A social realist account of the emergence of a formal academic staff development programme at a South African university

- Description: Using social realist theory and particularly the morphogenetic/morphostatic methodology advocated by Margaret Archer, this study offers a critical examination of the emergence of a formal academic staff development programme at a small South African university (SSAU). Archer’s morphogenetic approach enabled an investigation of the interface between culture, structure and agency (at macro, mezo and micro levels) in order to theorize about the material, ideational and agential conditions that obtained and which in turn enabled the emergence of the Postgraduate Diploma of Higher Education (PGDHE) at the SSAU. The study therefore advances concrete propositions about the cultural, structural and agential conditions for transformation which existed at a particular time in the history of higher education (and the subfield of educational development) which enabled the introduction of the PGDHE. Analysis of the data suggests that what occurred at SSAU was a disruption of the morphostatic synchrony between structure and culture brought about by new discourses and structures emanating from the broader international and national higher education context. In particular, it seems that attempts at reconciling the constraining contradictions between the discourses and structures related to quality assurance on the one hand and educational development on the other resulted in a conjunction between transformation at the levels of both the cultural system and social structure. This conjunction, along with the actions of key Institutional agents and the morphogenesis of the staff of the Educational Development Unit, created sufficiently enabling conditions in the Institution for the introduction of the PGDHE. The research adds to knowledge through insights into the contribution that the ideas, beliefs, values, ideologies and theories about higher education broadly and about educational development specifically make to enabling or constraining conditions for the professionalization of academic staff in higher education institutions. It uncovers how relevant structures at the international, national and institutional levels can shape the practice of educational development and specifically staff development. It has generated insights into how the relevant people and the positions they hold can impact on staff development practices. In summary, the research could contribute towards emancipatory knowledge which could be used by SSAU and educational development practitioners elsewhere to inform future planning and decision making in relation to educational development and more specifically staff development practices in their contexts.

Evolutionary and biogeographic studies in the genus Kniphofia moench (Asphodelaceae)

- Description: Kniphofia, a genus of approximately 71 species, is almost entirely African with two species occurring in Madagascar and one in Yemen. Commonly known as ‘red hot pokers’ they are popular among horticulturists. The genus is also well known for its complex alpha taxonomy. To date, no studies have examined the phylogenetic relationships among species or the evolutionary history of the genus, and little work has been done on their biogeography. The main focus of this study was (i) to review the alpha taxonomy, (ii) to assess diversity and endemism in Kniphofia, (iii) to use DNA sequence data to reconstruct a specieslevel phylogeny to understand intra-generic species relationships and evolutionary processes (iv) to use phylogeographic approaches to study the biogeography and evaluate biogeographical patterns, and (v) to assess anatomical variation and determine if anatomical characters are useful for species delimitation. It was found that the genus has six centres of diversity, five of which are centres of endemism. The South African Centre is the most speciose and is also the largest centre of endemism. Kniphofia shows a strong Afromontane grassland affinity in Tropical and East Africa. In South Africa, it is found from high altitudes to coastal habitats, with the most speciose regions being Afromontane grasslands. It is thus not considered to be an Afromontane element, but rather an Afromontane associate. Five major evolutionary lineages were identified using cpDNA sequence data (trnT-L spacer), four of which are southern African. The fifth lineage is represented by material from Madagascar, East and Tropical Africa. The nuclear ITS region failed to provide resolution, as many sequences were identical. The five lineages recovered using cpDNA showed some congruence with geographic origin rather than the taxonomic arrangement based on morphology. All of the species with multiple samples were non-monophyletic. This could be due to hybridisation and/or incomplete lineage sorting. The nested clade analysis, although preliminary, did not completely agree with the phylogenetic analyses. One of the three third level nested clades appears to show fragmentation between the Cape Region, KwaZulu-Natal and northern parts of southern Africa. Furthermore, another nested clades recovered suggest a range expansion and radiation from the Drakensberg into the adjacent Drakensberg-Maputoland-Pondoland transition. Morphological species of Kniphofia exhibited substantial leaf anatomical variation and anatomical characters do not cluster samples into their morphological species. The anatomical results do not fit any geographic pattern, nor do they correspond to the lineages recovered using molecular markers or the nested clades. Leaf anatomical variation does not appear to be influenced by geographical or environmental factors. However, hybridisation may play a role but was not tested in this study. In light of the above findings it is proposed that the evolutionary and biogeographic history of Kniphofia is strongly linked to tectonic events, and Quaternary climatic cycles and vegetation changes. Tectonic events (viz. uplifts) may have resulted in vicariance events that may account for the five cpDNA lineages recovered in phylogenetic analyses, while Quaternary climatic cycles and vegetation changes may have had a more recent impact on evolution and biogeography. It is hypothesised that the ancestral area for Kniphofia was much more widespread when Afromontane grasslands were more extensive during cooler and drier glacial episodes. Kniphofia on the high mountains of Tropical and East Africa would have tracked Afromontane grasslands as they expanded their ranges in cooler periods. While during wetter and warmer interglacial periods Kniphofia would have retreated into refugia on the mountains of Tropical and East Africa, with no gene flow possible between these refugia. In South Africa, where latitude compensates for altitude, Kniphofia may have maintained a distribution that extended into the lowlands even during interglacials. A cyclic climate change hypothesis implies that populations of Kniphofia (at different phases of the climatic cycle) would have experienced periods of contractions and fragmentation followed by periods of range expansion and coalescence or secondary contact. Altitudinal shifting is proposed to be the most likely mechanism for fragmentation and range expansion, and would would possibly promoted hybridisation. Within the five lineages there is evidence for recent differentiation as the branch lengths are short, there are numerous nonmonophyletic species and numerous identical haplotypes (cpDNA and ITS) which collectively indicate a recent radiation in southern Africa. A recent radiation would also account for the taxonomic confusion and difficulty in differentiating morpho-species. These climatic events may also account for the substantial anatomical variation in southern African Kniphofia species.

- Description: This thesis explored use of metallophthalocyanines as electrocatalysts towards thiol and thiocyanate oxidation, nitrosothiol decomposition and reduction of oxygen, as well as biomimetic and photo-catalysts of cyclohexene oxidation. 2-mercaptoethanol (2-ME), L-cysteine (CYS) and reduced glutathione (GSH) thiols were oxidized on cobalt tetra ethoxythiophene and cobalt tetra phenoxy pyrrole phthalocyanine modified glassy carbon electrodes, whose catalytic activity was found to depend on pH, film thickness and method of electrode modification. Oxidation of thiocyanate (SCN-), CYS and 2-ME was catalyzed by a selfassembled monolayer of cobalt tetraethoxythiophene Thiocyanate oxidation occurred via two electron transfer, whereas that of CYS and 2-ME required 1 electron. The oxidations of SCN- and 2-ME were catalyzed by ring based processes, while CYS was catalyzed by both Co[superscript III]/Co[superscript II] process and ring-based processes. Oxidation of GSH and 2-ME was conducted on screen printed graphite electrodes modified with cobalt phthalocyanine. Activity depended on method of electrode modification and CoPc % composition. Decomposition of Snitrosoglutathione occurred in the presence of copper ions and NaBH[subscript 4]. Reduced and oxidized glutathione were detected as products using cobalt phthalocyanine adsorbed on an ordinary pyrolytic graphite electrode. Reduction of oxygen was electro-catalyzed by adsorbed manganese phthalocyanine complexes on glassy carbon electrodes. FePc, FePc(Cl)[subscript 16], CoPc and CoPc substituted with phenoxypyrrole and ethoxythiophene ligands were also used as electro-catalysts. Oxygen reduction occurred via two electron transfer in acidic and neutral media forming hydrogen peroxide, while water was formed in basic media via four electron transfer. Cyclohexene oxidation using tert-butylhydroperoxide or chloroperoxy benzoic acid as oxidants in the presence of FePc, FePc(Cl)[subscript 16] and CoPc formed cyclohexene oxide, 2-cyclohexen-1-ol, 2- cyclohexen-1-one and adipic acid. Product selectivity depended on the nature of catalyst and oxidant. The FePc(Cl)[subscript 16] catalyst was transformed into a µ-oxo dimer during the oxidation process while M[superscript III]Pc intermediates were formed with Co[superscript II]Pc and Fe[superscript II]Pc catalysts. - Description: Heat shock protein 70 (called DnaK in prokaryotes) is one of the most prominent groups of chaperones whose role is to prevent and reverse protein misfolding. PfHsp70 is a heatinducible cytoplasm/nuclear localised Plasmodium falciparum Hsp70. PfHsp70 is thought to confer chaperone cytoprotection to P. falciparum during the development of malaria fever. The objective of this study was to examine the chaperone properties of PfHsp70 using a bioinformatics approach, coupled to in vivo and in vitro analysis. Structural motifs that qualify PfHsp70 as a typical Hsp70 chaperone were identified. Although PfHsp70 has a higher similarity to human Hsc70 than E. coli DnaK, in vivocomplementation assays showed that PfHsp70 was able to reverse the thermosensitivity of E. coli dnaK756 (a temperature sensitive strain whose DnaK is functionally compromised). Two residues (V401 and Q402) in the linker region of PfHsp70 that are critical for its in vivo function were identified. Constructs were generated that encoded the ATPase domain of PfHsp70 and the peptide binding domain of E. coli DnaK (to generate PfK chimera); and the ATPase domain of E. coli DnaK fused to the peptide binding domain of PfHsp70 (KPf). The two chimeras were tested for their ability to reverse the thermosensitivity of E. coli dnaK756 cells. Whilst KPf was able to reverse the thermosensitivity of the E. coli dnaK756 cells, PfK could not. Previously, PfHsp70 purification involved urea denaturation. Using a detergent, polyethylenimine (PEI), PfHsp70 was natively purified. Natively purified PfHsp70 had a basal ATPase activity approximately two times higher than the previously reported activity for the protein purified through urea denaturation. PfJ4, a type II Hsp40, could not stimulate the ATPase activity of PfHsp70 in vitro. Arch and hydrophobic pocket substitutions (A419Y, Y444A and V451F) were introduced in the PfHsp70 peptide binding domain. Similar substitutions were also introduced in the KPf chimera. PfHsp70-V451F (hydrophobic pocket mutant) had marginally compromised in vivo function. However, a similar mutation (V436F), introduced in KPf abrogated the in vivo function of this chimera. The arch and hydrophobic pocket derivatives of PfHsp70 exhibited marginally compromised in vivo function, whilst equivalent mutations in KPf did not affect its in vivo function. The ability of PfHsp70 and its arch/hydrophobic pocket mutants to suppress the heatinduced aggregation of malate dehydrogenase (MDH) in vitro was investigated. Whilst PfHsp70 arch mutants displayed marginal functional loss in vivo, data from in vitro studies revealed that their functional deficiencies were more severe. This is the first study in which an Hsp70 from a parasitic eukaryote was able to suppress the thermosensitivity of an E. coli DnaK mutant strain. Findings from the in vivo and in vitro assays conducted on PfHsp70 suggest that this protein plays a key role in the life-cycle of P. falciparum. Furthermore, this study raised insights that are pertinent to the current dogma on the Hsp70 mechanism of action.

The arch and hydrophobic pocket derivatives of PfHsp70 exhibited marginally compromised in vivo function, whilst equivalent mutations in KPf did not affect its in vivo function. The ability of PfHsp70 and its arch/hydrophobic pocket mutants to suppress the heatinduced aggregation of malate dehydrogenase (MDH) in vitro was investigated. Whilst PfHsp70 arch mutants displayed marginal functional loss in vivo, data from in vitro studies revealed that their functional deficiencies were more severe. This is the first study in which an Hsp70 from a parasitic eukaryote was able to suppress the thermosensitivity of an E. coli DnaK mutant strain. Findings from the in vivo and in vitro assays conducted on PfHsp70 suggest that this protein plays a key role in the life-cycle of P. falciparum. Furthermore, this study raised insights that are pertinent to the current dogma on the Hsp70 mechanism of action.

- Description: This thesis examines the 1998 complaint of racism against the Mail & Guardian, a leading exponent of South Africa's alternative press in the 1980s, and important contemporary producer of investigative journalism. The study is framed within a cultural studies approach, analysing the Mail & Guardian as constituted by a 'circuit of production': its social context, production, texts, and audiences. The thesis makes three main arguments. First, that the claim of racism cannot be understood outside of a consideration of both the changing political milieu, and subtle changes within the Mail & Guardian itself. Significant social changes relate to the reconfiguration of racial and class identities wrought by the 'Mbeki state'. Within the Mail & Guardian, the thesis argues for the importance of the power and subjectivity of the editor as a key 'factor' shaping the identity of the paper, evidenced in its production practices and textual outputs. In this regard, the thesis departs from a functionalist analysis of particular 'roles' within the newsroom, drawing instead on a post-structuralist approach to organisational studies. Based on this production and social context, the thesis examines key texts which deal with aspects of South Africa's social transformation, and which exemplify aspects of the Mail & Guardian's reporting which led to the complaint of racism by the Black Lawyers Association (BLA) and the Association of Black Accountants (ABASA). Their complaint was that the Mail & Guardian's reporting impugned the dignity of black people, and in so doing was a violation of their rights to dignity and equality which are constitutionally guaranteed. However, as freedom of the press is also guaranteed by the South African constitution, their complaint to the South African Human Rights Commission (SAHRC) resulted in public debate about these contending rights. My second argument relates to the jurisprudential approach to racism, and the related issue of affirmative action, which informed the complaint against the paper. Contrary to the 'normative', liberal approach to these issues, this thesis highlights Critical Race Theory as the jurisprudential basis for both the claimants' accusation of racism against the Mail & Guardian, and aspects of its implicit use in South African human rights adjudication. The thesis argues that in failing to recognise these different philosophical and political bases of legal reasoning, the media, including the Mail & Guardian, in reporting on these matters failed in their purported role of serving the public interest. The thesis concludes by applying Fraser's critique of Habermas's notion of a single, bourgeois public sphere to journalism, thereby suggesting ways in which the critiques of some of the Mail & Guardian's own journalists could be employed to enlarge its approach to journalism - giving voice to constituencies seldom heard in mainstream media.

Synthesis, properties and reactions of Novel Quinone Methides

- Description: Novel p-quinone methides have been synthesized by the dealkylation of 5-(p-alkyloxyaryl)- 10,11-dihydro-5H-dibenzo[a,d]cyclohepten-5-ols and related compounds. Aspects of the dealkylation reaction were investigated using computational methods in order to identify possible intermediates and postulate reasons for the observed reactivity patterns. This included studying the effect of varying the size of the central B-ring, changing the alkyloxy group, and altering the substitution pattern on the parent alcohols. We have assessed the relative energies of reaction intermediates and have also evaluated the influence of factors such as charge delocalisation, LUMO properties of the carbocations and thermodynamic factors on the dealkylation reaction. The use of different dealkylating reagents was also briefly investigated. Demethylation of 1,3- dimethyl-11-(4-methoxyphenyl)-6,11-dihydrodibenzo[b,e]oxepin-11-ol with pyridine hydrochloride led to acid-catalyzed ring-contraction of the parent alcohol to form a novel substituted anthraquinone, 9-(4-hydroxyphenyl)-1,3-dimethyl-anthracen-10-(9H)-one, in good yield. The general reactivity of the p-quinone methides of interest to us was explored by subjecting these compounds to reaction with a range of nucleophiles (bases, Grignard reagents and alcohols). A range of aryl Grignard reagents were reacted with the p-quinone methides, with the main product isolated in almost all cases being the aryl-coupled 1,2-addition product. The nucleophilic addition reactions of alcohols were supported by a computational study and a probable reaction mechanism has been postulated. A base-catalyzed rearrangement is proposed to account for the formation of products in which dehydrogenation of the ethane bridge was observed. These studies showed that in these p-quinone methides, chemical reactivity is strongly influenced by steric crowding, resulting in reversal of the normal 1,2- vs. 1,6- selectivities expected for nucleophilic addition. The ketalization process was explored further using diols and thiols. Products analogous to those obtained with the monohydric alcohols resulted from the diols, along with a series of novel bis-ethers. A range of miscellaneous reactions of 4-(dibenzo[a,d]cycloheptan-5-ylidene)cyclohexa-2,5,- dienone and related systems were investigated. Functionalization by epoxidation, dichlorocarbenation and Diels-Alder reactions, photochemical and [2+2] cycloaddition were attempted and reduction and oxidation reactions were also explored. Photochemical demethylation of an ortho-methoxyl substituent on the p-quinone methide system was observed to occur in good yield. The p-quinone methides underwent reductive coupling in the presence of Zn/AlCl3. The electronic spectra of highly conjugated carbocations were obtained and their potential as novel dyes evaluated. A low-temperature Grignard exchange reaction followed by spontaneous cyclization upon workup, was successful in synthesizing the lactone, spiro[10,11- dihydro-5H-dibenzo[a,d]cyclohepten-(3’,4’H)-phenyl-5,2’(5’H)-furan-5’-one], in one step from the starting ketone. A novel seven-membered Malachite Green dye analogue, 11-(4- dimethylamino-phenyl)-3-morpholin-4-yl-6,11-dihydro-dibenzo[b,e]oxepin-11-ol, was also synthesized and its electronic spectra compared to that of the unannulated Malachite Green dye series. All novel compounds synthesized were characterized using NMR, IR and HRMS-analysis.

- Description: PART A The syntheses, spectroscopic and electrochemical characterisation of a series of titanium and tantalum phthalocyanine complexes are reported. The complexes are unsubstituted and substituted at either the peripheral or non-peripheral positions with sulphonates, aryloxy, arylthio or amino groups. The complexes mostly exhibit Qbands in the near-infrared region as well as interesting properties in different solvents. The interaction of differently sulphonated titanium and tantalum phthalocyanine complexes with methyl viologen (MV[superscript 2+]), and hence the stoichiometry and association constants are evaluated. Detailed photophysicochemical properties of the complexes were investigated and are for the first time presented with fluorescence lifetimes easily obtained from fluorescence quenching studies. The transformation of 1-hexene photocatalysed by aryloxy- and arylthio-appended complexes is also presented for the first time. The electrochemical properties of the complexes are unknown and are thus presented. Cyclic (CV) and square wave (SWV) voltammetries, chronocoulometry and spectroelectrochemistry are employed in the study of the complexes. Two oneelectron reductions and a simultaneous 4-electron reduction are observed for the unsubstituted Cl[subscript 3]TaPc. Reduction occurs first at the metal followed by ring-based processes. The tetra- and octa-substituted derivatives however exhibit peculiar electrochemical behaviour where a multi-electron transfer process occurs for complexes bearing certain substituents. For all complexes, the first two reductions are metal-based followed by ring-based processes. A comparative study of the electrocatalytic activities of the complexes towards the oxidation of nitrite is also investigated. The complexes are immobilised onto a glassy carbon electrode either by drop-dry or electropolymerisation methods. All the modified electrodes exhibit improved electrocatalytic oxidation of nitrite than the unmodified electrodes by a twoelectron mechanism producing nitrate ions. Catalytic currents are enhanced and nitrite overpotential reduced to ~ 0.60 V. Kinetic parameters are determined for all complexes and a mechanism is proposed. PART B: The syntheses and electrochemical characterisation of chromium and titanium complexes for the selective trimerisation of ethylene to 1-hexene are presented. The synthesis of the chromium complex requires simple steps while tedious steps are used for the air-sensitive titanium complex. The spectroscopic interaction of the chromium complex with the co-catalyst methylaluminoxane is investigated. The complexes are characterised by electrochemical methods such as cyclic voltammetry and spectroelectrochemistry.

- Description: The prevention of outbreaks of waterborne diseases remains a major challenge to public health service providers globally. One of the major obstacles in this effort is the unavailability of on-line and real-time methods for rapid monitoring of faecal pollution to facilitate early warning of contamination of drinking water. The main objective of this study was to develop a β-glucuronidase (GUD)-based method that could be used for the on-line and real-time monitoring of microbial water quality. GUD is a marker enzyme for the faecal indicator bacteria Escherichia coli. This enzyme breaks down the synthetic substrate p-nitrophenyl-β-D-glucuronide (PNPG) to D-glucuronic acid and p-nitrophenol (PNP), which turns yellow under alkaline pH. The enzymatically produced PNP was used to detect GUD activity. In situ GUD assays were performed using running and stagnant water samples from the Bloukrans River, Grahamstown, South Africa. The physico-chemical properties of environmental GUD were determined, after which a liquid bioprobe and a microbial biosensor modified with Moraxella 1A species for the detection of the enzyme activity were developed. In order to determine the reliability and sensitivity of these methods, regression analyses for each method versus E. coli colony forming units (CFU) were performed. The storage stabilities of the bioprobe and biosensor were also investigated. The physico-chemical properties of in situ GUD were different from those of its commercially available counterpart. The temperature optimum for the former was between 35 and 40 °C while for the latter it was 45 °C. Commercial (reference) GUD had a pH optimum of 8.0 while the environmental counterpart exhibited a broad pH optimum of between pH 5.0 and 8.0. The liquid bioprobe had a limit of detection (LOD) of GUD activity equivalent to 2 CFU/100 ml and a detection time of 24 h. The method was less labour intensive and costly than the culturing method. The liquid bioprobe was stable for at least four weeks at room temperature (20 ± 2 °C). The biosensor was prepared by modifying a glassy carbon electrode with PNP degrading Moraxella 1A cells. The biosensor was 100 times more sensitive and rapid (5-20 min) than the spectrophotometric method (24 h), and was also able to detect GUD activity of viable but non-culturable cells. Thus it was more sensitive than the culturing method. Furthermore, the biosensor was selective and costeffective. The possibility of using a Pseudomonas putida JS444 biosensor was also investigated, but it was not as sensitive and selective as the Moraxella 1A biosensor. The Moraxella biosensor, therefore, offered the best option for on-line and real-time microbial water quality monitoring in South African river waters and drinking water supplies.

- Description: The prevention of outbreaks of waterborne diseases remains a major challenge to public health service providers globally. One of the major obstacles in this effort is the unavailability of on-line and real-time methods for rapid monitoring of faecal pollution to facilitate early warning of contamination of drinking water. The main objective of this study was to develop a β-glucuronidase (GUD)-based method that could be used for the on-line and real-time monitoring of microbial water quality. GUD is a marker enzyme for the faecal indicator bacteria Escherichia coli. This enzyme breaks down the synthetic substrate p-nitrophenyl-β-D-glucuronide (PNPG) to D-glucuronic acid and p-nitrophenol (PNP), which turns yellow under alkaline pH. A visual programming environment, which can be used to build context aware applications as well as a proof-of-concept machine-based memory manager, is conceptualized and implemented. An experimental machine-based memory manager is implemented and evaluated. The model describes a machine-based memory manager which manages an individual's external memories by context. It addresses the management of external memories which accumulate over long periods of time by proposing a context aware file system which automatically organizes external memories by context. It describes how personal memory management can be facilitated by machine using six entities (life streams, memory producers, memory consumers, a memory manager, memory fragments and context descriptors) and the processes in which these entities participate (memory capture, memory encoding and decoding, memory decoding and retrieval). The visual programming environment represents a development tool which contains facilities that support context aware application programming. For example, it provides facilities which enable the definition and use of virtual sensors. It enables rapid programming with a focus on component re-use and dynamic composition of applications through a visual interface. The experimental machine-based memory manager serves as an example implementation of the machine-based memory manager which is described by the model developed in this thesis. The hardware used in its implementation consists of widely available components such as a camera, microphone and sub-notebook computer which are assembled in the form of a wearable computer. The software is constructed using the visual programming environment developed in this thesis. It contains multiple sensor drivers, context interpreters, a context aware file system as well as memory retrieval and presentation interfaces. The evaluation of the machine-based memory manager shows that it is possible to create a machine which monitors the states of an individual and his environment, and manages his external memories, thus supporting and augmenting his biological memory.

Collaborative research with traditional African health practitioners of the Nelson Mandela Metropole : antimicrobial, anticancer and anti-diabetic activities of five medicinal plants

- Description: The promotion and development of indigenous knowledge pertaining to the traditional African healing system is one of the prime objectives set out by the South African government. Despite excellent research opportunities and funding, the biggest problem with ethnopharmacological research is a lack of interaction with indigenous communities, which tends to dilute the benefits this research has to offer these communities. The primary aim of this study was thus to promote the traditional African healing system through collaborative medicinal plant research with local traditional health practitioners. The research collaboration aimed to validate some biological activities of traditional remedies used by collaborating traditional health practitioners and ensured interactive sessions where scientific literature, research practices, findings and relevant legislation were discussed and debated. The joint development of a medicinal garden was a valuable tool in realising these goals. Aqueous and ethanol extracts of Bulbine frutescens, Ornithogalum longibracteatum, Ruta graveolens, Tarchonanthus camphoratus and Tulbaghia violacea were selected for antimicrobial, anticancer and anti-diabetic screening, because of their sustainable utilisation potential. The ethanol extract of T. violacea produced the best antimicrobial activity on Bacillus subtilis (100% growth inhibition) and Candida albicans (89% growth inhibition) at 250 μg/ml. The EC50 for the ethanol extract of T. violacea against HT29 colon cancer cells was 101 μg/ml. The aqueous extracts of B. frutescens and T. camphoratus (0.5 and 50 μg/ml) produced the highest overall glucose uptake activity in Chang liver and C2C12 muscle cells. T. camphoratus was unanimously chosen by participating practitioners as the plant to be investigated further. The aqueous extract of T. camphoratus increased glucose uptake in C2C12 muscle cells through increased translocation of GLUT4 to the plasma membrane and activation of the PI3-kinase and AMP-kinase pathways. It produced some alpha-glucosidase inhibitory activity at concentrations of ≥ 200 μg/ml. Apart from interactive feedback seminars at which the findings were presented to participating practitioners, all scientific literature regarding the plants was summarised, translated, compiled and given to participating practitioners in written format. An indigenous knowledge agreement has been negotiated and will formalise the collaboration in future. It is recommended that future research focuses on plants with economic development potential that can be cultivated in the medicinal garden.

- Description: My involvement with mathematics education amongst township educators and learners over the past seven years has highlighted the absence of sustained meaning and meaning making of mathematical concepts. It appears though that this instrumental rather than relational understanding of FET mathematics is not unique to township learners but is encountered amongst learners of all socio-economic classes and is representative of many FET mathematics learners. Given that the language of learning and teaching is a major contributory factor in the South African education system, it appears that the language of mathematics itself is a greater exacerbating factor for many learners of mathematics. The exclusive algorithmic approach to classroom mathematics further seems to alienate many learners from the essence of the meaning of mathematical tasks. This research undertakes to determine whether metateaching and metalearning as forerunners to metacognition facilitates the acquisition of the sustained meaning of mathematical concepts. Metateaching and metalearning refer to the acute and deliberate awareness by educator and learner as to what constitutes concepts. Teaching and learning therefore presupposes the deconstruction of concepts into its subsumed derivative roots. It also assumes an awareness of the tacit degrees of abstraction that characterise tasks and the content of tasks. This in turn has implications for the educator's adopted sequence of topics for instruction. Metacognition implies awareness on the part of the learner (and educator) as to how material is learned and a further awareness as to how that learning can be sustained. Whether we ascribe meaningful learning to radical or social constructivism, or to associationist didactive approaches, or a combination of these, we are making assumptions about how learners acquire and sustain mathematical meaning because mathematics is, by and large a symbolic language often devoid of affective connotation. Furthermore our assessments of learners' tasks amount to clinical corrections of austere formulae wrapped in algorithmic procedures which manifest nothing of a learner's experience of mathematics or the deeper understanding (or misunderstandings) which characterise a learning and/or assessment episode. To this end the research design of this interpretive case study requires learners to expound in textual accounts their thoughts as they describe the evolution of a mathematical process as they approach a solution and eventually interpret it. The textual account exposes the concept definition for what it really is in a learner's understanding of it and it is the expressiveness of language that indicates whether the understanding of a learner is approaching the concept image. The textual accounts vary in richness in terms of mathematical register and this in turn reflects the conceptual depth. The mechanism which seems to promote the conversion from concept definition to concept image is the graphical representation of the mathematical task or procedure, possibly because of its greater concreteness as opposed to the abstraction of its algebraic form.

- Description: This study originated from an evaluation of the performance of a commercially available high concentration point focus concentrator PV system. The effect of module design flaws was studied by using current-voltage (I-V) curves obtained from each module in the array. The position of reverse bias steps revealed the severity of mismatch in a string of series-connected cells. By understanding the effects of the various types of mismatch, power losses and damage to the solar cells resulting from hot spot formation can be minimized and several recommendations for improving the basic performance of similar systems were made. Concern over the extent and type of defect failure of the concentrator photovoltaic (CPV) cells prompted an investigation into the use of a light beam induced current (LBIC) technique to investigate the spatial distribution of defects. An overview of current and developing LBIC techniques revealed that the original standard LBIC techniques have found widespread application, and that far-reaching and important developments of the technique have taken place over the years. These developments are driven by natural progression as well as the availability of newly developed advanced measurement equipment. Several techniques such as Lock-in hermography and the use of infrared cameras have developed as complementary techniques to advanced LBIC techniques. As an accurate contactless evaluation tool that is able to image spatially distributed defects in cell material, the basis of this method seemed promising for the evaluation of concentrator cells.

A preliminary examination of selected biological links between four Eastern Cape estuaries and the inshore marine environment

- Description: The coastal transition zone (CTZ), which links the terrestrial and marine environments, has been identified as a region of high biological diversity and elevated production. Results of studies conducted in the northern hemisphere indicate that the links between estuaries and the adjacent marine environment is critical to ecological functioning within the CTZ. This study assessed the influence of selected estuaries with different hydrodynamic characteristics on the adjacent marine environment along the south-eastern coastline of southern Africa. Four estuaries were examined, including two permanently open systems, the fresh water deprived Kariega and fresh water dominated Great Fish, and two temporarily open/closed estuaries (TOCE), the Kasouga and East Kleinemonde. Results of the study indicated that outflow of estuarine water from the Great Fish Estuary contributed to a plume of less saline water being evident within the adjacent marine environment. The plume of water was associated with increased zooplankton biomass and particulate organic matter (POM) and chlorophyll-a concentrations. Adjacent to the Kariega Estuary, no evidence of fresh water outflow into the marine environment was observed. However, in the sea directly opposite the mouth of the estuary an increase in zooplankton abundance and biomass was evident. Results of numerical analyses indicated that the increase in zooplankton abundance observed adjacent to the mouth of both permanently open estuaries could not be attributed to the export of zooplankton from the estuary, but rather the accumulation of marine species within the region. The mechanisms responsible for this accumulation were not determined, but it was thought to be associated with increased food availability in the estuarine frontal zone. A similar, but less dramatic biological response was also observed in the marine environment adjacent to the two TOCEs. It is suggested that the increase in biological activity within these regions could be ascribed to seepage of estuarine or ground water through the sand bar that separates these estuaries from the sea. Results of stable carbon isotope analyses indicated that both the Great Fish and Kariega estuaries exported carbon to the nearshore marine environment. The area influenced by estuarine derived carbon was dependent on the volume of estuarine outflow to the marine environment. Adjacent to the fresh water dominated Great Fish Estuary, estuarine derived carbon was recorded up to 12km from the mouth, while adjacent to the fresh water deprived Kariega, estuarine derived carbon was only evident directly opposite the mouth. The recruitment of macrozooplankton (> 2cm) into the fresh water deprived Kariega Estuary was in the range recorded for other permanently open southern African estuaries with higher fresh water flow rates. This indicates that the mechanisms which allow estuarine dependent larvae to locate and enter estuaries are not related to fresh water inflow. In conclusion, this study has demonstrated that despite their small size relative to European and North American systems, South African permanently open and temporarily open/closed estuaries also influence biological activity within the adjacent nearshore marine environment.

- Description: The coastal transition zone (CTZ), which links the terrestrial and marine environments, has been identified as a region of high biological diversity and elevated production. Results of studies conducted in the northern hemisphere indicate that the links between estuaries and the adjacent marine environment is critical to ecological functioning within the CTZ. This study assessed the influence of selected estuaries with different hydrodynamic characteristics on the adjacent marine environment along the south-eastern coastline of southern Africa. Four estuaries were examined, including two permanently open systems, the fresh water deprived Kariega and fresh water dominated Great Fish, and two temporarily open/closed estuaries (TOCE), the Kasouga and East Kleinemonde. Results of the study indicated that outflow of estuarine water from the Great Fish Estuary contributed to a plume of less saline water being evident within the adjacent marine environment. The plume of water was associated with increased zooplankton biomass and particulate organic matter (POM) and chlorophyll-a concentrations. Adjacent to the Kariega Estuary, no evidence of fresh water outflow into the marine environment was observed. However, in the sea directly opposite the mouth of the estuary an increase in zooplankton abundance and biomass was evident. Results of numerical analyses indicated that the increase in zooplankton abundance observed adjacent to the mouth of both permanently open estuaries could not be attributed to the export of zooplankton from the estuary, but rather the accumulation of marine species within the region. The mechanisms responsible for this accumulation were not determined, but it was thought to be associated with increased food availability in the estuarine frontal zone. A similar, but less dramatic biological response was also observed in the marine environment adjacent to the two TOCEs. It is suggested that the increase in biological activity within these regions could be ascribed to seepage of estuarine or ground water through the sand bar that separates these estuaries from the sea. Results of stable carbon isotope analyses indicated that both the Great Fish and Kariega estuaries exported carbon to the nearshore marine environment. It has been suggested that all second language (L2) learners, in the process of mastering a target language (TL), develop a linguistic system that is self-contained and different from both the learner's native language and the TL. This system is referred to as 'interlanguage' (lL). In the process of SLA, IL evolves into an ever-closer approximation of the TL, and ideally, a learner's IL should continue to advance until it becomes equivalent to the TL. However, it has been observed that somewhere in the L2 learning process, IL may reach one or more plateaus during which the development of the IL is delayed or arrested. A permanent cessation of progress toward the TL is referred to as 'fossilization'. Researchers in SLA agree that motivation is one of the key factors influencing language-learning success and studies suggest that some language learning motivation may be related to the need for achievement. The purpose of this research was to establish if adult ESL learners are aware of fossilization and, to examine if motivation, and more specifically achievement motivation, is a factor in IL fossilization. The participants in this study consisted of 15 ESL learners in Puerto Rico who had at least eight years of formal ESL training. The instrument used to gather information included a questionnaire to obtain demographical and qualifying data, an 'English Language Proficiency Evaluation' to determine levels of IL fossilization, a 'Measure of Achievement Motivation' to ascertain achievement motive, and individual and group interviews in order to ascertain perception(s) regarding the role of motivation on fossilization and perceptions regarding the barriers to achieving TL competency. The research demonstrated that there is a moderate to strong positive relationship between IL fossilization and achievement motivation, i.e., high achievement motive is correlated to TL competency and descending levels of achievement motive are correlated to ascending levels of IL fossilization. The findings have significant implications for both ESL learning and instruction, and suggest that not all IL fossilization may be permanent.

- Description: Providing relevant and quality schooling for all South African learners is the paramount goal of the South African National Department of Education. South Africa 's historical and current socio-economic contexts provide many challenges for both the Department of Education and schools in this endeavour to provide quality teaching and learning. These challenges impact directly and indirectly on what happens in the classroom. Since 1994 a plethora of education and training policy has been introduced in South Africa to redress historical imbalances; to introduce a new education and training framework and approach; and to provide guidelines, principles and procedures for addressing some of the challenges that impact on schools. The National Whole School Evaluation Policy provides the legislative framework for the establishment of a quality assurance process in South African schools based on accountability and support. The subsequent Integrated Quality Management System attempts to provide a framework for integrating school evaluation and performance measurement. Policy frameworks are in place to guide quality assurance and school improvement, however, the reality of implementing this at a grass roots level is particularly challenging. The General Motors (GM) South Africa Foundation, a non-governmental development organisation, established by General Motors (GM) South Africa, commenced with the piloting the Learning Schools Initiative to investigate some of the challenges of whole school development and evaluation. This research documents the Learning Schools Initiative's intervention with the initial two pilot secondary schools situated in Port Elizabeth (Nelson Mandela Bay) over a four-year period. It reviews the relevant school reform and school development literature and adopts a critical realist evaluative research approach to investigate the key mechanisms that promote whole school development and change in this context. In keeping with this approach, the results of the research are analysed and discussed within a context-menchanism-outcome configuration that involves the identification of the key mechanisms that bring about desired outcome/s in a specific context. Seven key generative mechanisms are identified as critical at a school and classroom level (i) school culture, (ii) school structures, (iii) effective leadership and management, (iv) personal growth and meaning, (v) restoration of relationships, (vi) professional development of educators, and development of capacity to work together, and (vii) support and accountability. The need to structure school development interventions around the triggering of identified key mechanisms is also identified as an important overarching mechanism. Suggestions are made for further research required to facilitate a deeper understanding of how to bring about meaningful change that results in quality teaching and learning in South African schools.
