- Description: The extant coelacanths have a close resemblance to the coelacanth fossil records dating back to 230mya. Like their predecessors, the extant coelacanths inhabit rocky caves at a depth of 100-300m below sea level. In the Comoros, the water temperature at these depths is estimated to fluctuate between 14-20°C. High-level adaptation to these environment and lack of competition are thought to have led to the morphological uniformity and slow change throughout the history of the coelacanths. Under stress conditions, proteins unfold or misfold leading to the formation of aggregates. Molecular chaperones facilitate the correct folding of other proteins so that they can attain a stable tertiary structure. In addition, molecular chaperones aid the refolding of denatured proteins and the degradation of terminally misfolded protein after cellular stress. Heat shock proteins form one of the major classes of molecular chaperones. Here we show that, despite high-level adaptation to a unique habitat and slow change, the genome of the coelacanth encodes the major and highly conserved molecular chaperone, Hsp70. Latimeria menadoensis and Latimeria chalumnae contain intronless hsp70 genes encoding Hsp70 proteins archetypal of known Hsp70s. Based on the coelacanth codon usage, we have shown that bacterial protein expression systems, particularly Escherichia coli, may not be appropriate for the overproduction of coelacanth Hsp70s and coelacanth proteins in general. Also interesting, was the discovery that like the rat Hsc70, the L. menadoensis Hsp70 could not reverse thermal sensitivity in a temperate sensitive E. coli DnaK mutant strain, BB2362. We also report the successful isolation of a 1.2 kb region of L. menadoensis hsp70 upstream regulatory region. This region contain three putative heat shock elements, a TATA- box and two CAAT-boxes. This regulatory region resembled the Xenopus, mouse, and particularly tilapia hsp70 promoters, all of which have been shown to drive the expression of reporter genes in a heat dependent manner. Taken together, this data is the first to strongly suggest an inducible Hsp70-base cytoprotection mechanism in the coelacanth. It further provides basis to formulate testable predictions about the regulation, structure and function of Hsp70s in the living fossil, Latimeria.

Management of invasive aquatic weeds with emphasis on biological control in Senegal

- Description: In 1985 the Diama Dam was built near the mouth of the Senegal River to regulate flows during the rainy season and prevent the intrusion of seawater during the dry season. This created ideal conditions upstream of the dam wall for invasion by two highly invasive aquatic weeds, first by water lettuce Pistia stratiotes Linnaeus (Araceae) in 1993, and then by salvinia Salvinia molesta D.S. Mitchell (Salviniaceae) in 1999. This study was focused on the management of P. stratiotes and S. molesta. Following successes that were achieved elsewhere in the world, biological control programmes involving two weevil species were inaugurated against both weeds and research was focused on several aspects. These included pre-release studies to determine the weevils' host-specificity and impact on the plants in the laboratory, their subsequent mass-rearing and releases at selected sites and post-release evaluations on their impact on the weed populations in the field. Both programmes, which reprepresented the first biocontrol efforts against aquatic weeds in Senegal, proved highly successful with severe damage inflicted on the weed populations and complete control achieved within a relatively short time span. A laboratory exclusion experiment with N. affinis on P. stratiotes showed that in treated tubs, the weevil strongly depressed plant performance as measured by the plant growth parameters: mass, rosette diameter, root length, number of leaves and daughter plants whereas control plants were healthy. Field releases started in September 1994 and water coverage by P. stratiotes at Lake Guiers was reduced by 25% in January 1995 and 50% in April 1995. A general decline of 65% in water coverage by P. stratiotes was observed in June 1995 and by August 1995, eight months after releases P. stratiotes mats were destroyed. Further, although no releases were made there, good results were obtained within 18 months at Djoudj Park water bodies, located 150 km NW from Lake Guiers indicating the potential of the weevil to disperse long distances. In 2005, P. stratiotes reappeared and the weevil N. affinis has located and controlled all of these P. stratiotes recurrences after new releases. In 1999, S. molesta covered an estimated area of 18 000 ha on the Senegal River Left Bank and tributaries (Senegal) and 7 840 ha on the Senegal River Right Bank (Mauritania). Military and Civil Development Committee (CCMAD) and community volunteers made an effort to control S. molesta using physical removal, but this costly and labour-intensive approach was unsustainable. Hence, biological control was adopted by Senegal and Mauritania to manage the weed. Host range tests to assess feeding by C. salviniae on S. molesta and non-target plants and carried out on 13 crop species showed that no feeding damage was observed on the latter and weevils only fed on S. molesta. Field releases of some 48 953 weevils at 270 sites were made from early January 2002 to August 2002. Within one year, weevils were established and were being recovered up to 50 km from the release sites. In a case study conducted at one of the release sites, the S. molesta infestation was reduced from 100% to less than 3% 24 months after release. These results are discussed in the context of the weeds’ negative impact on aquatic systems and riverside communities, and in the involvement of these communities in the programmes.

Molecular characterisation of the chaperone properties of Plasmodium falciparum heat shock protein 70

- Description: Heat shock protein 70 (called DnaK in prokaryotes) is one of the most prominent groups of chaperones whose role is to prevent and reverse protein misfolding. PfHsp70 is a heatinducible cytoplasm/nuclear localised Plasmodium falciparum Hsp70. PfHsp70 is thought to confer chaperone cytoprotection to P. falciparum during the development of malaria fever. The objective of this study was to examine the chaperone properties of PfHsp70 using a bioinformatics approach, coupled to in vivo and in vitro analysis. Structural motifs that qualify PfHsp70 as a typical Hsp70 chaperone were identified. Although PfHsp70 has a higher similarity to human Hsc70 than E. coli DnaK, in vivocomplementation assays showed that PfHsp70 was able to reverse the thermosensitivity of E. coli dnaK756 (a temperature sensitive strain whose DnaK is functionally compromised). Two residues (V401 and Q402) in the linker region of PfHsp70 that are critical for its in vivo function were identified. Constructs were generated that encoded the ATPase domain of PfHsp70 and the peptide binding domain of E. coli DnaK (to generate PfK chimera); and the ATPase domain of E. coli DnaK fused to the peptide binding domain of PfHsp70 (KPf). The two chimeras were tested for their ability to reverse the thermosensitivity of E. coli dnaK756 cells. Whilst KPf was able to reverse the thermosensitivity of the E. coli dnaK756 cells, PfK could not. Previously, PfHsp70 purification involved urea denaturation. Using a detergent, polyethylenimine (PEI), PfHsp70 was natively purified. Natively purified PfHsp70 had a basal ATPase activity approximately two times higher than the previously reported activity for the protein purified through urea denaturation. PfJ4, a type II Hsp40, could not stimulate the ATPase activity of PfHsp70 in vitro. Arch and hydrophobic pocket substitutions (A419Y, Y444A and V451F) were introduced in the PfHsp70 peptide binding domain. Similar substitutions were also introduced in the KPf chimera. PfHsp70-V451F (hydrophobic pocket mutant) had marginally compromised in vivo function. However, a similar mutation (V436F), introduced in KPf abrogated the in vivo function of this chimera. The arch and hydrophobic pocket derivatives of PfHsp70 exhibited marginally compromised in vivo function, whilst equivalent mutations in KPf did not affect its in vivo function. The ability of PfHsp70 and its arch/hydrophobic pocket mutants to suppress the heatinduced aggregation of malate dehydrogenase (MDH) in vitro was investigated. Whilst PfHsp70 arch mutants displayed marginal functional loss in vivo, data from in vitro studies revealed that their functional deficiencies were more severe. This is the first study in which an Hsp70 from a parasitic eukaryote was able to suppress the thermosensitivity of an E. coli DnaK mutant strain. Findings from the in vivo and in vitro assays conducted on PfHsp70 suggest that this protein plays a key role in the life-cycle of P. falciparum. Furthermore, this study raised insights that are pertinent to the current dogma on the Hsp70 mechanism of action.

Networking: enabling professional development and institutionalisation of environmental education courses in Southern Africa

- Description: This study was aimed at understanding how networking can enable or constrain professional development and institutionalisation of environmental education courses in southern Africa in the context of the Course Development Network (CDN), a project of the Southern Africa Development Community Regional Environmental Education Programme (SADC REEP). The study involved 12 institutions (each institution was represented in the CDN by a staff member) in eight SADC member states. It was contextualized through a review of social, political, environmental and educational developments in southern Africa, with specific reference to regionalization processes, as the SADC REEF is constituted under this post-colonial political framework. Relational philosophy informed the research process and methodology. The philosophy underpinned three distinct, yet related theoretical perspectives namely: critical realism, which provided the ontological perspectives of the study; Actor Network Theory; and Community of Practice, which provided the epistemological perspectives. Data was generated during a 33-month period in southern Africa and 12 months in the United Kingdom (and on a one week visit to Italy). During a 12 month Split Site Commonwealth Scholarship award, tenable at Manchester Metropolitan University in the United Kingdom, I studied the Environment and School Initiative (ENSI) and the Sustainability Education for European Primary Schools (SEEPS), to examine whether the same mechanisms that made them successful would apply to the CDN in the context of southern Africa. While these two case studies provided useful insight into the relationship between networking, professional development and institutionalization of environmental education programmes, they were not the main focus of the research. Data analysis was mainly through inductive, abductive and retroductive modes of inference. Inductive data analysis was done by means of Nvivo – a computer software package used for qualitative data analysis. The software aided in revealing features and relationships in the data in more depth as it allowed flexibility in working with data. Abduction is the interpretation of a phenomenon by means of a conceptual framework. In this study, I used Actor Network Theory (ANT) (Latour & Woolgar, 1979; Callon, 1986) and Community of Practice (COP) (Lave, 1988; Lave & Wenger, 1991; Fullan, 2003) as analytical conceptual frameworks to probe networking and professional development respectively. Data on institutionalisation of environmental education courses was analysed by means of retroductive mode of inference, which is a thought operation that enables the understanding of social reality beyond what is empirically observable or experienced. This study reveals that there were a number of necessary (internal) and contingent (external) factors that enabled or constrained networking, professional development and institutionalisation of environmental education courses in the context of the CDN. The key factors included existing cultural capital, donor political economy, power relations, poverty related factors and social transformation trends. Opportunity and constraint : historicity, hybridity and notions of cultural identity among farm workers in the Sundays River Valley

- Description: This thesis focuses on relationships of opportunity and constraint among farm workers in the Sundays River Valley (SRV), Eastern Cape Province. Relationships of 'constraint' include those experiences of displacement and forced removal and war, including forced removals by the apartheid state in 1960 and 1970. Relationships of 'opportunity' include the ways in which residents in the SRV have contested their experiences of upheaval and domination, and the formation of a regional sense of place and belonging/ investigate how farm workers actually draw elements of locality and identity from their experiences of upheaval, and how displacement bolsters feelings of belonging and place. Instead of viewing displacement as a once-off experience, this thesis investigates displacement in historical terms, as a long-term, 'serial' experience of human movement, which is continued in the present- specifically through the creation of the Greater Addo Elephant National Park. I concentrate on developing a spatialised and cultural notion of movemenUplacement. 'Place' is investigated as a term that refers to rather indeterminate feelings of nostalgia, memory and identity, which depend on a particular connection to territory (ie: 'space'). I emphasise that elements of place in the SRV are drawn from and expressed along dualistic lines, which juxtapose situations of opportunity and constraint. In this way, farm workers' sense of connection to farms and ancestral territory in the SRV depends on their experiences of stable residency and work on farms, as well as their memories of removal from land in the area. I emphasise that those elements of conservatism (expressed as 'tradition' and Redness) among Xhosa-speaking farm workers are indications of a certain hybridity of identity in the region, which depend on differentiation from other groups (such as so-called 'coloured' farm workers and 'white' farmers), as well as associations between these groups. This thesis lays emphasis upon those less visible and definable 'identities' in the Eastern Cape Province, specifically by shifting focus away from the exhomeland states of the Ciskei and Transkei, to more marginal expressions of identity and change (among farm workers) in the Province. I point out that labourers cannot solely be defined by their positions as farm workers, but by their place and sense of cultural belonging in the area. In this sense, I use the idea of work as a loaded concept that can comment on a range of cultural attitudes towards belonging and place, and which is firmly embedded in the private lives of labourers - beyond their simple socio-economic conditions of farm work. I use Bourdieu's conception of habitus and doxa to define work as a set of dispositions that have been historicised and internalised by workers to such an extent, that relationships of domination are sometimes inadvertently obscured through their apparent 'naturalness'. Moreover, I point out that work can be related to ritualised action in the SRV through the use of performance and practice-based anthropological theory. Both work and ritual are symbolic actions, and are sites of struggle within which workers express themselves dualistically. - Description: This thesis focuses on relationships of opportunity and constraint among farm workers in the Sundays River Valley (SRV), Eastern Cape Province. Relationships of 'constraint' include those experiences of displacement and forced removal and war, including forced removals by the apartheid state in 1960 and 1970. Relationships of 'opportunity' include the ways in which residents in the SRV have contested their experiences of upheaval and domination, and the formation of a regional sense of place and belonging/ investigate how farm workers actually draw elements of locality and identity from their experiences of upheaval, and how displacement bolsters feelings of belonging and place. Instead of viewing displacement as a once-off experience, this thesis investigates displacement in historical terms, as a long-term, 'serial' experience of human movement, which is continued in the present- specifically through the creation of the Greater Addo Elephant National Park. I concentrate on developing a spatialised and cultural notion of movemenUplacement. - Description: Literature suggests that a small number of family businesses are able to make the transition from the founder to other family members with a common reason cited being the lack of planning. This study aims to build understanding of leadership succession in family businesses by focusing on the influence of a founder's family business value set on the succession planning process in Black family-owned businesses. Using the Strauss and Corbin (1990) grounded theory method, this study develops a theory of succession planning of Black familyowned businesses labelled Passing the Spear which is comprised of three stages based on an analysis of 21 qualitative interviews. The Spear was an analogy used to represent both the values of the founder and the family business, thus in Passing the Spear, founders where essentially performing a dual transfer of their values and leadership to the successor. The implementation of the process Passing the Spear was influenced by the family business value set of the founder, which in this study, were labelled as Traditional, Progressive, and Transitional. These value sets were distinguished by their behaviour regarding their choice of successor where founders with a traditional value set exhibited gatekeeping behaviour, while the behaviour of founders with a progressive value set was labelled navigating, and finally, founders with a transitional value set demonstrated behaviour labelled exploring. Furthermore, it was found that after the successor had been chosen, founders appeared to follow a generic succession planning process, however, the ease and timing of implementation was influenced by the family business context, evidence of being proactive and the degree of family business resilience. The process of Passing the Spear comprised of three stages of (a) Showing the spear where the founder's focus is on the induction and socialization of the successor using the strategies of bringing the successor into the family business and managing the family-business interface using relationships; (b) Explaining the spear entailed founders using the strategies of sharing knowledge and teaching the successors about the family business; and ( c) Sharing the spear was where founders focused on empowering the successors by implementing strategies of sharing responsibilities and learning from the successor.The process provides insights into the influence of family business values on the succession planning process and can be useful for founders of Black family-owned businesses in planning for succession in their businesses. In addition, the study provides another perspective of succession planning and offers a contribution to the literature for understanding succession in Black family-owned businesses.

- Description: There has been an upsurge in the use of Complementary and Alternate Medicines (CAMs) in both developed and developing countries. Although herbal medicines have been in use for many centuries, their quality, safety and efficacy are still of major concern. Many countries are in the process of integrating CAMs into conventional health care systems based on the knowledge and use of traditional medicines. The quality control (QC) of herbal products usually presents a formidable analytical challenge in view of the complexity of the constituents in plant material and the commercial non-availability of appropriate qualified reference standards. Sceletium, a genus belonging to the family Aizoaceae, has been reported to contain psychoactive alkaloids, specifically mesembrine, mesembrenone, mesembrenol and some other related alkaloids. Sceletium is marketed as dried plant powder and as phyto-pharmaceutical dosage forms. Sceletium products and plant material marketed through health shops and on the internet are associated with unjustified claims of specific therapeutic efficacy and may be of dubious quality. Validated analytical methods to estimate Sceletium alkaloids have not previously been reported in the scientific literature and the available methods have focused only on qualitative estimation. Furthermore, since appropriate markers were not commercially available for use as reference standards, a primary objective of this study was to isolate relevant compounds, qualify them as reference standards which could be applied to develop appropriate validated qualitative and quantitative analytical methods for fingerprinting and assay of Sceletium plant material and dosage forms. The alkaloidal markers mesembrine, mesembrenone and ∆⁷ mesembrenone were isolated by solvent extraction and chromatography from dried plant material. Mesembranol and epimesembranol were synthesised by hydrogenation of the isolated mesembrine using the catalyst platinum (IV) oxide and then further purified by semi-preparative column chromatography. All compounds were subjected to analysis by ¹H, ¹³C, 2-D nuclear magnetic resonance and liquid chromatography-tandem mass spectroscopy. Mesembrine was converted to hydrochloride crystals and mesembranol was isolated as crystals from the hydrogenation reaction mass. These compounds were analysed and characterised by X-ray crystallography. A relatively simple HPLC method for the separation and quantitative analysis of five relevant alkaloidal components in Sceletium was developed and validated. The method was applied to determine the alkaloids in plant material and dosage forms containing Sceletium. An LCMS method developed during the study provided accurate identification of the five relevant Sceletium alkaloids. The method was applied for the quantitative analysis and QC of Sceletium plant material and its dosage forms. This LCMS method was found to efficiently ionize the relevant alkaloidal markers in order to facilitate their detection, identification and quantification in Sceletium plant material as well as for the assay and QC of dosage forms containing Sceletium. The chemotaxonomy of some Sceletium species and commercially available Sceletium dosage forms were successfully studied by the LCMS method. The HPLC and LCMS methods were also used to monitor the bio-conversion of some of the alkaloids while processing the plant material as per traditional method of fermentation. Additionally a high resolution CZE method was developed for the separation of several Sceletium alkaloids in relatively short analysis times. This analytical method was used successfully to fingerprint the alkaloids and quantify mesembrine in Sceletium and its products. Sceletium species grown under varying conditions at different locations, when analyzed, showed major differences in their composition of alkaloids and an enormous difference was found to exist between the various species with respect to the presence and content of alkaloids. Sceletium and its products marketed through health shops and the internet may thus have problems with respect to the quality and related therapeutic efficacy. The QC of Sceletium presents a formidable challenge as Sceletium plants and products contain a complex mixture of compounds. The work presented herein contributes to a growing body of scientific knowledge to improve the QC standards of herbal medicines and also to provide vital information regarding the selection of plant species and information on the specific alkaloidal constituents to the cultivators of Sceletium and the manufacturers of its products.

Oxidation of thiocyanate (SCN-), CYS and 2-ME was catalyzed by a selfassembled monolayer of cobalt tetraethoxythiophene Thiocyanate oxidation occurred via two electron transfer, whereas that of CYS and 2-ME required 1 electron. The oxidations of SCN- and 2-ME were catalyzed by ring based processes, while CYS was catalyzed by both Co[superscript III]/Co[superscript II] process and ring-based processes. Oxidation of GSH and 2-ME was conducted on screen printed graphite electrodes modified with cobalt phthalocyanine. Activity depended on method of electrode modification and CoPc % composition. Decomposition of Snitrosoglutathione occurred in the presence of copper ions and NaBH[subscript 4]. Reduced and oxidized glutathione were detected as products using cobalt phthalocyanine adsorbed on an ordinary pyrolytic graphite electrode. Reduction of oxygen was electro-catalyzed by adsorbed manganese phthalocyanine complexes on glassy carbon electrodes. FePc, FePc(Cl)[subscript 16], CoPc and CoPc substituted with phenoxypyrrole and ethoxythiophene ligands were also used as electro-catalysts. - Description: This thesis examines the 1998 complaint of racism against the Mail & Guardian, a leading exponent of South Africa's alternative press in the 1980s, and important contemporary producer of investigative journalism. The study is framed within a cultural studies approach, analysing the Mail & Guardian as constituted by a 'circuit of production': its social context, production, texts, and audiences. The thesis makes three main arguments. First, that the claim of racism cannot be understood outside of a consideration of both the changing political milieu, and subtle changes within the Mail & Guardian itself. Significant social changes relate to the reconfiguration of racial and class identities wrought by the 'Mbeki state'. Within the Mail & Guardian, the thesis argues for the importance of the power and subjectivity of the editor as a key 'factor' shaping the identity of the paper, evidenced in its production practices and textual outputs. In this regard, the thesis departs from a functionalist analysis of particular 'roles' within the newsroom, drawing instead on a post-structuralist approach to organisational studies. Based on this production and social context, the thesis examines key texts which deal with aspects of South Africa's social transformation, and which exemplify aspects of the Mail & Guardian's reporting which led to the complaint of racism by the Black Lawyers Association (BLA) and the Association of Black Accountants (ABASA). Their complaint was that the Mail & Guardian's reporting impugned the dignity of black people, and in so doing was a violation of their rights to dignity and equality which are constitutionally guaranteed. However, as freedom of the press is also guaranteed by the South African constitution, their complaint to the South African Human Rights Commission (SAHRC) resulted in public debate about these contending rights. My second argument relates to the jurisprudential approach to racism, and the related issue of affirmative action, which informed the complaint against the paper. Contrary to the 'normative', liberal approach to these issues, this thesis highlights Critical Race Theory as the jurisprudential basis for both the claimants' accusation of racism against the Mail & Guardian, and aspects of its implicit use in South African human rights adjudication. The thesis argues that in failing to recognise these different philosophical and political bases of legal reasoning, the media, including the Mail & Guardian, in reporting on these matters failed in their purported role of serving the public interest. The thesis concludes by applying Fraser's critique of Habermas's notion of a single, bourgeois public sphere to journalism, thereby suggesting ways in which the critiques of some of the Mail & Guardian's own journalists could be employed to enlarge its approach to journalism - giving voice to constituencies seldom heard in mainstream media.

- Description: This thesis examines the 1998 complaint of racism against the Mail & Guardian, a leading exponent of South Africa's alternative press in the 1980s, and important contemporary producer of investigative journalism. The study is framed within a cultural studies approach, analysing the Mail & Guardian as constituted by a 'circuit of production': its social context, production, texts, and audiences. The thesis makes three main arguments. First, that the claim of racism cannot be understood outside of a consideration of both the changing political milieu, and subtle changes within the Mail & Guardian itself. Significant social changes relate to the reconfiguration of racial and class identities wrought by the 'Mbeki state'. While the hydantoin-hydrolyzing enzymes have been studied in detail, the mechanisms that control expression of the hyu genes have not. The research reported in this work elucidates some of the mechanisms involved in the regulation of the hyu genes in A. tumefaciens strains. The hydantoin-hydrolyzing enzyme activity from the environmental isolate A. tumefaciens RU-AE01 was characterized. A broad host range vector for the simultaneous analysis of divergent promoters was constructed. The promoter regions responsible for the activation of transcription of hyuH and hyuC were identified by deletion analysis. It was proposed that transcription of hyuH was activated by a putative σ[superscript 54]-dependent promoter or a putative σ[superscript 70]-dependent promoter identified upstream of the hyuH gene. The hyuC gene was activated by a putative σ[superscript 70]-dependent promoter identified upstream of the hyuC gene. The regulation of hydantoinase and N-carbamyl amino acid amidohydrolase enzyme activity was compared to the regulation of transcription from the RU-AE01 hyuH-hyuC region. Expression of the hydantoin-hydrolyzing enzymes was regulated by induction which correlated with reporter enzyme expression from the hyuH and hyuC promoter regions. However, the expression of the hydantoin-hydrolyzing enzymes was also regulated by nitrogen catabolite repression (NCR). This did not correlate to the reporter gene expression of the hyuH promoter region but did compare to the reporter gene expression of the hyuC promoter region. This suggested that NCR of hyuH was at the post-translational level whereas NCR of the hyuC promoter was at the transcriptional level. Pathways involved in the regulation of the hyu genes were characterized. The production of the hydantoin-hydrolyzing enzymes in both A. tumefaciens strains RU-AE01 and RU-OR were regulated by proteins involved in the global ntr pathway. The levels of the hydantoin-hydrolyzing enzymes in strain RU-AE01 were elevated in the presence of increased levels of NtrB and NtrC illustrating the importance of the ntr pathway in the regulation of the levels of the hydantoin-hydrolyzing enzymes. Similarly, in RU-OR the presence of exogenous NtrB and NtrC elevated levels of N-carbamyl amino acid amidohydrolase activity. However, the levels of hydantoinase enzyme activity in strain RU-OR were elevated in the presence of NtrC alone. In addition, the presence of a His6-tagged NtrC molecule abolished the elevation in the levels of the hydantoinase but not the N-carbamyl amino acid amidohydrolase enzyme activity in strain RU-OR. This suggests that NtrC has a direct role in the regulation of the expression of hyuH in RU-OR. In addition, it indicates that the hyu genes in the two A. tumefaciens strains RU-AE01 and RU-OR are different. The presence of the RU-AE01 hyuH-hyuC fragment caused a dramatic increase in the hydantoin-hydrolyzing enzyme activity in strain RU-OR but not strain RU-AE01. This implied the incidence of a possible repressor protein in RU-OR, which is titrated out by the presence of the RU-AE01 hyuH-hyuC fragment. Protein-DNA binding assays suggest that this putative repressor may be 38 kDa in RU-OR cells.

- Description: This study investigates the temporal, spatial and impact characteristics of severe convective storm hazard and risk in the Eastern Cape Province of South Africa. Using historical data on severe convective storms dating from 1897, patterns of the hazard threat and risk to various geographic populations were investigated. A conceptual framework that emphasises the combined role hazard and vulnerability play in defining risk was used for the study. A methodology for ranking the severity of the storms in the historical dataset, based on recorded damage/impact, was specifically developed for the study. It is intended that this methodology will have a potentially wider application and may be adapted to a range of hazard impact and risk studies in South Africa and internationally. The study was undertaken within the context of the South African Disaster Management Act of 2002. Findings of the study show that severe convective storms can occur throughout the province, but there are clearly demarcated areas of higher frequency and concentration. The impact of storms is particularly severe on impoverished and vulnerable rural populations in the eastern parts of the province, where there is an urgent need for building capacity in disaster risk management. A major outcome of the study is the production of a severe convective storm hazard/risk map of the Eastern Cape, which it is hoped will be of benefit to a number of stakeholders in the province, particularly disaster management, but also the South African Weather Service, agricultural organisations, development/planning authorities, educational authorities and risk insurers. It is hoped that this map and the study in general will assist in guiding the operational responses of the various authorities, especially in terms of those interventions aimed at disaster risk reduction in the Eastern Cape.

- Description: Sperm structure in five species of cicadine cicadas (Albanycada albigera, Azanicada zuluensis, Platypleura capensis, P. hirtipennis and Pycna semiclara) and five species of cicadettine cicadas (Melampsalta leucoptera, Quintilia walkeri, Stagira simplex, Xosopsaltria thunbergi and Monomatapa matoposa) was investigated by light and electron microscopy. In addition, spermiogenesis in cicadas was described; the information was derived from two cicadettines (Diceroprocta biconica and M. matoposa) and three cicadines (Kongota punctigera, P. capensis and P. semiclara). Mature spermatozoa of all species investigated are elongate and filiform, consisting of three distinct regions: the head (acrosome and nucleus), mid-piece and tail. All species produce more than one discrete length of nucleated, motile sperm, a form of sperm polymorphism termed polymegaly. Polymegaly is expressed in three ways; sperm have uni-, bi- or trimodal nucleus and tail lengths. Besides the differences in length, there are also notable differences in the size of nuclei. The anterior parts of sperm heads are embedded in an elongate homogenous matrix forming spermatodesmata. The conical acrosome is deeply invaginated posteriorly, and sits on top of the nucleus. The acrosomal contents are differentiated internally with a tubular substructure and a subacrosomal space. The anterior of the nucleus intrudes into the posterior section of the subacrosomal space. Anteriorly the acrosome is laterally flattened; posteriorly it extends as two tubular processes on either side of the nucleus that gradually decrease in diameter. The homogenously electron-dense nucleus is pointed anteriorly and is generally cylindrical, although posteriorly there is a lateral invagination that extends part-way along the nucleus. This invagination houses fine granular material of the putative centriolar adjunct which does not form in close proximity to the centriole and hence may not be a true centriolar adjunct. The lamellate disposition of the centriolar adjunct material within the sperm-midpiece of cicadettine cicadas is distinct, and separates these cicadas from their cicadine counterparts in which the centriolar adjunct material is non-lamellate. Vesicle-like elements that are associated with both the posterior nucleus and the centriolar adjunct are also found within the invagination. Immediately posterior of and adjoining the centriolar adjunct is a pair of mitochondrial derivatives that are elongated and extend for almost the entire length of the tail. Except for size the architecture of short and long spermatozoa is generally similar in all species. The absence of accessory bodies in cicada sperm suggests that within the Cicadomorpha, the families Cicadidae and Cercopidae are closely related. Only long nuclei were observed in the fertilized eggs of A. zuluensis indicating that sperm with long nuclei might be favoured for fertilization. Spermiogenesis involves: (a) development of the acrosome from a proacrosomal granule; (b) development of the nucleus, characterized by elongation and streamlining with a simultaneous condensation of chromatin; (c) development of the axoneme from the centriole; (d) amalgamation of individual small mitochondria to form elongated mitochondrial derivatives in which cristae are arranged into regularly spaced lamellae; and (f) elimination of cytoplasm. The presence of a manchette, a transient microtubular organelle, which surrounds the acrosome, nucleus and mitochondrial derivatives, is a characteristic feature of spermiogenesis. The gross morphology of the reproductive tract in both male and female cicadas exhibits an organization similar to that in most oviparous insects. The non-functional spermatheca is the only exceptional feature in the female reproductive tract. Its role has been taken over by the common oviduct which, subsequently, has become modified into a swollen, differentiated structure with a dual role of receiving oocytes from the paired ovaries and storage of spermatozoa. Testis mass varies between cicada species; this variation might be linked to the intensity of sperm competition which has been found to be positively correlated with relative investment in spermatogenesis. Based on the preliminary findings of this study, K. punctigera, with its larger testis relative to body size, would be the ideal candidate to show the greatest levels of sperm competition. Accessory glands in both male and female A. zuluensis, D. biconica, P. hirtipennis and O. quadraticollis are very long; this character might be of phylogenetic significance. The interaction of differently sulphonated titanium and tantalum phthalocyanine complexes with methyl viologen (MV[superscript 2+]), and hence the stoichiometry and association constants are evaluated. Detailed photophysicochemical properties of the complexes were investigated and are for the first time presented with fluorescence lifetimes easily obtained from fluorescence quenching studies. The transformation of 1-hexene photocatalysed by aryloxy- and arylthio-appended complexes is also presented for the first time. The electrochemical properties of the complexes are unknown and are thus presented. Cyclic (CV) and square wave (SWV) voltammetries, chronocoulometry and spectroelectrochemistry are employed in the study of the complexes. Two oneelectron reductions and a simultaneous 4-electron reduction are observed for the unsubstituted Cl[subscript 3]TaPc. Reduction occurs first at the metal followed by ring-based processes. The tetra- and octa-substituted derivatives however exhibit peculiar electrochemical behaviour where a multi-electron transfer process occurs for complexes bearing certain substituents. For all complexes, the first two reductions are metal-based followed by ring-based processes. A comparative study of the electrocatalytic activities of the complexes towards the oxidation of nitrite is also investigated. The complexes are immobilised onto a glassy carbon electrode either by drop-dry or electropolymerisation methods. All the modified electrodes exhibit improved electrocatalytic oxidation of nitrite than the unmodified electrodes by a twoelectron mechanism producing nitrate ions. Catalytic currents are enhanced and nitrite overpotential reduced to ~ 0.60 V. Kinetic parameters are determined for all complexes and a mechanism is proposed. The purpose of this study was therefore to compare biological, morphological and genetic variation of South African Syngnathus species among different populations/locations, and with European populations of S. acus. Sixteen meristic and ten morphometric characteristics were quantified from specimens obtained from field as well as various international natural history museum collections. Univariate (ANOVA) and multivariate (principal component analysis and discriminant analyses) analyses were used to assess morphological differences among the species. Morphometric variables were adjusted as ratios of the standard length and using an allometric procedure. ANCOVA analysis indicated significant differences between S. acus and S. temminckii for the relationships of the standard length (SL) and all morphological characters. There was no significant correlation between SL and snout length, snout depth, inter-orbital width and trunk depth for S. watermeyeri. The analyses provided evidence for distinct populations of S. acus, S. temminckii and S. watermeyeri although morphological character differentiation was greater between S. watermeyeri and the other two larger species. Although, significant differences were observed for meristic characters, pairwise comparisons did not reflect a clear pattern of variability. Most of the measured morphological characters contributed more than 70% to the morphological variation between the populations. Plot of the canonical scores for the variables resulted in the specimens clustering according to species groups and locations of S. temminckii. Sequences of 750 base pairs of the mitochondrial cytochrome gene from 11 localities were compared with published sequences of other species of Syngnathus. Phylogenetic analysis was performed using parsimony, maximum likelihood (ML) and Bayesian inference (BI). The South African species were revealed to be sister-taxa with about 6 % divergence, while S. temminckii and S. acus had about 11% sequence divergence. 20 haplotypes among 46 total specimens from the three species. Gene flow was estimated at approximately 3 migrants per generation between the two South African populations and about 1 per generation between S. temminckii and S. acus. Such strong stock structuring among presumably recently established post-Pliocene (< 2 Million years ago) populations suggests that these species are reproductively isolated. Morphological and genetic variation observed in this study combined with current knowledge of life history attributes of the South African pipefishes indicate that conservative management decisions are necessary until the patterns and extent of differentiation among populations species-wide can be investigated further. It is thus being proposed that the name of the South African population of S. acus be changed to Syngnathus temminckii (Kaup, 1856).

This gives substantial credibility to the information provided by the companies and highlights the importance of selecting appropriate methods. There are some variances between the research results and established literature on the development and transfer of core competencies. There are notable differences between the companies from South Africa and the US multinational on the effectiveness of methods used to develop and transfer core competencies. There are also areas where all the companies are in accord. "Roadmaps" covering the effectiveness of methods for developing and transferring core competencies in different locations in a value chain are presented. The microbial ecology of sulphidogenic lignocellulose degradation

- Description: Acid mine drainage is a well known environmental pollutant, not only in South Africa, but throughout the world, and the use of microbial processes in the treatment of these wastes has been the subject of investigation over past decades. Lignocellulose packed-bed reactors have been used in passive treatment systems, and, although effective initially, they show early decline in performance while the packing material remains largely un-utilized. Little is known about this phenomenon which remains a severe constraint in the development of efficient passive mine water treatment systems. It has been proposed that the degradation pathways of the complex lignocellulose substrate may be limited in some way in these systems during the manifestation of this effect. This study has addressed the problem using a molecular microbial ecology methodology in an attempt to relate trophic functions of the microbial population to the physico-chemical data of the system. A field-scale lignocellulose packed-bed reactor located at Vryheid Coronation Colliery (Northern Kwa-Zulu Natal province, South Africa) was monitored for six years and the results showed the classic profile of performance decline related to a slowdown in sulphate reduction and alkalinity production. The reactor was decommissioned , comprehensive samples were collected along the depth profile and the microbial populations investigated by means of 16S rRNA gene methodology. The population was found to include cellulolytic Clostridia spp., CytophagaIFlavobacterlBacteroidetes, Sphingomonadaceae and as yet uncultured microorganisms related to microbiota identified in the rumen and termite gut. These are all known to be involved as primary fermenters of cellulose. Oesulphosporosinus was present as sulphate reducer. A comparison of substrata sampling and population distribution suggested that spatial and temporal gradients within the system may become established over the course of its operation. Based on these findings, a laboratory-scale reactor was constructed to simulate the performance of the packed-bed reactor under controlled experimental conditions. The laboratory-scale reactor was operated for 273 days and showed comparable performance to that in the field in both biomolecular and physicochemical data. Clearly defined trophic niches were observed. These results suggested that a sequence of events does occur in lignocellulose degradation over time. Based on the spatial and temporal column studies, a descriptive model was proposed to account for these events. It was found that fermentative organisms predominate in the inlet zone of the system using easily extractable compounds from the wood, thus providing feedstock for sulphate reduction occurring in the succeeding compartments. Production of sulphide and alkalinity appears to be involved in the enhancement of lignin degradation and this, in turn, appears to enhance access to the cellulose fraction. However, once the readily extractables are exhausted, the decline in sulphide and alkalinity production leads inexorably to a decline in the overall performance of the system as a sulphate reducing unit operation. These observations led to the proposal that with the addition of a limited amount of a readily available carbon source, such as molasses, in the initial zone of the the reactor, the ongoing generation of sulphide would be sustained and this in turn would sustain the microbial attack on the lignocellulose complex. The Rhodes BioSURE process and the use of sustainability indicators in the development of biological mine water treatment

- Description: Polluted waters, arising from extensive past and ongoing mining operations in South Africa, pose serious environmental threats to the limited fresh water resource. The long time periods, of decades to centuries, over which decanting mine waters may be expected to flow raises additional concerns about the sustainability of these resources. Responses to the problem have thus increasingly been directed towards the long-term sustainability of mine water treatment technologies (MWTT) as a critical indicator in both their research and development, and application. Bioprocess treatments have been considered in this regard and, among these, the Rhodes BioSURE Process has been investigated in preliminary studies using complex organic carbon wastes as the carbon source and electron donor for the central sulphate reduction unit operation. Although both the mining industry and the related statutory/regulatory authority in South Africa share public commitment to sustainability in the treatment of mine waters, no systematic mechanism has emerged to enable the application of sustainability thinking as a guiding principle in the selection and application of MWTTs, nor in the research and development undertaking. This study undertook the development of a Sustainability Indicator Framework in order to provide a systematic basis for the incorporation of sustainability objectives in MWTT bioprocess development, and specifically to use this framework as an input to the investigation of the scaleup development of the Rhodes BioSURE Process. In the development of the MWTT Sustainability Indicator Framework, an initial survey of industry thinking in this area was undertaken and, based on these outcomes, a detailed questionnaire methodology was developed in order to identify and quantify critical sustainability indicators. These included analysis of environmental, economic, social and technical indicators used in sustainability accounting practice in the industry. Statutory/regulatory sustainability targets in the same categories were derived from State of the Environment Reports (SoER) from Provincial authorities where mining is undertaken in South Africa. A synthesis of industry and SoER values was derived from weighted averages and the Sustainability Indicator Framework based on these outcomes. A Conceptual Decision-Support System, to guide the selection and development of MWTTs, was proposed and also based on these results. In the development of the Rhodes BioSURE Process the use of primary sludge (PS) had been investigated as a potential complex carbon and electron donor source. In this regard the utility operator, and sewage treatment process infrastructure, was identified as potentially meeting aspects of the sustainability objectives identified for MWTT application development. Both the Sustainability Indicator Framework and the Conceptual Decision-Support System provided inputs in the formulation of the experimental programme relating to the scale-up development of the Rhodes BioSURE Process. Based on these outcomes, a series of single- and multi-stage reactor configuration, optimisation and enzymology studies were undertaken at bench-, pilot- and technical-scale operations. These units were operated at hydraulic retention times (HRT) ranging between 22 to 72 hours and at chemical oxygen demand to sulphate ratios (COD:SO[subscript 4]) ranging between 1:1 to 2:1. Studies undertaken in fed-batch, bench-scale reactors confirmed the preliminary feasibility of using established sewage treatment infrastructure as a replacement for novel reactor configurations that had been used in the initial studies. The results further indicated that the hydrolysis of PS occurred at different rates under biosulphidogenic conditions in the different reactor configurations investigated. Scale-up of these findings in multi-stage pilot- (7.4m[superscript 3]) and technical-scale plants (680m[superscript 3]) showed comparable performances between the unit operations in terms of SO[subscript 4] and COD removal. These results indicated no apparent advantages in the uncoupling of hydrolysis and sulphate reduction in separate unit operations as had been suggested in previous studies. Scale-down/scale-up studies were undertaken in a continuously fed single-stage reactor configuration and showed that the process could be effectively operated in this way. Previous proposals that chemical and biological gradients established in the sludge bed of the Recycling Sludge Bed Reactor (RSBR) exercised an influence on the rates of substrate hydrolysis were investigated and the relative activity of α- and β-glucosidase and protease enzymes was measured. Results provided additional support for this hypothesis and it was shown that enzyme assay may also provide a useful tool in process development and monitoring studies. While sulphide recovery, following the sulphate reduction step in the BioSURE Process, was not investigated as a component of this study, the treatment of final effluent or waste spills was identified as an important sustainability requirement given the toxicity of sulphide to human and ecosystem environments. A conventional trickle filter reactor system was evaluated for this purpose and showed close to 100% oxidation to sulphate in a short contact time operating regime. Although residual COD removal was low at ~20% of influent, it is considered that high rate recycle biofilter operation could achieve the COD discharge standard of 75 mg/l. The results of the above studies provided inputs into the design, construction and commissioning of the first full-scale commercial application of the Rhodes BioSURE Process for mine wastewater treatment using sewage sludge as the carbon and electron donor source. An adjacent mine and sewage works have been linked by pipeline and an operational capacity of 10 Ml/day water treated has been established with sulphate reduced from ~1300mg/l to <200mg/l. These developments constitute a novel contribution in the mine waste water treatment field.

- Description: Polluted waters, arising from extensive past and ongoing mining operations in South Africa, pose serious environmental threats to the limited fresh water resource. The long time periods, of decades to centuries, over which decanting mine waters may be expected to flow raises additional concerns about the sustainability of these resources. Responses to the problem have thus increasingly been directed towards the long-term sustainability of mine water treatment technologies (MWTT) as a critical indicator in both their research and development, and application. Bioprocess treatments have been considered in this regard and, among these, the Rhodes BioSURE Process has been investigated in preliminary studies using complex organic carbon wastes as the carbon source and electron donor for the central sulphate reduction unit operation. Although both the mining industry and the related statutory/regulatory authority in South Africa share public commitment to sustainability in the treatment of mine waters, no systematic mechanism has emerged to enable the application of sustainability thinking as a guiding principle in the selection and application of MWTTs, nor in the research and development undertaking. This study undertook the development of a Sustainability Indicator Framework in order to provide a systematic basis for the incorporation of sustainability objectives in MWTT bioprocess development, and specifically to use this framework as an input to the investigation of the scaleup development of the Rhodes BioSURE Process. In the development of the MWTT Sustainability Indicator Framework, an initial survey of industry thinking in this area was undertaken and, based on these outcomes, a detailed questionnaire methodology was developed in order to identify and quantify critical sustainability indicators. These included analysis of environmental, economic, social and technical indicators used in sustainability accounting practice in the industry. The role of achievement motivation on the interlanguage fossilization of middle-aged English-as-a-second-language learners

It has been suggested that all second language (L2) learners, in the process of mastering a target language (TL), develop a linguistic system that is self-contained and different from both the learner's native language and the TL. This system is referred to as 'interlanguage' (lL). In the process of SLA, IL evolves into an ever-closer approximation of the TL, and ideally, a learner's IL should continue to advance until it becomes equivalent to the TL. However, it has been observed that somewhere in the L2 learning process, IL may reach one or more plateaus during which the development of the IL is delayed or arrested. A permanent cessation of progress toward the TL is referred to as 'fossilization'. Researchers in SLA agree that motivation is one of the key factors influencing language-learning success and studies suggest that some language learning motivation may be related to the need for achievement. The purpose of this research was to establish if adult ESL learners are aware of fossilization and, to examine if motivation, and more specifically achievement motivation, is a factor in IL fossilization. The participants in this study consisted of 15 ESL learners in Puerto Rico who had at least eight years of formal ESL training. The instrument used to gather information included a questionnaire to obtain demographical and qualifying data, an 'English Language Proficiency Evaluation' to determine levels of IL fossilization, a 'Measure of Achievement Motivation' to ascertain achievement motive, and individual and group interviews in order to ascertain perception(s) regarding the role of motivation on fossilization and perceptions regarding the barriers to achieving TL competency. The research demonstrated that there is a moderate to strong positive relationship between IL fossilization and achievement motivation, i.e., high achievement motive is correlated to TL competency and descending levels of achievement motive are correlated to ascending levels of IL fossilization. The findings have significant implications for both ESL learning and instruction, and suggest that not all IL fossilization may be permanent.

The role of ants in structuring insect communities on the canopies of senegalia drepanolobium near Laikipia, Kenya

- Description: In the black cotton ecosystem of Laikipia, Kenya, four symbiotic ants coexist at a fine spatial scale on canopies of Senegalia drepanolobium. They exhibit different aggressive behaviours and modify their tree canopies differently. These diverse behaviours were expected to affect the associated canopy arthropod communities. At the Kenya long-term exclosure experiment (KLEE) and its immediate environs at Mpala Research Centre, Laikipia, the insect communities coexisting with each of the four ant species were characterized, and their response to different vertebrate herbivory. Other ant species inhabiting the tree canopies or the ground were surveyed too. Pitfall trapping was used in sampling terrestrial ants, while beating and mist-blowing were used in collecting arboreal insects. Different sampling methods had varying efficacies, revealing the importance of using several methods. There are at least sixteen ant species in this ecosystem, all occurring on the ground, but only ten species on the trees. Terrestrial ant communities in this ecosystem cannot be used as indicators of grazing pressure for range management. A total of 10,145 individual insects were collected from the tree canopies, comprising of 117 species from seven orders and 25 families, forming a complex community of species interacting at different levels. Symbiotic ant species had a significant effect on insect community structure and composition. Crematogaster sjostedti was associated with a community that was significantly different from the other ant species. There was no significant effect of vertebrate feeding pressure on the canopy insect community, but there was an interaction effect between ant species and treatments. Significant differences between ant species mostly occurred on treatment plots where only cows were allowed to graze. One or more of the ant species may be a keystone species in this ecosystem even though experimental manipulations failed to confirm earlier findings. It was concluded that the one-year period during which experimental manipulations were carried out was not long enough to reflect takeover effects on the insect community. The four symbiotic ant species colonizing S. drepanolobium comprises of two guilds, the hemipteran-tending ants (C. sjostedti and Crematogaster mimosae) and non-tending ants (Crematogaster nigriceps and Tetraponera penzigi). Communities associated with these guilds were found to be significantly different in all four diversity indices. The black cotton ecosystem is species-poor compared to other ecosystem such as forests. The number of insect species that colonizes S. drepanolobium and coexists with acacia-ants forms a large proportion of the invertebrate community. Therefore, this ecosystem should be conserved to safeguard this invertebrate community. This will also give scientists a chance to establish how the various insect species coexist with symbiotic ants on tree canopies.
