- Description: In South Africa, the baculovirus Cryptophlebia leucotreta granulovirus (CrleGV) is used to control the citrus pest Thaumatotibia leucotreta (Meyrick) (Lepidoptera: Tortricidae). However, the risk associated with continuous application of a single active ingredient is resistance. In order to manage resistance should it occur in field populations of T. leucotreta in South Africa, five new CrleGV genotypes from geographically distinct insect populations which were shown to exhibit some degree of phenotypic variation were recovered and genetically characterized. In droplet bioassays using seven CrleGV isolates against five T. leucotreta populations, some isolates were found to show higher virulence to some host populations than others. There were marked differences in the LD50 values of isolates and the number of occlusion bodies required per larva ranged between 0.79 and 3.12. The significance of these findings with respect to the application of CrleGV biopesticides and the management of resistance is discussed.

Single restriction endonuclease (REN) analysis of viral genomic DNA extracted from purified occlusion bodies showed that isolates differed in their DNA profiles. Partial sequencing of granulin and egt genes from the different isolates and multiple alignments of nucleotide sequences revealed the presence of single nucleotide polymorphisms (SNPs), some of which resulted in amino acid substitutions in the protein sequence. Based on these findings as well as comparisons with other documented CrleGV isolates, we propose two phylogenetic groups for CrleGV-SA isolates recovered in this study.

This study compared a variety of phenotypic traits between geographically distinct T. leucotreta populations collected from the Addo, Marble Hall, Citrusdal and Nelspruit regions of South Africa, and reared under laboratory conditions for several generations. Traits including pupal mass, female fecundity, egg hatch, pupal survival, adult eclosion and developmental time were used as parameters to measure biological performance and fitness. Insects from the Citrusdal region of the Western Cape exhibited significantly lower pupal mass, female fecundity, egg hatch, pupal survival, adult eclosion and the longest duration in larval and pupal development compared to the other colonies investigated. This is the first study to report differences in the performance of laboratory reared T. leucotreta from different geographic locations, and the findings may have important implications for the application of viral biopesticides for the control of this pest in South Africa.

In this study, we examined the effects of habitat condition (shadedvs. full-sun habitats) on plant traits and leaf characteristics of the invasive alien plant, Chromolaenaodorata (L.) King and Robinson (Asteraceae). In addition, the performance was evaluated in two generations of a specialist folivore, Pareuchaetes insulata (Walker) (Lepidoptera: Erebidae: Arctiinae), on leaves obtained from both shaded and full-sun habitats. The study was done in an area where the insect was introduced as a biological control agent. Leaves growing in shade were less tough, had higher water and nitrogen content, and lower total non-structural carbohydrate, compared with leaves growing in full sun. Plants growing in shade had longer leaves and were taller, but above-ground biomass was significantly reduced compared with plants growing in full sun. In both generations (parents and offspring), P. insulata developed faster and had larger pupal mass, increased growth rate, and higher fecundity when reared on shaded foliage compared with full-sun foliage. Although immature survival and adult longevity did not differ between habitats, Maw’s host suitability index indicated that shaded leaves were more suitable for the growth and reproduction of P. insulata. We suggest that the benefits obtained by P.insulata feeding on shaded foliage are associated with reduced toughness and enhanced nitrogen and water content of leaves. These results demonstrate that light-mediated changes in plant traits and leaf characteristics can affect insect folivore performance.

The efficiency of the sensitised photo-oxidative degradation of polychlorophenols depends not only on singlet oxygen quantum yield of sensitiser, but also on its stability. The influence of substrate structure and the pH of the solution on the photo-oxidation efficiency, as well as on the sensitiser photodegradation were studied. It was found that the contribution of the Type II (singlet oxygen-mediated) mechanism to the relative efficiency of the photosensitised photo-oxidation of the phenol, decreased from phenol to p-chlorophenol, TCP and PCP, whereas Type I radical pathway increased. The results obtained for PCP give evidence that electron transfer from the excited sensitiser to the substrate in its unionised form is responsible for enhanced sensitiser photodegradation.

Using cyclic voltammetry (CV) and Osteryoung square wave voltammetry (OSWV), hydrazine was detected over a linear concentration range of 1.3 × 10−5 to 9.2 × 10−5 mol/L with low limits of detection (ca. 5 and 11 μM for OSWV and CV, respectively). At concentrations higher than 1.2 × 10−4 mol/L the anodic peak potential shifted to 0.40 V (versus Ag|AgCl), and this was interpreted to be due to kinetic limitations resulting from the saturation of hydrazine and its oxidation products onto the redox-active monolayer film. This type of metallophthalocyanine-SAM-based electrode is a highly promising electrochemical sensor given its ease of fabrication, good catalytic activity, stability, sensitivity and simplicity.

- Description: Electrochemical properties of novel, peripherally substituted zinc phthalocyanine complex, octa(4-methylphenylthio-) phthalocyaninatozinc(II) [ZnPc(SC6H4CH3)8] (1a) in DMF solution are presented. This complex showed five quasi-reversible/reversible, diffusion-controlled redox couples. Solution voltammetry of 1a showed little contrast with that of its alkythiol-derivative, octabutylthiophthalocyaninatozinc(II) [ZnPc(SC4H9)8] (1b) in that both thiol-substituents tend to show electron-withdrawing influence on the phthalocyanine ligands; complex 1a showing easier reduction and more difficult to oxidation when compared to 1b and other alkyl derivatives. The voltammetric features of the solid ultrathin films of 1a and 1b, immobilized on gold electrodes via the self-assembling technique, are also presented. Interestingly, the self-assembled films are stable and reproducible and provide good suppression to the following Faradaic processes; gold surface oxidation, solution ion species and underpotential deposition (UPD) of copper.

- Description: Surface electrochemical behaviour of a self-immobilised iron phthalocyanine (FePc) ultrathin film, via axial ligation reaction, onto a preformed 4-mercaptopyridine self-assembled monolayer on gold electrode has been described. Electrochemical evidence for the sensor clearly suggested surface-confined, flat “umbrella”-oriented and densely-packed monolayer film structure. The proposed electrochemical sensor exhibited good catalytic activity towards the oxidation of thiocyanate in pH 4.0 medium over a linear range of three decades of concentration (ca. 10−6–10−3 mol dm−3) with a detection limit in the order of ∼10−7 mol dm−3. The sensor exhibited useful potential for the analysis of thiocyanate in human urine and saliva samples. The advantageous properties of this type of electrode as a sensor for thiocyanate lie in its ease of fabrication, excellent catalytic activity, stability, sensitivity and simplicity.

Surface electrochemical parameters of the modified electrodes showed that these MPc-SAMs are densely packed with flat orientations. The electrochemical, electrocatalytic and stability properties of these MPc complexes follow this order: FePc > MnPc > CoPc. This finding is remarkable as it suggests that the success of using this method of self-assembling of MPc onto gold electrode is largely dependent on the bond distance between the pyridine linker and the central metal of the MPc; the shorter the distance, the faster the co-ordination and the better the electrocatalytic properties towards L-cysteine and thiocyanate. Thus, the superiority of FePc-based SAM over those of the MnPc and CoPc, has been proposed to be the result of the more favorable axial co-ordination properties of FePc with pyridine (i.e. shorter Fe–N(pyridine) bond length.
