- Description: Tin(IV) complexes of a 4-methylthiophenyl functionalized porphyrin (1-Sn) and its corrole analogue (2-Sn) were synthesized so that their photophysicochemical properties and photodynamic activities against MCF-7 breast cancer cells could be compared. Singlet oxygen luminescence studies revealed that 1-Sn and 2-Sn have comparable ΦΔΦΔ values in DMF of 0.59 and 0.60, respectively, while the IC5050 values after irradiation of MCF-7 cells for 30 min with a Thorlabs 625 nm LED (432 J · cm−2)−2) were determined to be 12.4 and 8.9 μμM. The results demonstrate that the cellular uptake of 2-Sn and its molar absorptivity at the irradiation wavelength play a crucial role during in vitro cytotoxicity studies.

Structures of the isolated compounds were elucidated based on NMR spectroscopy and high resolution electron ionization mass spectrometry (HR-EI-MS) data. All compounds were tested against Trypanosoma brucei brucei and Plasmodium falciparum. At a single concentration (20 μM) in the antitrypanosomal and antiplasmodial assays, compound 4 exhibited remarkable activities against T. brucei brucei and P. falciparum with percentage inhibition of 97.3% and 83.0% respectively, whereas compounds 1, 2, 3 and 5 were inactive. In a dose response antiplasmodial assay compound 4 exhibited moderate activity against P. falciparum with an IC50 value of 7.20 μM.

This short communication reports technical details and preliminary results of a python-based tool (AMADAR) designed to generate any Diels–Alder (DA) transition state geometry (TS) and analyze determined IRC paths in a (quasi-)automated fashion, given the product SMILES. Two modules of the package are devoted to performing, from IRC paths, reaction force analyses (RFA) and atomic (fragment) decompositions of the reaction force F and reaction force constant κ. The performance of the protocol has been assessed using a dataset of 2000 DA cycloadducts retrieved from the ZINC database. The sequential location of the corresponding TSs was achieved with a success rate of 95%. RFA plots confrmed the reaction force constant κ to be a good indicator of the (non)synchronicity of the associated DA reactions. Moreover, the atomic decomposition of κ allows for the rationalization of the (a)synchronicity of each DA reaction in terms of contributions stemming from pairs of interacting atoms. The source code of the AMADAR tool is available on GitHub [CMCDD/AMADAR(github. com)] and can be used directly with minor customizations, mostly regarding the local working environment of the user.

Few studies have investigated the conservation contribution of extralimital white rhinos to the overall national herd. We aimed to determine whether the white rhinos introduced to a private game reserve in the Eastern Cape province have been successful from a reproductive perspective. We calculated inter-calving intervals, age at first calving, sex ratios of calves, and recruitment rates for white rhinos at a single site between 1992 and 2019. The average net annual population growth rate for the population was 10%, which is higher than the recommended 5% by the Biodiversity Management Plan for white rhinos. Trends in density-dependent parameters such as age at first calving and inter-calving intervals also indicated that the study population is still well below the density at which ecological constraints may manifest. We demonstrate that an extralimital white rhino population in the Eastern Cape can be successful from a reproductive perspective.

While this approach is helpful in software development and simulation, it is not easily adaptable to automotive design where the final model is constantly changing and being modified. Consequently, the aim of this study is to propose a method for conducting computational fluid dynamics and machine learning concurrently to accelerate the development process. The proposed method is used to design and improve the aerodynamic efficiency of an object. The approach focuses on developing, implementing, and comparing machine learning models capable of generating optimised three-dimensional objects with the required geometry to direct airflow paths required in applications such as pressure generation, as needed for both active and passive flow control. The study concludes that both decision tree regression and long short-term memory (LSTM) autoencoder models could be used to optimise the aerodynamic efficiency of solid bodies, but that the LSTM autoencoder performs better overall. An undesirable effect of the shape optimisation is an overall reduction in shape size as optimization increases.

Using Bernstein’s concepts of classification and his theories on recontextualisation, a content analysis study was carried out to compare the level of NRM integration within the Grade 10 Life Sciences syllabus, and a Grade 10 Life Sciences textbook. Results from the analysis of the syllabus showed that overall only 9% of all the knowledge statements analysed had a strong link to NRM and related issues. The highest percentage of such sentences was found in the Core Knowledge section of the syllabus (21%). For the textbook, only 8% of the analysed items had a strong link to NRM and related issues, with the highest percentage of such items occurring in the Suggested Activities section (16%). However, the level of NRM integration in both documents increased considerably when sentences that had only an implicit link to NRM and related issues were included. It was concluded that both documents provide ample opportunities for NRM learning, although the extent to which this occurs varies among their different sections. The recontextualising role of the Grade 10 Life Sciences textbook was reflected in its relatively higher level of NRM integration in the Suggested Activities category, and in the Glossary category. This study highlights the need for further strengthening of the position of NRM within the Grade 10 Life Sciences syllabus, and for more Bernstein-based research to inform South Africa’s curriculum reform initiatives in environmental education.

- Description: This special issue is the fourth decadal review of biological control of invasive alien plants (biocontrol of weeds) in South Africa, following those published in 1991, 1999 and 2011. Including this introduction, there are 24 papers covering the weed biocontrol programmes, or important developments in the science and practice, from the period 2011-2020. Seventy-two target weed species are covered, including 25 species on which projects were initiated during the past decade. Developments in regulations, mass-rearing and implementation, and community engagement are also reviewed. An updated catalogue of agents released, rejected and under consideration is presented and reflects the most recent methods of quantifying success in weed biocontrol. Key events over the last decade include the hosting of the XIV International Symposium on Biological Control of Weeds to celebrate 100 years of weed biocontrol in South Africa, as well as the establishment of the Centre for Biological Control at Rhodes University. The science and practice of weed biocontrol has expanded significantly in the past decade, with growth in the number of researchers and practitioners, increased funding, and an increased number of scholarly outputs. Unlike many other countries in the world, South Africa has largely avoided constraints due to restrictive and risk averse legislation and bureaucracy, and has continued to release new biocontrol agents at a similar rate to that in previous years. Much of the success of weed biocontrol in South Africa is due to the sustained and increasing support of the Natural Resource Management Programme of the Department of Forestry, Fisheries and the Environment (Working for Water Programme). However, gaps in funding, where no funds are available for months at a time, are a major concern as the weed biocontrol community loses human capital in these periods, and research programmes suffer significant set-backs. Weed biocontrol is an essential component of South Africa’s strategy to reduce the negative impacts of invasive alien plants and has contributed significantly towards the protection of the country’s ecosystems, indigenous biodiversity, water security, agricultural productivity, and society in general. If the trend of increasing support for weed biocontrol in South Africa continues, we can expect that the benefits for the country at large will increase substantially in the future.

- Description: This special issue is the fourth decadal review of biological control of invasive alien plants (biocontrol of weeds) in South Africa, following those published in 1991, 1999 and 2011. Including this introduction, there are 24 papers covering the weed biocontrol programmes, or important developments in the science and practice, from the period 2011-2020. Seventy-two target weed species are covered, including 25 species on which projects were initiated during the past decade. Developments in regulations, mass-rearing and implementation, and community engagement are also reviewed. An updated catalogue of agents released, rejected and under consideration is presented and reflects the most recent methods of quantifying success in weed biocontrol. Key events over the last decade include the hosting of the XIV International Symposium on Biological Control of Weeds to celebrate 100 years of weed biocontrol in South Africa, as well as the establishment of the Centre for Biological Control at Rhodes University. The science and practice of weed biocontrol has expanded significantly in the past decade, with growth in the number of researchers and practitioners, increased funding, and an increased number of scholarly outputs. Unlike many other countries in the world, South Africa has largely avoided constraints due to restrictive and risk averse legislation and bureaucracy, and has continued to release new biocontrol agents at a similar rate to that in previous years. Much of the success of weed biocontrol in South Africa is due to the sustained and increasing support of the Natural Resource Management Programme of the Department of Forestry, Fisheries and the Environment (Working for Water Programme). However, gaps in funding, where no funds are available for months at a time, are a major concern as the weed biocontrol community loses human capital in these periods, and research programmes suffer significant set-backs. The developing world poses some particular challenges in this regard, specifically in terms of the large number of local level subsistence resources users who are important de facto decision-makers. We examine four natural resource management case studies from South Africa that differ in spatial scale and complexity, ranging from a single village to a whole biome. We distil seven lessons to help guide development of social learning processes and organizations in similar situations relating to natural resource planning and management. The lessons pertain to: maintaining ‘key individuals’ within social learning processes; the role of researchers; the formulation of research questions that social learning processes require adaptive long-term funding and capacity support; that local resource users are key decision-makers in developing countries; some perspectives on knowledge; and the need to measure research success.

We examine this for Lantana camara invasion in southern India through assessment of the demand for and impacts of harvesting on this globally problematic IAS. We interviewed local artisans regarding their knowledge and quantities of Lantana used, along with forest surveys to estimate Lantana densities and size classes in harvested and unharvested sites, and lastly we undertook controlled cutting in moist and dry deciduous forests in both the wet and dry seasons to examine rate of regrowth and mortality. Over the entire study area the abundance of Lantana far outweighed local demand, but at small scales around villages, density and size classes were significantly reduced through harvesting. The controlled cutting experiment showed marked seasonal differences, with the most severe cutting intensity resulting in significant mortality when Lantana plants were cut in the wet season, but with limited effect when cut in the dry season. We conclude that promoting local use of IAS may be a feasible approach in controlling them and thereby limiting their impacts in forests.

The modified electrode was characterised using X-ray photoelectron spectroscopy and scanning electrochemical microscopy, and well as electrochemistry. The electrocatalytic ability of the modified electrode was tested using hydrazine as a sample analyte, and showed a significant 10-fold improvement in the detection of hydrazine compared to the surface without nanotubes, and comparable stability. The linear range for this surface was over 1.0 × 10−5 to 1.0 × 10−4 mol dm−3, with a limit of detection of 1.10 ± 0.06 μmol dm−3 and sensitivity of 15.61 μA mM−1.

- Description: Despite the well-known anti-mycobacterial actions of isoniazid (INH) and rifampicin (RIF), the clinical success of tuberculosis (TB) therapy requires prolonged administration of multiple drugs in high doses, which often result in frequent adverse effects and low patient adherence. Although liposomes are promising candidates for controlled delivery of anti-TB drug, the high cost of synthetic and highly purified natural lipids currently used in liposomal technology might preclude the universal application of therapeutic liposomes. This work aimed at evaluating the potential of a cost-effective lipid material, crude soybean lecithin (CL), to co-encapsulate RIF and INH for liposomal dual delivery. RIF was encapsulated in CL-liposomes with/without cholesterol using film hydration method, after which INH was incorporated using a freeze–thawing technique. Dynamic light scattering, differential scanning calorimetry, X-ray diffraction and dialysis were used for liposome characterization. Liposomes containing CL alone (CLL) exhibited 90%encapsulation efficiency for RIF and 59%for INH. The mean size and surface charge of CLL were 1114nm and –63mV, respectively. In addition, CLL showed a controlled release profile for the co-encapsulated drugs. CLL would be promising vehicles for macrophage-targeting drug delivery. The present findings demonstrate the feasibility of using CL for preparation of combination products for liposomal delivery.

In this study, Bacillus sp.-derived GH5 and GH26 endo-mannanases were comparatively analysed biochemically for their synergistic action with a commercial cellulase blend, CTec2, during pre-treated lignocellulose degradation. Substrate specificity and thermo-stability studies on mannan substrates showed that GH26 endo-mannanase was more catalytically active and stable than GH5. GH26 also exhibited higher binding affinity for mannan than GH5, while GH5 showed more affinity for lignocellulosic substrates than GH26. Applying the endo-mannanases in combination with CTec2 for lignocellulose degradation led to synergism with a 1.3-fold increase in reducing sugar release compared to when CTec2 was used alone. This study showed that using the activity of endo-mannanases displayed with model substrates is a poor predictor of their activity and synergism on complex lignocelluloses.
