Fucoidans from South African brown seaweeds: establishing the link between their structure and biological properties (anti-diabetic and anti-cancer activities)

- Description: Type 2 diabetes mellitus (T2DM) and cancer are major non-communicable diseases causing a heavy morbidity-mortality and economic burden globally. The therapeutic efforts in managing these diseases are primarily chemotherapeutic and are associated with demerits, including side effects and toxicity, limiting the prescribed amounts. These dosage limits may cause drug resistance, another major challenge in maintaining quality global health. The pursuit of novel natural bioproducts is a reasonable strategy to add to the arsenal against T2DM and cancer. Fucoidans, sulphated fucose polysaccharides abundant in brown seaweeds, have recently become popular for their biological activities, including anti-diabetic and anti-cancer properties. However, endemic South African brown seaweeds have not been adequately explored. Therefore, this study sought to characterise fucoidans extracted from South African brown seaweeds and elucidate their structure to their biological activities. Also, this study highlighted carbohydrate and glucose metabolism as major target processes in the control efforts of T2DM and cancer using fucoidans. Harvested brown seaweeds were identified as Ecklonia radiata and Sargassum elegans. E. maxima was kindly donated by KelpX. The fucoidans were then extracted using hot water, EDTA assisted, and acid extraction protocols. The integrity of the extracted fucoidan was confirmed through structural analysis using FTIR, NMR and TGA. The fucoidan extracts were then chemically characterised to determine their carbohydrate and monosaccharide composition and sulphate content. The characterised fucoidans were profiled for inhibiting the major amylolytic enzymes, namely α-amylase and α-glucosidase. The mode of inhibition by fucoidans and synergy experiments with the commercial anti-diabetic drug acarbose were also investigated. Furthermore, the fucoidans were screened for potential anti-cancer activities on the human colorectal HCT116 cancer cell line. The cytotoxicity of fucoidans was quantified using the resazurin assay. The effect of fucoidan on HCT116 cell adhesion on the tissue culture plastic was also investigated using the crystal violet-based cell adhesion assay. In addition, cancer antimigration properties of fucoidans were also investigated using 2D wound healing and 3D spheroid-based assays. Furthermore, the long-term survival of HCT116 cells was investigated through the clonogenic assay after treatment with fucoidans. Lastly, glucose uptake and lactate export assays revealed the influence of fucoidan on glucose uptake and the glycolytic flux of HCT116 cells. Fucoidans were successfully extracted with a yield between 2.2% and 14.2% on a dry weight basis. EDTA extracts produced the highest yields than the water and the acid extracts. Ecklonia spp. fucoidans displayed the highest total carbohydrate content, with glucose and galactose being the major monosaccharides. S. elegans and commercial Fucus vesiculosus had lower carbohydrate contents but contained more sulphates than the Ecklonia spp. fucoidans. Furthermore, the extracted fucoidan contained little to no contaminants, including proteins, phenolics and uronic acids. In addition, the extracted fucoidans were determined to be >100 kDa through ultracentrifugation. Mass spectrometry also detected the most abundant peak for all fucoidans to be around 700 Da (m/z). Extracted fucoidans inhibited the activity of α-glucosidase more strongly than the commercial anti-diabetic agent acarbose but were inactive on α-amylase. Fucoidans were also shown to be mixed inhibitors of α-glucosidase. Compellingly, fucoidans synergistically inhibited α-glucosidase in combination with the anti-diabetic agent acarbose, highlighting prospects for combination therapy. Finally, fucoidans demonstrated some anti-proliferative characteristics on HCT116 cancer cells by inhibiting their ability to adhere to the tissue culture plate matrix. Furthermore, some fucoidan extracts inhibited the migration of HCT116 cancer cells from 3D spheroids. Some of our fucoidan extracts also inhibited HCT116 colony formation, demonstrating inhibition of long-term cell survival. The E. maxima water extract also inhibited glucose uptake by HCT116 cells, thereby influencing the glycolytic flux. In conclusion, biologically active fucoidans were successfully extracted from South African brown seaweeds. Solms-Laub (Pontederiaceae), in the Lowveld region of Mpumalanga Province of South Africa. Occurrence and abundance of biocontrol agents were recorded at three sites on the Crocodile River and at three dams adjacent to citrus orchards. Leaves of P. crassipes and water samples were collected for insecticide residues and also nutrient status of the water and plants. Eccritotarsus catarinensis Carvalho (Hemiptera: Miridae), Neochetina spp. (combined) (Coleoptera: Curculionidae), and Orthogalumna terebrantis Wallwork (Sarcoptiformes: Galumnidae) were recorded with notable variation in abundance between the river and dams across regions. Insecticide residues were not detected on all leaves sampled across study regions, however, nutrients were detected with nitrate ranging between oligotrophic and mesotrophic. Phosphorus was also detected, but, neither of the nutrients correlated with the occurrence and abundance of naturalized biological control agents of P. crassipes. Bioassays were conducted to measure the effect of commonly used insecticides (viz. Methomyl and Chlorpyrifos) on the survival and feeding damage of biological control agents of P. crassipes. Survival of individual insects was recorded between 0.5 and 120 hours for Megamelus scutellaris and Neochetina eichhorniae Warner (Coleoptera: Curculionidae) adults for treatments where insecticides were topically applied onto the insects or leaves were dipped into the pesticides. Concentrations below field rates, recommended and above field rates of Methomyl and Chlorpyrifos on either exposure techniques significantly reduced survival and feeding of biocontrol agents. Methomyl was more toxic compared to Chlorpyrifos and it significantly reduced the survival of M. scutellaris and N. eichhorniae. Language Learning Anxiety: a bometric investigation of stress and language learning using wearable devices

- Description: As Sapolsky (2015) notes, stress research has long been characterised by definitional debates that gave rise to a lack of agreement between theorists. An example of this definitional confusion can be seen in the area of Language Learning Anxiety, where there appears to be confusion in the literature over terms such as anxiety and stress. This study investigated the link between stress and language learning using wearable devices measuring heart rate variance, as a means of establishing the feasibility of using this new technology in stress research. The results indicate that the contextualised longitudinal data delivered by wearable devices mitigates against the current dominant paradigm in Language Learning Anxiety, which postulates a straight-line negative correlation between stress and learning. Instead, the inverted U stress relationship proposed by theorists such as Hebb (1955) seem to be a better fit for the data. The nature of the contextualised data generated in this study allowed for comparisons between participants’ stress readings in academic contexts, such as language and non-language classes, and their free time. The findings suggest that certain long-held assumptions about heightened stress in academic contexts may not hold true. While the findings of this study did not reach the levels of statistical significance, they constitute proof of concept that the type of contextualised data delivered by wearable devices may allow for a new type of stress research that incorporates contextualising longitudinal perspectives on participants’ stress levels. In this study the inclusion of contextualising data led to fundamentally different conclusions about the relationship between stress and language learning. The same may be true of many areas of stress research. The findings presented in this study have broader paradigm-altering implications not only for educational policy, but also for stress research in general. Perhaps equally important was that the type of data delivered by wearable devices was qualitatively different from that normally associated with quantitative studies. As a citizen living in post-apartheid South Africa, I am captivated by the re-emerging wisdom and knowledge of Nguni cultural philosophies and practices. My practice-based research is interested in a creative interpretation of these ancient philosophies and practices in the context of postcolonial life. The project in many ways re-members through observation of socio-political issues such as inequality, violence, and vandalism as reflections of internal challenges of the individual self in current reality. In essence, my discussion in this paper highlights the need for healing grief and loss for the well-being – physical, emotional, mental, and spiritual – of society. Self-definition is the core of any community and we see this in the philosophy of the African proverb umntu ngumntu ngabantu, ‘I am because you are’. ‘I am’ is a set of beliefs that construct the individual self; our life’s course then becomes a constant molding of the body and the world around us. The wisdom of ubuntu was, and is, a catalyst for many African cultures, as this law recognizes each individual as important and equal in the community. As a descendant of various indigenous clans originating in Africa the well-being and balance of my being is determined by how much I know about who I am. Consequently, being a part of the generation that is experiencing the transitioning phases from apartheid I question the ways we are dealing with the residue of coloniality concerning all aspects of self; the physical, emotional, and mental bodies of self. My creative work is a reflection of the exercises I implemented to cultivate selfhood as a way to participate in and control my reality as a marginalized member of society. The different chapters touch on many intersecting theories about tools for healing/soothing the self. The creative process experimented with the various concepts embedded in the expression ‘I am because you are’. In the process of this research, I learned that knowing yourself is one of the principles of ubuntu, which teaches me that I cannot be or give what I do not have. The themes I explore through the above expression are self-love, self-acceptance, and self-respect using a creative lens. This practice-based research proposes a concept; the act of love as a decolonial gesture. My exhibition Vuleka Mhlaba Ndinegene1 utilizes visual language to convey the need for more conversations on healing using natural elements, as a way to make a connection between self, the natural environment, and community. In the first and second chapters, the paper focuses on practices employed by artists such as Lhola Amira, Guadalupe Maravilla, Buhlebezwe Siwani, Tony Gum, Lina Iris Viktor, and Aida Muleneh, to relate the various approaches to re-member the severed connection between self, nature, and community. This practice-based research proposes a concept; the act of love as a decolonial gesture. My exhibition Vuleka Mhlaba Ndinegene1 utilizes visual language to convey the need for more conversations on healing using natural elements, as a way to make a connection between self, the natural environment, and community. In the first and second chapters, the paper focuses on practices employed by artists such as Lhola Amira, Guadalupe Maravilla, Buhlebezwe Siwani, Tony Gum, Lina Iris Viktor, and Aida Muleneh, to relate the various approaches to re-member the severed connection between self, nature, and community. The synthesized porphyrins are conjugated to cancer selective biomolecules (folic acid and biotin) which are linked to nitrogen doped graphene quantum dots, as potential photosensitizers for photodynamic therapy (PDT). The symmetrical morpholine porphyrin complexes 2 (Sn(IV) 5,10,15,20 tetra-4-morpholinyl porphyrin) and 3 (Zn 5,10,15,20 tetra-4-morpholinyl porphyrin) had the same substituent but different central metals, and they were both conjugated to biotin decorated nitrogen doped graphene quantum dots (B-NGQDs), however complex 2 (Sn(IV) 5,10,15,20 tetra-4-morpholinyl porphyrin) was conjugated to B-NGQDs through an ester bond and complex 3 (Zinc 5,10,15,20 tetra-4-morpholinyl porphyrin) through 𝜋-𝜋 stacking. The effect of asymmetry was studied by comparing complex 3 (Zn 5,10,15,20 tetra-4-morpholinyl porphyrin) and complex 5 (Zn 5- bromophenyl-10-15-20-(tris-4- morpholinyl) porphyrin). Complex 5 (Zn 5- bromophenyl-10-15-20-(tris-4- morpholinyl) porphyrin) was an asymmetric porphyrin with morpholine and bromine as substituents. It was observed that asymmetry enhances singlet oxygen quantum yield and PDT activity. - Description: This thesis reports on the synthesis and characterisation of differently substituted aza-BODIPY dyes for use in photodynamic antimicrobial chemotherapy (PACT) and photodynamic therapy (PDT) activity studies. The aza-BODIPY dyes were synthesised by adding phenyl and naphthyl rings at the 3,5-positions of the aza-BODIPY core, with methylthiolphenyl (9a and 9b, respectively) or thien-2-yl rings (9c and 9d, respectively) at the 1,7-positions. 9a-c were iodinated at the 2-position to form 10a-c, respectively, while 9d was diiodinated at the 2,6-positions to form 10d. The methylthiolphenyl-substituted dyes (10a and 10b) were successfully conjugated to gold nanoparticles (AuNPs) to form nanoparticles conjugates (10a-AuNPs and 10b-AuNPs), while attempts to conjugate the thien-2-yl-substituted dyes were unsuccessful. The photophysicochemical properties of 9a-d, 10a-d and nanoconjugates 10a-AuNPs and 10b-AuNPs were investigated to determine their suitability for use in the applications. Adding heavy atoms such as iodine at the 2,6-positions of the aza-BODIPY led to enhanced singlet oxygen generation since these dyes displayed moderate to high singlet oxygen quantum yields. None of the dyes exhibited any fluorescence. The PACT activity studies for 9c-d, 10a-d, and the 10a-AuNPs and 10b-AuNPs were carried out against Staphylococcus aureus and Escherichia coli with a Thorlabs M660L3 light-emitting diode (LED) with an irradiance of 280 mW/cm2 for all dyes other than 10d. A Thorlabs M730L4 LED with an irradiance of 160 mW/cm2 was used for 10d. Time dependence studies were only carried out against Staphylococcus aureus, so very low log reductions were observed against Escherichia coli in initial concentration studies. The 10a-AuNP and 10b-AuNP nanoconjugates exhibited high antimicrobial activity with a log reduction of 9.41 and 0.00% viable colonies, while the iodinated aza-BODIPY had a log reduction of 8.94. The in vitro photodynamic therapy activities and dark cytotoxicity were investigated against human MCF-7 breast cancer cells for dyes 9c-d and 10c-d with a Thorlabs M660L3 LED (280 mW/cm2) for all dyes. The dark cytotoxicity was minimal in each case with IC50 > 50. Iodinated dyes 10c and 10d had IC50 values of 11.6 and 8.2 μM, respectively, and non-iodinated dyes 9c and 9d had IC50 values of 12.5 and 19.2 μM. A study on visual inspection of citrus sorters in a packhouse in the Eastern Cape of South Africa

- Description: Background and Purpose. The South African citrus industry is a major global producer of citrus as well as the second largest exporter of citrus fruit in the world. This industry provides employment to an estimated 112 000 workers in South Africa and is vital in providing economic opportunities to communities such as those in the Sunday’s River Valley in the Eastern Cape province of South Africa due to the low employment and education levels within this province. Citrus sorting is a visual inspection task that requires workers to sustain attention and remain vigilant for the duration of a working shift. Their role in the packhouse is to ensure that no defective products are packed for export or the local market and that the fruit meets the customers’ expectations. Since the demands of sorting citrus are poorly understood, this study attempted to assess the effect that sorting citrus has on vigilance performance as well as attempt to quantify perceived workload while taking into account different contextual factors that affect perceptions of this working task. Methods. This study was conducted within a citrus packhouse in the Eastern Cape province of South Africa. The sample was comprised of individuals employed by the packhouse as citrus sorters. In addition to a recruitment and information session, testing was conducted at three intervals during a working shift on four consecutive days. Each time, indicators of vigilance, such as correct responses, sensitivity, reaction time, false alarms and misses, were collected during a 5-minute computerised version of the Mackworth Clock-test, while perceived workload was assessed with the NASA-TLX scale, and sleepiness was recording using the WITS Sleepiness Scale. Furthermore, an open-ended question was asked to further understand factors affecting the ability of participants to concentrate during their task of sorting. Results. 18 participants consented to participate in this study. 16 of these individuals identified as female with an average age of 30 years (± 7). 83% of participants were of Black African descent and were isiXhosa speakers, while 17% of participants were of Coloured descent and Afrikaans speakers. This study found that performance, as measured by the percentage of correct responses, sensitivity and reaction time on the 5-minute long Mackworth Clock Test was not significantly affected as time-on-task for sorting citrus increased during a working shift, neither did performance on the vigilance test change as days of the week proceeded. However, ‘misses’ during the vigilance test were found to significantly decrease over the course of a working shift, thus contradicting the expected outcomes of this study. Perceived workload of sorting citrus, as well as perceived sleepiness, significantly increased with time-on-task during the course of a working shift. The major contributions for this increase in perceived workload while sorting citrus were derived from the Mental Demand and Physical Demand sub-scale for the NASA-TLX. However, responses from the open-ended question indicated that participants generally enjoyed their work, although there were factors, such as the nature of fruit and social set-up of work, that negatively affected this perception. Discussion. The results from this study contradicted the researcher’s expectations. It was anticipated that measures of correct responses and sensitivity would decrease and misses, and reaction time would increase with time-on-task for sorting citrus, yet vigilance indicators from the Mackworth Clock Test generally indicated no change in vigilance, and, according to “misses” even improved with time-on-task. A likely explanation for this is that the use of a 5-minute Mackworth Clock-test was unsuitable in this context, or rather, the time of this test was not long enough to induce a ‘vigilance decrement’. Furthermore, in order to administer this vigilance test, participants were taken off the production line and led to a testing room which most likely increased their levels of arousal, thus affecting performance on the vigilance test. However, even though perceived sleepiness was found to significantly increase over the course of a working shift, these data correlated poorly with the vigilance results, thus contradicting the expected outcomes of the study. Mental demand significantly increased, possibly due to the demand of attentional informational processing required for sorting, as well as various environmental factors present within the packhouse. Physical demand, too, significantly increased as workers were required to remain standing throughout their working task as well as to make use of their upper extremities to remove defective fruit. Correlation analyses showed significantly weak correlation for Mackworth Clock Test variables and sleepiness ratings to all other variables. NASA-TLX variables, however, generally showed significantly moderate correlations with one another. Conclusion. It remains unclear how vigilance changes and sustained attention while sorting citrus, since the experiment did not show changes in performance. This is largely attributed to the methodological set-up which could have increased arousal and therefore affected vigilance. It is recommended that future studies research vigilance and sustained attention during the sorting itself, rather than attempting to infer vigilance results from a computerised vigilance test. Perceived workload of sorting, however, increased over the course of a working shift due to significant increases in perceptions of the physical and mental demands of sorting. Future studies should address concerns relating to the physical demand of sorting. However, even though perceived sleepiness was found to significantly increase over the course of a working shift, these data correlated poorly with the vigilance results, thus contradicting the expected outcomes of the study. Mental demand significantly increased, possibly due to the demand of attentional informational processing required for sorting, as well as various environmental factors present within the packhouse. Physical demand, too, significantly increased as workers were required to remain standing throughout their working task as well as to make use of their upper extremities to remove defective fruit. Correlation analyses showed significantly weak correlation for Mackworth Clock Test variables and sleepiness ratings to all other variables. NASA-TLX variables, however, generally showed significantly moderate correlations with one another. Conclusion. It remains unclear how vigilance changes and sustained attention while sorting citrus, since the experiment did not show changes in performance. This is largely attributed to the methodological set-up which could have increased arousal and therefore affected vigilance. It is recommended that future studies research vigilance and sustained attention during the sorting itself, rather than attempting to infer vigilance results from a computerised vigilance test. Perceived workload of sorting, however, increased over the course of a working shift due to significant increases in perceptions of the physical and mental demands of sorting. Future studies should address concerns relating to the physical demand of sorting. - Description: Owing to the poor documentation of the phoscorite-carbonatite association present in the Schiel Complex and the associated economic potential of other known phoscorite-bearing complexes, the Schiel Complex is widely thought to have similar economic potential. This complex is often compared to the lucrative Phalaborwa Complex, as it is thought to have crystallised from a common parental melt, with a similar age of emplacement. This study aims to provide clarity on the physical and chemical characterisation of the various rock types present in the Schiel Complex, with this study being the first petrological investigation based on fresh in-situ samples gathered from 3 borehole cores which were drilled by FOSKOR in the 1960s. The sampled sections of the ultramafics from the Schiel Complex are comprised of end-member rock compositions of either magmatic phoscorites or pyroxenites or metasomatic glimmerites, where gradational contacts between these various end-members produce rock varieties that contain characteristics of one or more end-member types. Carbonatite rocks are present as medium-grained, coarse-grained and banded calcio-carbonatite varieties where the carbonatite rocks are proposed as being the metasomatic medium for glimmerite production. Contrary to previous research, the structure of the ultramafic and carbonatite bodies are present as vein and veinlet structures which seem to originate from a single pipe-like body, from which these rock types intruded into the surrounding syenitic country-rock. Metasomatic alteration of the ultramafic sections of the Schiel Complex also show that the carbonatite rocks must have intruded after some ultramafic magmatism. The presence of the same minerals, with similar chemistries, in both the ultramafic and carbonatite rocks as well as similar REE chondrite-normalised plots show that the various rock types may have originated from a common parental magma, where the accumulation and crystallisation of minerals is the most likely factor in producing the various Schiel Complex rock varieties, causing silicate minerals to be present in the carbonate fraction of the magma, and carbonate minerals in the silicate fraction of the magma. Apatite is the expected rare earth element (REE) mineralising mineral in phoscorites, but is shown to be depleted in REE content in the Schiel Complex due to metasomatic fluid infiltration causing the scavenging and dissipation of REEs. These rocks have also crystallised containing no significant copper-bearing mineralisation, contrary to that which is seen in the Phalaborwa Complex. A comparison of mica minerals between the Schiel Complex rocks and the Phalaborwa Complex rocks show that the two complexes have undergone unique emplacement processes and should not be considered as sister complexes. Efforts to date the glimmerite and carbonatite rocks based on zircon grain U/Pb geochronology proved unsuccessful in constraining the current ages of emplacement provided by previous researchers, but rock relationships show that the current accepted sequence of events cannot be correct, providing scope for further research. - Description: This study considers the experiences of teachers working with Fetal Alcohol Spectrum Disorders (FASD)-affected children in mainstream schooling. FASD refers to a range of conditions in children resulting from maternal alcohol consumption during pregnancy. Prevalence rates in South Africa are said to be among the highest in the world. There is a paucity of research on FASD-affected children in the Eastern Cape. This study was thus conducted in the Buffalo City Metropolitan Municipality (BCMM). Teachers told stories of their experiences of teaching undiagnosed FASD-affected children in mainstream schooling. The stories reveal the quality of life for children in classrooms and socio-contextual influences of their experiences at school, as well as the responsibilisation of teachers in these settings. This qualitative research is situated within a social constructionism paradigm. The social model of disability was utilised as a theoretical framework for this study to represent the systemic barriers against which FASD-affected learner support in the mainstream school can be conceptualised. A sample of five (5) Black female teachers were interviewed. Data gathering was done by conducting telephonic interviews using the single question inducing narrative, known as SQUIN. A thematic analysis of the data was conducted for the purpose of identifying themes within the data collected. Four superordinate themes emerged from the data, viz. narratives of FASD-affected learners, narratives of parental involvement, narratives of the role of teachers and narratives of stakeholder support. This study reveals a significant level of responsibilisation of teachers. Teachers appear to play a bigger role than just being teachers and they positioned themselves as playing a parental role in relation to FASD-affected learners. FASD-affected learners were positioned as “the problem” in the mainstream classroom whereas the environment is a disabling factor to FASD-affected learners. There is a need for a holistic approach in working with FASD-affected learners from all relevant stakeholders. Key recommendations are made for effective inclusion of FASD-affected learners in mainstream schooling. Future research recommendations include repeating this study in a different geographical area, a focus on senior phase learners and/or a more diverse sample. Importantly it also builds on my time working at the International Library of African Music (ILAM,) where the aforementioned instruments, as well as the dende (musical bow) and tshizambi (Vhavenda and Vatsonga mouth bow), are displayed in transparent glass cubicles with a note, “Do not touch, they are fragile”. This phrase is painfully apt because, as a musician, I have observed a decline in the availability and performance of these musical instruments. The truth is that, other than at ILAM, these instruments are hardly in circulation, let alone being performed. This fact ignited my interest in relearning some of the musical instruments I used to play and make while herding cows in Giyani. In the context of trends such as modernisation, rural–urban migration, and globalisation, I document my experiences as a musical-instrument maker, teacher, and performer in revitalising dende, tshipotoliyo, tshitiringo, and tshizambi through classroom practice, using Rhodes music students, instrument-making workshops, performances, and community collaborations as inspiration. - Description: The purpose of this study was to investigate the impact of three critical incidents with a direct effect on Small and Medium Enterprises (SMEs) within the Petroleum retail sector in South Africa. This study assessed three specific areas: (1) the impact of fuel levy increase on Petroleum retailers, (2) explore how fuel stock-outs are experienced by the SMEs, and (3) the effects of fuel distributors' strike on SMEs in the petroleum sector in South Africa. To address the research objectives, the study employed a qualitative research approach, based on a narrative research design. Primary data was collected through semi-structured interviews conducted with six SMEs in the fuel retail business in Johannesburg. An interview guide was used. Findings highlighted that the location of fuel retail SMEs is an important determinant of how the fuel retailers are affected by these three critical incidents of fuel levy increases, stock-outs and strikes. The study found that while the fuel levy increases are felt through reduced sales volumes, the impact is not the same across fuel retailers. The study revealed that the elasticity of demand varies depending on the location for which it is elastics for those retailers in high Living Standards Measure (LSM) locations and inelastic for those located in lower LSM locations. It was found that the impact is much less for those fuel retailers located in affluent suburbs with a high LSM. In locations such as Beyers Naude and Radiokop in Northern Johannesburg with a high LSM, the demand for fuel remains constant irrespective of price increases. The locations are also less likely to be adversely affected by protest actions. Concerning SMEs’ experiences of fuel stock-outs, the study found that stock-outs impact fuel retail business operations through overhead costs that are incurred while the fuel stations are out of stock. The small margins make it impossible for fuel retailers to quickly recover, as their profitability is dependent on sales volumes, which are compromised when there are stock-outs. Fuel retailers also suffer from reduced future sales volumes due to waning customer loyalty. Finally, the study found that the occurrence of wage-related strikes is minimal due to the nature of collective agreements that are signed within the sector. It was found that the impact is much less for those fuel retailers located in affluent suburbs with a high LSM. In locations such as Beyers Naude and Radiokop in Northern Johannesburg with a high LSM, the demand for fuel remains constant irrespective of price increases. The locations are also less likely to be adversely affected by protest actions. Concerning SMEs’ experiences of fuel stock-outs, the study found that stock-outs impact fuel retail business operations through overhead costs that are incurred while the fuel stations are out of stock. The small margins make it impossible for fuel retailers to quickly recover, as their profitability is dependent on sales volumes, which are compromised when there are stock-outs. Fuel retailers also suffer from reduced future sales volumes due to waning customer loyalty. Finally, the study found that the occurrence of wage-related strikes is minimal due to the nature of collective agreements that are signed within the sector. - Description: Despite the vast research on the negative impacts of invasive alien species on the environment, these species remain part of the rural communities due to their numerous livelihood uses. Thus, more research is required, focusing mainly on the impacts of invasive alien species on the livelihoods of rural communities. This study investigated the community perceptions of rosehip (Rosa rubiginosa) and its contribution to rural communities as an invasive alien species. Four community councils, Pitseng, Matlameng, Limamarela and Mphorosane in the Leribe District Lesotho, were assessed. The study followed the pragmatism paradigm. The contribution of rosehip to rural livelihoods was analysed by comparing income from rosehip with other income sources. The study used simple random sampling and snowball sampling to select a representative of 160 respondents. The primary data was collected using semi-structured questionnaires. Moreover, SPSS and Stata statistical package programs were used for statistical analyses. The results showed that rosehip's livelihood benefits, its negative impacts, the length of time it has been available in the area, and its abundance highly influence the social, economic and environmental perception of rural communities. Furthermore, the study revealed that although the income from rosehip is extremely low and available for only three months of the year, the income plays an important part to the poorer households who have no other income sources. The study found that the main reason for engagement in rosehip harvesting despite its challenging nature was unemployment. The study also revealed that rosehip is part of the risk-reducing strategy or income diversification. Some households used it to complement other sources of income, such as agricultural production. Rosehip trade, if well-controlled, has the potential to alleviate rural poverty by creating job opportunities, providing a source of household income, and acting as a safety net in the face of shocks such as limited job opportunities and food shortages. In conclusion, households’ perceptions of rosehip have proven that rosehip is a valuable resource that provides a supplementary income that contributes towards alleviating poverty in Lesotho’s rural communities. The study recommends the private sector to establish and manages small agro-processing industries focusing on products used daily. The effect of orchard sanitation and predatory ants on the eclosion of the internal feeding pests and Oriental fruit fly, in South Africa

Orchard sanitation can play an important role in suppressing the populations of these pests, however there are little data on the efficacy of sanitation techniques. Therefore, the current study investigated the effect of fruit disposal techniques and burying depths on the eclosion of the most important pests of citrus in South Africa, false codling moth Thaumatotibia leucotreta Meyrick (Lepidoptera: Tortricidae), Mediterranean fruit fly or Medfly Ceratitis capitata Wiedemann (Diptera: Tephritidae), Natal fruit fly Ceratitis rosa Karsh (Diptera: Tephritidae), and Oriental fruit fly Bactrocera dorsalis Hendel (Diptera: Tephritidae). Abscised C. sinensis fruits were inoculated with larvae of T. leucotreta, and eggs of C. capitata, C. rosa, and B. dorsalis, before being disposed as pulped, or whole, and buried at different depths (0 cm, 5 cm, 25 cm, and 50 cm). Abundance and richness of predatory ants were monitored using pitfall traps to ascertain their effect on the mortality of the immature stages of these pests. Ceratitis capitata and C. rosa failed to eclose from the inoculated fruits disposed at different depths, however, T. leucotreta and B. dorsalis adults did eclosed. Significantly fewer B. dorsalis eclosed from fruits that were pulped in comparison to eclosion where the fruit were left whole (F (3, 16) = 11.45, P < 0.01). Furthermore, depth of burial had a significant effect on the number of eclosed adults of Drosophila sp (F (3, 112) = 3.43, P < 0.01). Burying fruits at 50 cm suppressed the eclosion of all the internal feeding pests tested. Twenty-seven thousand seventy-three individual ants (Hymenoptera: Formicidae) were sampled from the same plots as used above, with at least 47% and 53% sampled from plots where pulped and whole C. sinensis fruits were disposed of, respectively. The ants were identified to morphospecies which included Pheidole1, Pheidole2, Formicinae1, Formicinae2, Formicinae3, and Myrmicinae1. The disposal of the inoculated C. sinensis fruits either as pulped or whole and burying at different depths significantly suppressed and/or delayed the eclosion of either of the tested internal feeding pests of citrus. Although, predacious ants were sampled from the same treatment plots they did not affect the survival or eclosion of the tested pests, and this could be attributed to the application of the slow toxic ant bait.
