This qualitative study pays attention to aspects of the choir’s history, its performance practice and of the “web” of community members connected to and involved in its activities. Through interviews and personal notes this in-depth study provides a “micronarrative” of this choir within the “web” of the Overberg community itself. It aims to not only interpret this narrative with regard to the meaning behind actions and their symbolic importance in society, but also to explore its relevance in the broader context of current South African cultural discourse. During this research it became evident that policy makers and potential funders regard this predominantly white choir with its Western repertoire as a form of undesirable exclusivity and elitism. This study opposes such a point of view, contending instead that elitism in the form of excellence has the power to defy barriers of social standing and ethnicity, and to unite people through a collective sense of ownership.

The botanical importance and health of the Bushmans estuary, Eastern Cape, South Africa

- Description: The Bushmans Estuary is one of the few permanently open estuaries in the Eastern Cape that is characterized by large intertidal salt marshes. Freshwater inflow to the estuary has decreased as a result of abstraction by more than 30 weirs and farm dams in the catchment. The mean annual run-off is naturally low (38 x 106 m3 y-1) and thus abstraction and reduction of freshwater inflow to the estuary is expected to cause a number of changes. The aims of this study were to determine the current health/status of the estuary based on the macrophytes and microalgae and identify monitoring indicators for the East London Department of Water Affairs, River Health Programme. Changes in the estuary over time were determined from available historical data which were compared with present data. This analysis showed that under normal average conditions freshwater inflow to the estuary is very low, less that 0.02 m3 s-1 most of the time. Under these conditions the estuary is in a homogenous marine state. Vertical and horizontal salinity gradients only form when high rainfall and run-off occurs (> 5 m3.s-1). Salinity gradients from 30.1 PSU at the mouth to 2.2 PSU in the upper reaches were measured in 2006 after a high flow event. However the estuary quickly reverted back to its homogenous condition within weeks after this flood. This study showed that freshwater inflow increased nutrient input to the estuary. Total oxidised nitrogen (TOxN) and soluble reactive phosphorus (SRP) concentrations were higher in August 2006, after the flood, than during the other low flow sampling sessions. TOxN decreased from a mean concentration of 21.6 μM in 2006 to 1.93 μM in February 2009. SRP decreased from 55.3 μM to 0.2 μM respectively. With the increased nutrient availability, the response in the estuary was an increase in phytoplankton biomass. After the 2006 floods the average water column chlorophyll-a was 9.0 μg l-1, while in the low freshwater inflow years it ranged from 2.1 to 4.8 μg l-1. The composition of the phytoplankton community was always dominated by flagellates and then diatoms, with higher cell numbers in the nutrient-enriched 2006 period. Although the water column nutrient data indicated that the estuary was oligotrophic, benthic microalgal biomass (11.9-16.1 μg.g-1) in the intertidal zone was comparable with nutrient rich estuaries. Benthic species indicative of polluted conditions were found (Nitzschia frustulum, Navicula gregaria, Navicula cryptotenelloides). These benthic species were found at the sites where wastewater / sewage seepage had occurred. Benthic diatom species also indicated freshwater inflow. During the high flow period in 2006 the dominant diatoms were fresh to brackish species that were strongly associated with the high concentrations of TOxN and SRP (Tryblionella constricta, Diploneis smithii, Hippodonta cf. gremainii, and Navicula species). During the freshwater limited period of 2008 and 2009 the benthic diatom species shifted to a group responding to the high salinity, ammonium and silicate concentrations. The species in this group were Nitzschia flexa, Navicula tenneloides, Diploneis elliptica, Amphora subacutiuscula and Nitzschia coarctata. Ordination results showed that the epiphytic diatom species responded to different environmental variables in the different years. Most of the species in 2008/2009 were associated with high salinity, temperature, dissolved oxygen, ammonium and silicate concentrations while the response was towards TOxN and SRP in 2006. The dominant species were Cocconeis placentula v euglyphyta in 2006; Nitzschia frustulum in 2008; and Synedra spp in 2009. The average biomass of the epiphytes was significantly lower in May 2008 than in both August 2006 and February 2009; 88.0 + 17.7 mg.m-2, 1.7 + 0.8 mg.m-2, and 61.8 + 14.4 mg.m-2 respectively. GIS mapping of past and present aerial photographs showed that submerged macrophyte (Zostera capensis) cover in 1966 and 1973 was less than that mapped for 2004. Salt marsh also increased its cover over time, from 86.9 ha in 1966 to 126 ha in 2004, colonizing what were bare sandy areas. Long-term monitoring of the health of the Bushmans Estuary should focus on salinity (as an indicator of inflow or deprivation of freshwater), benthic diatom identification and macrophyte distribution and composition (for the detection of pollution input), and bathymetric surveys (for shallowing of the estuary due to sedimentation).

- Description: This study investigated the role of selected ward committees 29 and 32 of Nompumelelo and Tsholomnqa respectively in the Buffalo City Municipality (BCM) in enhancing basic service delivery. In terms of the White Paper on Local Government (1998:4), developmental local government promotes a system that centres on working with local communities to find sustainable ways to meet their needs and improve the quality of their lives. The study on that basis examines the nature and extent to which wards 29 and 32 committees enhanced basic service delivery within BCM. The study is premised on the assumptions that: The involvement of party-elected Councillors in ward committees inhibits members of ward committees from playing an active role in their communities; Ward committees are not clearly communicating municipality programmes to their communities; BCM is biased in favour of urban wards in service delivery; If the committees of wards 29 and 32 were given more powers to play a much wider role in providing leadership and make decisions in their communities (being elevated from an advisory role to ward management structures), they would make an impact in enhancing basic service delivery; and, With additional decision-making powers ward committees could play a more effective role in local government matters. The perceived slow pace of service delivery by municipalities has resulted in growing impatience and dissatisfaction, in particular among poor communities. This has been demonstrated by the spontaneous protests and unrests directed at municipalities that have been taking place nationally since 2003. The uprisings explain two aspects, namely local government is considered by communities to be the delivery arm of government in South Africa and poor communities feel betrayed because their active participation in government-provided spaces for participation such as municipal elections, ward committees and IDPs did not yield the result of promised development (Theron, 2008:36). iv The study employed the qualitative research design using an interview survey as a method of data collection and the reviewing of existing study material and documents to test the validity of the afore-mentioned assumptions. Lastly, with the aim of assisting BCM in nurturing the potential of ward committees operating in its area of jurisdiction, the following recommendations based on the findings of the study are made: BCM considers subjecting ward committees to structured formal and accredited training; BCM delivers services in a legally compliant manner; BCM delegates sufficient powers to ward committees; Ward committees be trained together with officials that drive the CBP process when BCM starts implementing the process; and, BCM provides support to ward committees.

- Description: This interpretive case study sought to determine what influenced action competence-oriented teaching and learning processes in a high school environmental club. The goals were to describe a year of activities, within the TALON environmental club at Hudson Park High School in East London, South Africa and to identify how pedagogical, contextual and relational factors influenced action competence processes amongst club members. I explored how action competence theory and praxis have helped in other school education settings with the aim of establishing better environmental practices. The IVAC model of pedagogy (Jensen & Schnack, 1997) was used as a framework to operationalise action competence processes within the TALON Club. This entailed carefully examining investigative, visioning, action and change processes unfolding through the four themes of the club: plants, animal welfare, waste issues and our community outreach programme at Bongulethu High School. Using questionnaires, interviews, observation and a journal as the main methods of data collection I was able to make five analytical statements that related to the influence of pedagogy, the influence of context and the influence of relational dynamics within the TALON Club. The pedagogical processes were affected by the TALON members wanting to avoid any activities that were ‘too much like school’. However this served to compromise the action and visioning phases within the club, resulting in superficial and poorly-informed action-taking. The influence of context served to both enable and constrain action competence processes. The club and school structures therefore shaped the form that fledgling actions took as the TALON members participated in the meetings and outings and fulfilling the traditional expectations of the school community. Relational dynamics and a desire for fun took precedence over the action competence processes. Although the Club members were motivated by a sense of moral responsibility they were unable to articulate this concept into the club activities. In addition there were the power gradients between the educators and members to consider as determinants of the extent of participatory, democratic interactions. This complex interplay of factors influenced the action competence processes in the club. Recommendations were made on how best to work with the concepts of participation and democracy towards improving the reflexivity and knowledge base of both the educators and learners. In this way action competence processes will be better supported within the club in future.

The highly conserved, non-structural protein 2C has numerous roles during the virus life cycle and is essential for virus replication. Although the protein has been well studied in the case of PV, its interactions with the host cell during picornavirus infection is poorly understood. Theiler’s Encephalomyelitis virus (TMEV) is a picornavirus that infects mice, and is being used in our laboratory as a model in which to study the 2C protein. In this study, polyclonal antibodies against the TMEV 2C protein were generated and used to localise the protein in infected cells by indirect immunofluorescence. To produce antigen for immunisation purposes, the TMEV-2C protein sequence was analysed to identify hydrophilic and antigenic regions. An internal region of the 2C representing amino acid residues 31-210 was selected, expressed in bacteria and purified by nickel NTA affinity chromatography. Time course analysis of 2C (31-210) showed that the peptide was maximally expressed at 5 hours post induction. The communal farmers widely used conventional medicines however, proper administration methods were not followed. Ethnoveterinary medicines were used to prevent and treat disease, even by the younger stock farmers. The stock farmers used ethnoveterinary medicines mainly because of the lack of finance to purchase the conventional medicines, even though the latter was preferred. The stock farmers actively participated in state programmes but felt that they needed more state veterinarian visits and state provided medications because they could not afford private veterinarians and conventional medicines. Therefore, this study attempts to contribute to an understanding of the use of ethnoveterinary medicine and communal farmers’ animal healthcare practices.

This legacy posed a serious challenge to Namibia when it set out to achieve a national goal, Vision 2030. Vision 2030 inspires Namibia to be among the eveloped countries in 2030. In an attempt to achieve the goals outlined by the mission statement of Vision 2030, the Ministry of Education (MoE) introduced two national education initiatives: the Education and Training Sector Improvement Program (ETSIP), and the National Standards and Performance Indicators (NSPIs). These would be implemented in Namibian schools as, respectively, the Ministry’s strategic plan and its policy document. The NSPIs were introduced to address the fragmentation of standards and inequity in regions and schools in Namibia, as they aimed to standardise the provision of equal, quality education across the country (Namibia. MoE, 2005a, p. 1). The NSPI policy calls for schools to take quality assurance seriously, with particular emphasis on School Self-Evaluation (SSE), a process that encourages self-management (Namibia. MoE, 2007a, p. 3). Thus, the standards set out by the NSPIs became the yardstick by which internal and external school evaluators could assess whole school performance. The purpose of this present study was to investigate the perceptions and experiences of principals in the Ohangwena Region following the implementation of the NSPIs. It is a case study involving two Inspectors of Education (IoEs), twelve principals, and four Heads of Department (HODs). Interviews, document analysis, questionnaires, and a focus group interview were used to collect the data. The study found that principals acknowledge the importance of the NSPIs, and believe that they would succeed in improving the provision of quality education if they were fully implemented. The study found that schools implemented Performance Indicators 5.3 (School Administration) Aspect 5.3.2 (Storage and retrieval of information) in Key Area 5. However, the results of the study also showed that principals experienced many challenges in the implementation of the NSPIs. These included a lack of understanding among principals and teachers of the NSPIs, a lack of understanding of contemporary leadership theories (that would otherwise help principals to implement change effectively), and a lack of support in terms of skills and resources from both the Regional Office in Ohangwena and the MoE itself. The study found that the NSPIs have not yet improved school performance, and consequently that the pass rate remains the same as before the introduction of the NSPIs, four years ago. The SSE, which aims to help schools identify their strengths and weaknesses, and encourages them to establish and make use of a School Development Plan (SDP) to address weaknesses and sustain strengths, is characterised by a high degree of dishonesty. Thus, the information furnished by SSEs, SDPs, and Teachers Self-Evaluations (TSEs) is unreliable, lacks credibility, and has the further ill-effect of skewing schools’ performance targets. Therefore, this problem could be addressed when the rationale of completing the SSE is clearly explained to principals and teachers.

Suboptimal preparation for the Ironman triathlon can have detrimental effects on mental and physical condition. The purpose of this longitudinal investigation was to examine the relationship between a number of performance changes and fatigue markers during training for an Ironman as well as immediately after the event, in an attempt to better understand the effects of ultraendurance training. Eighteen athletes training for the Ironman; South Africa, 2009 were recruited for the study. Over the 6 month data collection period body mass, training load (TRIMP and Session x RPE methods), physiological responses (waking heart rate, postural dizziness, sleep ratings), changes in psychological state (profile of mood states - POMS), reported immunological responses (symptoms of illness), biochemical changes (salivary cortisol and alpha amylase) and performance (8 km submaximal running time trial (TT) and race day performance) were measured. These responses were compared to a control sample (n=15). Results show a significant increase (p<0.05) in training load (3899.4 ± 2517.8) four weeks prior to the event. Fatigue scores significantly increased (p<0.05) concurrently with this significant increase (p<0.05) in training. TT performance did not significantly (p<0.05) alter during the time course of training. It was however strongly correlated to training load (R2=0.85) and modestly related to race performance (R2=0.65). The signs and symptoms of upper respiratory tract infections (URTI) were prevalent during the training period, decreasing during the taper and race period. Large standard deviations were found within the majority of the responses. During the final two weeks of preparation, tension scores were significantly increased (p<0.05) while training load significantly decreased (p<0.05) during the final week of preparation. Cortisol increased significantly (p<0.05) immediately post race (0.507±0.15

- Description: In 1998, South Africa introduced outcomes-based education to affect the shifts in pedagogy considered necessary following the move to democracy in 1994. Across the globe, the use of the portfolio to assess learners is increasingly being promoted as a form of progressive pedagogy. Hence, its adoption by the country as a new form of assessment practice was warranted. However, how the portfolio is constructed and perceived by educators within the classroom can become problematic in practice. This was apparent in the South African context and justifies research into how the portfolio is constructed as an assessment method in educational policy and by educators in the classrooms. The Curriculum Guide Directive text and the transcripts of twenty-one interviews carried out in three Eastern Cape schools served as the source of data for this study. Discourses make up a powerful framework of spoken, written and symbolic texts of institutional bureaucracies. Within these institutions human subjects are defined and constructed. Therefore, discourse construction acts as institutional forms of knowledge which can exude power over the individual if not made transparent. I used critical discourse analysis to uncover the discourses that were embedded within the Curriculum Guide Directive and to identify the discourses entrenched within the educators’ perceptions of the portfolio. The discourses in the Curriculum Guide Directive suggest the construction of the portfolio as a method of surveillance to track whether the educators are preparing learners for the school leaving examination and instructing the educators to provide evidence in the form of a portfolio. Analysis of the transcripts of the twenty one interviews with educators from three schools in the Eastern Cape indicated that the portfolio was not seen as a form of assessment by the educators, but as a form of discipline put in place by the Department of Education to ensure that they are preparing the learners for the school leaving examination and are able to produce proof of this preparation in the form of a portfolio. The discourses from the research highlighted that the school leaving examination is hegemonised into South African educational pedagogy as a form of traditional assessment and is not challenged even when it is no longer bringing about the dominant ideological goals. The portfolio was not seen as an assessment method by the educators nor represented as such in the Curriculum Guide Directive. Therefore, the job of the portfolio became that of a policing tool. Thus, a school curriculum which has been introduced with the intent of furthering social justice can become exclusive in practice, even with the best intentions.

Isolation of xylanolytic multi-enzyme complexes from Bacillus subtilis SJ01

- Description: Cellulose and hemicellulose account for a large portion of the world‘s plant biomass. In nature, these polysaccharides are intertwined forming complex materials that require multiple enzymes to degrade them. Multi-enzyme complexes (MECs) consist of a number of enzymes working in close proximity and synergistically to degrade complex substrates with higher efficiency than individual enzymes. The cellulosome is a cellulolytic MEC produced by anaerobic bacteria that has been studied extensively since its discovery in 1983. The aim of this study was to purify a cellulolytic and/or hemicellulolytic MEC from an aerobic bacterium of the Bacillus genus. Several bacterial isolates were identified using morphological characteristics and 16S rDNA sequencing, and screened for their ability to degrade cellulose and xylan using a MEC. The isolate that produced a high molecular weight protein fraction with the greatest ability to degrade Avicel®, carboxymethyl cellulose (CMC) and birchwood xylan was identified as Bacillus subtilis SJ01. An optimised growth medium, consisting of vitamins, trace elements, birchwood xylan (as the carbon source), and yeast and ammonium sulphate (as the nitrogen sources), increased the production of CMCase and xylanase enzymes from this bacterium. The removal of a competing bacterial strain from the culture and the inhibition of proteases also increased enzyme activities. A growth curve of B. subtilis SJ01 indicated that xylanase production was highest in early stationary growth phase and thus 84 hours was chosen as the best cell harvesting time. To purify the MECs produced by B. subtilis SJ01 size-exclusion chromatography on a Sephacryl S-400 column was used. - Description: Cellulose and hemicellulose account for a large portion of the world‘s plant biomass. In nature, these polysaccharides are intertwined forming complex materials that require multiple enzymes to degrade them. Multi-enzyme complexes (MECs) consist of a number of enzymes working in close proximity and synergistically to degrade complex substrates with higher efficiency than individual enzymes. The cellulosome is a cellulolytic MEC produced by anaerobic bacteria that has been studied extensively since its discovery in 1983. The aim of this study was to purify a cellulolytic and/or hemicellulolytic MEC from an aerobic bacterium of the Bacillus genus. Several bacterial isolates were identified using morphological characteristics and 16S rDNA sequencing, and screened for their ability to degrade cellulose and xylan using a MEC. The isolate that produced a high molecular weight protein fraction with the greatest ability to degrade Avicel®, carboxymethyl cellulose (CMC) and birchwood xylan was identified as Bacillus subtilis SJ01. An optimised growth medium, consisting of vitamins, trace elements, birchwood xylan (as the carbon source), and yeast and ammonium sulphate (as the nitrogen sources), increased the production of CMCase and xylanase enzymes from this bacterium. The removal of a competing bacterial strain from the culture and the inhibition of proteases also increased enzyme activities. A growth curve of B. subtilis SJ01 indicated that xylanase production was highest in early stationary growth phase and thus 84 hours was chosen as the best cell harvesting time. To purify the MECs produced by B. subtilis SJ01 size-exclusion chromatography on a Sephacryl S-400 column was used. The intervention was carried out in a Grade 10 Life Sciences class which consisted of 35 learners. The first cycle consisted of seven lessons carried out over two weeks in normal school time. The main sources of data from my lesson interventions were the learners' written work, their journal entries, the researcher's journal, the field notes from the non-participant observer and interviews with two focus groups. Both the interviews and the lessons were recorded and transcribed. The research data reveals that the ability of the Junior Secondary learners to write explanations was hindered by language problems. In addition, a lack of integration of language issues into the subject content was a contributing factor to their inadequate writing skills. Spelling errors and lack of expression in English second language contribute to the problem of writing good explanations. The data has also revealed that learners‟ writing skills can be improved by implementing suitable teaching strategies, such as the curriculum cycle as proposed by Gibbons (2002). The curriculum cycle can be modified to fit the learners' needs and the real teaching situation. The amount of practice required to master the skill of writing in the genre of explanations should be considered.

There is general scholarly agreement that the fast track process has been accompanied by various socio-economic and political challenges, including an increase in levels of food insecurity. This thesis examines fast track reform in specific relation to the livelihoods of smallholder households and household food security amongst land beneficiaries. It argues that the problem of food insecurity in Zimbabwe is a complex social, political and economic issue, which cannot be simplistically reduced to the failures of fast track. Understanding household food insecurity post-2000 needs to go beyond the notion that the nation‟s food security hinges on overall levels of production alone. In particular, livelihoods and food security need to be conceptualised at community and household levels. Therefore, while addressing the broad macro-level analysis and discourse around the process of fast track, the study also adopts a micro-level analysis to look at the varied impact of fast track on the actual beneficiaries. The research focuses on small-scale beneficiaries in the Goromonzi District of Zimbabwe and, through the use of the sustainable livelihoods framework, looks at how their local contexts have been influenced by the wider socio-economic and political processes, and how beneficiaries have sought ways of coping with the challenges they face.

Data was collected from the x-ray departments of two training hospitals in Windhoek, Namibia over a five month period. A retrospective RFA was conducted to determine the department and student reject rates for both departments before intervention. Emphasis was placed on exposure related reject films. ECs were compiled and introduced at Katutura State Hospital (venue B) by the researcher. The students were instructed to use these charts. At Windhoek Central Hospital (venue A) no ECs were used. A prospective RFA was conducted to establish department and student reject rates at both hospitals after the intervention at venue B. Results: During the retrospective phase the department reject rate for venue A was 21 percent while the student reject rate was 23 percent. At venue B 24 percent and 26 percent were scored respectively. Students at venue A produced rejected radiographs due to overexposure (49 percent) and underexposure (23 percent), whilst 37 percent was recorded for both causes at venue B. At venue A, 35 percent of films were rejected due to incorrect mAs selection, at venue B the figure was 42 percent. Undiagnostic radiographs due to inaccurate kV selection comprised 62 percent for venue A and 59 percent for venue B. During the prospective phase the department reject rate for venue A was 20 percent and that of the students was 19 percent. For venue B 12 percent and 11 percent were scored respectively. At venue A radiographs rejected due to over and underexposure were 43 percent and 33 percent respectively while those at venue B were 33 percent and 34 percent. Incorrect mAs selection caused 33 percent of discarded films at venue A and 38 percent at venue B. The figures for inaccurate kV selection were 68 percent and 62 percent for venues A and B. Conclusions: The introduction and use of ECs lowered the student reject rate at venue B in the prospective phase.

However, as much as chronic illnesses are inevitable among employees, there should be ways at least to curb negative social and goal achievement impact which they bring in organisations. In this regard, a survey of 80 employees was conducted to examine the relationship between employees' knowledge about, contact with, and attitudes towards their co-employees with chronic illnesses (CI). Results indicated that employees' knowledge about and contact with individuals diagnosed with chronic illnesses were significantly related to better attitudes toward this population. The findings also revealed that knowledge was related to positive attitudes among employees who had more personal contact with individuals with Chronic illnesses (CI) and such findings indicate the importance of exposing employees to information about chronic illnesses and direct practice experience with individuals who have these illnesses. To be noted also is that 39% of the respondents preferred to be exposed to health bulletins and magazines; 18% of the respondents preferred the help of health workers and about a third (30%) favoured brochures, posters and printed materials such as flyers.
