- Description: The thesis presents a qualitative survey of web services' interoperability, offering a snapshot of development and trends at the end of 2005. It starts by examining the beginnings of web services in earlier distributed computing and middleware technologies, determining the distance from these approaches evident in current web-services architectures. It establishes a working definition of web services, examining the protocols that now seek to define it and the extent to which they contribute to its most crucial feature, interoperability. The thesis then considers the REST approach to web services as being in a class of its own, concluding that this approach to interoperable distributed computing is not only the simplest but also the most interoperable. It looks briefly at interoperability issues raised by technologies in the wider arena of Service Oriented Architecture. The chapter on protocols is complemented by a chapter that validates the qualitative findings by examining web services in practice. These have been implemented by a variety of toolkits and on different platforms. Included in the study is a preliminary examination of JAX-WS, the replacement for JAX-RPC, which is still under development. Although the main language of implementation is Java, the study includes services in C# and PHP and one implementation of a client using a Firefox extension. The study concludes that different forms of web service may co-exist with earlier middleware technologies. While remaining aware that there are still pitfalls that might yet derail the movement towards greater interoperability, the conclusion sounds an optimistic note that recent cooperation between different vendors may yet result in a solution that achieves interoperability through core web-service standards.

In vitro release of ketoprofen from proprietary and extemporaneously manufactured gels

- Description: Ketoprofen is a potent non-steroidal anti-inflammatory drug which is used for the treatment of rheumatoid arthritis. The oral administration of ketoprofen can cause gastric irritation and adverse renal effects. Transdermal delivery of the drug can bypass gastrointestinal disturbances and provide relatively consistent drug concentrations at the site of administration. The release of ketoprofen from proprietary gel products from three different countries was evaluated by comparing the in vitro release profiles. Twenty extemporaneously prepared ketoprofen gel formulations using Carbopol® polymers were manufactured. The effect of polymer, drug concentration, pH and solvent systems on the in vitro release of ketoprofen from these formulations were investigated. The gels were evaluated for drug content and pH. The release of the drug from all the formulations obeyed the Higuchi principle. Two static FDA approved diffusion cells, namely the modified Franz diffusion cell and the European Pharmacopoeia diffusion cell, were compared by measuring the in vitro release rate of ketoprofen from all the gel formulations through a synthetic silicone membrane. High-performance liquid chromatography and ultraviolet spectrophotometric analytical techniques were both used for the analysis of ketoprofen. The validated methods were employed for the determination of ketoprofen in the sample solutions taken from the receptor fluid. Two of the three proprietary products registered under the same manufacturing license exhibited similar results whereas the third product differed significantly. Among the variables investigated, the vehicle pH and solvent composition were found have the most significant effect on the in vitro release of ketoprofen from Carbopol® polymers. The different grades of Carbopol® polymers showed statistically significantly different release kinetics with respect to lag time. When evaluating the proprietary products, both the modified Franz diffusion cell and the European Pharmacopoeia diffusion cell were deemed adequate although higher profiles were generally obtained from the European Pharmacopoeia diffusion cells. Smoother diffusion profiles were obtained from samples analysed by high-performance liquid chromatography than by ultraviolet spectrophotometry in both diffusion cells. Sample solutions taken from Franz diffusion cells and analysed by ultraviolet spectrophotometry also produced smooth diffusion profiles. Erratic and higher diffusion profiles were observed with samples taken from the European Pharmacopoeia diffusion cell and analysed by ultraviolet spectrophotometry. The choice of diffusion cells and analytical procedure in product development must be weighed against the relatively poor reproducibility as observed with the European Pharmacopoeia diffusion cell.

Thermophilic lignin degrading enzymes from actinomycetes for biotechnological applications

- Description: Phenolic residues which accumulate in the environment as a result of agro-industrial practices has resulted in the need to find and use Eco-Friendly techniques, rather than the traditional methods of burning or burying this kind of waste. Bioremediation and bioconversion are attractive alternatives using whole cell or enzyme-based systems. The aims of this project were to isolate and uses thermophilic Actinomycetes, which produce thermo-tolerant oxidoreductase enzymes, which can be used to bioconvert a model industrial phenolic waste commonly genersated in the wine-making industry of South Africa. Current research in bioconversion and bioremediation focuses on mesophilic microbes in that their enzymes can catalyse reactions at higher temperatures without affecting its activity and lower contamination levels. Three novel Actinomycete isolates were isolated (RU-A0l , RU-A03 and RU-A06) from a compost site and characterized using a combination of conventional identification techniques and 16S rDNA methodology to identity the three isolates. All three isolates belong to the Streptomyces clade. In addition, five known Actinomycetes were selected from an internation culture collection and also screened for oxidoreductase activity in comparision to the three novel isolates. Although the five isolates were selected based on their ability to produce oxidoreductase enzymes, unexpectedly, no activity was detected. Screening assays for peroxidase, polyphenol oxidase and laccase on RU-AO 1, RU-A03 and RU-A06, showed that all three isolated produced peroxidases and peroxidases but no laccase. Substrate specificity studies revealed that the most suitable substrates to determine peroxidase and polyphenol oxidase activity on these isolates were catechol for polyphenol oxidase, 2,4-dichlorophenol for peroxidases and veratryl alcohol for lignin peroxidases. Previous studies have indicated that peroxidases and polyphenol oxidases are produced in Actinomycetes during the primary stage of growth. This was the case with RU-AOI , RU-A03 and RU-A06. Growth rates were higher that other Actinomycetes, with maxImum biomass being reached at 36 hours for the isolates RU-AOI and RU-A06 and 48 hours for isolate RUA03. pH studies showed that the three isolates were adaptable and could grow over a broad pH range. Catabolism studies of phenolic model compounds showed that the three isolates were capable of catabolizing the model phenolic compounds within a period of 24 hours. Further studies were carried out to determine the effect of these microbes and their enzymes in whole cell and enzyme-based systems on a model phenolic waste, graoe waste consisting of compressed grape skins, pips and stalks. Whole cell studies showed that the isolates were capable of bioconverting the waste at a maximum concentration of 30% grape waste (vol:vol). Peroxidase and polyphenol oxidase activity increased indicating induction of these enzymes in the presence of phenolic compounds, with a maximum increase of up to 15.9 fold increase in extracellular lignin peroxidase activity in RU-AO1. HPLC and phenolic determination assays indicated that bioconversion of the phenolic grape waste had occurred in the presence of the three isolates. Attempts were made to isolate and identify a peroxidase or phenol oxidase gene from one the isolates. As bacteria, Actinomycetes are amendable to gene manipulation making them suitable candidates for methods such as site directed evolution in comparison to fungi. Two clones were selected for sequencing based on positive activity results when assayed for peroxidase activity. However the resultant sequences did not identify a functional gene sequence. Southern Blotting was then carried out to determine the nature of the peroxidase gene. Previous studies have been focused on the catalase-peroxidase gene (CalC gene) found Actinomycetes and other bacteria. A probe was developed from the CalC gene. No hybridization occurred with any of the enzyme restricted DNA from the three isolates. The implications of these results are that the peroxidase genets in the three isolates are in fact lignin peroxidase in nature. This project has the potential in the bioconversion of phenolic wastes and is the first description of the use of thermophilic Actinomycetes in the bioconversion of an industrial phenolic waste.

The use of indigenous macroinvertebrates and Daphnia pulex in acute toxicity testing

- Description: Aquatic toxicology has been identified as a valuable tool in the identification and management of chemical pollution in aquatic ecosystems. Standardised methodologies for acute aquatic bioassays have been adopted from international agencies. As a result of these standard methods, the use of laboratory cultured organisms for toxicity testing has been more popular than that of indigenous field-caught organisms. Included in these adopted methods are those for the cultured crustacean Daphnia pUlex. D.pulex is adapted to living in standing water and the suitability of this species to determine toxic effects for South African riverine environments, which are largely flowing, has been questioned. Thus this thesis is a case-study ofthe use of D.pulex and indigenous site-specific macroinvertebrates as toxicity test organisms for setting acute water quality guidelines to protect aquatic ecosystems. The study highlights site-specific problems such as reference sites and organism identification. The acute tolerance of selected indigenous invertebrates was compared to that of D. pulex, using both a single-substance reference toxicant (zinc) and selected whole efiluents. The significance of source population and culture age as a potential source of biological variability between D.pulex cultures was also investigated. D.pulex cultures have been initiated in South Africa from females collected from a number of different local populations; also it is assumed that no genetic change (due to mutation) occurs within a D.pulex culture over time. In order to establish if source population and culture age are a source of biological variability between D.pulex experiments, the acute tolerJuce to zinc of two different D.pulex populations and three different generations within a population were compared. Due to experimental variability results were inconclusive, and differences in tolerance as a result of population difference or culture age could not be determined with confidence. The acute tolerance of D.pulex to a single reference chemical (zinc) and selected whole efiluents was compared to that of selected indigenous invertebrates. Acute 48 h D.pulex zinc tolerance (LC50 range: 0.22 - 0.60 mg/l Zn) was found to be more sensitive than acute 96 h tolerances shown by mayfly species A.fconurus peringueyi (Heptageniidae) (LC50: 17.42 mg/l Zn), Euthrauluselegans (Leptophlebiidae) (LC50: 0.98 mg/IZn), Ba~tidae (LC50: 0.94mg/IZn) and shrimp, Caradina nilotica (Atyidae) (LC50: 3.17 mg/l Zn). This result suggests that guidelines for zinc set using D.pulex will protect the selected indigenous invertebrates. Selected whole eftluents were not acutely toxic to either D.pulex or selected indigenous invertebrates. These experiments were used as a case study for method development regarding the comparative use of D.pulex and indigenous invertebrates in acute whole eftluent toxicity testing. Finally, it is recommended that a suite of indigenous organisms (e.g. macroinvertebrates, fish and algae), as well as laboratory cultured D.pulex, be used in the initial setting of guidelines and that D.pulex be used for routine compliance monitoring. It is futher recommended that a suite of available monitoring methods, such as chemical and biomonitoring methodologies, be used in conjuction with toxicity testing in water quality management.
