In vitro pharmacological screening of thiazolidinedione-derivatives on diabetes and Alzheimer’s potential therapeutic targets

- Description: There is an increased prevalence of diabetes and other non-communicable diseases in Sub-Saharan Africa and globally. In South Africa, the prevalence of type 2 Diabetes mellitus is currently estimated at 9.0% in people aged 30 and older and is expected to increase. Diabetes-related complications result in acute alterations in the mental state due to poor metabolic control as well as greater rates of decline in cognitive functioning with age, higher prevalence of depression and increased risk of Alzheimer’s disease. Alzheimer’s disease is the most common form of dementia in older adults and possibly contributes to 60 - 70% of cases. Alzheimer’s disease remains incurable, its progression inevitable with the currently available symptomatic therapies being palliative while the treatment of diabetes relies on insulin preparations and other glucose-lowering agents. Current treatment options have numerous side effects such as hypoglycaemia, diarrhoea, weight gain and abnormal liver function. This has geared the investigation of new generations of small molecules which exhibit improved efficacy and safety profiles. On this basis, several studies have shown that thiazolidinediones and their corresponding derivatives exhibit a broad spectrum of biological activities including anti-inflammatory, and antioxidant activities. Furthermore, recent evidence from experimental, epidemiological, and clinical studies highlight the utility of antioxidants for treating diabetes and its complications. Interestingly, there is increasing evidence that links diabetes and Alzheimer’s disease due to their pathophysiology and suppressing glycaemia has been shown to be beneficial in Alzheimer’s disease treatment. Accordingly, the aim of this study, was to evaluate the anti-diabetic and anti-Alzheimer’s properties of four novel synthesized thiazolidinedione-derivatives owing to their antioxidant properties. Methods The aim of this study was achieved through performing ferric reducing antioxidant power activity, 2,2’-Diphenyl-1-Picry Hydrazyl radical scavenging activity, α-amylase inhibition, α-glucosidase inhibition, aldose reductase inhibition, protein tyrosine phosphatase-1B inhibition, dipeptidyl peptidase-4 inhibition, acetylcholinesterase inhibition, matrix metalloproteinase-1 inhibition, and β-amyloid aggregation inhibition assays. In addition, peroxisome proliferator-activated receptor-γ activation was performed through docking studies. To establish physicochemical properties of TZD derivatives investigated, further in-silico studies were done using SwissADME tools. Results To this end, in-vitro and in-silico studies were successfully performed. In-silico ADME profiling predicted these derivatives to be drug-like with moderate to good solubility in the GI and not blood-brain barrier permeable. Furthermore, docking of these molecules against PPARγ predicted a similar mode of action to that of thiazolidinediones using Rosiglitazone as the standard drug with TZDD2 and TZDD4 forming equivalent conformations to that of Rosiglitazone in the same binding site and TZDD3 having an equivalent LBE to that of Rosiglitazone (-8.84 and -8.63kcal/mol respectively). In-vitro evaluation predicted a moderate antioxidant activity with TZDD2 and 3 exhibiting the highest FRAP activity and DPPH radical scavenging activity. Furthermore, enzymatic inhibition assays showed a relative inhibition activity with TZDD3 exhibited > 100% inhibition in concentrations ≥ 30 μg/mL and TZDD1, 2 and 4 exhibited ≥ 50% inhibition activity in all the concentrations (10, 20, 30, 40 and 50 μg/mL) in the α-amylase inhibition assay. Similarly, in the α-glucosidase inhibition assay, all the four derivatives exhibited a concentration dependent activity with TZDD3 showing the most activity. All the four derivatives exhibited ≥ 30% inhibition in the aldose reductase inhibition assay except TZDD1 at 10 μg/mL. TZDD4 exhibited a concentration dependent inhibition activity in the protein tyrosine phosphatase-1B inhibition assay. Interestingly, TZDD3 showed a decreasing inhibition activity as its concentration increased from 10 μg/mL through to 50 μg/mL. In the dipeptidyl peptidase–4 inhibition assay, TZDD2 and TZDD4 exhibited ≥ 20% inhibition activity across all the concentrations and in the acetylcholinesterase assay, TZDD1, 3 and 4 exhibited ≥ 25% across all the concentrations. Interestingly, in the matrix metalloproteinase-1 inhibition assay, some of these derivatives exhibited partial activation activity and partial inhibition with TZDD1 showing activation in concentrations 10 and 20 μg/mL and inhibition in concentrations 30, 40 and 50 μg/mL. TZDD4 showed activation in all the concentrations. In the β-amyloid aggregation assay, all the four derivatives showed inhibition activity ≥ 10% except TZDD1 at 50 μg/mL. Effects of climatic variability on maize productivity in South Africa from 1937-2018

- Description: Climate is an important factor in agricultural production. The world is facing climate change and variability, which result in high temperatures, low rainfall patterns, shortage of water and persistent droughts. Climate change poses a significant threat to South Africa’s water resources, food security, health, infrastructure, ecosystem services and biodiversity. Negative impacts of climate variability on agriculture, especially on maize the staple crop, will worsen the food security status of the nation as most of South Africa’s maize crop is produced in summer and highly depends on rainfall. This study attempted to assess the impact of climate on maize production in South Africa using secondary time series data for the period 1937 to 2018. Rainfall and temperature were used as proxies for climate variability. The Granger Causality Model was used to examine the causal linkages between climatic variables (temperature or rainfall) and maize output in South Africa for the study period. The major outcome of the analysis was that there is a two-way causal relationship between maize production and temperature. The results also indicated that there is uni-directional causality between maize yield and rainfall. Furthermore, the Variance Decomposition Model was used to forecast the relationship between climatic elements and maize production in South Africa. The result showed that all variables have an effect on maize yield, with temperature having the least effect. The last objective of the study was to profile the maize output trend for the period from 1937 to 2018. The main findings from the analysis indicate that maize production in South Africa has a general upward slope. The study recommends that the government should intensify the provision of irrigation systems for the farmers in the most vulnerable areas to mitigate the climate change. Government should also embark on massive campaigns using a variety of media to create the needed public awareness on climate change and its impact on food security. , Thesis (MSc) -- Faculty of Science and Agriculture, 2022

Water is a basic need that must be availed to humans, plants and animals at any given time. It is a requirement that each human on earth should get between 20-50 L of fresh, clean and pure water a day. It is very difficult to keep up with this need due to drastic increase in global pollution by emerging contaminants. The increase is due to human activities, modern way of life, industrialization and improper disposal of materials that contain these OPFRs. Organophosphate flame retardants OPFRs are the organic pollutants considered in this study. OPFRs are semi volatile compounds SVOCs that are used in many industries as flame retardants and additives. They were developed because of environmental and health concerns of previously used brominated and chlorinated flame retardants FRs. They show acute toxicity and exhibit adverse effects such as neurotoxicity, they are also carcinogenic to humans and animals. In recent years, some researchers have started investigating the occurrence and distribution of OPFRs in South African rivers. The aim of this study was to identify signatures of OPFRs in the aquatic environment of Buffalo River Estuary, and soil from a nearby dumpsite: and to develop a method for their removal using modified cyclodextrins. In this study, the concentration of OPFRs were investigated in sediments and soil samples collected from Buffalo River Estuary. This estuary is located close to a major port in East London city in the Eastern Cape Province, South Africa. Collection of samples was done across two seasons i.e. winter and spring. OPFRs in sediment and soil samples in this study were extracted using ultrasonic extraction method. Solid-phase extraction SPE was employed for the cleanup of the extracts, in order to reduce potential matrix interference and also for sample purification. The congeners of interest in this study were Tris 2-Choroethyl phosphate TCEP and Tris 1,3-Dicloro-2-propyl phosphate (TDCP). Samples were air dried immediately on arrival at the laboratory after being stored in an ice chest at 4 ℃ and further processed for further analysis.The concentrations of OPFRs were determined using GC-μECD (gas chromatograph coupled with micro electron capture detector) from soil and sediments in that area and they were found high in winter compared to spring owing to high level of discharge of industrial and domestic waste into the aquatic system and also the nearby dumpsite. The total concentrationof OPFRs Ʃ 2OPFRs for sediments and soil for both seasons was 39935±7619 μg/g and 10376±601 μg/g, respectively. The dominant congener in the Buffalo River Estuary was TCEP. The ecological risk assessment was estimated for the study area, and TCEP moderate risk while TDCP presented low risk. From this study, it could be inferred that the Buffalo River Estuary and the nearby dumpsite in East London are polluted by emerging pollutants i.e. OPFRs. Hence, strict control measures should be put in place to monitor all activities contributing to such magnitude of pollution in the estuary, which is of great economic value to the country. Furthermore in this study, an insoluble and hydrophilic polymer EDTA-Cysteine-β-Cyclodextrin was prepared and synthesized for the remediation of OPFRs in soil and sediments. - Description: Urban gardening in townships is a common coping strategy employed by poor inhabitants to improve food security, earn income through sales, and generate cost-savings through home consumption of fresh produce. Food security is a cornerstone of human development and welfare, and the achievement thereof remains a global challenge. The majority of academic research and policy documentation traditionally positions food insecurity as a rural phenomenon. However, recent trends point to an increase in food insecurity in urban areas, particularly in low-income areas such as townships. In South Africa, rapid urbanization has led to burgeoning townships and rapidly expanding low-income informal settlements characterised by poverty, unemployment, and food insecurity. Research concerned with the role that urban agricultural production plays in contributing to the food security of poor households is scarce and not well understood. This research, therefore, aimed to contribute to the limited research which specifically examines urban food security, and the role urban agriculture play in the achievement of household food security. The study employed collective action theory and utility theory to firstly provides an in-depth examination of the determinants of participation in urban gardening using Probit regression modeling. The second research objective was to perform an impact evaluation of participation in urban gardening on selected household food security indicators including Household Dietary Diversity Scores (HDDS), the Household Hunger Scale (HHS) and the Coping Strategy Index (CSI) using Propensity Score Matching (PSM). Purposive and snowball sampling methods were employed to obtain a sample of 60 urban gardeners and a control group of 55 non-gardening households from Extension 6, 7 and 10 townships as well as Joza Location in Makhanda East, Eastern Cape Province. Households were surveyed using a structured household food security questionnaire. A focus group discussion was held with the Linomtha Community Garden members and key informant interviews were undertaken with important stakeholders such as the local extension officers. Descriptive analysis revealed that urban gardening households (UGs) had older household heads who were less educated, and that land and water availability were the dominant constraints to increases in agricultural production. Results from the probit and average marginal effects models showed that the likelihood of participation significantly increased when household unemployment and dependency increased and that households who were engaged in off-farm economic activity were more likely to participate in urban gardening. The impact evaluation was performed using PSM and Average Treatment Effect on the Treated (ATT). Analysis illustrated that participation in urban gardening does significantly reduce both the full CSI and Reduced CSI. Thus, participation in urban gardening improves the food security of participating households by significantly reducing the frequency of participating households (UGs) applying undesirable coping strategies such as sending children elsewhere other than the household, borrowing food from other households and reducing the number of meals eaten in a day due to food shortages. This study posits that urban gardening alone will not eradicate the rampant food insecurity and poverty which pervades in South Africa’s townships. However, urban gardening participation does contribute to the construction of a sustainable, urban livelihood by reducing the number and severity of the undesirable coping strategies that food insecure households employ by providing some nutritious produce as well improved social capital through gardening networks and supporting institutions. It is recommended that policy makers, at a national and municipal level, need to create a clearer directive for the integration of urban food production into the urban food marketplace thus increasing access to income generating channels for small-scale home, and community gardeners. Co-operation between private and public institutions for food security and urban gardeners needs to be fostered and awareness of opportunities to participate in urban gardens needs to be improved. - Description: Urban gardening in townships is a common coping strategy employed by poor inhabitants to improve food security, earn income through sales, and generate cost-savings through home consumption of fresh produce. Food security is a cornerstone of human development and welfare, and the achievement thereof remains a global challenge. The majority of academic research and policy documentation traditionally positions food insecurity as a rural phenomenon. However, recent trends point to an increase in food insecurity in urban areas, particularly in low-income areas such as townships. In South Africa, rapid urbanization has led to burgeoning townships and rapidly expanding low-income informal settlements characterised by poverty, unemployment, and food insecurity. Research concerned with the role that urban agricultural production plays in contributing to the food security of poor households is scarce and not well understood. This research, therefore, aimed to contribute to the limited research which specifically examines urban food security, and the role urban agriculture play in the achievement of household food security. The study employed collective action theory and utility theory to firstly provides an in-depth examination of the determinants of participation in urban gardening using Probit regression modeling. The second research objective was to perform an impact evaluation of participation in urban gardening on selected household food security indicators including Household Dietary Diversity Scores (HDDS), the Household Hunger Scale (HHS) and the Coping Strategy Index (CSI) using Propensity Score Matching (PSM). Purposive and snowball sampling methods were employed to obtain a sample of 60 urban gardeners and a control group of 55 non-gardening households from Extension 6, 7 and 10 townships as well as Joza Location in Makhanda East, Eastern Cape Province. Households were surveyed using a structured household food security questionnaire. The paper employed a qualitative research technique and the explanatory research design to achieve its desired objectives. The results of the paper confirmed that skills training and development strategy, when implemented by institutions, ensures that staff is empowered, informed, and taught, which results in better efficiency and effectiveness. In this situation, spin-offs will benefit public organizations since employees’ increased abilities open new opportunities for long-term employment growth and organizational effectiveness. Furthermore, the knowledgeable and well-trained individual would make sound organizational decisions while successfully listening to the clients' problems. The benefits will flow down to the areas where these organizations are headquartered, and the improved image will be the goal for the ECDoE. The conclusion drawn is that Skills Development Act aims to provide skills to the South African workforce, ensure that employees have more opportunities for skill acquisition, provide opportunities for new entrants to gain work experience. NPM was developed in order to enhance performance and service delivery within the public sector through adopting principles of the public sector. The nature and process of the activities within the public sector and the traditional public administration have been seen as problematic. As such, the birth of NPM is grounded in the need to offer more efficient mechanisms for service delivery. This study aimed at assessing the implementation of NPM principles towards effective service delivery within the Eastern Cape government. The study was underpinned by the New Public Management Approach. A qualitative desktop review was adopted to find answers to research questions of the study. The data was evaluated qualitatively through thematic analysis. The study findings reveal that the Eastern Cape government is making strides to effectively implement NPM principles towards improving service delivery. The implemented principles are privatisation, managerialism, performance management, decentralisation and customer oriented. The effectiveness of strategies aimed at enhancing effective implementation of NPM principles for improved service delivery was also assessed. The findings show that the strategies are effective although there are some inherent issues that derail effective implementation. Poor reporting, corruption, poor management, shortage of trained managers and staff shortages were identified as challenges affecting effective implementation of NPM principles. Water governance and social equity in South Africa: a case study of Amathole District Municipality.

However, despite the new policy reforms and strategies adopted, inequities in water governance remain prevalent. Those residing in marginalised and poor rural areas are the most vulnerable and highly affected by this phenomenon. While this has been attributed to several factors, such as fragmentation of the water sector and lack of capacity, among other reasons, an analysis of the literature revealed that the absence of a water governance framework for social equity is highly prevalent in the context of Amathole District Municipality (ADM) where the research was conducted. Hence, the study's main objective was to develop a water governance framework for social equity that can be utilised to advise councils and policymakers on the attainment of social equity by water service providers at the local level. The study utilised the constructivist research philosophy by adopting a qualitative case study research design and an inductive research approach to address the research questions. Focus group discussions, semi-structured interviews, participant observations and document analysis were used to collect data. A total sample of thirty-four (n=34) participants was purposefully selected; twenty participants (n=20) participated in semi-structured interviews, while fourteen (n=14) participants were engaged through focus group discussions. Findings obtained from the thematic data analysis utilised revealed that while human rights principles and social equity values underpin South Africa’s water reforms, Amathole District Municipality is yet to fully absorb these foundational concepts into its water governance. The study revealed that this is largely attributed to a lack of meaningful participation, limited transparency and accountability in the processes and procedures of the municipality. Results also revealed that persistent inequities still exist in Amathole District Municipality as reflected by the unequal distribution of water services, inconsistency in services provision, lack of transparency and established procedures to guarantee procedural fairness and limited impact on interventions specifically in rural areas. Furthermore, the study revealed that the municipality’s efforts to address water inequities are highly undermined by various economic, environmental, socio- economic, and institutional governance factors. In South Africa, the QTL in secondary school remains low and compromised despite the implementation of EFA policy and use of EDOs in South Africa generally and King Williams Town (KWT) in particular. The drive of this study is to assess the role played by EDOs in supporting QTL in selected secondary schools in King Williams Town District. The study used a qualitative approach, a case study design was followed and the case in point for this study is EDOs in KWT district. Their roles where assessed between two secondary schools namely Enoch Sontonga high school and Nosizwe high school. Ten (10) interviews using face-to face and telephonic method. The study managed to conduct five face-to-face interviews among EDOs as they were conveniently located during a period of workshops in the circuit and two educators and three principals were interviewed electronically. Purposive and convenience sampling techniques were used to select participants. Using the Popper’s theory of learning perspective the study found out that quality teaching and learning in KWT schools is dependent on factors such as time on task, scope of learning, learning environment, teaching methods and strategies, support and monitoring programmes, the teacher preparedness and their motivation and availability of attractive incentives to retain quality practitioners in their classrooms. The study found out that presently the EDOs are performing various roles such as offering support to the SMT through helping them understand their function within the school and the broader education system and helping in developing systems for monitoring and recording progress made by learners towards achievement of targets set. However, there are challenges encountered by EDOs when supporting schools to improve the QTL that were discussed namely, lack of human resources and material resources; educator resistance; non-academic activities; learner and teacher absenteeism; programmes alignment; and work overload. These challenges can be addressed using strategies such as human resources training and recruitment; programmes coordination and stakeholder cooperation; transport provision; consequence management; attitude change and policy alignment and implementation. Using the Popper’s theory of learning perspective the study found out that quality teaching and learning in KWT schools is dependent on factors such as time on task, scope of learning, learning environment, teaching methods and strategies, support and monitoring programmes, the teacher preparedness and their motivation and availability of attractive incentives to retain quality practitioners in their classrooms. The study found out that presently the EDOs are performing various roles such as offering support to the SMT through helping them understand their function within the school and the broader education system and helping in developing systems for monitoring and recording progress made by learners towards achievement of targets set. However, there are challenges encountered by EDOs when supporting schools to improve the QTL that were discussed namely, lack of human resources and material resources; educator resistance; non-academic activities; learner and teacher absenteeism; programmes alignment; and work overload. These challenges can be addressed using strategies such as human resources training and recruitment; programmes coordination and stakeholder cooperation; transport provision; consequence management; attitude change and policy alignment and implementation. - Description: This research centres on the holistic assessments of spatial and temporal dimensions of soil erosion zones based on the parameters of geomorphometry, hydro-statistics, and land use/cover dynamics. The study used a case study approach based on a clustered framework model of soil erosion parameters in the Tyume River basin in Eastern Cape, South Africa. The methods adopted for the investigation are, namely; non-parametric time-series assessment of streamflow dataset, semidecadal assessment of land use/cover (LU/C) dynamics, geospatial analysis of geomorphometric variables, vulnerability analysis of soil erosion zones, regression analysis of determination coefficient, and Receiver Operating Characteristic Curve (ROC). The delineation of soil erosion zones was based on the integrated analysis of the parameters of geomorphometry, geology, hydrology, and land use/ cover. The result of the hydro-statistical analysis of the Tyume River reports a major decline in the inter-annual regime frequency of storm flow based on the Mann- Kendall (MK) test and Sen’s slope assessment in 1992 (p-value = 0.031), 1997 (p-value = 0.045), 2003 (p-value = 0.021), 2008 (p-value = 0.003), and 2016 (p-value = 0.002). The MK test depicted a recurrence of peak streamflow acceleration in every three years based on low-flow and highflow transition. The sensitivity of LU/C to temporal dynamics of streamflow trends shown by the coefficient of correlation of trends of the LU/C water bodies with streamflow semi-decadal acceleration indicates a moderately relevant relationship, R = 0.76. The temporal analysis of LU/C and hydro-statistical analysis shows that the Tyume basin was highly vulnerable to soil erosion by water in 1999, 2009, and 2019. The vulnerability of the Tyume River basin in 2019 is exceptional and this is due to the conversion of forested area (woodland) into a built-up environment and farmland, with a high vulnerability in 2019 due to the slump in the rate of change of woodland and precipitation, and the increase in the rate of built-up and agricultural activities. The soil erosion vulnerability mapping divides the river basin into the critical high, high, moderate, low, nonvulnerable zones that cover 40 km2, 135 km2, 209 km2, and 186 km2 respectively. Regression analysis shows that the areas of soil erosion in the Tyume basin are moderately represented by the model (R2 = 56) while the model performance assessment based on success rate and prediction rate estimation from the area under the ROC curve shows that the model is good, Area Under Curve of the ROC = 0.899, and 0.897. The analysis suggests that soil erosion is driven by the impact of land use/land cover change, particularly in areas of high drainage density. Significantly, high vegetation density played a vital role in lowering high-flow on the hill-slope and low topographic wetness area as well as in areas with erodible geologic properties. The study, therefore, recommends the advocacy of crop rotation method of agricultural practice in the highly critical areas of soil erosion and recommends the development of riparian forests around the Tyume River. Development and optimisation of a qPCR assay for the enumeration of Cryptophlebia leucotreta granulovirus (CrleGV) used for commercial applications

- Description: The citrus industry contributes significantly to the South African agricultural sector. Thaumatotibia leucotreta (Meyrick) (Lepidoptera: Tortricidae) is highly important to the South African citrus industry as it is classified as a phytosanitary pest by most international markets. Thaumatotibia leucotreta has caused an estimated annual loss of up to R100 million to the industry. In order to control T. leucotreta in South Africa, an integrated pest management (IPM) programme has been used. One of the components of this programme is Cryptophlebia leucotreta granulovirus (CrleGV), which has been formulated to a registered biopesticide namely Cryptogran and has been successfully applied in the field for over 15 years. To use CrleGV as biopesticides, quantification of the viral particles is required to perform bioassays for field trials and formulation, among other applications. Darkfield microscopy is a traditional method used for the quantification of CrleGV; however, the method is characterised as being subjective, tedious, labour intensive, and time-consuming. This study aims to develop and optimise a qPCR technique to accurately quantify CrleGV-SA OBs using plasmid DNA for downstream applications. Firstly, lef-8, lef-9, and granulin conserved genes from CrleGV-SA and CrleGV-CV3 genome sequences were analysed by performing multiple alignments to evaluate the degree of identity between these genes. This was done to design two sets of oligonucleotides (internal and external) from regions with the highest identity. Subsequently, in silico testing was done to evaluate the designed oligonucleotides to determine whether they specifically bind to the selected target regions. Secondly, three sets of DNA plasmids (pJET1.2-Gran, pJET1.2-lef-9, and pJET1.2-lef-8) were constructed, each containing a target region for either granulin, lef-9, and lef-8 genes for use as standards in a downstream qPCR assay. This was achieved by first extracting gDNA from CrleGV-SA OBs and using the gDNA as a template to PCR amplify the target regions of the selected gene regions with the designed oligonucleotides. Subsequently, the PCR amplified regions were then directly ligated into the pJET1.2/blunt vector, and the plasmids were confirmed by colony PCR, restriction enzyme digestion, and Sanger sequencing. Thirdly, two different methods of CrleGV-SA gDNA extraction were compared to determine which method has the best yields in terms of concentration. The extraction methods compared were the Quick-DNA Miniprep Plus kit according to manufacturer’s instructions (Method 1a), pre-treatment with Na2CO3 prior to using the Quick-DNA Miniprep Plus kit (Method 1b), pre- treatment with Na2CO3, and neutralisation with Tris-HCl prior to gDNA extraction using the Quick-DNA Miniprep Plus kit (Method 1c) and the CTAB method (Method 2). The gDNA concentration and purity for all samples were determined using a Nanodrop spectrophotometer. Method 1c (Na2CO3 and Tris-HCl pre-treated plus Quick-DNA Miniprep Plus kit) was the most efficient at extracting genomic DNA compared with the other methods, resulting in the highest DNA concentration in short processing time. Fourthly, plasmid standards were evaluated for use in the qPCR assay. This was done as it was important to consider the efficacy of the oligonucleotides; including the ability of the oligonucleotides to anneal to the appropriate segment of DNA without extensive formation of oligonucleotides dimers, non-specific annealing, or formation of secondary structure. In addition, it was done to ensure that highly accurate standard curves were generated. The standard curves were to be utilised in the downstream qPCR assay to determine the quantity of test samples by interpolation, reading from the values within the standard curve. Lastly, darkfield microscopy and qPCR methods of enumeration were compared to verify their accuracy and determine the most consistent and comparable method. This was achieved by quantifying the purified, crude-purified, and viral formulated CrleGV-SA suspensions using these methods. Subsequently, a statistical analysis was conducted to compare the results produced by the two enumeration methods. The obtained results showed that the granulin, lef- 8 and lef-9 qPCR values did not significantly differ from the darkfield microscopy results. The findings of this study revealed that the two assays, lef-8 qPCR and lef-9 qPCR, were more robust, sensitive, and efficient for the quantification of CrleGV-SA. Thus, this study has successfully developed a qPCR assay that is comparable with the traditional darkfield microscopy counting technique. This is the first study to use the qPCR technique to enumerate CrleGV-SA using plasmid standards. Assessment of cytotoxic artemisinin and its derivatives as DNA damaging inducing agents in triple-negative breast cancer cells

- Description: In developing countries, including South Africa, breast cancer is the primary cause of cancer-related deaths among women. TNBC (triple-negative breast cancer) is an aggressive breast cancer subtype that is more prevalent in women of African descent. This subtype lacks the key receptors, namely the estrogen receptor (ER-), progesterone receptor (PR-), and human epidermal growth factor receptor 2 (HER2-) that are the basis of successful targeted therapies for other subtypes of the disease. To date, there are no effective, standardized targeted therapies for TNBC. Artemisinin is an anti-malarial drug and numerous derivatives of the compound have been developed to improve the potency and solubility of the parent compound. Artemisinin and its derivatives have gained attention as potential anti-cancer agents; however, such studies have not yet progressed to clinical trials and the precise mechanism of action of these compounds is yet to be fully explained. In this study, artemisinin, and its known derivative artesunate, as well as a novel derivative, WHN11, were investigated as DNA damage-inducing agents in TNBC. WHN11 was found to be the most potent of the three compounds, displaying an IC50 of 3.20 μM against HCC70 cells, artemisinin displayed an IC50 of 214.70 μM and artesunate displayed an IC50 of 25.48 μM. The compounds were less toxic to the MCF12A non-cancerous cells, with IC50 values 298.30, 87.53, and 8.35 μM for artemisinin, artesunate, and WHN11, respectively, and displayed selectivity indices of 1.39, 3.44 and 2.61 μM for artemisinin, artesunate, and WHN11, respectively. In silico and in vitro studies revealed that the artemisinin compounds bind to DNA through the minor groove. While all three compounds were able to bind to DNA, a comet assay revealed that only artemisinin and artesunate, and not WHN11, were able to cause DNA damage compared to the vehicle control, DMSO. Finally, a topoisomerase I (TOPO I) enzyme assay demonstrated that while the compounds appeared to display a degree of inhibition of TOPO I, as evidenced by a downward shift in the plasmid band on the agarose gel, they were not able to fully inhibit the enzyme to return the plasmid to the supercoiled conformation. In addition, combination studies revealed that artemisinin, artesunate, and WHN11 acted synergistically in combination with camptothecin, but displayed either an additive (artemisinin) or antagonistic (artesunate and WHN11) relationship when used in combination with etoposide. In conclusion, artemisinin, its known derivative artesunate, and novel and highly toxic derivative WHN11, all bind to DNA via the minor groove, however only artemisinin and artesunate, and not WHN11, cause DNA damage, indicating a potentially different mechanism of action of the three artemisinins. All three compounds act synergistically with camptothecin, which suggests interference with topoisomerase activity, partially supported by slight inhibition of TOPO I activity, and could indicate either direct inhibition of the enzyme or interference with enzyme function by competitive binding to the DNA. - Description: The research presented in this thesis is based on my interest and experience in forms of African music, ethnomusicology, and studying mbira with Professor Emeritus Andrew Tracey. When I arrived in South Africa in 2019 to join Rhodes University’s Applied Ethnomusicology programme, I chose to study the mbira with Tracey as the idea of learning more about Zimbabwe through music was important to the formation of my identity. Through the lens of embodied learning and a practice-based approach in this research, I evaluate how Tracey’s numerous contributions to African music pedagogy have improved prospects for African music scholars and students in terms of contributing to the goals of applied ethnomusicology. The primary purpose of this thesis is to respond to the absence of serious scrutiny of existing pedagogical approaches to African music at universities across South Africa. The contribution this research makes will be valuable to African music programmes across the continent as well as to practitioners of African traditional instruments, such as the marimba, mbira, timbila xylophones, nyanga pan pipes, and valimba xylophones. The thesis comprises five chapters. The first presents an introduction to the research, and its goals, procedures and approaches, along with an outline of the subsequent chapters. Tracey’s biography is covered in the second chapter. A consideration of the state of African music teaching in other African countries such as Ghana, Kenya, and Zimbabwe; the history of African music; and the state of African music pedagogy in tertiary institutions in South Africa constitutes the third chapter. Chapter 4 comprises an analysis of Tracey’s articles and data gathered from interviews, as well as my personal reflections as Tracey’s student. The final chapter presents a summary of the preceding chapters, the study’s findings, and suggestions for further research. A multidisciplinary approach was used for this thesis. The results finds that Tracey’s articles had six common themes which he wrote about and are a contribution to African music pedagogy. The contribution of efficient financial management on public health service delivery at King Cetshwayo District in KwaZulu Natal

- Description: The aim of this research is to develop strategies that the DOH could put into place to improve its internal controls in financial management in order to contribute towards service delivery at Ngwelezane, Queen Nandi and Eshowe hospitals within King Cetshwayo District. The study used a mix of qualitative and quantitative study methods, however using one research instrument to collect both qualitative and quantitative data from 37 financial managers in these three hospitals. Data analysis using content analysis (of qualitative data) and percentage frequency analysis (of quantitative date) yielded study findings. This study found that financial managers are fairly competent regarding ‘effective communication’, ‘team work’, and ‘financial leadership’; the study results did not show respondents’ clear comprehension of Auditor General findings and processes followed to implement the recommendations in order to assess internal controls; and findings also indicate that financial managers meet objectives of financial management through cash flow, budget forecasts and expenditure. Developing a framework that will guide and monitor the application of financial strategies with the aim of improving service delivery will involve enhancing financial managers’ knowledge of District and Institution Service Delivery Plans, allocating resources properly and efficiently during the budgeting exercise, establishing internal strategies to maximise accountability and transparency, having fraud prevention strategies in these institution, and putting measures in place to be able to forecast budgetary adverse. Conceptualising mental distress from an African psychology paradigm: using an interpretative phenomenological analysis of the views of traditional healers

- Description: With South Africa's long history of colonialism and racial oppression, there are still services in the country that many South Africans cannot relate to, including psychology. Research shows that many South Africans experience and are affected by mental distress due to several factors, including poverty, unemployment, and traumatic experiences. Managing and treating such distress has always been challenging for most South Africans. Some debates question the relevance of psychological services from the West in a South African context. This study explores other approaches to psychology that look beyond the Biopsychosocial model when dealing with certain types of disorders in an African context. African psychology, or the Afrocentric approach, looks at what is beneath the surface, not just the presenting problem. Mainstream psychology strives to be universal and applicable to all. However, African psychology disagrees with this notion. African psychology perceives human beings as strongly influenced by social and cultural influences. The focus of this approach includes the spiritual realm and the attached meanings. There is evidence for the need to merge Traditional and Western medicine. The research methodology for this study is qualitative, using Interpretative Phenomenological Analysis. IPA allows for a critical engagement with the ways in which participants construct their reality. The researcher carried out semi-structured interviews to enable participants, all traditional healers (3 female and 2 male), to narrate their experiences dealing with mentally distressed clients. The accounts of these traditional healers were analysed focusing on people who have experienced mental distress. From the analysis and synthesis of the themes, findings illustrate how traditional healers conceptualise and construct mental distress from an African Psychology paradigm. An emerging core theme was the importance of the divine call and its influence on the chosen treatments. More studies are needed to illustrate the potential for collaboration between African Traditional healing and EuroAmerican healing practices, to provide holistic services to people in need. , Thesis (MA) -- Faculty of Humanities, Psychology, 2022

An appraisal of strategies to mitigate related to non-communicable diseases of the eye: a case study of Michael Mapongwane Health Clinic in Khayelitsha, Western Cape Metro District

- Description: Preventive and curative eye-care services are a critical component of care for patients diagnosed with chronic diseases. The South African constitution guarantees health as a fundamental human right of all citizens. While most chronically ill patients are managed at the primary health care centers, eye care services are currently contracted to private service providers. The extent to which people, especially the chronically ill patients, have access to eye-care service under the current structure is less understood. The study aims to establish the effectiveness and sustainability of the eye-care service delivery model at the Michael Mapongwane CHC, with specific reference to access to eye-care services. Specifically, the study examined the rate and determinants of eye-care service utilisation, assess user’s satisfaction and perspectives of primary health workers and private service providers regarding the current structure of eye-care services. The study adopted a descriptive and explanatory design, which involved a survey and semi-structured interviews. The survey was conducted among 423 patients attending the two primary health centers selected for this study. Patients with chronic illness were recruited over a period of one month. Overall, 365 patients with chronic illness and 58 patients without chronic illness took part in the study. Descriptive and inferential statistics were used to analyse the quantitative date. Purposive sampling was used to select nine health workers and 10 private eye-care service providers for the semi-structured interviews. The data obtained were transcribed and coded. Thematic content analysis was performed on the qualitative data. The results show that less than half of the respondents (n=168) had ever had an eye test, and an even lower proportion (n=115) had had an eye test in the past two years. Age, female sex, formal rural dwelling, marital status, and income were significantly associated with ever having an eye test done. The results reveal a gross inequality in access to eye care test. Only 19.2% of patients diagnosed with hypertension had had their eyes examined. Even fewer proportions of the patients (8.5%) had had their eyes examined during or after diagnosis for hypertension. Likewise, only one in three patients diagnosed with diabetes has had their eyes examined before or after they were diagnosed with diabetes. The majority of patients who had used the services affirmed that it was easy to access eye care services. While most respondents agreed that waiting time to make an appointment was not too long, over half of them affirmed that waiting to see an eye doctor was too long. Most users were reasonably satisfied with the eye care services received. Primary health care providers highlighted challenges such as work overload, poor infrastructure, and patient’s forgetfulness of the appointment dates as factors affecting access to eye care services in their clinic. The primary challenges the private eye-care service providers experienced were small rooms or inappropriate clinic rooms, language barrier and inadequate support from the clinics. In conclusion, this study has shown that access to preventive and curative health care services was among patients attending primary health care centres, and especially those with chronic illness. - Description: Since the dawn of democracy in 1994, the government of the Republic of South Africa (RSA) has launched numerous development programmes. These include youth empowerment initiatives aimed at developing the capacity of the youth in order for them to contribute to the economy. Optimistic prospects for any country depend on developing its young people. The measures to drive youth-focused delivery and to ensure youth development ought to move further than the establishment of structures and the delivering of policy statements. Government departments and municipalities are expected to implement meaningful youth empowerment programmes and ventures that are visible and that are adequately monitored. This study explored and assessed the effects of the Expanded Public Works Programme (EPWP) on youth empowerment. It used a case study of Amathole District in the Eastern Cape, South Africa province. The study utilised the qualitative technique to provide solutions to the research problem and to provide responses to the research hypothesis of the study. The sample comprised ten public representatives and thirty youth beneficiaries. In line with the purposive sampling approach, two sets of data were generated: the first set from public managers of the programme, and the second from programme beneficiaries. The researcher made use of the qualitative thematic content analysis approach, where data was analysed to arrive at emerging themes. The study was delimited to the effects of EPWP on youth empowerment. It was also limited to Amathole District within the Eastern Cape province. The results showed that most of the beneficiaries were engaged on a period of 1 to 12 months. This shows that most of participants were not engaged for viable and long periods, which they would need to truly increase their financial or monetary position in their households and have ample time to attain expertise. The results further indicated that in some cases the EPWP provides financial and income support to beneficiaries, but in some it does not. In general, the results showed that the EPWP reduces poverty because participants are able to look after their families and relatives after taking part in the EPWP. However, the training and skills participants attained in the EPWP interventions did not help them find employment elsewhere. This is linked to the findings that showed that beneficiaries were not provided with training certificates. Therefore, to enhance the execution of the EPWP and speed up poverty reduction, equity and joblessness in South Africa, the study recommends that ECDPW in ADM Area should design projects/programmes that are attractive to youth and that will equip them with necessary skills to empower youth to participant in the job market after the leave the programme.
