  • Description: Over the last few years, there has been a notable increase in popularity in the use of cannabidiol (CBD) as a form of alternative medicinal treatment for various illnesses. CBD, a by-product of the cannabis plant, is an isolate and does not contain the psychoactive agent, tetrahydrocannabinol (THC). Endometriosis and Polycystic Ovarian Syndrome (PCOS) are chronic reproductive health sicknesses that are increasingly experienced by women. In the absence of cures, biomedical treatment for these diseases aim to manage symptoms, for example; heavy bleeding, heightened levels of pain, and insomnia. CBD offers an alternative to women who feel that biomedical interventions are no longer able to maintain their health and well-being. CBD positions itself as a natural remedy claiming to be safe and effective. This research study, mainly through qualitative data collection, focused on experiences of Zimbabwean and South African women living with endometriosis and/ or PCOS, who have turned to CBD to manage their symptoms. The importance of this study was to position itself within patients’ lived experiences. The research study found that CBD indeed has numerous benefits, including pain management, alleviating stress, and anxiety. Through the emergent themes from the data, it became clear that women are marginalised and treated unequally in the biomedical healthcare sphere. An exploratory study of King IV™: The effect of Principle Four and its practices in achieving good performance by selected JSE-listed companies

  • Description: The research conducted for the dissertation entitled “An exploratory study of King IVTM: The effect of principle four and its practices in achieving good performance by selected JSE-listed companies”, aimed to determine whether principle four, namely the creation of value, and the application by an organisation of the recommended practices as set out in King IVTM, contribute to the achievement of good performance. The research outlined the background to the research by describing the history of the development of corporate governance internationally and in South Africa, dating back to 1932 and culminating with the King IV™ report. It described the problem statement, defined the aims and objectives of the study, and demonstrated that the significance of the study lies in its exploration of the relationship between the King IV™ practices and good performance. The research also provided a review of the related literature, describing the theoretical background for the research, which includes the conceptual framework and propositions of the research. The research was situated in the post-positivist paradigm, the methodology was qualitative, and the method adopted was a theory-based evaluation, based on a deductive thematic analysis with pattern matching. From a detailed reading of the integrated reports and the King IVTM compliance registers of the fifteen selected listed companies for the years 2017 to 2019, hits and misses in relation to the six capitals and the eight associated practices were recorded. From this, the resulting descriptive statistics were calculated and set out in the form of tables, and a regression analysis was used to support the findings. The research revealed, based on the descriptive statistics data that there is a moderate relationship between good performance and the application of the eight practices set out in King IVTM, that contribute to the achievement of good performance. It was, therefore, concluded that the adoption of corporate governance principles and practices has an effect on performance, although it may not be significant. It is also concluded that the application of the practices in terms of principle four as set out in King IVTM is achieving its intended outcome - the creation of value. The research also concluded that good performance constitutes more than just a company performing financially and that all companies need to comply with the requirements of King IVTM, as the Code is designed to guide organisations in achieving good corporate governance. Governance principles and practices should be adopted in a way that does not unduly constrain them and is appropriate to a company’s particular circumstances. Complying with corporate governance principles and implementing the recommended practices might not guarantee positive outcomes (good performance), but it will guide the board of directors in the pursuit of ethical and effective leadership, and that of sustainable development in order to meet the needs of all stakeholders. , Thesis (MBA) -- Faculty of Commerce, Rhodes Business School, 2022

  • Description: The purpose of this study was to investigate the impact of three critical incidents with a direct effect on Small and Medium Enterprises (SMEs) within the Petroleum retail sector in South Africa. This study assessed three specific areas: (1) the impact of fuel levy increase on Petroleum retailers, (2) explore how fuel stock-outs are experienced by the SMEs, and (3) the effects of fuel distributors' strike on SMEs in the petroleum sector in South Africa. To address the research objectives, the study employed a qualitative research approach, based on a narrative research design. Primary data was collected through semi-structured interviews conducted with six SMEs in the fuel retail business in Johannesburg. An interview guide was used. Findings highlighted that the location of fuel retail SMEs is an important determinant of how the fuel retailers are affected by these three critical incidents of fuel levy increases, stock-outs and strikes. The study found that while the fuel levy increases are felt through reduced sales volumes, the impact is not the same across fuel retailers. The study revealed that the elasticity of demand varies depending on the location for which it is elastics for those retailers in high Living Standards Measure (LSM) locations and inelastic for those located in lower LSM locations. It was found that the impact is much less for those fuel retailers located in affluent suburbs with a high LSM. In locations such as Beyers Naude and Radiokop in Northern Johannesburg with a high LSM, the demand for fuel remains constant irrespective of price increases. The locations are also less likely to be adversely affected by protest actions. Concerning SMEs’ experiences of fuel stock-outs, the study found that stock-outs impact fuel retail business operations through overhead costs that are incurred while the fuel stations are out of stock. The small margins make it impossible for fuel retailers to quickly recover, as their profitability is dependent on sales volumes, which are compromised when there are stock-outs. Fuel retailers also suffer from reduced future sales volumes due to waning customer loyalty. Finally, the study found that the occurrence of wage-related strikes is minimal due to the nature of collective agreements that are signed within the sector.   • Description: Malaria is caused by Plasmodium parasites, spread to people through the bites of infected female Anopheles mosquitoes. This study focuses on all 5 (Plasmodium falciparum, Plasmodium knowlesi, Plasmodium malariae, Plasmodium ovale and Plasmodium vivax) parasites that cause malaria in humans. Africa is a developing continent, and it is the most affected with an estimation of 90% of more than 400 000 malaria-related deaths reported by the World Health Organization (WHO) report in 2020, in which 61% of that number are children under the ages of five. Malaria resistance was initially observed in early 1986 and with the progression of time anti-malarial drug resistance has only increased. As a result, there is a need to study the malarial proteins mechanism of action and identify alternative treatment strategies for this disease. Type II NADH: quinone oxidoreductase (NDH2) is a monotopic protein that catalyses the electron transfer from NADH to quinone via FAD without a proton-pumping activity, and functions as an initial enzyme, either in addition to or as an alternative to proton-pumping NADH dehydrogenase (complex I) in the respiratory chain of bacteria, archaea, and fungal and plant mitochondrial. The structures for the Plasmodium knowlesi, Plasmodium malariae, Plasmodium ovale and Plasmodium vivax were modelled from the crystal structure of Plasmodium falciparum (5JWA). Compounds from the South African natural compounds database (SANCDB) were docked against both the NDH2 crystal structure and modelled structures. By performing in silico screening the study aimed to find potential compounds that might interrupt the electron transfer to quinone therefore disturbing the enzyme‟s function and thereby possibly eliminating the plasmodium parasite. CHARMM-GUI was used to create the membrane (since this work is with membrane-bound proteins) and to orient the protein on the membrane using OPM server guidelines, the interface produced GROMACS topology files that were used in molecular dynamics simulations. Molecular dynamics simulations were performed in the Centre for high performance computing (CHPC) cluster under the CHEM0802 project and the trajectories produced were further analysed. In this work not only were hit compounds from SANCDB identified, but also differences in behaviour across species and in the presence or absence of the membrane were described.   • Description: In contemporary South Africa, a country in transition, destruction rather than reconstruction seems commonplace. Electricity supply is at an all-time low and ‘load shedding’ is an almost daily occurrence. Similarly, more fragility of water delivery and sanitation service is being reported with the likelihood of ‘water shedding’ a soon-to-be reality. In view of the ever-increasing reported mismanagement of South Africa’s water and sanitation infrastructure, which is likely nearing the point of collapse if not already collapsed, this thesis set out to interrogate at a provincial and municipal level the status of water and sanitation in Chris Hani District Municipality (CHDM), Eastern Cape Province. One major driver behind this study was to evaluate provincial and municipal water/wastewater infrastructure as a platform to support implementation of locally developed bioprocess technologies. First, publicly available historical data was used to derive a baseline or benchmark from which to determine gains/losses in compliance, water quality and efficiency. The period 2009-2013/4 during which Blue and Green Drop reporting was routine, was used as an appropriate start point. Data thus derived indicated that, at provincial and district municipality (DM) level, potable water supply was by bulk water schemes and, sanitation was typically by waste stabilization pond (WSP) treatment of municipal sewage. The derived benchmark for the period 2009-2013/14 indicated that most plants were not compliant (~75% of the Eastern Cape Province water treatment plants (WTP) operating between average performance to critical state), operated either at- or above design capacity (Eastern Cape Province, 16%; CHDM, 19%). A large number of plants for the province (62.4%) and district (81.25%) had hydraulic design capacity or average daily flows that were unknown (information not provided in available literature or plant reports) and final effluent did not always meet the general standard (70.3% of the CHDM wastewater treatment works (WWTW)). To determine the contemporary state (2020 - 2022) of water and sanitation within CHDM, a scoping exercise of WTP and WWTW in selected local municipalities was carried out. This was done along a west-east transect by appraising WTP and WWTW in the towns of Cradock, Tarkastad, Komani, Tsomo, Cacadu, and Ngcobo. Furthermore, targeted assessment of the Queenstown WTP and the Cradock WWTW was used to derive real-time data on the status of water and sanitation infrastructure. Results showed that water and sanitation services within CHDM had, in the years between 2009-2013/14 to date, deteriorated. Freshwater demand significantly exceeded capability of water supply schemes, where demand was as much as three times greater than reported available supply. For most WWTW, operation was in excess of capacity and between 1.52 and 12 times installed hydraulic loading. The targeted scoping exercise revealed that Queenstown WTP is a moderate risk plant (Cumulative risk rating, CRR = 11 and maximum risk rating, MRR = 47.8%), whereas Cradock WWTW was in working condition but with challenges and some infrastructural dysfunction. Assessment of water/effluent quality revealed that turbidity and TOC were above SANS 241 general limit. Non-compliance in terms of nitrate/nitrite-N, ammonium-N, phosphate-P, TSS, total coliforms, E. Coli, and free chlorine was evident at Cradock WWTW. Unstructured interviews with plant operators corroborated these findings. The overwhelming support for bulk schemes for potable water provision and WSP for sanitation indicated a partially water secure municipal district but derelict in terms of its sanitation services. The later, it was concluded, in particular provides an ideal opportunity for implementation of platform technologies to support bioprocesses for entrepreneurship, employment, economic benefit and to secure a closed circular economy for regional water and sanitation through valorisation of co-product streams.   • Description: The purpose of this study is to investigate and understand how specialists who are newly appointed team leaders in an automotive manufacturing organisation experience the transition from specialists to team leaders. The role of leading others in an organisation is linked to human resource leadership. The human resource is highlighted as a valuable resource as it is the only thinking resource in an organisation. It also influences cost and quality and it is central to the organisation's competitive advantage. Consequently, any interruptions or lack of effective management of the human resource will result in a loss of value that the human resource brings into the organisation since they maximise the use of all other resources. The role transitioning process takes place in three phases. These are the separation, transitioning and incorporation phases. In the separation phase, the transitioning individuals have difficulty detaching from their previous role; the individuals still having the urge to do the work for their subordinates instead of delegating illustrate this. In the transition phase, the individuals in the study experienced the anxiety of wanting to make the new role a success whilst experiencing a sense of lack of belonging due to not being in the familiar old role and not entrenched in the new role. In the incorporation phase, the transitioning individuals experience a sense of stability where they are comfortable acting in the new role. Problem: This study aims to answer the question of how newly appointed team leaders in an automotive manufacturing organisation experience role transition from specialists to team leaders Method: The study will be employing the qualitative thematic deductive method. The target population consists of team leaders who have transitioned into the team leader position in the past two years in different departments. The data was collected by way of face-to-face semistructured interviews with a sample size of seven participants. Data analysis was conducted with a method that is consistent with thematic analysis. 3 Key Findings: Individuals transitioning have urgencies to perform their previous roles in the early phase; during the transition phase, they have anxiety because they are not yet successful in the new role, and in the final phase, they have become comfortable with their new roles and their new identities. Implications: The study's outcome is to identify insights and resultant recommendations for non-leading specialists transitioning into roles where they have to lead others.   • Description: The study critically examines and discusses language choice on twitter, a social media platform that has a vast number of people from different language groups. This diverse platform allows people to tweet, communicate, and share information online. There are tweets that trend and attract more engagement across different language groups. O jewa ke eng? which translates to ‘what’s bothering you?’ is one of those tweets, and it is communicated in Sesotho. (More discussion about the tweet in the introduction section that follows). According to Eisenstein, O’ Connor, Smith & Xing (2010) & Eisenstein (2016), “the popularity and ubiquity of social media allows access to a wide variety of spontaneous language enabling researchers to study language variation across space and time at large scale”, (Preotiuc-Pietro & Ungar, 2018: 1534). The introduction to the internet and socially mediated platforms such as Twitter and others has changed how information is produced and consumed. Social media allow people to communicate and engage in languages they are comfortable and familiar with. The study discusses language choice at three levels: language choice between isiXhosa verses English; isiXhosa verses other African languages and isiXhosa verses other varieties. The study is pursued with the overarching aim to investigate the possibility of African languages such as isiXhosa being used as dominant languages on social media. Thus, the study explores language use by isiXhosa speaking users on twitter that engaged with the tweet. Moreover, the study investigated why they choose to use one language over the other or multiple languages. To give a broader perspective on the study, language choice, language shift and language attitude are discussed and critically evaluated. Theoretically, the concept of code-switching and code-mixing is used when looking at bilingualism and multilingualism. It is imperative to look at multilingualism because the prevalent use of English is no longer a barrier to communication, social media platforms have created a space where African language users are able to engage in their native languages. Moreover, the study explores the use of standard and non-standard isiXhosa languages. There are varieties of languages such as tsotsitaal, slang and others, and these language varieties are not recognised as standard, used in educational settings and so on. In doing so, language attitude is used as the concept to investigate what propels isiXhosa users to or not to use these language varieties in the online communities. This is an important part of the mathematics curriculum as it is the beginning of statistical literacy learning. We need to be able to question, evaluate claims based on data, create arguments we can defend and use data meaningfully, it is thus crucial that learners acquire statistical literacy. This research sets out to examine the text in the data handling sections of four Intermediate Phase Mathematics book series in order to answer the following research question: What are the language comprehension demands of English mathematical texts on data handling that are used in South African Intermediate Phase Mathematics? The theories framing the study are Vygotsky’s sociocultural theory and Cummins’ second language acquisition theory. It is an interpretivist mixed method case study that takes the form of a document analysis.. The findings indicate that many units in the books analysed have a higher readability level than the grade level and will thus present a challenge to learners in terms of their ability to access the mathematical content. An analysis of the linguistic complexity revealed that the features contributing most to the complexity of the texts included words with seven or more letters, prepositional phrases, infinitives, complex verbs and complex/compound sentences. An examination of the non-textual elements revealed that most of them are accurate, connected, concise, contextual and these add to the comprehensibility although there were a few which could be possibly be distractors. In this study, systems theory was adapted for analysing the sustainability focus of the Faculty of Economics and Information Technology Systems (FEITS) at Walter Sisulu University. A qualitative research method was used during this research, and data collected from a purposive sample of staff, activities and documentation. In this study, Von Bertalanffy’s systems theory is applied to analyse the sustainability foci and practices of the FEITS. The study describes sustainability practices and concerns that emerge from research activities, community engagement activities and work-integrated learning. The theory is also used to analyse the connection between various subsystems of the faculty when addressing emerging local sustainability concerns. Education for sustainable development is an important aspect of this study as it considers the sustainability concerns in the curriculum from the perspective of current and future alternatives for the economy and socio-ecological aspects of communities. The following findings have emerged from the study: Contextual realities related to inequality were foci of discussion and concern in various qualifications. Socioecological issues and risks were found to be related to water and sanitation, food security and climate change. Sustainability discourse is related to health and wellbeing, peace, and security. There is an alignment of green skills to the curriculum of the faculty and pedagogical practices which relate to sustainability. The faculty participates in stakeholder platforms to respond local sustainability challenges and is developing courses in response to local development needs. Assessment of cytotoxic artemisinin and its derivatives as DNA damaging inducing agents in triple-negative breast cancer cells

  • Description: In developing countries, including South Africa, breast cancer is the primary cause of cancer-related deaths among women. TNBC (triple-negative breast cancer) is an aggressive breast cancer subtype that is more prevalent in women of African descent. This subtype lacks the key receptors, namely the estrogen receptor (ER-), progesterone receptor (PR-), and human epidermal growth factor receptor 2 (HER2-) that are the basis of successful targeted therapies for other subtypes of the disease. To date, there are no effective, standardized targeted therapies for TNBC. Artemisinin is an anti-malarial drug and numerous derivatives of the compound have been developed to improve the potency and solubility of the parent compound. Artemisinin and its derivatives have gained attention as potential anti-cancer agents; however, such studies have not yet progressed to clinical trials and the precise mechanism of action of these compounds is yet to be fully explained. In this study, artemisinin, and its known derivative artesunate, as well as a novel derivative, WHN11, were investigated as DNA damage-inducing agents in TNBC. WHN11 was found to be the most potent of the three compounds, displaying an IC50 of 3.20 μM against HCC70 cells, artemisinin displayed an IC50 of 214.70 μM and artesunate displayed an IC50 of 25.48 μM. The compounds were less toxic to the MCF12A non-cancerous cells, with IC50 values 298.30, 87.53, and 8.35 μM for artemisinin, artesunate, and WHN11, respectively, and displayed selectivity indices of 1.39, 3.44 and 2.61 μM for artemisinin, artesunate, and WHN11, respectively. In silico and in vitro studies revealed that the artemisinin compounds bind to DNA through the minor groove. While all three compounds were able to bind to DNA, a comet assay revealed that only artemisinin and artesunate, and not WHN11, were able to cause DNA damage compared to the vehicle control, DMSO. Finally, a topoisomerase I (TOPO I) enzyme assay demonstrated that while the compounds appeared to display a degree of inhibition of TOPO I, as evidenced by a downward shift in the plasmid band on the agarose gel, they were not able to fully inhibit the enzyme to return the plasmid to the supercoiled conformation. In addition, combination studies revealed that artemisinin, artesunate, and WHN11 acted synergistically in combination with camptothecin, but displayed either an additive (artemisinin) or antagonistic (artesunate and WHN11) relationship when used in combination with etoposide. In conclusion, artemisinin, its known derivative artesunate, and novel and highly toxic derivative WHN11, all bind to DNA via the minor groove, however only artemisinin and artesunate, and not WHN11, cause DNA damage, indicating a potentially different mechanism of action of the three artemisinins. All three compounds act synergistically with camptothecin, which suggests interference with topoisomerase activity, partially supported by slight inhibition of TOPO I activity, and could indicate either direct inhibition of the enzyme or interference with enzyme function by competitive binding to the DNA.   • Description: The main objective of this study was to investigate the use of baseline assessments for reading in isiXhosa in Foundation Phase. It focused on how teachers conduct baseline assessments, the assessment tools they use, and their responsiveness to baseline assessments data in terms of curriculum planning and practice, given the lack of explicit guidelines and tools specifically developed for African languages. The lack of clarity may lead teachers to assume that the same assessment approaches and practices apply across all official languages despite their linguistic and sociolinguistic differences. Vygotsky’s socio-cultural theory of learning and Biggs’ concept on constructive alignment are used as the theoretical and conceptual framework for this study. The premise for this study was that it would be problematic to apply similar assessment approaches and use the same tools for all official South African languages. The socio-cultural approach emphasises the importance of language learning and learning generally according to the context within which language occurs. The main research sites are two Quintile 1 schools in a township in Makhanda District. These schools have been receiving external intervention which includes a baseline assessment for isiXhosa reading from Non-Governmental Organisations (NGOs). This study sought to provide some insights on whether previous assessments have informed subsequent teaching practices. The findings of the study revealed that the most prominent tool for assessing reading in isiXhosa is Early Grade Reading Assessment (EGRA) and the tools used by external literacy interventions in the two schools were adapted versions of EGRA. Additionally, the subtasks in the baseline assessment tools used were aligned to the linguistic features of isiXhosa besides the nonsense word component. Furthermore, teachers were assessing and teaching reading at lower levels than expected in the grades 2 and 3 in this study. This study recommends that teachers respond to the baseline assessment data appropriately to develop reading competencies.   • Description: The 21st century has seen a dramatic increase in chronic non-infectious diseases, especially in the area of mental health. Medical anthropologists have seen a rise in the development of mental illnesses in both developed and developing nations. There is, however, little research conducted on trauma narratives that do not stem from political violence from an anthropological lens. South Africa has various understandings of trauma depending on the cultural context and it is crucial to examine these narratives as this provides vital information of the daily lived experiences of trauma survivors. Key themes draw on issues of trauma denialism, communicating distress, traumatic symptoms and the development of mental illnesses as a result of traumatic exposure. The data was analysed through Goffman’s (1959) Presentation of Self in Everyday Life illustrating various ways how survivors present themselves depending on the particular audience. This research employed both qualitative and quantitative research methods to gather a holistic understanding of trauma survivors.   • Description: This thesis is divided into two main sections: Part 1 describes the design, and evaluation of the accuracy of a new web server – PRotein Interactive MOdeling (PRIMO-Complexes) for modeling protein complexes and biological assemblies. The second part describes the development of bioinformatics tools to predict HIV-1 drug resistance and support bioinformatics research and education. Recent technological advances have resulted in a tremendous increase in the number of sequences and protein structures deposited in the Universal Protein Resource Knowledgebase (UniProtKB) and the Protein Data Bank (PDB). However, the number of sequences has increased at a higher rate compared with the experimentally solved multimeric protein structures. This is partly due to advances in high-throughput sequencing technology. To fill this protein sequence-structure gap, computational approaches have been developed to predict protein structures from available sequences. Computational approaches include template-based and ab initio modeling with the former being the most reliable. Template-based modeling process can be achieved using either standalone software or automated modeling web servers. However, using standalone software requires familiarity with command-line interfaces as well as utilising other intermediate programs which could be daunting to novice users. To alleviate some of these problems, the modeling process has been automated, however, it still has numerous challenges. To date, only a few web servers that support multimeric protein modeling have been developed and even these provide little, if any user involvement in the process. To address some of these issues, a new web server – PRIMO-Complexes – was developed to model protein complexes and biological assemblies. The existing PRIMO web server could only model monomeric proteins. Part 1 of this thesis provides a detailed account of the development and evaluation of PRIMO-Complexes. The rationale for developing this new web server was based on the understanding that most proteins function as protein multimers and often the ligand-binding sites, and enzyme active sites are located at the protein-protein interfaces. It, therefore, necessitated developing capabilities for modeling multimeric proteins. PRIMO-Complexes web server was developed using the Waterfall system development life cycle model, is based on the Django web framework and makes use of high-performance computing resources to execute jobs. The accuracy of the algorithms embedded in PRIMO- Complexes was evaluated and the results were promising. Additionally, PRIMO-Complexes performs comparatively well in relation to other web servers that offer multimeric protein modeling. Another unique feature of PRIMO-Complexes is its interactivity. The webserver was developed with capabilities for allowing users to model multimeric proteins with an appreciable degree of control over the process. In the second part of the thesis several other bioinformatics tools are described, for example, a webserver for predicting HIV-1 drug resistance, the RUBi protein model repository, and a bioinformatics web portal for education and research resources. RUBi protein model repository stores verified theoretical models built using various modeling approaches. This enables users to easily access models to reproduce and/or further the research. This is described in chapter 5. Chapter 6 describes the design and development of the Human Immunodeficiency type 1 Resistance Predictor (HIV-1 ResPredictor), a web application that employs artificial neural networks (ANN) to predict drug resistance in patients infected with HIV-1 subtype B. The ANNs and subtype classifiers performed well making this web application potentially useful to both clinicians and researchers in this era of personalised medicine. Finally, chapter 7 describes a bioinformatics education web portal that equips students with information on how to use bioinformatics online resources. Being aware of these resources is not enough without a deeper understanding and guidance on how to apply bioinformatics methods to solve practical problems. This web portal was aimed at familiarising students with the basic terminology and approaches in structural bioinformatics.   • Description: Bitcoin is defined as a virtual cryptocurrency that solely exists in electronic form. Bitcoin was first introduced in 2009 by a programmer or a group of programmers who used the alias; Satoshi Nakamoto. Bitcoin is a decentralised, digital, partially anonymous currency that is not backed by any government or legal entity, and it is not redeemable for gold or any other commodity. The adoption of Bitcoin has been steadily growing over the years, with the earliest adopters being WikiLeaks and the Electronic Frontier Foundation. Ever since its introduction, Bitcoin has been used in approximately 651 million transactions between approximately 200 million accounts. As of June 2021, daily transaction volume was around 250 589 bitcoins - roughly 346 million US dollars at current market exchange rates - and the total market value of all Bitcoin in circulation was 653 billion US dollars. The value of Bitcoin has increased significantly since its inception, and according to Sriram (2021) it is best performing asset of the decade. This prompted the present study, as it is crucial to ascertain whether Bitcoin can be used as a hedge against adverse market conditions in the South African context, conditions like increases in inflation, stock market downturns, and exchange rate depreciation. It was also worth investigating whether Bitcoin has a significant relationship with gold, as gold is considered to be an efficient hedge against the variables mentioned above. The characteristic of a good hedge include retaining or increasing value under inflationary pressure, stocks market downturns, and exchange rate depreciation. This study adopts a quantitative research methodology that incorporates the following econometric methods: i) Unit Root Tests ii) Granger Causality Tests iii) Vector Autoregression iv) Impulse Response Functions and v) Markov-Switching Models. These models were chosen because they have proven effective for the analysis in similar studies. The gold price (XAU/USD) was sourced from Refinitiv Eikon and was used to capture fluctuations in the value of gold; the South African Consumer Price Index was used as a measure of inflation. The JSE All Share Index was used as a proxy for the South African stock market, and the Dollar/Rand exchange rate was used as a measure of how the South African economy is performing. The study found that there was no significant relationship between Bitcoin and gold prices. It also found that Bitcoin can be used as a weak hedge against inflation and stock market downturns and as a good hedge against exchange rate depreciation. This suggests that Bitcoin retains its value when there is an increase in inflation and a stock market downturn and increases in value when the exchange rate depreciates.   • Description: Freshwater fishes are in global decline and fish as a group are the most imperilled of all vertebrates. Freshwater systems are among the most threatened globally, largely owing to their comparatively high species and habitat diversity while occupying a minute fraction of the Earth’s surface. In South Africa, invasion by non-native fishes has had a devastating effect on freshwater systems across the country. Numerous other stressors and anthropogenic impacts continue to impact these systems, including habitat degradation, water abstraction and global change effects. In general, South African freshwater systems are under-studied and there is a lack of baseline biological and ecological studies on many freshwater fish species. The Cape Fold Ecoregion (CFE) of South Africa is a particularly vulnerable region, with many range-restricted species and highly fragmented native fish ranges following high invasion rates. Within the CFE, the Olifants-Doring River System (ODRS) is of primary concern owing to the high endemism and imperilled status of its freshwater fish species. The Rondegat River in the ODRS is of notable conservation value, as it hosts populations of important endemic CFE species. This river is unique, being the site of the first alien fish eradication programme of its kind in South Africa. Thus the Rondegat River, and in particular its imperilled cyprinid assemblage, is used here as a case study of the responses and community dynamics of recovering freshwater fish species. The members of this cyprinid assemblage are Sedercypris calidus, Pseudobarbus phlegethon and Labeobarbus seeberi. Sedercypris calidus and L. seeberi are listed by the International Union for the Conservation of Nature (IUCN) as “Near-Threatened”, while P. phlegethon is listed as “Endangered”. Understanding the dynamics and responses to understudied vulnerable fish communities in the wake of restoration efforts was the overarching goal of this thesis. The first chapter served as a review of current knowledge of the CFE, the Rondegat River, the myriad stressors that have impacted on or are projected to affect this region, and the freshwater fish species on which those stressors act. My first investigations served as an extension of the existing Rondegat River monitoring programme, making use of remote underwater video (RUV) data to assess relative abundance and habitat associations of the focal species (Chapter 2). A relatively limited spatial range of P. phlegethon was established, which was suggested to be a result of relatively highly specialised habitat requirements and sensitivity to disturbance. Sedercypris calidus was confirmed as a relatively more abundant and ubiquitous species across the length of the Rondegat River, sharing much of the lower and middle reaches with L. seeberi. I sought to use experimental trials of functional response, as a proxy for feeding performance, across representative temperatures and relevant prey types, in the spatially overlapping S. calidus and L. seeberi (Chapter 3). Labeobarbus seeberi generally outcompeted S. calidus across temperature treatments and prey types. The ecomorphology and diet of all three species were used to construct trophic profiles, which suggested that there was a high degree of feeding capacity overlap between L. seeberi and S. calidus, while P. phlegethon diverged from the other two species (Chapter 4). Gut content suggested that all three species overlapped broadly in diet. This indicated that the realised trophic niche of these species is similar, despite some morphological specialisation. I then used RUV data to investigate in-situ feeding behaviours, with the aim to disentangle the nuances of community dynamics and mechanisms of coexistence in the cyprinid assemblage (Chapter 5). I found that, despite the higher feeding performance of L. seeberi (Chapter 3) and its overlaps in diet and feeding capacity with S. calidus (Chapter 4), S. calidus is able to mitigate competitive pressures through foraging mode switching and exploitation of allochthonous food inputs. Evidence for further habitat and prey selectivity in Pseudobarbus phlegethon was gathered based on dependence on complex habitats and pool refugia for the majority of its feeding, supporting this species as a headwater specialist; alongside signals of its spatial and habitat use patterns (Chapter 2). While S. calidus and L. seeberi were found to be less habitat-specific than P. phlegethon, caution was noted in the potential for ongoing stressors, such as habitat destruction, loss of river connectivity and global change effects, to impact on the reproductive success of these two species. Stressors affecting the habitats and sensitive invertebrate taxa upon which all three species depend continue to threaten the Rondegat system, highlighting the need to maintain ecosystem integrity through conservation interventions. There remains significant scope to maintain restoration efforts in the Rondegat River and other river systems of the CFE, through direct conservation actions, enhanced community awareness, indigenous riparian vegetation restoration and involvement of local stakeholders in various conservation-centred activities. , Thesis (PhD) -- Faculty of Science, Ichthyology and Fisheries Science, 2022

  • Description: Introduction: School going adolescents have been identified as a group that is at risk of obtaining insufficient sleep due to shortened and irregular sleep, all of which can affect the overall wellbeing of adolescents. While there has been extenstive research on adolescent sleep around the world, research in South Africa has been limited. Of the research performed in this context, most has been conducted in city or urban contexts, but there has been comparatively less research on sleep in adolescents from rural and township areas. Therefore, this study focused on characterising and comparing the sleep-wake behaviour of late adolescents from selected rural and township areas in Alice in the Eastern Cape of South Africa. Methods: An adapted version School Sleep Habits Survey (SSHS) was included in the study and assessed demographic, academic, sleep-wake behaviour, lifestyle, and behavioural information of adolescents from rural and township schools in and around Alice. The survey was distributed physically to all the learners from four co-educational rural and township schools. The responses to the survey were analysed with descriptive and non-parametric inferential statistics, as all data were not normally distributed. Results: A total of 123 learners completed the survey. All the participants were in Grade 12, aged 16 to 26 years (median =18, IQR= 18-19) from rural (n=28) and township (n=95) schools. Seventy-four female and 49 male learners participated in the study. Within the sample, there were also a group of participants who were not adolescents (n=22) whose age ranged between 20 and 26. Reported sleep during both the week and weekend (7hours) was lower than both the recommended duration for adolescents and their own self-reported sleep need of a median of 8 hours and 14 minutes. Bedtimes were significantly earlier on weekdays than weekends. The need to do homework and feeling sleepy were cited as the main reasons for going to sleep during the week and the weekends. Wake times on weekdays and weekends did not differ, with most learners citing being woken up by an alarm clock as the main reason for waking during both weekdays and weekends. Most learners (n=47; 38%) reported walking to get to school, while 41 (33%) relied on public transport to get to school. There were no significant effects of gender or of being rural and township scholars in relation to their sleep variables. Comparisons between non-adolescents and adolescents revealed that non-adolescents reported sleeping longer (7 hours and 30 minutes) compared to adolescents (7 hours) than adolescents did on weekends. As a group, the learners in this sample were morningness-orientated in terms of chronotype, presented with low day time sleepiness, some evidence of sleep-wake behaviour problems and of depressive mood symptoms. Nearly half (n=53; 43%) the group reported napping occasionally on school days. In terms of caffeine consumption, just over half (n=64; 52%) of the learners reported never consuming caffeinated substances such as soda, coffee, or tea. Conclusions: The reported sleep duration of learners in this study was consistent on both the week and weekend but was shorter than the recommended 8-10 hours for late adolescents. While learners in this sample reported early bedtimes, sleep was likely curtailed by the early rise times due to the contextual constraints where scholars had to commute, by either walking or taking public transport, to make the early school start times. There was a high prevalence of sleep disturbances in this sample however, the effects of this (increased daytime sleepiness) were lower which can be attributed to low reported prevalence of caffeine consumption combined with the high prevalence of reported day time napping on school days and weekends. While only a snapshot, the results of this study support previous research that has highlighted the challenges that many adolescents across the globe experience when it comes to obtaining enough sleep. The results of this study highlighted the need for more research to understand the sleep-wake behaviours of learners and their lifestyles more objectively. Given the early starts for many participants in this study, interventions such as delaying school start times may help to improve the amount of sleep that learners get during the week. However, extensive consultations would be needed to ensure that systemic changes that aim to improve sleep of adolescents are applicable for the context of rural and townships areas in South Africa. , Thesis (MSc) -- Faculty of Science, Human Kinetics and Ergonomics, 2022
