In addition, older people often do not seek help throughout their lives for various reasons. While there is research available about older people and trauma, little is known about Eye Movement Integration(EMI)therapy as a treatment intervention in treating symptoms of trauma in older persons. The purpose of this research was an explorative and descriptive study of older people’s responses to EMI therapy as a treatment intervention for their symptoms of trauma. More specifically, the study was designed to determine how older people process their symptoms through EMI therapy and whether they view it as valuable. This qualitative study used an exploratory-descriptive research design with a non-probability, purposive sampling method. The EMI therapy sessions were offered to older adults in the Knysna area who presented with symptoms of trauma. From this group, only those who were able and willing to engage in therapeutic interviews formed part of the research. As part of the study, they consented to the use of their inventories from the EMI sessions as a qualitative document analysis and taking part in qualitative interviews to explore their views on the usefulness of EMI therapy to deal with their trauma. The findings emphasise that older people are not familiar with EMI. However, they view their experience of it in a positive light and regard it as a useful treatment intervention for treating their symptoms. Moreover, EMI seems to be a brief and cost-effective trauma intervention with practical benefits for older adults. The results from this study can assist social work professionals, specifically those working with older people, to address and improve the overall health and emotional wellbeing of older people. Data verification methods ensured the validity of this study

If South African macro and social policy explicitly aims to be ‘inclusive’, it is important to know how the average [median] South African income level changed over the first fifteen years of democracy. Common summary measures of economic progress such as GDP per capita may mask the standard of living experienced by the majority of the population in any particular period. Tracking the progress in the middle strata of South Africa’s income distribution provides a fresh perspective on the nature of economic progress in the country. This is against a worrying backdrop of rising income inequality post-1994 (Van der Berg and Louw, 2004; Hoogeveen and Özler, 2006; Leibbrandt et al, 2010). The paper is structured as follows. Section 2 motivates for the importance of focussing on the actual middle of the income distribution and highlights the emphasis on reducing income inequality within South African macroeconomic and social policy. Issues of comparability in the data and the choice of definition for the middle are discussed in section 3. Section 4 presents descriptive statistics relating to changes in incomes, and ends with a brief analysis of non-income measures of progress. The final section summarises the main findings and concludes.

- Description: Driven by the needs of growing populations, industrial and governing powers are successfully accelerating the rate of industrial consumption, production and employment as if the earth’s resources are in unlimited supply. In contrast, a range of international sustainable development forums, inspired by visionary individuals, have made significant progress in creating awareness that the footprint of human activity is exceeding the earth’s sink and source capacity; and educating people in government, workplaces and communities to slow down industrial consumption and clean up production. Turning around conventional and short sighted ‘business as usual’ logic, and directing economies toward greener, long-term sustainability outcomes, still meet with resistance and hidden unsustainable agendas. The ‘green economy’ drive nevertheless since 2008 attracts financial and human resources and bold action in favour of more sustainable management of human-nature relations. The sustainable development movement for example advocates a ‘triple bottom line’ approach, holding that socially and ecologically responsible economic development would be sustainable. The sustainability movement has attained significant buy-in among governments and business communities. It forms the under-labouring philosophy of the programmes reviewed in this case study. The thesis reviews social-economic events paving the way for a global green economy. Taking a leadership role in the sustainable development movement the United Nations (UN) and the Organisation for Economic Co-Operation and Development (OECD) concurred to respond to the 2008 world-wide economic meltdown with a two-pronged ‘Green New Deal’ (UNEP, 2008). The Green New Deal financial package helps restore multi-national economic growth, employment and markets while re-shaping economies to follow an ecologically and socially responsible growth trajectory. South Africa implements green economy principles as part of the 2010 The New Growth Path overarching policy framework, with an implementation strategy embedded in the 2011 National Development Plan (NDP) (RSA. The Presidency, 2010; 2011). The New Growth Path emphasises that the transformation of South Africa’s un-sustainable economic and educational legacy to a more sustainable future is not expected to follow a smooth, linear process. The transition to a green economy is rather expected to be an event of “… noisy, healthy democracy” (RSA. The Presidency, 2010). A green, low carbon economy particularly constitutes a pledge to slow down and turn the human induced climate change trajectory around. McKinsey (2009) argues that this pledge is attainable on a world-wide scale, as sufficient and suitable environmentally sound techniques and technologies are already in place. Attaining buy-in from business stakeholders toward re-thinking and amending an economy’s self-defying large environmental footprint (inclusive of carbon, water and waste footprints) however requires education starting with awarenessraising followed by educational programmes and official curricula aimed at implementation and continuous improvement of green practices in day to day ‘doings and beings’ (Sen, 1997). This study at implementation level reviews two green economy training programmes and their emergence in South Africa around this rationale. The awareness generation and training programmes elected as case study examples are the ‘Resource Efficient and Cleaner Production’ (RECP) and ‘Industrial Energy Efficiency’ (IEE) programmes, of the hosting agency National Cleaner Production Centre of South Africa (NCPC-SA). The RECP and IEE teams reach out to decision makers, engineers and artisans at industrial workplaces and workplace related events to add green competences to their business-as-usual skill sets. Implementing green options in industry typically slows down industrial scale resource consumption, pollution, waste generation and green-house gas (GHG) emissions while optimising resource productivity and enterprise excellence. Optimised supply side systems allow industry to reduce energy and material intensity of products thus reducing cost and producing more with less. In transitioning to a ‘Green GDP’ economy South Africa is awakening to the reality that natural resources constitute the original, albeit limited feedstock for growth and employment. The RECP and IEE approach also contribute to reduction of industrial waste, waste-to-landfill, and energy and resource security. Literature reviewed for this research provides evidence that the green economy’s triple bottom line philosophy is quantifiable thus manageable. A range of green economy management tools are emerging, including guidelines for carbon, water and environmental footprinting and the green-house gas abatement cost curve (see section 2.2.5) (McKinsey and Company, 2009). Transitioning from business as usual to ecologically sustainable industrial sectors however requires visionary, educated leadership, willing and capable of introducing modern and more efficient techniques and technologies. The boundaries of this half thesis embrace the globally and historically significant Tbilisi Declaration and other education and sustainable development agreements produced by United Nations and OECD mechanisms. Participating nations like South Africa incorporate the essence of these agreements into domestic policies and strategies, and align industries to remain competitive in international markets, which are increasingly enforcing green standards like ISO 14001 and ISO 50001. The focus of this case study guided by inductive, abductive and retroductive inference is to understand how the two sustainability learning programmes for people in industrial workplaces, supported by the United Nations Industrial Development Organisation (UNIDO) and relevant donors, and overseen by the South African Department of Trade and Industry (the dti), relate to emerging green learning aspects. Producing a review of this nature requires a framework of laterally understanding emerging green learning aspects, for which I have reviewed green economy literature and also green learning and conventional education and training literature respectively. Themes emerging from the literature review informed an analytical instrument (questionnaire) in Phase One. In Phase Two the questionnaire was applied through nested case study methodology to show how the educational content and approaches of the RECP and IEE programmes relate to emerging green learning aspects and as such is suitable for mainstreaming in the national educational system. From an explicit educational perspective potential partners for collaboration include the Department of Higher Education and Training (DHET) the South African Qualifications Authority’s (SAQA’s) Quality Council for Trades and Occupations (QCTO) and the Sector Education and Training Authorities (SETAs) representing the training needs of the industrial workforce but affiliated to QCTO and SAQA. In the extended scenario the NCPC-SA as a dti programme recognises the Department of Environmental Affairs (DEA) as lead agency guiding implementation of South Africa’s green economy, and specifically DEA’s National Environmental Sector Skills Planning Forum (NESPF), a national leader in green skills development in South Africa, as conduit for productively mainstreaming relevant RECP and IEE content and approaches toward green skills development for the green economy.

Taking a leadership role in the sustainable development movement the United Nations (UN) and the Organisation for Economic Co-Operation and Development (OECD) concurred to respond to the 2008 world-wide economic meltdown with a two-pronged ‘Green New Deal’ (UNEP, 2008). The Green New Deal financial package helps restore multi-national economic growth, employment and markets while re-shaping economies to follow an ecologically and socially responsible growth trajectory. South Africa implements green economy principles as part of the 2010 The New Growth Path overarching policy framework, with an implementation strategy embedded in the 2011 National Development Plan (NDP) (RSA. The Presidency, 2010; 2011). The New Growth Path emphasises that the transformation of South Africa’s un-sustainable economic and educational legacy to a more sustainable future is not expected to follow a smooth, linear process. The transition to a green economy is rather expected to be an event of “… noisy, healthy democracy” (RSA. The Presidency, 2010). A green, low carbon economy particularly constitutes a pledge to slow down and turn the human induced climate change trajectory around. McKinsey (2009) argues that this pledge is attainable on a world-wide scale, as sufficient and suitable environmentally sound techniques and technologies are already in place. Attaining buy-in from business stakeholders toward re-thinking and amending an economy’s self-defying large environmental footprint (inclusive of carbon, water and waste footprints) however requires education starting with awarenessraising followed by educational programmes and official curricula aimed at implementation and continuous improvement of green practices in day to day ‘doings and beings’ (Sen, 1997). This study at implementation level reviews two green economy training programmes and their emergence in South Africa around this rationale. The awareness generation and training programmes elected as case study examples are the ‘Resource Efficient and Cleaner Production’ (RECP) and ‘Industrial Energy Efficiency’ (IEE) programmes, of the hosting agency National Cleaner Production Centre of South Africa (NCPC-SA). The RECP and IEE teams reach out to decision makers, engineers and artisans at industrial workplaces and workplace related events to add green competences to their business-as-usual skill sets. Implementing green options in industry typically slows down industrial scale resource consumption, pollution, waste generation and green-house gas (GHG) emissions while optimising resource productivity and enterprise excellence. Optimised supply side systems allow industry to reduce energy and material intensity of products thus reducing cost and producing more with less. In transitioning to a ‘Green GDP’ economy South Africa is awakening to the reality that natural resources constitute the original, albeit limited feedstock for growth and employment. The RECP and IEE approach also contribute to reduction of industrial waste, waste-to-landfill, and energy and resource security. Literature reviewed for this research provides evidence that the green economy’s triple bottom line philosophy is quantifiable thus manageable. A range of green economy management tools are emerging, including guidelines for carbon, water and environmental footprinting and the green-house gas abatement cost curve (see section 2.2.5) (McKinsey and Company, 2009). Transitioning from business as usual to ecologically sustainable industrial sectors however requires visionary, educated leadership, willing and capable of introducing modern and more efficient techniques and technologies. The boundaries of this half thesis embrace the globally and historically significant Tbilisi Declaration and other education and sustainable development agreements produced by United Nations and OECD mechanisms. Participating nations like South Africa incorporate the essence of these agreements into domestic policies and strategies, and align industries to remain competitive in international markets, which are increasingly enforcing green standards like ISO 14001 and ISO 50001. The focus of this case study guided by inductive, abductive and retroductive inference is to understand how the two sustainability learning programmes for people in industrial workplaces, supported by the United Nations Industrial Development Organisation (UNIDO) and relevant donors, and overseen by the South African Department of Trade and Industry (the dti), relate to emerging green learning aspects. Producing a review of this nature requires a framework of laterally understanding emerging green learning aspects, for which I have reviewed green economy literature and also green learning and conventional education and training literature respectively. Themes emerging from the literature review informed an analytical instrument (questionnaire) in Phase One. In Phase Two the questionnaire was applied through nested case study methodology to show how the educational content and approaches of the RECP and IEE programmes relate to emerging green learning aspects and as such is suitable for mainstreaming in the national educational system. From an explicit educational perspective potential partners for collaboration include the Department of Higher Education and Training (DHET) the South African Qualifications Authority’s (SAQA’s) Quality Council for Trades and Occupations (QCTO) and the Sector Education and Training Authorities (SETAs) representing the training needs of the industrial workforce but affiliated to QCTO and SAQA. In the extended scenario the NCPC-SA as a dti programme recognises the Department of Environmental Affairs (DEA) as lead agency guiding implementation of South Africa’s green economy, and specifically DEA’s National Environmental Sector Skills Planning Forum (NESPF), a national leader in green skills development in South Africa, as conduit for productively mainstreaming relevant RECP and IEE content and approaches toward green skills development for the green economy.

Documents such as the National Curriculum, Science Curriculum, Syllabi, Grade 6 Natural Science textbooks and SATs results were analysed to gain some insight and understanding of the issue before conducting interviews. In the second phase of this study I conducted semi-structured interviews with the Topnaar community members and two teachers. This was followed by classroom observations. Additionally, I used stimulated recall interviews (SRI) to provide clarity on how teachers use prior knowledge and mediate learning when teaching the topic water conservation using indigenous knowledge. The study is informed by Vygotsky’s (1978) socio-cultural theory in conjunction with Shulman’s (1986) pedagogical content knowledge (PCK). Since a classroom is a social unit where interactions occur between teachers and learners, and learners and learners, these theories provided a useful lens for observing lessons and analysing teachers’ content knowledge and how they mediate learning using local indigenous knowledge. A thematic approach to data analysis was adopted and qualitative data were analysed inductively. The findings of the study revealed that the indigenous knowledge amongst the Topnaars are governed by the ideals of Ubuntu. Findings also revealed that proper planning is needed to incorporate IK into science lessons as it may clear up misconceptions that may arise from indigenous knowledge. The study recommends that teachers and curriculum planners should explore different indigenous practices that are linked to the topics in the curriculum or syllabus to enhance learning and teaching in their science classrooms. It also recommends that curriculum developers, with the help of subject advisors, teachers and community members, should start planning on how indigenous knowledge can best be integrated into science textbooks as it has become a hindrance for the teachers to incorporate it.

The study focuses on South African scholars attending school in rural areas where the contrast between learning in formal and informal learning contexts is more pronounced than that in urban areas. The research draws on rural scholars' local knowledge of formal and informal learning contexts in order to gain a rich insight into how cognition is situated in different learning contexts. This is accomplished through investigating the structure of the respective learning tasks, the mediators involved, the task objectives and the means for achieving these objectives in the different learning contexts. The thesis draws on a socio-cultural approach to the study of cognitive development to probe the activity of learning in a formal and informal learning context. Through the use of a context sensitive methodological methods especially Participatory Rural Appraisal (PRA) tools and techniques, it was possible to illuminate tacit local knowledge structures and to get participants to actively explicate their understandings related to learning in different contexts The research results illustrate the assertion that the activity of learning is fundamentally situated in the learning context from which it arises. Learning is framed by the community of practice which structures affordances for situated learning, through mediation, within zones of proximal development. Learning in a formal context such as the school is often abstract, rule-based, standardised and theory related. Learners also find it difficult to reflect on the learning tasks and the mediational means used in a formal learning context. In contrast, the learning which takes place in an informal setting is often practical, individualised, flexible and environment based. This learning is structured around everyday activities and is dynamically defined and supported. In a situation where a learner is exposed to dislocated learning contexts, the essential goal of educational initiatives is to bridge the gap between the two. This can be achieved through mediators creating effective zones of proximal development which facilitate the individuals adaptation between learning contexts. Exposing rural scholars' local knowledge of learning in formal and informal contexts allows for a fuller understanding of the cognitive development structured within formal and informal communities of practice. It is this understanding that is necessary to address the situation where learning contexts, drawing on different knowledge bases find ways of thinking, prove challenging and/or conflicting to the scholar.

Evaluation of model systems for the study of protein association / incorporation of Beta-Methylamino-L-Alanine (BMAA)

- Description: β-methylamino-L-alanine (BMAA) is thought to be a contributing factor of Amyotrophic Lateral Sclerosis-Parkinsonism Dementia Complex (ALS/PDC). It has been shown that the levels of toxin ingestion by humans are too low to cause disease. However, it has recently been theorized that this toxin is bioaccumulated within cells. Via a process of slow release from this reservoir, the BMAA is able to bring about neurotoxicity. Mechanisms of uptake and bioaccumulation of BMAA have been proposed in several publications; however the mechanism of protein incorporation of BMAA has not yet been identified. Identifying suitable model systems would be a prerequisite in order for future studies on BMAA protein incorporation. Three specific models were therefore chosen for investigation; mammalian cell lines including C2C12 and HT29, a prokaryotic (E. coli) expression system and yeast cells. The cytotoxity of BMAA was established for the mammalian cell lines and further investigation of BMAA incorporation into cellular proteins was performed on all three above mentioned models. Samples were run on HPLC-MS in order to determine uptake of BMAA into cells or lack thereof. Results indicate negligible cytotoxicity as measured by MTT and CellTitre Blue assays, limited uptake and protein incorporation of BMAA within the prokaryotic model and insignificant uptake of BMAA by yeast cells. Although the uptake of BMAA in the prokaryotic model was not extensive, there was indeed uptake. BMAA was not only taken up into the cells but was also observed in inclusion body protein samples after hydrolysis. After further investigation and use, this model could very well provide researchers with information pertaining to the mechanism of association of BMAA with proteins. Although the other models provided negative results, this research was valuable in the sense that one can narrow down the number of possible model systems available. Also, in seeking models for studying protein association/incorporation, the use of the final target cell is not relevant or necessary as the purpose of the research was to identify a model system in which the mechanism of protein association/incorporation can, in future, be studied.

It has been shown that the levels of toxin ingestion by humans are too low to cause disease. However, it has recently been theorized that this toxin is bioaccumulated within cells. Via a process of slow release from this reservoir, the BMAA is able to bring about neurotoxicity. Mechanisms of uptake and bioaccumulation of BMAA have been proposed in several publications; however the mechanism of protein incorporation of BMAA has not yet been identified. Identifying suitable model systems would be a prerequisite in order for future studies on BMAA protein incorporation. Three specific models were therefore chosen for investigation; mammalian cell lines including C2C12 and HT29, a prokaryotic (E. coli) expression system and yeast cells. The cytotoxity of BMAA was established for the mammalian cell lines and further investigation of BMAA incorporation into cellular proteins was performed on all three above mentioned models. Samples were run on HPLC-MS in order to determine uptake of BMAA into cells or lack thereof. Results indicate negligible cytotoxicity as measured by MTT and CellTitre Blue assays, limited uptake and protein incorporation of BMAA within the prokaryotic model and insignificant uptake of BMAA by yeast cells. Although the uptake of BMAA in the prokaryotic model was not extensive, there was indeed uptake. BMAA was not only taken up into the cells but was also observed in inclusion body protein samples after hydrolysis. After further investigation and use, this model could very well provide researchers with information pertaining to the mechanism of association of BMAA with proteins. Although the other models provided negative results, this research was valuable in the sense that one can narrow down the number of possible model systems available. Also, in seeking models for studying protein association/incorporation, the use of the final target cell is not relevant or necessary as the purpose of the research was to identify a model system in which the mechanism of protein association/incorporation can, in future, be studied.

Only 42% of the people who require anti-retrovirals (ARVs) in low to middle income countries are receiving anti-retroviral therapy (ART). There is a need to develop novel and cost effective methods for producing antiretroviral drugs. Stavudine and azidothymidine (AZT) were identified as potential targets because they could both be produced through a common intermediate – 5 methyluridine (5-MU). It has been established that the biocatalytic production of 5-methyluridine is possible through a reaction known as transglycosylation, in a process which has not previously been demonstrated as commercially viable. A selection of biocatalysts were expressed either in recombinant E. coli strains or in the wild type organisms, purified and then screened for their ability to produce 5-MU. A combination of Bacillus halodurans purine nucleoside phosphorylase 1 (BHPNP1) and E. coli uridine phosphorylase (EcUP) gave the highest 5-MU yield (80%). This result represents the first combination of free enzymes from different organisms, giving high yields of 5-MU under high substrate conditions. Both enzymes were purified and successfully characterised. The established pH optimum was pH 7.0 for both enzymes. Temperature optima and stability data for BHPNP1 (70 C and t1/2 at 60 C of 20.8 h) indicated that the biocatalytic step was operating within the capabilities of this enzyme and would operate well at elevated temperatures (up to 60 C). Conversely, the temperature optimum and stability data for EcUP (optimum of 40 C and t1/2 at 60 C of 9.9 h) indicated that the enzyme remained active at 40 C for the duration of a 25 h biotransformation, but at 60 C would only be operating at 20% of its optimum activity and would lose activity rapidly. BHPNP1 and EcUP were used in a bench scale (650 ml) transglycosylation for the production of 5-MU. A 5-MU yield of 79.1% was obtained at this scale with a reactor productivity of 1.37 g.l-1.h-1. Iterative saturation mutagenesis was used to rapidly evolve EcUP for improved thermostability. A moderately high throughput colorimetric method was developed for screening the mutants based on the release of p-nitrophenol upon phosphorolysis of a pyrimidine nucleoside analogue. By screening under 20 000 clones the mutant UPL8 was isolated. The mutant enzyme showed an optimum temperature of 60 C and improved stability at 60 C (t1/2 = 17.3 h). The increase in stability of UPL8 is due to only 2 mutations (Lys235Arg, Gln236Ala). These mutations may have caused an increase in stability due to interactions with other structural units in the protein, stabilization of the entrance to the binding pocket, or by decreasing the flexibility of the α-helix at the N-terminus. Transglycosylation experiments showed that the mutant enzyme UPL8 is a superior catalyst for the production of 5-MU. A 300% increase in reactor productivity was noted when free enzyme preparations of UPL8 was combined with BHPNP1 at 1.5% m.m-1 substrate loading. The high yield of 5-MU (75-80% mol.mol-1) was maintained at 9% m.m-1 substrate loading. A commercially viable productivity of 31 g.l-1.h-1 was thus realised. Further optimisation of the process could produce still higher productivities. Future work in directed evolution of nucleoside phosphorylases is envisaged for improved stability and enhanced substrate range for application to other commercially relevant transglycosylation reactions.

An exploration of the nature of a private general medical practice as a social system : a case study

- Description: This research study explores in general the nature of a private general medical practice [PGMP] and whether analysis of the PGMP as a social system can lead the Group Dynamics Practitioner towards developing interventions that will enhance group effectiveness in the PGMP support staff group. The main assumption is that, through the application of a framework of analysis based on that of G. C. Homans and the AGIL functional prerequisites developed by T. Parsons, a structured analysis of the external and internal variables that impact on the PGMP as a social system can be undertaken. The findings of the analysis would lead to the formulation of interventions that would improve the performance effectiveness of the PGMP as a social system. Following a two-questionnaire survey of 17 practices that provided demographic information as well as soft skills training needs, a single PGMP was identified for the case study. Data pertaining to the group as a social system were collected, and by using direct observation, content analysis and a sociometric test, the practice support staff sub-system, being the main focus of this research, could be analysed. By linking the findings to the elements of the framework of analysis, areas of ineffective group functioning could be identified and interventions suggested. This research indicates that the choice of soft skills is associated with the nature and size of the practice, as well as the dynamics of the sociometric patterns characteristic of the relations within the practice support staff subsystem; that while some practice support staff have preferences for sociometric task and socio-emotional relations outside their work clusters, these seem to serve as a buffer against clique forming, thus enhancing the function of integration within the social system as a whole; and that the physical practice layout, and the interaction dynamics that it creates, tend to hinder integration between the members of the practice support staff group, as a social subsystem.
