- Description: Terfezia pfeilii is an edible mycorrhizal fungus that thrives in the Kalahari Desert of southern Africa. It is best known by desert dwellers for its flavour and as a source of nutrition. Although the genus Terfezia is generally regarded as being an ectomycorrhizal mycobiont, the exact mycorrhizal type formed by T. pfeilli and its' associated host plants remains uncertain. Discovery of the host plants for T. pfeilii would first be required in order to further investigate the life cycle and cultivation of this truffle. This study focussed on the isolation of mycelia from the ascocarp, optimising the growth conditions of the mycelial cultures, rapid molecular identification of T. pfeilii, investigation of potential helper bacteria and mycorrhizal synthesis experiments. T. pfeilii ascocarps were harvested from the Spitskop Nature Reserve in Upington, South Africa. Ascocarps were successfully identified using both morphological and molecular methods. Despite the delayed growth mostly caused by contaminating microorganisms, the isolation of T. pfeilii mycelia culture was successful. Molecular techniques were used to confirm the identity of the pure culture. Further studies were conducted on ways to improve the growth conditions of the mycelial culture on Fontana medium. An optimum temperature of 32°C, the addition of Bovine Serum Albumin as a nitrogen source and a pH of 7.5 significantly improved the growth of T. pfeilii in vitro. A rapid PeR-based molecular method was developed to speed up the identification of T. pfeilii. Specific primers that can exclusively amplify the ITS region of T. pfeilii were designed and used to identify both the ascocarps and the mycelial culture. The specificity of these primers was confirmed by their inability to amplify DNA from the isolates of contamining fungi obtained during the isolation process. Molecular comparison was made to confirm the reclassification of South African samples of T. pfeilii as Kalaharituber pfeilii as proposed by Ferdman et al.,(2005). However, in this study, the name T. pfeilii has been retained. A total of 17 bacterial isolates were obtained from the fruiting bodies of T. pfeaii and these were tested for stimulation of mycelial growth in vitro, indole production and phosphate solubilising capabilities. Bacterial isolates that showed potential to be Mycorrhization Helper Bacteria (MHB) were identified as Paenibacillus sp., Bacillus sp. and Rhizobium tropici. Selected plant seedlings were inoculated with T. pfeilii cultures or ascocarp slurry in order to re-establish the mycorrhizal association. After 8 months, light microscopy observations revealed an endomycorrhizal type association between Cynodon dactylon and T. pfeilii. This was confirmed with molecular analysis using specific T. pfeilii ITS primers. After 15 months, molecular methods confirmed Acacia erioloba as another host plant. The perceived economic impact of the city of Johannesburg's storm water attenuation policy on private property developers

- Description: Stormwater attenuation policy has, over recent years, become a highly contentious issue for the property development community. Increased urbanisation, locally and internationally, has forced municipal authorities to reconsider the role of stormwater management in this evolving urban landscape. It is within this context that the legislative support and municipal policy for stormwater management in the City of Johannesburg has been explored, particularly in terms of the economic impact on property developers. The research aimed to explore the relationship between stormwater attenuation policy and the economic impact thereof on private property developers. Consideration was given to the cost, risk and time factors of legislative and policy requirements within the development process. Further consideration was given to the physical environment that has brought about current pressures. Based upon research of international issues highlighted in countries with well developed attenuation policy, a questionnaire was constructed to evaluate the response of local private property developers to selected issues as key sub-problems. The responses were tested against the developed set of hypotheses. The research indicated that developers had a generally poor level of knowledge with regard to the stormwater management policy of the City of Johannesburg, as well as the supporting legislative requirements of provincial and national policy documents. The results of the survey also indicated that developers were strongly opposed to the loss of developable area, but indicated a limited financial impact of the current stormwater attenuation policy. The risk inherent in incorporating an attenuation facility within a development was identified as being low with little perceived impact. Developers further indicated that the inclusion of attenuation facilities was seen as a significant contributing factor in the delay, approval and acceptance of new developments in which attenuation facilities were required, while maintenance costs were indicated to be of a low level of importance. Recommendations in response to the research findings included the establishment of detailed policy documentation and support for effective distribution channels in conjunction with industry and public focused information campaigns, improved municipal capacity and a greater level of technical support. A need for additional stormwater research and an increase in the capture of relevant data for GIS purposes was identified.

- Description: Construction Planning will increasingly play a more critical role within the realm of the Built Environment. Existing practices used to plan and communicate the construction process to be undertaken are failing to deliver the desired results for construction companies and clients alike. At a time of unprecedented growth in the industry around the world, which is leading to a general skills shortage, especially in management positions, construction planners are increasingly being asked to deal with more responsibility. As with other industries, technological improvement in the tools at their disposal is one way to address the inadequacies of the present situation. Increasingly, three dimensional design packages are being used to generate construction information which can then be used for quantities calculations, automated manufacturing processes and construction simulation. The latter forms the basis for their use in the process of planning, through new technologies being developed as virtual construction tools or 4D planning, the addition of time to the 3D model environment, but using the elements within the model as the basis for the construction programme. The benefit of using the design information to form the basis of the programme is that the interface tasks and logistical activities, as well as location related constraints, can be identified and then communicated to all levels within the construction team through a time based visual image. The purpose of this study is therefore to establish a scientifically analysed alternative method for the creation, review and delivery of construction programmes. In order to achieve the research objectives, three methodologies have been employed. Firstly, the literature review in the fields of planning including existing methodologies and previous research of 4D related techniques has been analysed. An overview of the perceived weaknesses to current practises and proposed solution will be explored and best case scenarios outlined and further investigated. Secondly, the 51 Lime Street project provides an environment in which the proposed 4D planning techniques have been implemented and the benefits of the process can, through observation / participation methodology, be validated. Thirdly, through interview questionnaires, with Lime St contractors and management, and e-mail questionnaires to a broader sample stratum, data on the ability of the tools, the techniques employed on 51 Lime Street and similar projects have been collated and statistically analysed to validate the reliability and relevance for future implementation. The result of the research will provide management teams with a practical alternative to existing planning methodologies. Construction planners will have alternative technique that can further enhance their role within the project team whilst increasing their ability to communicate the team’s vision to a wider audience, making them and the project more efficient and effective in the process. It has been proposed that clients insist on the use of 3D from the commencement of the design process so that this information can be passed downstream through the construction process and onto facilities management. Planners need to be able to communicate their requirements better and the 4D planning models provide both a more inclusive way of planning alongside a better communications medium in the form of moving images. The use of thermal modelling in improving rural subsistence aquaculture

Many of these attempts have failed due to inadequate support from government and support agencies. There has been a recent revived interest in promoting rural aquaculture development. Presented in this project are six scenarios demonstrating the potential application of thermal modelling techniques to improving subsistence rural aquaculture. These scenarios were based on a model validated against a real life system, and run with environmental data for the year 2005. Bovine manure, grass thatch, cereal straw and Typha spp. water reeds were tested for thermal performance. These results were then applied to the scenarios. The open and kraal enclosed pond scenarios yielded the widest deviation in temperatures throughout the year, with an average temperature of 3.8oC and 6.0oC above ambient and a temperature range of 10.6oC and 12.4oC for the year respectively. Eliminating solar radiation and reducing wind speed resulted in an average difference of 1.5oC below ambient and a temperature range of 7.5oC for the year. When completely enclosed in a well-sealed traditional Xhosa style hut, the pond had an average difference of 5.6oC below outside ambient temperature, with a range of 7.8oC for the year. A passive solar heating scenario added to the sealed hut scenario, which when run continuously maintained an average difference of 34.7oC above outside ambient temperatures, with a range of 56oC for the year. When a hypothetical 25oC temperature control was included in the model, the system maintained an average of 7.1oC above ambient outside temperatures, with a temperature range of 0.8oC throughout the year. The wider range of temperatures made available through the use of thermal modelling approaches provides a more diverse range of species available for subsistence rural aquaculture at any site, significantly improving its potential.

These low values were confirmed by direct measurements of ωp in reflectance spectra obtained in the range 15-200 cm-1. The simulated carrier concentration and mobility values determined optically were compared to the values determined by Hall measurements at room temperature and previously reported values by other researchers. The simulated values obtained were in reasonable agreement with the Hall values. The simulated and measured carrier concentrations obtained for InAs layers were significantly higher than the intrinsic carrier concentration for InAs at room temperature, indicating notable concentrations of donors resulting from the growth process.

The ligand dab is coordinated to the rhenium(V) centre through a dianionic imido nitrogen only, in a distorted octahedral coordination geometry around the metal ion. The complex trans-[Re(ada)Cl3(PPh3)2] (H2ada=2-aminodiphenylamine) was prepared from the reaction of trans-[ReOCl3(PPh3)2] with H2ada in acetonitrile. The reaction of equimolar quantities of cis-[ReO2I(PPh3)2] with 5,6-diamino-1,3- dimethyluracil (H2ddd) in acetonitrile led to the formation of [Re(ddd)(Hddd)I(PPh3)2](ReO4). The X-ray crystal structure shows that the ligand ddd is coordinated monodentately through the doubly deprotonated amino nitrogen and is therefore present as an imide. The chelate Hddd is coordinated bidentately via the neutral amino nitrogen, which is coordinated trans to the imido nitrogen, and the singly deprotonated amido nitrogen, trans to the iodide. The reaction of equimolar quantities of [NH4(ReO4)] with H2ddd in methanol under reflux conditions led to the isolation of [C12H12N6O4] as only product. The [ReO4]- ion is therefore instrumental in the formation of [C12H12N6O4], and since the product contains no rhenium in any oxidation state, the conclusion is that [ReO4]- catalyses the oxidative deamination of H2ddd. The X-ray crystal structure consists of two centrosymmetric, tricyclic rings, comprising a central pyrazine ring and two terminal pyrimidine rings. The reaction of a twofold molar excess of H2apb (H2apb=2-(2-aminophenyl)-1Hbenzimidazole) with trans-[ReO2(py)4]Cl in ethanol gave the green product of the formulation [ReO(Hapb)(apb)] in good yield. The rhenium atom lies in a distorted trigonal-bipyramidal environment. The two imidazole N(2) atoms lie in the apical positions trans to each other, with the oxo-oxygen and two amido N(1) atoms in the trigonal plane. The complex has C2-symmetry. The two amino groups are singly deprotonated and provide a negative charge each, so that they are coordinated as amides. The oxo group provides two negative charges. In order to obtain electroneutrality for the rhenium(V) complex, the two coordinated imidazole nitrogens provide one negative charge. The complex salt trans-[Re(mps)Cl(PPh3)2](ReO4) (H3mps=N-(2-amino-3- methylphenyl)salicylideneimine) was prepared by the reaction of trans- [ReOCl3(PPh3)2] with a twofold molar excess of H3mps. The Xray crystal structure shows that the mps ligand coordinates as a tridentate chelate via the doubly deprotonated 2-amino nitrogen, the neutral imino nitrogen and the phenolate oxygen. The imide and phenolate oxygen coordinate trans to each other in a distorted octahedral geometry around the rhenium(V) centre, with the two chlorides in cis positions.

Individuals of similar functional groups were divergent in their peak flowering times and limited their degree of flowering overlap further by spatial aggregation and niche separation, within the Thicket of the Gamtoos River Valley. The nectar properties were especially useful in structuring the bird pollinator community, which resulted in greater ethological isolation and hence, greater reproductive assurance in the mixed co-flowering plant communities. Choice array experiments revealed that it was the fine scale aggregation of flowering individuals that ensured that bird-pollinators feed selectively, since when equal choice was available, interspecific visitation increased significantly compared to natural scenarios. Bird behaviour and the ecological intermediateness of one to the species explained its prominence in hybrid combinations. The spatial occurrence of hybrid individuals can be traced back to the energetics of foraging and its influence on bird floral constancy. The pollination ecology of similar South African Aloe species were extrapolated from these and recent findings by various authors, but emphasises the need for a robust natural phylogeny of the Aloaceae in order to draw comprehensive conclusions on the evolutionary radiation of this highly charismatic group.

Having a system that notifies the user of important messages using the most appropriate medium and device will benefit users that rely on time critical information. There are several existing systems and models for mobile notification as well as for adaptive mobile notification using context awareness. Current models and systems are typically designed for a specific set of mobile devices, modes and services. Communication however, can take place in many different modes, across many different devices and may originate from many different sources. The aim of this research was to develop a model for adaptive mobile notification using context awareness. An extensive literature study was performed into existing models for adaptive mobile notification systems using context awareness. The literature study identified several potential models but no way to evaluate and compare the models. A set of requirements to evaluate these models was developed and the models were evaluated against these criteria. The model satisfying the most requirements was adapted so as to satisfy the remaining criteria. The proposed model is extensible in terms of the modes, devices and notification sources supported. The proposed model determines the importance of a message, the appropriate device and mode (or modes) of communication based on the user‘s context, and alerts the user of the message using these modes. A prototype was developed as a proof-of-concept of the proposed model and evaluated by conducting an extensive field study. The field study highlighted the fact that most users did not choose the most suitable mode for the context during their initial subscription to the service. The field study also showed that more research needs to be done on an appropriate filtering mechanism for notifications. Users found that the notifications became intrusive and less useful the longer they used them.

- Description: The black rhinoceros remains one of the world's extremely endangered species despite a variety of policies to protect it. The black rhinoceros population at the Great Fish River Reserve (GFRR) in the Eastern Cape in South Africa has increased steadily since their re-introduction in 1986. This megaherbivore is a browser, with a diet obtained largely from the short and medium succulent thicket of the GFRR. Knowledge of the preferential diet of the black rhinoceros on the reserve is an important factor for the effective management of the land and the herbivores that compete for its resources. The dietary preferences of the black rhinoceros at the reserve have been established using backtracking methods. In this study the rbcL gene was used to establish an rbcL gene database of the plants from the GFRR and determine the botanical composition of the black rhinoceros dung from the GFRR. Due to the limited number of rbcL gene plant sequences from the GFRR deposited in the GenBank database, 18 plant species from the GFRR were sequenced. Sequence analyses between the partial rbcL gene sequences generated were able to distinguish between plants down to species level. Plant species from the family Euphorbiaceae and Fabaceae showed sequence variation at intra-specific level compared to those of Tiliaceae which were more conserved. The generated rbcL gene sequences from seasonal dung samples were compared to the rbcL gene sequenced from 18 plant species obtained from the GFRR and those from the GenBank database. A wide range of plant species were identified from the dung samples. There were no major differences in botanical composition between the dung samples, except that Grewia spp. were found to dominate in almost all seasons. The results obtained on the free radical scavenging activity of the extracts against 2,2-Diphenyl-l-picrylhydrazyl (DPPH) increased in the order of methanol > ethyl acetate > chloroform. The DPPH free radical scavenging activity of the methanol plant extracts increased in the order Brachylaena elliptica > Plumbago auriculata > Grewia robusta > Azima tetracantha. Methanol extracts on the TLC plate sprayed with Fe³⁺-2,4,6-Tri-2-pyridyl-s-triazine (TPTZ) showed that the compounds present in the extracts react differently to ferric ion, with most compounds unable to reduce ferric ion. Furthermore the methanol extracts were able to exhibit reduction potentials vs. Ag/AgCl at low concentrations. The compounds in the extracts were shown to be phenolic acids and flavonoid glycosides.

- Description: The human skin blanching assay (HSBA) is a well-researched and validated method for the bioequivalence assessment of topical corticosteroids. Traditionally, visual assessment of skin blanching has been used. Such testing methods are not conducive for interlaboratory comparisons. Regulatory bodies prefer less subjective methods of analysis. The FDA released guidelines on the assessment of bioequivalence for topical corticosteroids that recommends the use of a chromameter as a reliable method to measure skin blanching although the use of visual assessment with acceptable validation is also provided for. However, the FDA does not elucidate on the manipulation and handling of the chromameter during skin blanching measurements. The purpose of this project was several fold, which included investigations to standardize the manipulation and handling of a chromameter. In particular, measures to avoid skin whitening resulting from the effects of pressure on the skin during chromameter use were investigated. Other methods of analysis should surpass or at least be comparable to the HSBA if such methods are to be used for the assessment of topical corticosteroids. Microdialysis is a relatively new technique for assessing the rate at which drug penetrates the skin. The advantage of using this method is that there are fewer restrictions for selection of an appropriate study population unlike those required for the HSBA where one has to be both a ‘responder’ and a ‘detector’ for their results to be used in data analysis. Microdialysis was investigated by initially conducting experiments in which microdialysis probes were embedded into topical formulations containing mometasone furoate (MF) and the initial results revealed that relatively low drug was released from the formulations. These results indicated that should microdialysis be applied to measure the in vivo release of MF from such topical formulations following application to the skin, even lower concentrations of MF would likely result in the dialysate, necessitating the need for ultra-high sensitive methods of analysis. Typically, the availability of an appropriate analytical technique such as liquid chromatography coupled with mass spectrometry (LCMS) would be a pre-requisite for such in vivo studies. However, only high-pressure liquid chromatography (HPLC) and other less sensitive equipment was available in the laboratories. The study objectives were therefore focussed on in vitro assessment of the release of MF from topical formulations using microdialysis and Franz cells. In addition, the in vivo release of MF was also studied using the HSBA. Data obtained from the microdialysis experiments were compared with the data obtained from the Franz cell diffusion studies in order to provide information on the pharmaceutical availability of MF from the various topical MF dosage forms. Subsequently, pharmaceutical equivalence was investigated from the comparative pharmaceutical availability data using statistical analysis. An additional objective was to attempt to correlate in vitro with in vivo data (IVIVC) to establish a model that could be used to assess safety and efficacy of generic topical drug products. The in vivo data obtained from the HSBA were processed according to the FDA requirements and these pharmacodynamic data were subsequently compared with the microdialysis and Franz cell results. In summary the objectives of this project were: 1. To develop a system to improve the reproducibility of the use of a Minolta® chromameter and compare this with the standard/normal manipulation and handling of such instruments. 2. - Description: The human skin blanching assay (HSBA) is a well-researched and validated method for the bioequivalence assessment of topical corticosteroids. Traditionally, visual assessment of skin blanching has been used. Such testing methods are not conducive for interlaboratory comparisons. Regulatory bodies prefer less subjective methods of analysis. The FDA released guidelines on the assessment of bioequivalence for topical corticosteroids that recommends the use of a chromameter as a reliable method to measure skin blanching although the use of visual assessment with acceptable validation is also provided for. However, the FDA does not elucidate on the manipulation and handling of the chromameter during skin blanching measurements. The purpose of this project was several fold, which included investigations to standardize the manipulation and handling of a chromameter. In particular, measures to avoid skin whitening resulting from the effects of pressure on the skin during chromameter use were investigated. Other methods of analysis should surpass or at least be comparable to the HSBA if such methods are to be used for the assessment of topical corticosteroids. Microdialysis is a relatively new technique for assessing the rate at which drug penetrates the skin. The advantage of using this method is that there are fewer restrictions for selection of an appropriate study population unlike those required for the HSBA where one has to be both a ‘responder’ and a ‘detector’ for their results to be used in data analysis. Microdialysis was investigated by initially conducting experiments in which microdialysis probes were embedded into topical formulations containing mometasone furoate (MF) and the initial results revealed that relatively low drug was released from the formulations. These results indicated that should microdialysis be applied to measure the in vivo release of MF from such topical formulations following application to the skin, even lower concentrations of MF would likely result in the dialysate, necessitating the need for ultra-high sensitive methods of analysis. Typically, the availability of an appropriate analytical technique such as liquid chromatography coupled with mass spectrometry (LCMS) would be a pre-requisite for such in vivo studies. However, only high-pressure liquid chromatography (HPLC) and other less sensitive equipment was available in the laboratories. The study objectives were therefore focussed on in vitro assessment of the release of MF from topical formulations using microdialysis and Franz cells. In addition, the in vivo release of MF was also studied using the HSBA. Data obtained from the microdialysis experiments were compared with the data obtained from the Franz cell diffusion studies in order to provide information on the pharmaceutical availability of MF from the various topical MF dosage forms. Subsequently, pharmaceutical equivalence was investigated from the comparative pharmaceutical availability data using statistical analysis. An additional objective was to attempt to correlate in vitro with in vivo data (IVIVC) to establish a model that could be used to assess safety and efficacy of generic topical drug products. The in vivo data obtained from the HSBA were processed according to the FDA requirements and these pharmacodynamic data were subsequently compared with the microdialysis and Franz cell results. In summary the objectives of this project were: 1. To develop a system to improve the reproducibility of the use of a Minolta® chromameter and compare this with the standard/normal manipulation and handling of such instruments. 2. To develop and validate an HPLC method for the analysis of MF for use with in vitro diffusion studies using microdialysis and Franz cells. 3. The three security methods implemented and studied were IPSec (Internet Protocol Security), SRTP (Secure Real-time Transport Protocol), and SIAX2 (Secure Inter-Asterisk eXchange 2 protocol). From the experiments, it was found that bandwidth and CPU usage were significantly affected by the addition of CIA. In ranking the three security methods in terms of these two resources, it was found that SRTP incurs the least bandwidth overhead, followed by SIAX2 and then IPSec. Where CPU utilisation is concerned, it was found that SIAX2 incurs the least overhead, followed by IPSec, and then SRTP.

- Description: The Semantic Web is a vision to create a “web of knowledge”; an extension of the Web as we know it which will create an information space which will be usable by machines in very rich ways. The technologies which make up the Semantic Web allow machines to reason across information gathered from the Web, presenting only relevant results and inferences to the user. Users of the Web in its current form assess the credibility of the information they gather in a number of different ways. If processing happens without the user being able to check the source and credibility of each piece of information used in the processing, the user must be able to trust that the machine has used trustworthy information at each step of the processing. The machine should therefore be able to automatically assess the credibility of each piece of information it gathers from the Web. A case study on advanced checks for website credibility is presented, and the site presented in the case presented is found to be credible, despite failing many of the checks which are presented. A website with a backend based on RDF technologies is constructed. A better understanding of RDF technologies and good knowledge of the RAP and Redland RDF application frameworks is gained. The second aim of constructing the website was to gather information to be used for testing various trust metrics. The website did not gain widespread support, and therefore not enough data was gathered for this. Techniques for presenting RDF data to users were also developed during website development, and these are discussed. Experiences in gathering RDF data are presented next. A scutter was successfully developed, and the data smushed to create a database where uniquely identifiable objects were linked, even where gathered from different sources. Finally, the use of digital signature as a means of linking an author and content produced by that author is presented. RDF/XML canonicalisation is discussed in the provision of ideal cryptographic checking of RDF graphs, rather than simply checking at the document level. The notion of canonicalisation on the semantic, structural and syntactic levels is proposed. A combination of an existing canonicalisation algorithm and a restricted RDF/XML dialect is presented as a solution to the RDF/XML canonicalisation problem. We conclude that a trusted Semantic Web is possible, with buy in from publishing and consuming parties.

- Description: Geographic Information System technology today allows for the rapid analysis of vast amounts of spatial and non-spatial data. The power of a GIS can only be effected with the rapid collection of accurate input data. This is particularly true in the case of the South African National Working for Water (WFW) Programme where large volumes of spatial data on alien vegetation infestations are captured throughout the country. Alien vegetation clearing contracts cannot be generated, for WFW, without this data, so that the accurate capture of such data is crucial to the success of the programme. Mapping Invasive Alien Plant (IAP) data within WFW is a perennial problem (Coetzee, pers com, 2002), because not enough mapping is being done to meet the annual requirements of the programme in the various provinces. This is re-iterated by Richardson, 2004, who states that there is a shortage of accurate data on IAP abundance in South Africa. Therefore there is a need to investigate alternate methods of data capture; such as remote sensing, whilst working within the existing WFW data capture standards. The aim of this research was to investigate the use of Landsat ETM imagery as a data capture source for mapping alien vegetation for the WFW Programme in terms of their approved mapping methods, for both automated and manual classification techniques. The automated and manual classification results were compared to control data captured by differential Global Positioning Systems (DGPS). The research tested the various methods of data capture using Landsat ETM images over a range of study sites of varying complexity: a simple grassland area, a medium complexity grassy fynbos site and a complicated indigenous forest site. An important component of the research was to develop a mapping (classification) Ranking System based upon variables identified by WFW as fundamental in data capture decision making: spatial and positional accuracy, time constraints and cost constraints for three typical alien invaded areas. The mapping Ranking System compared the results of the various mapping methods for each factor for the study sites against each other. This provided an indication of which mapping method is the most efficient or suitable for a particular area.

Experimental procedures were conducted utilising the Chatillon ™ Dynamometer to measure force output in the initial, sustained and ending phases. The K4b2 Ergospirometer was used to assess physiological responses (heart rate and oxygen consumption [V02])' Nine recorded forces and nine experimental conditions formed the basis of this study, with subjects required to push three loads (200kg, 350kg, 500kg) at three frequencies (1120 sec, 1/40 sec, 1/60 sec) at a speed of 3.6km.h-1 over 14 metres on a co-efficient of friction controlled walkway for six minutes. Gait analysis, along with perceptions of exertion (,Central ' and 'Local' RPE) were collected during the third and sixth minutes of each condition . Body discomfort and contribution were identified upon completion of each condition. The results demonstrated that load and frequency interacted to influence responses within each domain. Increasing loads required increased force output during each stage of the push, which had a concomitant effect on physiological and perceptual responses. Significant differences arose between the initial, sustained and ending forces for each load, showing the direct relationship between load and force exertion. The combination of heaviest load/quickest frequency required the greatest physiological output, exceeding recommended guidelines for heart rate, V02 and energy expenditure responses. Intermediate combinations required moderate and acceptable energy cost. Linear relationships were established between heart rate and oxygen consumption , as well as between load and V02 , thus providing industrial practitioners an opportunity to evaluate task demands in situ. The combination of high forces and elevated physiological responses increased the subjective rating of the condition. The results emphasise the need to holistically consider all contributing factors in a dynamic pushing task. Dynamic pushing tasks place biomechanical, physiological and perceptual demands on the human operator, which must be minimised in order to ensure that this form of manual materials handling becomes sustainable in the long term.

It had two aims. Firstly, to determine the role incentives could play in mitigating these pressures. Secondly, to determine the characteristics of an incentive programme that would most effectively achieve this. In order to do this, an understanding of landowner activities, needs, opinions and barriers to behaviour; the nature of the pressures on thicket and the nature of the required behaviour to reduce these pressures; and current and past institutional arrangements needed to be achieved. This was done predominantly through a current literature review and personal interviews with landowners and key informants. These findings were used to make recommendations for an effective incentive programme. Landowners showed a preference towards tangible incentives, in particular management assistance, financial compensation and law enforcement. They indicated an aversion to an incentive programme implemented by a government agency, particularly district and provincial government. Rather, landowners showed a propensity towards a nongovernment organisation (NGO) or a farmers group implementing an incentive programme. It was recommended that the two major pressures, namely land transformation and alien invader plants, required different interventions by different agencies in order to be mitigated. The pressure of land transformation required a stewardship model response, with the primary drive being a non-contractual environmental extension service to landowners. The extension service should focus on promoting pro-conservation practises, raising awareness and disseminating information. It should also build a relationship of trust between landowners and the implementing agency. The pressure of alien invader plants would be most effectively addressed through the Working for Water programme. Tangible incentives must be provided to the landowner to induce the costly exercise of alien invader plant control. In particular, the high cost of labour must be addressed. The regulatory incentive of applying laws requiring landowners to control alien invader plants on their land should also be enforced.

Tangible incentives must be provided to the landowner to induce the costly exercise of alien invader plant control. In particular, the high cost of labour must be addressed. The regulatory incentive of applying laws requiring landowners to control alien invader plants on their land should also be enforced.

- Description: Acquired Immunodeficiency Syndrome (AIDS) is a human disease caused by the human immunodeficiency virus type 1 (HIV-1) and it is one of the biggest social, economic and health challenges in the world. The Joint United Nations Programme on HIV/AIDS (UNAIDS) and World Health Organization (WHO) estimated that between 33.4 to 46.0 million people around the world were living with HIV/AIDS in December 2005 and the highest estimates are in the Sub-Saharan Africa (around 25 million). In more developed countries a combined antiretroviral therapy called highly active antiretroviral therapy (HAART) is used, which results in reduced progression to AIDS in most patients. Despite the beneficial effects of HAART, significant side effects are experienced by treated patients. In addition, most infected people live in countries where the treatment is very expensive or, in many cases, not available at all. These people therefore rely on medicinal plants for health care. In this study, aqueous extracts from Sutherlandia frutescens and Lobostemon trigonus were screened for potential anti-HIV activities in a series of in vitro enzymatic assays, including reverse transcriptase, HIV-1 protease and glycohydrolases. Two extracts of Sutherlandia leaves (SFL-1 and SFL-2) were prepared that inhibited HIV reverse transcriptase and a Lobostemon leaf extract (LTL) was shown to also inhibit this enzyme. All extracts were assayed at 1.25mg/ml. Tannin content was determined for all active extracts using a tannic acid assay. SFL-1 and SFL-2 were found to contain about 6 percent and 7 percent tannins, respectively, and LTL contained 31% tannins by weight. Tannins were removed using polyamide columns and three fractions were collected for each. The extracts were also fractionated with Sephadex G-25, Amberlite IR 120 and Dowex 1-X8 as size exclusion, cation exchange and anion exchange, respectively. Extracts were also fractionated by preparative thin layer chromatography where two compounds were separated from S. frutescens extract with high activity against reverse transcriptase while showing insignificant inhibition towards other enzymes tested. SFL-BFW-10 and SFL-WEF-7 inhibited reverse transcriptase by almost 100 percent and the IC50 values calculated for these compounds were 0.34 and 0.23mg/ml, respectively. Cytotoxicity of these compounds was evaluated on Chang liver cells and peripheral blood mononuclear cells (PBMCs). None of these compounds showed any significant inhibition of cell proliferation. The purity of these compounds could not be confirmed because there was insufficient material to use in the techniques required to show purity and identification. Therefore, TLC was used to determine the nature of these compounds. SFL-BFW-10 was identified as an organic acid and SFL-WEF-7 was identified as flavonoid.

Endozoochory in the subtropical thicket: comparing effects of species with different digestive systems on seed fate

This is especially true in the Subtropical Thicket biome where many plant species are adapted for endozoochory. Endozoochorous seed dispersal is carried out by a wide range of thicket fauna, but changes in the faunal assemblage associated with transformed thicket threatens to disrupt the process of endozoochorous seed dispersal and in turn the regeneration dynamics of thicket plants. Outside of conservation areas, indigenous seed dispersers are often replaced by domestic species, such as goats, which are hypothesized to differ in their seed dispersal efficiencies. The dispersal efficiencies of important thicket seed dispersers that occur in transformed and untransformed thicket were therefore investigated. The influence of gut morphologyphysiology on seed gut passage time, seed mortality and germination in animal species representing different digestive models was measured, as well as the role of black-backed jackal in thicket plant seed dispersal. Black-backed jackal were found to disperse the seeds of few thicket plant species and are therefore not considered important thicket seed dispersers under the climatic conditions prevailing during the study. The average gut passage time of seeds was found to differ significantly between birds and mammals, but not between elephants (hindgut fermentor), goats (ruminant) and pigs (omnivore, foregut fermentor) as was expected considering the differences in their digestive morphology. A significantly shorter gut passage of large seeds compared with small seeds was observed in goats, due to the regurgitation of larger seeds. Seed regurgitation in ruminants may prove to be an important factor contributing to their quality as seed dispersers in transformed thicket. The mortality of seeds was significantly higher in goats than in elephants, probably due to the efficient digestion associated with ruminants. The effect of gut passage on seed germination was not consistent and varied between animal species and across different thicket plant species. It was concluded that seed dispersers that occur in transformed and untransformed thicket differ in their seed dispersal efficiency due, in part, to differences in the nature of their digestive tracts. This has important implications for the process of seed dispersal in thicket, and the rehabilitation of degraded thicket vegetation.

- Description: This thesis presents a history of the landscape of Nqabara, an administrative area in a rural and coastal area of the Eastern Cape Province, South Africa. In the process of investigating landscape history, the inquiry engages with a range of data sources from diverging discursive contexts, including data from ethnographic fieldwork, from the consultation of archival documents and historical reports as well as from comparative historic and ethnographic research, necessitating a critical consideration of the epistemological contexts of data production and the dialogue between researcher and data. Furthermore, in its aim to move beyond historical description towards explanation, the study interrogates the dualist ontological conceptualisations of nature and culture, society and ecology, object and meaning upon which are built three dominant conceptual frameworks concerned with human-environment relationships: social-ecological systems theory, transdisciplinary landscape studies and political ecology. Drawing primarily upon the works of James Gibson, Anthony Giddens and Tim Ingold, an ontological foundation is developed to guide the enquiry and move towards an alternative understanding of the relationship of people’s livelihoods with respect to the landscape in which it is lived, which I call here the praxisembodiment perspective. This ontology takes the situated patterns of action of a situated agent-in-its-environment as its point of departure and proceeds to develop a framework explaining how relations among the patterns of action of different agents-in-their-environment, emerge in structures that simultaneously enable and constrain future action. The foundation is thereby provided for a monist understanding of how landscape and social structure emerge simultaneously from the complex intersection of patterns of actions and interactions of agents in their environment. This framework calls for an understanding of time, space and scale, not as independent variables influencing process and action, but as emergent properties arising from the patterns of actions of situated agents. Finally the alternative ontology is applied to the history of landscape and livelihoods of people of Nqabara. It is concluded that an appropriate understanding and explanation of the critical transformations in the landscape as well as in social institutions, should be sought through analysis of the complex ways in which patterns of action of multiple spatial and temporal rhythms and between multiple agents in an environment, intersect and resonate.

Pollinator mediated selection in Pelargonium reniforme Curtis (Geraniaceae) : patterns and process

- Description: Pelargonium reniforme is currently divided into two subspecies, P. reniforme subsp. reniforme and P. reniforme subsp. velutinum. The species falls into section Reniformia along with the closely-related P. sidoides. Observations of the section showed some discrepancies in the current taxonomy; mainly floral variation that was not recorded in the descriptions of the subspecies of P. reniforme, particularly the differences in hypanthium lengths. Patterns of variability were analysed using both morphometrics and inter simple sequence repeat (ISSR) data for P. reniforme (both subspecies), and P. sidoides. Results showed no support for the current subspecific division of P. reniforme but also no support for the current status of P. sidoides as a separate species. However, both morphometric and ISSR data show some evidence for the existence of two distinctly separate groups within P. reniforl1le subsp. velutinum as two distinct types have been recognized: those with bright pink flowers with long tubes (up to 70mm), and those with pale pink flowers with shorter tubes (as little as 8mm). These two forms have been found in a single population (Grahamstown), where they occur sympatrically, as well as in monomorphic populations. Analyses of the processes thought to be responsible for the observed patterns were conducted on the Grahamstown population. Observations of pollinators suggest that long-tongued insects (Butterflies, Nemestrinid flies) pollinate the long-tubed flowers and short-tongued insects (Bombyliid flies, long-tongued bees) pollinate short-tubed flowers. However, analysis of selection gradients in the population, indicate no directional selection is occurring for hypanthium lengths. The results of this thesis show that selection is occurring within Pelargonium reniforme, but at this time, selection is not strongly directional and floral differences are maintained, even when floral forms occur sympatrically.
