- Description: The use of the yield curve in forecasting economic recessions is well established in the literature. A new avenue of use for the yield curve has emerged in the form of using it to forecast bull and bear stock markets. This has the potential to change how investors manage portfolios. A dynamic market-timing strategy would allow investors to shift out of or in to stock markets based on the probability of bear stock market in the future. The relationship between the yield curve and the stock market is tested using an adapted probit model. This has proven positive with encouraging results for the US, India and Spain. This is tested for South Africa using the adapted probit model and the SA yield spread. Bear stock markets are identified on the JSE and forms part of the probit modelling process. Bear markets are identified using a six- and four-month criteria. As South Africa is a small, open and developing economy, the probit is also modelled using the US yield spread. The three probit models do not appear to track bear markets well. This is substantiated through the Henriksson-Merton parametric model test which tests for market timing ability. The results for the SA yield spread using both bear market criteria do not show market timing ability, however, the SA and US yield spread model does show potential market timing ability.

- Description: This study investigates the literary activities of two emerging female Kenyan writers, Claudette Okwiri Oduor and Jacqueline Ndinda Kioko, both of whom are award-winning authors. Oduor won the 2014 Caine Prize for African Writing while Kioko bagged the Wasafiri New Writing Fiction Award 2017. It examines specifically how the writers deal with memory and mourning in negotiating between the past and future. I explore how their fictional and non-fictional narratives assist individuals and groups to confront loss, reconstruct new identities, and renegotiate belonging amidst personal and social upheaval. The fictional narratives at the centre of this research are Oduor’s “The Plea Bargain” (2011), “My Father’s Head” (2013) and “Rag Doll” (2014), and Kioko’s “Sometime Before Maulidi” (2014) and “Some Freedom Dreams” (2017). The study explores the themes of mental illness, existential crisis, and fragmentation, and considers bereavement, queer relationships, cultural freedom, and social recognition. The research further considers the active participation of these two writers in Kenya’s contemporary literary-cultural conversations, which span different genres and various media platforms, including blogs, YouTube clips, online magazines, and social media networks in dialogue with other writers. I trace the significance of the literary-cultural link these authors have with their local, continental, and global counterparts in countries like Uganda, Nigeria, and South Africa. The link finds expression through their (in)direct association with some of the new online publishing outlets in Kenya like Jalada Africa, Enkare Review, and Kikwetu. More importantly, their shared participation in and association with such international awards and scholarships as the Commonwealth Short Story Prize, Kwani Trust Manuscript Project, and Miles Morland Foundation is integral in apprehending contemporary literary exchanges and multidirectional flows of publishing in Africa and beyond. I equally illustrate how mentorship of younger writers through local writers’ organisations and collectives like AMKA and Writivism help in the formation of an alternative canon other than the mainstream. The study affirms that the authors seem to transcend the boundaries of production and circulation by fluidly moving between electronic and non-electronic platforms, thus mimicking the memory production of remembering, repeating, and working through.

Technology in conservation: towards a system for in-field drone detection of invasive vegetation

- Description: Remote sensing can assist in monitoring the spread of invasive vegetation. The adoption of camera-carrying unmanned aerial vehicles, commonly referred to as drones, as remote sensing tools has yielded images of higher spatial resolution than traditional techniques. Drones also have the potential to interact with the environment through the delivery of bio-control or herbicide, as seen with their adoption in precision agriculture. Unlike in agricultural applications, however, invasive plants do not have a predictable position relative to each other within the environment. To facilitate the adoption of drones as an environmental monitoring and management tool, drones need to be able to intelligently distinguish between invasive and non-invasive vegetation on the fly. In this thesis, we present the augmentation of a commercially available drone with a deep machine learning model to investigate the viability of differentiating between an invasive shrub and other vegetation. As a case study, this was applied to the shrub genus Hakea, originating in Australia and invasive in several countries including South Africa. However, for this research, the methodology is important, rather than the chosen target plant. A dataset was collected using the available drone and manually annotated to facilitate the supervised training of the model. Two approaches were explored, namely, classification and semantic segmentation. For each of these, several models were trained and evaluated to find the optimal one. The chosen model was then interfaced with the drone via an Android application on a mobile device and its performance was preliminarily evaluated in the field. Based on these findings, refinements were made and thereafter a thorough field evaluation was performed to determine the best conditions for model operation. Results from the classification task show that deep learning models are capable of distinguishing between target and other shrubs in ideal candidate windows. However, classification in this manner is restricted by the proposal of such candidate windows. End-to-end image segmentation using deep learning overcomes this problem, classifying the image in a pixel-wise manner. Furthermore, the use of appropriate loss functions was found to improve model performance. Field tests show that illumination and shadow pose challenges to the model, but that good recall can be achieved when the conditions are ideal. False positive detection remains an issue that could be improved. This approach shows the potential for drones as an environmental monitoring and management tool when coupled with deep machine learning techniques and outlines potential problems that may be encountered.

Towards development of a malaria diagnostic: Generation, screening and validation of novel aptamers recognising Plasmodium falciparum lactate dehydrogenase

- Description: Malaria, caused by infection with the Plasmodium parasite, is one of the leading causes of death in under-developed countries. Early detection is crucial for the effective treatment of malaria, particularly in cases where infection is due to Plasmodium falciparum. There is, therefore, an enduring need for portable, sensitive, reliable, accurate, durable, self-validating and cost-effective techniques for the rapid detection of malaria. Moreover, there is a demand to distinguish between various infectious species causing malaria. Research in the area of malarial biomarkers has identified a unique, species-specific, epitope of P. falciparum lactate dehydrogenase (PfLDH), enhancing prospects for the development of diagnostics capable of identifying the species causing malarial infection. In recent years, improvements have been made towards the development of rapid diagnostic tests for detecting malarial biomarkers. Owing to their low cost, ease of labeling, and high thermal stability (relative to antibodies), the development and synthesis of aptamers that target the malarial lactate dehydrogenase represents one of the key innovations in the field of rapid diagnostics for malaria. This study explored the generation of aptamers that specifically target P. falciparum. Two sets of aptamers with diagnostically-supportive functions were generated independently, through parallel SELEX of recombinantly-expressed, full-length Plasmodium falciparum lactate dehydrogenase (rPfLDH), and an oligopeptide comprising the P. falciparum-specific epitope on lactate dehydrogenase (LDHp). The latter offers a promising solution for generating aptamers capable of binding with high specificity to P. falciparum. In this work, an rLDH class of aptamers was generated when SELEX was performed using the full-length rPfLDH protein as the target and the LDHp class of aptamers was generated when SELEX was performed using the oligopeptide LDHp as a target. Aptamers were successfully generated through the process of SELEX (systematic evolution of ligands through exponential enrichment) following the study and application of several optimisation steps, particularly during the amplification stage of SELEX. Optimisation steps included the study of improvements in PCR conditions; role of surfactants (Triton-X), modifying the PCR clean-up protocol; and agarose gel excision. Structurally-relevant moieties with particular consensus sequences (GGTAG and GGCG) were found in aptamers both reported here and previously published, confirming their importance in recognition of the target. Novel moieties particular to this work (ATTAT and poly-A stretches) were identified. Clades of consensus sequences were identified in both the rLDH and LDHp groups of aptamers, where sequences in the rLDH clade did not show preferential binding to rPfLDH while those in the LDHp clade (particularly LDHp 3 and 18) were able to recognise and bind only LDHp. Of the 19 sequences returned from the parallel SELEX procedures for rPfLDH (11 sequences) and LDHp (8 sequences), six rPfLDH and all eight LDHp sequences underwent preliminary screening and those with low responses eliminated. Of the eight LDHp-targeting aptamer sequences, five were preliminarily shown to bind to LDHp, whereas only two rPfLDH-targeting sequences were shown to bind to the target (rLDH 4 and 7). To this small selection of rPfLDH oligonucleotide sequences, two more (rLDH 1 and 15) were chosen for further study based on their sequences, secondary and predicted tertiary conformations. Sequences chosen for further study were therefore: rLDH 1, 4, 7 and 15 in the rLDH class, and LDHp 1, 3, 11, 14 and 18 in the LDHp class. Binding properties of the aptamers towards their targets were investigated using enzyme-linked oligonucleotide assays (ELONA), fluorophore-linked oligonucleotide assays (FLONA), electromobility shift assays (EMSA), surface plasmon resonance (SPR), and GelRed dissociation assays, while applications towards aptasensors were explored using electrochemical impedance spectroscopy (EIS) and fluorescent microscopy. Some inconsistencies were seen for specific aptamer to target binding interactions using specific techniques; however, generally, binding to the targets was observed across the techniques assessed. These varied responses demonstrate the need to screen and validate aptamers using a variety of techniques and platforms not necessarily specific for the proposed application. From the aptamer binding screening studies using ELONA, the most promising aptamers generated were identified as LDHp 11, rLDH 4, rLDH 7 and rLDH 15. Aptamer rLDH 4, which was generated against rPfLDH, exhibited preferential and specific binding to the lactate dehydrogenase from P. falciparum, over the recombinantly-expressed lactate dehydrogenase from Plasmodium vivax (rPvLDH), albeit with lowered responses compared to LDHp 11 in ELONA and EMSA studies. However, in kinetic ELONA studies rLDH 4 showed binding to both rPfLDH and rPvLDH. Aptamer rLDH 7 showed high affinity for rPfLDH and rPvLDH in kinetic studies using ELONA. However, screening studies with ELONA indicates that aptamer rLDH 7 may not be suitable for diagnostic tests in serum samples given its non-specific binding to human serum albumin (HSA). Aptamer rLDH 15 exhibited species specificity for rPfLDH in screening studies using ELONA but showed affinity towards rPvLDH (albeit lower relative to its affinity for rPfLDH) in kinetic studies using ELONA. LDHp 11, generated against the PfLDH peptide, showed a clear preference for rPfLDH when compared to rPvLDH and other control proteins, in both sets of ELONA studies conducted, as well as EMSA, thus possessing a strong ability to identify the presence of Plasmodium falciparum owing to its generation against the species-specific epitope. While LDHp 1 demonstrated binding to plasmodial LDH in a flow-through system (SPR), so reiterating ELONA responses, it did not perform well in the remaining methodologies. Aptamers rLDH 1 and 15 and LDHp 3, 14 and 18 exhibited a mixed set of results throughout the target protein screening analyses and were, thus, not considered for selective binding in P. falciparum parasite bodies. In studies aimed at exploring biosensor assemblies utilising the developed aptamers, both rLDH 4 and LDHp 11, along with rLDH 7, LDHp 1 and pL1, demonstrated in situ binding to the native PfLDH in fluorescent microscopy. LDHp 11 exhibited FITC-based fluorescence equivalent to the anti-rPfLDHp IgY antibody in confocal fluorescent microscopy indicating superior binding to the native PfLDH compared to the remaining aptamers. An examination of electrochemical impedance as a platform for a biosensor assembly did not, in these studies, exhibit the required sensitivity using physiologically relevant concentrations of analyte expected for pLDH following infection with Plasmodium spp. Malstat/LDH activity was explored for application in a colorimetric aptasensor. A decrease in both rPfLDH and rPvLDH activity was observed following incubation with the tested aptamers, but rLDH 1, rLDH 7 and LDHp 14 did not exhibit similar decreases in rPvLDH activity. Aptamers rLDH 1, 4 and 7 and LDHp 11 and 14 were, therefore, not selected as candidates for LDH capture in LDH activity-based diagnostic devices for P. falciparum. The decreases in pLDH activity in the presence of aptamers could hold promise as direct or antagonistic malaria therapeutic agents. Preliminary studies on the application of aptamers as malaria therapeutic agents, while of interest, should be viewed with due caution given the challenges of aptamers reaching the intracellular native plasmodial LDH hosted within the red blood cells. In conclusion, this work has shown the ability of the LDHp 11 aptamer, generated in these studies, to selectively bind rPfLDH over rPvLDH, and to bind to the native PfLDH in fluorescent microscopy, indicating that this aptamer holds promise as a biorecognition element in malaria biosensors and other diagnostic devices for the detection, and differentiation, of P. falciparum and P. vivax. The use of a species-specific epitope of P. falciparum as a target in aptamer generation paves the way for similar such studies aimed at generating aptamers with species selectivity for other Plasmodium species.

Aptamer rLDH 4, which was generated against rPfLDH, exhibited preferential and specific binding to the lactate dehydrogenase from P. falciparum, over the recombinantly-expressed lactate dehydrogenase from Plasmodium vivax (rPvLDH), albeit with lowered responses compared to LDHp 11 in ELONA and EMSA studies. However, in kinetic ELONA studies rLDH 4 showed binding to both rPfLDH and rPvLDH. Aptamer rLDH 7 showed high affinity for rPfLDH and rPvLDH in kinetic studies using ELONA. However, screening studies with ELONA indicates that aptamer rLDH 7 may not be suitable for diagnostic tests in serum samples given its non-specific binding to human serum albumin (HSA). Aptamer rLDH 15 exhibited species specificity for rPfLDH in screening studies using ELONA but showed affinity towards rPvLDH (albeit lower relative to its affinity for rPfLDH) in kinetic studies using ELONA. LDHp 11, generated against the PfLDH peptide, showed a clear preference for rPfLDH when compared to rPvLDH and other control proteins, in both sets of ELONA studies conducted, as well as EMSA, thus possessing a strong ability to identify the presence of Plasmodium falciparum owing to its generation against the species-specific epitope. While LDHp 1 demonstrated binding to plasmodial LDH in a flow-through system (SPR), so reiterating ELONA responses, it did not perform well in the remaining methodologies. Aptamers rLDH 1 and 15 and LDHp 3, 14 and 18 exhibited a mixed set of results throughout the target protein screening analyses and were, thus, not considered for selective binding in P. falciparum parasite bodies. In studies aimed at exploring biosensor assemblies utilising the developed aptamers, both rLDH 4 and LDHp 11, along with rLDH 7, LDHp 1 and pL1, demonstrated in situ binding to the native PfLDH in fluorescent microscopy. LDHp 11 exhibited FITC-based fluorescence equivalent to the anti-rPfLDHp IgY antibody in confocal fluorescent microscopy indicating superior binding to the native PfLDH compared to the remaining aptamers. An examination of electrochemical impedance as a platform for a biosensor assembly did not, in these studies, exhibit the required sensitivity using physiologically relevant concentrations of analyte expected for pLDH following infection with Plasmodium spp. Malstat/LDH activity was explored for application in a colorimetric aptasensor. A decrease in both rPfLDH and rPvLDH activity was observed following incubation with the tested aptamers, but rLDH 1, rLDH 7 and LDHp 14 did not exhibit similar decreases in rPvLDH activity. Aptamers rLDH 1, 4 and 7 and LDHp 11 and 14 were, therefore, not selected as candidates for LDH capture in LDH activity-based diagnostic devices for P. falciparum. The decreases in pLDH activity in the presence of aptamers could hold promise as direct or antagonistic malaria therapeutic agents. Preliminary studies on the application of aptamers as malaria therapeutic agents, while of interest, should be viewed with due caution given the challenges of aptamers reaching the intracellular native plasmodial LDH hosted within the red blood cells. In conclusion, this work has shown the ability of the LDHp 11 aptamer, generated in these studies, to selectively bind rPfLDH over rPvLDH, and to bind to the native PfLDH in fluorescent microscopy, indicating that this aptamer holds promise as a biorecognition element in malaria biosensors and other diagnostic devices for the detection, and differentiation, of P. falciparum and P. vivax. The use of a species-specific epitope of P. falciparum as a target in aptamer generation paves the way for similar such studies aimed at generating aptamers with species selectivity for other Plasmodium species.

- Description: In South Africa, freshwater ecosystems are under pressure due to increasing anthropogenic factors degrading river systems. The Cape Fold, the lower Southern Temperate Highveld and the Amatolo – Winterberg Highlands freshwater ecoregions contain some of the heavily impacted river systems in the country. One such river system is the Great Fish River system, which has been affected by anthropogenic modification (hydrological modification, water pollution and invasion by non-native fish species). These modifications have raised conservation concerns on an imperilled anabantid fish, the Eastern Cape rocky Sandelia bainsii that is confined to the Kat and Koonap rivers, which are tributaries of the Great Fish River. Historically, the species was reportedly common and abundant, with a range spanning across seven river systems (Kowie, Great Fish, Keiskamma, Igoda, Gxulu, Buffalo and Nahoon rivers) in the Eastern Cape Province. However, surveys have indicated that the species has experienced decline in population size and distribution range, with localised extirpations being reported. This resulted in its listing on the IUCN Red List of threatened species as Endangered. The proposed shale gas exploration and potential infrastructure development in the Karoo basin, which encompasses the headwaters of the Kat River, represent a potential future threat to the remnant populations of this species. The aim of the present study was to review the past and present distribution range of S. bainsii, determine the habitat associations and assess the conservation of S. bainsii in the Great Fish River system. This information is essential for informing conservation and management decisions for this species in the Great Fish River system. This study undertook a comprehensive survey of the Kat and the Koonap rivers in 2017, and augmented this information with data from more recent surveys that were conducted from 2009 - 2014 to map the distributions freshwater fishes in the Great Fish River system. Data from these recent surveys (2009 – 2017) were compared with historical records (1961 – 2005) obtained from the South African Institute for Aquatic Biodiversity to evaluate changes in distribution patterns of S. bainsii. Historical records indicated that S. bainsii occurred at 11 localities in the Kat River and three localities in the Koonap River. Data from recent surveys indicated that the species has persisted at these historical localities, except one locality in the Koonap River where no individuals of S. bainsii were captured, despite the use of multiple sampling approaches and gears. Although the species still persists at two of the three historical localities in the Koonap River, the population sizes have considerably declined as the species was reportedly in high abundance at these localities in the 1980s. Factors threatening the continued existence of this species in the Koonap River likely include non-native species, extreme drought conditions and the associated excessive abstraction of water which has resulted in the drying of some of the refugia pools where the species was historically common and abundant. In contrast to the Koonap River, the 2017 survey indicated that in the Kat River, S. bainsii had a similar distribution range compared to historical records. The species occurred at 11 of the 41 localities that were sampled in the Kat River, and the sampled populations consisted of all size classes. Sandelia bainsii occurred with other native fish species in the system, including Labeo umbratus, Enteromius anoplus and Glossogobius callidus. A comparison of historical and recent data indicates that non-native fishes have likely not expanded their ranges in the Kat River. This is likely due to the presence of weirs and dams that have prevented upstream movement of non-native fish species L. aeneus, C. gariepinus and T. sparrmanii which are now abundant in the lower Kat and mainstem Great Fish River. Surveys from the present study indicated that although S. bainsii still persists within the Kat River, there are a number of factors that pose a considerable threat to the continued existence of this species in the Great Fish River system. Chapter 3 evaluated the habitat associations of S. bainsii based on 10 physical and chemical variables and distribution data collected from 30 localities in the Upper Kat River and seven of its tributaries. A non-metric multidimensional scaling model was used to assess the distribution patterns of S. bainsii in relation to environmental variables. Furthermore, a negative binomial generalised linear model (GLM) was used to assess the relationship between the abundance of S. bainsii and environmental variables. Results from the present study indicated that S. bainsii was a habitat generalist as the distribution of this species showed no specific association with any of the habitat variables that were considered in this study. The species occurred in a wide range of habitats, including rocky sections in the upper Kat River and its tributaries as well as the mainstem sections. The GLM however indicated that the abundance of S. bainsii was negatively associated with increasing conductivity. In addition, the Kat River contains multiple weirs of variable sizes. Future studies should therefore evaluate the effects of these weirs on population fragmentation, gene flow and long-term evolutionary potential of S. bainsii through the application of techniques such as mitochondrial DNA and microsatellites.

- Description: The present study investigated the semantic classification of the Setswana noun class system. This enquiry falls under the broad area of the noun classification system in Bantu languages, psycholinguistics and lexicogrpahy. Specifically it explores the basis of noun classification in Setswana making indications that Setswana noun classification is based on a partial semantic classification. Data for the study was drawn from the Setswana Oxford Dictionary. Sixty Setswana nouns, from class 1, 3, 5, and 7, were selected and analysed and then grouped into semantic categories (i.e., PERSON, DEROGATION, TRANSPORATION and so forth). The study adopted Kgukutli’s (1994) semantic classification in performing the dictionary analysis. The rest of the data was drawn from the intuitions of thirty-nine contemporary speakers of Setswana, with the aid of a linguistic test which was fashioned according to Selvik’s (2001) psycholinguistic test. The language test required participants to match the predetermined Setswana definitions with hypothetical Setswana nouns with selected class prefixes attached to them. The results from the empirical study showed that speakers were associating prefixes to certain semantic values, suggesting that each noun class had specific semantic content that was unique to that class. The semantic categories created through the dictionary analysis were then compared to those given by the thirty-nine Setswana speakers, to analyse whether there were any similaritires in the semantic classification of the noun classes. The findings of the dictionary analysis and linguistic test revealed that there were certain semantic characteristics that each class was associated with that seemed to be unique to the class. However, there were various semantic overlaps in the semantic categories associated with the different noun classes, which brings into question whether a semantic classification is viable in the classing of nouns. The study suggests that prior classification of Setswana nouns are not precise enough and that additional semantic categories are needed to offer a more precise classification of nouns in this language.

Satire in J.J. R. Jolobe's literary works : a critique in relation to contemporary South Africa

- Description: J.J.R. Jolobe is regarded as one of the individuals who made a valuable contribution to the development of isiXhosa literature through his works, notably in his poetry (Ilitha, Umyezo; Jolobe 1936). His poetry ranges from abstract subjects to more philosophical matters. This study is aimed at decoding the manner in which he employs satire to conscientise African people of the then horrible situation that they were facing. With that said, poetry will not be the only work that this thesis analyses in the process of evaluating Jolobe’s satire; his essays will also be examined (Amavo; Jolobe 1940). Based on the writings of various authors specialising in the subject, satire has been deemed to be a style of literary writing, one which involves invective satire. For the researcher, that statement will be rebutted, as it will be argued that the mode of satire need not be wholly invective. Jolobe’s light-hearted satire not only showcases the amusing side of his writings, but also indicates the seriousness with which they were intended. Themes covered in Jolobe’s satire have inspired the researcher to evaluate these literary texts in relation to modern contexts, especially when it comes to the relationship between the lines of the author’s experience and the public. With that said, the social role of satire is something that one cannot deny. One could therefore say that there is an urgent need for African satirists to face the existing social and economic reality as authentically as possible. The voice of a satirist should also echo the voice of their society as a whole. Satirical study in post-colonial Africa, in South Africa in particular, is useful due to the idea that the works of the likes of Jolobe may diminish in significance due to neo-colonialism. In fact, this is the point which is considered in this study of Jolobe’s satire. This study also examines stages afforded to the development of satire in Africa, especially in the post-colonial era. The purpose is to identify the effects of satire that are related to socio-political as well as religious factors. These factors are often seen as those that play a vital role is one’s personal morals, and those that are meant to shape the whole community. Jolobe addresses imperialism and the class struggle, which speaks to the society’s loyalties regarding the mobilization toward realizing the dream of being independent. This speaks to the works analysed, revealing protests against oppression and exploitation by imperialists; such works show how inhumane people could be against those who they deem to be beneath their standards. Researchers like Mahlasela (1973), Sirayi (1985), Kwetana (2000) and Khumalo (2015) are amongst those who have made it a point to study Jolobe to ensure that these works are kept alive, along with their significance. Other prospective researchers can follow suite in researching the great Jolobe. In ensuring that the aims of this study come to light, the researcher will be using socialist realism as a way of seeing that the works of Jolobe are realistic in nature. With that said, there will be an exploration of allegoric satire. Satiric allegory will be evaluated with regard to the view that it represents a unique slant on satire, whereby it deems satire to be more than just a supportive method of literary criticism. This allows the researcher to hold the view that satire should not be a restrictive framework when dealing with African literature. - Description: J.J.R. Jolobe is regarded as one of the individuals who made a valuable contribution to the development of isiXhosa literature through his works, notably in his poetry (Ilitha, Umyezo; Jolobe 1936). His poetry ranges from abstract subjects to more philosophical matters. This study is aimed at decoding the manner in which he employs satire to conscientise African people of the then horrible situation that they were facing. With that said, poetry will not be the only work that this thesis analyses in the process of evaluating Jolobe’s satire; his essays will also be examined (Amavo; Jolobe 1940). Based on the writings of various authors specialising in the subject, satire has been deemed to be a style of literary writing, one which involves invective satire. For the researcher, that statement will be rebutted, as it will be argued that the mode of satire need not be wholly invective. Jolobe’s light-hearted satire not only showcases the amusing side of his writings, but also indicates the seriousness with which they were intended. Themes covered in Jolobe’s satire have inspired the researcher to evaluate these literary texts in relation to modern contexts, especially when it comes to the relationship between the lines of the author’s experience and the public. The spread of publication dates across the three novels also allows for an examination of morphing social critique from 2000-2013.

Mathematics educationists therefore advocate that schools should make extensive and deep efforts to rethink their instructional programmes. Research furthermore suggests that increasingly, indicators on school performance and teaching reveal largely unacknowledged problematic teaching of mathematics in the great majority of South African schools. This research study is therefore a contribution towards rethinking the teaching strategies within mathematics classrooms. The study examined the use of visualisation processes in order to understand how these interact with the pedagogy of selected mathematics teachers when they teach number sense after participating in an intervention programme. This study argues that the effective use of visualisation processes enhanced the teaching of number sense. The research study was framed as a case study that was grounded within the interpretive paradigm. The study was located in classrooms where the participating teachers promoted active learning after taking part in an intervention programme. A constructivist theoretical underpinning was therefore adopted. At the heart of the study was the Visualisation Intervention Programme (VIP), which involved seven Rhodes University Mathematics Education Project (RUMEP) teachers within the John Taolo Gaetsewe District of the Northern Cape Province. The content of the VIP was informed by initially working with five teachers of well-resourced schools within the Northern Cape who made interesting use of visualisation processes and manipulatives to teach number sense. With the assistance of these five teachers, the VIP was then implemented by seven selected RUMEP teachers to investigate the role of visualisation processes in the teaching of number sense understandings. The study employed a mixed method approach. Qualitative data was collected through observations and interviews, while quantitative data was collected with a series of pre- and post-tests. The analysis of the findings of this research study revealed that the effective use of visualisation processes was instrumental in enhancing the teaching of number sense understandings. A comparative mineralogical and geochemical study of manganese deposits in the Postmasburg Manganese Field, South Africa

- Description: The Postmasburg Manganese Field (PMF), located in the Northern Cape Province of South Africa, is host to some of the largest deposits of iron and manganese metal in the world. These deposits are restricted to a geographical area known as the Maremane Dome, an anticlinal structure defined by folded dolostones of the Campbellrand Subgroup and overlying ironformations of the Asbestos Hills Subgroup of the Neoarchaean-Palaeproterozoic Transvaal Supergroup. Manganese ores associated with the Maremane Dome have been divided into two major classes in the literature: the Wolhaarkop breccia-hosted massive ores of the Eastern Belt, as well as the shale-associated ores of the Western Belt. The Eastern Belt ores have been classed as siliceous in nature, while the Western Belt deposits are reported to be typically ferruginous. These divisions were made based on their varying bulk chemical and mineralogical compositions in conjunction with their different stratigraphic sub-settings. Presently, both deposit types are explained as variants of supergene mineralisation that would have formed through a combination of intense ancient lateritic weathering in the presence of oxygen, extreme residual enrichments in Mn (and Fe), and accumulations in karstic depressions at the expense of underlying manganiferous dolostones. This study revisits these deposits and their origins by sampling representative end-member examples of both Eastern Belt and Western Belt manganese ores in both drillcore (localities Khumani, McCarthy and Leeuwfontein), and outcrop sections (locality Bishop). In an attempt to provide new insights into the processes responsible for the genesis of these deposits, the possibility of hydrothermal influences and associated metasomatic replacement processes is explored in this thesis. This was achieved using standard petrographic and mineralogical techniques (transmitted and reflected light microscopy, XRD , SEM-EDS and EMPA), coupled with bulk-rock geochemical analysis of the same samples using a combination of XRF and LAICP- MS analyses. Combination of field observations, petrographic and mineralogical results, and geochemical data allowed for the re-assessment of the different ore types encountered in the field. Comparative considerations made between the bulk geochemistry of the different end-member ore types revealed no clear-cut compositional distinctions and therefore do not support existing classifications between siliceous (Eastern Belt) and ferruginous (Western Belt) ores. This is supported by trace and REE element data as well, when normalised against average shale. The geochemistry reflects the bulk mineralogy of the ores which is broadly comparable, whereby braunite and hematite appear to be dominant co-existing minerals in both Eastern Belt (Khumani) and Western Belt (Bishop) ore. In the case of the McCarthy locality, manganese ore is cryptomelane-rich and appears to have involved recent supergene overprint over Eastern Belt type ore, whereas the Leeuwfontein ores are far more ferruginous than at any other locality studied and therefore represent a more complex, hybrid type of oxide-rich Mn mineralisation (mainly bixbyitic) within massive hematite iron ore. In terms of gangue mineralogy, the ores share some close similarities through the omnipresence of barite, and the abundance of alkalirich silicate minerals. Eastern Belt ores contain abundant albite and serandite whereas the main alkali-rich phase in Western Belt ores is the mineral ephesite. In both cases, Na contents are therefore high at several wt% levels registered in selected samples. The afore-mentioned alkali enrichments have been variously reported for both these deposit types. The occurrence of high alkalis cannot be explained through classic residual or aqueous supergene systems of ore formation, as proposed in prevailing genetic models in the literature. Together with the detection of halogens such as F and Br through SEM-EDS analyses of ore from both belts, the alkali enrichments suggest possible hydrothermal processes of ore formation involving circulation of metalliferous sodic brines. Selected textural evidence from samples from both ore belts lends support to fluid-related models and allow the proposal for a common hydrothermal-replacement model to have been responsible for ore formation across the broader Maremane Dome region.

Mangrove forests afford juvenile fish and invertebrates with nursery and recruitment habitats, facilitated by the fine scale configuration of their specialised root systems. Although the importance of mangroves for resident and transient juveniles is well recognised, the roles that mangrove microhabitats play for larvae is not yet comprehensively understood. This study aimed to determine how microhabitats with varying degrees of complexity influence the composition, abundance and distribution of larval communities that inhabit mangrove forests and the physiological responses of larvae to acute temperature variations in relation to ontogenetic stage and microenvironment exposure. Two relatively pristine study sites were selected to represent a warm temperate and subtropical mangrove system in the Eastern Cape and KwaZulu-Natal on the east coast of South Africa, respectively. The differences in complexity among the root systems of Rhizophora mucronata, Avicennia marina and Bruguiera gymnorhiza were assessed using 3D scanning and the computed 3D models were then analysed using four complexity metrics. Results indicated that A. marina is the most complex in terms of surface-volume ratio, R. mucronata has the most interstitial space among its roots and B. gymnorhiza and R. mucronata differ in their fractal dimensions. Larvae collected in each microhabitat at each site using light traps showed that, despite temperature and salinity homogeneity across microenvironments, spatio-temporal differences occurred in both fish and invertebrate assemblages. This trend suggests that microhabitat structural complexity exerts an influence on larval community composition by acting as a microscape of available habitat, which ensures ecological linkages within and among the mangrove forest and adjacent ecosystems. In addition, the oxygen consumption rates of mangrove-associated brachyuran larvae varied according to mangrove microhabitat, whereby larvae collected at less complex environments had the highest metabolic rates at increased temperatures. Moreover, ontogenetic shifts in physiology were prevalent as older brachyuran larvae were more eurythermal than earlier stages, suggesting that thermally stressful events will have a greater impact on recently spawned larvae. Overall, the interstitial spaces within individual root systems are the most important complexity measure, as utilisation of these mangrove microhabitats is scale-dependent, and larvae will most likely occupy spaces inaccessible to large predators. Likewise, microscale variation in the environmental conditions and ontogenetic stage of brachyuran larvae within the mangrove microscape, can amplify the physiological responses to rapid temperature variations. Results suggest that early stage larvae are the most vulnerable to mass-mortality, and if thermally stressful events increase in frequency, duration and magnitude, the larval supply for the successful recruitment into adult populations could be under threat. Through linking how mangrove microhabitat complexity influences larvae in terms of community metrics and physiology, this study paves the way for further advancement of our understanding of how microscale processes emerge into meso- and macroscale patterns and influence the stability and functioning of highly productive ecosystems.

The differences in complexity among the root systems of Rhizophora mucronata, Avicennia marina and Bruguiera gymnorhiza were assessed using 3D scanning and the computed 3D models were then analysed using four complexity metrics. Results indicated that A. marina is the most complex in terms of surface-volume ratio, R. mucronata has the most interstitial space among its roots and B. gymnorhiza and R. mucronata differ in their fractal dimensions. Larvae collected in each microhabitat at each site using light traps showed that, despite temperature and salinity homogeneity across microenvironments, spatio-temporal differences occurred in both fish and invertebrate assemblages. This trend suggests that microhabitat structural complexity exerts an influence on larval community composition by acting as a microscape of available habitat, which ensures ecological linkages within and among the mangrove forest and adjacent ecosystems. In addition, the oxygen consumption rates of mangrove-associated brachyuran larvae varied according to mangrove microhabitat, whereby larvae collected at less complex environments had the highest metabolic rates at increased temperatures. Moreover, ontogenetic shifts in physiology were prevalent as older brachyuran larvae were more eurythermal than earlier stages, suggesting that thermally stressful events will have a greater impact on recently spawned larvae. Overall, the interstitial spaces within individual root systems are the most important complexity measure, as utilisation of these mangrove microhabitats is scale-dependent, and larvae will most likely occupy spaces inaccessible to large predators. Likewise, microscale variation in the environmental conditions and ontogenetic stage of brachyuran larvae within the mangrove microscape, can amplify the physiological responses to rapid temperature variations. Results suggest that early stage larvae are the most vulnerable to mass-mortality, and if thermally stressful events increase in frequency, duration and magnitude, the larval supply for the successful recruitment into adult populations could be under threat. Through linking how mangrove microhabitat complexity influences larvae in terms of community metrics and physiology, this study paves the way for further advancement of our understanding of how microscale processes emerge into meso- and macroscale patterns and influence the stability and functioning of highly productive ecosystems.

The study was conducted along a gradient of impacts from less impacted agricultural headwaters to highly impacted urban sites located immediately downstream of the city of Makhanda. Macroinvertebrates were separated into functional feeding groups (FFGs) (i.e. collector-gatherer, collector-filterer, scraper, shredder, and predator) which were then used to assess the effects of selected physico-chemical variables and riparian zone condition on FFG organization. Collector-gatherers were the most abundant in the Bloukrans River and represented 71.3 % of the macroinvertebrate assemblages. Stream order 1species such as Nitzschia palea, Gomphonema parvulum, Tryblionella apiculata, Diploneis vulgaris and Staurosira elliptica. Multivariate analysis (Canonical correspondence analysis (CCA)) indicated that differences in diatom community assemblages were best explained by calcium, magnesium, pH, phosphate, nitrate, dissolved oxygen, sediment nitrate, conductivity and salinity. These results indicate that diatoms can be used as bioindicators for monitoring highly impacted river systems and to also further examine pollution gradients and impacts of specific/point pollution sources. In order to further test the application of diatom indices, nine sites with contrasting water quality were sampled along the length river system in February, May and July 2018. Diatom-based indices incorporated in OMNIDIA software were applied to assess the integrity of the water quality as indicated by diatom communities. For comparative purposes, several foreign indices (e.g. the trophic diatom index (TDI), the percentage pollution-tolerant valves (%PTV), biological diatom index (BDI)) and the South African Diatom Index (SADI) were used in the study. From the results, the Percentage Pollution-Tolerant Valves (%PTV) of most urban sites in the Bloukrans River was above the 20% limit indicating the presence of organic pollutants. Although the foreign diatom indices were applicable in the study, the SADI had significant correlations with most water quality variables (p < 0.05) compared to other indices such as Watanabe Index (WAT), Biological Index of Water Quality Trophic Index (BIWQ) and Trophic Index (TI)). These results support wider use of the SADI as an indicator of water quality conditions in South African river systems. Finally, the observed variations in diatom community structure and composition as a result of changes in water quality were broadly in agreement with the results of macroinvertebrate FFG structure indicating that the two biological indicators can, and should, be used as complementary techniques in the biomonitoring of rivers and streams in South Africa.

- Description: Climate change and variability is a global phenomenon which has deeply localised patterns, dynamics and effects. Amongst those people who are particularly vulnerable to climate change effects are small-scale farmers who are dependent in large part on rain-fed agriculture in pursuing their livelihoods. This is true of small-scale farmers in contemporary Zimbabwe and, more specifically, farmers in communal areas. At the same time, at international and national levels, there are attempts currently to minimise the effects of, and to adapt to, climate change. However, adaptation measures also exist at local levels amongst small-scale farmers, such as communal farmers in Zimbabwe. In this context, as its main objective, this thesis examines climate change and small-scale farmer livelihood adaptation to climate change with specific reference to communal farmers in Chivi District in Zimbabwe and, in particular, in Ward 25 which is popularly known as Ngundu. In pursuing this main objective, a number of subsidiary objectives are addressed, including a focus on the established livelihoods of Ngundu farmers, the perceptions and concerns of Ngundu farmers about climate change, the coping and adaptation measures of Ngundu farmers, and the enablements and constraints which affect attempts by Ngundu farmers to adopt such measures. The fieldwork for the thesis involved a diverse array of research methods, such as a questionnaire survey, life-history interviews, key informant interviews, focus group discussions and transect walks. In terms of theoretical framing, the thesis makes use of both middle-level theory (the Sustainable Livelihoods Framework) and macro-theory in the form of the sociological work of Margaret Archer. Combined, these two theories allow for a focus on both structure and agency when seeking to understand livelihood adaptations to climate change by communal farmers in Ngundu. The thesis concludes that there are massive constraints inhibiting adaptation measures by Ngundu farmers, but that this should not distract from the deep, often historically-embedded, concerns of Ngundu farmers about climate change and the multiple ways in which they express agency in and through adaptation and coping activities. It also highlights the need for more specifically sociological investigations of climate change and small-scale farmer adaptation, as well as the need for localised studies which are able to identify and analyse the specificities of adaptation.

Stable isotope analysis identified substantial ontogenetic dietary shifts in all species, corresponding to changes in body size. Stable isotope analysis revealed that the niche space occupied by G. affinis was broad and overlapped with that of the other three focal species. Stable isotope metrics showed that G. affinis and O. mossambicus utilised a wide range of resources compared to G. callidus and G. aestuaria. Stomach content analysis showed that G. callidus, O. mossambicus and G. affinis fed predominantly on benthic resources, while G. aestuaria fed mainly plankton resources. Functional response experiments revealed that G. callidus and G. affinis both displayed Type II functional responses. In single fish trials, G. affinis had significantly higher functional responses than G. callidus. In heterospecific G. callidus-G. affinis combinations the functional response of G. callidus was reduced by the presence of G. affinis, whereas, this combination greatly enhanced G. affinis functional response magnitudes. The functional response of G. callidus, O. mossambicus and G. affinis under two temperature treatments along with fish abundance data was used to determine temporal differences in the ecological impacts of each fish species between seasons. The relative impact potential of O. mossambicus was consistently higher than that of G. callidus and G. affinis. This study demonstrates how seasonal temperature fluctuations affect the relative impact capacities of introduced species. Overall, this thesis showed that high temperature along with life-history traits contributes to the biotic interactions between native and non-native species in novel environments.

The problem of student retention is not new in our institutions of higher learning. Support structures, strategies, and policies need to be developed for different academic and social challenges that students face daily. This study explores the need for academic support at Rhodes University; it also explores the internal and external problems that students face, the perceptions and experiences of students on academic support available to them. The study implemented a narrative research approach implanted within a qualitative research framework. Data was collected through a questionnaire, and narrative interviews. Tinto’s Student Integration Model was used to analyse the data in this study and was supplemented by Louw’s conceptual framework model and Swail’s Geometric Design of Student Persistence. These three models position the student experience as the foundation for academic support programmes and development. The study argues that as long as effective and adequate academic support is lacking, students will continue to experience difficulties in completing their studies. The findings from the study reveal that at present, Rhodes University provides support to students who are in extended studies. However, this support system disadvantages many students, notably those not in the extended studies programme. As a result, such students are not supported through strategies and structures that extended studies students receive. Furthermore, the data generates internal and external problems that student face daily, as a result, the findings showed that there is a need for academic support at Rhodes. The study recommends that Rhodes University must establish an academic unit that will provide academic support to all students registered in the university.

- Description: The primary objective of this study was to assess the pattern of change in neurocognitive performance for adolescent athletes on baseline measures of the Immediate Post Concussion Assessment and Cognitive Testing (ImPACT) test, over five consecutive years, with a view to providing an indication of the optimal interval for repeat baseline testing of high school athletes. Participants were non-clinical, predominantly South African high school athletes in the overall age range 13 to 18 years (N = 108), divided into five groups (Grades 8,9, 10, 11 and 12), and tested at five test intervals. Repeated-measures ANOVA analyses examined differences in score performance across the test intervals for each of the five composite scores of the ImPACT test (Verbal Memory, Visual Memory, Visual Motor Speed, Reaction Time, Impulse Control). For the Verbal Memory, Visual Memory, Visual Motor Speed and Reaction Time composites there were significant neurocognitive score changes between several test intervals. Taking these results into account, in conjunction with substantial variability in performance, it is concluded that there is a need for annual baseline testing throughout the high school years. The secondary objective was to generate normative tables (Means and Standard Deviations) on the ImPACT test for the five participant groups at each of the five test intervals, including data for: the five composite scores (Verbal Memory, Visual Memory, Visual Motor Speed, Reaction Time, Impulse Control); for the twelve subtest scores test that go to make up the composite scores; and for four additional memory subcomponent scores (Word Memory immediate recall, Word Memory delayed recall, Design Memory immediate recall, Design Memory delayed recall). The results provide a clinical and heuristic normative platform for future use with brain injured individuals, which can be used to facilitate clinical interpretations of postconcussion assessments.

- Description: The primary objective of this study was to assess the pattern of change in neurocognitive performance for adolescent athletes on baseline measures of the Immediate Post Concussion Assessment and Cognitive Testing (ImPACT) test, over five consecutive years, with a view to providing an indication of the optimal interval for repeat baseline testing of high school athletes. Participants were non-clinical, predominantly South African high school athletes in the overall age range 13 to 18 years (N = 108), divided into five groups (Grades 8,9, 10, 11 and 12), and tested at five test intervals. - Description: This study focuses on the influence of clientelism in the informal sector of Zimbabwe in Glen View 8 (Complex). The study used the case of Glen view 8 (complex) in Harare Province. The study focused on political dynamics in the informal sector; livelihood strategies employed by informal sector operators; manifestation of “Big Men”, social networks in the informal sector; and different strategies employed by operators to overcome the problems of political manipulation, clientelism and patronage. It employs a qualitative research methodology to enable a nuanced comprehension of the clientelistic relationships that take place in the informal sector of Zimbabwe. Through in-depth interviews, focus group discussions, narratives and in-depth interviews with key informants, the study explored the clientelistic nature of the informal sector. The major findings of the study are that the informal sector in Zimbabwe is influenced by political patronage. It was established that patronage influences the informal sector in Zimbabwe in general and at Glen View Complex 8 in particular. Some of the operators revealed that patronage negatively affects their business as they are sometimes forced to attend political party meetings either at the complex or at ZANU-PF star rallies in town. The operators experience a plethora of problems such as lack of security, poor sanitation, stiff competition, poor infrastructure, lack of insurance and fire outbreaks. It has been revealed that most of the problems experienced at the complex are a result of the politicisation of the informal sector particularly by the ZANU-PF party. Operators at the complex have described the politicisation of the informal sector as a major drawback to their efforts of realising maximum benefits from their work.
