- Description: Diabetes mellitus (DM) is a group of endocrine and metabolic disorders characterised and identified by the presence of hyperglycaemia over a long period and, to an extent, accompanied by hyperlipidaemia. CVD has been reported to be the leading cause of mortality in patients with DM. Several antidiabetic agents are available for managing DM, but these agents are not for curative therapy and present with undesirable side effects. In addition, these agents become less effective as the patient's condition progresses to complete beta-cell failure. Therefore, developing newer antidiabetic agents with minimal undesirable side effects, prolonged efficacy and protection against the development of DM complications are necessary. This study was conducted to identify potential novel antidiabetic agents with cardiovascular-protective activity. When fish are subjected to thermal stress, they may primarily respond by changing their behaviour. Species that have broad phenotypic behavioural plasticity (i.e. defined as the ability to adjust behavioural activity in presiding environmental conditions in order to remain within their optimal thermal range) may have a competitive advantage. Fish behavioural plasticity may take many forms. Some species may seek out thermal refugia by changing their phenology or distribution, while others alter the timing of their seasonal and spawning migrations in response to a changing environment. Although fishes can use behavioural changes to cope with climate change impacts, there does appear to be variability in the behavioural responses within species. However, if alterations in behaviour are insufficient to ensure that the individual remains within their optimal thermal range, physiological acclimation (i.e. defined as the process in which an organism adjusts to prevailing conditions by broadening their thermal performance curve so that their performance is maximized in the new thermal environment) may be required. Making the personal political: understanding the impacts of participation in an anti-rape protest for women who have experienced sexual violence

The majority of those who experience sexual violence do not acknowledge, disclose or report it, and do not access support. Many factors have been identified which can inhibit acknowledgement and disclosure. While silence is to be honoured as a chosen response, unwanted silencing is of concern. The annual Silent Protest at Rhodes University is a day-long anti-rape protest which draws attention to the attitudes and practices which silence victim-survivors, and enacts solidarity with victim-survivors of sexual violence. I conducted a three year “critically engaged, activist ethnography” with the community of Silent Protestors and organisers. My focus was on the stories that victim-survivors told about the impacts of participating in the protest. Changes in the research context allowed for a participatory action research cycle to be incorporated into the methodology. This offered an opportunity to implement and evaluate some changes suggested by my research contributors. My contributor’s narratives highlighted the ways in which as a society we position those who have been subjected to sexual violence in ways that are limited and limiting and which diminish victim-survivors’ sense of agency and value. This malignant positioning restricts the choices available to victim-survivors. The anticipation of being positioned in negative ways can inhibit victim-survivors from disclosing a violation and accessing care and justice. Participation in the Silent Protest seems to stand against some of the factors which inhibit acknowledgement and disclosure. Through their participation many women took up opportunities to share their story with at least one other person and to receive and offer care. Most described participation as having been beneficial and having helped them move towards living well again. The ways in which the Silent Protest positioned victim-survivors and the choices they were offered in relation to that positioning seemed to open up opportunities for thinking, feeling and acting in ways that they preferred. - Description: This study focused on investigating and developing an Afrophilic orientation to a sociocultural approach to philosophy for children. The main aim was to foster a critical and generative approach to considering the heritage-based curriculum foundations of Zimbabwean primary schools, focusing on Sebakwe resettlement primary schools (the case study area). Afrophilia foundations in the study are regarded as “the discourses that are the medium of philosophical reflexion” (Rettova, 2004, p. 4) in each African society. As articulated in the study, such discourses include African proverbs, poems, stories, music, and folktales which are useful in the initiation of philosophical engagements with children in a community of inquiry approach. A community of inquiry is a framework that reflects a collaborative-dialogical approach to teaching and learning. Curriculum reviews in postcolonial Zimbabwe have revealed an unconscious misalignment of the Zimbabwean education system's philosophical underpinnings because it has continued to align itself with imperial British colonial philosophy, which contradicts the country's developmental needs (Presidential Commission of Inquiry into Education and Training [CIET], 1999, Zimbabwe Ministry of Primary and Secondary Education [MoPSE], 2014). The philosophical misalignment is an extension of the colonial history of Zimbabwe showing that vestiges of colonial rule still shape the education sector. This leaves the education system focusing mainly on abstracted concepts with continued marginalisation of local cultures, discourses, and knowledge. Consequently, it leaves Zimbabwe together with other postcolonial education systems with the task of dealing with the phenomenon of colonial continuity. One implication of these colonial continuities, is learner disinterest in learning, which affects the educational process negatively, affecting the learning of critical reflexive thinking, an issue which I have observed in the Sebakwe resettlement schools involved in this study where I have been teaching for 15 years. To address, this, the study sought to answer the following questions: What historical and contemporary barriers are affecting the promotion of a culture of learning in Zimbabwean resettlement primary schools, particularly as they relate to learners' underdevelopment of critical reflexive thinking skills? In addition, How can the Philosophy for Children Afrophilia curriculum intervention promote a culture of learning in Sebakwe resettlement primary schools that is oriented toward the development of critical reflexive thinking skills in children? In approaching the research questions, I applied postcolonial and decolonisation theory, sociocultural learning theory and curriculum theory aimed at transformative change, that is oriented towards achieving a more contextually oriented approach to teaching and learning, and a paradigm of ‘learning as connection’ (Lotz-Sisitka & Lupele, 2017; Shumba, 2017). This has been identified as a ‘missing’ discourse in mainstream educational quality discourses in southern Africa (Lotz-Sisitka & Lupele, 2017). Furthermore, to create space for curriculum development using multi-actor groups in a community of inquiry, I used decolonial research methodologies in the data generation process. The study is constituted as an interventionist case study, and I applied a four-stage process comprising document analysis, workshops, participant observations involving children between 8-11 years, and reflective interviews with parents, educators and children as instruments for data gathering. I also used process-based analytical tools developed in Philosophy of Children research to analyse the processes of critical reflexive thinking development that emerged from the Philosophy for Children pedagogy involving ten lessons which I facilitated, and videorecorded. Moreover, I used postcolonial and decolonial discourse analysis to provide the broader analytical insights that informed the interpretations of the lesson analysis data from the perspective of the research problem that I address across the research project. This PhD is produced as a PhD by publication, which involves publishing of papers, and orientation to, and interpretation of the papers. The main findings of this study are reported in articles that I prepared for publication, three of which have already been published (see Appendices A1, A4 and A5), and four which have been submitted for publication (A2, A3, A6, A7) with some already at an advanced stage of finalisation via review. The conceptual paper (A1) that served as the foundation for the publication journey revealed that, in addition to the weight of cultural technologies of domination, the curriculum is shaped by the paradox of a superficial interpretation of unhu/ubuntu educational philosophy. As a result, the curriculum becomes disconnected from the learners' real-world experiences. The second paper (A2), which focuses on why Zimbabwe needs the Philosophy for Children approach with a sociocultural medley, unveils Zimbabwe's complex decolonial curriculum reforms and their many contradictions and paradoxes. However, it also emerged that the approach used in this study empowers teachers, is relevant to the emerging constellation of practices in the Sebakwe resettlement, and influences power sharing beyond teacher-child relationships. The third paper (A3), based on children’s philosophy for children practice, defends the study’s decision to bring children’s heritage and cultural lens to bear on curriculum and pedagogical praxis. In essence, the article explores a synergy between philosophy for children and the Zimbabwean heritage-based educational curriculum, serving to enrich both. The fourth paper (A4) makes the case that philosophy for children could be a viable pedagogy for transformative education, and it provides evidence-based implementation of a context-based philosophy for children. According to this paper, the approach is effective in strengthening strong community relationships, instilling pride in local heritage, and advancing curriculum transformation. The fifth paper (A5) focusses on the approach's implications for teachers' roles, practices, and competencies. Six dimensions of teacher roles, practices, and competences surfaced, including the role of the teacher as a decoloniser and pedagogical innovator, among others. The sixth paper (A6), influenced by the previous papers' findings, focused on decolonisation and improving learning opportunities for children in the Sebakwe area using the philosophy for children approach. This paper's data depicts a ‘third space’ in which learners consolidate their cultural capital and curriculum content into their own meaning construction. The implication is that schools become neutral sites that improve learners' interdependence and inclusivity while also taking contextual realities into account. The findings of the seventh and final paper (A7) presented in this write-up advance the idea that a Philosophy for Children approach with a sociocultural medley influences an ethics of care by demonstrating how Afrophilia experiences influence a new path to wildlife conservation and sustainability. The study highlights that integrating Philosophy for Children and Afrophilia foundations of knowledge into the school curriculum promotes critical reflexive thinking skills, helps to address real-life problems and adds relevance to the curriculum. The study further shows that the integration of philosophy for children in the advancement of curriculum transformation in Zimbabwe is a successful formative interventionist approach in the resettlement schools that are characterised by a critical shortage of teaching and learning resources. In essence, the research opens an understanding that curriculum transformation and decolonisation are context-based and multi-actor processes, as showcased in the experiences of parents, teachers, education inspectors, and children in this study. Furthermore, this study posits that situating curriculum decolonization and transformation within unhu/ubuntu dialectical rationality and advancing diversity in reasoning necessitates deeper engagement with heritage-based curriculum and provides teachers with appropriate agency to modify and adapt their pedagogies in alignment with the learners' life world. According to the study, this emerged as a rational possible solution to the problem of curriculum decontextualisation. Curriculum decontextualisation as highlighted in the study via the problem of colonial continuity mentioned above, appears to be further influenced by the emphasis on examination assessment scores which seem to widen the gap between the adult and child worlds, as well as the gap between contextual realities and [curriculum] examination content. Overall, the study offers an approach that can deepen an unhu/ubuntu foundation for the heritage-based curriculum reform in Zimbabwe, and strengthen the learning of children in the resettlement schools, where the case was explored. - Description: Mathematics is a language that is rich in visual representations (Mudaly & Rampersad, 2010). Visual Representations assist us in developing our reasoning skills when solving a problem and our understanding of the relationships between concepts (Ozkan et al., 2018). This thesis focuses on the different visual representations (VR) in South African Foundation Phase mathematics textbooks and workbooks. Textbooks and workbooks play an important role in developing an understanding of mathematical concepts for both teachers and learners (Harries & Spooner, 2000). While teachers generally rely heavily on textbooks, they were a key resource while schools were closed due to COVID-19 lockdown regulations. The theory of Constructivism forms part of the theoretical framework for this study. Constructivism advocates that learners actively construct knowledge through experiences rather than passively receiving knowledge from the outside (Von Glaserfeld, 2001). Vygotsky believed that social interactions create experiences that facilitate the learning and meaning-making process (Vygotsky, 1978). This case study is underpinned by an interpretivist paradigm as it sought to examine the nature of VRs in three Grades 1 - 3 textbooks/workbooks. My research approach is primarily qualitative with descriptive statistics to assist in developing a more comprehensive understanding of the research questions. The study was guided by the analytic tool designed by Fotakopoulou and Spiliotopoulou (2008) which I adapted for Foundation Phase mathematics use. The framework provides insight on the type of VR, VRs relation to content, VRs relation to reality, the function of the VR and dimensionality of a VR. While the workbooks had many more VRs than textbooks, the dominant type of VR in textbooks and workbooks are images. Microbial and extracellular polymeric substance dynamics in arid–zone temporary pan ecosystems

In addition, microbiomes produce extracellular polymeric substances (EPS) which encompass a hydrated exopolymer mainly constituted of carbohydrates and proteins. The exopolymer aid proliferation and persistence of biofilms on their resident surfaces. There is however paucity of data on functional diversity of microbiomes in arid zone temporary wetlands with previous research having mainly focused on permanent systems in the northern hemisphere. In the face of ongoing climatic changes and anthropogenic threats to wetlands, it is imperative to assess the health status of aquatic systems in relation to microbial productivity dynamics. In this thesis, colorimetric methods and sequence–based metagenomics were conducted to quantify microbial EPS production and bacterial metagenome functions, respectively. This study was conducted in Khakhea–Bray region (North–West, South Africa) in June 2021 and January 2022 with a focus on evaluating microbial patterns of distribution between seasons (i.e., Dry and Wet) and varying depth i.e., deepest zones (Deep), intermediate depth (Mid) and shallowest regions (Edge). Additionally, potential relationships between EPS and either water content or organic matter content (OM content) were evaluated. In this study it was hypothesized that wet phases and deeper zones will have high EPS production and support more functions in comparison to shallowest regions and dry phases. Carbohydrates and proteins were quantified using the Dubois method and modified Lowry procedure, respectively. Carbohydrates generally occurred in higher proportions than proteins, suggesting that EPS found in these systems was largely diatom produced. The wet phases (wet season and inundation periods) supported more EPS production compared to the dry phases. The results of principal components analysis (PCA) and Spearman’s correlations suggested that EPS was highly correlated with sediment water content among other assessed variables. No significant associations were established between EPS and organic matter content. Spatial distribution of EPS demonstrated similar patterns between the deepest (Deep) and the intermediate depth zones (Mid) however the shallow regions (Edge) had significantly lower concentrations. Bacterial characterization was established by amplification of the 16S rRNA gene using illumina–sequencing protocol. Enzyme functions associated with biogeochemical pathways were predicted in PICRUSt2 bioinformatics pipeline. A total of 15 042 Unique Amplicon Sequence Variants (ASVs) were observed to be affiliated to 51 bacterial phyla and 1 127 genera. All top genera had commonality in heat tolerance. Firmicutes, dominated at phyla level with 59 % (mean ± sd, 19 ± 13 %) relative abundance followed by Actinobacteria and Proteobacteria both at 34 % (18 ± 7 %) and (18 ± 6 %), respectively. Microbial diversity matrices highlighted significant differences in beta diversity more than alpha diversity. Bacterial microbiomes were more distinct between seasons compared to within season, suggesting that functions were seasonally driven. These findings were supported by highest rates of denitrification, carbohydrate degradation and EPS production by core microbiomes in the wet season as compared to low rates of nitrogen mineralisation, carbon fixation and nitrification in the dry season. The present findings represent a first attempt in evaluating sequence–based metagenomics in semi–arid southern African temporary pan ecosystem. Both microbial EPS and bacterial functional potential were highly driven by water availability, with highest rates mainly associated with maximum inundation compared to dry states of pans. A Pedagogy of Love: reflections on 25 years of informal vocational education and training practices in the commercial fishing industry in South Africa

- Description: This thesis is a reflection of informal vocational education and training (IVET) practices conducted by the Researcher in the commercial fishing industry between 1995-2021. Fourteen interventions took place during this time which involved several hundred sea-going employees who are disadvantaged by low levels of literacies and low/no Internet Computer Technology connectivity. The Production Programme was chosen as the focus of study and draws upon the influence of the other thirteen programmes. The purpose of the Production Programme was to teach technical fish processing skills to the production management teams and factory hands who work in the factories on board the vessels. The programme ran for five years and evolved through three distinct generations. At the heart of this work lies the question: ‘What made these programmes successful?’ This is an intra-programmatic study and seeks to identify the pedagogical practices which promoted or confounded the efficacy of the Production Programme; and based upon this understanding how such programmes could be improved, transferred, and taken to scale. These questions are both descriptive and explorative in nature. IVET is regarded as training which takes place outside of the formal South African National Qualifications Framework. This work is relevant because approximately 80% of sea-going staff neither finish school, nor get the opportunity of Post School Education and Training by going to a university, a technical institution, or a community college. This statistic is reflected in the general population (Department of Higher Education and Training [DHET], 2022). There is a fine line between being employed and unemployed for people working at this level in the formal economy which makes this project relevant to youth or persons who are ‘not in employment, education or training’ too. This means that for most South African adults IVET presents a significant opportunity for post-school education. Therefore, it is important to answer the questions raised above to rapidly improve inclusion of the majority of South Africans into meaningful education which improves livelihoods. The Theory of Practice Architectures (TPA), from the collection of social-material practice theories, is the conceptual framework for this thesis. The smallest unit of analysis of TPA are practices which may be ‘sayings/thinkings’, ‘doings’, or ‘relatings’. These practices bundle together into practice arrangements and form practice architectures. The reason that TPA was chosen was that practices were the only data available as we (learners, managers, facilitators, and me) knew what we had said/thought or done, and we were aware of the relationships between us over the years as the fourteen interventions played out. Under conventional research circumstances data would be collected in real time, however in this project, most of the data is historical. In addition to the fundamental building blocks of TPA, the theory is embedded in a Theory of Education. There have also been contemporary enhancements to the TPA which were significant to this study, for example, the Ecologies of Practices, a Trellis of Practices which Support Professional Learning, Middle Leadership, Travelling Practices, and moves towards transformative or transgressive education using the TPA in IVET. This is empirical, qualitative research and an ethnographic case study was chosen as the research design which is a methodology particularly suited to answering both descriptive and explorative questions. Nine methods for data collection were used, namely an historical reflective narrative; two focused-group interviews; three individual interviews; four Whatsapp videos; one WhatsApp voice note; two mobisodes; ten questionnaires; 29 documents; and 16 photographs. Because this data was collected under Covid-19 pandemic conditions, two conceptions were employed to guide the generation of data under these uncertain and constrained conditions. These were firstly, the ‘methodology of chance’ which allowed for a ‘methodological agility’; and secondly, the idea of ‘information power’ which is helpful in deciding on how much data is enough. In order to be explicit concerning a key research activity, the approach and method used to review the literature is explicated. Key practices were identified in the data set and described; and then the data was analysed using heuristics provided by TPA theorists. Seven Tables of Invention were used to synthesise the data arising from these practices. An eighth Table of Invention was used to synthesise all the practices and practice arrangements characterised; and to indicate how these evolved over time and space. The data description and analysis is supported by eight Analytic Memos, a comprehensive Data Code Table and a hyperlink to a data repository which provides access to oral and video material. The findings distinguished five key practices and practice arrangements which were: Practices of the Creation of Courseware; Practices of Teaching and Learning; Practices of Assessment; Practices of Love; Practices of Management. The thesis title is reflective of the impact which love has upon the pedagogical process of IVET. Based upon the analysis and synthesis of the corpus of data, practices which either promoted or confounded the Production Programme became visible; it is these insights which inform future improvements to similar programmes. Emanating from these findings, two overarching practice architectures (PA) were identified which restrained the Production Programme in the same manner that the banks of a river restrain a river, and yet simultaneously, are changed by the river over time. These are the PA of Methodology and Methods and the PA of Maturing Ecologies of Practices. The inferences drawn from the data were achieved through the use of deductive, inductive, and abductive reasoning. My claim to new knowledge is a lamination of a practical contribution on one side of the coin, and a theoretical contribution on the other side of the coin. The PA of Methodology and Methods provides a practical mechanism to create, deliver and assess IVET. This is done by explicating the three practice architectures which constitute the overarching PA of Methodology and Methods which are, the PA of Informality; the PA of Range, and the PA of Relationality. An IVET programme constitutes Ecologies of Practices. The theoretical conception of the overarching PA of Maturing Ecologies of Practices provides a conceptual tool which enables the transferring and scaling of IVET programmes. It does this by providing theoretical indicators to establish the ‘state’ of an IVET programme as it matures over time from a pioneer state to a settler state. An IVET educator can then work towards creating a PA which is conducive for a mature ecologies of practices to form; and the programme can then be transferred and/or taken to scale, if this is desirable in the particular context. The power of my claim to new knowledge does not lie on one side or the other of the coin, but in the lamination of the practical and theoretical contributions put to use in the service of IVET. This thesis concludes with a number of theoretical and practical recommendations which are loosely grouped according to ‘sayings/thinkings’, ‘doings’ and ‘relatings’ in deference to the value of TPA to this thesis. An urgency is conveyed in these recommendations as there is an immediate need to improve the livelihoods of ordinary South Africans. The influence of immuno-contraception on the ecology of African savanna elephants (Loxodonta africana) in a small game reserve in the Eastern Cape, South Africa

- Description: African savanna elephants (Loxodonta africana) can alter the vegetation structure on a landscape which has both positive and negative consequences for biodiversity. This ecological role can be dependent on the density of elephants in a given area and how the area is transformed. Elephants are currently listed as an endangered species on the International Union for Conservation of Nature (IUCN) global red list. Despite this endangered status, strong conservation efforts in southern Africa have increased elephant numbers in protected areas. In South Africa, many of these protected areas are small (<1000 km2) and fenced. Thus, without the freedom to roam over large areas, elephant impacts on vegetation and other ecosystem processes may be exacerbated. Increased growth rates and higher population densities, have prompted wildlife managers to explore several population-level management techniques to slow elephant growth rates in small, protected areas. One such technique is the use of immuno-contraception. My study focused on three datasets (Pre, During and Post porcine zona pellucida (PZP) treatments) of one elephant population over an 18-year period in a small game reserve in the Eastern Cape, South Africa. The first aim of my study was to compare the female demography and reproductive parameters of treated (with PZP) and non-treated females (no PZP). I predicted that the population growth would decrease as more elephant cows were contracepted as well as them having longer inter-calving intervals than non-treated cows. Further, I predicted that there would be a skewed sex ratio of male calves born to treated cows. Lastly, I predicted that fertility rates would be lower in treated cows than in non-treated cows. I calculated the annual population growth rates, inter-calving intervals, sex ratios, age at first calving, fecundity rates and fertility rates of all breeding cows. Population growth rates were similar to other managed populations although, compared to a relatively untouched elephant population, annual growth rates in my study site were high. The inter-calving intervals and age at first birth were low compared to other African protected areas which were most likely influenced by the use of PZP, rainfall and a lack of density dependence. Treated cows did not have longer inter-calving intervals compared to non-treated cows nor was there a male biased sex ratio of the offspring of treated cows. The second aim was to assess the spatial ecology of this elephant population and how it may be influenced by immuno-contraception. I predicted that the use of PZP would increase the elephant herd’s core range as there were fewer reproducing cows. I also predicted that PZP would alter the elephant’s habitat use making them less selective in terms of the vegetation types they selected. - Description: African savanna elephants (Loxodonta africana) can alter the vegetation structure on a landscape which has both positive and negative consequences for biodiversity. This ecological role can be dependent on the density of elephants in a given area and how the area is transformed. Elephants are currently listed as an endangered species on the International Union for Conservation of Nature (IUCN) global red list. Despite this endangered status, strong conservation efforts in southern Africa have increased elephant numbers in protected areas. In South Africa, many of these protected areas are small (<1000 km2) and fenced. Thus, without the freedom to roam over large areas, elephant impacts on vegetation and other ecosystem processes may be exacerbated. Increased growth rates and higher population densities, have prompted wildlife managers to explore several population-level management techniques to slow elephant growth rates in small, protected areas. One such technique is the use of immuno-contraception. My study focused on three datasets (Pre, During and Post porcine zona pellucida (PZP) treatments) of one elephant population over an 18-year period in a small game reserve in the Eastern Cape, South Africa. The first aim of my study was to compare the female demography and reproductive parameters of treated (with PZP) and non-treated females (no PZP). I predicted that the population growth would decrease as more elephant cows were contracepted as well as them having longer inter-calving intervals than non-treated cows. Further, I predicted that there would be a skewed sex ratio of male calves born to treated cows. Lastly, I predicted that fertility rates would be lower in treated cows than in non-treated cows. MtHsp70 self-aggregates, a property unique to this isoform, and an Hsp70 escort protein (Hep1) is required to maintain the molecular chaperone in a soluble, functional state. This study aimed to compare the solubilizing interaction of Hep1 from T. brucei and Homo sapiens (H. sapien). The recently introduced Alphafold program was used to analyze the structures of mtHsp70 and Hep1 proteins and allowed observations of structures unavailable to other modelling techniques. The GVFEV motif found in the ATPase domain of mtHsp70s interacted with the linker region, resulting in aggregation, the Alphafold models produced indicated that the replacement of the lysine (K) residue within the KTFEV motif of DnaK (prokaryotic Hsp70) with Glycine (G), may abrogate bond formation between the motif and a region between lobe I and II of the ATPase domain. This may facilitate the aggregation reaction of mtHsp70 orthologues and provides a residue of interest for future studies. Both TbHep1 and HsHep1 reduced the thermal aggregation of TbmtHsp70 and mortalin (H. sapien mtHsp70) respectively, however, TbHep1 was ~ 15 % less effective than HsHep1 at higher concentrations (4 uM). - Description: The landscape of the Upper Zambezi Catchment in the remote North-Western Province of Zambia retains many natural features, but current and future anthropogenic activities threaten its integrity. Lack of information relating to the basic ecological functioning of the region hampers conservation efforts. Flooding from the adjacent woody-savanna and open grasslands in the Upper-Zambezi Catchment in the wet season drives crucial processes in the aquatic ecosystems such as the Kabompo River. This thesis describes aquatic food webs and describes the nutrient cycling in this river. A general introduction was based on the seasonality, river morphology, and biodiversity of the Kabompo River. There is no aquatic food web information available on this river. To address this knowledge gap, sampling of the principal food web components– vegetation, invertebrates, and fish – were collected in the dry season from August to September 2019 across six sites in the Kabompo River. Stable isotope analyses provided proxies for the food web structure at each site, and were the basis of longitudinal comparisons. The analyses suggest that the Kabompo River food webs follow the trophic ordination and nutrient cycling characteristic of the revised-Riverine Productivity Model. Food webs and community assemblages remain structurally similar between sites and provide some evidence of bottom-up productivity-driven trophic dynamics. To predict the possible landscape-scale processing of nutrient changes present for the aquatic ecosystems of the Kabompo River, a longitudinal comparison between a herbivore, Labeo cylindricus, and omnivore, Synodontis spp., fish species and their resources (primary producers and invertebrates) was done and showed archetypal fish food web trophic separation (2 ‰ to3 ‰) between species which remains consistent for the headwater sites. The nutrient values change at the convergence between the Kabompo Bridge and the Mwinilunga branches and show a decrease in the trophic separation of the sites below. This change becomes apparent from the last site, Watopa, where the trophic separation re-establishes (2 ‰ to 3 ‰) between the species. The trend suggests primarily autochthonous production in the headwater reaches, changing to allochthonous and autochthonous nutrient incorporation after the convergence of the two main branches. Permanent wetlands surrounding the convergence zones may be conducive to more primary producer activity and increased nutrient turnover. Permanent wetlands are a common feature of the Upper Zambezi Catchment tributaries, with the most notable lentic system the Barotse Flood Plains in the south, which shares reach-adjacent characteristics with the Kabompo River. Trends from the 2015 dry season identified for the Barotse Flood Plains support the findings of this thesis, where the δ13C values recorded reach depletion of -45 ‰. Management in the Upper Zambezi Basin and the Kabompo River comprises biannual assessments of water quality and quantity by the Zambezi River Authority, but not of the ecosystem process. This thesis may provide information to address the ecological (food webs and nutrient cycling) dynamics of the Basin rivers. Species that have been introduced to new habitats can either be alien (not causing any harm to the new environment) or invasive alien species (destroying or competing with the indigenous species for resources). Investigating the source of these introductions is important as it provides baseline information about their biology and evolution. It also allows for better prevention measures for future invasions and for effective conservation strategies. In this study, I investigate the source of the introduced populations of tropical house geckos Hemidactylus mabouia in the Eastern Cape Province using two mitochondrial gene regions, ribosomal 16S and NADH dehydrogenase subunit 4 (ND4). We also look at the potential spread of these species using species distribution models (SDMs). The phylogeny showed that the introduced populations into the Eastern Cape were genetically similar to both H. mabouia lineages found in Central & Southern Africa. The phylogeny produced two clades from which the majority of the introduced samples were grouped in a clade with samples from Angola, DRC, and Mozambique, and only a few nested within the second clade with the South African native population. Haplotype networks from both genes also showed two clades, matching those found in the phylogeny. The introduced samples were associated with samples from Angola and those from South Africa (native) as these localities had a number of shared haplotypes. SDMs showed less evidence that these individuals could naturally move west down the coast as the probability of occurrence was below 0.4 from the Eastern Cape to the Western Cape. Our conclusion was that the introductions may be due to multiple avenues like species moving down the east coast or human translocation including trade routes. - Description: The new generation of radio telescopes, such as the MeerKAT, ASKAP (Australian Square Kilometre Array Pathfinder) and the future Square Kilometre Array (SKA), are expected to produce vast amounts of data and images in the petabyte region. Therefore, the amount of incoming data at a specific point in time will overwhelm any current traditional data analysis method being deployed. Deep learning architectures have been applied in many fields, such as, in computer vision, machine vision, natural language processing, social network filtering, speech recognition, machine translation, bioinformatics, medical image analysis, and board game programs. They have produced results which are comparable to human expert performance. Hence, it is appealing to apply it to radio astronomy data. Image segmentation is one such area where deep learning techniques are prominent. The images from the new generation of telescopes have a high density of radio sources, making it difficult to classify the sources in the image. Identifying and segmenting sources from radio images is a pre-processing step that occurs before sources are put into different classes. There is thus a need for automatic segmentation of the sources from the images before they can be classified. This work uses the Unet architecture (originally developed for biomedical image segmentation) to segment radio sources from radio astronomical images with 99.8% accuracy. After segmenting the sources we use OpenCV tools to detect the sources on the mask images, then the detection is translated to the original image where borders are drawn around each detected source. Identifying and segmenting sources from radio images is a pre-processing step that occurs before sources are put into different classes. There is thus a need for automatic segmentation of the sources from the images before they can be classified. This work uses the Unet architecture (originally developed for biomedical image segmentation) to segment radio sources from radio astronomical images with 99.8% accuracy. After segmenting the sources we use OpenCV tools to detect the sources on the mask images, then the detection is translated to the original image where borders are drawn around each detected source. The test was found to favour learners within the central band on the ability scale. An exploratory five dimensional item response theory model was then developed to investigate the alignment with the test specifications as evidence for construct validity. Significant discrepancies between the factor structure and the specifications of the test were identified. Notably, the results suggest that some items measured an ability that was not purely mathematical, such as reading ability, which would distort the test’s representation of Mathematics ability, disadvantage learners with lower English literacy, and reduce the construct validity of the test. Further validity evidence was obtained by differential item functioning analyses which revealed that fourteen items function differently for learners from different provinces. Although possible reasons for the presence of differential item functioning among provinces were not discussed, its presence provided sufficient evidence against the validity of the test. In conclusion, multidimensional item response theory provided an effective and rigorous approach to establishing the validity of a large-scale assessment. MeerKAT observations of three high-redshift galaxy clusters

- Description: Galaxy clusters are the largest, gravitationally-bound structures in the Universe, formed through the hierarchical merger of smaller structures. The most accepted view is that the merging process injects energy into the intracluster medium (ICM) and re-accelerates pre-existing particles and compresses magnetic fields, generating, eventually, synchrotron emission. Such radio emission appears as radio halos, i.e. central Mpc-size diffuse structures, mostly visible in merging or unrelaxed clusters and with a spatial correspondence with the thermal gas component of the ICM. Observations have probed radio halo properties mostly for clusters withM500 > 6×1014 M⊙ at intermediate redshifts (0.3 < z < 0.4), providing support to their connection between mergers, which provide the necessary energy to re-accelerate particles via turbulence. Probing the redshift evolution of radio halos is an important test of the turbulent re-acceleration scenario, as fewer halos are expected at high redshift, given the same mass interval. In this thesis, we present MeerKAT observations at 1.28 GHz of three high-redshift (PSZ2G254.08- 58.45, PSZ2G255.60-46.18 and PSZ2G277.76-51.74, in the 0.42 ≲ z ≲ 0.46 range) clusters, with masses M500 ≳ 6.2 × 1014 M⊙, selected for their disturbed dynamical state – inferred from existing X-ray observations. Our observations reached rms noise values between 20 and 23.5 μJy beam−1, with ∼ 4′′ angular resolution. No evidence of diffuse emission is found at ii full resolution. Low-resolution (∼ 30′′) images achieved rms noise levels of 30-50 μJy beam−1, amongst the deepest observations of high-redshift targets. One radio halo was detected in the least massive cluster PSZ2G254.08-58.45 extending over ∼ 500 kpc, with a 1.20 } 0.08 mJy integrated flux density. We placed a ∼1 mJy upper limit at 95% confidence level on the radio halo flux density for the other two targets. The radio-halo detection is consistent with the recent P1.4 GHz − M500 correlation from Cuciti et al. (2021b), while the upper limit on PSZ2G255.60-46.18 is consistent with being on the correlation. On the other hand, the upper limit on PSZ2G277.76-51.74 places the radio halo well below the correlation. Recently a 1.5 GHz survey (Giovannini et al., 2020) detected a slightly higher fraction of radio halos in clusters in the same redshift range, with power and size typically higher than what we found in our observations. Both observations are, however, not inconsistent with each other. Distribution and conservation status assessment of the freshwater fishes in the Krom River system in the Eastern Cape Province, South Africa

- Description: The Cape Fold Ecoregion (CFE) contains the highest number of endemic freshwater fishes in Africa, south of the Zambezi River system. The CFE’s unique fish fauna has, however, suffered severe decline in historical distribution ranges and population sizes due to multiple impacts, with the two main threats being introduced invasive piscivores and habitat degradation. Growing evidence shows that biodiversity is being lost at unprecedented levels due to increased human pressure and demand for limited resources. Future projections also indicate high extinction risk, particularly for freshwater ecosystems, as a result of global climate change. Yet for many regions, including the CFE, there is still limited biodiversity knowledge to guide decision making processes. The Krom River system in the eastern Cape Fold Ecoregion (CFE) contains three native freshwater fish species. These are Pseudobarbus senticeps, which was recently revalidated, Sandelia capensis and a galaxiid lineage currently informally referred to as Galaxias zebratus sp. ‘Joubertina’. Two of these taxa are listed under threatened categories of the International Union for Conservation of Nature (IUCN). These are P. senticeps, which is listed as Critically Endangered and Galaxias zebratus sp. ‘Joubertina’, which is listed as Endangered. Although S. capensis is currently listed as Data Deficient, the most recent information indicates that the species comprises three allopatric lineages, with the Krom River population belonging to a lineage that is distributed across a number of river systems on the south coast. Two non-native fishes, Micropterus salmoides and Lepomis macrochirus, have also been introduced into this river system. Updated information on distribution patterns and habitat utilisation is required to guide authorities to develop effective management plans and conservation of the threatened fishes of the Krom River system. The aim of this study was to determine the current distribution of the fishes of the Krom River system and to provide recommendations for in situ conservation measures to protect remnant populations and promote recovery and range expansion of the native fishes. The primary objective of the study was to use historical and present data to map the distributions patterns. Data on the distribution of the fishes of the Krom River system were obtained from various sources, including the NRF-SAIAB database, studies published in peer reviewed literature and unpublished reports. Systematic sampling was conducted in the Krom River system and several of its tributaries to provide a snapshot of the current distribution of both native and non-native fishes in this system at the time of this study. The presence of instream physical barriers was recorded, and habitat and water quality were visually assessed. Only four species of freshwater fishes were recorded during the surveys. These were two native species, the Krom River redfin P. senticeps and the Cape kurper Sandelia capensis, and two introduced species, the black bass M. salmoides and bluegill sunfish L. macrochirus. Pseudobarbus senticeps was the most common and widely distributed species in the system, and was recorded at 20 localities (four mainstem and 16 tributary localities). Sandelia capensis was less common and was recorded at only eight localities (four mainstem and four tributary localities). These two native species co-occurred at all the eight localities where S. capensis was recorded, but P. senticeps was always more abundant than Sandelia. Micropterus salmoides was recorded at six localities (four mainstem and 2 tributary localities) while L. macrochirus was recorded at five localities (four mainstem and one off-stream dam site). Native fishes were not recorded at sites where non-native fishes were present except at two localities in the Wit Els River where juvenile M. salmoides were found amongst the native fish samples. However, unlike the other localities where all size classes (i.e., young of the year, juveniles, subadults and adults) were present, only adult redfins were found in the Wit Els River where juveniles of largemouth bass occurred. A comparison of the past and present distribution patterns of the native fishes indicates a considerable decline in distribution range, and remnant populations are now fragmented compared to past observations. The major threats and impacts on the Krom River system are the presence of non-native piscivores, construction of instream physical barriers, and agricultural activities. The information from this study could form a basis for establishing long-term conservation measures that should focus on preventing the spread of non-native fishes, and rehabilitating critical habitats for the future persistence of remnant populations of native fishes. The study also evaluated the implications of incomplete taxonomy on conservation status assessments and prioritisation, by evaluating case studies of species complexes of freshwater fishes whose taxonomy has been recently resolved, as well as two complexes with lineages that await formal recognition as distinct species. The aim was to demonstrate how incomplete knowledge of taxonomy affects the assessment of extinction risk and can potentially misdirect conservation prioritisation. Specifically, the study examined how change in taxonomy or recognition of undescribed genetic lineages in faunal listings affects range size and IUCN Red List risk category. The study taxa were Pseudobarbus afer sensu lato (sl), Enteromius anoplus sl, Amphilius natalensis sl, Sandelia capensis and S. bainsii, and the study assessed the species complexes as Least Concern or Near Threatened. The majority of these taxonomically revised species and genetic lineages were determined to be Critically Endangered (CR), Endangered (EN) and Vulnerable (VU), as the extent of occurrence was estimated to be < 100 km2; < 5000 km2 and < 20 000 km2 respectively. The species and lineages have been observed in fewer than ten locations, and the populations have been observed to be in decline. The results of this study indicate that the current taxonomic status of the native species in the Krom River system obscures the diversity of these fishes and affects conclusions for conservation assessments. - Description: The Cape Fold Ecoregion (CFE) contains the highest number of endemic freshwater fishes in Africa, south of the Zambezi River system. The CFE’s unique fish fauna has, however, suffered severe decline in historical distribution ranges and population sizes due to multiple impacts, with the two main threats being introduced invasive piscivores and habitat degradation. Growing evidence shows that biodiversity is being lost at unprecedented levels due to increased human pressure and demand for limited resources. Future projections also indicate high extinction risk, particularly for freshwater ecosystems, as a result of global climate change. Yet for many regions, including the CFE, there is still limited biodiversity knowledge to guide decision making processes. The Krom River system in the eastern Cape Fold Ecoregion (CFE) contains three native freshwater fish species. These are Pseudobarbus senticeps, which was recently revalidated, Sandelia capensis and a galaxiid lineage currently informally referred to as Galaxias zebratus sp. ‘Joubertina’. Two of these taxa are listed under threatened categories of the International Union for Conservation of Nature (IUCN). These are P. senticeps, which is listed as Critically Endangered and Galaxias zebratus sp. ‘Joubertina’, which is listed as Endangered. Although S. capensis is currently listed as Data Deficient, the most recent information indicates that the species comprises three allopatric lineages, with the Krom River population belonging to a lineage that is distributed across a number of river systems on the south coast. Two non-native fishes, Micropterus salmoides and Lepomis macrochirus, have also been introduced into this river system. Updated information on distribution patterns and habitat utilisation is required to guide authorities to develop effective management plans and conservation of the threatened fishes of the Krom River system. The aim of this study was to determine the current distribution of the fishes of the Krom River system and to provide recommendations for in situ conservation measures to protect remnant populations and promote recovery and range expansion of the native fishes. The primary objective of the study was to use historical and present data to map the distributions patterns. Data on the distribution of the fishes of the Krom River system were obtained from various sources, including the NRF-SAIAB database, studies published in peer reviewed literature and unpublished reports. Systematic sampling was conducted in the Krom River system and several of its tributaries to provide a snapshot of the current distribution of both native and non-native fishes in this system at the time of this study. The presence of instream physical barriers was recorded, and habitat and water quality were visually assessed. Only four species of freshwater fishes were recorded during the surveys. These were two native species, the Krom River redfin P. senticeps and the Cape kurper Sandelia capensis, and two introduced species, the black bass M. salmoides and bluegill sunfish L. macrochirus. Pseudobarbus senticeps was the most common and widely distributed species in the system, and was recorded at 20 localities (four mainstem and 16 tributary localities). Sandelia capensis was less common and was recorded at only eight localities (four mainstem and four tributary localities). These two native species co-occurred at all the eight localities where S. capensis was recorded, but P. senticeps was always more abundant than Sandelia. Micropterus salmoides was recorded at six localities (four mainstem and 2 tributary localities) while L. macrochirus was recorded at five localities (four mainstem and one off-stream dam site). Native fishes were not recorded at sites where non-native fishes were present except at two localities in the Wit Els River where juvenile M. salmoides were found amongst the native fish samples. However, unlike the other localities where all size classes (i.e., young of the year, juveniles, subadults and adults) were present, only adult redfins were found in the Wit Els River where juveniles of largemouth bass occurred. A comparison of the past and present distribution patterns of the native fishes indicates a considerable decline in distribution range, and remnant populations are now fragmented compared to past observations. The major threats and impacts on the Krom River system are the presence of non-native piscivores, construction of instream physical barriers, and agricultural activities. The information from this study could form a basis for establishing long-term conservation measures that should focus on preventing the spread of non-native fishes, and rehabilitating critical habitats for the future persistence of remnant populations of native fishes. The study also evaluated the implications of incomplete taxonomy on conservation status assessments and prioritisation, by evaluating case studies of species complexes of freshwater fishes whose taxonomy has been recently resolved, as well as two complexes with lineages that await formal recognition as distinct species. The aim was to demonstrate how incomplete knowledge of taxonomy affects the assessment of extinction risk and can potentially misdirect conservation prioritisation. Specifically, the study examined how change in taxonomy or recognition of undescribed genetic lineages in faunal listings affects range size and IUCN Red List risk category. The study taxa were Pseudobarbus afer sensu lato (sl), Enteromius anoplus sl, Amphilius natalensis sl, Sandelia capensis and S. bainsii, and the study assessed the species complexes as Least Concern or Near Threatened. The majority of these taxonomically revised species and genetic lineages were determined to be Critically Endangered (CR), Endangered (EN) and Vulnerable (VU), as the extent of occurrence was estimated to be < 100 km2; < 5000 km2 and < 20 000 km2 respectively. The species and lineages have been observed in fewer than ten locations, and the populations have been observed to be in decline. The results of this study indicate that the current taxonomic status of the native species in the Krom River system obscures the diversity of these fishes and affects conclusions for conservation assessments. Factors influencing the number of non-timber forest product species used at the individual, household and village levels in Vhembe, Limpopo Province, South Africa

- Description: Non-Timber Forest Products (NTFPs) are wild biological resources obtained from natural landscapes, agricultural systems, and rural and urban regions for a variety of uses. NTFPs contribute significantly to livelihoods and living standards by offering a variety of socioeconomic benefits such as food, health, income generation, and safety nets. These multiple benefits are provided by a wide diversity of plant, animal and fungal species in different settings. However, the precise number of NTFP species used depends on the scale of assessment, namely use by individuals, by households and by villages. Consequently, it is difficult to compare numbers of species used between studies because of the differing scales. As a result, the objective of this study was to determine the number of NTFP species used at three different scales and how each is correlated with appropriate scale-dependent contextual variables. This objective was met by sampling individuals and whole households in five villages in the Vhembe District in Limpopo Province, South Africa. Twenty households were randomly selected in each village, providing a total sample of 100 households and 179 individuals in those households. In each household, every adult member was interviewed individually about the NTFPs that they used or knew. The major questions covered the NTFP species used, the respondents' socioeconomic level, the NTFP species used previously, and if the number of NTFP species are changing in the landscape. Data were reported per individual as well as per household and per village. The household level was derived by adding the responses of all the adults in each household but excluding duplicates. The same approach was used at the village scale, namely, the data across the 20 households were pooled, and duplicates were omitted. Patterns of NTFP use were examined by type of NTFP, and analysis using standard statistical tests, along with a Principal Components Analysis and a multivariate Detrended Correspondence Analysis of the NTFP species composition across villages. A total of 275 species of NTFPs were mentioned across the entire sample, including 81 species of firewood, 48 species of edible insects, 43 species of wild vegetables, 40 species of edible fruits, 38 species of wood utensils, 28 species of medicinal plants, 23 species of thatch grass, 19 species of poles, 19 species of wood for carvings, nine species of reeds, eight species of mushroom, and eight species of fodder. Of these species, some species had more than one use. Approximately 90% of the species used by the respondents were native species. Wild vegetables (e.g., Corchorus olitorius, Amaranthus capensis, Bidens pilosa), firewood (e.g., Colophospermum mopane, Parinari curatellifolia, Combretum hereroense), wild fruit (e.g., Landolphia kirkii, Vangueria infausta), and edible insects (e.g., Imbrasia belina, Macrotermes michaelseni, Encosternum delegorguei) were the most often used NTFPs by communities in the Vhembe area. Most of the households relied on NTFPs for subsistence. At the scale of an individual, the mean number of NTFP species used was 26.8±12.6 across the five villages. When considering a household, the mean number was 36.1±10.7 species. When aggregated across the 20 households per village, the mean number per village was 105.2±34.1 species. The species accumulation curve showed that 20 households per village were reasonably representative for four of the five villages. The results from the principal component analysis (PCA) and correlation analysis revealed that age, education, and employment status were significantly associated with the number of species used by individuals, whereas gender showed no clear correlation. At the household scale, household size was positively related with the number of species used, but income level and income source were not. At the village scale, attributes such as proximity to a town, settlement type, vegetation type, and distance to a nearby savanna all had some influence on the number of NTFP species listed. Most of the respondents felt that the number of species used had increased over the last 10–15 years, although some stated the opposite. Increases were attributed by traditional councils bans, the establishment of protected areas, and restoration. Activities eroding the number of NTFP species were identified as clear felling for farming, climate change, and over-harvesting.
