Populations from North and South America, Europe and Asia have been extensively covered in terms of their anthropometric characteristics, physical abilities, technical competency, tactical understanding as well as various personality traits. In describing these details, and especially how each is affected during the ageing and pubertal process, a more informed understanding of the talent development systems is created. A holistic approach to talent development is required to effectively and efficiently produce elite level footballers. An important consideration is the unique socio-economic environment many youth footballers experience which may impact on the talent development process. The present study therefore sought to quantify the anthropometric characteristics and the physical ability level of youth South African footballers from Local Football Association in Makhanda in the Sarah Baartman Region of the Eastern Cape province. In order to achieve this a two-factorial design was used with age and maturity status as independent variables. Anthropometric characteristics were tested by finding the height, weight, body mass index and total fat percentage of participants. Physical ability was determined by results for aerobic capacity, power, acceleration, speed and agility. All tests were football specific and had been widely used in a variety of footballing studies. Additional demographic and socio-economic information was also recorded. A total of 136 participants were placed in their respective Under 11, 13, 15 or Under 19 age groups, while the maturity status of 96 participants were stratified into five distinct categories. Statistical analyses was conducted using p-value significance, with appropriate Tukey post-hoc tests administered when necessary, while effect sizes were calculated using Cohen’s d. Significant changes and practical effect sizes were present for nearly all dependent variables when either stratified by age or by maturity. Total fat percentage was the only dependent variable which was not influenced by either age or maturity. Age and maturity effect sizes for total body fat percentage indicated practical changes present which did not occur during statistical analysis. The present results indicate poor values when compared to normative data or that of other football playing study populations. Further longitudinal research is required to better understand individual growth rates for youth South African footballers, particularly from the Makana region. Implications of the thesis may determine long term development pathways, while the delayed maturity rate is a critical finding which needs to be further researched.

In respect of "Lappies, kan jy my hoor, Lappies?" special attention is paid to similarities and contrasts between this text and the traditional suspense story, notably the way in which conventional techniques are employed to create suspense, as well as to generate an entire subtext which eventually "relocates" the text on the niveau of the murderer's psychological dilemma. In discussing "Voorgevoel" emphasis is not placed primarily on what is conveyed by the narrator, but on the way in which his intentions are subverted both by the window pane through which he is looking and by the narration as such. In this way he is foregrounded and revealed as narrator, just as the text is foregrounded and revealed as literature, with the emphasis, in both cases, not only on their defence mechanisms but also on their impotence. Ultimately, they may be seen as three narratives juxtaposed through irony and relativism. "Gustav gaan speel" is based loosely on Barthes's lexia model, in order to determine the signifying process in the text, and also to demonstrate the way in which the text presupposes rereading. In the discussion of the title text it is revealed how the text is centered in the basic dichotomy between the narrator-as-writer and the journalist, and the way in which this polarity is relativized by the text as such. The text is demonstrated to be the credo of the volume as a whole as well as of the fiction of the Seventies in Afrikaans.

The integration of effluent treatment using constructed wetlands, with crop production and aquaculture

- Description: Breweries are major consumers of water and produce nutrient rich wastewater. Therefore, it is important to research technologies that optimise the reuse and recycling of this waste. This study compared different constructed wetlands (CWs) in terms of their potential for cleaning brewery effluent and simultaneously producing crops Beta vulgaris and fish Oreochromis mossambicus. Filling and draining times (FDT; 15 min, 30 min and 60 min) in tidal CWs were compared in Experiment 1. The 15 min FDT (6.52 ± 0.09 mg/l) and 30 min FDT (5.74 ± 0.09 mg/l) had higher dissolved oxygen (DO) than the 60 min FDT (5.40 ± 0.09 mg/l; p<0.05). This resulted in the 15 and 30 min FDT treatments reaching ammonia effluent discharge standards sooner than the 60 min FDT. Total plant harvest increased with increasing FDT; therefore, 15 min FDT was used as the FDT in tidal treatments in the following experiments. The aerated CW (5.81 ± 0.07 mg/l) and tidal CW (5.67 ± 0.07 mg/l) treatments had higher DO concentrations than the unaerated CW treatment (3.76 ± 0.07 mg/l; p<0.05) in Experiment 2. This resulted in lower ammonia concentrations on day 5 in the aerated and tidal CWs compared with the unaerated treatment (p<0.05). The tidal CW (23.97 ± 2.57 kg) had a total harvest that was approximately four times higher than the unaerated CW (p<0.05), which had the highest frequency of chlorosis and plant mortality; and was unable to treat ammonia to discharge standards. In Experiment 3, the aerated and tidal CW were compared with municipal-water as water sources for aquaculture. There were no differences in fish growth (p > 0.05). However, there were differences in water quality; with the municipal treatment having the lowest pH, EC and nitrate concentration (p<0.05); but all water quality parameters remained in a range suitably for fish production. Due to the tidal CW having the highest plant harvest and lowest frequency of chlorosis and mortality; it was the most suitable CW technology to clean the brewery effluent, and to produce B. vulgaris and water that could be used downstream in aquaculture.

- Description: This thesis considered the income tax implications for South African tax resident individuals who render services abroad. The research included an analysis of the impact that the amendment to the section 10(1)(o)(ii) exemption has on individuals rendering services abroad and companies who send their employees abroad. In doing so, this thesis sought to highlight the key factors for consideration, for both employers and individuals. A doctrinal methodology was applied, and an analysis was carried out of relevant tax legislation, commentary of experts in the field of tax law and the relevant case law of South Africa, the United Kingdom (UK), Australia and the United States of America (US), where relevant. It was established that residency is key to determining the tax liability of a person and has an impact on the relief mechanisms that are available where double taxation arises. In addition, the amendment to section 10(1)(o)(ii) was considered. It was concluded that when rendering services abroad, both the employer and employee need to consider the tax consequences that may arise and highlights the factors which may be relevant. The thesis illustrates that, whilst the R1 million exemption alleviates the double tax consequences to a certain extent, further guidance is needed as to how the R1 million threshold will be calculated.

- Description: Global climate changes have been associated with ocean warming and sea-level rise. Armouring of coastlines has become common practice with the increasing threat of coastal erosion. The transformation of soft sediment habitats to hard, artificial habitats because of coastline armouring can lead to changes in species diversity, composition and distribution. It is, therefore, essential to assess changes to habitats from coastal development as well as the ecological impact erosion control structures have within coastal systems. Ecological engineering attempts to combine engineering principals and ecological processes to reduce environmental impacts from coastal development and the implementation of artificial structures. Estuaries are particularly vulnerable to anthropogenic impacts through development, and are extremely important systems offering nursery and foraging grounds for many species. These systems are, however, particularly vulnerable to anthropogenic impacts from urbanisation and development. Within South Africa many estuaries are being transformed by the addition of artificial structures to combat erosion, one such structure is the Reno mattress (a flattened wire box filled with rocks). This study compared the fish diversity and abundances of existing Reno mattress structures and natural eelgrass (Zostera capensis) habitat in the Keurbooms Estuary, South Africa. Benthic invertebrates were sampled using standard core sampling and an adapted suction sampling approach within the two habitats. The non-destructive method of mini Baited Remote Underwater Video Systems (BRUVs) was used to sample fish. Seasonal benthic invertebrate and fish abundances and assemblages were assessed from winter 2018 to spring 2019 with greater abundances of both recorded in summer. Significantly greater abundances, diversity, and richness of fish were found in the Reno mattress habitat compared to Z. capensis. Invertebrate taxa displayed some overlap between habitats, however, three higher taxonomic groups were only recorded within Reno mattress habitat and one only within the eelgrass habitat. Fish assemblages differed significantly between the two established habitats. A Before-After-Control-Impact (BACI) investigation was used to assess the ecological impact of newly installed Reno mattresses in the Keurbooms Estuary. Zostera capensis extent was sampled by determining the percentage cover of 0.5 m X 0.5 m quadrats and measurements of eelgrass blades. Percentage cover and blade length decreased during the installation of Reno mattress, but then recovered shortly after completion of the installation. Abundances, richness and diversity of invertebrates and fish were found to be similar before and after the installation which suggests that the installation had no net negative impact on the site. The Reno mattresses were found to attract fauna typical of rocky shore environments as well as a few invasive alien invertebrate species. This study noted that a hybrid habitat of Reno mattress and eelgrass was created and may in fact provide the positives of both to a system. In any coastal development it will be important to balance the demands of a growing population and the protection of natural habitats. The results of this study suggest that complex artificial structures such as Reno mattresses do provide habitat for fish and invertebrates. However, the use of these structures should be in combination with natural vegetation (e.g. as a hybrid habitat) and not one that replaces intertidal and subtidal natural habitat especially eelgrass. There is limited information regarding the ecological impacts of using Reno mattresses in estuaries and this study provides new information on their ecological efficacy that should be valuable for future coastal erosion control practices.

Invertebrate taxa displayed some overlap between habitats, however, three higher taxonomic groups were only recorded within Reno mattress habitat and one only within the eelgrass habitat. Fish assemblages differed significantly between the two established habitats. A Before-After-Control-Impact (BACI) investigation was used to assess the ecological impact of newly installed Reno mattresses in the Keurbooms Estuary. Zostera capensis extent was sampled by determining the percentage cover of 0.5 m X 0.5 m quadrats and measurements of eelgrass blades. Percentage cover and blade length decreased during the installation of Reno mattress, but then recovered shortly after completion of the installation. Abundances, richness and diversity of invertebrates and fish were found to be similar before and after the installation which suggests that the installation had no net negative impact on the site. The Reno mattresses were found to attract fauna typical of rocky shore environments as well as a few invasive alien invertebrate species. This study noted that a hybrid habitat of Reno mattress and eelgrass was created and may in fact provide the positives of both to a system. In any coastal development it will be important to balance the demands of a growing population and the protection of natural habitats. The results of this study suggest that complex artificial structures such as Reno mattresses do provide habitat for fish and invertebrates. However, the use of these structures should be in combination with natural vegetation (e.g. as a hybrid habitat) and not one that replaces intertidal and subtidal natural habitat especially eelgrass. There is limited information regarding the ecological impacts of using Reno mattresses in estuaries and this study provides new information on their ecological efficacy that should be valuable for future coastal erosion control practices.

Hop binds to Hsp70 and Hsp90 and facilitates the transfer of client proteins (proteins undergoing folding) from the former to the latter and also regulates nucleotide exchange on Hsp90. Due to its role in correcting protein misfolding during cell stress, Hsp90 is being pursued as a cancer drug target and compounds that inhibit its ATPase activity have entered clinical trials. However, it has been proposed that inhibiting the interaction between Hsp90 and Hop may be alternative approach for inhibiting Hsp90 function for cancer therapy. The malaria parasite Plasmodium falciparum experiences temperature fluctuations during vector-host transitions and febrile episodes and cell stress due to rapid growth and immune responses. Hence, it also depends on chaperones, including PfHsp90, to maintain protein functionality and pathogenesis, demonstrated inter alia by the sensitivity of parasites to Hsp90 inhibitors. In addition, PfHsp90 exists as a complex with the malarial Hop homologue, PfHop, in parasite lysates. Consequently, the purpose of this study was to explore P-PI assay formats that can confirm the interaction of PfHsp90 and PfHop and can be used to identify inhibitors of the interaction, preferably in a medium- to high-throughput screening mode. As a first approach, cell-based bioluminescence and fluorescence resonance energy transfer (BRET and FRET) assays were performed in HeLa cells. To facilitate this, expression plasmid constructs containing coding sequences of P. falciparum and mammalian Hsp90 and Hop and their interacting domains (Hsp90 C-domain and Hop TPR2A domain) fused to the BRET and FRET reporter proteins – yellow fluorescent protein (YFP), cyan fluorescent protein (CFP) and Renilla luciferase (Rluc) - were prepared and used for HeLa cell transient transfections. The FRET assay produced positive interaction signals for the full-length P. falciparum and mammalian Hsp90-Hop interactions. However, C-domain-TPR2A domain interactions were not detected, no interactions could be demonstrated with the BRET assay and western blotting experiments failed to detect expression of all the interaction partners in transiently transfected HeLa cells. Consequently, an alternative in vitro FRET assay format using recombinant proteins was investigated. Expression constructs for the P. falciparum and mammalian C-domains and TPR2A domains fused respectively to YFP and CFP were prepared and the corresponding fusion proteins expressed and purified from E. coli. No interaction was found with the mammalian interaction partners, but interaction of the P. falciparum C-domain and TPR2A domain was consistently detected with a robust Z’ factor value of 0.54. A peptide corresponding to the PfTPR2A domain sequence primarily responsible for Hsp90 binding (based on a human TPR2A peptide described by Horibe et al., 2011) was designed and showed dose-dependent inhibition of the interaction, with 53.7% inhibition at 100 μM. The components of the assay are limited to the purified recombinant proteins, requires minimal liquid steps and may thus be a useful primary screening format for identifying inhibitors of P. falciparum Hsp90-Hop interaction.

Thereafter the classical or traditional comparative method is used to propose seven new species Scarabaeolus soutpansbergensis (Deschodt and Davis 2015), Scarabaeolus megaparvulus (Davis and Deschodt 2015), Scarabaeolus niemandi (Deschodt and Davis 2015), Scarabaeolus carniphilus (Davis and Deschodt 2015), Scarabaeolus ermienae (Deschodt and Davis 2015), Scarabaeolus planipennis (Davis and Deschodt 2015) and Scarabaeolus afronitidus (Davis and Deschodt 2015) and formally synonymise Scarabaeolus vansoni (Ferreira, 1958) with Scarabaeolus lucidulus (Boheman, 1860) and Scarabaeolus xavieri (Ferreira, 1968) with Scarabaeolus andreaei (zur Strassen, 1963). Morphometric measurements of external structures of a group of flightless relict beetles in the tribe Canthonini are used to compile a nexus file which is analysed with computer software. The interpretation of these results is used here to support the erection of a new genus Drogo Deschodt, Davis & Scholtz 2016, Lastly I analysed the DNA sequences of specimens from different species belonging to a species complex in the genus Epirinus Reiche, 1841 occurring over a wide geographic range. These sequences are used together with external morphological characters to propose the synonymy of Epirinus hluhluwensis Medina & Scholtz 2005 and Epirinus ngomae Medina & Scholtz 2005 with Epirinus davisi Scholtz & Howden 1987.

This thesis explores the developmental and commercialisation processes involved in the transfer of a potentially disruptive bioprocessing technology for beverage alcohol production. Ethanolic fermentation strategies are of interest due to their global economic importance and their potential to produce clean renewable fuels in the future. Currently used methods are both energetically wasteful and economically inefficient. To this end more effective bioprocessing methods and implementation strategies are required to enable commercially viable decentralised small-scale ethanol production. Perfusion reactors have a number of advantages over batch and other continuous fermentation strategies. This study aimed to develop and study the fermentative efficiency of a perfusion tower bioreactor system at the bench scale, and subsequently through a scale up process to a low level commercial capacity. An HPLC method was developed for the Simultaneous quantification of common fermentation analytes; this was used to determine bench scale fermentation efficacies over an operational period. At steady state the ethanol volumetric productivity of the bench scale bioreactor system was 3.40 g. L-1.h-1, the average yield of ethanol to consumed sugar was 0.467 g.g -1, with an average sugar conversion percentage of 96%. Results showed that the tower perfusion bioreactor was appropriate for high performance ethyl alcohol fermentations. This reactor design was then scaled up to pilot scale and then commercial scale ca pacity. Similar efficienCies were achieved with these larger systems. Based on the process performance data obtained, a commercialisation strategy was developed and market performance was projected. It was found that productivity rates per unit volume were favourable, and the bioreactor system was determined to be very cost effective for a decentralised ethanolic beverage manufacturing model.

It has been widely acknowledged and observed that an organisation that crafts an effective organisational culture should be well positioned to react to these threatening changes, and to proactively seize opportunities to adapt and learn. This research study seeks to investigate how an organisational culture that supports organisational capacity for change can be developed. It analyses what a banking organisation’s espoused culture is made up of and how it was formulated. A thematic analysis of how the bank’s aspirational and innovative culture contributes to the organisation’s capacity for change is also presented. Organisational management should take a leading role in developing the values and practices that foster participation, which are then imparted to the entire organisation. The research study thus only focused on analysing the development of organisational culture from the management perspective. An interpretivist research approach was adopted, with a case study design as the research method. Data was gathered through conducting personal interviews with twelve managers from a Retail and Business Banking cluster of one of the big South African banks. Numerous interventions were employed in the organisation to develop espoused values, aspirational culture and to encourage all employees to champion innovations and constantly search for better ways to offer delighting value propositions to clients. The research findings can assist managers and organisational leaders in building their organisational capacity for change through developing an effective organisational culture that supports change initiatives. Since the research study only focused on analysing the development of organisational culture from the perspective of management, further research studies may involve employees as well, due to the fact that everyone is expected to take an active role for an organisation to display the overall capability to effectively respond to a progressively volatile environment. The research was able to reveal how key initiatives that are designed from theoretical constructs can be used to deliver on an innovative organisational culture that contributes to organisational capability for change.

It has been widely acknowledged and observed that an organisation that crafts an effective organisational culture should be well positioned to react to these threatening changes, and to proactively seize opportunities to adapt and learn. This research study seeks to investigate how an organisational culture that supports organisational capacity for change can be developed. It analyses what a banking organisation’s espoused culture is made up of and how it was formulated. A thematic analysis of how the bank’s aspirational and innovative culture contributes to the organisation’s capacity for change is also presented. Organisational management should take a leading role in developing the values and practices that foster participation, which are then imparted to the entire organisation. Different antimalarial compounds with different mechanisms of action are ideal as this decreases chances of resistance occurring. Inhibiting DXR and consequently the MEP pathway is a good strategy to find a new antimalarial with a novel mode of action. From literature, all the enzymes of the MEP pathway have also been shown to be indispensable for the synthesis of isoprenoids. They have been validated as drug targets and the X-ray structure of each of the enzymes has been solved. DXR is a protein which catalyses the second step of the MEP pathway. There are currently 255 DXR inhibitors in the Binding Database (accessed November 2017) generally based on the fosmidomycin structural scaffold and thus often showing poor drug likeness properties. This study aims to research new DXR inhibitors using in silico techniques. We analysed the protein sequence and built 3D models in close and open conformations for the different Plasmodium sequences. Then SANCDB compounds were screened to identify new potential DXR inhibitors with new chemical scaffolds. Finally, the identified hits were submitted to molecular dynamics studies, preceded by a parameterization of the manganese atom in the protein active site.

Added to this, interest in the history of the Eastern Cape has been stimulated by the building of the 1820 Settlers' Monument, which was opened in 1974, to mark the hundred and fiftieth anniversary of the arrival of the British Settlers. In order to set the social and political scene for the beginnings of cultural activity in the schools, I have given a brief account of the historical events leading up to the English settlement in the Eastern Cape. Because the evolution of the towns has affected that of the schools, an account of this development has been included¹. To prevent this work from becoming unwieldy, the detailed investigation of the cultural milieu has been limited to that of two frontier towns and two seaports, as being the most representative in the Eastern Cape². So much that was done in the schools had grown out of the ideas brought from Britain and its public schools that it was necessary also to look at the basis upon which the schools in the Eastern Cape were founded, and the lines along which their ethos developed. To facilitate a consideration of the work done in play production and related activities over a period of approximately a hundred and fifty years, I have separated the account into four natural historical divisions: from 1820 until the turn of the century; 1900 to 1918; the period between the two world wars; and that which follows the Second World War. Of these, the first three have been considered from the historical point of view; but with regard to the years between 1940 and 1977 it seemed more interesting and profitable to examine specific trends and developments in dramatic activity within the schools. He also wrote plays intended to ‘rally people to action’ during the apartheid years. The changes in the political and social situation in South Africa since 1994 have had significant implications for those writers and artists who produced protest literature and art. The changes in Mda’s own practice and approach to art are themselves quite telling. His experimental novels place him among those African artists pioneering a new chapter for black South African art and the self-reflexive nature of his novels suggest that he is aware of the fact and is consciously forming and reforming his ideas about what it means to be an artist in post-apartheid South Africa. This study will unpack the role of the artist and the function of art in the becoming new South Africa as represented in Zakes Mda’s novels, thereby hypothesizing Mda’s aesthetic philosophy, as may be deduced from his practice, for what an African artist and art should be. This will be done first by locating Mda in the debates around art and literature within the sociopolitical context of a South Africa in transition. Despite the fact that when it comes to public action in the post-apartheid situation, Mda distinguishes between his own role in society as an artist who is a social activist and the role intended for his work, his own novels reveal a desire for the artefact (or artwork) to have a developmental, educational or conscientizing function. This is evident in representations of the effects of art in what this study proposes to be his extended South African black Kunstlerroman, which spans three novels. It is also demonstrated in his ekphrastic novel, The Madonna of Excelsior, in which visual art is interpreted in the process of description, thereby educating the reader. Not only that, but the reader is made into an ‘almost viewer’ and taught how to ‘see’ art. What emerges in the process of this study is Mda’s aesthetic philosophy or what may be termed his ‘aesthetics of liberation’ concerning the role of the artist in post-apartheid South Africa, a suitable African audience and how art works theoretically, as expressed through his fiction.

Further, reason and imagination are used in Harry Potter as modes of resistance against the prejudice which shapes much of the society of the magical world and so will be examined. In the Harry Potter series, Rowling combines fantasy traditions with realism and in so doing ensures her wizarding world mirrors the world of the reader. Rowling enacts a re-creation of the real world of the reader through a recombination of realistic and fantasy elements. This thesis will call on fantasy theorists Rosemary Jackson and Dimitra Fimi as well as the fantasy and science fiction writer Ursula Le Guin to examine how Rowling conforms to and expands on the fantasy tradition in which she writes in her creation of the magical world. It is made evident through Rowling’s treatment of Harry’s friends Hermione Granger and Luna Lovegood that combining reason and imagination is more beneficial than choosing one over the other. The girls’ ways of thinking, seeing and interacting with those around them are complex and he learns from their combined wisdom how to navigate challenges and trials. Criticism focusing primarily on the secondary character of Luna is relatively scarce, despite her impact on Harry’s views regarding death and the afterlife. This thesis offers a new perspective on the importance to Rowling’s narrative of this open-minded, idiosyncratic figure. Rational and imaginative ways of thinking are necessary modes to use in the resistance to prejudice in wizarding society since this pervasive privileging of wizards over other magical beings espoused by the magical government inspires Lord Voldemort to kill or subjugate those whose magical heritage falls short of pure-blooded wizarding ancestry. In analysing the ostensibly conflicting rational and imaginative modes of thought, I examine Rowling’s unconscious use of shadow theory through her treatment of Harry’s dreams and visions – a direct connection between Harry and Lord Voldemort. Harry confronts his antagonist – and addresses the prejudices pervading wizarding society – through making rational decisions that require imaginative action.

However, according to some reports on Grade 12 English First Additional Language in the Eastern Cape, poetry is a particularly challenging genre to both teachers and learners. This background has given rise to this study, which aimed to investigate the critical and meaningful teaching of poetry in Grade 11, a class preparing learners for Grade 12. This interpretive qualitative case study drew on Vygotsky’s (1978) socio-cultural theory of cognitive development, which states that learners’ best knowledge is produced when co-operative learning takes place, with the help of the teacher. Purposive sampling was used to select two high schools from Lusikisiki district in the Eastern Cape, and one Grade 11 class and one teacher from each school. Lesson observations, document analysis, semi-structured interviews with the teachers, and focus group discussions with the learners were used to collect data. Cultural-Historical Activity Theory, Critical Discourse Analysis, Dutta’s (2001) model of poetry learning, as well as insights from Vygotsky’s socio-cultural theory were used to analyse data. In addition, Four Reader Roles by Freebody and Luke (1990) were employed as a data analysis tool to find out the degree to which the poetry teaching developed learners into code breakers, text users, text participants and text analysts. The data revealed that both teachers and learners had a limited understanding of and negative attitudes towards poetry, and this affects the way teachers teach as well as how learners learn poetry. It also indicated that a teacher-centred approach constrains meaning and critical poetry teaching and learning. The findings suggested that in place of the traditional way of poetry teaching, teachers need to learn, develop and use innovative teaching strategies to strengthen poetry understanding in learners.
