- Description: The preferential in vitro adsorption of apolipoprotein E (Apo E) onto the surface of colloidal drug carriers may be used as a strategy to evaluate the in vivo potential for such systems to transport drugs to the brain. The aim of this research was to investigate the in vitro protein adsorption patterns of didanosine-loaded nanostructured lipid carriers (DDI-NLCs), using two-dimensional polyacrylamide gel electrophoresis (2-D PAGE), in order to establish the potential for NLCs to deliver DDI to the brain. NLC formulations were manufactured using high-pressure homogenization using a lipid matrix consisting of a mixture of Precirol® ATO 5 and Transcutol® HP. The 2-D PAGE analysis revealed that NLCs in formulations stabilized using Solutol® HS 15 alone or with a ternary surfactant system consisting of Solutol® HS 15, Tween® 80, and Lutrol® F68, preferentially adsorbed proteins, such as Apo E. Particles stabilized with Tween® 80 and Lutrol® F68 did not adsorb Apo E in these studies, which could be related to the relatively large particle size and hence small surface area observed for these NLCs. These findings have revealed that DDI-loaded NLCs may have the potential to deliver DDI to the brain in vivo and, in addition, to Tween® 80, which has already been shown to have the ability to facilitate the targeting of colloidal drug delivery systems to the brain. Solutol® HS 15–stabilized nanoparticles may also achieve a similar purpose.

Separation was achieved using a Phenomenex® Luna 5 μm (C18) column and a mobile phase comprised of phosphate buffer (adjusted to pH 3.0): acetonitrile in a ratio of 70:30 (v/v). Detection was accomplished using a full scan multi channel ESA Coulometric detector in the “oxidative-screen” mode with the upstream electrode (E1) set at +600 mV and the downstream (analytical) electrode (E2) set at +950 mV, while the potential of the guard cell was maintained at +1050 mV. The detector gain was set at 300. Experimental design using central composite design (CCD) was used to facilitate method development. Mobile phase pH, molarity and concentration of acetonitrile (ACN) were considered the critical factors to be studied to establish the retention time of CPT and cyclizine (CYC) that was used as the internal standard. Twenty experiments including centre points were undertaken and a quadratic model was derived for the retention time for CPT using the experimental data. The method was validated for linearity, accuracy, precision, limits of quantitation and detection, as per the ICH guidelines. The system was found to produce sharp and well-resolved peaks for CPT and CYC with retention times of 3.08 and 7.56 min, respectively. Linear regression analysis for the calibration curve showed a good linear relationship with a regression coefficient of 0.978 in the concentration range of 2–70 μg/mL. The linear regression equation was y = 0.0131x + 0.0275. The limits of detection (LOQ) and quantitation (LOD) were found to be 2.27 and 0.6 μg/mL, respectively. The method was used to analyze CPT in tablets. The wide range for linearity, accuracy, sensitivity, short retention time and composition of the mobile phase indicated that this method is better for the quantification of CPT than the pharmacopoeial methods.

Eudragit NE 30D (ethyl acrylate–methyl methacrylate copolymer in a 2:1 ratio) aqueous dispersion was used as a granulating binder for the manufacture of VRP mini-matrix sustained-release tablets. The wet granulation process was performed to prepare free-flowing granules that were blended with Carbopol. The tablets were manufactured using a single-punch press by compression of the granules with magnesium stearate as a lubricant. Drug release was determined in phosphate buffer solution using USP Apparatus 2 and 3. Dissolution data were fitted to zero- and first-order models; in addition, the kinetic data were determined by evaluation of Higuchi release kinetics. The mechanism of drug release was established using the Korsmeyer–Peppas model. In general, all tablets showed high mechanical resistance with less than 1% friability. There was no significant difference between the dissolution profiles of the formulation manufactured in-house and the commercially available product. The release mechanism of the formulated and marketed products was controlled by anomalous non-Fickian diffusion. VRP release was prolonged for 12 h indicating the usefulness of the formulation as a twice-daily dosage form. The mechanism of drug release for the dosage forms was unaffected by the choice of apparatus.

The main purpose of this study was to explore the impact of Organizational commitment on Organizational Citizenship Behaviour (OCB). In this study, age and gender effects on OCB of tertiary institution employees were also examined. The data was collected from 68 academic and administration staff from the University of Fort Hare, Alice campus through questionnaires. The results show that 68 respondents are of the view that organizational commitment impacts positively in enhancing the organizational citizenship behaviour of the employees. The findings also reveal that age and gender play no role in the practice of employees OCB. Management can increase the levels of OCB in their employees by recognizing employees who engage themselves in such behaviour. The Global drive for an interlinked world economy has increased the need for monetary authorities and Governments to able to effectively deal with any negative spins off from capital flows and also be able to take advantage of positive effects capital flows may have on an economy. The study seeks to understand how the change to lift restrictions on capital flows into the South African economy may have impacted on economic growth. The study analyses the relationship that existed between capital flows, that is to say foreign direct investment (FDI) and portfolio investment (P_I) and economic growth under the period of capital controls (1975 Q1 to 1994Q1). Then study will then analyse the same relationship but this time under the liberalised period (1994 Q2 to 2010 Q2) and compare how the long run relationship has changed after capital account liberalisation. The study uses an endogenous model to determine the relationship. The study unlike most will focus on a single economy, which is South Africa and not use panel data like most previous studies. The study found that in the short run capital account liberalising aided economic growth as both FDI and P_I became significant, with positive coefficients and also found that there is long run relationship between economic growth and capital flows. In the long run FDI is significant while P_I is not. After liberalisation FDI adjusted faster in the long run than before liberalisation on its impact on economic output. Also of note was that the study found that under capital controls the conditional variance was constant but after liberalisation the relationship between capital flows and economic growth became more sensitive to negative news and the conditional variance was not constant thus indication of increased volatility. To maximise from opening up of capital accounts the economy should maintain sound macroeconomic policies. Equality is our Constitution’s focus and organising principle.” Given the foundational role of equality in the South African constitutional framework, the drafters of the South African Constitution (the Constitution) directed the South African Parliament (Parliament) to enact legislation to “prevent or prohibit unfair discrimination” between individuals within three years of the enactment of the Constitution.3 Under great pressure, Parliament finalised and passed the Promotion of Equality and Prevention of Unfair Discrimination Act (the Equality Act) within two days of the constitutional deadline.4 The Equality Act, as the title indicates, addresses the promotion of equality on the one hand, and provides for reactive measures where the equality right is breached, on the other. The reactive provisions include the prohibition of unfair discrimination and related infringements of the equality right. The Equality Act expressly provides for the enforcement of its provisions in specifically created equality courts. The majority of the reactive provisions of the Equality Act have been operational since 16 June 2003. More than a decade after the enactment of the legislation, the promotional aspects of the Equality Act are yet to come into operation. Eighteen landscape-scale case studies around the world, in Vietnam, India, Réunion, Madagascar, South Africa, Congo, Niger, Ethiopia, Israel, France, Portugal, Brazil, Chile, Dominican Republic and Hawai‘i. Qualitative comparison of case studies, based on questionnaire sent to network of acacia researchers. Information based on individual knowledge of local experts, published and unpublished sources. We propose a conceptual model to explain current uses and perceptions of introduced acacias. It highlights historically and geographically contingent processes, including economic development, environmental discourses, political context, and local or regional needs. Four main groupings of case studies were united by similar patterns: (1) poor communities benefiting from targeted agroforestry projects; (2) places where residents, generally poor, take advantage of a valuable resource already present in their landscape via plantation and/or invasion; (3) regions of small and mid-scale tree farmers participating in the forestry industry; and (4) a number of high-income communities dealing with the legacies of former or niche use of introduced acacia in a context of increased concern over biodiversity and ecosystem services. Economic conditions play a key role shaping acacia use. Poorer communities rely strongly on acacias (often in, or escaped from, formal plantations) for household needs and, sometimes, for income. Middle-income regions more typically host private farm investments in acacia woodlots for commercialization. Efforts at control of invasive acacias must take care to not adversely impact poor dependent communities.

The first, the Arkwork Collective, is an art-junk process that engages marginalised youth in Grahamstown, South Africa in a process that uses creative sculpture and drama to explore personal and social issues that exist in their immediate context. The second, Jonga Phambili Sinethemba looks into the impact of climate change and HIV/AIDS (amongst other issues) in the rural and peri-urban communities of Willowvale and Lesseyton in the Eastern Cape, South Africa. It seeks to provide a platform where members of each community can define the vulnerabilities, capabilities, social networks in their areas with the aim of bolstering the adaptive capacity of these communities. Snippets of my experiences in these projects are shared with the intention of demonstrating constraints to learning in a social context. Key ideas that the paper explores include honouring the lived experiences of participants as part of the process, prioritising the participation of each individual present as part of the ongoing conversation, the challenge of surfacing the vital independent links of a collective, drawing on the reflective capacity of a diverse group, assessing the quality of participation, building capabilities for ‘response-ability’ and rethinking facilitation. Each section sets out challenges and questions for practitioners in this field to reflect on. The paper suggests that in order to achieve the laudable aims of social learning, we need to peel back the common rhetoric of its participatory aims and acknowledge the complexity, flexibility and dedication that it requires.

Feminist debates on cosmetic surgery have focused on the question of whether to regard women who modify their bodies in this way as active agents engaged in liberatory ‘body projects’, or whether such projects are evidence of their subjection to oppressive stereotypes and beauty norms. The latter perspective is challenged here by the participants’ characterisation of breast reduction surgery as profoundly ‘freeing’. The article deals in particular with the conscious choice of participants to knowingly risk not being able to breastfeed children in future in order to achieve a body type which conforms to their understanding of youthful beauty and sexuality.

It reviews and reports on the history of environment and sustainability education in teacher education, and from this, the national case study begins to conceptualise a new approach to environment and sustainability teacher education within a new curriculum policy environment, and a new teacher education and development policy environment.Action research case study methodology is used to document the first phase of the emergence of this network, and this report covers Phase 1 of the initiative, which covers formation of the network, review of previous practices, three conceptual development pilot studies undertaken in both in-service and pre-service teacher education environments and a piloting of a ‘Train the Trainers’ or ‘Educate the Teacher Educators’ programme, which complements and extends the actual teacher education and development (TED) programme under development.The study highlights critical insights of relevance to the shift to a content referenced curriculum in South Africa, and shows how the ‘knowledge mix’ which forms the foundation of the new Teacher Education Qualifications Framework can be engaged. It also highlights some features of the changing knowledge environment, and what dominant knowledge practices are in environment and sustainability-related teaching and teacher education practices, opening these up for further scrutiny. It raises concerns that dominant knowledge work, while integrating a range of forms of knowledge (as is expected of the teacher education system under the new policy), tends to be limited by content on problems and issues for raising awareness, and fails to develop deeper conceptual depth and understanding of environment and sustainability, as issues based knowledge dominates. Similarly, it fails to support social innovation as a response to environment and sustainability concerns, as awareness raising dominates in dominant knowledge work. The study provides a revised conceptual framework for the Teacher Development Network (TEDN) programme, with guidance on key elements necessary to take the programme forward in Phase 2.

- Description: This national case study reports on the development of a national network, curriculum framework and resources for teacher education, with specific focus on the inclusion of environment and sustainability, also known as education for sustainable development (ESD) in the South African teacher education system. It reviews and reports on the history of environment and sustainability education in teacher education, and from this, the national case study begins to conceptualise a new approach to environment and sustainability teacher education within a new curriculum policy environment, and a new teacher education and development policy environment. Action research case study methodology is used to document the first phase of the emergence of this network, and this report covers Phase 1 of the initiative, which covers formation of the network, review of previous practices, three conceptual development pilot studies undertaken in both in-service and pre-service teacher education environments and a piloting of a ‘Train the Trainers’ or ‘Educate the Teacher Educators’ programme, which complements and extends the actual teacher education and development (TED) programme under development. The study highlights critical insights of relevance to the shift to a content referenced curriculum in South Africa, and shows how the ‘knowledge mix’ which forms the foundation of the new Teacher Education Qualifications Framework can be engaged. - Description: A series of water-soluble tetrasulfonated metallophthalocyanines (MPcs) dyes have been studied to be used as a drug or photosensitizer (PS) in photodynamic therapy (PDT) for the treatment of cancers. During PDT the PS is administrated intravenously or topically to the patient before laser light at an appropriate wavelength is applied to the cancerous area to activate the PS. The activated PS will react with oxygen typically present in the cancerous tissue to generate reactive oxygen species for the destruction of the cancerous tissue. This in vitro study aimed at investigating the cytotoxic effects of different concentrations of zinc tetrasulfophthalocyanines (ZnTSPc) activated with a diode laser (λ = 672 nm) on melanoma, keratinocyte and fibroblast cells. To perform this study 3 × 104 cells/ml were seeded in 24-well plates and allowed to attach overnight, after which cells were treated with different concentrations of ZnTSPc. After 2 h, cells were irradiated with a constant light dose of 4.5 J/cm2. Post-irradiated cells were incubated for 24 h before cell viability was measured using the CellTiter-Blue Viability Assay. Data indicated high concentrations of ZnTSPc (60–100 μg/ml) in its inactive state are cytotoxic to the melanoma cancer cells. Also, results showed that photoactivated ZnTSPc (50 μg/ml) was able to reduce the cell viability of melanoma, fibroblast and keratinocyte cells to 61%, 81% and 83% respectively. At this photosensitizing concentration the efficacy the treatment light dose of 4.5 J/cm2 against other light doses of 2.5 J/cm2, 7.5 J/cm2 and 10 J/cm2 on the different cell lines were analyzed. ZnTSPc at a concentration of 50 μg/ml activated with a light dose of 4.5 J/cm2 was the most efficient for the killing of melanoma cancer cells with reduced killing effects on healthy normal skin cells in comparison to the other treatment light doses. Melanoma cancer cells after PDT with a photosensitizing concentration of 50 μg/ml and a treatment light dose of 4.5 J/cm2 showed certain apoptosis characteristics such as chromatin condensation and fragmentation of the nucleus. This concludes that low concentrations of ZnTSPc activated with the appropriate light dose can be used to induce cell death in melanoma cells with the occurrence of minimal damage to surrounding healthy tissue.
