Development of an enzyme-synergy based bioreactor system for the beneficiation of apple pomace lignocellulosic waste

- Description: Due to the finite supply of non-renewable fossil fuels, agro-industrial wastes are identified as alternate, renewable sources for energy supply. Large amounts of fruit waste are generated in South Africa due to fruit juice and wine processing from apples, grapes and citrus fruit. Apple pomace is the solid residue that is left over after juice, cider and wine processing and constitutes between 25-30% of the total fruit. On a global scale millions of tonnes of apple pomace are produced; between 2006-2007 over 46 million tonnes were produced. In South Africa a total production of 244 469 tonnes were produced during the 2011- 2012 season. Initially, apple pomace was regarded as a waste by-product used for animal feed and compost in soil, however presently it is considered a source of dietary fiber and natural antioxidants like polyphenols. In addition, apple pomace has a high carbohydrate content and can be enzymatically hydrolysed to produce sugar monomers which, in turn, can be fermented by yeasts to produce bioethanol. The polyphenols present in apple pomace can be used for their health properties, and the bioethanol can be used as a replacement for fossil fuel. Apple pomace is lignocellulosic in nature and consists of hemicellulose, cellulose, lignin and pectin. A combination of enzymes such as cellulases, hemicellulases, pectinases and lignases are required to operate in synergy for the degradation of lignocellulosic biomass. This is due to the recalcitrant nature of lignocellulose. This study investigated the degradation of apple pomace using a combination of commercially obtained enzyme cocktails viz. Viscozyme L , Celluclast 1.5L and Novozyme 188. The commercial enzymes Viscozyme L and Celluclast 1.5L were added in a ratio of 1:1 (50%:50%). The final concentrations of the enzymes were 0.019 mg/ml each. Novozyme 188 was added to provide a final concentration of 0.0024 mg/ml. A novel cost effective 20L bioreactor was designed, constructed and implemented for the degradation of apple pomace to produce value added products. The hydrolysis of the apple pomace was performed initially in 1 L flasks (batch fed) and, once optimized, scaled up to a 20 L bioreactor in batch mode. The bioreactors were operated at room temperature (22 ± 2ºC) and in an unbuffered system. The sugars released were detected and quantified using an optimized validated HPLC method established in this study. The sugars released in the bioreactors were mainly glucose, galactose, arabinose, cellobiose and fructose. The polyphenols released in this study were gallic acid, catechin, epicatechin, chlorogenic acid, rutin and phloridzin, which have a number of health benefits. The simultaneous analyses of the polyphenols were performed using a newly developed and validated HPLC method established in this study. This method was developed to detect nine polyphenols simultaneously. The two HPLC methods developed and validated in this study for the analysis of sugars and polyphenols demonstrated good accuracy, precision, reproducibility, linearity, robustness and sensitivity. Both analytical methods were validated according to the International Convention on Harmonization (ICH). The HPLC parameters for sugar analysis were: refractive index (RI) as the detection mode, the stationary phase was a ligand-exchange sugar column (Shodex SP0810) and an aqueous mobile phase in isocratic mode was used. The HPLC method for polyphenols employed UV diode array detection (DAD) as the detection mode, a reverse phase column as the stationary phase and a mobile phase of consisting of 0.01 M phosphoric acid in water and 100% methanol using gradient elution mode. The highest concentrations of sugars released in the novel 20 L bioreactor with 20% apple pomace (w/v) substrate loading were as follow: glucose (6.5 mg/ml), followed by galactose (2.1 mg/ml), arabinose (1.4 mg/ml), cellobiose (0.7 mg/ml) and fructose (0.5 mg/ml). The amounts of polyphenols released at 20% (w/v) apple pomace substrate were epicatechin (0.01 mg/ml), catechin (0.002 mg/ml), rutin (0.03 mg/ml), chlorogenic acid (0.002 mg/ml) and gallic acid 0.01 (mg/ml). Two mathematical models were developed in this study for kinetic analysis of lignocellulose (apple pomace) hydrolysis in the novel 20 L bioreactor, using the experimental data generated by the above HPLC analyses. The first model, modelling with regression, defines the hydrolysis of the sugars glucose, galactose, cellobiose and arabinose produced in the novel 20 L bioreactor at 5%, 10%, 15% and 20% (w/v) substrate concentrations. The regression model describes the sugars produced in the 20 L bioreactor by minimizing the error of the sugars released by finding a value for K which minimises the function which computes the sum of squares of errors between the solution curves and the data points. The second, more complex, model developed in this study used a system of differential equations model (ODE). This model solved the system by using a numerical method, such as the Runge-Kutta method, then fitted the solution curves to the data. Both models simulated (and had the ability to predict) the production of sugars in the novel 20 L bioreactor for apple pomace hydrolysis. These two models also revealed the time at which the maximum amount of sugars were released, which revealed the optimum time to run the 20 L bioreactor in order to be more cost effective. The optimum time for maximum glucose (the main sugar used in fermentation for biofuel production) release was determined to be around 60 h. The ODE model, in addition, determined the rate at which the substrate became depleted, as well as the rate at which the enzymes became deactivated for the various substrate loadings in the 20 L bioreactor. A third model was developed to determine the optimal running cost of the bioreactor which incorporated the substrate loading and the amount of glucose (g/L) produced. The novel 20 L bioreactor constructed from cost effective materials demonstrated that agro-industrial waste can be converted to value-added products by lignocellolytic enzymes. The sugars released from apple pomace can be used in biofuel production and the polyphenols as food supplements and nutraceuticals for health benefits. This novel study contributes to agro-industrial waste beneficiation via fuel production. In addition, using agro-industrial waste for the generation of value added products (instead of mere disposal) will help prevent environmental pollution.

Measuring the performance of the banking sector in Zimbabwe

- Description: The measurement of the banking sector performance in Zimbabwe is motivated by the unique developments that typified the sector during the period 2009-2014 after emerging from an economic crisis. The Zimbabwean economy returned to stability and growth in 2009, after a decade long economic decline. Economic stability brought about growth in deposits, loans, assets, capitalization and profits during this period. The banking sector has been accused of excessive profiteering through overpricing their products, which culminated in the intervention by the authorities in the sector. The interest rates spread, fees and other charges were presumed to be high which motivated the need to understand whether the banking sector is efficient or inefficient given the high interest rate spreads between the deposit rates and lending rates. Furthermore the high interest rates have raised the question of whether banks were exploiting their market power to price their products highly or whether their prices were determined by the dictates of market forces. Continued profitability of the sector also called for an investigation into what was driving the persistence of profitability over time. The primary objective of this research was to measure the performance of the banking sector during the period 2009-2014. The study contributes to the empirical literature by measuring and assessing the drivers of banking sector competition, efficiency and profitability and applying them at much disaggregated levels. This study also contributes to the debate on the relationships among the performance measures of competition, profitability and efficiency. The study adopted a number of methods which contributes to the array of tools central banks can employ to measure bank performance. The study employed a number of methodologies to measure the competition, efficiency and profitability performance of the banking sector. Competition was estimated using the new empirical industrial organisation methods of Panza and Rose (1987) and the Lerner (1934) Index was used. Cost and revenue efficiency was estimated using the two step methods of Data Envelopment Analysis followed by the Tobit regression method. An assessment of the persistence and drivers of profitability was measured using the Generalised Method of Moments. This study shows that the banking sector was operating under monopolistic competition market structure. This implies that banks held some market power as a result of product differentiation due to unique features such as brands, image and advertising, among others. The study indicates that competition increased during the period 2009-2014. Market power/competition in the banking sector during the study period was driven by capital adequacy, non-performing loans, liquidity risk, cost-income ratio, economic growth and government policy on pricing of bank products. The study suggests that the banking sector experienced an average inefficiency level of approximately 35 per cent in relationship with the best performing institutions in the sample. As a result of stability experienced in the economy, the average revenue and cost efficiency increased between 2009 and 2014. The study further established that the discord around the implementation of the indigenisation and empowerment law, coupled with the government intervention in the banking sector had a negative impact on the banking sector efficiency. It also found that efficiency is determined by market power, capital adequacy, cost income ratio, economic growth, inflation, market share and profitability. The Granger Causality test between cost efficiency and market power suggests that causality is bidirectional. On the other hand granger causality between revenue efficiency and market power is unidirectional and positive, running from revenue efficiency to market power. The result implies that policy measures should bring a balance between increasing competition and improving the revenue efficiency. The study shows that the banking sector was profitable during the period 2009 to 2014. The profitability was a reflection of a stable macroeconomic environment, typified by low inflation levels, despite the crises during this period. It further reveals that the banking sector‟s profitability persisted over time, reflecting the regulatory structure of the sector. The study established that profitability was determined by market power, non-performing loans, liquidity risk, capital adequacy, bank size and cost efficiency. This implies profitability was driven by bank specific determinants. There are a number of policy implications derived from the study. Regulatory measures such as forced consolidations can lead to excessive market power by the banking institution; hence it should be moderated. Banks should enhance credit risk because NPLs has been dragging profits. Banks should take advantage of the various measures introduced, such as the setting up of the special purpose vehicle and credit reference bureau. The government should avoid tampering with market forces as this reduces competition, efficiency and profitability and put in place measures that grow the economy as it increases the efficiency and profitability of the banking sector.

Anti-diabetic and phytochemical analysis of sutherlandia frutescens extracts

- Description: In Africa, the importance of medicinal plants in folklore medicine and their contribution to primary healthcare is well recognized. Across the continent, local herbal mixtures still provide the only therapeutic option for about 80% of the population. The vast floral diversity and the intrinsic ethnobotanical knowledge has been the backbone of localized traditional herbal medical practices. In Africa, an estimated 5400 of the 60000 described plant taxa possess over 16300 therapeutic uses. Similarly, with a therapeutic flora comprising of approximately 650 species, herbal medical practitioners in South Africa, make use of a plethora of plants to treat different human diseases and infections. Over the years, studies have identified numerous plant species with potential against chronic metabolic diseases including type 2 diabetes mellitus (T2DM). Globally, the incidence and prevalence of T2DM have reached epidemic proportions affecting people of all ages, nationalities and ethnicity. Considered the fourth leading cause of deaths by disease, T2DM is a global health crisis with an estimated diagnosis and mortality frequency of 1 every 5 seconds and 1 every 7 seconds respectively. Though the exact pathophysiology of T2DM is not entirely understood, initial peripheral insulin resistance in adipose tissue, liver, and skeletal muscle with subsequent pancreatic β-cell dysfunction resulting from an attempt to compensate for insulin resistance is a common feature of the disease. The current approach to treating T2DM is the use of oral antidiabetic agents (OAAs), insulin, and incretin-based drugs in an attempt to achieve glycaemic control and maintain glucose homeostasis. However, conventional anti-T2DM drugs have been shown to have limited efficacies and serious adverse effects. Hence, the need for newer, more efficacious and safer anti-T2DM agents. Sutherlandia frutescens subsp. microphylla is a flowering shrub of the pea family (Fabaceae/Leguminaceae) found mainly in the Western Cape and Karoo regions of Southern Africa. Concoctions of various parts of the plant are used in the management of different ailments including T2DM. However, despite extensive biological and pharmacological studies, few analyses exist of the chemical constituents of S. frutescens and no Triple Time of Flight Liquid Chromatography with Mass Spectrometry (Triple TOF LC/MS/MS) analysis has been performed. The initial aim of this study was to investigate the phytochemical profile of hot aqueous, cold aqueous, 80% ethanolic, 100% ethanolic, 80% methanolic and 100% methanolic extracts of a single source S. frutescens plant material using colorimetric and spectrophotometric analysis. The hot aqueous extractant was found to be the best extractant for S. frutescens, yielding 1.99 g of crude extract from 16 g fresh powdered plant material. This data suggests that application of heat and water as the extractant (hot aqueous) could play a vital role in extraction of bioactive compounds from S. frutescens and also justifies the traditional use of a tea infusion of S. frutescens. Colorimetric analysis revealed the presence of flavonoids, flavonols, tannins, and phenols in all extracts with varying intensity. The organic extracts 100% methanol, 80% and 100% ethanol exhibited high color intensity (+++) for flavonoids and flavonols respectively, while all the extracts exhibited a moderate color intensity (++) for tannins and phenols. Spectrophotometric analysis of S. frutescens extracts revealed that all the organic extracts contained a significantly higher concentration (in mg/g of extract) of flavonols and tannins when compared to the aqueous extracts. All extracts contained approximately equal levels of phenols. These data confirm the presence of all four groups of bioactive phytocompounds in the S. frutescens extracts used in this study, and also confirm that different solvent extractants possess the capability to differentially extract specific groups of phytocompounds. in individual extracts. Further comparison of these compounds with online databases of anti-diabetic phytocompounds led to the preliminary identification of 10 possible anti-diabetic compounds; α-Pinene, Limonene, Sabinene, Carvone, Myricetin, Rutin, Stigmasterol, Emodin, Sarpagine and Hypoglycin B in crude and solid phase extraction (SPE) fractions of S. frutesecens. Furthermore, using two hepatic cell lines (Chang and HepG2) as an in-vtro model system, the anti-T2DM properties of crude aqueous and organic extracts of S. frutescents was investigated and compared. Both aqueous and organic extracts of S. frutescens were found to decrease gluconeogenesis, increase glucose uptake and decrease lipid accumulation (Triacylglycerol, Diacylglycerol, and Monoacylglycerol) in Chang and HepG2 hepatic cell cultures made insulin resistant (IR) following exposure to high concentration of insulin and fructose. Using real-time quantitative reverse transcriptase polymerase chain reaction (qRT-PCR), the aqueous and organic extracts of S. frutescens were confirmed to regulate the expression of Vesicle-associated membrane protein 3 (VAMP3), Mitogen-activated protein kinase 8 (MAPK8), and Insulin receptor substrate 1 (IRS1) in insulin resistant hepatic cells. IR-mediated downregulation of VAMP3, MAPK8, and IRS1 mRNA in IR HepG2 hepatic cell cultures was reversed in the presence of aqueous and organic extracts of S. frutescens. The hot aqueous extract displayed the highest activity in all the assays, while all the organic extracts displayed similar potency. In conclusion, this study reports that aqueous and organic extracts of S. frutescens possess numerous anti-diabetic compounds that can be further investigated for the development of new, more efficacious and less toxic anti-diabetic agents. The presence of multiple compounds in a single extract does suggest a synergistic or combinatorial therapeutic effect. These findings support the burgeoning body of in-vivo and in-vitro literature evidence on the anti-diabetic properties of S. frutescens and its use in folklore medicine.

- Description: In Africa, the importance of medicinal plants in folklore medicine and their contribution to primary healthcare is well recognized. Across the continent, local herbal mixtures still provide the only therapeutic option for about 80% of the population. The vast floral diversity and the intrinsic ethnobotanical knowledge has been the backbone of localized traditional herbal medical practices. In Africa, an estimated 5400 of the 60000 described plant taxa possess over 16300 therapeutic uses. Similarly, with a therapeutic flora comprising of approximately 650 species, herbal medical practitioners in South Africa, make use of a plethora of plants to treat different human diseases and infections. Over the years, studies have identified numerous plant species with potential against chronic metabolic diseases including type 2 diabetes mellitus (T2DM). Globally, the incidence and prevalence of T2DM have reached epidemic proportions affecting people of all ages, nationalities and ethnicity. Considered the fourth leading cause of deaths by disease, T2DM is a global health crisis with an estimated diagnosis and mortality frequency of 1 every 5 seconds and 1 every 7 seconds respectively. Though the exact pathophysiology of T2DM is not entirely understood, initial peripheral insulin resistance in adipose tissue, liver, and skeletal muscle with subsequent pancreatic β-cell dysfunction resulting from an attempt to compensate for insulin resistance is a common feature of the disease. The current approach to treating T2DM is the use of oral antidiabetic agents (OAAs), insulin, and incretin-based drugs in an attempt to achieve glycaemic control and maintain glucose homeostasis. However, conventional anti-T2DM drugs have been shown to have limited efficacies and serious adverse effects. Hence, the need for newer, more efficacious and safer anti-T2DM agents. Sutherlandia frutescens subsp. microphylla is a flowering shrub of the pea family (Fabaceae/Leguminaceae) found mainly in the Western Cape and Karoo regions of Southern Africa. Concoctions of various parts of the plant are used in the management of different ailments including T2DM. However, despite extensive biological and pharmacological studies, few analyses exist of the chemical constituents of S. frutescens and no Triple Time of Flight Liquid Chromatography with Mass Spectrometry (Triple TOF LC/MS/MS) analysis has been performed. The initial aim of this study was to investigate the phytochemical profile of hot aqueous, cold aqueous, 80% ethanolic, 100% ethanolic, 80% methanolic and 100% methanolic extracts of a single source S. frutescens plant material using colorimetric and spectrophotometric analysis. The hot aqueous extractant was found to be the best extractant for S. frutescens, yielding 1.99 g of crude extract from 16 g fresh powdered plant material. This data suggests that application of heat and water as the extractant (hot aqueous) could play a vital role in extraction of bioactive compounds from S. frutescens and also justifies the traditional use of a tea infusion of S. frutescens. Colorimetric analysis revealed the presence of flavonoids, flavonols, tannins, and phenols in all extracts with varying intensity. The organic extracts 100% methanol, 80% and 100% ethanol exhibited high color intensity (+++) for flavonoids and flavonols respectively, while all the extracts exhibited a moderate color intensity (++) for tannins and phenols. Spectrophotometric analysis of S. frutescens extracts revealed that all the organic extracts contained a significantly higher concentration (in mg/g of extract) of flavonols and tannins when compared to the aqueous extracts. All extracts contained approximately equal levels of phenols. These data confirm the presence of all four groups of bioactive phytocompounds in the S. frutescens extracts used in this study, and also confirm that different solvent extractants possess the capability to differentially extract specific groups of phytocompounds. in individual extracts. Further comparison of these compounds with online databases of anti-diabetic phytocompounds led to the preliminary identification of 10 possible anti-diabetic compounds; α-Pinene, Limonene, Sabinene, Carvone, Myricetin, Rutin, Stigmasterol, Emodin, Sarpagine and Hypoglycin B in crude and solid phase extraction (SPE) fractions of S. frutesecens. Furthermore, using two hepatic cell lines (Chang and HepG2) as an in-vtro model system, the anti-T2DM properties of crude aqueous and organic extracts of S. frutescents was investigated and compared. Both aqueous and organic extracts of S. frutescens were found to decrease gluconeogenesis, increase glucose uptake and decrease lipid accumulation (Triacylglycerol, Diacylglycerol, and Monoacylglycerol) in Chang and HepG2 hepatic cell cultures made insulin resistant (IR) following exposure to high concentration of insulin and fructose. Using real-time quantitative reverse transcriptase polymerase chain reaction (qRT-PCR), the aqueous and organic extracts of S. frutescens were confirmed to regulate the expression of Vesicle-associated membrane protein 3 (VAMP3), Mitogen-activated protein kinase 8 (MAPK8), and Insulin receptor substrate 1 (IRS1) in insulin resistant hepatic cells. IR-mediated downregulation of VAMP3, MAPK8, and IRS1 mRNA in IR HepG2 hepatic cell cultures was reversed in the presence of aqueous and organic extracts of S. frutescens. The hot aqueous extract displayed the highest activity in all the assays, while all the organic extracts displayed similar potency. In conclusion, this study reports that aqueous and organic extracts of S. frutescens possess numerous anti-diabetic compounds that can be further investigated for the development of new, more efficacious and less toxic anti-diabetic agents. The presence of multiple compounds in a single extract does suggest a synergistic or combinatorial therapeutic effect. These findings support the burgeoning body of in-vivo and in-vitro literature evidence on the anti-diabetic properties of S. frutescens and its use in folklore medicine.

A morphogenic and laminated system explanation of position-practice systems and professional development training in mainstreaming education for sustainable development in African universities

- Description: This research focuses on Education for Sustainable Development (ESD) in Higher Education Institutions (HEIs), particularly in Africa. It explores the roles and practices of these institutions, especially their professionals, in the Anthropocene era where increasing concern for contemporary environmental and sustainability issues and risks emerge. The study presents a longitudinal case study of institutions and participants of the Swedish/African/Asian International Training Programme (ITP) on ESD in Higher Education (HE), who are mostly university educators. This thesis however focuses on African ITP participants only. At a macro level, the research sought to examine how African university educators have contributed to the United Nations Decade of Education for Sustainable Development (UNDESD) through their participation in the ITP (which is a change oriented professional development training programme on ESD) and the associated ESD ‘change projects’. The change projects are ITP participants’ direct attempts to mainstream environment and sustainability issues, concerns, and concepts into core university functions and practices: teaching, research, community engagement, and management operations and policy engagement. At a meso level the study sought insight into how educators in national institutions were supported by sub-regional and regional initiatives, institutions and organisations, including the Mainstreaming of Environment and Sustainability in African (MESA) Universities Partnership programme, especially an initiative supported by the Southern African Development Community Regional Environmental Education Programme to provide (limited) seed funding to three southern African universities to establish what are known as ‘MESA Chairs’, with dedicated time and support for MESA activities in their universities . At a micro level, this research sought to investigate how the position-practice systems and the ITP shape (enable or constrain) effective ESD mainstreaming in higher education, and how the morphogenetic approach and laminated system can be used to understand and explain these dynamics and their relations with meso and macro level engagements. The research sought to understand these dynamics through empirical investigations using survey questionnaires, interviews, document analysis and field visits. The research is constituted as theoretical, conceptual, methodological and analytical exploration using a singular and nested case study research approach, underlaboured by a critical realist ontology, and drawing on a social learning epistemology and social realist morphogenetic interpretive lens. In particular, ontological depth was sought via critical realist laminated system explanation. See Chapter Two for details. This study was carried out in three phases. Phase one encapsulates the investigation of all ITP ESD in higher education alumni who were Asian and African participants from the inception of the ITP to its completion, over a six-year period (2008-2013). This included 280 academics from Asia and Africa in 35 countries in Asia and Africa from 106 institutions in Asia and Africa with their 139 change projects. The outcome of phase one of the research is only included in this thesis as an appendix (see Appendix 3; Agbedahin & Lotz-Sisitka, 2015). However, this phase provided and formed the foundational data that was expanded in phases two and three for the purpose of this study. Phase two of this research concentrated on a less broad population of research participants comprising only all African ITP alumni, from all regions in Africa. The overall data collection and analysis included 162 academics in 23 African countries from 66 institutions with their 81 change projects. The aim was to investigate and provide a morphogenetic explanation of their change projects and how the relationship between participants’ positions and practices (and that of others) may influence ESD mainstreaming in universities. The outcome of this phase two investigation is presented in Chapter Four. In phase three, (nested) case studies of Swaziland, Zambia, and Botswana (in the southern Africa region), which included all the ESD ITP HE participants therein and the three corresponding EE/ESD MESA Chairs, were developed. The population sample in this phase three therefore contained 20 academics, from six institutions with their nine change projects. This phase was characterised by field trips to these countries and in-depth data collection and analysis in order to investigate and deepen the morphogenetic explanations of their change projects and how the relationship between participants’ positions and practices (and that of others) have indeed influenced the ESD mainstreaming in universities. The outcome of this phase three research is presented in Chapters Five, Six and Seven. The final Chapter Eight of this thesis focuses on the seven scalar laminated system perspective and reflections on this research and discussion of these perspectives for supporting the mainstreaming of ESD in African higher education institutions and more specifically in the three case countries and respective institutions presented in Chapters Five, Six, and Seven. The seven scalar laminated system is presented in relation to the position-practice system, and draws on morphogenetic social realist and social learning theory to provide perspective on the actual change processes. Chapter Eight also includes a discussion on social learning and its implication for ESD mainstreaming, and provides recommendations for further research. The outcome of the theoretical exploration underpinning this study provided a potential model for understanding ESD learning and change processes that are facilitated by professional development training programmes in the context of ESD in HE. This study also provides a model for appraising educational changes in time and in space, especially in relation to ESD, or the types of changes that can be brought about by professional development interventions such as those provided by the ITP and how they can be tracked, monitored and documented. For the field of professional or academic development in higher education, this research highlights the significance of the relationship between position-practice systems, professional development interventions and institutional transformation. For the field of ESD in higher education, this study shows the need for in-depth consideration of the position-practice system and sphere of influence of change agents and related stakeholders in and around their institutions in the design and development of professional development programmes. It further sheds light on the laminated system of factors that contextually constrain and/or enable effective ESD mainstreaming at individual, collective, institutional, national, regional and global levels.

Scalable chemistry involving diazonium salts

- Description: Herein an alternative approach aimed at reducing the cost of numbering up technique as a scale up strategy for chemical processes from the laboratory bench top to the industry is explored. The effect of increasing channel size on the reaction conversion of the synthesis of azo compounds is investigated. This was achieved via a systematic investigative understanding of the synthesis in microreactors where a proof of concept study was performed to determine the optimum reaction parameters in azo coupling reactions involving couplers with aminated or hydroxylated groups in Little Things Factory-MS microreactors (Channel diameter: 1.0 mm) It was found that at slightly alkaline conditions (pH 8.55) and at a temperature of 25 °C, excellent conversions were attained in the azo coupling reaction of the diazonium salt solution of 2,4-dimethylaniline to 2-naphthol. On the other hand, the azo coupling reaction of the diazonium salt solution of p-nitroaniline to diphenylamine was found to thrive at a pH of 5.71 and at a temperature of 25 °C. Using, these optimized reaction parameters, the in-situ and reactive quench of diazonium salts in LTF-MS microreactors was investigated where it was found that at a flow rate of 0.2 ml/min, 0.03 ml/min and 0.07 ml/min of diazotizable amine & HCl, sodium nitrite and coupler solutions respectively, a conversion of 98% is achieved in approximately 2.4 minutes. A library of azo compounds was thus generated under these reaction conditions from couplers with aminated or hydroxylated aromatic aromatic systems. The scaled up synthesis of these compounds in a homemade PTFE tubing (ID 1.5 mm) reactor system was thereafter investigated and comparable conversions were observed. Capitalizing on the benefits of a large surface area and the short molecular diffusion distances observed in microreactors, in-situ phase transfer catalyzed azo coupling reaction of diphenylamine to p-nitroaniline was also explored. In this investigation a rapid and easy optimization protocol that yielded a 99%, 22% and 33% conversion of diphenylamine, carbazole and triphenylamine respectively in approximately 2.4 minutes using Chemtrix microreactors was established. On increasing the microreactor channel internal diameter in the scaled up synthesis approach, it was found that a 0.5 mm increase in channel internal diameter does result in lower reaction conversions.

- Description: Spices are of great importance in the indigenous culinary and traditional medicine systems of the people of Nkonkobe Municipality of the Eastern Cape, South Africa. The present investigation evaluated the ethnopharmacological potentials of two indigenous South African spices- Foeniculum vulgare and Lippia javanica. The pharmacological investigations on these two plant species include ultra-morphology, nutrient and mineral analysis, evaluation of the essential oil, phytochemical and antioxidant assays, antimicrobial, anti-inflammatory as well as anti-urolithiatic assay of the acetone and aqueous extracts of the two plants. The species were selected for study as the most cited plants after an ethnobotanical survey conducted on the indigenous knowledge of plants used as spices and medicine in Nkonkobe Municipality, Eastern Cape of South Africa. Among the plants cited Foeniculum vulgare(Apiaceae) and Lippia javanica (Verbenaceae) stood out as the most commonly used spices. Others were members of the families; Solanaceae, Apiaceae, Amaryllidaceae, Amaranthaceae and Lamiaceae. Ultra-morphological studies conducted on the leaves of the two selected plants using scanning electron microscope revealed the presence of non-glandular and glandular trichomes, stomata and crystals. The leaf surfaces of these herbs may serve as secretory sites where aromatic secondary metabolites are produced. Analyses of the proximate, mineral, vitamin and anti-nutrients contents of these two spices showed that both species are good sources of these phytochemicals and may be used to enrich the human diet. Lippia javanica possesses lower lipid (0.50 percent), fibre (5 percent) and carbohydrate (64.96 percent) contents than Foeniculum vulgare. On the other hand, protein (20.54 percent), ash (11.60 percent) and moisture content (11.69 percent) were higher in Foeniculum vulgare than in Lippia javanica. Foeniculum vulgare showed higher N (3286 mg/100g), Mg (386.7 mg/100g), K (3187 mg/100g) and Na (1383 mg/100g) content while Lippia javanica was higher in Ca (1833 mg/100g), Zn (4.7 mg/100g), Cu (0.9 mg/100g) and Fe (78.4 mg/100g). Vitamins Aand E were also higher in Lippia javanica (1.31 mg/100g; 2.52 mg/100g) while Foeniculum vulgare (0.45 mg/100g) had higher vitamin C content. No significant differences were observed in the phytate, oxalate and tannin contents of the two spices, but saponin and cyanide were significantly lower in Lippia javanica (268.5 mg/100g; 8.45 mg/100g) than in Foeniculum vulgare (1855 mg/100g; 10.5 mg/100g) Evaluation of the essential oil component, cytotoxicity and antimicrobial activities of both fresh and dried leaves of Lippia javanica and Foeniculum vulgare revealed that the dried samples yielded more oil and also contain more chemical than the fresh samples of both plants. The overall antimicrobial activity evaluated using susceptibility and microdilution assays revealed that the oils of F. vulgare and L. javanica exhibited high antifungal and antibacterial activity, compared to the reference drugs. In addition, essential oil from fresh leaves of both spices was less toxic compared to the oil from the dried leaves. Polyphenolic evaluation of the aqueous and acetone extracts of the plants revealed that the acetone extract had higher phenol, flavanol, flavonoid and proanthocyanidin contents than the aqueous extracts. The total phenolic content of acetone and aqueous extracts were 4.49 ± 0.411 mg/g and 3.73 ± 0.498 mg/g tannic acid equivalent (TAE) respectively for Lippia javanica. The same trend was also observed in Foeniculum vulgare with total phenolic content of acetone and aqueous extracts were 4.22 ± 0.325 mg/g and 4.17 ± 0.651 mg/g tannic acid equivalent (TAE) respectively. - Description: Spices are of great importance in the indigenous culinary and traditional medicine systems of the people of Nkonkobe Municipality of the Eastern Cape, South Africa. The present investigation evaluated the ethnopharmacological potentials of two indigenous South African spices- Foeniculum vulgare and Lippia javanica. The pharmacological investigations on these two plant species include ultra-morphology, nutrient and mineral analysis, evaluation of the essential oil, phytochemical and antioxidant assays, antimicrobial, anti-inflammatory as well as anti-urolithiatic assay of the acetone and aqueous extracts of the two plants. The species were selected for study as the most cited plants after an ethnobotanical survey conducted on the indigenous knowledge of plants used as spices and medicine in Nkonkobe Municipality, Eastern Cape of South Africa. Among the plants cited Foeniculum vulgare(Apiaceae) and Lippia javanica (Verbenaceae) stood out as the most commonly used spices. Others were members of the families; Solanaceae, Apiaceae, Amaryllidaceae, Amaranthaceae and Lamiaceae. Ultra-morphological studies conducted on the leaves of the two selected plants using scanning electron microscope revealed the presence of non-glandular and glandular trichomes, stomata and crystals. The leaf surfaces of these herbs may serve as secretory sites where aromatic secondary metabolites are produced. Analyses of the proximate, mineral, vitamin and anti-nutrients contents of these two spices showed that both species are good sources of these phytochemicals and may be used to enrich the human diet. Lippia javanica possesses lower lipid (0.50 percent), fibre (5 percent) and carbohydrate (64.96 percent) contents than Foeniculum vulgare. On the other hand, protein (20.54 percent), ash (11.60 percent) and moisture content (11.69 percent) were higher in Foeniculum vulgare than in Lippia javanica. Foeniculum vulgare showed higher N (3286 mg/100g), Mg (386.7 mg/100g), K (3187 mg/100g) and Na (1383 mg/100g) content while Lippia javanica was higher in Ca (1833 mg/100g), Zn (4.7 mg/100g), Cu (0.9 mg/100g) and Fe (78.4 mg/100g). Vitamins Aand E were also higher in Lippia javanica (1.31 mg/100g; 2.52 mg/100g) while Foeniculum vulgare (0.45 mg/100g) had higher vitamin C content. No significant differences were observed in the phytate, oxalate and tannin contents of the two spices, but saponin and cyanide were significantly lower in Lippia javanica (268.5 mg/100g; 8.45 mg/100g) than in Foeniculum vulgare (1855 mg/100g; 10.5 mg/100g) Evaluation of the essential oil component, cytotoxicity and antimicrobial activities of both fresh and dried leaves of Lippia javanica and Foeniculum vulgare revealed that the dried samples yielded more oil and also contain more chemical than the fresh samples of both plants. The overall antimicrobial activity evaluated using susceptibility and microdilution assays revealed that the oils of F. vulgare and L. javanica exhibited high antifungal and antibacterial activity, compared to the reference drugs. In addition, essential oil from fresh leaves of both spices was less toxic compared to the oil from the dried leaves. Polyphenolic evaluation of the aqueous and acetone extracts of the plants revealed that the acetone extract had higher phenol, flavanol, flavonoid and proanthocyanidin contents than the aqueous extracts. The total phenolic content of acetone and aqueous extracts were 4.49 ± 0.411 mg/g and 3.73 ± 0.498 mg/g tannic acid equivalent (TAE) respectively for Lippia javanica. The same trend was also observed in Foeniculum vulgare with total phenolic content of acetone and aqueous extracts were 4.22 ± 0.325 mg/g and 4.17 ± 0.651 mg/g tannic acid equivalent (TAE) respectively. But empirical research suggests that making the right decision for effective grain commodities trading has been a difficult task for stakeholders due to high volatility of grain commodities prices. Studies have shown that this is more challenging among grain commodities farmers because of their lack of skills and the time to sift through and make sense of the datasets on the plethora of factors that influence the grain commodities market. This thesis focused on providing an answer for the main research problem that grain farmers in South Africa do not take full advantage of all the available strategies for trading their grain commodities because of the complexities associated with monitoring the large datasets that influence the grain commodities market. The main objective set by this study is to design a framework that can be followed to collect, integrate and analyse datasets that influence trading decisions of grain farmers in South Africa about grain commodities. This study takes advantage of the developments in Big Data and Data Science to achieve the set objective using the Design Science Research (DSR) methodology. The prediction of future prices of grain commodities for the different trading strategies was identified as an important factor for making better decisions when trading grain commodities and the key factors that influence the prices were identified. This was followed by a critical review of the literature to determine how the concepts of Big Data and Data Science can be leveraged for an effective grain commodities trading decision support. This resulted in a proposed framework for grain commodities trading. The proposed framework suggested an investigation of the factors that influence the prices of grain commodities as the basis for acquiring the relevant datasets. The proposed framework suggested the adoption of the Big Data approach in acquiring, preparing and integrating relevant datasets from several sources. Furthermore, it was suggested that algorithmic models for predicting grain commodities prices can be developed on top of the data layer of the proposed framework to provide real-time decision support. The proposed framework suggests the need for a carefully designed visualisation of the result and the collected data that promotes user experience. Lastly, the proposed framework included a technology consideration component to support the Big Data and Data Science approach of the framework. To demonstrate that the proposed framework addressed the main problem of this research, datasets from several sources on trading white maize in South Africa and the factors that influence market were streamed, integrated and analysed. Backpropagation Neural Network algorithm was used for modelling the prices of white maize for spot and futures trading strategies were predicted. There are other modelling techniques such as the Box-Jenkins statistical time series analysis methodology. But, Neural Networks was identified as more suitable for time series data with complex patterns and relationships. A demonstration system was setup to provide effective decision support by using near real-time data to provide a dynamic predictive analytics for the spot and December futures contract prices of white maize in South Africa. Comparative analysis of predictions made using the model from the proposed framework to actual data indicated a significant degree of accuracy. A further evaluation was carried out by asking experienced traders to make predictions for the spot and December futures contract prices of white maize. The result of the exercise indicated that the predictions from the developed model were much closer to the actual prices. This indicated that the proposed framework is technically capable and generally useful. It also shows that the proposed framework can be used to provide decision support about trading grain commodities to stakeholders with lesser skills, experience and resources. The practical contribution of this thesis is that relevant datasets from several sources can be streamed into an integrated data source in real-time, which can be used as input for a real-time learning algorithmic model for predicting grain commodities prices. This will make it possible for a predictive analytics that responds to market volatility thereby providing an effective decision support for grain commodities trading. Another practical contribution of this thesis is a proposed framework that can be followed for developing a Decision Support System for trading in grain commodities. This thesis made theoretical contributions by building on the information processing theory and the decision making theory. The theoretical contribution of this thesis consists of the identification of Big Data approach, tools and techniques for eradicating uncertainty and equivocality in grain commodities trading decision making process.

In this regard, the municipalities are viewed as the most affected institutions, with the Eastern Cape Province identified as culprit number one engaging in corrupt activities. There are a number of contributing factors, as attributed in the study, but most prevalent among these are capacity issues in most of these municipalities, with fewer personnel and less capacity. Challenges of corruption in municipalities are perpetrated mainly by municipal officials, councilors and members of the public. This study, identified corruption as one of the challenges facing municipalities in the Eastern Cape. It involves both the achievements and limitations that have been experienced; and indeed, there are still numerous issues to be addressed and accomplished. It discusses some critical issues such as how corruption has been the mother of the day in South Africa and its effect on the public sector in general. Inadditon, misappropriation of public resources and corruption immersed the nation as a whole into a greater challenge rather than focusing on service delivery issues. There are situations, within municipalities, where the majority of senior officials, who may be in possession of valuable information, are always under suspension. The findings of the study also show that all the municipalities operating in this new democaticatic dispensation have not been able to prepare a transparent annual financial year report at the end of the year. In fact, the majority of them always obtain disclaimer or adverse reports from the Office of Auditor-General (AG), despite numerous attempts and interventions by the AG‟s office and the Provincial Treasury to rectify the situation. The challenges of leadership crises in some of these municipalities, both administratively and politically have been critically examined. The findings show that between 2012 and 2015 financial years, some of these municipalities have been without full-time Accounting Officers; people were only seconded or in acting capacity, and some of them were under adminisistration, which is section 139 (c) of the constitution. There were numerous attempts of interventions by the national leadership of the ANC in the ANC regional structures which in many instances were the root cause of the problems or divisions, as well as in municipalities where in Buffalo City Metro and Nelson Mandela Metro respectively there were leadership changes or redeployments. The issue of corruption in the municipalities is compromising and impact negatively in the service delivery. It can be concluded that municipalities are characterized by a number of instances of corruption perpetrated mainly by municipal officials, councilors and members of society. Bribery, fraud, nepotism, and systematic corruption are some of the forms of corruption that take place in municipalities in South Africa. There are systems in place to fight corruption, but the reality is that they are very weak and ineffective. The anti-corruption strategies put in municipalities have general functioned poorly hence the prevailing the situation. There is glaring non-compliance with the Municipal Finance Management Act (MFMA), the Treasury Regulations and their own Procurement Procedures and Procurement Policies. It is evident that despite the achievement by an ANC led government for almost over 21 years of democracy, there remain many political and administrative challenges especial in municipalities. The fundamental question need to be posed and address therefore is, how best these challenges can be addressed and turn around service delivery to a tangible outcome. The Constitution of the Republic of South Africa provides for the promotion and maintenance of a high standard of professional ethics in the public service. Public resources are vulnerable to abuse by unscrupulous elements, both inside and outside government and therefore requires careful monitoring and control. The existing legislative framework is solid but fragmented and requires review and consolidation to improve its efficiency. To achieve that government requires an approach that is developmental in nature. Governance should be able to take control of the organization’s systems and at the same time adaptable to the environment in a way strengthens the capacity to achieve goals.

Influence of increased processing speed on the microstructure evolution and mechanical property relationship in friction stir welding of AA5182-H111 (T500)

- Description: Friction stir welding of 5xxx series aluminium is generally carried out at low travel speeds, which limits the commercial applications of this process. This manuscript discusses the dynamic eformation characteristics of AA5182-H111 (T500), which allowed a travel speed of 1500 mm/min to be realised. This was achieved by using a spindle speed of 500 rev/min; the same as that required to make a weld of 200 mm/min. The thermal cycles, microstructural evolution due to tool / work-piece interaction, and the local microstructural and mechanical properties of the completed weld are discussed for a travel speed for 200 mm/min and 1500 mm/min. At a rate of 200 mm/min, heat generated due to contact between the shoulder and work-piece surface was transferred ahead of the tool, driving dynamic recrystallization which lowered the flow stress. Hereafter the material entered the stir zone where it was extruded around the pin during a process of continuous dynamic crystallization. At a travel speed of 1500 mm/min, the rate of heat conduction ahead of the tool was not rapid enough, and the material was deformed at low temperatures. A low weld pitch rotations per unit length travelled) was required to ensure that the material was sufficiently strained, to allow the formation of geometric necessary boundaries, which resulted in grain subdivision. The number of high angle grain boundaries was increased through subdivision, thus, increasing the nucleation sites for dynamically recrystallized necklace grains to form. In the absence of heat conduction from the shoulder, the high plastic strain ahead of the tool provided the energy required to drive dynamic recrystallization. Heat was generated from the high plastic strain and during recrystallization, where the stored plastic energy was adiabatically released. Once the material entered the stir zone, the high strain rates associated with the tool rotation which drove dynamic recrystallization that allowed the material to reach high levels of strain, resulted significant grain refinement. The weld zone of the 200 mm/min displayed weld a slight increase in yield strength, with respect to the parent material, due to grain refinement to 10 μm, while the yield strength of the 1500 mm/min weld was significantly increased due to grain refinement to 5 μm. The high weld speed did, however, generate high tensile residual stresses. This work is not only significant for the field of friction stir welding, but also in terms of material processing. The dynamic deformation characteristics associated with Al-5Mg-Mn, alloys which was observed ahead of the tool, is of high value in processes where plastic deformation is applied to improve the mechanical properties of these alloys.

The high weld speed did, however, generate high tensile residual stresses. This work is not only significant for the field of friction stir welding, but also in terms of material processing. The dynamic deformation characteristics associated with Al-5Mg-Mn, alloys which was observed ahead of the tool, is of high value in processes where plastic deformation is applied to improve the mechanical properties of these alloys.

The primary host of Mycobacterium bovis is cattle. It remains a major global health problem. About 2 billion people are thought to be infected with TB and about 1.3 million decease each year from the disease. This study is aimed at determining the prevalence of the disease, characterizing the MTBC isolates and increasing the understanding of the genetic diversity MTBC in the Eastern Cape Province of South Africa, from studying both samples from both humans and cattle. A total of 376 cattle lymph nodes were collected from two commercial abattoirs for investigations. Suspected 6 000 sputum specimens were also collected from humans (cattle farmers). All collected samples were cultured, and isolates were confirmed with a multiplex polymerase chain reaction (PCR) targeting the mpb64 and IS6110 genes. Positive isolates were tested for resistance against anti-TB drugs using the standard Lowenstein Jensen proportion method. Isolates from cattle lymph nodes were further characterized to species level, using the region of deletion 1 (RD1) and oxyR restriction fragment length polymorphism analyses. MIRU-VNTR typing and spoligotyping assays were used to further examine the genetic diversity of all strains. Obtained spoligotype patterns were matched with isolates published in the universal spoligotyping database (SITVIT2). MTBC was detected in 162 (43.09 percent) cattle lymph nodes and 156 sputum specimens. Some isolates 25.9 percent (42/162) from the cattle lymph nodes samples were resistant to rifampicin (RIF) and 98.7 percent (154/156) were resistant to isoniazid (INH) and rifampicin (RIF) (of the human sputum specimens). All cattle lymph nodes isolates exhibited the RD1 gene, and the oxyR analyses classified 96.9 percent of the isolates as Mycobacterium tuberculosis complex and 3.1 percent as Mycobacterium bovis. Twenty seven spoligotype patterns were isolated from cattle lymph nodes, with 10 shared types (SIT) consigned to 5 lineages, including Bov_4-Caprea (1.9 percent), Microti (0.6 percent) and Beijing (17.9 percent) constituting East Asian, Latin-American-Mediterranean (LAM) (3.7 percent), X (4.3 percent), MANU (3.1percent), S (0.6 percent) and 67.9 percent were orphans that were further categorized to their designated lineages (Indo-Oceanic, Euro-American and West-African 2). Ninety one percent of the isolates from humans (farmers) were grouped into 7 clusters by spoligotyping, while the remaining 9 percent was unclustered. SITs were observed in 89.6 percent of the isolates from the Beijing, LAM, X, T and S families. The Beijing family was the most prevalent strain from both human isolates (77.2 percent) and cattle lymph nodes isolates (17.9 percent), and the T (0.6 percent) and Microti (0.6 percent) families were the least detected genotypes in both humans and cattle lymph nodes. In the cattle lymph nodes, spoligotyping showed a higher clustering rate of 82.1 percent, with the lowest (HGDI) = 0.485. The 12 MIRU-VNTR clustering rate was 64.8 percent, showing a higher HGDI of 0.671. However, a combination of both methods showed a decreased clustering rate of 53.7 percent; however these methods resulted in a higher HGDI (0.676). Isolates from human sputum specimens spoligotyping showed a higher clustering rate of 89 percent with an HGDI = 0.822. The 12 MIRU-VNTR showed the lowest clustering rate of 69 percent and an HGDI = 0.940 and the combination of the methods showed a clustering rate of 78 percent and an HGDI = 0.951. The results of this study show the diversity of MTBC strains in the Eastern Cape Province and the low clustering rate indicates continuing transmission in the province. The detection of MTBC strains from slaughtered cattle lymph nodes in abattoirs confirms the risk of providing TB infected meat in markets which has serious implications for the control of TB in South Africa. The continuing spread of MTBC in South Africa threatens both the public and economic health.

- Description: This thesis investigates the chemistry of 25 ascidian species collected from Algoa Bay, South Africa with a concerted focus on metal accumulation by these ascidians and the possible interaction of these metals with ascidian metabolites. Chapter 2 details the screening techniques employed to establish the presence of nitrogenous metabolites (1H- 15N HMBC), hyper-accumulated metal ions (ICP-MS) and potential metal ion/ ascidian metabolite complexes (LC-ICP-MS/ESI-MS). Unfortunately, exhaustive attempts to detect intact metal ion/ascidian metabolite complexes through the use of liquid chromatography with parallel inductively coupled plasma mass spectrometry/electrospray mass spectrometry (LC-ICPMS/ ESI-MS) were unsuccessful. However, the LC-ICP-MS/ESI-MS data obtained for the crude organic extracts of six of the Algoa Bay ascidian species, Distaplia skoogi, Aplidium monile, Aplidium sp., Didemnum sp., Leptoclindines sp. and Polycitor sp. enabled identification of a number of ten halogenated metabolites, namely the indoles 2.28-2.30, and the tyramine and tyrosine derivatives (2.31-2.33, 2.41, 2.43, 2.44 and 2.46), within the ascidian extracts. This study confirmed that LC-ICP-MS/ESI-MS is a powerful tool for the dereplication of halogenated metabolites in complex mixtures especially where these compounds are present in very small amounts. This study is also the first report of these compounds (eight of which are known) in African ascidians. Compounds 2.32 and 2.46 have not been reported before from a marine source. Compounds 2.28-2.30 and 2.33 were present in sufficient amounts in the respective ascidian extracts to allow their isolation and structure elucidation using standard spectroscopic techniques Chapter 3 explores the ability of ascidians to accumulate a wide range of metal ions at concentrations which are often orders of magnitude higher than those of the surrounding sea water. Inductively coupled plasma mass spectrometry (ICP-MS) was used to determine the total ion concentrations of 24 metals in 25 Algoa Bay ascidian species. To the best of our knowledge this is the largest and most extensive investigation of metal concentrations in a group of different ascidians occurring in the same area. Hypotheisizing that the metal ion concentrations for each ascidian specimen screened may represent a unique fingerprint for each specimen principal component analysis (PCA) was used in an attempt to establish whether there were spatial, temporal or phylogenetic relationships associated with the metal concentration fingerprints of the ascidians that formed part of this study. The PCA results showed that there were no statistically significant relationships between ascidian metal ion concentrations and either the collection year or the collection site of the ascidians. However, species from the family Didemnidae provided the clearest statistical evidence supporting a phylogenetic relationship between these ascidians and their hyperaccumulated metal ion profiles. Furthermore, these results suggested that ascidian species are indeed actively concentrating metal ions from the surrounding sea water and are not simply sinks for passively accumulated metal ions. Interestingly, the concentration of vanadium in the set of ascidians studied did not appear to correlate with any of the other metals accumulated by these ascidians suggesting that there is possibly a unique method employed for the accumulation of vanadium by ascidians. Chapter 4 investigated this possibility further after the nucleosides 4.10, 4.11, 4.13, 4.15, 4.17 and 4.40 were isolated from the vanadium accumulating ascidian Aplidium monile. Studies into the interactions between nucleosides and vanadyl are unfortunately rare and usually qualitative in nature with limited information provided about the stability or structures of the complexes formed. The vanadyl accumulating aplousobranch ascidians e.g. Aplidium monile dominated our study of Algoa Bay ascidians therefore providing us with the rationale to investigate the relatively little studied binding ability and stability of vandyl-nucleoside complexes. Potentiometric studies were conducted to determine the stability constants of complexes formed between the oxovanadium ion vanadyl (VO2+) and the commercially available nucleosides 4.10-4.14. The data afforded by this analysis clearly confirmed the complexity of the vanadyl/nucleoside complexation and suggested that guanosine (4.12) formed the most stable complex with oxovanadium ions. We were also able to establish a third protonation constant for the hydroxyl moiety in 4.12 with a logK 8.87 which has not been previously reported. Finally, Chapter 5 revisited the cytoxicity two Algoa Bay ascidians, Clavelina sp. and Atriolum marinense the extracts from which produced promising bioactivity results in previous studies against oesophageal cancer cells. The HP-20 fractionated extracts of Clavelina sp. and Atriolum marinense proved to be similalrly cytotoxic to breast cancer cells. With the exception for the 100% acetone(aq)fractions the NMR data for both species suggested that most active non polar fractions were dominated by what appeared to be structurally unremarkable fatty acid glycerides and as such were not pursued further. Purification of the 100% acetone(aq)fraction of A. marinense resulted in the isolation of a styrene trimer, 5.1, common to both ascidian extracts. The NMR simulation software WIN-DAISY was employed to confirm the structure of 5.1. Attempts to establish if 5.1 was an isolation artefact or a product of marine pollution were inconclusive

Financial characteristics of the nonprofit organisation: theory and evidence for the assessment of the financial condition of South African public universities

Foundational constructs of nonprofit economics are applied in the consideration of financial theories of nonprofit organisations in general, and public universities in particular. From this review, a number of hypotheses are developed. Each of these specifies a positive or negative association between a university's financial condition and a particular dimension of its assets, liabilities, equity, revenues, expenses and surplus. From the nonprofit financial analysis literature, ratios and indicators relevant to these hypotheses are selected. Audited data from the annual financial statements of the universities for the seven year period 2007 to 2013 are substantially transformed in mitigation of failures in accounting, auditing and accountability. The adjusted accounting numbers are used to calculate the financial indicators applicable to each university. Exploratory factor analysis is implemented to categorise and organise this large indicator set on the basis of identified associations with a smaller number of factors. It is found that the financial condition of South African public universities is defined by two broad financial characteristics, capital and revenue. Assessment of the capital dimension is informed by a focus on institutional equity, with particular emphasis on expendable equity and its proportionate relationships with surplus, total capital, and total expenses. The revenue dimension is appropriately evaluated in the context of a comparative and interactive consideration of the three main components of South African public university revenue, as well as the proportionate relationship between non-staff operating expenses and total expenses. The framework displays considerable levels of stability and consistency over the seven year review period, and its constructs are, in addition, robust to the application of multiple alternative confirmatory tests involving financial data that are independent of the factor solutions. The financial condition assessment framework developed in this thesis offers a contribution to a broader discourse in nonprofit finance and accounting, with a focus on public university finances.

The framework displays considerable levels of stability and consistency over the seven year review period, and its constructs are, in addition, robust to the application of multiple alternative confirmatory tests involving financial data that are independent of the factor solutions. The financial condition assessment framework developed in this thesis offers a contribution to a broader discourse in nonprofit finance and accounting, with a focus on public university finances.

Ways of seeing over time: the construction and imagination of ‘historic separation’ in Israeli and Palestinian cultures

Zionism is argued to function as a cultural hegemony that operates in a mercurial, selfsustaining and vibrant manner that has the effect of what this thesis terms ‘centrifugal magnetism’ on discourse in the region. Palestinian national narratives are held to be in tangential relation to Zionism (a classic colonial master-narrative), thus entering into a ‘terrible embrace’ of destructive colonial/postcolonial repetition that tends towards violent conflict and the discrimination of minorities. This thesis then proposes a ‘way out’ of this historiographical pattern that is argued to tangibly inform the cultural fabric of the region. By drawing on the later works of Mahmoud Darwish, Mustaffa Hallaj and Said, it is proposed that there are traces of a notion of self and community that can be described as postnational. This demands a reconstruction of narratives of the past in the region in a pluralistic fashion that is based upon shared exilic identity in flux over what Darwish termed an ‘open historical space’. Crucially, this alternative postnational narrative opens up conceptualisations of the future and is founded upon a renewed disposition to temporality. This thesis thus concludes by proposing that an understanding of temporality as ‘ecstatic’ and essential to being (Martin Heidegger) should be included as a crucial consideration for the end to conflict and the attainment of just and equitable futures.

Large unamplified transmission reach is another contributor in reducing deployment costs of an optical communication system. Spectrally efficient modulation formats are suggested as a solution to overcome the problems associated with limited channels and bandwidth of dense wavelength division multiplexing (DWDM) optical communication systems. Higher order modulation formats which are considered to be spectrally efficient and can increase the transmission capacity by transmitting more information in the amplitude, phase, polarization or a combination of all was studied. Different detection technologies are to be implemented to suit a particular higher order modulation format. In this research multilevel modulation formats, different detection technologies and a digital signal processing aided receiver were studied in a practical optical transmission system. The work in this thesis started with the implementation of the traditional amplitude shift keying (ASK) modulation and a differential phase shift keying (DPSK) modulation systems as they form the basic building block in the design of higher order modulation formats. Results obtained from using virtual photonics instruments (VPI)simulation software, receiver sensitivity for 10Gbpsnon-return-to-zero (NRZ), amplitude phase shift keying (ASK) and DPSK signals were measured to be -22.7 dBm and -22.0 dBm respectively. Performance comparison for the two modulation formats were done over different transmission distances. ASK also known as On-Off keying (OOK) performed better for shorter lengths whereas DPSK performed better for longer lengths of up to90km.Experimental results on a 10 Gbps NRZ- ASK signal gave a receiver sensitivity of -21.1 dBm from digital signal processing (DSP) aided receiver against -19.8 dBm from the commercial bit error ratio tester (BERT) yielding a small difference of 1.3 dB hence validating the reliability and accuracy of the digital signal processing (DSP) assisted receiver. Traditional direct detection scheme and coherent detection scheme performances were evaluated again on a 10 Gbps NRZ ASK signal. Coherent detection that can achieve a large unamplified transmission reach and has a higher passive optical splitting ratio was first evaluated using the VPI simulation software. Simulation results gave a receiver sensitivity of -30.4 dBm forcoherent detection and -18.3 dBm for direct detection, yielding a gain in receiver sensitivity of 12.1 dB. The complex coherently detected signal, from the experimental setup gave a receiver sensitivity of -20.6 dBm with a gain in receiver sensitivity of 3.5 dBm with respect to direct detection. A multilevel pulse amplitude modulation (4-PAM) that doubles the data rate per channel from10 Gbps to 20 Gbps by transmitting more information in the amplitude of the carrier signal was implemented. This was achieved by modulating the optical amplitude with an electrical four level amplitude shift keyed (ASK) signal. A receiver consisting of a single photodiode, three decision circuits and a decoding logic circuit was used to receive and extract the original transmitted data. A DSP aided receiver was used to evaluate the link performance. A receiver sensitivity of -12.8 dBm is attained with a dispersion penalty of about 7.2 dB after transmission through 25 km of G.652 fibre.

Predators of aerial insects and riparian cross-boundary trophic dynamics: web-building spiders, dragonflies and damselflies

Using web-building spiders and odonates (dragonflies and damselflies) as model organisms, I sought to investigate whether the diets of predators of aerial insects could be used to assess the strength of the trophic connectivity between freshwater and terrestrial systems in relation to variables such as stream width, distance from the river and aquatic insect emergence rates and abundances. Predator diet composition was determined by using a combination of diet analysis tools: direct observations of cross-subsidies, naturally-abundant stable (carbon and nitrogen) isotope analysis and fatty acid analysis. I also sought to reveal feeding niches and guilds among riparian aerial predators and investigate how the environment influenced predators’ access to aquatic prey subsidies. As emergent aquatic insect abundances decreased with an increase in distance from the river, and increased with stream width and seasonal changes from winter to summer, stable isotope and fatty acid analyses revealed distinct changes in web-building spider diet composition. Examination of the fatty acid eicosapentaenoic acid, a component commonly used as an indicator of consumer reliance on aquatic nutritional subsidies, showed that aquatic subsidies extended further inland at the wider sections of the river. Spiders and odonates at the wider sections of the Kowie River generally received more subsidies (56 – 70%) than those at the narrower sections (25 – 60%). When terrestrial insect biomass was distinctly low in winter, the benefit of aquatic subsidisation to spiders was relatively lower at the narrower sections of the Kowie River relative to the wide sections. As such, riparian areas adjacent to wide parts of the river were more likely to support larger populations of aerial predators than those at the narrow sections. Apart from the diet changes across time and space, there was evidence of inter-specific niche partitioning in both spiders and odonates, but no differences were observed between males and females of the same species. Results showed odonates of different sizes and hunting strategies had separate dietary niches, hence varied access to aquatic nutritional subsidies. The larger odonate taxa that frequently foraged mid-air had more varied diets and relied less on aquatic emergent insects than the smaller odonates that foraged from perches near the river. There was also evidence of niche partitioning amongst the spiders, as those that built horizontal webs captured more aquatic insects (40 – 78%) than the vertical orb-web builders (20 – 66%). This study showed that the nature and extent of trophic cross-boundary linkages in riparian areas largely depended on the availability of subsidies that varied seasonally and spatially. The width of the stream and seasonal variability emerged as important predictors of emergent insect abundances/biomasses that influenced predator feeding niches. The high mobility of odonates made their reliance on aquatic nutritional subsidies different from the less mobile spiders. The link between the width of the river and the extent of trophic connectivity has implications for riparian area management and definition of riparian buffer zones. However, the variation in diet niches amongst terrestrial consumers makes the results area-specific, and more studies are required that incorporate additional terrestrial predators in other fluvial systems so that we can make some generalizations on the dynamics of riparian trophic cross-boundary links.

The effects of land use on the avifauna and its conservation in a Kenyan coastal forest ecosystem, and the significance of the Arabuko Sokoke Forest to the local community

- Description: This study examines the effects of land use on the bird community of Arabuko Sokoke Forest, the largest area of coastal forest remaining in East Africa and a major Important Bird Area in mainland Kenya. Bird species diversity in three land use types (primary forest, plantation and farm lands) was compared using multivariate analysis to determine the response of different feeding guilds to habitat characteristics. The effect of habitat characteristics on overall bird diversity and specific feeding guilds was tested using linear mixed models. A total of 2600 bird observations were recorded during point counts, representing 97 bird species including 25 fruit-eating birds, 17 nectar feeders; and 60 species belonging exclusively to other feeding guilds. Land use had a significant effect on overall bird diversity and abundance. The distribution of frugivorous birds was primarily influenced by the presence of fruiting trees rather than land use type, while nectarivores were significantly affected by vertical habitat heterogeneity and vegetation type. Although the distribution of insectivorous birds is influenced by many habitat factors, proximity to natural forest, habitat heterogeneity, and the presence of large trees and fruiting trees appear to be most important to this guild. The natural forest has the greatest avian diversity and a distinctive community compared to plantation and farmlands. Patterns of habitat use by birds in the area suggest that vertical vegetation heterogeneity and complexity is especially significant in sustaining diverse and abundant bird populations, if they are in close proximity to native forests. Improvement of conservation management for the plantation and farmlands is thus critical for connectivity with other remnant primary forest patches in the area. Socio-economic data was collected from 109 forest adjacent households to determine the value of the forest to the local community and their perception of conservation issues. Arabuko Sokoke Forest is important in supplementing the livelihood needs of the local community. However, the community lacks information on the forest management plan and many people have little knowledge of local birds, which could limit their capacity to participate in conservation projects. Drivers for local community participation in conservation projects are primarily a sustainable income and the fulfillment of basic household needs. Community conservation education is needed to promote local knowledge of forest biodiversity, as well as clear frameworks for the active involvement of the local community in forest management. Support of community based projects is vital to achieve both the conservation and livelihood objectives of the Arabuko Sokoke Forest management plan.

An exploration of how consistently and precisely mathematics teachers code-switch in multilingual classrooms

This study adopts the notion that code switching is a potential resource that teachers can use when teaching multilingual mathematics classes. The aim of this study is to determine how precisely and consistently selected teachers of multilingual mathematics classes code switched during teaching of trigonometry and geometry at secondary school. This study is informed by socio-cultural theory in general and Vygotsky’s work in particular. It focussed specifically on the critical role that language plays in the teaching and cognitive development of mathematics. My study situated within an interpretivist paradigm, used a case study research design and a mixed method research approach. Data were obtained through document collection, observing and interviewing three Grade 11 Mathematics teachers purposively selected from three secondary schools in Grahamstown and King Williamstown education districts of the Eastern Cape Province, South Africa. Data were quantitatively and qualitatively analysed. Findings from this study revealed that the frequency of code switching was not consistent across teachers, topics and lessons. Teachers taught predominantly in the public domain exposing students to compromised mathematical content through their code switching practices. Borrowing code switching was prevalently employed consistently across the participating teachers. Very little transparent code switching, from mainly those mathematical terms commonly used in the foundation and the intermediate phases, was evident in teacher language. No Grade 11 trigonometry and geometry terms in isiXhosa were transparently and consistently code switched. The data suggested that while precision was observed in some cases, it was not consistent. Inconsistencies were caused by lack of planning for code switching, lack of teaching materials in indigenous languages, selective code switching, and ‘safe mode’ code switching strategies which affected teachers’ pedagogical practices. Overall results in this study illustrate that the lack of planning for code switching and the lack of explicit policies and clear-cut official positions on code switching for teaching has contributed to inconsistent and imprecise code switching by the participating teachers. This study concludes that the development of supporting mechanisms, identifying and documenting best practices to encourage transparent, meaningful and beneficial code switching is urgently required to aid and promote conceptual understanding of strongly bounded sub-registers of secondary school mathematics such as trigonometry and geometry. It is anticipated that this study will contribute significantly to the ongoing debate on language use in education and to the institution of best practices for judicious, consistent and precise use of students’ home language during the teaching of mathematics in South Africa.

My study situated within an interpretivist paradigm, used a case study research design and a mixed method research approach. Data were obtained through document collection, observing and interviewing three Grade 11 Mathematics teachers purposively selected from three secondary schools in Grahamstown and King Williamstown education districts of the Eastern Cape Province, South Africa. Data were quantitatively and qualitatively analysed. Findings from this study revealed that the frequency of code switching was not consistent across teachers, topics and lessons. Teachers taught predominantly in the public domain exposing students to compromised mathematical content through their code switching practices. Borrowing code switching was prevalently employed consistently across the participating teachers. Very little transparent code switching, from mainly those mathematical terms commonly used in the foundation and the intermediate phases, was evident in teacher language. No Grade 11 trigonometry and geometry terms in isiXhosa were transparently and consistently code switched. The data suggested that while precision was observed in some cases, it was not consistent. Inconsistencies were caused by lack of planning for code switching, lack of teaching materials in indigenous languages, selective code switching, and ‘safe mode’ code switching strategies which affected teachers’ pedagogical practices. Overall results in this study illustrate that the lack of planning for code switching and the lack of explicit policies and clear-cut official positions on code switching for teaching has contributed to inconsistent and imprecise code switching by the participating teachers. This study concludes that the development of supporting mechanisms, identifying and documenting best practices to encourage transparent, meaningful and beneficial code switching is urgently required to aid and promote conceptual understanding of strongly bounded sub-registers of secondary school mathematics such as trigonometry and geometry. It is anticipated that this study will contribute significantly to the ongoing debate on language use in education and to the institution of best practices for judicious, consistent and precise use of students’ home language during the teaching of mathematics in South Africa.

Catering for children with special education needs in the provision of early childhood development programme in Zimbabwe : towards a holistic and inclusive framework

A mixed method design located in the pragmatism paradigm was used to carry out the study. Post-positivist is an interactive process which produced in-depth, detailed, rich data from personal perspectives and experiences that resulted in realistic understanding of phenomenon, interpreted through the social and cultural context of respondents’ lives. By adopting quantitative and qualitative approaches, the researcher was able to ensure a high level of reliability in the collected data, at the same time obtaining comprehensive information on how primary schools attaching preschool classes in Chiredzi district cater for ECD “A” and “B” children with special education needs. Purposive sampling and cluster sampling procedures were used to solicit information from ECD teachers, school heads, parents, the District Education Team, NGOs and critical government ministries through semi-structured questionnaires, face-t-face interviews, focus group discussions, observations and document analysis. The study established that, while the government carries out its mandate of providing social services to learners with special education needs, they are incapacitated due to lack of resources. The majority of schools are not providing adequate health facilities for learners with special education needs. Furthermore, most schools are not providing nutritional supplementation to vulnerable learners, situations which deprive the learners with the much needed opportunity to grow and develop to the anticipated potential. In addition, the study noted that, ECD “A” and “B” children with special education needs enrolled in classes attached to primary schools do not access Basic Education Assistance Module (BEAM) or Cash Transfers to alleviate the children that are in need. It came out that such a situation impacts on learners’ attendance and subsequently their learning outcomes. Regardless of the stipulations by international treaties on the nature of inclusive education, the study established that, the education system has not yet modified curriculum to accommodate learners with diversified forms of special education needs. The situation is made worse by the fact that, heads of schools, and teachers are not adequately trained to handle learners with special education needs in early childhood development programmes. In addition, schools face the challenge of scarcity of learning resources that support ECD learners with special education needs which seriously impact on the growth and development of young learners at this critical stage of their development. Regardless of the essence of continuous professional development programs, in schools whose purpose is to redress the professional gap in schools, the study found that the CPD are doing very little to transform the mind-set of teachers. The study concluded that learners with special education needs are not benefiting from the current inclusive education programme. The study recommends that following; Stakeholders work together to promote the health and nutritional needs including other social services of ECD learners in classes attached to primary schools. There be a national teacher in-service programme for those teachers deployed in schools, to equip them with the desired skills, and knowledge to handle learners with special education needs, especially at this critical stage of their development. Teachers’ Colleges redesign their curriculum to address inclusive education in schools. Teachers’ Colleges need to empower new graduates with skills and knowledge to handle children with special education needs in schools. Policy makers to advocate for redesigning the curriculum to include children with special education needs to ensure availability of resources in inclusive schools to alleviate the challenges faced by these children. Those policy makers should advocate for a way to ensure that all children with special education be enrolled in mainstream schools. Thus, the study proposed an alternative, holistic, and inclusive education model for schools attaching ECD “A” and “B”

The relationship between leaders’ emotional intelligence and followers’ motivational behaviour and organisational commitment

- Description: Over the past few years, emotional intelligence (EI) has generated significant interest and a wealth of research as a possible area of insight into what determines outstanding performance in the workplace (Ashworth, 2013:8; Pillay, Viviers and Mayer, 2013:1). The internal environment of organisations in the labour-intense hospitality industry is complex and dynamic. Given the unpredictability of change, the researcher found the hospitality industry to be an intriguing milieu within which to ascertain the importance of EI in predicting leaders’ effectiveness as a measure of outstanding performance. As employees are the internal customers of any hotel organisation, representing many hotel organisations’ only true competitive advantage, the purpose of this quantitative investigation was to examine the relationship between leaders’ EI and followers’ motivational behaviour and organisational commitment. Despite the intuitive plausibility of the assumption that leaders who exhibit EI competencies contribute to outstanding performance, the issue of followers’ motivational behaviour and organisational commitment as leadership indices has received little empirical attention. This study sets out to integrate prior findings on EI, motivation and organisational commitment, to support these findings in literature, and to incorporate these findings into a comprehensive conceptual framework. Using critical realists’ post-positivistic philosophical assumptions, the researcher used the Emotional and Social Competencies Inventory (ESCI) to assess leaders’ EI. Furthermore, the Motivational Sources Inventory (MSI) was used to assess followers’ motivational behaviour, while Organisational Commitment Scales (OCS) were used to assess followers’ organisational commitment. The survey respondents consisted of 120 leaders and 435 followers from 13 hotels in four prominent hotel groups in South Africa. The quantitative data collected from the surveys was analysed quantitatively using SPSS to reach substantial results with inferences. The analysis of variance revealed an overall positive relationship between demographic variables and Leaders EI, followers’ motivational behaviour and followers’ organisational commitment. The correlational analysis revealed positive relationships between leaders’ EI and followers’ motivational behaviour and organisational commitment (R= 0.05-, p<0.01) except for instrumental motivation. The correlation between leaders’ emotional self-awareness and followers’ intrinsic process motivation was somewhat weak while the relationship between leaders’ emotional self-awareness and instrumental motivation was found to be sufficient, but statistically not significant. The researcher can conclude that generally the results of this study reveal that organisational leaders can positively influence the motivational behaviour and organisational commitment of their followers by enhancing their own EI competencies. The results add to the leadership literature by illuminating possible antecedents to leadership effectiveness. It is believed that this research will help the hospitality industry at large in clarifying the importance of EI competencies in leadership as a means of obtaining positive motivation behaviour and commitment from followers. Furthermore, the findings have both managerial and research implications for hospitality operations strategy formulation in order to gain competitive advantage and improve the financial position of the businesses.
