Assay techniques have been developed which permitted assay of individual grains. Thus during the investigation both groups and individual grains were used as samples. The use of groups permitted control of assay technique. Length of radicle, coleoptile and lateral roots were recorded in order to permit quantitative estimation of correlation between growth and the concentration of the various carbohydrates. Initially, during the study of changes in the carbohydrate concentration in Hickory King grains, total reducing sugar, sucrose and dextrin concentrations were estimated. However, the results obtained for changes in dextrin concentration, although reproduceable, thereby indicating reliable assay technique, presented a confusing picture and, in view of the apparent importance of sucrose and reducing sugar concentration, assay of dextrin concentration was discontinued in a subsequent study of Early Pearl. Instead changes in total reducing sugar, sucrose and glucose concentrations were followed. The results revealed that there is very considerable variability in physiological activity between grains subjected to the same germination conditions. However, all, irrespective of variety, follow the same basic metabolic pattern during germination. The trends observed were: (i) Reducing sugar accumulates slowly during the first 72-96 hours germination, but thereafter accumulation is very rapid, although concentration may decrease towards the end of the gestation period. (ii) Glucose follows a similar pattern to reducing sugar, accumulating slowly during the early stages of germination, followed by a period of rapid increase in concentration, which may decrease towards the end of the germination period. (iii) Sucrose concentration in dormant grains is fairly high, but it decreases markedly during the first 96 hours germination. This is followed by a phase of sucrose accumulation. (iv) Dextrin concentration shows two peaks. Initial level is low, but it accumulates rapidly during the first 72 hours. The level decreases between 72 and 120 hours but increases when the germination period is increased to 192 hours, after which there is a marked decrease. It was impossible, from the data relating to the study of individual grains, to discern a trend in dextrin concentration. With the exception of dextrin, about which there is little information, the results are in general agreement with the literature. Investigation of correlation between the various carbohydrates and between these and growth revealed that: (i) reducing sugar concentration and growth are positively correlated ; (ii) glucose concentration and growth are positively correlated; (iii) sucrose and reducing sugar concentrctions are negatively correlated during the initial stages of germination; (iv) sucrose and glucose concentrations are negatively correlated during early germination; (v) glucose and reducing sugar are positively correlated; (vi) in general, correlation between growth and concentration of the carbohydrates studied, decreases during the later periods of germination. These observations suggested that growth was, at least during the early stages of germination, dependent on the level of reducing sugar, and more particularly on the level of glucose, and that sucrose is the principal source of reducing sugar during this period. The relationship between amylase activity (total alpha- and beta-amylase activity) and reducing sugar concentration tends to be curvilinear, which suggests that amylolytic activity produces relatively little reducing sugar during early gennination, even though amylase activity and growth may be positively correlated. The results suggest, contrary to the observations of previous workers, that alpha-amylolytic activity may be present in dormant grains and that maize is not characterised by low levels of beta-amylase activity during germination. From the observations it is concluded that the initinl accumulati on of reducing sugar is the result of sucrose hydrolysis, and therefore sucrose is an importnnt metabolite durjng early germination. Amylolytic activity contributes little reducing sugar durlng the initia1 stages of germinatIon but that after approximately 72 hours it represents the major source of reducing sugar.

De Bary (1887) recognised the fruiting bodies of the Ascomycetes as 'compound sporophores' made up of interwoven hyphae or of pseudoparenchyma consisting of a peripheral layer separate from an inner tissue. These fruiting bodies included the discocarp or apothecium, the pyrenocarp or perithecium and the cleistocarp or cleistothecium. The name, Pyrenomycetes, has been variously applied to a group of Ascomycetes but mainly to the perithecial types, inclusive of the true perithecia and the loculate forms. The perithecium itself was described as a 'cup-shaped discomycetous' sporocarp with margins incurved to form a pyriform structure (de Bary, 1887). The presence or absence of a stroma had been long considered as a basis for the separation of the major groups of the Ascomycetes. Separation on these grounds was found unacceptable as it grouped together clearly unrelated species or separated related ones. As the subdivisions of the pyrenomycetous Ascomycetes are dependent upon the morphological features of the ascocarp, it would be pertinent to discuss the following: a) The Stroma. b) The various forms of ascocarp which mayor may not be associated with such a stroma. i) Perithecia: free or immersed in a stroma. ii) Uni- or multiloculate stromata. c) The centrum, the details associated with the centrum and the use of these details in the taxonomy of the pyrenomycetes.

- Description: That certain bacteria in the rumen of sheep and cattle are attached to solid particles in the ruminal ingesta has been known for many years. In 1942 Baker published direct microscopical evidence that bacteria were attached to cellulose food particles and to starch granules in the rumen. The sites of attachment of these bacteria corresponded to sites of disintegration of the particles when viewed by polarised light. This indicated that at least bacteria attacking solid substrates such as cellulose and starch were attached to particles of ruminal ingesta. Van der Wath (1942) found rumen bacteria attached to particles of chemically pure cellulose and of crushed maize which he suspended in separate compartments of a pure silk bag inside the rumen of sheep. The bacteria associated with the particles of cellulose were mainly Gram negative rods , while clusters of iodophilic cocci were observed in most instances around the maize kernels . The latter organisms were isolated in pure culture and found to be heat-tolerant, short-chain, Gram positive cocci fermenting glucose, maltose, and other soluble sugars as well as starch. It was thus not surprising that many years later Schwartz et al (1964) obtained evidence which suggested that bacteria metabolising soluble substrates such as glucose also showed marked attachment to solid particles of ingesta.

It is the most distant area from the equator in which pineapples are grown commercially, most other areas lying between latitudes 25°N and 25°S (Collins, 1960). The prevailing climatic conditions may be considered as being adverse to the growth of a plant which originated in the tropics. Girton, (1962) considers the area to be unsuitable for commercial production because of the cold weather experienced during winter. Seasonal fluctuations in growth and nutrient levels of the plant have been referred to by van Lelyveld, (1964), but these have never been studied locally. Because of the profound effect of temperature on the growth and nutrient requirements of any plant, it was deemed necessary to investigate the effects of the cooler conditions on the growth and nutrient levels of the pineapple plant, the results of which may give some indications of seasonal nutrient requirements.

- Description: This report contains the results of some ecological and systematic studies on the diatoms from the Vaal Dam catchment area in the Transvaal, and the Bloukrans River in the Eastern Cape Province. In Part 1 the effects of high concentrations of nitrogen were studied in relation to the composition of the diatom associations. Water samples from four stations on the Bloukrans River were analysed chemically at certain intervals during the months of April and August 1967. Diatom samples collected from these stations at the beginning and end of each of these sampling periods were subjected to a "Thomasson Analysis" to determine the relative densities of the various species in the diatom associations. A statistical analysis of the results reflected a poor but positive correlation between the two variables, i.e. high numbers of nitrogen heterotrophic Nitzschiae were correlated with high concentrations of nitrogen, while low numbers were correlated with low concentrations. Part 2 presents the results of the ecological studies on the diatom associations of the Vaal Dam Catchment Area. In this section the diatom associations from each sampling point or station were subjected to a "Thomasson Analysis" to determine the relative densities of the different species in the associations. All species described as new or having interesting features are illustrated in the plates.

During the course of this study 20 species have been described as new to science; the descriptions of some have been published, while the descriptions of the others will be published formally in the future. All species described as new or having interesting features are illustrated in the plates.

The effect of varying heavy metal balances in the nutrient medium, on the growth and development of Aspergillus Sp. and Penicillium Sp.

- Description: The study was conducted to investigate the effects of: (i) varying the level of supply of the heavy metals iron, manganese, copper and zinc. (ii) varying the ratio between different pairs of metals in the medium. In particular the iron:manganese and zinc:copper ratios were studied. Initially the two fungi Aspergillus niger van Tieghem, (variety and strain) and Penicillium notatum Westling, were used. Penicillium notatum Westling was subsequently discarded, in favour of Penicillium glancum Link, because it did not sporulate freely in liquid culture. The fungi were grown in controlled nutrient solutions, and during the course of tho growth and development, the form and sporulation of the felts was noted. After a period of growth, the felts were removed, dried and weighed. The pH of the liquor was measured. the results were studied to determine the effect of varying levels of supply of the heavy metals, and of the varying heavy metal ratios in the culture solution. In the investigation of the effect of varying the level of supply of individual heavy metals, optimum concentrations were demonstrated for copper and manganese. Increaning the concentration of pairs of heavy metals cimultaneously was found to influence the appearance and degree of symptoms of toxicity. Cultures of Aspergillus and Penicillium were found to be able to tolerate concentrations of copper, in particular, considerably greater than the observed optima, when zinc was present in equal concentration. Citric acid, and subsequently ethylene- diaminetetra-acetic acid, were used as chelating agents, in order to prevent the precipitation of the metals in the culture solution during autoclaving. It was found that the use of chelating agents markedly reduced symptoms of toxicity. There was no conclusive evidence that the iron:manganese ratio in the culture medium was an important factor in the growth and development of cultures of Aspergillus and Penicillium. However there is considerable evidence that in cultures of Penicillium, the zinc:copper ratio in the medium is of some importance in the determination of the dry weight yield trends. This effect was not demonstrated in cultures of Aspergillus.

Grazing sequence pattern and species selection by cattle in the Dohne sourveld

- Description: A detailed investigation into the harvesting of the standing grass crop within camps by cattle showed the following: 1. Cattle have a definite order of preference for the components of the Dohne Sourveld grass sward. The most palatable species in order of preference (based on Corrected Species Importance rating) are Themeda triandra, Tristachya hispida, Andropogon appendiculatus, Alloteropsis semialata and Heteropogon contortus. The least preferred species is Elionurus argenteus. 2. The key species, which is defined as the most abundant and productive palatable species, has been identified as Themeda triandra. 3. In harvesting the grass crop, cattle not only have a set preference for species, but also have a set grazing sequence pattern. This can be divided into three main phases. On entering the camp the animals first select the species of their choice, "creaming" these plants (grazed less than 50%) to obtain the most nutritious feed. In their daily search for food they spread their grazing over an increasingly wider area and over a wider range of species. When they can no longer obtain their daily requirements from this "creaming" operation of the more preferred species they return to the most preferred species , which are then heavily grazed (more than 50%) producing the common pattern of area grazing within a camp. These areas increase in size as the cattle graze down the species of their choice, moving to less preferred species as soon as they can no longer obtain their daily intake requirements from the more preferred species. Finally when they have no alternative they graze the least preferred species. 4. Increasing the stocking density does not alter the grazing sequence pattern or change the order of preference for the component species in the sward. It only increases the rate of utilization of the more preferred species and the various phases in the grazing sequence pattern are reached more rapidly. 5. At higher stocking densities the palatable species are more evenly utilized than at lower stocking densities. 6. Cattle graze a greater variety of species when the sward is young and actively growing. As the maturity of the sward increases the cattle concentrate on the more preferred species and are more selective in their grazing. However, their order of preference remains the same. Species that are highly preferred early in the season retain their ranking when the sward is mature. The only exception to this rule is Alloteropsis semialata. Early in the growing season this species is high on the preference list, but as it matures less is taken until when fully mature, it is rejected by cattle. 7. Elionurus argenteus is the least preferred of all the species in the Dohne Sourveld. Cattle will graze this plant as a last resort when grazing a camp early in the season, but reject the plant when it is fully mature. 8. To retain vigour and production of the Dohne Sourveld results of this investigation support the view that grazing management should be based on the principle of controlled selective grazing. Having shown that the more preferred species are over-utilized when attempting to utilize the least preferred species any grazing management that calls for total utilization of all standing vegetation should be rejected. 9. The results of this investigation supports a multicamp system for the Dohne Sourveld where high stocking densities are employed. This will result in the rapid and even utilization of the more preferred species. Management should be based on short period of stay and long period of absence. Cattle should be moved as soon as the key species have been well utilized, but not overgrazed. Cattle should only be returned to the camp when key species have made adequate regrowth following grazing. 10. A method, that is simple to use, is proposed for determining utilization of the veld and key species at different times of the year using the relationship between percentage of ungrazed plants and utilization of the sward and of Themeda triandra, the key grazing species in the sward. 11. Results show that when rested camps are grazed after May, the number of Themeda triandra inflorescences are drastically reduced. Veld that is specifically rested for the production of Themeda triandra seed should be rested from March of the year preceding expected seeding.
