Quantifying variability of emissions of greenhouse gas (CO2& CH4) across selected soils and agricultural practices

- Description: Increasing concentrations of greenhouse gases (GHG’s)in the atmosphere are warming the planet, and agriculture is responsible for about 30% of these emissions. Soils act as a host for greenhouse gases, since both their storage and emission capacities are large, accounting for two-times the amount of carbon in the atmosphere and in plant and animal life. It sequesters large amounts of carbon, and because agricultural practices depend on soil for production, the practices influence the soil’s ability to store the carbon effectively. Production soils emit greenhouse gas, predominantly carbon dioxide and methane, which are assessed for emissions in this study. Climate change creates unpredictability in precipitation and temperature; farmers need to be flexible and adapt production methods to such environmental changes in order to continue producing sustainably. Global food production needs to grow drastically to meet the projected demands for rising population and diet shifts; studies have shown that feeding a more populated and a more affluent, equal, world will require roughly a doubling of agricultural production by 2050, which means more GHG emissions from the soil. To enable better control on these emissions, their links to agricultural practices need to be better quantified. The study was done in two areas: (1) long-term comparative farming systems research trial with controlled vegetable plots, in the agricultural school of Nelson Mandela University, in George, Western Cape province and (2) long-term wheat research trial of the Free State University, in Bethlehem, Free State province. The objective in study area one is to assess and compare GHG emissions from conventional and organic systems. Temperature and soil moisture were measured during gas samples to establish the influence they have on gas emissions. The objective in study area two is to assess and compare GHG emissions from no-till, plough, and stubble mulch. Stubble mulch refers to crop residue left in place on the land as a surface cover during fallow periods. Two polypropylene canisters are placed in a sampled plot to trap gas emitted from the soil. Analyses of the trapped gases in the headspace gives concentrations of CO2and CH4that was emitted during the duration the canister was closed. The gas is analysed by a G2201-i Picarro gas analyser, presently the only such instrument in South Africa. The analyser’s near-infrared Cavity Ring Down Spectroscopy technology is capable of simultaneous measurements of CO2and CH4down to parts per million. In study area one, conventional plots (R2T6 & R1T6) emitted 65.089ppm CO2and 61.159ppm CO2, and 0.0010ppm CH4and 0.0004ppm CH4, respectively. Organic plots (R1T3 & R2T3) emitted 53.264ppm CO2and 47.885ppm CO2, and 0.0023ppm CH4and 0.0019ppm CH4respectively. Thus, conventional plots emitted 19.98% & 30.98% more CO2than organic plots; but organic plots emitted 81.97% & 155.5% more CH4thanconventional plots. In study area two, ploughed soils emitted 38.727ppm CO2and 0.015ppm CH4, no-tilled soils emitted 31.798ppm CO2 and 0.011ppm CH4andstubble mulched soils emitted 28.373ppm CO2 and 0.009ppm CH4. Thus, ploughed soils emitted 19.65%more CO2than no-tilled soils, no-tilled soils emitted 11.38% more CO2 than stubble mulched soils and ploughed soils emitted 30.36% more CO2 than stubble mulched soils. Ploughed soils emitted 30.77% more CH4 than no-tilled soils, no-tilled soils emitted 20% more CH4 than stubble mulched soils and ploughed soils emitted 50% more CH4than stubble mulched soils. Moist soils result in decreased CO2emissions in conventional plots and increased CH4emissions in organic plots. Increasing temperature patterns are followed by a trend of increasing gas emissions. Reducing GHG emissions from agriculture and developing sustainable tillage practices can help mitigate climate change and increases the chances of stabilizing GHG concentrations and temperature control within a required range. , Thesis (MSc) -- Faculty of Science, School of Environmental Sciences, 2021

Conceptualisations and pedagogical practices of academic literacy in Namibian higher education

- Description: The purpose of this study was to investigate academic literacy development lecturers’ conceptualisations of academic literacy and resultant pedagogical practices in academic development courses at three different Higher Education Institutional types in Namibia. The research sites were a Traditional University, a University of Technology and a Comprehensive University. The focus was to understand the extent to which the academics’ conceptions of academic literacy and the resultant pedagogical practices in the academic development courses at these three Higher Education Institutions (HEIs) facilitate epistemological access into students’ chosen fields of study. Bernstein’s Pedagogical theory (1990), Genre theory (1996) and Halliday’s Systemic Functional Linguistics (1978) were used as the study’s theoretical lenses and analytical framework. An interpretative paradigm and a qualitative case study design were employed as the research approach. Semi-structured interviews, classroom observations and documentary evidence were used to generate data. Research findings revealed a common (mis)conception of the nature of academic literacy, the resultant inadequate learning support offered to students in the selected academic literacy development courses, and a clear divorcing of academic literacy interventions from the students’ ‘home’ or mainstream disciplines at the three HEIs. The participants understood academic literacy from an autonomous position as a set of generic skills which could be taught outside of mainstream classes. Moreover, findings revealed that this understanding impacted on the design and assessments of all the academic literacy courses across the three universities under study. An investigation into yeast-baculovirus synergism for the improved control of Thaumatotibia leucotreta, an economically important pest of citrus

- Description: A mutualistic association between Cydia pomonella and yeasts belonging to the genus Metschnikowia has previously been demonstrated. Larval feeding galleries inoculated with M. andauensis, reduced larval mortality and enhanced larval development. Additionally, adult C. pomonella female oviposition preference was also shown to be influenced by the volatiles produced by M. andauensis. This mutualistic relationship was manipulated for biological control purposes, by combining M. pulcherrima with the baculovirus Cydia pomonella granulovirus. The combination of M. pulcherrima with brown cane sugar and CpGV in laboratory assays and field trials resulted in a significant increase in larval mortality. A similar observation was made when M. pulcherrima was substituted for Saccharomyces cerevisiae. This indicates that yeasts harbour the potential for use in biological control, especially when combined with other well-established biocontrol methods. Thaumatotibia leucotreta is a phytophagous insect endemic to southern Africa. It is highly significant to the South African citrus industry due to its classification as a phytosanitary pest by most international markets. An integrated pest management programme has been implemented to control T. leucotreta. The baculovirus Cryptophlebia leucotreta granulovirus forms one component of this programme and is highly effective. In this study, we proposed to determine which yeast species occur naturally in the gut of T. leucotreta larvae and to examine whether any of the isolated yeast species, when combined with the CrleGV-SA, enhance its effectiveness. Firstly, Navel oranges infested with T. leucotreta larvae were collected from geographically distinct citrus-producing regions across South Africa. This led to the isolation and identification of six yeast species from the gut of T. leucotreta larvae via PCR amplification and sequencing of the internal transcribed spacer region and D1/D2 domain of the large subunit. Six yeast species were identified, viz. Meyerozyma guilliermondii, Hanseniaspora uvarum, Clavispora lusitaniae, Kluyveromyces marxianus, Pichia kudriavzevii and Pichia kluyveri. Additionally, Saccharomyces cerevisiae was included as a control in all trials due to its commercial availability and use in the artificial diet used to rear T. leucotreta. Secondly, larval development and attraction assays were conducted with the isolated yeast species. Thaumatotibia leucotreta larvae that fed on Navel oranges inoculated with M. guilliermondii, P. kluyveri, H. uvarum, and S. cerevisiae had accelerated developmental periods and reduced mortality rates. Additionally, it was demonstrated that T. leucotreta neonates were attracted to YPD broth cultures inoculated with P. kluyveri, H. uvarum, P. kudriavzevii and K. marxianus for feeding. Thirdly, oviposition preference assays were conducted with adult T. leucotreta females to determine whether the isolated yeast species influence their egg-laying in two-choice and multiple-choice tests. Navel oranges were inoculated with a specific yeast isolate, and mated adult females were left to oviposit. Meyerozyma guilliermondii, P. kudriavzevii and H. uvarum were shown to influence adult T. leucotreta female oviposition preference in two-choice tests. However, multiple-choice tests using the aforementioned yeast species did not mimic these results. Lastly, a series of detached fruit bioassays were performed to determine the optimal yeast:virus ratio, test all isolated yeast species in combination with CrleGV-SA and to further enhance yeast/virus formulation through the addition of an adjuvant and surfactant. CrleGV-SA was applied at a lethal concentration that would kill 50 % of T. leucotreta larvae. The optimal yeast concentration to use alongside CrleGV-SA was determined. Pichia kluyveri, P. kudriavzevii, K. marxianus and S. cerevisiae in combination with CrleGV-SA increased larval mortality compared to CrleGV-SA alone. The inclusion of molasses and BREAK-THRU® S 240 to P. kudriavzevii and S. cerevisiae plus CrleGV-SA formulations greatly enhanced their efficacy. Additionally, semi-field trials were initiated using P. kudriavzevii and S. cerevisiae, with promising preliminary results being obtained, although more replicates need to be performed. In this study, we proposed to determine which yeast species occur naturally in the gut of T. leucotreta larvae and to examine whether any of the isolated yeast species, when combined with the CrleGV-SA, enhance its effectiveness. Firstly, Navel oranges infested with T. leucotreta larvae were collected from geographically distinct citrus-producing regions across South Africa. This led to the isolation and identification of six yeast species from the gut of T. leucotreta larvae via PCR amplification and sequencing of the internal transcribed spacer region and D1/D2 domain of the large subunit. Six yeast species were identified, viz. Meyerozyma guilliermondii, Hanseniaspora uvarum, Clavispora lusitaniae, Kluyveromyces marxianus, Pichia kudriavzevii and Pichia kluyveri. Additionally, Saccharomyces cerevisiae was included as a control in all trials due to its commercial availability and use in the artificial diet used to rear T. leucotreta. Secondly, larval development and attraction assays were conducted with the isolated yeast species. Thaumatotibia leucotreta larvae that fed on Navel oranges inoculated with M. guilliermondii, P. kluyveri, H. uvarum, and S. cerevisiae had accelerated developmental periods and reduced mortality rates. Additionally, it was demonstrated that T. leucotreta neonates were attracted to YPD broth cultures inoculated with P. kluyveri, H. uvarum, P. kudriavzevii and K. marxianus for feeding. Thirdly, oviposition preference assays were conducted with adult T. leucotreta females to determine whether the isolated yeast species influence their egg-laying in two-choice and multiple-choice tests. Navel oranges were inoculated with a specific yeast isolate, and mated adult females were left to oviposit. Meyerozyma guilliermondii, P. kudriavzevii and H. uvarum were shown to influence adult T. leucotreta female oviposition preference in two-choice tests. However, multiple-choice tests using the aforementioned yeast species did not mimic these results. Lastly, a series of detached fruit bioassays were performed to determine the optimal yeast:virus ratio, test all isolated yeast species in combination with CrleGV-SA and to further enhance yeast/virus formulation through the addition of an adjuvant and surfactant. CrleGV-SA was applied at a lethal concentration that would kill 50 % of T. leucotreta larvae. The optimal yeast concentration to use alongside CrleGV-SA was determined. Pichia kluyveri, P. kudriavzevii, K. marxianus and S. cerevisiae in combination with CrleGV-SA increased larval mortality compared to CrleGV-SA alone. The inclusion of molasses and BREAK-THRU® S 240 to P. kudriavzevii and S. cerevisiae plus CrleGV-SA formulations greatly enhanced their efficacy. Additionally, semi-field trials were initiated using P. kudriavzevii and S. cerevisiae, with promising preliminary results being obtained, although more replicates need to be performed. The study used a descriptive survey research design. The sampling method used for this study was convenience sampling. A questionnaire was used for data collection. The researcher cleansed and coded the data, whilst a statistician entered the collected data into STATISTICA. The data was analysed descriptively and also by means of inferential analysis method that is. The descriptive data was presented in the format of pie charts and tables with percentages, means and standard deviations as measures for distribution mechanisms. While inferential analysis presented the regression in table formats but with a clear indication of significant levels and adjusted R-square for all the independent variables. The sample that participated in this study consisted of 288 respondents from the IDC. Empirical findings revealed that the working environment, work-life balance, compensation and employee engagement positively influenced employee retention. Career development was not significantly related to employee retention. The study concluded and made recommendations to four stakeholders, namely the government to have roadshows and marketing activities to promote the Skills Development Act and skills Levies Act, IDC to review their employee retention strategy, Managers or Supervisors to support their employees by providing market related salary packages and provide session where they engage the employees in the organisations developments, and the HR Practitioners must ensure that the right fit of candidates are recruited to the organisation. , Thesis (MBA) -- Faculty of Business and Economic Sciences , Business Administration, 2021

In South Africa more broadly there is a lack of information on levels of physical activity and sedentary behaviour as well as physical fitness measures particularly in children. Objective: The purpose of this study was to enhance the existing body of knowledge on this, by measuring the prevalence of physical inactivity, sedentary behaviour and screen time of primary school children in a private school from the Makana Municipal region in the Eastern Cape Province of South Africa, where the evidence is most sparse. A secondary aim was to assess levels of physical fitness in this cohort, as well as to compare across grades and sexes. Method: After ethical approval was granted, stature and body mass measurements were taken and body mass index was computed for primary school children (Grade one to seven) from a private school in the Makana Municipality. Sitting stature was measured in order to determine each child’s peak height velocity and maturity offset. Five different physical fitness tests were conducted on the children and blood pressure and heart rate values were measured at rest. Physical activity and screen time questionnaires were completed by the children’s parents for a seven day period. Results: Females were found to mature faster than males. In addition females were more physically active (Females= 916 min/week; Males= 669 min/week) while also accumulating more sedentary time (Females= 1241 min/week; Males= 970 min/week) as compared to males, although no significant differences were found between males and females for these variables. The sample participated in more than 60 minutes of moderate-to-vigorous physical activity and less than two hours of screen time daily, which is in accordance with guidelines. No differences were found between grades and sexes for accumulated screen time. Strength increased significantly with grade and age, and there were also statistical differences between the sexes for all three strength measures with males being stronger than females. Females were more flexible than males and no statistical differences were found between sexes for the shuttle run test. Blood pressure correlated positively with mass, as did all the strength measures as well as maturity offset. No correlations were found between screen time and all other variables. A negative relationship was recorded between screen time and physical activity, although not significant. For females a strong positive correlation was found between sedentary behaviour and physical activity. Conclusion: The study was successful in adding to the body of knowledge regarding the prevalence of physical inactivity, sedentary behaviour and screen time within school children from the Makana Municipality in the Eastern Cape Province of South Africa. The sample was considered physically active as they accumulated on average more than 60 minutes of moderate-to-vigorous physical activity daily (Males= 96 min/day; Females 130 min/day), which is in accordance with guidelines recommended by the World Health Organization (World Health Organization, 2018). For example, many corporations go to great lengths to present themselves as being environmentally friendly when in fact they are not. Many corporations have also been forced to recall dangerous products which they at one time or another insisted were safe. Research teams have misled participants with regard to what they can expect from their participation in studies, with grave consequences. Governments throughout the world are mired in corruption, and yet deny that this is so. One possible explanation is that in such situations these institutions are simply lying. However, another possible explanation is that these institutions are self-deceived, or lying to themselves. Recently it has been suggested that self-deception is something that affects certain groups as well as individuals. Given that institutions can wield a great deal of political, social and economic power, if institutions are capable of self-deception there is room for things to go awry on a very large scale with potentially dire consequences. Yet the explanations currently on offer for group-level self-deception appear to amount to instances of individual self-deception (either to certain key individual members of those groups being self-deceived, or to all or most members of a group sharing the same self-deceptive belief), and as such I do not regard the explanations currently on offer as satisfactory. I propose that there are certain situations in which we ought to see institutions themselves as self-deceived or lying to themselves. While the terms ‘self-deception’ and ‘lying to oneself’ are often used interchangeably, I differentiate between the two and argue that both institutional self-deception and an institution lying to itself are institution-level phenomena, and do not rely on any individual within the institution being self-deceived or lying to themselves. - Description: The COVID-19 virus showed the world how under-prepared it was to cope with a pandemic. It also raised an awareness in the world that a highly communicable disease can begin anywhere, and without proper monitoring and reporting can spread to become a global pandemic. The South African Health Department has been under significant stress and on the brink of collapse during the successive waves of the pandemic due to the lack of healthcare workers, medical doctors, infectious disease specialists and epidemiologists as well inadequate medical and clinical infrastructure. The infectious disease specialty in South Africa is also in crisis, prompting the Infectious Disease Society of South Africa to plead with the Health Department and universities to fund projects that support learning and research into infectious diseases, virology, and epidemiology. This project proposes the design of an Infectious Disease Centre of Excellence for the Study of Infectious Diseases at Livingstone Hospital, Gqeberha which focuses on the research, teaching and learning, and healing of infectious diseases in a ho listic building design incorporating biosafety research laboratories, an infectious disease ward and a centre of excellence. The methodology used was to research existing precedent studies that are focused on the research, teaching and learning, and healing of infectious diseases as well as researching a theoretical framework based on holism and systemic architec ture. This theoretical framework, coupled with a careful analysis of the relation ship of each highly specialised space with the other space components leads to the design of an Infectious Disease Centre of Excellence for the Study of Infectious Diseases which responds to the urgent need expressed above as well as providing a design which promotes the well-being of all users. - Description: The COVID-19 virus showed the world how under-prepared it was to cope with a pandemic. It also raised an awareness in the world that a highly communicable disease can begin anywhere, and without proper monitoring and reporting can spread to become a global pandemic. The South African Health Department has been under significant stress and on the brink of collapse during the successive waves of the pandemic due to the lack of healthcare workers, medical doctors, infectious disease specialists and epidemiologists as well inadequate medical and clinical infrastructure. The infectious disease specialty in South Africa is also in crisis, prompting the Infectious Disease Society of South Africa to plead with the Health Department and universities to fund projects that support learning and research into infectious diseases, virology, and epidemiology. This project proposes the design of an Infectious Disease Centre of Excellence for the Study of Infectious Diseases at Livingstone Hospital, Gqeberha which focuses on the research, teaching and learning, and healing of infectious diseases in a ho listic building design incorporating biosafety research laboratories, an infectious disease ward and a centre of excellence. The methodology used was to research existing precedent studies that are focused on the research, teaching and learning, and healing of infectious diseases as well as researching a theoretical framework based on holism and systemic architec ture. This theoretical framework, coupled with a careful analysis of the relation ship of each highly specialised space with the other space components leads to the design of an Infectious Disease Centre of Excellence for the Study of Infectious Diseases which responds to the urgent need expressed above as well as providing a design which promotes the well-being of all users. The facility also enhances the Livingstone Hospital precinct. - Description: This thesis explores the possibility of marrying repressed African indigenous knowledge systems with the abrasive, post- colonial environment that is Central, such that an African identity is represented in a built environment that was shaped and formed by European influences. This is aligned with the African renaissance paradigm. Set in the backdrop of Central, Port Elizabeth’s disparaged CBD, the study culminates in a successful and appropriate architectural design for an Afro- indigenous pharmacy and a vernacular architecture for Port Elizabeth. The design process will take cues from the social, economic, political, physical and urban CBD context of the chosen site, such that the final proposal is practical and usable by the public for which it is intended, while simultaneously being environmentally sensitive. Furthermore, the research aims to shed light upon the importance of urban regeneration through the repurposing of an abandoned Brownfield site into a productive landscape and a sustainable enterprise. The qualitative methodological approach is followed to gather primary data, which is obtained by visiting existing herbalist practices and conventional pharmacies in Port Elizabeth, and will be used as case studies from which interpretations and information will be drawn. The effect of limestone ammonium nitrate and chicken manure fertilizers on growth, yield and quality of two cabbage cultivators

- Description: Cabbage is a well-known and popular crop in South Africa. As a highly nutritious leafy green vegetable, it is produced and sold to a variety of markets, from formal markets to hawkers, for human consumption and as animal feed. The cabbage crop contributes positively to food security, health, and households’ financial wellbeing. As a crop, cabbage is known to be a heavy feeder due to its high nutritional requirements (in terms of fertiliser) particularly nitrogen. This high requirement can be supplied by either synthetic or by organic fertilisers in the form of animal manures. However, fertiliser has proven to be a challenge due to its high costs, as its prices increase rapidly each year. The current study aimed to determine the most effective fertilisation method for the growth, quality, and yield of cabbage (Brassica oleracea var. capitata). Two fertiliser types (chicken manure and limestone ammonium nitrate) were applied, and the treatments were repeated five times over two planting sessions on two cultivars, Grandslam and Empowa, at concentrations of 0%, 100% and 150%. A control for each treatment / cultivar was also grown. The parameters plant height, head diameter, pests and disease development, quality and dry mass were measured to determine the effect on growth, yield, and quality of the cabbage crop. From each planting, a total of 60 cabbages served as specimens for data collection purposes. The research trial was conducted in open field and laid out in a split-plot block method. The cultivar was allocated to the main plot, while the fertiliser treatments served as the sub-plots. The study was carried out for a period of six months, which allowed for data collection to be done in two different growing conditions. Planting was done manually by hand in planting bags, and washed river sand was used as a substrate (growing medium). The data collected during the experiment was analysed by means of statistical data analysis software STATISTICA and the methods applied were a three-way analysis of variance (ANOVA). The analysis of variance was performed at a significance level of p as a means to determine a correlation of relationships for all set parameters (variables) for both cultivars, treatments and plantings. The results of the study showed that the two fertiliser types, namely limestone ammonium nitrate (LAN) and chicken manure, both had a significant effect (p on plant height, head diameter, dry mass. Also, the pest and disease development and quality of cabbage were significantly influenced by the fertiliser types and season (climate). The interactive effect of cultivar and fertiliser was statistically significant. Empowa showed better results on chicken manure compared to LAN. In the case of Grandslam, however, no significant fertiliser effect was observed. The study also showed that fertilisation with a fertiliser that contains high nitrogen (N) content promotes pest and disease incidence. Increasing N application rate resulted into an increase of pest and disease incidence. The change of season, which promotes different growth conditions (in terms of temperature, water requirements, pests, and diseases) for the plants was found to have had an influence on the study results. Lasty, it was observed that chicken manure fertiliser yields a cabbage with a good internal and external quality compared to limestone ammonium nitrate fertiliser.
