially a digital database and a set of functions and procedures to capture, analyse and manipulate spatially related data. A GIS is therefore ideally suited to the study and analysis of maps. A digital map database was established, using modem GIS technology, to facilitate the collation of existing maps of an area in the Central Namaqua Province (CNP). This database is based on a lithological classification system similar to that used by Harris (1992), rather than on an interpretive stratigraphic model. In order to establish the database, existing geological maps were scanned into a GIS, and lines of outcrop and lithological contacts were digitised using a manual line following process, which is one of the functions native to a GIS. Attribute data were then attached to the resultant polygons. The attribute database consists of lithological, textural and mineralogical data, as well as stratigraphical classification data according to the South African Committee for Stratigraphy (SACS), correlative names assigned to units by the Precambrian Research Unit, the Geological Survey of South Africa, the Bushmanland Research group and the University of the Orange Free State. Other attribute data included in the database, are tectonic and absolute age information, and the terrane classification for the area. This database reflects the main objective of the project and also serves as a basis for further expansion of a geological GIS for the CNP. Cartographic and database capabilities of the GIS were employed to produce a collated lithological map of the CNP. A TNTmipsTM Spatial Manipulation Language routine was written to produce a database containing two fields linked to each polygon, one for lithology and one for a correlation probability factor. Correlation factors are calculated in this routine from three variables, namely the prominence a worker attached to a specific lithology within a unit or outcrop, the agreement amongst the various workers on the actual lithology present within an outcrop, and the correspondence between the source of the spatial element (mapped outcrop) and the source of the attribute data attached to it. Outcrops were displayed on the map according to the lithology with the highest correlation factor, providing a unique view of the spatial relationships and distribution patterns of lithological units in the CNP. A second map was produced indicating the correlation factors for lithologies within the CNP. Thematic maps are produced in a GIS by selecting spatial elements according to a set of criteria, usually based on the attribute database, and then displaying the elements as maps. Maps created by this process are known as customised maps, since users of the GIS can customise the selection and display of elements according to their needs. For instance, all outcrops of rock units containing particular lithologies of a given age occurring in a specific terrane can be displayed - either on screen or printed out as a map. The database also makes it possible to plot maps according to different stratigraphic classification systems. Areas where various workers disagree on the stratigraphic classification of units can be isolated, and displayed as separate maps in order to aid in the collation process. The database can assist SACS in identifying areas in the CNP where stratigraphic classification is still lacking or agreements on stratigraphic nomenclature have not yet been attained. More than one database can be attached to the spatial elements in a GIS, and the Namaqua-GIS can therefore be expanded to include geochemical, geophysical, economic, structural and geographical data. Other data on the area, such as more detailed maps, photographs and satellite images can be attached to the lithological map database in the correct spatial relationship. Another advantage of a GIS is the facility to continually update the database(s) as more information becomes available and/or as interpretation of the area is refined.
