- Description: The bacterium Clostridium acetobutylicum produces acetone and n [subscript] - butanol from molasses in an industrial fermentation system. Although the bacterium has been cultured in liquid media it does not grow well on agar plates and requires high concentrations of hydrogen. Pretreatment of agar plates with bovine catalase improves growth on agar media. The bacteria produce an area of clearing (halo) on Potato agar plates due to butyric acid (the precursor of n [subscript]-butanol) and ß -amylase production. This characteristic will be used as a plate screening assay for the selection of high solvent producing mutants. A laboratory scale fermentation system was developed and detailed studies including pH, turbidity and cell morphology changes, and the details of solvent production were undertaken. The fermentation was optimized for mutant selection. The production of normal solvent yields by isolated clones is required for the mutant selection programme. Studies revealed that sporulation of the clones increased their solvent yield although solvent yields were still lower than normal. Efficient sporulation is therefore a prerequisite for clone fermentation. The origin of the phage infection during the factory outbreak was determined and resistant clones obtained. The presence of a bacteriocin-like toxin causing decreases in turbidity was identified during the final fermentation stage. The strain sensitivity, optimum conditions for stability as well as the kinetics of inactivation and lethality have been investigated. Preliminary characterization and purification studies indicate the proteinaceous nature of the toxin. , KMBT_363

It has been suggested that palmiet (Prionium serratum, Thurniaceae) works as an ecosystem engineer, shaping peat wetlands in South Africa. However, there is currently a paucity of evidence supporting this claim. Palmiet has a dense root, rhizome and stem system that forms dense stands, growing from channel banks into fast flowing river channels. This slows river flows, traps sediment, which builds up riverbeds and ultimately blocks river channels, turning the river into a wetland. The aim of this study was to determine if palmiet is an ecosystem engineer and to document its pattern of colonisation and the nature of its control of a fluvial system. This was achieved by undertaking vegetation surveys in the Kromrivier Wetland in the Eastern Cape. The data was analyzed using vegetation classification and ordination, where vegetation communities were linked to environmental factors. It was found that palmiet occupied three distinctive habitats; 1) on near-horizontal valley- bottom habitats filled with sediments that are a mixture of autochthonous organic sediment and allochthonous clastic fines, 2) the bed of gullies that have recently filled with coarse grained clastic sediment, and 3) open water bodies. Three conceptual models were developed, one that accounts for the process of gully bed colonisation, sediment trapping and gully filling, another involving rapid colonisation of sedimentary fill from tributary sediment sources that block a gully, and the third involving colonisation of open-water areas that form in former gullies upstream of the blockage. The study suggests that the wetland has been characterised by repeated cutting and filling cycles, despite which, palmiet has repeatedly reinstated diffuse flow conditions across the valley floor. Palmiet was indicated to exert a key control on fluvial form and dynamics of the wetlands in the Kromrivier valley.

Deep learning applied to the semantic segmentation of tyre stockpiles

This push is further intensified by the health and safety risks discarded tyres pose to the surrounding population. Waste tyre recycling initiatives in South Africa are on the increase; however, there is still a growing number of undocumented tyre stockpiles developing throughout the country. The plans put in place to eradicate these tyre stockpiles have been met with collection, transport and storage logistical issues caused by the remoteness and distant locales. Eastwood (2016) aimed at optimising the logistics associated with collection, by estimating the number of visible tyres from images of tyre stockpiles. This research was limited by the need for manual segmentation of each tyre stockpile located within each image. This research proposes the use of semantic segmentation to automatically segment images of tyre stockpiles. An initial review of neural network, convolutional network and semantic segmentation literature resulted in the selection of Dilated Net as the semantic segmentation architecture for this research. Dilated Net builds upon the VGG-16 classification architecture to perform semantic segmentation. This resulted in classification experiments which were evaluated using precision, recall and f1-score. The results indicated that regardless of tyre stockpile image dimension, fairly accurate levels of classification accuracy can be attained. This was followed by semantic segmentation experiments which made use of intersection over union (IoU) and pixel accuracy to evaluate the effectiveness of Dilated Net on images of tyre stockpiles. The results indicated that accurate tyre stockpile segmentation regions can be obtained and that the trained model generalises well to unseen images.

- Description: Background: The musculoskeletal demands placed on the lower limb musculature of fast bowlers over time have not received much attention. In particular, measures of muscle recruitment changes have, to the author’s knowledge, not been considered. Objective: The present study, therefore sought to establish any associations between an eight over, simulated fast bowling spell, and muscle activation patterns, power output, perceptual demands, and changes in performance. This will enable improvements in the development of training programmes. Methods: Players’ were required to attend two sessions in total. The purpose of the initial session was to collect specific demographic, anthropometric and physiological data and injury history information from each player. In addition, this first session allowed for habituation with the treadmill, the jump meter and all other equipment involved in experimentation. The second testing session involved electrode attachment sites being identified on player’s dominant leg. The areas were then shaved, wiped with an alcohol swab and left to dry, to ensure good connectivity. Pre- and post- measures of muscle activity and functional strength of the lower limbs were recorded in the Department of Human Kinetics and Ergonomics. The protocol took place at the Kingswood High Performance Centre, which is in close proximity to the initial testing site. The protocol involved players bowling eight overs (48 balls). During the protocol, accuracy, ball release speed and perceptual measures were recorded at the end of each over. After the protocol, players were driven back to the Human Kinetics and Ergonomics Department where post-testing measures were completed. The dependable variables of interest were muscle activation, functional strength of the lower limbs, ‘local’ ratings of perceived exertion (RPE), body discomfort, accuracy, and ball release speed. Results: For all muscles it was shown that, as the speed increased so did the muscle activity in players’ lower limbs. There were no significant changes in muscle activity preversus post-protocol. There was however, a general trend of decreasing muscle activity post protocol at higher testing speeds. There were significant (p<0.05) decreases in peak power following the simulated eight over bowling spell. ‘Local’ RPE displayed a significant (P<0.05) increase with each additional over and were observed to reach the ‘heavy’ category. The players’ highest discomfort area was in the lower back, with 13 players perceiving discomfort in this region following the eight over spell. The shoulder and chest were another two areas player’s indicated discomfort with eight players selecting the dominant shoulder. Seven players complained of the dominant side pectoral muscle, leading foot and dominant latissimus dorsi muscle being uncomfortable. Interestingly, the dominant pectoral showed the highest body discomfort ratings amongst players. There were no significant changes in accuracy between overs although there were large interindividual differences in accuracy points between players. The decrease in ball release speed observed during over seven was shown to be significantly (p<0.05) lower than overs one to four. Conclusion: The power output and perceived strain results of the players, appears to indicate the presence of fatigue in players. However, the results are not conclusive, as the fatigue was not shown in muscle recruitment patterns, as well as the body discomfort ratings. There was a non-significant trend observed in the lower limb muscle activation decreasing at higher speeds. Players were able to maintain accuracy. With many South Africans experiencing poverty and food insecurity, the National School Nutrition Programme aims to provide a social safety net and address short term hunger in children. However, challenges are faced regarding the implementation thereof, such as a lack of training of food handlers. This results in poor hygiene and food safety knowledge and behaviour. Subsequently, the risk of transmission of foodborne pathogens and the contamination of food rises. Foodborne illnesses are a major cause of morbidity and mortality, specifically in children, due to weak immune systems. Therefore, the study was conducted in order to develop and test a hygiene and food safety manual so that it can be made available to the South African Department of Education to be used by teachers in order to train the food handlers of the National School Nutrition Programme. The manual can then be used to train all newly recruited food handlers, and therefore ensure that school meals are safe to consume by learners.

Using an intelligent self-service solution to support telecommunication service provisioning

- Description: The increasing competition between telecommunication service providers (TSPs) and the large amounts of human resource costs incurred to provision telecommunication services to small, medium and micro enterprises (SMMEs), highlight the need for effective self-service provisioning methods to reduce costs. Existing service provisioning methods do not effectively support SMMEs in the service provisioning process. The current methods used are manual and Web-based. The manual service provisioning method is labour and time-intensive. TSPs are being forced to cut human resource costs as the profit margins for provisioning services are narrow due to competition. The current web-based systems do not allow SMMEs to effectively purchase services as they provide long textual lists to consult and limited information about services. The objective of this research was to develop an intelligent web-based self-service system to support SMMEs in telecommunication service provisioning, using a Design Science Research methodology. A constraint-based, knowledge-based recommender system was selected as the most appropriate intelligent system to support telecommunication service provisioning. A prototype called SelPro was developed as a solution to address the shortcomings of the current ineffective service provisioning methods available for SMMEs. SelPro incorporates an interactive wizard-style user interface with dynamic recommendations to support novice users as well as an expert interface which provides less user support. An expert review and a user study were conducted to determine if SelPro was usable, useful and effective. SelPro was perceived as being usable for SMMEs wishing to purchase telecommunication services. Expert participants were satisfied with the usability of SelPro and perceived it as being useful, easy to use and easy to learn. SelPro was considered effective because of the high task success ratings provided by the participants using the two interfaces. The research determined that intelligent web-based self-service provisioning can be used to effectively support SMMEs in telecommunication service provisioning. General design recommendations for similar systems were proposed and future work will involve using intelligent systems to support large enterprises in telecommunication service provisioning.

- Description: The increasing competition between telecommunication service providers (TSPs) and the large amounts of human resource costs incurred to provision telecommunication services to small, medium and micro enterprises (SMMEs), highlight the need for effective self-service provisioning methods to reduce costs. Existing service provisioning methods do not effectively support SMMEs in the service provisioning process. The current methods used are manual and Web-based. The manual service provisioning method is labour and time-intensive. TSPs are being forced to cut human resource costs as the profit margins for provisioning services are narrow due to competition. The current web-based systems do not allow SMMEs to effectively purchase services as they provide long textual lists to consult and limited information about services. The objective of this research was to develop an intelligent web-based self-service system to support SMMEs in telecommunication service provisioning, using a Design Science Research methodology. A constraint-based, knowledge-based recommender system was selected as the most appropriate intelligent system to support telecommunication service provisioning. A prototype called SelPro was developed as a solution to address the shortcomings of the current ineffective service provisioning methods available for SMMEs. SelPro incorporates an interactive wizard-style user interface with dynamic recommendations to support novice users as well as an expert interface which provides less user support. An expert review and a user study were conducted to determine if SelPro was usable, useful and effective. SelPro was perceived as being usable for SMMEs wishing to purchase telecommunication services. Expert participants were satisfied with the usability of SelPro and perceived it as being useful, easy to use and easy to learn. SelPro was considered effective because of the high task success ratings provided by the participants using the two interfaces. The research determined that intelligent web-based self-service provisioning can be used to effectively support SMMEs in telecommunication service provisioning. General design recommendations for similar systems were proposed and future work will involve using intelligent systems to support large enterprises in telecommunication service provisioning.

These components are often volatile in nature and dissipate in a matter of hours. When considering the different types of slow release systems, an overwhelming variety of these systems is available. Each one of the systems is unique in a way, and is designed to perform a particular function, whether it facilitates the controlled release of an active into the body via the skin surface (transdermal delivery) or whether it reduces the rate of loss of an active from the skin surface to the surrounding environment. For the purpose of this study, a previously existing fixative formulation which is believed to reduce the rate of loss of an active component to the environment, through film formation on the skin surface, was investigated. Alternative ingredients or components were incorporated together with the original fixative formulation ingredients into an experimental design which investigates the effect of each group of the components present. 18 formulations with various concentrations of the components within the groups and specified upper and lower limits for each component were formulated. The fixative properties of the formulations were analysed through the incorporation of a fixed amount of a simple fragrance molecule, 4- methoxybenzaldehyde, into each formulation and evaporation studies were conducted in an environmental room at 28±1° C over a period of 5 hours followed by gas chromatography analysis and finally data analyses using statistical methods. The most efficient fixative formulation was established using regression analysis. The fragrance compound in this formulation was found to evaporate at a rate of 0.47 g/L per hour. The least efficient fixative formulation lead to the loss of 0.78 g/L of the fragrance component per hour. From the calculated fragrance concentrations, the rate constant for each individual fixative formulation could be calculated and response surface 8 modelling by backward regression was used in order to determine how each component contributes to the rate of loss of the fragrance compound. Since the sum of the original ingredient and its alternative was constant, each of the original ingredients was coupled directly to its alternative and no conclusion could be made about the contribution of individual components. By increasing the concentration of Hydroxypropylcellulose (HPC) 100K and its alternative HPC 140K, while keeping the effects of the other components constant, a decrease in the rate of fragrance loss was observed. The same conclusion could be made when increasing the concentrations of PEG-12 Dimethicone and its alternative cetyl dimethicone (decreases the evaporation rate). An interaction took place between HPC 100K and PEG-12 dimethicone and their alternatives. The negative effect was, however, not as strong as the combined positive effect on the rate of fragrance loss of the individual components HPC and PEG-12 dimethicone. Evidence suggested that the removal of the components polyvinylpyrrolidone and its alternative, polyurethane-32 (Baycusan® C1003), would improve the effectiveness of the fixative formulation in terms of its slow release properties. - Description: Background: The steady increase in deaths due to non-communicable diseases (NCDs) in South Africa is raising concern, as 63% of all deaths worldwide are attributed to NCDs and these diseases cause 36 million deaths each year, with 80 % of incidences occurring in low-and middle-income countries. In rural communities where income is increasing, the prevalence of NCDs are increasing as well. Due to the lack of NCD prevention and treatment in rural communities, it is expected that, if health promotion strategies are not implemented and socio-economic development in South Africa continues, there will soon be an uncontrollable NCD epidemic. It has been found that dietary risks affected people’s healthirrespective of their age, gender or residential sociodemographic development and that dietary improvement might prevent one in every five deaths worldwide.Aim:The aim of this study was to determine the factors that influence the perceptions and behaviour in a rural community with a low socio-economic status regarding dietary habits and to use the knowledge to formulate an effective dietary modification strategy.Designand methodology:For this study,action research with a quantitative and qualitative component was followed.The quantitative component focuses on measureable aspects, and the qualitative component focuses on meaning. Data was collected following the standard operating procedure for anthropometry, blood pressure and blood glucose. Further methods used were questionnaires, 24-hour dietary recalls and focus group discussions. Data were analysed using Microsoft Excel, FoodFinder3 and other calculations.Sample and setting:A convenience sample (n = 16) wasused and this study was undertaken on request of the non-governmental organization, Karoo Angels and included all participants who earn an income and who do not work for themselves, that were willing to participate and living in Rietbron and Vondeling, in the Sarah BaartmanDistrict in the Eastern Cape. Results:Weight changes took place, both the crafters in the mild thinness category gained weight, one gained 3.9 kg and the other 0.5 kg. The three crafters in the normal category did not have to gain or lose weight, but all three ofthem gained an average of 3.6 kg. For the group who had to lose weight, no statistically significant weight change took place from before the intervention (76.08 ±17.67 kg) to after the intervention (75.81 ± 17.81 kg, t(10) = 0.692, p = 0.505). The maximum weight lost ivwas 3.1 kg and the maximum weight gained was 3.2 kg. Changes took place regarding BMI, waist circumference, waist-to-hip ratio and neck circumference. Some positively changed their BMI status.After the implementation of the intervention, there was a slight decrease in frequency of alcohol consumption, but smoking and physical activity remained mostly the same. Hypertension was the most prevalent NCD and is linked to smoking and alcohol consumption which is prevalent in this study. Fruit and vegetable consumption is low, and a slight increase in consumption was evident. A maximum dietary diversityscore of seven and minimum of two, where the mean was 3.81was found. The crafters’ diet did not meet the DASH diet recommendation,the mean energy distribution is of all macronutrients differ with ±4% and none of the participants’ mean micronutrients intake met the recommendations of the DASH diet or RDA. Factors that prevent them from reaching their goals included low income, low availability of fruit, vegetables and transport and no storage for food.Conclusion:It is important to consider the individuals’ living conditions, resources, knowledge, motivation and social group. The environment and resources available have a significant impact on the lifestyle of the individuals. Directing efforts of behaviour change to increase dietary diversity as the main objective, might be a more efficient way to change behaviour.Recommendations:Future studies should consider doing this study over a longer time period. The involvement of a psychologist would also be beneficial to assist the process of behaviour change.

- Description: Cyclopia intermedia E. Mey is harvested from the wild to produce honeybush tea. The presence of a lignotuber and its slow growth seems to hinder the cultivation of this species and wild harvesting is likely to continue. Species distribution modelling indicated that C. intermedia has a climate envelope that spans from the Eastern Cape to the Western Cape along the Cape Fold mountains. This regional distribution is threatened under future climate change scenarios with a range loss and shift identified for climate conditions predicted for 2050. More specifically, its niche was identified as south-facing slopes with sandy to loamy sand soils and water holding capacity that is higher than the surrounding areas. Some keystone Fynbos species were found to share this niche: Leucadendron salignum, Leucospermum cuneiforme, Protea neriifolia, Protea repens, Elegia filacea and Rhodocoma fruticosa. Cyclopia intermedia grows throughout the summer with flowering buds developing in autumn. These enlarge during winter with the flowers opening in early spring. Pods develop in November and seed set occurs during December. Harvesting plants increases their fecundity with cut plants producing more than twice the number of pods and seeds compared to their non-harvested counterparts. This is because resprouting plants produce more flowering stems than uncut plants. Controlled harvesting or localised short-cycle burning could be considered as a management option to improve fecundity of declining Cyclopia intermedia populations in protected areas. C. intermedia harvesting could occur every third year and stem colour was identified as a measure of harvest maturity. Where at least 30 percent of the population has orange (RHS colour 163A) stems the population is suitable for harvesting.

- Description: The black wildebeest Connochaetes gnou, a southern African endemic largely confined to the Grassland biome, was used as a model species to assess the response of a specialised grazer to grass availability, as an indicator of habitat quality. The occurrence of animals in low-quality habitats may have detrimental effects on population performance and persistence. Robust techniques are therefore required to assess the response of animals to varying habitat quality, to ensure sustainability of populations and species. Black wildebeest habitat use was assessed in relation to grass availability and visibility, and faecal analysis was used to estimate variation in diet composition and quality, parasite loads, and faecal pellet dimensions, across a grass gradient. Grass consumption, diet quality, and faecal pellet size and weight of black wildebeest increased with increasing grass availability. While black wildebeest consumed up to 55% browse at grass-poor sites, they consistently preferred grasses, potentially resulting in an increased intake of low-quality grasses at grass-poor sites. This, combined with the increased intake of indigestible fibres in browse, may explain the observed trends in diet quality, which in turn may explain variation in faecal pellet size and weight (indicators of body size and weight). Furthermore, black wildebeest population trends within the study area suggest that populations in grass-poor habitats, outside their natural distribution range, may exhibit reduced population performance. These findings suggest that grass availability may be a useful indicator of habitat quality for specialised grazers. The study also provided an opportunity to explore the utility of various diet assessment techniques for estimating herbivore diet composition and quality. Caution should be applied when interpreting and comparing estimates of diet composition and quality that were estimated using different techniques, because different techniques may yield different diet estimates. The study also suggests that black wildebeest may switch to C3 grasses in the winter (as indicated by isotope analysis).

A comprehensive evaluation framework for system modernization : a case study using data services

- Description: Modernization is a solution to migrate cumbersome existing systems to a new architecture for improved longevity of business processes. Three modernization approaches exist. White-box and black-box modernization are distinct from one another. Grey-box modernization is a hybrid of the white-box and black-box approaches. Modernization can be utilised to create data services for a Service Oriented Architecture. Since it is unclear which modernization approach is more suitable for the development of data services, a comprehensive evaluation framework is proposed to evaluate which of the white- or black-box approaches is more suitable. The comprehensive framework consists of three evaluation components. Firstly, developer effort to modernize existing code is measured by acknowledged software metrics. Results indicated that had only a single evaluation component been used, inconclusive results of the more suitable approach may have been obtained. The findings of this research contribute a comprehensive evaluation framework which can be applied to compare modernization approaches and measure modernization success.

- Description: Modernization is a solution to migrate cumbersome existing systems to a new architecture for improved longevity of business processes. Three modernization approaches exist. White-box and black-box modernization are distinct from one another. Grey-box modernization is a hybrid of the white-box and black-box approaches. Modernization can be utilised to create data services for a Service Oriented Architecture. Since it is unclear which modernization approach is more suitable for the development of data services, a comprehensive evaluation framework is proposed to evaluate which of the white- or black-box approaches is more suitable. The comprehensive framework consists of three evaluation components. Firstly, developer effort to modernize existing code is measured by acknowledged software metrics. In 2017, TB was responsible for an estimated 1.3 million deaths among both the HIV negative and positive populations worldwide (WHO, 2018). Approximately 23% (roughly 1.7 billion) of the world’s population is estimated to have latent TB with a high risk of reverting to active TB infection. In 2017, an estimated 558,000 people developed drug resistant TB worldwide with 82% of the cases being multi-drug resistant TB (WHO, 2018). South Africa is ranked among the 30 high TB burdened countries with a TB incidence of 322,000 cases in 2017 accounting for 3% of the world’s TB cases. TB is curable and is clinically managed through a combination of intensive and continuation phases of first-line drugs (isoniazid, rifampicin, ethambutol, and pyrazinamide). Second-line drugs which include fluoroquinolones, injectable aminoglycoside and injectable polypeptides are used in cases of first line drug resistance. The third-line drugs include amoxicillin, clofazimine, linezolid and imipenem. These have variable but unproven efficacy to TB and are the last resort in cases of total drug resistance (Jilani et al., 2019). TB drug resistance to first-line drugs especially isoniazid in M. tuberculosis has been attributed to single nucleotide polymorphisms (SNPs) in the catalase peroxidase enzyme (katG), a protein important in the activation of the pro-drug isoniazid. The SNPs especially at position 315 of the katG enzyme are believed to reduce the sensitivity of the M. tuberculosis to isoniazid while still maintaining the enzyme’s catalytic activity - a mechanism not completely understood. KatG protein is important for protecting the bacteria from hydro peroxides and hydroxyl radicals present in an aerobic environment. This study focused on understanding the mechanism of isoniazid drug resistance in M. tuberculosis as a result of high confidence mutations in the katG through modelling the enzyme with its respective variants, performing MD simulations to explore the protein behaviour, calculating the dynamic residue network analysis (DRN) of the variants in respect to the wild type katG and finally performing alanine scanning. From the MD simulations, it was observed that the high confidence mutations i.e. S140R, S140N, G279D, G285D, S315T, S315I, S315R, S315N, G316D, S457I and G593D were not only reducing the backbone flexibility of the protein but also reducing the protein’s conformational variation and space. All the variant protein structures were observed to be more compact compared to the wild type. Residue fluctuation results indicated reduced residue flexibility across all variants in the loop region (position 26-110) responsible for katG dimerization. In addition, mutation S315T is believed to reduce the size of the active site access channel in the protein. From the DRN data, residues in the interface region between the N and C-terminal domains were observed to gain importance in the variants irrespective of the mutation location indicating an allosteric effect of the mutations on the interface region. Alanine scanning results established that residue Leucine at position 48 was not only important in the protein communication but also a destabilizing residue across all the variants. The study not only demonstrated change in the protein behaviour but also showed allosteric effect of the mutations in the katG protein.

Secondly, we consider quadratic Hamilton-Poisson systems on the (minus) Lie-Poisson space se(1,1)*. These systems are classified up to an affine isomorphism. Six normal forms are identified for the homogeneous case, whereas sixteen representatives (including several infinite families) are obtained for the inhomogeneous systems. Thereafter we consider the stability and integration of the normal forms obtained. For all homogeneous systems, and a subclass of inhomogeneous systems, we perform a complete stability analysis and derive explicit expressions for all integral curves. (The extremal controls of a large class of optimal control problems on SE(1,1) are linearly related to these integral curves.) Lastly, we discuss the Riemannian and sub-Riemannian problems. The (left-invariant) Riemannian and sub-Riemannian structures on SE(1,1) are classified, up to isometric group automorphisms and scaling. The addition of an A₄ family symmetry and extended Higgs sector to the Standard Model can generate this mixing pattern, assuming the correct vacuum expectation value alignment of Higgs scalars. The effect of deviating this alignment is studied, for different types of A₄ models, with a phenomenological emphasis: the effect of perturbations on the model predictions for the neutrino oscillation and neutrino mass observables. The standard theoretical description of neutrino oscillations is presented, along with a summary of the past, present and future experimental efforts aimed at measuring the neutrino mixing parameters. Additionally, the current constraints on the sum of absolute neutrino masses and the amplitude for neutrinoless double beta decay, which is yet to be observed, are discussed. These constraints provide a model-independent test of family symmetery models. The Standard Model is reviewed, and extensions to the Standard Model such as the seesaw mechanism(s) are discussed: these are designed to endow neutrinos with mass, and can be incorporated into A₄ symmetry models. Models with different A₄ particle assignments are analysed for deviations from tribimaximal mixing. There are nine models presented in Chapter 5, with lepton doublets transforming as 3 (underlined) and right-handed charged leptons transforming as 1, 1', 1" (all underlined) ; five of these include right-handed neutrinos transforming as 3 (underlined) and make use of the seesaw mechanism. Chapter 6 contains the analysis of six models that assign all leptons to the 3 (underlined) representation, with four of these utilising the seesaw mechanism. The models are tested for any degree of fine tuning of the parameters that define the mass matrices. Studies have shown that plants grown above 600 ppm tend to have an increased growth rate and invest more in carbon-based defences. This has important implications for the management of invasive alien plants, especially for the field of biological control which is mostly dependent on herbivorous insects. This is because insects reared on such plants have been shown to have reduced overall fitness. Nevertheless, most of the studies on potential changes in plant-insect interactions under elevated CO2 are based on agricultural systems, with only a limited number of these types of studies conducted on alien invasive weeds. However, climate change and invasive species are two of the most prevalent features of global environmental change. Therefore, this also warrants active research and experimental studies to better understand how these systems will be affected by future climates. Thus, the aim of this study was to investigate the effects of elevated atmospheric CO2 on the biological control of four invasive aquatic weeds (Azolla filiculoides, Salvinia molesta, Pistia stratiotes, and Myriophyllum aquaticum). These species are a threat to natural resources in South Africa but are currently under successful control by their biological control agents (Stenopelmus rufinasus, Cyrtobagous salviniae, Neohydronomus affinis, and Lysathia n. sp.). To achieve this, the selected plant species were grown in a three-factor experimental design in winter (CO2 X nutrients X herbivory), and another two-factorial design in summer (CO2 X herbivory). Atmospheric CO2 concentrations were set at ambient (400 ppm) or elevated (800 ppm), as per the predictions of the IPCC. As per my hypothesis, the results suggest that these species will become more challenging in future due to increased biomass production, asexual reproduction and a higher C: N ratio which is evident under high CO2 concentrations. Although the biological control agents were in some instances able to reduce this CO2 fertilisation effect, their efficacy was significantly reduced compared with the levels of control observed at ambient CO2. These results suggest that additional biological control agents and other management methods may be needed for continued control of these invasive macrophytes, both in South Africa and further afield where they are problematic.

- Description: Background: Agriculture plays a vital role in the economy of any industrially developing country, including South Africa. In the Eastern Cape of South Africa citrus farming is a significant contributor to the local economy (Johnson et al., 2005). The harvesting phase of citrus farming is performed manually and exposes workers to physical risks, which can lead to the development of musculoskeletal disorders. In particular, the standard harvesting bag comprises of a single shoulder strap and promotes asymmetrical load carriage which results in shoulder and lower back pain complaints. The current study compared the physiological (EMG), perceptual (RPE), usability (PUEU) and productivity effects of two new harvesting bag designs (a hip belt and a backpack bag design) to the standard harvesting bag design. This was performed in a laboratory as well as a field setting. Methods (Laboratory phase): 36 participants (12 males and 24 females) were assigned to one worker group. The “tall ladder worker” group was comprised of only males and the “step ladder worker” and “ground worker” group of females. Each participant was required to simulate a citrus harvesting task while utilizing each of the bag designs on different days. On each day/test session, participants performed three harvesting cycles. Muscle activity was measured throughout the entire testing session and RPE were recorded at the end of each cycle. Results (Laboratory phase): The EMG and RPE results indicate that the backpack design was the most ideal design to reduce asymmetry, while the standard harvesting bag design was the worst. Although not significant, there was greater muscle asymmetry (p=0.109) and a significantly higher perceived exertion when using the standard bag (p=0.0004), in comparison to using the backpack. Methods (Field phase): 17 Xhosa-speaking citrus harvesters (6 females and 11 males) participated in this study. Each harvester worked with one of the three bag designs on a different day. Productivity of each worker was assessed every hour by recording the number of bags filled with fruit and at the end of the shift. A Perceived Usefulness & Ease of Use questionnaire was presented to each participant to obtain feedback on worker acceptance to the new bag designs. Results (Field phase): A general trend in support of the hip belt bag design over the other two bag designs were found, even within the different worker demographic groups (age, sex and worker experience). The workers perceived less exertion (7.98 ± 1.86) and were more productive (9.90 ± 2.11 bags/hour) when using the hip belt design; they also found this bag the most useful (1.02 ± 0.09) and easy to use (1.07 ± 0.25). In contrast, the backpack bag design had significantly poorer responses when compared to the other two bag designs and this was evident in all the dependent variables assessed (RPE, productivity and PUEU). Conclusion: The results from the laboratory phase supported the expectation that the backpack bag design reduces asymmetry and hence, is more suitable than the standard harvesting bag. - Description: Background: Agriculture plays a vital role in the economy of any industrially developing country, including South Africa. In the Eastern Cape of South Africa citrus farming is a significant contributor to the local economy (Johnson et al., 2005). The harvesting phase of citrus farming is performed manually and exposes workers to physical risks, which can lead to the development of musculoskeletal disorders. In particular, the standard harvesting bag comprises of a single shoulder strap and promotes asymmetrical load carriage which results in shoulder and lower back pain complaints. The current study compared the physiological (EMG), perceptual (RPE), usability (PUEU) and productivity effects of two new harvesting bag designs (a hip belt and a backpack bag design) to the standard harvesting bag design. This was performed in a laboratory as well as a field setting. Methods (Laboratory phase): 36 participants (12 males and 24 females) were assigned to one worker group. The “tall ladder worker” group was comprised of only males and the “step ladder worker” and “ground worker” group of females. Each participant was required to simulate a citrus harvesting task while utilizing each of the bag designs on different days. On each day/test session, participants performed three harvesting cycles. Muscle activity was measured throughout the entire testing session and RPE were recorded at the end of each cycle. Results (Laboratory phase): The EMG and RPE results indicate that the backpack design was the most ideal design to reduce asymmetry, while the standard harvesting bag design was the worst. Although not significant, there was greater muscle asymmetry (p=0.109) and a significantly higher perceived exertion when using the standard bag (p=0.0004), in comparison to using the backpack. Methods (Field phase): 17 Xhosa-speaking citrus harvesters (6 females and 11 males) participated in this study. Each harvester worked with one of the three bag designs on a different day. Productivity of each worker was assessed every hour by recording the number of bags filled with fruit and at the end of the shift. A Perceived Usefulness & Ease of Use questionnaire was presented to each participant to obtain feedback on worker acceptance to the new bag designs. Results (Field phase): A general trend in support of the hip belt bag design over the other two bag designs were found, even within the different worker demographic groups (age, sex and worker experience). The workers perceived less exertion (7.98 ± 1.86) and were more productive (9.90 ± 2.11 bags/hour) when using the hip belt design; they also found this bag the most useful (1.02 ± 0.09) and easy to use (1.07 ± 0.25). In contrast, the backpack bag design had significantly poorer responses when compared to the other two bag designs and this was evident in all the dependent variables assessed (RPE, productivity and PUEU). Conclusion: The results from the laboratory phase supported the expectation that the backpack bag design reduces asymmetry and hence, is more suitable than the standard harvesting bag. The use of building information modelling as a tool to improve informed design communication between student and lecturer during critique sessions

- Description: Within the design studio at the Department of Architecture, Nelson Mandela University, the timeous submission of design projects is a problem. After close inspection, it is noted that students spend weeks on conceptual design ideas with no progressions towards completion. The purpose of the study is thus to determine whether building information modelling (BIM), as opposed to the traditional method of design, can be used to improve informed design communication during conceptual design for the first-year master of architecture students and contribute to timeous design project submission. A cohort comparison rubric will be used as the method for looking at the qualitative attributes of design intention, function, aesthetics, and sustainability. The 2016 cohort made use of the traditional design method, whereas for the 2017 cohort the BIM course was introduced in the computer module with implementation in the design studio. The work of nine students from both cohorts was analysed and findings include the following: improved drawing type availability, accuracy, and quality; reduced time spent on redundant work; inclusive discussions compared to isolated critique sessions; reduced printing costs - and the establishment of “what if” scenarios early in the project. Based upon the research findings it can be concluded that BIM as a teaching strategy and process has the capability of improving the communication between student and lecturer in the conceptual stage, thereby leading to a more timeous submission. It is recommended that BIM education must be incorporated in the undergraduate programme thereby providing students with an appropriate platform for expressing conceptual ideas more effectively within the design course.
