- Description: Picornaviruses belong to the Picornaviridae family which are one of the largest and most diverse family of RNA viruses that cause a broad spectrum of infections in both humans and animals. These diseases range from severe infections such as poliomyelitis, meningitis, myocarditis to mild illnesses such as the common cold. Picornavirus outbreaks are a worldwide threat as they are continuously occurring. A recent outbreak of foot-and-mouth disease caused by a picornavirus occurred in South Africa, resulting in a temporary ban on the movement of cattle. Currently, the FDA has not approved any antiviral drugs against this virus, increasing the urgency for identifying effective antivirals. Picornaviruses have similar genomes and capsid organisation as such, those that are non-hazardous to humans can be used as a model system. A Theiler’s murine encephalomyelitis virus (TMEV) strain GDVII and Baby Hamster Kidney fibroblasts (BHK-21 cells) was used as a replication system to develop and optimise a medium-throughput antiviral screening assay. The TMEV GDVII replication system in BHK-21 cells was validated, and preliminary experiments were performed that were necessary for the development of the TMEV GDVII antiviral assay. This was achieved by conducting a CPE assay to visually monitor the onset and development of CPE induced by TMEV GDVII. Plaque assays accurately quantified the number of infectious virus particles required for calculating the MOI in downstream experiments. Lastly, indirect immunofluorescence and Western blot analysis detected the expression of viral proteins using previously generated antibodies against the TMEV GDVII VP1 capsid and 2C protein, thereby confirming infection in BHK-21 cells. The development of robust and reproducible assays is an essential component in antiviral drug discovery. Therefore, the confirmed replication system was then used as a foundation to develop a medium-throughput CPE-based TMEV GDVII antiviral assay whereby the parameters were optimised to produce one of high quality. Firstly, the quantitation of viral-induced CPE was examined and confirmed in a 96-well plate using resazurin as a cell viability indicator. Each parameter was tested at varying conditions, and the optimal was concluded as 2 % FBS in the assay media, a 15 000 cells/well seeding density, infecting the cells with TMEV GDVII at an MOI of 0.00625 and measuring resazurin at an endpoint of 72 hpi. Furthermore, the parameters were ii validated by calculating the Z’- factor, which consistently produced scores above 0.5, indicative of a reliable, robust, reproducible antiviral assay. Currently, there are no inhibitors against TMEV GDVII that have been reported or confirmed in cell lines, animal models or clinical trials. Therefore, once the optimal assay parameters were selected, it presented an opportunity to assess whether potential compounds, including itraconazole (ITZ) and dipyridamole (DIP), possessed antiviral activity that could firstly, be utilised as a control inhibitor when screening compounds against TMEV GDVII and secondly, contribute to research on this virus. Additionally, the previously produced anti-TMEV GDVII capsid antibody was shown to neutralise viral infection and was also included as a potential control. The sensitivity of the cells towards DMSO, a solution in which the compounds were solubilised, was first investigated. It was found that concentrations above 1 % are toxic to the cells; as such, the final DMSO concentrations were always kept below 1 % when screening compounds. Lastly, the generation of dose-response curves aided in the conclusion that the antibody was the most suitable control inhibitor as it displayed potent antiviral activity and no cytotoxicity towards the cells. In contrast, ITZ and DIP did not possess effective antiviral action and were toxic to cells at high concentrations. Finally, after all the components of the medium-throughput TMEV GDVII antiviral assay were identified, it was possible to screen 24 compounds from a coumarin and marine natural product library for cell cytotoxicity and antiviral activity. After generating dose-response curves, it was concluded that no compound effectively inhibited virus-induced CPE, and most were toxic to cells at relatively high concentrations. The effect of piospheres on the ecology of insectivorous birds and their arthropod prey

- Description: Desertification is the degradation of arid ecosystems that result in the loss of biodiversity. Piospheres are areas of local degradation around a central point due to overgrazing and increased herbivore presence. There is a paucity of information regarding the effect of localised degradation on arthropods and insectivorous birds. Both of these organisms play crucial roles in ecosystem functioning and stability and can be used as models to study ecosystem functioning. I investigated the effect of piospheres on arthropods and birds in the Eastern Cape province of South Africa. I found that the abundance and diversity of arthropods were significantly reduced inside the piospheres due to the lack of vegetation. Termites were a group specifically negatively impacted by piospheres, with a significant reduction in their presence inside the piosphere. The family composition of arthropods also changed inside and outside the piospheres, with Caelifera, Diptera and Formicidae being the most dominant groups. The diversity of birds was also significantly reduced due to the degradation inside the piospheres. Looking at insectivorous birds, I found that the reduction in both vegetation and arthropod prey availability resulted in non-random avoidance of piospheres. This shows that piospheres negatively impact both arthropods and birds. The results from my study are supported by other literature studying the effects of habitat degradation associated with desertification. Due to the similarities of degradation between piospheres and desertification I make the argument that piospheres can be studied as localised models of desertification. The decrease in vegetation and arthropod abundance and diversity was found to further impact the feeding success of insectivorous birds. Using piospheres as a model for desertification, I found that the foraging effort of birds is significantly reduced within a degraded area due to the lack of vegetation providing safety to arthropod prey species. In addition to this, the foraging efficiency of insectivorous birds is significantly reduced inside the piospheres due to the decreased arthropod abundance and diversity. This shows that inside the piospheres birds spend less time searching for insects and have fewer successful feeds. The impacts of game farming on worker’s livelihoods: a case of a private game reserve in South Africa’s Eastern Cape Province

- Description: Land use shifts from commercial agricultural farming to game farming through wildlife tourism, has been prevalent in post-apartheid South Africa. Scholars have argued that wildlife tourism to be more profitable than other forms of commercial farming. As such, game farming has been promoted as a community-engaged vehicle for rural development through the creation of better employment opportunities. This study explored the implications of these shifts on the livelihoods of farm workers living on the game farms. Unlike labour tenants, farm workers who live on the private game, earn a wage. However, farm workers’ accommodation on the game farm is dependent on them being employed on the farm. The aim of the study was to explore the impacts of game farms on the livelihoods of farm workers since the shift from commercial agricultural practices to tourism services in the form of private game reserves. A case study research design was used to gather in-depth information about the multiple realities of the participants, not only about their employment relationships, but also their experiences living on the property on which they are employed. I collected data mainly through semi-structured interviews. I also collected and analysed company documents. The study adopted a Sustainable Livelihoods Framework as a theoretical schema to analyse the livelihood assets readily available to the farm dwellers and the various livelihood strategies they used to make ends meet. The findings of the study revealed that residing on the game farm, although it provided access to free accommodation, tended to pose several challenges to the workers, including i) exploitative employment structures and ii) insecure land rights. The findings also revealed human capital development initiatives mandated for employees were not formally documented. As a result, employees were confined in their current employment. Consequently, the study revealed that participants who had been employed by the game farm for over a decade showed little evidence of enhanced livelihoods. The livelihood strategies of the participants were their basic employment wage, the use of money borrowing systems through stokvels, and informal trading such as selling sweets and chips in the game reserve villages where other workers live with their families. , Thesis (MA) -- Faculty of Humanities, Sociology, 2023

- Description: Mental distress is a universal phenomenon experienced by many individuals despite age, race, gender, occupation, or socio-cultural context and is slowly becoming a major contributor to the burden of disease in South Africa. However, mental distress fails to take precedence in SA because of inherent intricacies in understandings about it, as a result of ways of being conceptualised and interpreted differently across cultures. This research study explored amaXhosa healthcare workers’ understandings, knowledge, practices, and attitudes regarding mental distress amongst some amaXhosa people. The study aimed to investigate what mental distress means for some people who belong to the amaXhosa ethnic group, to uncover how they conceptualise mental distress, seek help or what behaviours prevent help-seeking. It aimed to highlight any prevalent attitudes of stigma and discrimination, to build insight into overlooked aspects in psychotherapy practice especially when dealing with non-western populations. The study utilised interpretative phenomenological analysis (IPA) as its analytical lens; to explore and to enter as far as possible into the worlds of participants to generate rich data and in-depth analysis. Four participants were recruited through both purposive and snowball sampling and data were gathered using individual semi-structured interviews. From the data collected, the findings illustrate a limited understanding of mental distress amongst some amaXhosa people due to lack of education and awareness on the subject, leading to the apparent silence in discussing such matters and inadvertently predisposing it to being viewed as taboo. The predominant themes as evidenced by the data were the use of language that sensationalises mental distress; misinformation; Afrocentric beliefs that rationalise mental distress; alienation and segregation of those affected; and the primary healthcare system as a source of reinforcing prevalent stigma and discrimination. - Description: Very few students in South Africa (SA) receive adequate career counselling at school and often arrive at institutions of higher learning without a clear sense of what their prospective careers will entail. The limited access to career counselling and assessment is further compounded by criticisms of popular career assessment instruments used in SA, where the primary goal of career counselling and assessment is to match clients to careers for job placement. This approach no longer serves the needs of a diverse SA population and disregards contextual influences on careers. Therefore, a need exists to understand and inform career counselling interventions and to guard against the provision of de-contextualised and contextually insensitive approaches. Moreover, changes in the workplace, where career change has become the norm as the world of work has moved away from stability and permanency to fluidity, calls for a contemporary response from career counselling theories, practitioners as well as researchers, to equip clients with the necessary skills to respond to these changes. Thus, SA institutions of higher learning need to innovate their practices to more inclusively, effectively, and justly serve the needs of a diverse student population for work success in a developing nation and a complex world. This study aimed to evaluate the relevance and effectiveness of a university’s career counselling services from the students’ perspectives. It focuses on gaining an indication of the perceived effectiveness of assessment measures used, and to ascertain the influences that impact on individual students’ career decision-making over the course of undergraduate studies. Due to the increasing recognition for the need to develop a contextualised approach to career development interventions, this study included the SACII, a locally developed interest inventory as part of a career assessment battery, with a group of university students. A pragmatic approach using mixed-methods was used. Multiple case studies of the career trajectories of a cohort of undergraduate students who had undertaken career assessments at the university career centre, were tracked longitudinally over the course of undergraduate studies. Data were generated through vignettes that included the 13 participants' career assessment reports, a service evaluation questionnaire and two successive follow-up interviews. Each participant's first follow-up interview occurred six months after their career assessment feedback session; and the second follow-up was in the final year of undergraduate studies, after eighteen months. Descriptive statistical analysis summarised the basic features of the quantitative data from the evaluation questionnaire. Thematic analysis was used for organisation and analysis of the follow-up interview data. Findings were that the career counselling and assessment facilitated greater self-knowledge in relation to career decision-making, assisted with selecting degree major subjects and enhanced career planning abilities. Using locally developed assessment measures in career counselling proved to be useful. The use of dynamic software to potentially enhance conceptual understanding and a productive disposition in the visual learning of algebra: an interventionist case study

The introduction and development of technology into mathematics is one of the occurrences which has also contributed to a new discourse in teaching mathematics – in this case the teaching of algebra. Algebra is still seen as a gatekeeper and remains as one of the key reasons for a negative disposition amongst learners towards learning the subject. Across the globe psychologists, philosophers and educators continue to engage in debates and research projects in search of answers and solutions for the improvement of algebra teaching and an improvement in dispositions towards learning algebra. This thesis reports on a research project that focused on the use of dynamic software to enhance the conceptual understanding and productive dispositions of selected learners through the visual learning of abstract algebraic concepts. The research was executed as an interventionist case study. A case study methodological strategy was adopted with two groups of 30 Grade 9 learners. One group was a Grade 9 mathematics class of a school in Windhoek who scored above average in algebra but showed a very low disposition score, while the other group was made up of learners from a community project who scored high on the disposition scale but achieved below average results in algebra. The analytical framework of the case study is structured around a combination of complementary algebraic topics presented through visual learning, with GeoGebra as a medium of instruction. With the focus on visualisation and the use of technology the study investigated and attempted to understand how participants processed and internalised algebraic concepts to make sense of abstract algebraic concepts and eventually gain sustained conceptual understanding. The study was framed by the theoretical theories of constructivism and the Dual Coding Theory. For the collection of data, a mixed methods approach was adopted following three cycles. Three algebraic topics were taught with GeoGebra applets yielding both qualitative and quantitative data, through the observation of participants, screen captures and reflective interviews, using instruments designed specifically for the study and collecting quantitative achievement test results. The study, a journey that both the participants and the researcher embarked upon, revealed that the use of technology enhanced conceptual understanding for both groups and both groups showed a positive change in disposition towards learning algebra. The intervention with GeoGebra consistently and progressively improved in terms of conceptual understanding and dispositions towards learning algebra significant improvements in results were achieved. The findings showed that this approach to teaching algebra yielded positive results and gave new insights into visual teaching with technology. Visual representations of linear algebraic expressions: a case study in a Grade 9 after-school mathematics club

- Description: Visualisation is commonly used as a tool in introducing algebra through visual or kinaesthetic sequences designed to prompt learners' development of a general rule for moving from a term's position to its output value. Fluency in both the concepts and the conventions of elementary algebra are essential to learners, as algebra forms the language in which many advanced mathematical ideas are encoded. Moreover, algebraic fluency is often associated with an ability to think abstractly about arithmetic processes. In many classrooms, however, research has shown that learners often focus on fluency in algebraic conventions rather than concepts, learning how to manipulate expressions without understanding the algorithms they are taught to follow. This trend can be linked to several causes, including teacher-centred mathematics classrooms in which learners are – whether implicitly or explicitly – encouraged to copy formulae and methods in order to ‘get it right in a test’ without necessarily grasping the underlying logical relationships. This case study, therefore, aimed to determine whether there was value in using visual, kinaesthetic models to broaden and deepen learners' use of algebra. To that end, in the context of an extra-curricular mathematics club that aimed to decentre the teachers and demand innovative ideas of the participants, six pairs of Grade 9 learners were tasked with creating visual representations of a linear algebraic expression using coloured building cubes. The responses to this task over the course of five assignments were many and varied and almost universally displayed a sustained internal logic that the learners were able to explain and develop. Most pairs began with a visual list of terms arranged in sets of towers, pyramids or, in one case, a spiral. At the end of the study, all but one of the pairs had settled on a Visual Expression, in which various colours were used to represent elements of the algebra such as the values of the coefficient, the variable and the constant term. The participants' representations grew in complexity over the course of the study and the conformity of the final responses showed that the club was a collaborative space in which learners shared ideas. This dissertation addresses this gap by examining the municipal public resource management processes from an oversight perspective. The study sought to analyse the effectiveness of year-end oversight at Amathole District Municipality (ADM) over water and sanitation services. This research aimed to study the year-end oversight over service delivery with reference to the Municipal Public Resource Management (MPRM) model. According to the model, effective service delivery includes inter alia effective oversight, as oversight forms part of the entire system of the municipal resource management cycle. Thus, for the MPRM model, oversight should be exercised on all these processes. The research was a desktop analysis that used both a qualitative and quantitative research methodology. The primary document analysed was the ADM Oversight Report (OR). This report is produced by the Municipal Public Accounts Committee (MPAC) and reflects the year-end oversight conducted by the MPAC on the municipality's activities. In the analysis of the OR, it was concluded that the ADM MPAC had failed to conduct effective year-end oversight of W&S services. The efficacy of the ADM MPAC's year-end oversight was measured against guidelines for effective year-end oversight from the National Treasury and the MPRM model. A native weevil and an exotic planthopper: investigating potential biological control agents for nymphaea mexicana zuccarini (nymphaeaceae) and its hybrids in South Africa

- Description: Nymphaea mexicana Zuccarini (Nymphaeaceae) is an invasive plant originating from southern USA and Mexico that has become problematic in South Africa, invading several water bodies around the country. Manual removal of this plant is very labour intensive and is not cost efficient or effective for long term control, while the use of herbicides is damaging to the environment and expensive. Consequently, this plant is a desirable candidate for biological control, which takes advantage of enemy release of the target weed and aims to re-establish population suppression induced by host specific natural enemies. Initiating biological control requires that several steps are followed to maximise the success of the programme, and the first few of these, including overseas surveys in the native range of the plant, have already been completed. This thesis aimed to continue biological control research for this species to take further steps at effectively managing the plant. Firstly, pre-release surveys in the invaded range are necessary to: determine what factors (including enemy release) contribute to the invasiveness of the target weed; establish a baseline of information to allow for comparison after biological control agents have been released; and identify any insect herbivores that may already be present in the country. The pre-release surveys conducted in this study revealed useful information about N. mexicana invasions in South Africa and provided evidence that enemy release is applicable to this case. However, these studies determined that a native weevil, Bagous longulus Gyllenhal (Coleoptera: Curculionidae), has expanded its host range to include the exotic N. mexicana at three sites, and may thus have potential for management of the species through augmentative releases. The invasion of N. mexicana in South Africa is further complicated by the presence of several Nymphaea hybrids originating from a complex history of horticultural trade. Although previous research has shown that several hybrid groups are present in South Africa, their parentage is not known. As biological control requires the use of host specific insects adapted to overcome the unique chemical and morphological defences utilised by plant species, hybrids are notoriously difficult to manage because they possess intermediate characters inherited from parent species to which natural enemies may not have adapted. Although biological control of hybrids is challenging, other case studies have demonstrated that it is possible to find suitable agents, but the chances of success are increased if putative parents of the hybrids are known so that they can be surveyed for natural enemies. Further molecular studies including possible parents of the Nymphaea hybrids in South Africa were thus carried out in this thesis to focus future surveying efforts. Two main hybrid groups were identified with genetic similarity to two tested putative Nymphaea parents, and this will allow further investigations of these species to improve the chances of successfully managing these hybrid groups. Some of the tested hybrids showed genetic contributions from multiple groups, some of which were unidentified, so it is necessary to prioritise the most problematic hybrids for biological control. With more insight into the genetic makeup of the Nymphaea hybrids in South Africa, investigations into the host specificity of potential biological control agents can be conducted. The ideal biological control agent should have a broad enough host range to impact and survive on both N. mexicana and its hybrids, but without a host range so broad that it would pose risk to native South African species. Host specificity trials are thus necessary to determine the suitability of potential agents. The identification of B. longulus feeding on N. mexicana during pre-release surveys motivated further investigations to determine the natural distribution, field host range, and host specificity of B. longulus in experimentally controlled conditions. Further surveys were therefore conducted at native Nymphaea sites around South Africa in addition to host specificity trials using the native Nymphaea nouchali Burm. f. (Nymphaeaceae), two populations of N. mexicana, and a cultivated hybrid. Results from the surveys and host specificity tests suggest that B. longulus is widely distributed across South Africa, is specific to Nymphaea with no observed preference between N. mexicana and the native N. nouchali, and does not perform well on Nymphaea hybrids. Hence, B. longulus is promising for use in new association biological control through augmentative releases but is not suitable for management of hybrids. In addition to the potential use of the South African B. longulus, it is necessary to conduct host specificity trials for natural enemies from the native range of N. mexicana that were prioritised in previous studies. Megamelus toddi Beamer (Hemiptera: Delphacidae) is one such species that was imported into quarantined laboratory conditions from Florida, USA. Host specificity trials were conducted using the same test plants as described for the studies on B. longulus, in addition to multigeneration trials to determine how long M. toddi could survive on non-target host plants. As with the B. longulus studies, no statistically significant differences in preference were observed between N. mexicana and N. nouchali, but M. toddi could not complete development on the test hybrid, indicating that this species is also unsuitable for the management of Nymphaea hybrids. Despite suboptimal plant health, M. toddi completed development for three generations on the native N. nouchali. This lack of host specificity deems M. toddi unsafe for release in South Africa but highlights the importance of following predefined steps to develop a biological control programme. The concluding chapter of this thesis discusses the aforementioned findings in a broader context by considering the driving forces of plant invasions in general and specifically for N. mexicana in South Africa. - Description: Nymphaea mexicana Zuccarini (Nymphaeaceae) is an invasive plant originating from southern USA and Mexico that has become problematic in South Africa, invading several water bodies around the country. Manual removal of this plant is very labour intensive and is not cost efficient or effective for long term control, while the use of herbicides is damaging to the environment and expensive. Consequently, this plant is a desirable candidate for biological control, which takes advantage of enemy release of the target weed and aims to re-establish population suppression induced by host specific natural enemies. Initiating biological control requires that several steps are followed to maximise the success of the programme, and the first few of these, including overseas surveys in the native range of the plant, have already been completed. This thesis aimed to continue biological control research for this species to take further steps at effectively managing the plant. Firstly, pre-release surveys in the invaded range are necessary to: determine what factors (including enemy release) contribute to the invasiveness of the target weed; establish a baseline of information to allow for comparison after biological control agents have been released; and identify any insect herbivores that may already be present in the country. The pre-release surveys conducted in this study revealed useful information about N. mexicana invasions in South Africa and provided evidence that enemy release is applicable to this case. However, these studies determined that a native weevil, Bagous longulus Gyllenhal (Coleoptera: Curculionidae), has expanded its host range to include the exotic N. mexicana at three sites, and may thus have potential for management of the species through augmentative releases. The invasion of N. mexicana in South Africa is further complicated by the presence of several Nymphaea hybrids originating from a complex history of horticultural trade. Although previous research has shown that several hybrid groups are present in South Africa, their parentage is not known. As biological control requires the use of host specific insects adapted to overcome the unique chemical and morphological defences utilised by plant species, hybrids are notoriously difficult to manage because they possess intermediate characters inherited from parent species to which natural enemies may not have adapted. Although biological control of hybrids is challenging, other case studies have demonstrated that it is possible to find suitable agents, but the chances of success are increased if putative parents of the hybrids are known so that they can be surveyed for natural enemies. Further molecular studies including possible parents of the Nymphaea hybrids in South Africa were thus carried out in this thesis to focus future surveying efforts. Two main hybrid groups were identified with genetic similarity to two tested putative Nymphaea parents, and this will allow further investigations of these species to improve the chances of successfully managing these hybrid groups. Some of the tested hybrids showed genetic contributions from multiple groups, some of which were unidentified, so it is necessary to prioritise the most problematic hybrids for biological control. With more insight into the genetic makeup of the Nymphaea hybrids in South Africa, investigations into the host specificity of potential biological control agents can be conducted. The ideal biological control agent should have a broad enough host range to impact and survive on both N. mexicana and its hybrids, but without a host range so broad that it would pose risk to native South African species. Host specificity trials are thus necessary to determine the suitability of potential agents. The identification of B. longulus feeding on N. mexicana during pre-release surveys motivated further investigations to determine the natural distribution, field host range, and host specificity of B. longulus in experimentally controlled conditions. Further surveys were therefore conducted at native Nymphaea sites around South Africa in addition to host specificity trials using the native Nymphaea nouchali Burm. f. (Nymphaeaceae), two populations of N. mexicana, and a cultivated hybrid. Results from the surveys and host specificity tests suggest that B. longulus is widely distributed across South Africa, is specific to Nymphaea with no observed preference between N. mexicana and the native N. nouchali, and does not perform well on Nymphaea hybrids. Hence, B. longulus is promising for use in new association biological control through augmentative releases but is not suitable for management of hybrids. In addition to the potential use of the South African B. longulus, it is necessary to conduct host specificity trials for natural enemies from the native range of N. mexicana that were prioritised in previous studies. Megamelus toddi Beamer (Hemiptera: Delphacidae) is one such species that was imported into quarantined laboratory conditions from Florida, USA. Host specificity trials were conducted using the same test plants as described for the studies on B. longulus, in addition to multigeneration trials to determine how long M. toddi could survive on non-target host plants. As with the B. longulus studies, no statistically significant differences in preference were observed between N. mexicana and N. nouchali, but M. toddi could not complete development on the test hybrid, indicating that this species is also unsuitable for the management of Nymphaea hybrids. Despite suboptimal plant health, M. toddi completed development for three generations on the native N. nouchali. This lack of host specificity deems M. toddi unsafe for release in South Africa but highlights the importance of following predefined steps to develop a biological control programme. The concluding chapter of this thesis discusses the aforementioned findings in a broader context by considering the driving forces of plant invasions in general and specifically for N. mexicana in South Africa. Themes of change that emerged from the nurses’ experiences were rendering counselling services in a non- judgemental, non-directive way, being acknowledged and recognized for the services they offer as abortion providers, and accepting that abortion is a reproductive legal right that can be accessed whenever a woman in need of the service requests it. The findings of this research also indicate that, despite the controversy and stigma attached to providing abortion services, the participants were motivated to continue offering abortion services because they felt equipped since undergoing abortion counselling training. A Pedagogy of Love: reflections on 25 years of informal vocational education and training practices in the commercial fishing industry in South Africa

- Description: This thesis is a reflection of informal vocational education and training (IVET) practices conducted by the Researcher in the commercial fishing industry between 1995-2021. Fourteen interventions took place during this time which involved several hundred sea-going employees who are disadvantaged by low levels of literacies and low/no Internet Computer Technology connectivity. The Production Programme was chosen as the focus of study and draws upon the influence of the other thirteen programmes. The purpose of the Production Programme was to teach technical fish processing skills to the production management teams and factory hands who work in the factories on board the vessels. The programme ran for five years and evolved through three distinct generations. At the heart of this work lies the question: ‘What made these programmes successful?’ This is an intra-programmatic study and seeks to identify the pedagogical practices which promoted or confounded the efficacy of the Production Programme; and based upon this understanding how such programmes could be improved, transferred, and taken to scale. These questions are both descriptive and explorative in nature. IVET is regarded as training which takes place outside of the formal South African National Qualifications Framework. This work is relevant because approximately 80% of sea-going staff neither finish school, nor get the opportunity of Post School Education and Training by going to a university, a technical institution, or a community college. This statistic is reflected in the general population (Department of Higher Education and Training [DHET], 2022). There is a fine line between being employed and unemployed for people working at this level in the formal economy which makes this project relevant to youth or persons who are ‘not in employment, education or training’ too. This means that for most South African adults IVET presents a significant opportunity for post-school education. Therefore, it is important to answer the questions raised above to rapidly improve inclusion of the majority of South Africans into meaningful education which improves livelihoods. The Theory of Practice Architectures (TPA), from the collection of social-material practice theories, is the conceptual framework for this thesis. The smallest unit of analysis of TPA are practices which may be ‘sayings/thinkings’, ‘doings’, or ‘relatings’. These practices bundle together into practice arrangements and form practice architectures. The reason that TPA was chosen was that practices were the only data available as we (learners, managers, facilitators, and me) knew what we had said/thought or done, and we were aware of the relationships between us over the years as the fourteen interventions played out. Under conventional research circumstances data would be collected in real time, however in this project, most of the data is historical. In addition to the fundamental building blocks of TPA, the theory is embedded in a Theory of Education. There have also been contemporary enhancements to the TPA which were significant to this study, for example, the Ecologies of Practices, a Trellis of Practices which Support Professional Learning, Middle Leadership, Travelling Practices, and moves towards transformative or transgressive education using the TPA in IVET. This is empirical, qualitative research and an ethnographic case study was chosen as the research design which is a methodology particularly suited to answering both descriptive and explorative questions. Nine methods for data collection were used, namely an historical reflective narrative; two focused-group interviews; three individual interviews; four Whatsapp videos; one WhatsApp voice note; two mobisodes; ten questionnaires; 29 documents; and 16 photographs. Because this data was collected under Covid-19 pandemic conditions, two conceptions were employed to guide the generation of data under these uncertain and constrained conditions. These were firstly, the ‘methodology of chance’ which allowed for a ‘methodological agility’; and secondly, the idea of ‘information power’ which is helpful in deciding on how much data is enough. In order to be explicit concerning a key research activity, the approach and method used to review the literature is explicated. Key practices were identified in the data set and described; and then the data was analysed using heuristics provided by TPA theorists. Seven Tables of Invention were used to synthesise the data arising from these practices. An eighth Table of Invention was used to synthesise all the practices and practice arrangements characterised; and to indicate how these evolved over time and space. The data description and analysis is supported by eight Analytic Memos, a comprehensive Data Code Table and a hyperlink to a data repository which provides access to oral and video material. The findings distinguished five key practices and practice arrangements which were: Practices of the Creation of Courseware; Practices of Teaching and Learning; Practices of Assessment; Practices of Love; Practices of Management. The thesis title is reflective of the impact which love has upon the pedagogical process of IVET. Based upon the analysis and synthesis of the corpus of data, practices which either promoted or confounded the Production Programme became visible; it is these insights which inform future improvements to similar programmes. Emanating from these findings, two overarching practice architectures (PA) were identified which restrained the Production Programme in the same manner that the banks of a river restrain a river, and yet simultaneously, are changed by the river over time. These are the PA of Methodology and Methods and the PA of Maturing Ecologies of Practices. The inferences drawn from the data were achieved through the use of deductive, inductive, and abductive reasoning. My claim to new knowledge is a lamination of a practical contribution on one side of the coin, and a theoretical contribution on the other side of the coin. The PA of Methodology and Methods provides a practical mechanism to create, deliver and assess IVET. This is done by explicating the three practice architectures which constitute the overarching PA of Methodology and Methods which are, the PA of Informality; the PA of Range, and the PA of Relationality. An IVET programme constitutes Ecologies of Practices. The theoretical conception of the overarching PA of Maturing Ecologies of Practices provides a conceptual tool which enables the transferring and scaling of IVET programmes. It does this by providing theoretical indicators to establish the ‘state’ of an IVET programme as it matures over time from a pioneer state to a settler state. An IVET educator can then work towards creating a PA which is conducive for a mature ecologies of practices to form; and the programme can then be transferred and/or taken to scale, if this is desirable in the particular context. The power of my claim to new knowledge does not lie on one side or the other of the coin, but in the lamination of the practical and theoretical contributions put to use in the service of IVET. This thesis concludes with a number of theoretical and practical recommendations which are loosely grouped according to ‘sayings/thinkings’, ‘doings’ and ‘relatings’ in deference to the value of TPA to this thesis. Biological longitudinal aspects of the Kabompo River, a significant tributary of the upper Zambezi sub-catchment, North-west Province, Zambia

- Description: The landscape of the Upper Zambezi Catchment in the remote North-Western Province of Zambia retains many natural features, but current and future anthropogenic activities threaten its integrity. Lack of information relating to the basic ecological functioning of the region hampers conservation efforts. Flooding from the adjacent woody-savanna and open grasslands in the Upper-Zambezi Catchment in the wet season drives crucial processes in the aquatic ecosystems such as the Kabompo River. This thesis describes aquatic food webs and describes the nutrient cycling in this river. A general introduction was based on the seasonality, river morphology, and biodiversity of the Kabompo River. There is no aquatic food web information available on this river. To address this knowledge gap, sampling of the principal food web components– vegetation, invertebrates, and fish – were collected in the dry season from August to September 2019 across six sites in the Kabompo River. Stable isotope analyses provided proxies for the food web structure at each site, and were the basis of longitudinal comparisons. The analyses suggest that the Kabompo River food webs follow the trophic ordination and nutrient cycling characteristic of the revised-Riverine Productivity Model. Food webs and community assemblages remain structurally similar between sites and provide some evidence of bottom-up productivity-driven trophic dynamics. To predict the possible landscape-scale processing of nutrient changes present for the aquatic ecosystems of the Kabompo River, a longitudinal comparison between a herbivore, Labeo cylindricus, and omnivore, Synodontis spp., fish species and their resources (primary producers and invertebrates) was done and showed archetypal fish food web trophic separation (2 ‰ to3 ‰) between species which remains consistent for the headwater sites. The nutrient values change at the convergence between the Kabompo Bridge and the Mwinilunga branches and show a decrease in the trophic separation of the sites below. This change becomes apparent from the last site, Watopa, where the trophic separation re-establishes (2 ‰ to 3 ‰) between the species. The trend suggests primarily autochthonous production in the headwater reaches, changing to allochthonous and autochthonous nutrient incorporation after the convergence of the two main branches. Permanent wetlands surrounding the convergence zones may be conducive to more primary producer activity and increased nutrient turnover. Permanent wetlands are a common feature of the Upper Zambezi Catchment tributaries, with the most notable lentic system the Barotse Flood Plains in the south, which shares reach-adjacent characteristics with the Kabompo River. Trends from the 2015 dry season identified for the Barotse Flood Plains support the findings of this thesis, where the δ13C values recorded reach depletion of -45 ‰. Management in the Upper Zambezi Basin and the Kabompo River comprises biannual assessments of water quality and quantity by the Zambezi River Authority, but not of the ecosystem process. This thesis may provide information to address the ecological (food webs and nutrient cycling) dynamics of the Basin rivers. - Description: The landscape of the Upper Zambezi Catchment in the remote North-Western Province of Zambia retains many natural features, but current and future anthropogenic activities threaten its integrity. Lack of information relating to the basic ecological functioning of the region hampers conservation efforts. Flooding from the adjacent woody-savanna and open grasslands in the Upper-Zambezi Catchment in the wet season drives crucial processes in the aquatic ecosystems such as the Kabompo River. This thesis describes aquatic food webs and describes the nutrient cycling in this river. A general introduction was based on the seasonality, river morphology, and biodiversity of the Kabompo River. There is no aquatic food web information available on this river. To address this knowledge gap, sampling of the principal food web components– vegetation, invertebrates, and fish – were collected in the dry season from August to September 2019 across six sites in the Kabompo River. Stable isotope analyses provided proxies for the food web structure at each site, and were the basis of longitudinal comparisons. The analyses suggest that the Kabompo River food webs follow the trophic ordination and nutrient cycling characteristic of the revised-Riverine Productivity Model. Food webs and community assemblages remain structurally similar between sites and provide some evidence of bottom-up productivity-driven trophic dynamics. To predict the possible landscape-scale processing of nutrient changes present for the aquatic ecosystems of the Kabompo River, a longitudinal comparison between a herbivore, Labeo cylindricus, and omnivore, Synodontis spp., fish species and their resources (primary producers and invertebrates) was done and showed archetypal fish food web trophic separation (2 ‰ to3 ‰) between species which remains consistent for the headwater sites. The nutrient values change at the convergence between the Kabompo Bridge and the Mwinilunga branches and show a decrease in the trophic separation of the sites below. This change becomes apparent from the last site, Watopa, where the trophic separation re-establishes (2 ‰ to 3 ‰) between the species. The trend suggests primarily autochthonous production in the headwater reaches, changing to allochthonous and autochthonous nutrient incorporation after the convergence of the two main branches. Permanent wetlands surrounding the convergence zones may be conducive to more primary producer activity and increased nutrient turnover. Permanent wetlands are a common feature of the Upper Zambezi Catchment tributaries, with the most notable lentic system the Barotse Flood Plains in the south, which shares reach-adjacent characteristics with the Kabompo River. Trends from the 2015 dry season identified for the Barotse Flood Plains support the findings of this thesis, where the δ13C values recorded reach depletion of -45 ‰. Management in the Upper Zambezi Basin and the Kabompo River comprises biannual assessments of water quality and quantity by the Zambezi River Authority, but not of the ecosystem process. This thesis may provide information to address the ecological (food webs and nutrient cycling) dynamics of the Basin rivers. Factors influencing the number of non-timber forest product species used at the individual, household and village levels in Vhembe, Limpopo Province, South Africa

NTFPs contribute significantly to livelihoods and living standards by offering a variety of socioeconomic benefits such as food, health, income generation, and safety nets. These multiple benefits are provided by a wide diversity of plant, animal and fungal species in different settings. However, the precise number of NTFP species used depends on the scale of assessment, namely use by individuals, by households and by villages. Consequently, it is difficult to compare numbers of species used between studies because of the differing scales. As a result, the objective of this study was to determine the number of NTFP species used at three different scales and how each is correlated with appropriate scale-dependent contextual variables. This objective was met by sampling individuals and whole households in five villages in the Vhembe District in Limpopo Province, South Africa. Twenty households were randomly selected in each village, providing a total sample of 100 households and 179 individuals in those households. In each household, every adult member was interviewed individually about the NTFPs that they used or knew. The major questions covered the NTFP species used, the respondents' socioeconomic level, the NTFP species used previously, and if the number of NTFP species are changing in the landscape. Data were reported per individual as well as per household and per village. The household level was derived by adding the responses of all the adults in each household but excluding duplicates. The same approach was used at the village scale, namely, the data across the 20 households were pooled, and duplicates were omitted. Patterns of NTFP use were examined by type of NTFP, and analysis using standard statistical tests, along with a Principal Components Analysis and a multivariate Detrended Correspondence Analysis of the NTFP species composition across villages. A total of 275 species of NTFPs were mentioned across the entire sample, including 81 species of firewood, 48 species of edible insects, 43 species of wild vegetables, 40 species of edible fruits, 38 species of wood utensils, 28 species of medicinal plants, 23 species of thatch grass, 19 species of poles, 19 species of wood for carvings, nine species of reeds, eight species of mushroom, and eight species of fodder. Of these species, some species had more than one use. Approximately 90% of the species used by the respondents were native species. Wild vegetables (e.g., Corchorus olitorius, Amaranthus capensis, Bidens pilosa), firewood (e.g., Colophospermum mopane, Parinari curatellifolia, Combretum hereroense), wild fruit (e.g., Landolphia kirkii, Vangueria infausta), and edible insects (e.g., Imbrasia belina, Macrotermes michaelseni, Encosternum delegorguei) were the most often used NTFPs by communities in the Vhembe area. Most of the households relied on NTFPs for subsistence. At the scale of an individual, the mean number of NTFP species used was 26.8±12.6 across the five villages. When considering a household, the mean number was 36.1±10.7 species. When aggregated across the 20 households per village, the mean number per village was 105.2±34.1 species. The species accumulation curve showed that 20 households per village were reasonably representative for four of the five villages. The results from the principal component analysis (PCA) and correlation analysis revealed that age, education, and employment status were significantly associated with the number of species used by individuals, whereas gender showed no clear correlation. At the household scale, household size was positively related with the number of species used, but income level and income source were not. At the village scale, attributes such as proximity to a town, settlement type, vegetation type, and distance to a nearby savanna all had some influence on the number of NTFP species listed. Most of the respondents felt that the number of species used had increased over the last 10–15 years, although some stated the opposite. Increases were attributed by traditional councils bans, the establishment of protected areas, and restoration. Activities eroding the number of NTFP species were identified as clear felling for farming, climate change, and over-harvesting. Integrating music and mathematics for connecting across multiple constructs of fractional understanding: an RME task design journey

- Description: Two South African curricular aims: appreciating creativity in mathematics and developing conceptual understanding, motivated this study. Negative views towards mathematics and challenges in teaching and learning fractions at primary school level are reported in literature, with the part-whole construct of fractions often the sole teaching focus. Despite challenges in curriculum integration (high demands on teachers and diluting disciplines), benefits, such as motivation and creative thinking, are noted. I recognised music-mathematics integration as an opportune context for designing tasks to support learners in moving flexibly between the fraction as ratio, fraction as measure and part-whole constructs. Guided by Realistic Mathematics Education principles, I embarked on a participatory dual-design experiment in task design, grappling within three micro-Communities of Practice (micro-CoPs) and across two planes: the Design-Theorising Plane and the Grounded-Practice Plane. In the Design-Theorising Plane, I worked with my two doctoral supervisors, grappling with design obstacles and finding resolutions. COVID-19 restrictions shifted our meetings to online platforms, allowing documentation and analysis of the task design process through recording functions. In the Grounded-Practice Plane, I worked within two separate micro-CoPs, both at independent schools (eight and two participating teachers respectively). Data on the teachers’ interrogation and implementation of the designed tasks were obtained via formal and informal interviews. Their reflections informed ongoing adaptations to the task design. Data were analysed in a matrix I designed and via NVivo coding. Findings include both the product of the task design journey (eight music-mathematics lessons, resources, and representations) and the process (ten groupings of Obstacle-Resolution Cycles). Three key questions (relating to music-mathematics fidelity; to task simplification for implementation; and to appropriate music-mathematics representation) were used in addressing each Obstacle-Resolution Cycle. Designing tasks to teach the part-whole construct of fractions was relatively straightforward, but designing tasks to teach the fraction as ratio and fraction as measure constructs was more challenging. These constructs could not be conflated by superimposing the music and mathematical linear representations. Aligning them, however, allowed for moving flexibly between the constructs. However, there seems to be contradictions between the intended curriculum, the enacted curriculum and the attained curriculum. This disjuncture is exacerbated in part by the fact that science teachers seem to be struggling to be cultural knowledge brokers. It is against this backdrop that this formative interventionist study sought to leverage a peer-learning community and expert community members in the integration of IK into the learning and teaching of Grade 10 Chemistry on the rate of reactions. To achieve this, we mobilised the indigenous technologies of preserving and pounding Mahangu and making Oshikundu to mediate learning of the rate of reactions. The study was guided by the broad overarching research question: How does a peer-learning community and expert community members leverage the integration of indigenous knowledge into the learning and teaching of Grade 10 Chemistry on the rate of reactions? In this study, I used two complementary paradigms, viz. the transformative research paradigm and the indigenous research paradigm. Within these paradigms, I employed a qualitative case study research design using the community of practice and participatory action research as research approaches. Five Grade 10 Chemistry teachers from three schools in the Ohangwena region were involved in this study. Data were generated through semi-structured interviews, co-analysis of curriculum documents, workshop presentations and discussions, practical demonstrations, participatory observation, lesson observation, stimulated recall interviews, and participants’ reflections. Vygotsky’s sociocultural theory and Shulman’s Pedagogical Content Knowledge (PCK) were employed as theoretical frameworks in this study. Additionally, within PCK, Mavhunga and Rollnick’s Topic-Specific Pedagogical Content Knowledge components were used as an analytical framework. I used an inductive-deductive approach to data analysis to come up with sub-themes and themes. The main finding of this study revealed that leveraging a peer-learning community and the expert community members (ECMs) empowered the Chemistry teachers involved in this study to be cultural knowledge brokers and their understanding of how to integrate IK in their teaching improved. Both their subject matter knowledge and pedagogical content knowledge improved through co-developing and enacting exemplar lessons that integrated IK from the expert community members as well as from their own environments. A main insight of this study is that Chemistry teachers should seek opportunities to create peer-learning communities that engage with expert community members who are the custodians of the cultural heritage. Pharmaco-chemical investigation of Erythrina caffra: extracts, isolated compounds and their biological activities

- Description: In this study, secondary metabolites isolated from Erythrina caffra, a medicinal plant indigenous to South Africa, were investigated. E. caffra is well-known for its healing properties and it is traditionally used for treating bacterial infections like tuberculosis (TB), abscesses, tooth aches and ear infections. Its extracts have also been used to treat cancer. Though many studies have been done on this plant, most of them tended to focus solely on the isolated compounds. In the present study however, extracts, fractions and isolated compounds from E. caffra were evaluated for their anticancer, anti-oxidant, anti-enzymatic, antibacterial and cytotoxicity. The methanol crude extract (B1) from the stem bark of E. caffra was used to extract alkaloidic fractions (B2 and B3) using ethyl acetate and n-butanol respectively, a third fraction (B4) was also extracted using ethyl acetate this fraction was called a neutral fraction. The neutral fraction (B4) was fractionated and through a sequence of column chromatography three active secondary metabolites were isolated. The isolated compounds included Lupeol (1), stigmasterol (2) and 5,7-Dihydroxy-4'-methoxy-3',5'-diprenylflavanone (3). These isolated compounds were characterized and identified using spectroscopic techniques including IR, NMR and high-resolution Mass Spectrometry. Using the cell line HCC-70, isolated from a primary ductal carcinoma, in vitro anticancer assays were carried out on the crude extract from the bark, fractions, isolated compounds and an unseparated mixture of two compounds. These samples were also evaluated for their anti-oxidant, anti-enzymatic, antibacterial and cytotoxicity activities. The crude extract inhibited the cell viability by over 30% and had no effect on the HeLa cells at concentrations of 20μM. Abyssinone V’ 4-methyl-ether (3) and the mixture of stigmasterol (2) and an unidentified compound exhibited potent anticancer activity against the HCC-70 cell line with IC50 of 18.05μM and 9.04μM respectively. Antibacterial assays were also carried out on the crude extracts, fractions and concoctions made from the fractions with the best activity combined with the ones that performed poorly. The concoctions were prepared as two separate series (S and N series). The crude extract inhibited more than 80% of the Staphylococcus aureus cells at a concentration of 20μM with only minimal damage to the HeLa cells. In the concoctions however, the N series managed to inhibit over 96% of the S. aureus while exhibiting no cytotoxicity towards HeLa cells. The extract and its fractions also showed good anti-oxidant activities. Molecular docking of these compounds was done on the Human estrogen receptor (PDB ID:3ERT) and Abyssinone V’ 4-methyl-ether (3) showed the best docking score of -6.6 Kcal/mol, for the simulation against Epidermal growth factor receptor (PDB ID: 1M17) Stigmasterol (2) showed the best docking score of -3.8 Kcal/mol. In silico docking on 3ERT and 1M17 were done to test the binding affinity of the isolated compounds to the proteins which are well known to be overexpressed in some types of cancer. Flavonoids isolated from Erythrina species have been reported to possess good antiplasmodial activity. However, due to the minute amounts isolated in the present study in-vitro assays could not be carried out. Nevertheless, in-silico assays were conducted on the most prominent protozoal parasite which causes malaria in the majority of African countries. In-silico simulations were done against Plasmodium falciparum protein (PDB ID: 7KJH), of the tested compounds Abyssinone V’ 4-methyl-ether (3) was found possess the best docking score of -4.4 Kcal/mol. The molecular docking of 7KJH was done to assess the inhibitory potential of the isolated compounds on protozoal parasites. Pharmacokinetic properties of the isolated compounds were also assessed in silico to assist in evaluating the drug likeness of these compounds. The compounds showed a percent human oral absorption of 100% except for Abyssinone V’ 4-methyl-ether (3), which showed 93.83%, this indicates a remarkable oral bioavailability. Stigamsterol (2) exhibited a Caco-2 cell permeability (QPPCaco) greater than 500 which indicates outstanding results for good intestinal absorption. The compounds also displayed a blood-brain partition co-efficient (QPlogBB) ranging from -1.433 to 0.128 suggesting they will have less potential to cross the blood-brain barrier, thus reducing any CNS related toxicity. Molecular networking of the crude extracts and the fractions was done through GNPS which allowed the identification of known compounds including one isolated in the present study, Abyssinone V’ 4-methyl-ether (3). - Description: In this study, secondary metabolites isolated from Erythrina caffra, a medicinal plant indigenous to South Africa, were investigated. E. caffra is well-known for its healing properties and it is traditionally used for treating bacterial infections like tuberculosis (TB), abscesses, tooth aches and ear infections. Its extracts have also been used to treat cancer. Though many studies have been done on this plant, most of them tended to focus solely on the isolated compounds. In the present study however, extracts, fractions and isolated compounds from E. caffra were evaluated for their anticancer, anti-oxidant, anti-enzymatic, antibacterial and cytotoxicity. The methanol crude extract (B1) from the stem bark of E. caffra was used to extract alkaloidic fractions (B2 and B3) using ethyl acetate and n-butanol respectively, a third fraction (B4) was also extracted using ethyl acetate this fraction was called a neutral fraction. The neutral fraction (B4) was fractionated and through a sequence of column chromatography three active secondary metabolites were isolated. The isolated compounds included Lupeol (1), stigmasterol (2) and 5,7-Dihydroxy-4'-methoxy-3',5'-diprenylflavanone (3). These isolated compounds were characterized and identified using spectroscopic techniques including IR, NMR and high-resolution Mass Spectrometry. Using the cell line HCC-70, isolated from a primary ductal carcinoma, in vitro anticancer assays were carried out on the crude extract from the bark, fractions, isolated compounds and an unseparated mixture of two compounds. These samples were also evaluated for their anti-oxidant, anti-enzymatic, antibacterial and cytotoxicity activities. The crude extract inhibited the cell viability by over 30% and had no effect on the HeLa cells at concentrations of 20μM. Abyssinone V’ 4-methyl-ether (3) and the mixture of stigmasterol (2) and an unidentified compound exhibited potent anticancer activity against the HCC-70 cell line with IC50 of 18.05μM and 9.04μM respectively. Antibacterial assays were also carried out on the crude extracts, fractions and concoctions made from the fractions with the best activity combined with the ones that performed poorly. The concoctions were prepared as two separate series (S and N series). The crude extract inhibited more than 80% of the Staphylococcus aureus cells at a concentration of 20μM with only minimal damage to the HeLa cells. In the concoctions however, the N series managed to inhibit over 96% of the S. aureus while exhibiting no cytotoxicity towards HeLa cells.
