The jackal as a mesopredator : dietary response to an apex predator and role as an endozoochore

- Description: Interactions between apex predators, mesopredators and their impacts on trophic dynamics have been well documented. However, the influence of apex predators, through the provision of carrion, on the foraging ecology of African mesopredators is poorly understood. To investigate such resource supplementation by apex predators for mesopredators, the black-backed jackal Canis mesomelas, a generalist mesopredator, was used as a model species to improve our understanding of this topic in South African ecosystems. To do this, I compared the diet of jackal at replicated sites with and without cheetah in the eastern Karoo, South Africa. Scat samples were macroscopically categorized into six broad dietary categories: mammal, reptile, bird, fruit, forage and insect. Mammalian prey was further identified to species and grouped into six functional dietary categories: carnivore, large ungulate, small ungulate, small mammal, and unknown mammal. Jackal diets were dominated by mammal prey, with significant differences in the biomass and frequency of occurrence of mammals by jackal at sites with cheetah and without cheetah. Mammal consumption, showed limited variation amongst seasons. Resource supplementation provided by predator derived carrion is an important factor influencing jackal foraging ecology. The emergent hypothesis that jackal may show demographic responses (increased fecundity, survival, density) to this supplementation needs to be tested. This study also provided an opportunity to investigate the potential role of jackal as an endozoochore. Jackal consume fruits and their seeds, yet to our knowledge, an investigation into their seed dispersing abilities is yet to be investigated. This study assessed the viability and germination capacity of ingested seeds extracted from jackal scat samples using the tetrazolium tests and germination trials. Seed viability and germination of all three species (Grewia occidentalis, Pappea capensis and Diospyros lycioides) were significantly lower than uningested seeds. These results suggest that jackal gut passage is acting severely on ingested seeds. This study represents a novel investigation into differences in jackal diet in sites with and without cheetah as well as the role jackal play in endozoochory, contributing to improved understanding of the ecological role of jackal.

- Description: The concept of sustainability has received considerable attention over the last few decades (Scoones, 2007). International awareness and demands for sustainable development have pushed the sustainable narrative into the forefront of developmental goals. The Brundtland Report of 1987, released by the United Nations entitled “Our Common Future” resulted in the classic modern definition of sustainable development. Agriculture is the primary source of food, shelter and fibre for the world and therefore has a leading role to play in the realisation of a sustainable future (Van Calker, Berentsen, Giesen and Huirne, 2005). Most sustainability research in agriculture has adopted a particular scale and dimension, leading to a host of definitions and methodologies for its assessment (Hayati, 2017, Janker and Mann, 2018, Latruffe, Diazabakana, Bockstaller, Desjeux and Finn, 2016, Smith and McDonald, 1998). Different approaches to assessing sustainable agriculture have developed in hand with various conceptual methods resulting in a lack of a common understanding of what sustainable agriculture means (Janker and Mann, 2018, Smith and McDonald, 1998). Agriculture has a leading role to play in sustainable development in the South African context; however, commercial agriculture in South Africa has been associated with human rights violations as well as unsustainable social practices and exploitation of employees (Mather, 2007, Muller, Vermeulen and Glasbergen, 2012b). This research was conducted in an agri-processing company that had expanded rapidly and been experiencing high absenteeism and increased conflict in the work environment in an attempt to identify how many of the chosen indicators of on-farm internal social sustainability were present in the company. The literature review helped to contextualise the subject of sustainability, agricultural sustainability, agricultural social sustainability and various methods for assessing agricultural sustainability. The subject of agriculture social sustainability within the South African context was expanded upon, and a framework was selected from the literature to develop the aims of this research. The Barrientos and Visser (2013) framework had been utilised in evaluating social sustainability in the South African agricultural context before, and when compared with relevant literature was deemed appropriate for this research. Through the use of a case study approach, thirty employees were interviewed on a one on one basis with close ended questions. Data was collected through the use of an odd-numbered Likert scale and simple descriptive statistics were used to answer questions regarding which socially sustainable on-farm internal indicators, identified in the literature and in the chosen framework, occurred in the company in question. The research looked to further critically evaluate the aspects of social sustainability that were implemented by the company and discuss the challenges relating to those identified indicators that were not implemented. Finally, the research made recommendations to improve the on-farm internal social sustainability of the company in question. The results of this research showed that out of the ten indicators of on-farm internal social sustainability that were relevant in the framework, five were present in the company in question and five were absent. Those indicators that were present were: contracts, wages, benefits, communication with management and adequate representation. Indicators that were not present included health and safety, employee wellbeing, the level of discrimination, the level of harsh treatment and employee decision making. It was apparent from the results that there are many challenges to implementing on-farm internal socially sustainable policies and practices in the company. Chief amongst these challenges was communication with employees concerning policies that have been implemented and the information that is available to the employees regarding these policies. The company at the centre of this research should ensure that policies are put in place that helps to convey information to employees and ensure that the policies are implemented efficiently and effectively. It was recommended that the company initiate a Human Resources division that would be responsible for communication and the implementation of policies to increase on-farm internal social sustainability. By increasing social sustainability in the workplace, the company should be able to increase its efficiencies and effectiveness in a competitive global market. The researcher is aware that the findings of this research are not generalizable and are specific to the company in question both in terms of scale and location. Further research is needed to determine the impact of individual indicators and the weighting of each indicator in a specific environment. Further research should be conducted in a company that is in a similar industry and has similar staff demographics to the company at the centre of this research, as the scale of measure will influence the results.

Applying high-resolution remote sensing to quantify baboon damage at a sub-compartment level in pine stands in the Mpumalanga escarpment region of South Africa

- Description: Managing risk in intensively managed monoculture plantation forests is an essential task to ensure sustainable yield and a continuous flow of forest products. However, since risks can be either biotic or abiotic, not all of them have a predictable pattern of spread, which can cause severe losses if management does not have the chance to implement mitigation action. Monitoring the change in forest health is vital as this provides the opportunity for preventative management and quantifies the amount of damage that management has to deal with. To provide this window of opportunity for appropriate action, constant monitoring is required. Until recently, forest health was measured through field surveys which provided adequate data. This procedure, however, is time consuming. Remote sensing has become very popular as a monitoring tool, due to its ability to provide assessment data in a fraction of the time. In this study, baboon damage in plantations along the Mpumalanga escarpment area of South Africa was monitored using remote sensing methods. While there are many methods of forest health monitoring using remote sensing, some approaches are less suitable as they either monitor damage caused at a plantation level, use lower spatial resolution (>10m) datasets or map damage using one available time period. The purpose of this study was first to establish the impact of baboon damage through time, using Sentinel-2 satellite imagery with all vegetation indices available, and the Extreme Gradient Boosting (XGboost) algorithm. The second part focused on analysing the damage at a tree level using PlanetScope imagery using a deep Learning approach. Overall, the study found that the use of Sentinel-2 data and PlanetScope data could accurately distinguish between the varying severity of baboon damage, achieving an accuracy of 95% and 82%. The processing time of the deep learning Artificial Neural Network (ANN) was greatly affected by the number of hidden layers and neurons used. Implementation of techniques used in this study has the potential to improve the accuracy of forest health monitoring in compartment forestry in South Africa.

- Description: This study redefines the political in dance by drawing on the scholarly concept of the “choreopolitical” (André Lepecki) and extending it into analysing related concepts such as the “postdramatic” (Hans-Thies Lehmann), performance and/as research, among others from Performance Studies scholarship as well as from First Physical Theatre Company’s pioneering legacy of production, pedagogy and research in making Phyical Theatre performance. Following from the notion that performance is both a site and a method of study/knowing, the research invites a rethinking of the relationship between art (performance), epistemology and the political, in the sense that performance becomes a way, not of simply re-presenting the political but, as its own way of knowing, actively questioning the very categories on which the political is premised. The argument for Physical Theatre as having nascent potential to invoke what I call “the power of the small” is analysed as a choreopolitical method and community of practice that has a generative capacity to produce the “intimate revolts” (Julia Kristeva) or body of questions that can perform the imaginative curiosities/forms required to create provocative, subversive, ethical, reflexive and charged performance. My argument is supported by critical commentary, insight, choreological analysis and reflection on the dramaturgical strategies and choreopolitics of selected commissioned choreographers and dance forms that extended FPTC’s manifesto and production between 1993 and 2015. My project has the following three goals: (i) to contextualise, conceptualise and identify key issues in the identity, pedagogy and performance ethos of Physical Theatre as a performance philosophy and form; (ii) to engage critically with the praxis of Physical Theatre within the contextual, cultural, historical and political relationships between Physical Theatre and other performance practices in South Africa; and (iii) to document, analyse and interpret selected claims, works and performance processes from the archive of FPTC’s repertory and training manifesto from 1993 to 2015. The research evaluates the political significance and consequence of FPTC’s heritage and legacy problematising constraints, possibilities, tensions, failures and proposing the hope of imaginative entanglements with practising freedoms.

‘Regulated Flexibility’ and labour market regulation: a case Study of Twizza Soft Drinks in the Eastern Cape, South Africa

- Description: Following the negotiated settlement, which led to the ANC assuming power in South Africa in1994, debates concerning the nature of the South African labour market ensued between policy makers and economists alike. Central to understanding the South African labour market was the policy objective of regulated flexibility that has guided the formation of labour legislation in the post-1994 period, including the Labour Relations Act of 1995, the Basic Conditions of Employment Act of 1997, the Employment Equity Act of 1998 and the Skills Development Act of 1998. Regulated flexibility attempts to accommodate the interests of the employer for flexibility and the interests of the employee in regulation or security. These four Acts and the relevant provisions contained within them are the central focus of this research paper, in particular how they affect the case study firm Twizza Soft Drinks. An interpretivist approach was utilised as the preferred research methodology with in-depth, semi-structured interviews being the primary source of data collection. This research paper attempts to situate more clearly the impact of South Africa’s macro-economic policies since 1994 on labour market policy and undertakes an exploration of internal dynamics of firms in response to exogenous factors, such as government regulation. The key finding suggest that some Acts (BCEA, LRA) do not impose a significant burden on the firm and some provisions can lead to beneficial outcomes such as business modernisation and the adoption of formal Human Resource Practices. Conversely, some provisions contained in the EEA increase the administrative burden and therefore increase the indirect cost on the firm.

- Description: Learners in South Africa continue to underperform in the international and national mathematics benchmarking tests. In the primary school, poor performance in mathematics is viewed as an indicator of limited number sense. Since the end of Apartheid, there has been a proliferation of classroom-based research that attempts to explain why learners are underperforming and find solutions to the problem. Research that seeks to explain learner underperformance attributes poor learner performance to social-economic issues, teachers’ poor content and pedagogical knowledge, the complexity of the Language of Learning and Teaching, and insufficient support for learners with Mathematics Learning Difficulties (MLD). With regards to the latter, research suggests that Foundation Phase teachers are not equipped to assist learners with MLD develop their number sense. This qualitative case study aims to investigate the knowledge that an expert Foundation Phase teacher draws on, in the process of teaching, to assist learners with MLD develop their number sense. Data generated from observations and interviews with a Grade One teacher was analysed using Rowland, Turner and Thwaites’ (2013) Knowledge Quartet. The study found that the participant Grade One teacher employed all four categories of the Knowledge Quartet when developing her learners’ number sense. In particular, she placed strong emphasis on vocabulary development as a means of circumnavigating MLD when developing number sense in a Grade One mathematics lesson. She demonstrated knowledge of: the importance of vocabulary in learning mathematics; how to develop the learners’ understanding of mathematics vocabulary (and concepts); and how to adapt her approach to support the number sense development of learners with MLD. This research has value for teacher education programmes, both pre- and in-service, as it highlights the knowledge that a Grade One teacher draws on as she develops the number sense of all her learners, including those with MLD.

Dynamics of stimulated luminescence in natural quartz: Thermoluminescence and phototransferred thermoluminescence

We investigate the dynamics of stimulated luminescence on this material through thermoluminescence (TL) and phototransferred thermoluminescence (PTTL). Measurements were made on unannealed natural quartz as well as quartz annealed at 800 and 1000̊C. The samples were annealed for 10 minutes and for 1 hour. The material, in its un- and annealed state has its main peak between 68 and 72̊C when measured at 1Cs ̃1 after a dose of 50 Gy. A study of dosimetric features and kinetic analysis was carried out on two prominent peaks, peak I and III for all the samples. The peaks show a sublinear dose response for irradiation doses between 10 and 300 Gy. Kinetic analysis shows that peak I is a first-order peak and peak III a general-order peak. Interestingly, we observe for peak I for the sample annealed at 800̊C for 1 hour an inverse thermal quenching behaviour. We demonstrate that a peak affected with an inverse thermal quenching-like behaviour can still show effect of thermal quenching when the dose the sample is irradiated to is significantly reduced. We ascribe the apparent dependence of thermal quenching on dose to competition between radiative and non-radiative transitions at the recombination centre. Peaks I, II, and III for all the samples were reproduced under phototransfer when the peaks, initially removed by preheating to a certain temperature are exposed to 470 and 525 nm light. The infuence of duration of illumination on the PTTL intensity of these peaks corresponding to various preheating temperatures is modelled using coupled first-order dfferential equations. The model is based on systems of acceptors and donors whose number and role depends on preheating temperature

Both function as extensions of masculinity, representing an ideological binary of male superiority and female inferiority. This manifests concretely in the perceived disparity regarding gender representation amongst rock musicians, particularly guitarists, and often leads to somewhat one-dimensional views about the misogyny associated with the genre (Gracyk 2001: 182-183). - Description: The research reported on in this thesis explored teacher mediation when a biliteracy approach through task-based teaching and learning is used in a Foundation Phase classroom in a township school in the Eastern Cape. It is an action research aimed at understanding and systematically investigating how and what it means to work with bilingual mediation to ensure cognition, with emphasis on task design, facilitation for cognition, mediation forms and language use. This thesis was motivated by three issues that are still not being adequately addressed: the low level of cognitive work in South African schools, the failure to use the home languages of children throughout schooling as a medium of instruction and assessment (while providing excellent access to English as subject), and a top-down approach to both policy development and teacher professionalisation. These three issues drove me to explore theories that can help address them, and that is how I came to rely mostly on mediation, biliteracy and a task-based approach to teaching. For this research, on data handling I prepared and taught six lessons, but only three of the six lessons were recorded, transcribed and analysed for empirical data. I chose data handling, as in my previous experience I found it to include all the mathematical problem-solving skills which involve addition subtraction, analysing and comparing information. It also offered opportunities for language use, and meaningful interactive co-construction and acquiring of knowledge in the process of teaching and learning. This turned into a form of theory-driven action research, which was also developmental. I was critically reflective on my practices, and my facilitation for cognition and how I use language to make cognition possible. I also looked at the types of activities that I gave learners to help reach maximum development. The data collected from the classroom interactions, shows how I, in some instances, would take decisions, implement them and then find them not to be effective. It also shows some of the challenges I came across, from myself and the learners. Learners challenges were, unfamiliarity with the systematic build-up of data handling, filling in tables, transferring information from one form into a different form, and constructing and analysing bar graphs. This was part of pedagogynot the policy, which indicated inadequate teacher development. This could be because data handling is allocated minimal weighting from the CAPS document, and teachers do not go as in depth as they need to in dealing with data handling. My challenge was to prepare the grade 3 class for more data handling encounters in the higher grades. I had to ensure they grasped data handling concepts in their mother tongue before the switch to English as LoLT, as prescribed by policy. Learners proved to have little or no knowledge with regards to data handling concepts, which meant I had to start from the basics, as I had nothing to build on, and then progress to grade 3 level in one year. This study suggests that for any concept that has to be taught, cognition must be a priority, and strategies on how to facilitate that needs to be well thought out. Teachers need to be aware of theories that can positively impact on their practices. Teacher development is key to improvement of education, especially in the Eastern Cape. That cannot be done in isolation, but in partnership with relevant stakeholders.

I chose data handling, as in my previous experience I found it to include all the mathematical problem-solving skills which involve addition subtraction, analysing and comparing information. It also offered opportunities for language use, and meaningful interactive co-construction and acquiring of knowledge in the process of teaching and learning. This turned into a form of theory-driven action research, which was also developmental. I was critically reflective on my practices, and my facilitation for cognition and how I use language to make cognition possible. I also looked at the types of activities that I gave learners to help reach maximum development. The data collected from the classroom interactions, shows how I, in some instances, would take decisions, implement them and then find them not to be effective. It also shows some of the challenges I came across, from myself and the learners. Learners challenges were, unfamiliarity with the systematic build-up of data handling, filling in tables, transferring information from one form into a different form, and constructing and analysing bar graphs. This was part of pedagogynot the policy, which indicated inadequate teacher development. This could be because data handling is allocated minimal weighting from the CAPS document, and teachers do not go as in depth as they need to in dealing with data handling. My challenge was to prepare the grade 3 class for more data handling encounters in the higher grades. I had to ensure they grasped data handling concepts in their mother tongue before the switch to English as LoLT, as prescribed by policy. Learners proved to have little or no knowledge with regards to data handling concepts, which meant I had to start from the basics, as I had nothing to build on, and then progress to grade 3 level in one year. This study suggests that for any concept that has to be taught, cognition must be a priority, and strategies on how to facilitate that needs to be well thought out. Teachers need to be aware of theories that can positively impact on their practices. Teacher development is key to improvement of education, especially in the Eastern Cape. That cannot be done in isolation, but in partnership with relevant stakeholders.

These were compared to the standard parametric approach common in practice which makes use of the Normal distribution for modelling lead time demand. The three reorder point calculation methods were each incorporated into the inventory policy simulations using data supplied by a South African automotive spare parts business. The simulations covered a period of twelve months and were repeated for multiple service levels ranging from 70 to 99 percent. Results of the simulations were compared at a high level as well as for groups of items identified using segmentation techniques which considered different item demand and lead time characteristics. Key findings were that the Normal approximation method was far superior in terms of the service level metric, while the variation of the Bookbinder and Lordahl [1989] method adopted in this study presented possible cost benefits at lower service levels.

Early detection is crucial for the effective treatment of malaria, particularly in cases where infection is due to Plasmodium falciparum. There is, therefore, an enduring need for portable, sensitive, reliable, accurate, durable, self-validating and cost-effective techniques for the rapid detection of malaria. Moreover, there is a demand to distinguish between various infectious species causing malaria. Research in the area of malarial biomarkers has identified a unique, species-specific, epitope of P. falciparum lactate dehydrogenase (PfLDH), enhancing prospects for the development of diagnostics capable of identifying the species causing malarial infection. In recent years, improvements have been made towards the development of rapid diagnostic tests for detecting malarial biomarkers. Owing to their low cost, ease of labeling, and high thermal stability (relative to antibodies), the development and synthesis of aptamers that target the malarial lactate dehydrogenase represents one of the key innovations in the field of rapid diagnostics for malaria. This study explored the generation of aptamers that specifically target P. falciparum. Two sets of aptamers with diagnostically-supportive functions were generated independently, through parallel SELEX of recombinantly-expressed, full-length Plasmodium falciparum lactate dehydrogenase (rPfLDH), and an oligopeptide comprising the P. falciparum-specific epitope on lactate dehydrogenase (LDHp). The latter offers a promising solution for generating aptamers capable of binding with high specificity to P. falciparum. In this work, an rLDH class of aptamers was generated when SELEX was performed using the full-length rPfLDH protein as the target and the LDHp class of aptamers was generated when SELEX was performed using the oligopeptide LDHp as a target. Aptamers were successfully generated through the process of SELEX (systematic evolution of ligands through exponential enrichment) following the study and application of several optimisation steps, particularly during the amplification stage of SELEX. Optimisation steps included the study of improvements in PCR conditions; role of surfactants (Triton-X), modifying the PCR clean-up protocol; and agarose gel excision. Structurally-relevant moieties with particular consensus sequences (GGTAG and GGCG) were found in aptamers both reported here and previously published, confirming their importance in recognition of the target. Novel moieties particular to this work (ATTAT and poly-A stretches) were identified. Clades of consensus sequences were identified in both the rLDH and LDHp groups of aptamers, where sequences in the rLDH clade did not show preferential binding to rPfLDH while those in the LDHp clade (particularly LDHp 3 and 18) were able to recognise and bind only LDHp. Of the 19 sequences returned from the parallel SELEX procedures for rPfLDH (11 sequences) and LDHp (8 sequences), six rPfLDH and all eight LDHp sequences underwent preliminary screening and those with low responses eliminated. Of the eight LDHp-targeting aptamer sequences, five were preliminarily shown to bind to LDHp, whereas only two rPfLDH-targeting sequences were shown to bind to the target (rLDH 4 and 7). To this small selection of rPfLDH oligonucleotide sequences, two more (rLDH 1 and 15) were chosen for further study based on their sequences, secondary and predicted tertiary conformations. Sequences chosen for further study were therefore: rLDH 1, 4, 7 and 15 in the rLDH class, and LDHp 1, 3, 11, 14 and 18 in the LDHp class. Binding properties of the aptamers towards their targets were investigated using enzyme-linked oligonucleotide assays (ELONA), fluorophore-linked oligonucleotide assays (FLONA), electromobility shift assays (EMSA), surface plasmon resonance (SPR), and GelRed dissociation assays, while applications towards aptasensors were explored using electrochemical impedance spectroscopy (EIS) and fluorescent microscopy. Some inconsistencies were seen for specific aptamer to target binding interactions using specific techniques; however, generally, binding to the targets was observed across the techniques assessed. These varied responses demonstrate the need to screen and validate aptamers using a variety of techniques and platforms not necessarily specific for the proposed application. From the aptamer binding screening studies using ELONA, the most promising aptamers generated were identified as LDHp 11, rLDH 4, rLDH 7 and rLDH 15. Aptamer rLDH 4, which was generated against rPfLDH, exhibited preferential and specific binding to the lactate dehydrogenase from P. falciparum, over the recombinantly-expressed lactate dehydrogenase from Plasmodium vivax (rPvLDH), albeit with lowered responses compared to LDHp 11 in ELONA and EMSA studies. However, in kinetic ELONA studies rLDH 4 showed binding to both rPfLDH and rPvLDH. Aptamer rLDH 7 showed high affinity for rPfLDH and rPvLDH in kinetic studies using ELONA. However, screening studies with ELONA indicates that aptamer rLDH 7 may not be suitable for diagnostic tests in serum samples given its non-specific binding to human serum albumin (HSA). Aptamer rLDH 15 exhibited species specificity for rPfLDH in screening studies using ELONA but showed affinity towards rPvLDH (albeit lower relative to its affinity for rPfLDH) in kinetic studies using ELONA. LDHp 11, generated against the PfLDH peptide, showed a clear preference for rPfLDH when compared to rPvLDH and other control proteins, in both sets of ELONA studies conducted, as well as EMSA, thus possessing a strong ability to identify the presence of Plasmodium falciparum owing to its generation against the species-specific epitope. While LDHp 1 demonstrated binding to plasmodial LDH in a flow-through system (SPR), so reiterating ELONA responses, it did not perform well in the remaining methodologies. Aptamers rLDH 1 and 15 and LDHp 3, 14 and 18 exhibited a mixed set of results throughout the target protein screening analyses and were, thus, not considered for selective binding in P. falciparum parasite bodies. In studies aimed at exploring biosensor assemblies utilising the developed aptamers, both rLDH 4 and LDHp 11, along with rLDH 7, LDHp 1 and pL1, demonstrated in situ binding to the native PfLDH in fluorescent microscopy. LDHp 11 exhibited FITC-based fluorescence equivalent to the anti-rPfLDHp IgY antibody in confocal fluorescent microscopy indicating superior binding to the native PfLDH compared to the remaining aptamers. An examination of electrochemical impedance as a platform for a biosensor assembly did not, in these studies, exhibit the required sensitivity using physiologically relevant concentrations of analyte expected for pLDH following infection with Plasmodium spp. Malstat/LDH activity was explored for application in a colorimetric aptasensor. A decrease in both rPfLDH and rPvLDH activity was observed following incubation with the tested aptamers, but rLDH 1, rLDH 7 and LDHp 14 did not exhibit similar decreases in rPvLDH activity. Aptamers rLDH 1, 4 and 7 and LDHp 11 and 14 were, therefore, not selected as candidates for LDH capture in LDH activity-based diagnostic devices for P. falciparum. The decreases in pLDH activity in the presence of aptamers could hold promise as direct or antagonistic malaria therapeutic agents. Preliminary studies on the application of aptamers as malaria therapeutic agents, while of interest, should be viewed with due caution given the challenges of aptamers reaching the intracellular native plasmodial LDH hosted within the red blood cells. In conclusion, this work has shown the ability of the LDHp 11 aptamer, generated in these studies, to selectively bind rPfLDH over rPvLDH, and to bind to the native PfLDH in fluorescent microscopy, indicating that this aptamer holds promise as a biorecognition element in malaria biosensors and other diagnostic devices for the detection, and differentiation, of P. falciparum and P. vivax. The use of a species-specific epitope of P. falciparum as a target in aptamer generation paves the way for similar such studies aimed at generating aptamers with species selectivity for other Plasmodium species.

Aim: This study aims to determine the knowledge, attitudes and behaviours that motivate food preparers in safe food preparation. Method: Building on existing work in understanding this phenomenon it asks: A purposive sample of male and female participants who prepare food at home were asked to complete a questionnaire. The questionnaire items were developed through focus group interviews and after a pilot study, presented to participants to complete. The items were subjected to principal component analysis (factor analysis), yielding a six-factor solution. The factor means, standard deviations were calculated and reliability was determined for each factor using Cronbach’s alpha as a measure of internal consistency. Group differences were determined through independent samples t-tests and anova. Results: The quantitative analysis demonstrated that food preparers have a fair knowledge and perception of how to prevent contamination. However, their behaviour is various aspects of food preparation might at times put them at risk for contamination. With regards to attitude, when comparing groups, people who prepare food with love and care demonstrated a more positive attitude towards food preparation than those who are less passionate to prepare food. On this basis, hygiene education should focus on the perceived threat to health as cues to action in changing attitudes and adherence to recommendations on hygiene. Conclusion: Further research is required to solicit the views of respondents below the age of 18 with regard to food-handling practices in pre-prepared food to ascertain and prevent further contamination and ensure food safety.

Applied phylogeography : mapping the genetic resource of Honeybush across the Cape Floristic Region

- Description: Aim: Honeybush tea is a herbal infusion made from members of the Cape Floristic Region (CFR) endemic Cyclopia Vent. Consting of 21 extant species, six are anthropogenically redistributed across the CFR for Honeybush cultivation. However, with no information regarding the distribution and levels of genetic diversity among wild Honeybush populations, anthropogenic translocation of cultivated genotypes may risk disrupting natural genetic diversity patterns. In this thesis, an applied phylogeographic approach is used to provide baseline insights into the spatial structuring of Honeybush genetic diversity. Location: The Cape Floristic Region (CFR), located along the southern Cape of South Africa. Methods: Spatial structuring of evolutionary lineages within Cyclopia are explored at a genus level using data generated from the combination of Anchored Hybrid Enrichment library preparation and high through put sequencing. A High Resolution Melt analysis (HRM) toolkit is then developed to screen haplotype variation across three non-coding chloroplast loci. This HRM toolkit coupled with haplotype confirmation Sanger sequencing is then applied to describe the spatial structuring of genetic diversity in wild populations for two Honeybush species (C. intermedia and C. subternata) and to compare genetic diversity among wild and cultivated populations of three Honeybush species (C. intermedia, C. subternata, and C. longifolia). A reanalysis of published phylogeographic data focusing on CFR plant taxa is used to test if phylogeographic breaks occur across the boundaries among adjacent Centers of Endemism (CoEs). Finally, the low-copy nuclear data set generated via AHE and high throughput sequencing is summarized. Results: Phylogeographic structuring was detected at the inter- and intraspecific levels in Cyclopia. The genus level analysis revealed a strong phylogenetic split between western CFR and eastern CFR endemic species in the nuclear and chloroplast genomes. At the intraspecific level, HRM proved to be a high throughput and accurate tool for haplotype detection, revealing phylogeographic structuring of genetic diversity in C.intermediaand C.subternata. This genetic diversity and structuring was not detected in cultivated Honeybush populations, which appear to have originated from a small number of founding individuals and have experienced a genetic bottleneck. Spatial structuring of genetic lineages is common among CFR plant taxa and the reanalysis of existing data found a significant trend for phylogeographic breaks tooccuracrosstheboundariesamongadjacentCoEs. Finally, over200000baseswere sequenced across 445 low-copy nuclear loci for 14 Cyclopia species. Conclusions: This thesis provides important baseline information on the spatial distribution of wild Honeybush genetic diversity. Phylogeographic structuring is present in wild Honeybush populations. This however is not accounted for in cultivated populations, which may place wild genetic diversity at risk of being disrupted if geneflow among wild and cultivated populations occurs. The results from this thesis should therefore be used to develop precautionary guidelines for the anthropogenic redistribution of Honeybush genetic material for cultivation. Furthermore, the novel insights into regional patterns of phylogeohraphic structuring of plant populations should encourage more efficient and hypothesis driven sampling designs in future phylogeographic work in the CFR. The high throughput sequence data generated in this thesis should transform Cyclopia to a model organism for the study of molecular evolution in the CFR.

- Description: Aim: Honeybush tea is a herbal infusion made from members of the Cape Floristic Region (CFR) endemic Cyclopia Vent. Consting of 21 extant species, six are anthropogenically redistributed across the CFR for Honeybush cultivation. However, with no information regarding the distribution and levels of genetic diversity among wild Honeybush populations, anthropogenic translocation of cultivated genotypes may risk disrupting natural genetic diversity patterns. In this thesis, an applied phylogeographic approach is used to provide baseline insights into the spatial structuring of Honeybush genetic diversity. Location: The Cape Floristic Region (CFR), located along the southern Cape of South Africa. Methods: Spatial structuring of evolutionary lineages within Cyclopia are explored at a genus level using data generated from the combination of Anchored Hybrid Enrichment library preparation and high through put sequencing. The paper explores what Black women students perceive to be Rhodes University's institutional culture and its impact on their lived realities. Subsequently, these women have learned who they are, and what place they occupy in South Africa today, through navigating a space not necessarily accommodating to Blackness and difference. There exists a plethora of literature on the issues which Black women scholars systematically encounter daily in the academy, in sub-Saharan Africa and beyond. Nonetheless, there needs to be a further inquiry on the question of belonging of Black womanhood in HWU post the student-led movements of the past few years that have renewed the challenge to South Africa's colonial past, its neoliberal present, and its scourge of gender-based violence. This paper captures an ongoing conversation around the role of Black women in addressing transformation in HWU. As a Black woman in an HWU, I found myself wondering whether there are certain experiences students like me have in common – realities with nuances we call to identify with to some extent. I collected data from six Black women with whom I conducted interviews, and used it to compile this report and its analysis. I believe that the social significance of this study speaks to the importance of hearing the stories of subaltern groups that are positioned in spaces of privilege, yet continue to be defined by the disadvantage of their gender, race, and various other factors.

- Description: Over the past several decades, the world’s population has been rapidly urbanising, which has resulted in a marked shift of global population from rural to urban areas. About half of the global population now reside in urban areas and is projected to increase to two-thirds by the year 2050. However, the majority of the future urban population growth is projected to be largely concentrated in Asia and sub-Saharan Africa. Despite the economic prospects normally synonymous with urbanisation, the current speed and scale of urban transformation comes with formidable challenges to contemporary urban society in these two continents. Urbanisation may presents social, economic, health and environmental challenges in urban areas, not least being a shift of the locus of food insecurity from rural to urban areas, leading to poor dietary diversity among many urban dwellers. Similarly, urbanisation alters key ecosystem services and drives habitat alteration and fragmentation, biodiversity loss, proliferation of non-native species and changes on species diversity and richness. The aforementioned changes can result in a deterioration of living conditions and decline on the quality of life for the urban dwellers. However, on the other hand, urban green infrastructure is posed as vital tool in promoting liveable cities and enhancing livelihood resilience. Against this backdrop, this study portrays wild plants, a component of urban green infrastructure, as a key resource in promoting food security and dietary diversity, thereby promoting livelihood resilience and reduced vulnerability, especially of the urban poor. Framed under the ‘right to the city’ approach, this study examined urban foraging practices and their potential contribution to dietary diversity and how they are shaped by and respond to urbanisation in two medium-sized South African towns. The study was conducted in the towns of Potchefstroom and Thabazimbi, South Africa. An explanatory sequential mixed method design was employed for data collection. Multi-stage sampling was employed in selecting the study participants. Firstly, the study towns were stratified into four socio-economic zones: informal, reconstruction and development programme, township and affluent. These zones resemble high density (informal, reconstruction and development programme), medium density (township) and low density (affluent) areas. Then, a total of 374 households were randomly sampled across the socio-economic zones for the main survey (Chapter 3 and 5). A subset of this sample, i.e. 72 participants was considered for dietary recalls, alongside with a random sample of 65 non-foragers (Chapter 4). An inventory of plant species available in a given foraging space was conducted from a total of 136 plots spread across different urban spaces (Chapter 2). Semi-structured interviews were conducted with a total of 81 participants (sub population of the main survey) for elucidating data on urban dynamics (Chapter 6). Lastly, complementary in-depth interviews with 46 participants were conducted. The findings showed that different urban spaces were constituted by diverse vegetation, with 262 distinct plant species encountered across the spaces. The majority (60.7%) were indigenous to South Africa. Species composition and diversity varied with space type, being significantly high in protected areas as compared to the other space types. About half (53%) of the identified plant species had at least one documented use, with medicine, food and firewood being modal, in order of frequency. Species composition and diversity differed between forageable and non-forageable species, being significantly high for forageable species. Urban foraging was widespread, about 68% of the respondents reported foraging. Foraging provided food, fuel energy, medicine, and cultural affirmation, among others. Despite the high prevalence rate of foraging, the study did not find any meaningful contribution of wild foods to overall diets. Nonetheless, wild foods demonstrated a substantial contribution in diversifying diets within particular food groups consumed such as vegetables, thus emerging essential in mitigating monotonous urban diets, particularly of the urban poor. Besides, the prevalence of foraging differed significantly between and within towns, being high for Thabazimbi (54.7%) and residents in the outskirts of town (86.2%) than Potchefstroom (45.3%) and residents in the inner part of town (33.3%). Participation in foraging was primarily a function of childhood (91.8%) exposure and experience with foraging, positive perceptions (79%) towards practice, resident in the outskirts of town (86.2%) and low household income (49.2%). The findings indicated that foraging was driven by multifaceted motivations, with culture (91.8%), health (88.7%), livelihoods needs (75.0%) and leisure (73.8%) being the most common. Vacant spaces (54.7%), riparian areas (30%), and domestic gardens (17.9%) were the most preferred foraging spaces, albeit with differences within towns. Similarly, the mean frequency of access varied with foraging spaces, being slightly high for vacant spaces than riparian areas and domestic gardens. The majority of the respondents were unaware of both formal and informal regulations managing urban landscapes, with those acknowledging some awareness falling short of articulating on the specificities of the regulations. Besides, the findings showed that foraging practice was threatened by urban transformation. Foragers perceived that the practice has changed over time, in terms of spaces, participants and regulations. There was a decreasing trend on the number and size of foraging spaces as well as shifting demographic trends of foragers. Moreover, urban transformation brought about a new set of regulations used for managing certain spaces within the urban landscape. However, some regulations prohibited or restricted access and rights to certain spaces for foraging. Hence, this prompted foragers to devise adaptation strategies to cope with the said changing dynamics. Exploring new foraging sites within and outside forager’s neighbourhoods and negotiating access entry were the most frequently mentioned adaptation strategies. This study contributes to the emerging body of knowledge on urban foraging, which has being seldom understood in the Global South.

To that effect, financial institutions have credit scoring systems in place to be able to estimate the credit risk associated with a customer. Various classification models are used to develop credit scoring systems such as k-nearest neighbours, logistic regression and classification trees. This study aims to assess the performance of different classification models on the prediction of credit card payment default. Credit data is usually of high dimension and as a result dimension reduction techniques, namely principal component analysis and linear discriminant analysis, are used in this study as a means to improve model performance. Two classification models are used, namely neural networks and support vector machines. Model performance is evaluated using accuracy and area under the curve (AUC). The neuarl network classifier performed better than the support vector machine classifier as it produced higher accuracy rates and AUC values. Dimension reduction techniques were not effective in improving model performance but did result in less computationally expensive models.
