- Description: This study contributes to the literature on quality improvement in higher education by examining the structural, cultural and agential constraints and enablements on a quality process at a university in South Africa. It examined four cases and developed an understanding of the complex interaction of structure, culture and agency and the mechanisms that enable or constrain quality improvement in higher education. The study drew on the literature on higher education quality for the theoretical basis for what is known contributes to the way in which quality assurance and improvement is implemented and its impact on the higher education context. Critical Realism provided the ontological framework and conceptual tools to understand and explore the complex social world within which the quality process took place. The literature on the morphogenetic approach provided the analytical framework for the data analysis and findings. The data consisted of a set of documents from a quality process that took place over a five-year period. The data analysis revealed that different departmental contexts impact on how mechanisms are activated. Each school context shapes the way in which people engage with the review process and consequently, processes and procedures are mediated in each context. This research therefore adds to the understanding of the way in which quality processes take place at a micro-level within an institutional context and informs the approach to quality improvement more broadly, nationally and internationally. - Description: Urban gardening in townships is a common coping strategy employed by poor inhabitants to improve food security, earn income through sales, and generate cost-savings through home consumption of fresh produce. Food security is a cornerstone of human development and welfare, and the achievement thereof remains a global challenge. The majority of academic research and policy documentation traditionally positions food insecurity as a rural phenomenon. However, recent trends point to an increase in food insecurity in urban areas, particularly in low-income areas such as townships. In South Africa, rapid urbanization has led to burgeoning townships and rapidly expanding low-income informal settlements characterised by poverty, unemployment, and food insecurity. Research concerned with the role that urban agricultural production plays in contributing to the food security of poor households is scarce and not well understood. This research, therefore, aimed to contribute to the limited research which specifically examines urban food security, and the role urban agriculture play in the achievement of household food security. The study employed collective action theory and utility theory to firstly provides an in-depth examination of the determinants of participation in urban gardening using Probit regression modeling. The second research objective was to perform an impact evaluation of participation in urban gardening on selected household food security indicators including Household Dietary Diversity Scores (HDDS), the Household Hunger Scale (HHS) and the Coping Strategy Index (CSI) using Propensity Score Matching (PSM). Purposive and snowball sampling methods were employed to obtain a sample of 60 urban gardeners and a control group of 55 non-gardening households from Extension 6, 7 and 10 townships as well as Joza Location in Makhanda East, Eastern Cape Province. Households were surveyed using a structured household food security questionnaire. A focus group discussion was held with the Linomtha Community Garden members and key informant interviews were undertaken with important stakeholders such as the local extension officers. Descriptive analysis revealed that urban gardening households (UGs) had older household heads who were less educated, and that land and water availability were the dominant constraints to increases in agricultural production. Results from the probit and average marginal effects models showed that the likelihood of participation significantly increased when household unemployment and dependency increased and that households who were engaged in off-farm economic activity were more likely to participate in urban gardening. The impact evaluation was performed using PSM and Average Treatment Effect on the Treated (ATT). Analysis illustrated that participation in urban gardening does significantly reduce both the full CSI and Reduced CSI. Thus, participation in urban gardening improves the food security of participating households by significantly reducing the frequency of participating households (UGs) applying undesirable coping strategies such as sending children elsewhere other than the household, borrowing food from other households and reducing the number of meals eaten in a day due to food shortages. This study posits that urban gardening alone will not eradicate the rampant food insecurity and poverty which pervades in South Africa’s townships. However, urban gardening participation does contribute to the construction of a sustainable, urban livelihood by reducing the number and severity of the undesirable coping strategies that food insecure households employ by providing some nutritious produce as well improved social capital through gardening networks and supporting institutions. It is recommended that policy makers, at a national and municipal level, need to create a clearer directive for the integration of urban food production into the urban food marketplace thus increasing access to income generating channels for small-scale home, and community gardeners. Co-operation between private and public institutions for food security and urban gardeners needs to be fostered and awareness of opportunities to participate in urban gardens needs to be improved. - Description: Cancer is a devastating disease that is a leading cause of death worldwide. Despite the available cancer treatments, there is a significant need to improve the therapeutic approach towards this disease. Photodynamic therapy (PDT) is an alternative approach for treating cancer, which requires a photosensitiser, molecular oxygen and light. Although some porphyrin-based derivatives have been approved by the United States Food and Drug Administration (FDA) and other similar agencies elsewhere for photodynamic therapy, their relatively poor photophysicochemical properties mean that there is an ongoing need for new photosensitiser dyes. Singlet oxygen photosensitiser dyes can also be used to treat bacteria that develop antimicrobial resistance in the context of photodynamic antimicrobial chemotherapy (PACT). The main aim of this study was to synthesise and characterise a series of porphyrin dyes with 4-quinolinyl, thien-2-yl and 4-bromo-thien-2-yl meso-aryl groups and their Sn(IV) and In(III) complexes, as well as their corrole analogues. Corroles are contracted macrocycles that have interesting optical properties. The corroles selected for study were found to be difficult to synthesise and purify and had unfavourable photophysicochemical properties and were thus omitted from the PDT and PACT biological applications within this thesis. High- and low-symmetry A4 and ABAB type meso-tetraarylporphyrins porphyrins were synthesised to improve the photophysicochemical properties of the photosensitisers; the utility of these dyes as photosensitisers was studied against the MCF-7 breast cancer cell line for PDT and against Staphylococcus aureus and Escherichia coli for PACT. The thienyl-2-yl rings were introduced to red shift the lowest energy Q band towards the phototherapeutic window, while quaternisation of the nitrogen and sulfur atoms of the 4-quinolinyl and thien-2-yl rings to introduce a cationic nature was explored to improve the bioavailability of the drugs and uptake into the target cell walls for improved efficacy. Heavy Sn(IV) and In(III) central metal ions were introduced to enhance the singlet oxygen quantum yields and limit aggregation through axial ligation. The bromine atoms of the 4-bromo-thien-2-yl meso-aryl rings were also introduced to enhance the singlet oxygen quantum yields of the dyes. Furthermore, the utility of the porphyrin and corrole molecules for optical limiting properties to limit laser radiation to protect optical devices, including eyes, was explored by the z-scan technique. - Description: My thesis is a collection of poems that is written in isiXhosa and English. It explores African spirituality, my relationship with God, heartache from romantic and platonic relationships. I make use of dual languages because some themes that I write about such as African spirituality I find words that capture their truest emotion in my Xhosa vocabulary, and I cannot find them in English. My poems use the narrative form because it allows me to tell stories through poetry without having to commit my writing to musicality or rhyming that a lyric poetry normally has. My work is shaped by writers such as Kate Beinhemer, Mangaliso Buzani, Amy Saul Zerby, Nontsizi Mgqwethio, Simphiwe Nolutshungu and Oiu Miaojin. Buzani makes use of images and few lines in his writing but still manages to capture a story with brevity. Saul-Zerby makes use of text lingo in some of her poems, and I make use of it to close a gap that I have identified with the books that I was reading that are all written in a formal and serious format. Mgqwetho’s poems explore Christianity and African Spirituality which is one of the subjects that my work is exploring too but in a manner that differs. I talk about how they have been my guidance instead of praising their powers like Nontsizi normally does in her poems. Simphiwe Nolutshungu’s poems has influenced the structure of my poems. Fairy tales written by writers like Kate helps my writing to bring to life issues that sound too dreamy to be true but have manifested as visions and memories that I cannot wipe out from my conscience. Microbial and extracellular polymeric substance dynamics in arid–zone temporary pan ecosystems

- Description: Microbial communities of bacteria, viruses, algae, protozoans and fungi participate profoundly in aquatic systems, particularly in mediating processes such as primary production, decomposition, and biogeochemical cycles. In addition, microbiomes produce extracellular polymeric substances (EPS) which encompass a hydrated exopolymer mainly constituted of carbohydrates and proteins. The exopolymer aid proliferation and persistence of biofilms on their resident surfaces. There is however paucity of data on functional diversity of microbiomes in arid zone temporary wetlands with previous research having mainly focused on permanent systems in the northern hemisphere. In the face of ongoing climatic changes and anthropogenic threats to wetlands, it is imperative to assess the health status of aquatic systems in relation to microbial productivity dynamics. In this thesis, colorimetric methods and sequence–based metagenomics were conducted to quantify microbial EPS production and bacterial metagenome functions, respectively. This study was conducted in Khakhea–Bray region (North–West, South Africa) in June 2021 and January 2022 with a focus on evaluating microbial patterns of distribution between seasons (i.e., Dry and Wet) and varying depth i.e., deepest zones (Deep), intermediate depth (Mid) and shallowest regions (Edge). Additionally, potential relationships between EPS and either water content or organic matter content (OM content) were evaluated. In this study it was hypothesized that wet phases and deeper zones will have high EPS production and support more functions in comparison to shallowest regions and dry phases. Carbohydrates and proteins were quantified using the Dubois method and modified Lowry procedure, respectively. Carbohydrates generally occurred in higher proportions than proteins, suggesting that EPS found in these systems was largely diatom produced. The wet phases (wet season and inundation periods) supported more EPS production compared to the dry phases. The results of principal components analysis (PCA) and Spearman’s correlations suggested that EPS was highly correlated with sediment water content among other assessed variables. No significant associations were established between EPS and organic matter content. Spatial distribution of EPS demonstrated similar patterns between the deepest (Deep) and the intermediate depth zones (Mid) however the shallow regions (Edge) had significantly lower concentrations. Bacterial characterization was established by amplification of the 16S rRNA gene using illumina–sequencing protocol. Enzyme functions associated with biogeochemical pathways were predicted in PICRUSt2 bioinformatics pipeline. A total of 15 042 Unique Amplicon Sequence Variants (ASVs) were observed to be affiliated to 51 bacterial phyla and 1 127 genera. All top genera had commonality in heat tolerance. Firmicutes, dominated at phyla level with 59 % (mean ± sd, 19 ± 13 %) relative abundance followed by Actinobacteria and Proteobacteria both at 34 % (18 ± 7 %) and (18 ± 6 %), respectively. Microbial diversity matrices highlighted significant differences in beta diversity more than alpha diversity. Bacterial microbiomes were more distinct between seasons compared to within season, suggesting that functions were seasonally driven. These findings were supported by highest rates of denitrification, carbohydrate degradation and EPS production by core microbiomes in the wet season as compared to low rates of nitrogen mineralisation, carbon fixation and nitrification in the dry season. The present findings represent a first attempt in evaluating sequence–based metagenomics in semi–arid southern African temporary pan ecosystem. Both microbial EPS and bacterial functional potential were highly driven by water availability, with highest rates mainly associated with maximum inundation compared to dry states of pans. It can therefore be suggested that extended dry periods are threatening to microbially mediated processes in temporary wetlands, with implications to loss of biodiversity due to desiccation resulting in poor nutrient cycling. , Thesis (MSc) -- Faculty of Science, Zoology and Entomology, 2023

In addition, microbiomes produce extracellular polymeric substances (EPS) which encompass a hydrated exopolymer mainly constituted of carbohydrates and proteins. The exopolymer aid proliferation and persistence of biofilms on their resident surfaces. There is however paucity of data on functional diversity of microbiomes in arid zone temporary wetlands with previous research having mainly focused on permanent systems in the northern hemisphere. In the face of ongoing climatic changes and anthropogenic threats to wetlands, it is imperative to assess the health status of aquatic systems in relation to microbial productivity dynamics. In this thesis, colorimetric methods and sequence–based metagenomics were conducted to quantify microbial EPS production and bacterial metagenome functions, respectively. This study was conducted in Khakhea–Bray region (North–West, South Africa) in June 2021 and January 2022 with a focus on evaluating microbial patterns of distribution between seasons (i.e., Dry and Wet) and varying depth i.e., deepest zones (Deep), intermediate depth (Mid) and shallowest regions (Edge). Additionally, potential relationships between EPS and either water content or organic matter content (OM content) were evaluated. In this study it was hypothesized that wet phases and deeper zones will have high EPS production and support more functions in comparison to shallowest regions and dry phases. Carbohydrates and proteins were quantified using the Dubois method and modified Lowry procedure, respectively. Carbohydrates generally occurred in higher proportions than proteins, suggesting that EPS found in these systems was largely diatom produced. The wet phases (wet season and inundation periods) supported more EPS production compared to the dry phases. The results of principal components analysis (PCA) and Spearman’s correlations suggested that EPS was highly correlated with sediment water content among other assessed variables. No significant associations were established between EPS and organic matter content. Spatial distribution of EPS demonstrated similar patterns between the deepest (Deep) and the intermediate depth zones (Mid) however the shallow regions (Edge) had significantly lower concentrations. Bacterial characterization was established by amplification of the 16S rRNA gene using illumina–sequencing protocol. Enzyme functions associated with biogeochemical pathways were predicted in PICRUSt2 bioinformatics pipeline. A total of 15 042 Unique Amplicon Sequence Variants (ASVs) were observed to be affiliated to 51 bacterial phyla and 1 127 genera. All top genera had commonality in heat tolerance. Firmicutes, dominated at phyla level with 59 % (mean ± sd, 19 ± 13 %) relative abundance followed by Actinobacteria and Proteobacteria both at 34 % (18 ± 7 %) and (18 ± 6 %), respectively. Microbial diversity matrices highlighted significant differences in beta diversity more than alpha diversity. Bacterial microbiomes were more distinct between seasons compared to within season, suggesting that functions were seasonally driven. These findings were supported by highest rates of denitrification, carbohydrate degradation and EPS production by core microbiomes in the wet season as compared to low rates of nitrogen mineralisation, carbon fixation and nitrification in the dry season. The present findings represent a first attempt in evaluating sequence–based metagenomics in semi–arid southern African temporary pan ecosystem. Both microbial EPS and bacterial functional potential were highly driven by water availability, with highest rates mainly associated with maximum inundation compared to dry states of pans. Mayer, Davis and Schoorman (1995, p.709) define trust as “the willingness of a party to be vulnerable to the actions of another party based on the expectation that the other party will perform a particular action important to the trustor irrespective of the ability to monitor or control that other party”. This study took the form of an explanatory case study that focussed on an interaction between a firm and a salient stakeholder and attempted to explain how this interaction influenced the perceived trustworthiness of the firm in stakeholders that were observing the interaction. The study attempted to add to the work of Crane (2020), whose research had focussed on stakeholder connectedness, and specifically how this could be utilized by firms interacting with stakeholders to engender trust in a wider group of stakeholders. The research aimed to evaluate how the Sterile Insect Release programme, operating by the firm in the Sunday’s River Valley, was given a second chance through engagement with a salient stakeholder and how the interaction was observed by a wider group of stakeholders leading to perceived stakeholder trustworthiness and social capital. The study had five objectives, namely to; (1) analyse how a firm’s ability, benevolence, and integrity enables (or does not engender) the development of the firm’s trustworthiness amongst its stakeholders; (2) analyse how the firm’s engagement with a salient stakeholder enables (or does not engender) increased levels of trustworthiness in the firm by the stakeholders that are observing its interaction with a salient stakeholder; (3) analyse the benefits of social capital gained through increased levels of stakeholder trust; (4) identify good practice guidelines for firm engagement with salient stakeholders to ensure the correct cues are portrayed to observing stakeholders for the development of the trustworthiness of the firm; and (5) make recommendations related to management practices that will support the development of trust. The research adopted a qualitative approach and data were collected through semi-structured interviews with three groups of stakeholders that were present at the meeting. A review of the literature was undertaken to identify various propositions for the research and from which a theory-driven coding memo was developed. The derived propositions were as follows: (1) observing a firm’s engagement with a single salient stakeholder will provide cues to observing stakeholders about the trustworthiness of the firm, and (2) perceptions of a firm’s trustworthiness will result in increased social capital between the firm and its stakeholders. The first proposition was divided into three sub-propositions for the study which stated; (a) observing a firm’s engagement with a single salient stakeholder will provide cues to observing stakeholders about the ability of the firm; (b) observing a firm’s engagement with a single salient stakeholder will provide cues to observing stakeholders about the benevolence of the firm; and (c) observing a firm’s engagement with a single salient stakeholder will provide cues to observing stakeholders about the integrity of the firm. From the literature analysis, various themes were identified which include, observed interactions, ability, benevolence, integrity, trustworthiness, and increased social capital. The research findings confirmed the propositions as being relevant to the case study. The findings confirmed that ability, benevolence, and integrity are key attributes in portraying trustworthiness when interacting with a salient stakeholder. All three attributes were noted by the interviewees as being important to observing stakeholders in the development of trust in a firm, although ability and integrity were found to be the dominant attributes in this case study. The findings did present some evidence that an increased level of perceived trustworthiness in the firm did positively influence the levels of social capital, however, this was not a common theme among all the interviewees. In the light of these findings, it is recommended that firms utilize stakeholder models to identify salient stakeholders that are influential within the markets in which they operate. The firm should actively pursue a strong relationship with these stakeholders, and further attempt to generate opportunities to interact with these stakeholders in a public forum where stakeholder connectedness could be utilized to develop perceived trust in the wider group of stakeholders. The firm should focus on open, honest, and transparent communication which are key aspects for portraying the attributes of ability, benevolence, and integrity. - Description: Tourism is not only the world’s largest traded commodity in which many countries have gained economic advantage and stability, it is also a phenomenon of great economic and social significance. Tourism’s contribution, especially within developing countries and within emerging markets like South Africa, has become increasingly significant as it has created major economic growth and has highlighted areas of potential new sustainable development. The ultimate goal of achieving an increase in GDP and the creation of new employment opportunities is enhanced through the development of tourism. Considering the importance of the above and because the impact of tourism development in small towns is of significant economic benefit, research into this field is of value. This is the core of this study. As a result of tourism demand continually changing in small towns, mostly due to globalisation, demographic changes and agricultural changes, the need to continually update tourism data to ensure that tourism products remain highly rated is vital. This requires identifying tourists’ needs and supplying exactly what tourists demand. More specifically, evaluating tourist satisfaction and dissatisfaction will give accurate indications of a destination’s ability to satisfy its tourists, which is what will encourage repeat visits. The first objective of this dissertation was to qualitatively evaluate, through determining 27 product owners’ and 206 tourists’, opinions/perceptions of the tourism product offering in the Karoo. This study examined the accommodation sector of the tourism industry within 13 small towns in the Karoo. This is a fast growing and maturing industry. The study revealed that product owners and tourists expressed a positive attitude towards the growth of tourism in the Karoo, which they said was due to its heritage and authenticity, climate, desert tourism, natural beauty, ecotourism, remoteness, peace and quiet and acting as a refuge in an unspoilt natural environment. Amongst other recommendations, this result highlighted the importance of keeping closed the ‘gap’ between the expected product/level of service and the customer’s perception of the actual product/service received, by paying special attention to each of the identified tourist benefits. Most importantly, product owners, local tourism organisations (LTOs), municipalities, the government and all tourism stakeholders need to ensure that they work efficiently and effectively through transparent and collaborative relationships to ensure that they generate the highest possible returns from tourism investment through strategies that take cognizance of the above. The second objective of this dissertation was to quantitatively evaluate the economic impact of tourism through gathering and calculating 107 tourists’ spending data and 15 product owners’ expenditure data over a six month period between December 2010 and May 2011 in Graaff-Reinet. It was found that there was an increase over time in tourist spending, which led to an increase in direct income (net injection of visitor spending) and thus there was an increase in indirect income(affecting the level of product owners’ income)in the Graaff-Reinet economy. The key calculations in this study reflected a high multiplier of 1.2and a small amount of leakage (21%);and the final tourism economic impact of the accommodation sector in Graaff-Reinet wasR8940 982.00. This implies a positive economic contribution to this town. A focus on growing the tourist market is therefore a goal worth pursuing. This study provides up-to-date, statistically valid numerical/quantitative data as well as current qualitative data. However, gathering data for a longitudinal study would be highly beneficial to determine trends of how the tourism market in small towns is developing. This field has been given minimal attention, specifically within small towns. This dissertation was thus an attempt to initiate and motivate further ongoing efforts with regards to gathering, specifically economic, tourism data. Showing the positive economic contribution, and the multiple ripple effects that attracting tourists to an area can have, and the ultimate measurable economic benefit created through having a strong multiplier effect, will hopefully encourage all stakeholders to be confident and take the tourism industry more seriously. This highlights the need to recognise the economic impact potential of the tourism sector and to use the positive results identified in this study as a tool to lobby for tourism investment, which is unfortunately not reflected currently in many governmental programmes. Learning to make a difference: Small-scale women farmers in social learning spaces for climate action

- Description: How do women farmers in Africa learn about climate change? What is quality climate change learning for farmers? How do farmers interface new knowledge with their long-held and trusted traditional knowledge? How do we evaluate learning at farm level and beyond? Using Okoli’s theory mining review, I untangled a tripartite knot of social learning literature to find Social Learning Theory (SLT) suitable for a study to explore my practical and scholarly curiosity as reflected in the above questions. Wenger’s theory of Social Learning emerged as the most appropriate for my research. The second phase of my study explored the climate change learning and practice terrain for small-scale women farmers, analysing the connection between learning, practice, and the resultant value in two case study areas, municipalities in the Amathole District of the Eastern Cape Province of South Africa. In addition to a paper on SLT mining review that unravels and chooses between the strands of social learning, the two case studies resulted in three articles that responded to the study’s objectives and the research questions. The thesis is introduced and synthesised through five 'book-end' chapters, as well as through these four articles. What were my findings? In the first case study, in the drought-stricken Raymond Mhlaba Municipality, I gathered the data through individual semi-structured interviews with farmers, extension officers and representatives of the involved organisations. I also conducted a group interview with farmers and analysed documents to supplement interview data. I analysed the data using concepts of Communities of Practice (CoP) and SLT to map out the learning and practice landscape. I discovered a constellation of CoPs interconnected by the shared drive for adaptive water management. The constellation is made up of tertiary institutions, government departments, non-governmental organisations and farmers of varying experiences and competencies, with women emerging as the more proactive gender, and state-led extension services being willing but overstretched and under-resourced. SLT effectively traced the apparent fragmented learning within and outside the CoPs and the sudden and extensive shifts in the CoP boundaries, especially in the context of COVID-19 pandemic lockdowns and the increased adoption of digital learning platforms. Despite the richness and diversity brought by the emergent new learning networks that involve participants in the province and further afield, the adoption of digital learning platforms worsened the existing generational digital divide among farmers. iii In the second case study, in the water scarce Buffalo City Metropolitan Municipality, I adopted the Value Creation Framework (VCF) to conduct an ethnographic evaluation that used semi-structured interviews, participant observation and document analysis of the learning experiences of women farmers in a social movement on agroecology. I found that the farmer-centred learning approach of the movement has created value for the farmers involved, evidenced by the adoption of agroecology by over 2700 members (including new urban farmers who are occupying open spaces typically used as dumpsites). The learning approach has facilitated expansive learning, enhanced resource mobilisation, new collaborations, partnerships, and seed sharing networks. Additionally, it necessitated context-appropriate and transformative changes to intersectional justice issues associated with historical inequalities in access to land and water and gender discrimination, leading to improved practices, new access to markets and improved quality yields. These are examples of immediate, potential, applied, realised, orienting, enabling and transformative as well as strategic value, as defined by the VCF. In reflecting on how women farmers learn in these social learning spaces I elucidate the learning impact pathways and local contextual influences in shifting CoP boundaries, domains, and practices during the climate crisis as it intersects with other compounding factors. I generated insights that could be useful for stakeholders in the agricultural (extension) sector to build better pathways for emancipatory and empowering expansive social learning in contexts characterised by resource constraints, but also by strong women-led agency. Such learning could make a difference and cushion small-scale farming from collapse especially in times of unprecedented changes. - Description: How do women farmers in Africa learn about climate change? What is quality climate change learning for farmers? How do farmers interface new knowledge with their long-held and trusted traditional knowledge? How do we evaluate learning at farm level and beyond? Using Okoli’s theory mining review, I untangled a tripartite knot of social learning literature to find Social Learning Theory (SLT) suitable for a study to explore my practical and scholarly curiosity as reflected in the above questions. Wenger’s theory of Social Learning emerged as the most appropriate for my research. The second phase of my study explored the climate change learning and practice terrain for small-scale women farmers, analysing the connection between learning, practice, and the resultant value in two case study areas, municipalities in the Amathole District of the Eastern Cape Province of South Africa. In addition to a paper on SLT mining review that unravels and chooses between the strands of social learning, the two case studies resulted in three articles that responded to the study’s objectives and the research questions. The thesis is introduced and synthesised through five 'book-end' chapters, as well as through these four articles. What were my findings? In the first case study, in the drought-stricken Raymond Mhlaba Municipality, I gathered the data through individual semi-structured interviews with farmers, extension officers and representatives of the involved organisations. I also conducted a group interview with farmers and analysed documents to supplement interview data. I analysed the data using concepts of Communities of Practice (CoP) and SLT to map out the learning and practice landscape. I discovered a constellation of CoPs interconnected by the shared drive for adaptive water management. - Description: Malaria continues to be a burden globally with a myriad of challenges deterring eradication efforts. With most antimalarials facing drug resistance, such as atovaquone (ATQ), alternative compounds that can withstand resistance are warranted. The Plasmodium falciparum cytochrome b (PfCytb), a subunit of P. falciparum cytochrome bc1 complex, is a validated drug target. Structurally, cytochrome b, cytochrome c1, and iron sulphur protein (ISP) subunits form the catalytic domain of the protein complex having heme bL, heme bH and iron-sulphur [2FE-2S] cluster cofactors. These cofactos have redox centres to aid in the electron transfer (ET) process. These subunits promote ET mainly through the enzyme’s ubiquinol oxidation (Qo) and ubiquinone reduction (Qi) processes in the catalytic domain. ATQ drug has been used in the prevention and treatment of uncomplicated malaria by targeting PfCytb protein. Once the mitochondrial transmembrane ET pathway is inhibited, it causes a collapse in its membrane potential. Previously reported ATQ drug resistance has been associated with the point mutations Y268C, Y268N and Y268S. Thus, in finding alternatives to the ATQ drug, this research aimed to: i) employ in silico approaches incorporating protein into phospholipid bilayer for the first time to understand the parasites’ resistance mechanism; ii) determine any sequence and structural differences that could be explored in drug design studies; and iii) screen for PfCytb-iron sulphur protein (Cytb-ISP) hit compounds from South African natural compound database (SANCDB) and Medicines for Malaria Venture (MMV) that can withstand the identified mutations. Using computational tools, comparative sequence and structural analyses were performed on the cytochrome b protein, where the ultimate focus was on P. falciparum cytochrome b and its human homolog. Through multiple sequence alignment, motif discovery and phylogeny, differences between P. falciparum and H. sapiens cytochrome b were identified. Protein modelling of both P. falciparum and H. sapiens cytochrome b - iron sulphur protein (PfCytb-ISP and HsCytb-ISP) was performed. Results showed that at the sequence level, there were few amino acid residue differences because the protein is highly conserved. Important to note is the four-residue deletion in Plasmodium spp. absent in the human homolog. Motif analysis discovered five unique motifs in P. falciparum cytochrome b protein which were mapped onto the predicted protein model. These motifs were not in regions of functional importance; hence their function is still unknown. At a structural level, the four-residue deletion was observed to alter the Qo substrate binding pocket as reported in previous studies and confirmed in this study. This deletion resulted in a 0.83 Å structural displacement. Also, there are currently no in silico studies that have performed experiments with P. falciparum cytochrome b protein incorporated into a phospholipid bilayer. Using 350 ns molecular dynamics (MD) simulations of the holo and ATQ-bound systems, the study highlighted the resistance mechanism of the parasite protein where the loss of active site residue-residue interactions was identified, all linked to the three mutations. The identified compromised interactions are likely to destabilise the protein’s function, specifically in the Qo substrate binding site. This showed the possible effect of mutations on ATQ drug activity, where all three mutations were reported to share a similar resistance mechanism. Thereafter, this research work utilised in silico approaches where both Qo active site and interface pocket were targeted by screening the South African natural compounds database (SANCDB) and Medicines for Malaria Venture (MMV) compounds to identify novel selective hits. SANCDB compounds are known for their structural complexity that preserves the potency of the drug molecule. Both SANCDB and MMV compounds have not been explored as inhibitors against the PfCytb drug target. Molecular docking, molecular dynamics (MD) simulations, principal component, and dynamic residue network (DRN; global and local) analyses were utilised to identify and confirm the potential selective inhibitors. Docking results identified compounds that bound selectively onto PfCytb-ISP with a binding energy ≤ -8.7 kcal/mol-1. Further, this work validated a total of eight potential selective compounds to inhibit PfCytb-ISP protein (Qo active site) not only in the wild-type but also in the presence of the point mutations Y268C, Y268N and Y268S. The selective binding of these hit compounds could be linked to the differences reported at sequence/residue level in chapter 3. DRN and residue contact map analyses of the eight compounds in holo and ligand-bound systems revealed reduced residue interactions and decreased protein communication. This suggests that the eight compounds show the possibility of inhibiting the parasite and disrupting important residue-residue interactions. Additionally, 13 selective compounds were identified to bind at the protein’s heterodimer interface, where global and local analysis confirmed their effect on active site residues (distal location) as well as on the communication network. Based on the sequence differences between PfCytb and the human homolog, these findings suggest these selective compounds as potential allosteric modulators of the parasite enzyme, which may serve as possible replacements of the already resistant ATQ drug. - Description: This study explored the development of reflective practice in foundation phase pre-service teachers in the context of video-based lesson analysis at a university in South Africa. The study was conducted in the field of mathematics education, responding to the urgent need to equip pre-service South African teachers with the knowledge and skills for effective mathematics teaching. The research is foregrounded by the continuing poor performance of South African learners in mathematics at all levels of education in the country, which has been linked to the inadequate knowledge and skills of mathematics teachers. Pre-service teacher education is putting considerable effort into improving the preparation of mathematics teachers and developing their ability to reflect on their teaching practice is one of the strategies being employed for this purpose. Research has demonstrated the importance of reflective practice (RP) in both developing and extending teachers’ mathematical knowledge for teaching. This study therefore contributes to current research that supports the development of RP as a professional skill for promoting the acquisition of knowledge for teaching in pre-service teacher education. The study adopted a qualitative case study approach with two phases of data collection. In Phase 1 I collected and analysed three sets of 19 pre-service teachers’ written reflections to establish the nature of the reflections that they developed when analysing video-recorded mathematics lessons of experienced teachers’ practice. Phase 2 was conducted with four PSTs who reflected on video-recorded mathematics lessons of their own practice, and similarly sought to investigate the nature of the reflections they developed when reflecting on practice. The four PSTs wrote one set of reflections on their own lessons, went through three sessions of facilitator-guided reflections, then wrote another set of reflections to establish if the support provided in small group facilitator-guided sessions improved their reflections. Iterative content analysis was employed to analyse the PSTs’ written reflections, using an analytic tool that I developed for this purpose through merging Lee’s (2007) and Muir and Beswick’s (2007) levels of reflection frameworks. My model had four levels of reflection: description, explanation, suggestion and reflectivity. The names of each of the levels connect to the key indicator for that level. PSTs’ written reflections were coded and analysed according to these levels. The study found that PSTs’ initial reflections were mostly description of general classroom events with little reflection at the levels of explanation and suggestion, and an absence of reflectivity. Most reflections focused on general events in the lesson rather than mathematical events, even though the six lens framework they were given to guide their reflections prompted them to steer their attention towards mathematical events. The second and third sets of reflections, although mostly still at level 1, showed some shifts towards explanation and suggestion, although an increased focus on mathematical events though reflectivity was still largely absent. No PST reached the fourth level of reflectivity in Phase 1. However, in Phase 2, the PSTs’ reflections after the three small group facilitator-guided sessions included some evidence of reflectivity. The findings suggest the need for pre-service teacher educators to make a concerted effort to teach PSTs what reflection is and how to reflect on their practice. Phase 2 was conducted with four PSTs who reflected on video-recorded mathematics lessons of their own practice, and similarly sought to investigate the nature of the reflections they developed when reflecting on practice. The four PSTs wrote one set of reflections on their own lessons, went through three sessions of facilitator-guided reflections, then wrote another set of reflections to establish if the support provided in small group facilitator-guided sessions improved their reflections. Iterative content analysis was employed to analyse the PSTs’ written reflections, using an analytic tool that I developed for this purpose through merging Lee’s (2007) and Muir and Beswick’s (2007) levels of reflection frameworks. My model had four levels of reflection: description, explanation, suggestion and reflectivity. The names of each of the levels connect to the key indicator for that level. PSTs’ written reflections were coded and analysed according to these levels. The study found that PSTs’ initial reflections were mostly description of general classroom events with little reflection at the levels of explanation and suggestion, and an absence of reflectivity. Most reflections focused on general events in the lesson rather than mathematical events, even though the six lens framework they were given to guide their reflections prompted them to steer their attention towards mathematical events. The second and third sets of reflections, although mostly still at level 1, showed some shifts towards explanation and suggestion, although an increased focus on mathematical events though reflectivity was still largely absent. No PST reached the fourth level of reflectivity in Phase 1. However, in Phase 2, the PSTs’ reflections after the three small group facilitator-guided sessions included some evidence of reflectivity. The findings suggest the need for pre-service teacher educators to make a concerted effort to teach PSTs what reflection is and how to reflect on their practice. Business leaders need to demonstrate their willingness to consider their influence on society and not merely consider the demands and expectations of shareholders. Leaders must consider the ethical lens through which their decisions and actions are perceived. Service as a leadership competency can develop ethical leadership. Currently, service is not clearly defined and understood, and Pearse (2017) proposes five interrelated elements of service as a competency, namely individualised consideration, compassion, motivation to lead, humility and integrated thinking. The study examined if the five interrelated service elements are present when a leader at Director/CEO level exercises an act of service towards a follower. The study adopted a qualitative deductive thematic approach, collecting data through semi-structured interviews and using the critical incident technique to guide the interviews. The study's findings support the research proposition that service as a leadership competency consists of the five elements, but with some of these elements expanded. Service as a leadership competency is recommended to develop high-quality social relationships within an organisation, which will positively impact the corporate culture. Organisations can recruit and select leaders based on ethical leadership requirements and integrate these into performance management systems. - Description: The use of network telescopes to collect unsolicited network traffic by monitoring unallocated address space has been in existence for over two decades. Past research has shown that there is a lot of activity happening in this unallocated space that needs monitoring as it carries threat intelligence data that has proven to be very useful in the security field. Prior to the emergence of the Internet of Things (IoT), commercialisation of IP addresses and widespread of mobile devices, there was a large pool of IPv4 addresses and thus reserving IPv4 addresses to be used for monitoring unsolicited activities going in the unallocated space was not a problem. Now, preservation of such IPv4 addresses just for monitoring is increasingly difficult as there is not enough free addresses in the IPv4 address space to be used for just monitoring. This is the case because such monitoring is seen as a ’non-productive’ use of the IP addresses. This research addresses the problem brought forth by this IPv4 address space exhaustion in relation to Internet Background Radiation (IBR) monitoring. In order to address the research questions, this research developed four mathematical models: Absolute Mean Accuracy Percentage Score (AMAPS), Symmetric Absolute Mean Accuracy Percentage Score (SAMAPS), Standardised Mean Absolute Error (SMAE), and Standardised Mean Absolute Scaled Error (SMASE). These models are used to evaluate the research objectives and quantify the variations that exist between different samples. The sample sizes represent different lens sizes of the telescopes. The study has brought to light a time series plot that shows the expected proportion of unique source IP addresses collected over time. The study also imputed data using the smaller /24 IPv4 net-block subnets to regenerate the missing data points using bootstrapping to create confidence intervals (CI). The findings from the simulated data supports the findings computed from the models. The CI offers a boost to decision making. Through a series of experiments with monthly and quarterly datasets, the study proposed a 95% - 99% confidence level to be used. It was known that large network telescopes collect more threat intelligence data than small-sized network telescopes, however, no study, to the best of our knowledge, has ever quantified such a knowledge gap. Supervised machine learning techniques (ML) used in a NIDS can be affected by different scenarios. Thus, dataset recency, size, and applicability are essential factors when selecting and tuning a machine learning classifier. This thesis explores developing and optimising several supervised ML algorithms with relatively new datasets constructed to depict real-world scenarios. The methodology includes empirical analyses of systematic ML-based NIDS for a near real-world network system to improve intrusion detection. The thesis is experimental heavy for model assessment. Data preparation methods are explored, followed by feature engineering techniques. The model evaluation process involves three experiments testing against a validation, un-trained, and retrained set. They compare several traditional machine learning and deep learning classifiers to identify the best NIDS model. Results show that the focus on feature scaling, feature selection methods and ML algo- rithm hyper-parameter tuning per model is an essential optimisation component. Distance based ML algorithm performed much better with quantile transformation whilst the tree based algorithms performed better without scaling. Permutation importance performs as a feature selection method compared to feature extraction using Principal Component Analysis (PCA) when applied against all ML algorithms explored. Meta-ethnography is a methodology used to synthesise qualitative material by means of seven distinct phases: getting started; deciding what is relevant to the initial interest; reading the studies; determining how they are related; translating the studies into one another; weaving them together into a synthesis; and expressing the synthesis. This study synthesises five articles discovered through a deep search of the literature. Through the process of synthesis, a narrative emerges that connects past prejudice in music education in South Africa to present day educational inequality, one that looks towards a future in which children’s agency is harnessed in the multicultural world we live in to teach music in a relevant, contextual way. The synthesis extracts perceptions on colonialism and apartheid; educational access; post-apartheid curricula; music education pedagogies; informal music making; children’s games and agency; into the classroom; and how to assess. These themes weave a clear perspective on African musical arts education, and a reciprocal synthesis of the views of the articles’ authors.
