Genetic diversity, evolutionary relationships and conservation of southern African Labeo fishes in relation to water management

- Description: Labeo spp. are large, herbivorous fishes that are important components of aquatic ecosystems and are a high conservation priority in South Africa. This thesis contributes to determination of conservation priorities for Labeo umbratus (Smith 1841) by resolving the taxonomic status of this species in the evolutionary context of southern African Labeo spp., assessing the presence of unique lineages in historically isolated river basins, and assessing the threat of intra- and interspecific hybridisation associated with introductions. Phylogenetic analyses of five DNA sequence data sets (cytochrome c oxidase subunit I gene [COI], cytochrome b gene [Cyt b], Recombination activating gene 1 [Rag1], COI+Rag1 and COI+Cyt b+Rag1) showed that the Labeo umbratus group (sensu Reid, 1985), which comprises the species Labeo umbratus, Labeo capensis (Smith 1841), Labeo seeberi Gilchrist and Thompson 1911 and Labeo rubromaculatus Gilchrist and Thompson 1913, is monophyletic, morphologically distinct and geographically disjunct from other African Labeo spp. groups except in the Tugela River system were L. rubromaculatus co-occurs with Labeo molybdinus Du Plessis 1963. Phylogeographic analysis of mitochondrial DNA (Cyt b) sequence data demonstrated that the populations of the L. umbratus from the Orange and the southward-flowing river systems are reciprocally monophyletic and were identified as evolutionary significant units. The populations in the southward-flowing river systems were further divided into southwestern (Gourits and Gamtoos) and southeastern (Sundays, Bushmans, Great Fish, Keiskamma, Buffalo and Nahoon) polyp hyletic sublineages. Four management units (Gourits Basin; Gamtoos Basin; Sundays+Bushmans+Great Fish River Basins; and Keiskamma+Buffalo+Nahoon River Basin) were not reciprocally monophyletic but were proposed on the basis of containing unique haplotype frequencies for conservation purposes. To evaluate the threat of hybridisation to the genetic integrity of L. umbratus, the occurrence of Labeo umbratus x L. capensis hybrids was investigated using mtDNA Cyt b and nDNA S7 intron sequence data and morphological data. Genetic evidence for interspecific hybridisation was detected for populations in two impoundments, Hardap Dam (Orange River Basin) and Darlington Dam (Sundays River Basin, Eastern Cape, South Africa). Some putative hybrids were identifiable morphologically on account of intermediacy between the parental species in meristic and morphometric characters. Translocation via direct stocking (Hardap Dam) or via an inter-basin water transfer scheme (Darlington Dam) was identified as a driver for hybridisation. Introductions associated with an inter-basin water transfer scheme has resulted in introgression between the previously isolated Orange River and southern lineages of L. umbratus. Further translocation of fish from these affected areas to non-contaminated river systems and impoundments such as Kat River (Great Fish River) and Slagboom (Sundays River) should be avoided.

The impact of emotional intelligence on the work and carrer performance of early career academics at the University of Fort Hare

- Description: Orientation – Early Career Academics (ECAs) in their quest to make significant strides in their academic career are faced with a changing academic context, limited support from employers and circumstances emanating from globalization. All these may arouse emotions, fears, anxieties and uncertainties. Many authorities have exhibited emotional intelligence (EI) to be important in terms of self-management, coping and adaptation. In this regard EI may be influential in the career performance of ECAs. Research Purpose – The main aim of this study was to investigate the effect of EI on the career and work performance of ECAs at the University of Fort Hare. Motivation of the Study – EI is criticized for not being easily measurable and also for not being a sufficient predicting factor to individual performance. Little research exists on EI as a predictor to ECAs career progression. Research Approach, Design and Method – The study utilized quantitative data measurement scales. The Schutte Emotional Intelligence Scale was used to measure EI whilst career performance was measured using the Perceived Career Success Scale. The principles of structural equation modelling were applied in formulating the research hypotheses and in data analysis. Main Findings - Exploratory factor analysis extracted four factors from the SEIS, which were named expression of emotion, perception of emotion, use of emotion and regulation of emotion. Expression, perception and regulation of emotions were found to significantly influence job success, interpersonal success, non-organisational success and hierarchical success. The overall EI was not significant to explain change in ECAs career performance. The researcher also found no significant differences in the EI scores on the basis of ECAs age, gender and work experience. Practical/Managerial Implications - The findings to this study may be useful for career counselling and personal development such that an individual may be able to maximize performance and achievement of career goals. The study recommends that EI may be incorporated in learning programs so that ECAs and other professionals may improve their EI. Future research is encouraged on both ECAs and their senior counterparts in the same context of EI. Contribution or value-add – The study contributes to the debate on the predictive power of EI which is criticised by some authorities in the field.

- Description: Concentrating on the Katberg and Burgersdorp Formations, this research examined the sedimentary environments inter alia by conducting heavy mineral analysis, interpreting provenance and determining stratigraphic interrelationships of the two formations in the Tarkastad Subgroup (Beaufort Group, Karoo Supergroup) around the town of Whittlesea in the Eastern Cape Province of South Africa. It has been observed that the strata of the Katberg Formation are dominated by sandstones of a braided river system. On the other hand, the Burgersdorp Formation comprises mainly dark greyish-red mudstone, red shale with subordinate siltstone and red sandstone deposited on floodplains and channels of meandering rivers. The Katberg Formation becomes progressively less arenaceous to the north of the study area until it is virtually indistinguishable from the overlying Burgersdorp Formation. Dolerite dykes and sills that intruded the Karoo Supergroup disturbed and baked the sedimentary strata of the Tarkastad Subgroup as well as other Karoo successions. The Katberg formation is mostly found in elevated areas, commonly metamorphosed and capped by dolerite sills while the Burgersdorp is found in low lying areas and is less metamorphosed or not metamorphosed at all. Lithologies of the two formations and the Karoo dolerites have been geologically mapped and the interfingering relationship between the Burgersdorp and Katberg Formation is also shown on the map. Horizontal laminae, planar bedding and planar and trough cross-bedding characterise lithofacies in the two formations. The fossil hunt done during the course of the study did not yield any positive results in both formations. Initially, the pebbles from outcrops of the Katberg Formation in the East London area were described in hand specimen by Mountain (1939). This study took it further with petrographic- and modal analysis as well as grain size analysis for the sedimentary pebbles. Microscopic studies done on the rock samples collected in the study area revealed quartz and feldspar as the dominant constituents. Most of the feldspar is hazy as a result of dissolution and alteration to clays. Sparse sedimentary rock fragments and quartz overgrowths in some of the rocks and grains respectively, indicate phases of recycling from older sedimentary sources. Furthermore, the samples have a wide range of grain sizes, sorting, grain shapes and classification categories. Textural attributes of sediments such as mean grain size, sorting, skewness and kurtosis were used to characterise the nature of the transport processes and to reconstruct the depositional environment of the sediments. The sandstones of both the Katberg and Burgersdorp Formations are medium-grained and moderately sorted to moderately well sorted but the pebbles are very coarse- to coarse-grained. Deducing sediment provenance from the final product of a basin fill is anything but straight-forward because rocks break down and unstable minerals change during transportation along the pathways from source to basin and afterwards during diagenesis. Heavy mineral studies coupled with geochemical sandstone analysis were used to unravel the sources of sediments. Among the heavies found were opaque and non-opaque minerals which are highly resistant to weathering with zircon being the most dominant. The heavy mineral grains vary in shape from subround to angular-subround in shape. The integrated results of petrography, XRD, XRF and SEM/EDX were used to identify the source rock character and to evaluate the relative role of tectonics and climate in determining the geochemical composition of the Katberg and Burgersdorp sandstones. Sandstone composition was used to decipher the provenance and tectonic setting of source areas by standard triangular QtFL and QmFLt diagrams, tectonic setting discrimination diagrams and source rock composition diagrams to characterise the source rocks.

The current role of farmers’ associations in the Albany Area

- Description: The primary aim of this study was to analyse the current roles that farmers’ associations in the Albany Area fulfil, by determining their current practices, describing the internal measures of effectiveness, and to explain why there are some farmers’ who do not belong to farmers’ associations. In doing this, the researcher was able to determine whether maintaining the status quo would effectively satisfy the members of farmers’ associations. In turn, recommendations were made to farmers’ associations on the roles which could be improved, in order to obtain a higher level of satisfaction among members. In order to achieve the primary aim of the study, interviews were conducted and questionnaires were administered to members of, non-members of, and chairpersons of farmers’ associations in the Albany Area, thus implying that a pragmatic research paradigm was employed to conduct the study. The key findings of this study revealed that farmers’ associations in the Albany Area do perform the five roles of organised associations. However, the manner in which the roles are performed appears to be of a subpar standard. There are 29 services which are aligned with the five roles which are to be performed by organised associations. With regards to farmers’ associations, 18 of the 29 services are able to obtained by non-members through alternative mediums, at a cost lower than that of the membership fee. This does not entice members to join farmers’ associations because the benefits of membership are not seen. Using the competing values framework, we were able to determine the values incorporated into the daily management of farmers’ associations in the Albany Area. Based on the data analysis farmers’ associations were found to have an internal outlook. This could be the overarching reason as to why non-members of farmers’ associations are able to access 18 of the 29 services offered without actually being a member of the respective associations. It was concluded that while farmers’ associations do perform the five roles of organised associations, it does not imply that they do so effectively. This is due to the services offered falling under both public and private good categories. The results showed that 18 of the services offered were classified as public goods, which could be the overarching reason for low levels of membership in farmers’ associations in the Albany Area. An investigation into the feasibility of incorporating ketoconazole into solid lipid microparticles

- Description: One of the major challenges of the oral administration of ketoconazole (KTZ), an inhibitor of sterol 14α demethylase, used in the management of systemic and topical mycoses in immuno-compromised and paediatric patients is the lack of availability of liquid dosage forms. In order to overcome this challenge, extemporaneous preparations have been manufactured by care-givers and health care providers by crushing or breaking solid oral dosage forms of KTZ and mixing with a vehicle to produce a liquid dosage form that can be swallowed by patients. However, the use of extemporaneous preparations may lead to under or over-dosing if the care-givers are not guided accordingly. Furthermore, the dearth of information on the stability of these KTZ-containing extemporaneous preparations may lead to ineffective antifungal therapy and complicate the problems of resistance as it is difficult to estimate the shelf-lives of these extemporaneous products under varying storage conditions due to the susceptibility of KTZ to chemical degradation. Therefore, there is a need for formulation scientists to develop novel drug delivery systems that avoid the need for extemporaneous preparations, possess well-established limits of stability and minimize the risks of systemic adverse effects to facilitate KTZ therapy. The use of solid lipid microparticles (SLM) as potential carriers for the oral administration of KTZ was investigated since solid lipid carriers are known to exhibit the advantages of traditional colloidal carriers. The research undertaken in these studies aimed to investigate the feasibility of developing and manufacturing solid lipid microparticles (SLM), using a simple micro-emulsion technique, as a carrier for KTZ. Prior to pre-formulation, formulation development and optimization studies of KTZ-loaded SLM, it was necessary to develop and validate an analytical method for the in vitro quantitation and characterization of KTZ in aqueous dispersions of SLM during development and assessment studies. An accurate, precise, specific and sensitive reversed-phase high performance liquid chromatographic (RP-HPLC) method coupled with UV detection at 206 nm was developed, optimized and validated for the analysis of KTZ in formulations. Formulation development studies were preceded by solubility studies of KTZ in different lipids. Labrafil® M2130 CS was found to exhibit the best solubilising potential for KTZ. Pre-formulation studies were also designed to determine the polymorphic behavior and the crystallinity of KTZ and Labrafil® M2130 CS that was used for subsequent manufacture of the solid lipid carriers. DSC and FTIR studies revealed that there were no changes in the crystallinity of KTZ or Labrafil® M2130 CS following exposure to a temperature of 60°C for 1 hour. In addition the potential for physicochemical interaction of KTZ with the lipid Labrafil® M2130 CS was investigated using DSC and FTIR and the results revealed that KTZ was molecularly dispersed in Labrafil® M2130 CS and that it is unlikely that KTZ would interact with the lipid. - Description: One of the major challenges of the oral administration of ketoconazole (KTZ), an inhibitor of sterol 14α demethylase, used in the management of systemic and topical mycoses in immuno-compromised and paediatric patients is the lack of availability of liquid dosage forms. In order to overcome this challenge, extemporaneous preparations have been manufactured by care-givers and health care providers by crushing or breaking solid oral dosage forms of KTZ and mixing with a vehicle to produce a liquid dosage form that can be swallowed by patients. However, the use of extemporaneous preparations may lead to under or over-dosing if the care-givers are not guided accordingly. Furthermore, the dearth of information on the stability of these KTZ-containing extemporaneous preparations may lead to ineffective antifungal therapy and complicate the problems of resistance as it is difficult to estimate the shelf-lives of these extemporaneous products under varying storage conditions due to the susceptibility of KTZ to chemical degradation. Therefore, there is a need for formulation scientists to develop novel drug delivery systems that avoid the need for extemporaneous preparations, possess well-established limits of stability and minimize the risks of systemic adverse effects to facilitate KTZ therapy. The use of solid lipid microparticles (SLM) as potential carriers for the oral administration of KTZ was investigated since solid lipid carriers are known to exhibit the advantages of traditional colloidal carriers. The research undertaken in these studies aimed to investigate the feasibility of developing and manufacturing solid lipid microparticles (SLM), using a simple micro-emulsion technique, as a carrier for KTZ. Prior to pre-formulation, formulation development and optimization studies of KTZ-loaded SLM, it was necessary to develop and validate an analytical method for the in vitro quantitation and characterization of KTZ in aqueous dispersions of SLM during development and assessment studies. An accurate, precise, specific and sensitive reversed-phase high performance liquid chromatographic (RP-HPLC) method coupled with UV detection at 206 nm was developed, optimized and validated for the analysis of KTZ in formulations. Formulation development studies were preceded by solubility studies of KTZ in different lipids. Labrafil® M2130 CS was found to exhibit the best solubilising potential for KTZ. Pre-formulation studies were also designed to determine the polymorphic behavior and the crystallinity of KTZ and Labrafil® M2130 CS that was used for subsequent manufacture of the solid lipid carriers. DSC and FTIR studies revealed that there were no changes in the crystallinity of KTZ or Labrafil® M2130 CS following exposure to a temperature of 60°C for 1 hour. It examines how the creative process of desire is forestalled (reified) by habitual representations of whiteness as a privileged position, and proposes that performance can be a valid form of resistance to static conceptions of race and subjectivity. The discussion takes into account how the privilege of whiteness finds representation through forms of neo-liberalism and neo-colonialism in the post apartheid context. The analysis focuses on the work of white South African artists whose work offers a critique from within the privileged “centre” of whiteness. The research is situated within the inter-disciplinary field of performance studies entailing a reading and application of critical texts to the analysis. Alongside this qualitative methodology surfaces a subjective dialogue with the information presented on whiteness. Part Two includes an analysis of Steven Cohen’s The Cradle of Humankind (2011), Brett Bailey’s Exhibit A (2011) and Michael MacGarry’s LHR-JNB (2010). Each section examines the way in which the respective works engage in a questioning of whiteness through performance. Part Three investigates South African rap-rave duo, Die Antwoord and how their appropriation of Zef interrogates desires for an essential authenticity. Part Four focuses on my own performance practice and the proposed value of engaging with a form of practice-led research. This is particularly relevant in relation to critical race studies that require a level of self-reflexivity from the researcher. It presents an analysis of the work entitled Villain (2012) as a disturbance of theatrical desire through a process of ‘becoming’. This notion of meaning and identity as ‘becoming’ is argued as a strategy to challenge prevailing modes of perception which can possibly restore the production of desire to the viewer. The thesis concludes with the notion that performance can offer a mode of immanent ethics which is significant in creating both vulnerable and critical forms of whiteness.

A comparative polarimetric study of the 43 GHz and 86 GHz SiO masers toward the supergiant star VY CMa

- Description: The aim of this thesis is to perform observational tests of SiO maser polarisation and excitation models, using component-level comparisons of multiple SiO maser transitions in the 43 GHz and 86 GHz bands at milliarcsecond resolution. These observations reqwre very long baseline interferometric imaging with very accurate polarimetric calibration. The supergiant star VY CMa was chosen as the object of this study due to its high SiO maser luminosity, many detected SiO maser lines, and intrinsic scientific interest. Two epochs of full-polarisation VLBA observations of VY CMa were performed. The Epoch 2 observations were reduced using several new data reduction methods developed as part of this work, and designed specifically to improve the accuracy of circular polarisation calibration of spectral-line VLBI observations at millimetre wavelengths. The accuracy is estimated to be better than 1% using these methods. The Epoch 2 images show a concentration of v= l and v=2 J= 1-0 SiO masers to the east and northeast of the assumed stellar position. The v=l J=2-1 masers were more evenly distributed around the star, with a notable lack of emission in the northeast. There is appreciable spatial overlap between these three lines. The nature of the overlap is generally consistent with the predictions of hydrodynamical circumstellar SiO maser simulations. Where the v=l J = 1-0 and J =2-1 features overlap, the v=l J = 2-1 emission is usually considerably weaker. This is not predicted by current hydrodynamical models, but can be explained in the context of collisional pumping in a low density environment. Six observational tests of weak-splitting maser polarisation models were performed, including intercomparisons of linear polarisation in the v=l J=1-0 and J=2-1lines, linear polarisation versus saturation level, linear polarisation versus distance from the star, circular polarisation in the v= l J = 1-0 and J=2-1 lines, circular versus linear polarisation and modeling of ~ 900 electric-vector position angle rotations. The polarisation model tests generally do not support non-Zeeman circular polarisation mechanisms. For the linear polarisation tests, the results are more consistent with models that predict similar linear polarisation across transitions. The scientific importance of these tests is described in detail and avenues for future work are described.

However, evaluations in South Africa such as the Annual National Assessments (ANAs) and the Progress in International Reading Literacy Study (PIRLS) (2006) show that reading achievement in the Foundation Phase is low, especially for children learning with African languages as their home language. This thesis examines pacing and sequencing in three Grade 1 literacy classes where isiXhosa is the language of learning and teaching (LOLT). The research took the form of a case study, focusing on the teachers’ classroom practice and how they understand their practice. Documents related to reading development in the Foundation Phase are analysed with regard to pacing and sequencing, examining how the curriculum frames literacy for teachers in the Foundation Phase. The teachers’ understanding in relation to their social context is also an important part of this research. In the thesis the way in which the teachers sequence and pace learning is analysed, drawing on Bernstein’s notions of internal and external framing, and Bourdieu’s notion of the habitus. The intended curriculum and the implemented curriculum are then compared. The results confirm that in low socio‐economic status (SES) schools, the level of poverty has an impact on teaching practice. The results also show that pacing and sequencing are reliant on the degree of planning amongst teachers. The level of development amongst learners also plays a role in how teachers organise their practice in order to differentiate the teaching of reading for all learners. The understanding that teachers have about their teaching practice are layered and influenced by their experiences, history and memories as teachers. The thesis concludes that there is a need to develop teachers’ understanding of planning and organising the teaching of reading, especially when the learners are from poor communities and do not get support in the home. In order for teachers to improve their practice, it is important for them to have the opportunity to reflect on and understand their practice.

- Description: The combined potential of two crucial factors in 1993, which afforded the promotion of socially just and ecologically sustainable ways of living, led to the instigation of this research project. The first was the imminence og our first democratic election; the second was the possible introduction of environmental education into formal education. In the light of these momentous shifts, it became apparent to me that teachers would have to radically transform their practices in order to play their part in transforming society from the dark days of apartheid into one of equity and harmony. The implications of these factors precipitated the falling into place of the rationale for my research: teachers had to look for professional development experiences which could facilitate the creation of alternative ways of thinking and doing. As a result, I approached a group of biology and geography teachers on the Cape Flats and, after protracted discussions, we decided to examine the potential of the urban environment for the teaching of ecology from the perspective of socially just and environmentally sustainable living. Out of this decision was born this study, which aimed at examining whether this process could, as a means to professional development, be a 'moment' in our journey to becoming transformative intellectuals. From this aim, the central research question emerged: Can emancipatory action research play a role in empowering teachers to become transformative intellectuals? The study consisted of five stages: - exploring the problem by reviewing the literature on the research problem; - the semi-structured interviews; - five workshops; - the 'sensing the urban environment' fieldtrip; and, - the various evaluation sessions. What we achieved during this research project, firstly, was a better understanding of our practices, which led us to seeing our roles as teachers differently and altered our pedagogical approaches. Secondly, this process developed the belief within ourselves that we, as teachers, can and should make a difference to the educational world in which we live. Lastly, this process laid the foundation for continued collaborative action by the participants. This process taught us that educational transformation is difficult and painful, and that present educational structures are not conducive to change. Nonetheless, in the historical context of this research, emancipatory action research was successful in giving us a consciousness-raising experience and closed the rhetoric-reality gap as we engaged in praxis (the practitioners developing and implementing their own curriculum). If we, as transformative intellectuals, are to engage in intellectual labour in the future, we are not only going to need to change our way of thinking and doing but will have to create an enabling infrastructure to realise this as well. We will, in addition, have to change the structures of the institutions in which we work in order to practise as transformative intellectuals.

Nonetheless, in the historical context of this research, emancipatory action research was successful in giving us a consciousness-raising experience and closed the rhetoric-reality gap as we engaged in praxis (the practitioners developing and implementing their own curriculum). If we, as transformative intellectuals, are to engage in intellectual labour in the future, we are not only going to need to change our way of thinking and doing but will have to create an enabling infrastructure to realise this as well. We will, in addition, have to change the structures of the institutions in which we work in order to practise as transformative intellectuals.

The investigation constituted a critical evaluation of current ESL TOL research, which was essentially a theoretical examination of emerging problems derived from an in-depth assessment of current ESL TOL studies. The theoretical examination was supplemented by the practical implementation of the technique in a case study research, utilising three verbal protocols obtained from ESL students at the University of Fort Hare. The close observation afforded by the evaluative case study research paradigm provided the writer - as participant observer - with a further means of judging the merit of TOLs, which corroborated findings from the theoretical evaluation, and enabled a consideration of unanticipated issues which emerged from the practical implementation of the technique. The conclusion was that TOLs seem to have a unique ability to identify aspects of the nature of ESL reading gained from an on-line assessment of reader interaction, provided that TOL research is conducted within certain methodological and analytic research constraints. The writer has proffered suggestions for future ESL TOL research, and feels that the combination of TOL research findings with other measures of reading comprehension could elucidate aspects of ESL comprehension, making a valuable contribution to ESL reading theory and practice.

The emergence of various schemes for the preparation of South African primary teachers is outlined. The Cape Province provides the major exemplar. The nature of educational theory, and its precise contribution to enhanced teaching ability has been a subject of fierce debate within educational circles. An analysis of theory, in general, and as it applies in the educational context, is undertaken. The evolution of the core theoretical disciplines of education - history, philosophy, psychology and sociology of education - is traced. The current status of these disciplines at selected South African institutions undertaking primary teacher preparation is then investigated, and the views of lecturing staff at these institutions on the optimal approach to the teaching of educational theory are presented. The disciplines still play an important role in the South African Bachelor of Primary Education degree courses surveyed. In British teacher preparation courses, however, disenchantment with the disciplines' format has given rise to demands for new approaches to the teaching of educational theory. Some of these alternative approaches are evaluated. The most important changes to content in the Senior Secondary Mathematics Syllabus (July 1984) are the introduction of calculus and linear programming, the substitution of a section on analytical geometry for vector algebra and the recall of the remainder and factor theorems. The way in which these changes were introduced left the task of integrating them into the teaching process in the hands of individual teachers. This is a task of extreme importance. If one's classroom practice is to simply plough one's way through the syllabus, one loses many opportunities to make the study of mathematics meaningful and worthwhile. Accepting the view of the spiral nature of the curriculum where one returns to concepts and procedures at increasing levels of sophistication, one needs to identify the position of topics in this spiral and to trace their conceptual foundations. Analytical geometry is in particular need of this treatment. Similarly there are many opportunities for preparing for the introduction of calculus. If the teaching of calculus is left until the last moments of the Standard 10 year without proper groundwork, the pupil will be left with little time to develop an understanding of the concepts involved. It is the advent of calculators which presents the greatest challenge to mathematics education. We ignore this challenge to the detriment of our teaching. Taken seriously calculators have the potential to exert a radical influence on the content of curricula and examinations. They bring into question the time we spend on teaching arithmetic algorithms and the priority given to algebraic manipulation. Numercial methods gain new prominence. Calculators can even breathe new life into the existing curriculum. Their computing power can be harnessed not only to carry out specific calculations but also to introduce new topics and for concept reinforcement. The purpose of this study has been to bring about a proper integration of the new sections into the existing syllabus and to give some instances of how the calculator can become an integral part of the teaching/learning process
