Structure and functioning of fish assemblages in two South African estuaries, with emphasis on the presence and absence of aquatic macrophyte beds

- Description: Temporarily open/closed estuaries (TOCEs) are the dominant estuary type in South Africa. These systems are often characterized by extensive beds of submerged macrophytes, which form important foraging and shelter habitats for fishes, especially for estuary-dependent fish species such as the Cape stumpnose Rhabdosargus holubi and Cape moony Monodactylus falciformis that are commonly associated with them. A loss of submerged macrophytes from an estuary has been shown to affect the fish community as well as reducing overall system productivity. The TOC East Kleinemonde Estuary, situated in the warm-temperate biogeographic region of South Africa has been subject to an ongoing long-term fish monitoring project since 1995. During the period 1995 to 2002, this estuary contained large beds of the submerged macrophytes Ruppia cirrhosa and Potamogeton pectinatus. However, subsequent to a major flood event in 2003 these macrophytes have been largely absent from this system. The effect of the loss of submerged macrophytes on the East Kleinemonde fish assemblage was investigated through an analysis of seine and gill net catch data. Seine net catches for a 12 year period, encompassing six years of macrophyte presence and six years of macrophyte senescence, revealed changes in the relative abundance of certain fish species. Vegetation-associated species such as R. holubi and M. falciformis decreased in abundance whereas sediment-associated species, especially members of the family Mugilidae, increased in abundance following loss of the macrophytes in this estuary. The critically endangered pipefish Syngnathus watermeyeri was only recorded in catches during years in which macrophyte beds were present. In addition to the analysis of catch data, the importance of macrophytes as a primary energy source for selected estuarine fishes was explored through the analysis of carbon and nitrogen stable isotopes. Prior to conducting these analyses, common methodological practices to address the presence of carbonates and lipids within isotope samples were evaluated. A subset of samples were either acid washed to remove carbonates, or lipids were removed according to the method of Bligh and Dyer (1959) as both of these compounds have been shown to affect stable carbon and nitrogen isotope ratios. The suitability of the lipid normalization models of Fry (2002) and Post et al. (2007) for samples of estuarine fish muscle were also tested. Based on this evaluation both models are suitable for use with estuarine fish muscle tissue, however since neither carbonate nor lipid content of any of the samples used in this study was high all samples were left untreated in the following analysis. Carbon isotope ratios from a wide range of fish species collected from the East Kleinemonde Estuary during the macrophyte-senescent phase were compared with individuals of the same species from the neighbouring West Kleinemonde Estuary (where extensive beds of R. cirrhosa and P. pectinatus were present) and revealed the influence of submerged macrophyte material in the diet of fishes in the latter system. However, it was apparent that these plants are not directly consumed but rather contribute to the detrital pool that forms a food source for most invertebrate and some fish species. The most significant source of carbon for East Kleinemonde fishes during the macrophyte senescent phase appeared to have a more depleted origin; probably from benthic or pelagic microalgae. In conclusion, while the importance of macrophyte beds as shelter and foraging habitats for estuarine fishes are well documented, their role in terms of the structuring and functioning of fish assemblages in TOCEs remains somewhat uncertain. The findings of this study were possibly masked by the resilience of vegetation-associated species to the loss of this habitat, as well as by life history characteristics of species such as R. holubi that allow their numerical dominance despite habitat change. Nonetheless, macrophyte senescence in the East Kleinemonde Estuary resulted in the loss of at least one species and the reduced abundance of vegetation-associated species, probably reflective of reduced food resources and/or increased vulnerability to predation. As a result, beds of submerged macrophytes are an important habitat within TOCEs.

The isolation of muscle activity and ground reaction force patterns associated with postural control in four load manipulation tasks

- Description: Although much effort has been placed into the reduction of risks associated with manual materials handling, risk of musculoskeletal disorder development remains high. This may be due to the additional muscle activity necessary for the maintenance of postural equilibrium during work tasks. This research proposes that postural control and subsequent additional muscle activity is influenced by the magnitude of the external load and the degree of body movement. The objective of this research was to identify whether performing tasks with increased external load and with a greater degree of trunk motion places additional strain on the musculoskeletal system in excess of that imposed by task demands. Twenty-four male and twenty-four female subjects performed four load manipulation tasks under three loading conditions (0.8kg, 1.6kg, and 4kg). Each task comprised of a static and dynamic condition. For the static condition, subjects maintained a stipulated posture for ten seconds. The dynamic condition required subjects to move and replace a box once every three seconds, such that a complete lift and lower cycle was performed in six seconds. Throughout task completion, muscle activity of six pairs of trunk muscles were analysed using surface electromyography. This was accompanied by data regarding ground reaction forces obtained through the use of a force platform. After the completion of each condition subjects were required to identify and rate body discomfort. Differential analysis was used to isolate the muscle activity and ground reaction forces attributed to increased external load and increased trunk movement. It was found that the heaviest loading conditions (4kg) resulted in significantly greater (p<0.05) muscle activation in the majority of muscles during all tasks investigated. The trend of muscle activity attributed to load was similar in all significantly altered muscles and activation was greatest in the heaviest loading condition. A degree of movement efficiency occurred in some muscles when manipulating loads of 0.8kg and 1.6kg. At greater loads, this did not occur suggesting that heavier loading conditions result in additional strain on the body in excess of that imposed by task demands. In manipulated data, trend of vertical ground reaction forces increased with increased load in all tasks. Sagittal movement of the centre of pressure attributed to load was significantly affected in manipulated data in the second movement phase of the “hip shoulder” task and the second movement phase of the “hip twist” task. The “hip reach” task was most affected by increased load magnitude as muscle activity attributed to load was significantly different (p<0.05) under increased loading conditions in both movement phases in all muscles. Further, a significant interactional effect (p<0.05) between condition and data point was found in all muscles with the exception of the right and left lumbar erector spinae during the second movement phase of the “hip reach” task. Muscle activity associated with increased trunk motion resulted in additional strain on the trunk muscles in the “hip shoulder” and “hip reach” tasks as muscle activity associated with the static component of each of the above tasks was greater than that of the dynamic tasks. Trend of ground reaction forces attributed to increased trunk motion generally increased under increased loading conditions. - Description: The spatial and temporal patterns in selected components of the ichthyofauna and piscivorous avifauna in the small temporarily open/closed Riet River Estuary located on the eastern seaboard of southern Africa was investigated monthly over the period August 2005 to July 2006. The ichthyofauna within the littoral zone of the estuary was sampled using a 5 m seine net (8 stations) while a 30 m seine net (4 stations) was employed to sample the fish in the channel. Bird counts were made along repeat transects along the length of the estuary. Total ichthyofaunal abundances and biomass ranged between 1.60 and 8.67 individuals m⁻² and 0.45 to 21.76 g wwt m⁻² within the littoral zone, and between 0.08 and 0.44 individuals m⁻² and 0.58 and 36.52 g wwt m⁻² in the channel of the estuary. The highest values were generally recorded during the summer months. Results of the numerical analysis indicated that the breaching events recorded over the study period did not lead to a common trend in the ichthyofaunal community. In the absence of a link to the marine environment, the ichthyofaunal community in the littoral zone was numerically dominated by the estuarine resident species, Gilchristella aestuaria and to a lesser extent by Glossogobius callidus, which collectively accounted for ca. 54% of the total ichthyofauna sampled. The establishment of a link to the marine environment coincided with increased numbers of marine breeding species including Atherina breviceps and Rhabdosargus holubi to total fish counts within the estuary. Hierarchical cluster analysis did not identify any spatial patterns in the community structure of the ichthyofauna in the littoral zone or channel zone of the estuary, which could likely be linked to the absence of any distinct horizontal patterns in salinity and temperature within the system. A total of thirteen piscivorous bird species was recorded over the study period. Of the recorded species, six species were wading piscivores, four species were aerial divers and the remaining three species were pursuit swimmers. There were no significant correlations between the estimates of the ichthyofaunal abundance and biomass and bird numbers evident during the study (P> 0.05 in both cases). The Reed Cormorant (Phalacrocorax africanus) was the dominant species throughout the study, with a mean of 8.25 (SD ± 7.90) individuals per count. Mean values of the Pied Kingfisher (Ceryle rudis) and Giant Kingfisher (Megaceryle maximus) were 3.42 (SD ± 1.20) and 1.17 (SD ± 0.60) individuals per count, respectively. The remaining species revealed mean values < 0.5 individuals per count. The highest bird numbers were recorded in winter reflecting the migration of large numbers of the Reed Cormorant into the system. Breaching events were associated with a decrease in total bird numbers, which was most likely due to loss of potential foraging habitat (littoral zone) for waders resulting from reduced water levels. Monthly food consumption by all piscivorous birds revealed large temporal variability, ranging from 26.35 to 140.58 kg per month. The observed variability could be linked to mouth phase and bird numbers.

- Description: Municipal commonage in South Africa offers previously disadvantaged, landless residents access to both direct ecosystem goods and services (EGS) that provide additional income options and indirect social and cultural services. Given that EGS production is a function of ecosystem health, it is imperative that commonage land be managed to maximize current local benefit streams while ensuring future options through the maintenance of natural ecosystem functions. The payments for ecosystem services (PES) model potentially offers an opportunity for contributing to local economic development while providing fiscal incentives for environmentally sustainable natural resource management. PES depends on the demonstration of quantifiable changes in EGS delivery due to improvement in or maintenance of high ecosystem health that are a verifiable result of modifications in management behavior. This thesis therefore compared spatial variations in (i) ecosystem health and (ii) nine direct and indirect EGS values derived from natural resources on the Bathurst municipal commonage and neighboring Waters Meeting Nature Reserve (NR) to explore how different land use intensities affect ecosystem health and the resulting provision of EGS. The results indicate that the total economic value of annually produced EGS on the study site is nearly R 9.8 million (US$ 1.2 million), with a standing stock of natural capital worth some R 28 million (US$ 3.4 million). Nearly 45% of the total annual production is attributed to Waters Meeting NR, with roughly 34% from the low use zone of the commonage and the remaining 22% from the high use zone. Of the total annual production value on the study site, roughly 59% is derived from indirect (non-consumptive) uses of wildlife for the study site as a whole, though this proportion varies from 25% in the high use zone of the commonage to 94% on Waters Meeting NR. The two largest annual production values on the study site derive from ecotourism (R 3.5 million, US$ 0.4 million) and livestock production (R 2.6 million, US$ 0.3 million), suggesting that while increased production of indirect EGS could generate significant additional revenues, especially on Waters Meeting NR and in the low use zone of the commonage, direct (consumptive) EGS will likely remain an important component of land use on the commonage. A PES project to support the adoption of silvo-pastoral practices could provide positive incentives for improved land use practices on the commonage and potentially be financed by conservation-friendly residents of the Kowie River catchment and/or increased ecotourism revenues from Waters Meeting NR. Allowing carefully designed and monitored local access to natural resources within Waters Meeting NR could also reduce pressure on commonage resources. Together, these approaches could lead to a more sustainable subtropical thicket landscape and ensure that critical natural resources remain available to support local livelihoods in the long-term.

- Description: Municipal commonage in South Africa offers previously disadvantaged, landless residents access to both direct ecosystem goods and services (EGS) that provide additional income options and indirect social and cultural services. Given that EGS production is a function of ecosystem health, it is imperative that commonage land be managed to maximize current local benefit streams while ensuring future options through the maintenance of natural ecosystem functions. The payments for ecosystem services (PES) model potentially offers an opportunity for contributing to local economic development while providing fiscal incentives for environmentally sustainable natural resource management. PES depends on the demonstration of quantifiable changes in EGS delivery due to improvement in or maintenance of high ecosystem health that are a verifiable result of modifications in management behavior. This thesis therefore compared spatial variations in (i) ecosystem health and (ii) nine direct and indirect EGS values derived from natural resources on the Bathurst municipal commonage and neighboring Waters Meeting Nature Reserve (NR) to explore how different land use intensities affect ecosystem health and the resulting provision of EGS. The results indicate that the total economic value of annually produced EGS on the study site is nearly R 9.8 million (US$ 1.2 million), with a standing stock of natural capital worth some R 28 million (US$ 3.4 million). Nearly 45% of the total annual production is attributed to Waters Meeting NR, with roughly 34% from the low use zone of the commonage and the remaining 22% from the high use zone. Of the total annual production value on the study site, roughly 59% is derived from indirect (non-consumptive) uses of wildlife for the study site as a whole, though this proportion varies from 25% in the high use zone of the commonage to 94% on Waters Meeting NR. - Description: Despite the many changes that have occurred in South Africa since the end of apartheid, there are still residual effects of it, as is evidenced in the disparity of living conditions between different racial groups. It is also evident that there are differences in the work tasks and working conditions of nurses working in different work environments. This project looks at how living conditions as well as working conditions interactively affect the health, subjective well-being, and work ability of nurses. Questionnaires were completed by, and interviews were conducted with nurses from Settlers Hospital and seven municipal clinics within Grahamstown (n=152). The participation rate was approximately 71%. The questionnaires included self-report, forced-choice questions regarding basic demographics of the nurses, work conditions, living conditions, subjective satisfaction levels, as well as a simplified version of the Nordic Questionnaire of Musculoskeletal Strain (Kuorinka et al., 1987), and the Work Ability Index (WAI) (Tuomi et al., 2006). The questionnaires were translated into Afrikaans and IsiXhosa. One-on-one interviews were conducted with the participants, in order to obtain a 24-hour dietary recall, an indication of physical activity levels, as well as measurements of stature, mass, waist girth and hip girth. Factor analysis was performed to identify common variance from amongst the variables, while canonical correlations examined the interaction between the sets of factors. It was found that variables relating to demographic factors, living conditions, and working conditions were closely linked to each other. Factors from each of these groups were associated with life, health, and job satisfaction, anthropometric measures, musculoskeletal strain, and WAI scores. Satisfaction levels appeared to be largely determined by socioeconomic status, while anthropometrics, WAI scores, and levels of musculoskeletal strain were associated with levels of smoking and drinking, race, age, stature, position and tenure.

It has been found that hand injuries are among the most frequent injuries that occur to the body, predominantly during industrial activities. It has therefore been concluded that more research is needed into protective factors, such as glove use. The design features of a glove emphasise either protection or performance. There is often a trade-off between increased safety and performance capability when donning gloves. It has been determined that gloves which are fitted and comfortable for the worker may provide the best compromise between protective functions and decreased performance. This investigation aimed to assess the influence of glove fit on the performance attributes of industrial tasks, as well as on the responses of the human operator. Glove fit was analysed as 35 male participants donned three different glove sizes during each test, including a best-fitting glove, a glove one size smaller than best-fitting, and a glove one size larger than best-fitting. For each glove size, gloves of two differing materials were tested, namely nitrile and neoprene. A barehanded condition was also tested, totalling seven gloved/barehanded conditions for each test. The seven conditions were assessed in a laboratory setting in a battery of tests. This consisted of components of task performance, including maximum pulling and pushing force, maximum torque, precision of force, tactility, speed and accuracy and dexterity. The performance responses were recorded, as well as participants’ perceptual responses using the Rating of Perceived Exertion scale, and muscle activity. Six muscles were selected: Flexor Digitorum Superficialis, Flexor Pollicus Longus, Extensor Carpi Ulnaris, Extensor Carpi Radialis, Flexor Carpi Ulnaris and Flexor Carpi Radialis. The results revealed that glove fit does affect certain spects of performance, and influences human operator responses for selected task components. Furthermore, discrepancies were distinguished between orking barehanded and working with an optimally fitted glove. There was also a glove material effect established. Overall, it was found that muscle activity when exerting maximum force in a pushing and pulling direction was optimal with the nitrile glove material. Maximum torque performance was enhanced with the use of a best-fitting glove, as compared with an ill-fitting glove or barehanded work. Force precision was preferable when barehanded, as opposed to the tactility task which rendered optimal results with a best-fitting glove. The same was found for speed and accuracy results, as glove fit appeared to have no effect on performance, but performance was improved when participants were barehanded. Dexterity performance was the most conclusively influenced by the conditions, resulting in barehanded performance being optimal. However, should a glove be necessary for a given task, an optimally-fitted glove which is of a thinner material would be recommended. - Description: The Picornavirus family of positive sense RNA viruses includes some significant human and animal pathogens including Poliovirus (PV), Foot-and-Mouth disease virus (FMDV) and Human Rhinovirus (HRV). The genome is translated within the host cell into a polyprotein that is proteolytically cleaved into the structural and nonstructural proteins. The highly conserved, non-structural protein 2C has numerous roles during the virus life cycle and is essential for virus replication. Although the protein has been well studied in the case of PV, its interactions with the host cell during picornavirus infection is poorly understood. Theiler’s Encephalomyelitis virus (TMEV) is a picornavirus that infects mice, and is being used in our laboratory as a model in which to study the 2C protein. In this study, polyclonal antibodies against the TMEV 2C protein were generated and used to localise the protein in infected cells by indirect immunofluorescence. To produce antigen for immunisation purposes, the TMEV-2C protein sequence was analysed to identify hydrophilic and antigenic regions. An internal region of the 2C representing amino acid residues 31-210 was selected, expressed in bacteria and purified by nickel NTA affinity chromatography. Time course analysis of 2C (31-210) showed that the peptide was maximally expressed at 5 hours post induction. The peptide was solubilised using a mild detergent and 1.5 mg of purified antigen was used for immunisation of rabbits. Western blot analysis confirmed that the antibodies could detect both bacteriallyexpressed antigen, and virally-expressed 2C. Examination of virus-infected baby hamster kidney cells by immunofluorescence and confocal microscopy using the antiserum (anti-TMEV 2C antibodies) showed that the protein had a diffuse distribution upon early infection and at later stages it was located in a large perinuclear structure representing the viral replication complex. Furthermore, 2C localised to the Golgi apparatus as revealed by dual-label immunofluorescence using anti-TMEV 2C antibodies and wheat germ agglutinin (WGA). Furthermore, it was shown that TMEV infection results in changes in cell morphology and a redistribution of the cytoskeletal protein, β-actin. The successful production of antibodies that recognise TMEV 2C opens the way for further studies to investigate interactions between 2C and hostencoded factors.

The impact of a one-hour self-selected nap opportunity on physiological and performance variables during a simulated night shift

- Description: Napping has been explored extensively as a means of counteracting the negative effects associated with shift work. A significant amount of this research has focused on the implementation of scheduled naps, with few studies considering flexible nap schemes. The current study therefore aimed to assess the effects of a flexible nap opportunity on the physiological, cognitive, performance, neurophysiological and subjective responses of a group of non shift workers over the course of a three-day simulated night shift regime. Additional foci were the effects of the nap condition on the extent of the circadian adaptation of the subjects to the irregular work schedule and the circadian-related influences associated with being awake during the night. 36 subjects – 18 males and 18 females – were recruited to participate in the current study. The data collection spanned twelve days, during which four, three-day long shift cycles were set up: three night shift cycles and one day shift cycle. During each night shift cycle, three separate experimental conditions were staggered, namely the nap condition, the no nap condition and a booster break condition (a collaborative study that completed the setup). The day shift served as a further comparison. Each cycle comprised of 12 subjects, which meant there were four subjects per condition during each cycle. The shifts were 8 hours in duration, with the no nap group following a standard break schedule evinced in industry. The three breaks taken during the shifts amounted to a total time of 1 hour. The nap group was afforded a 1 hour flexible nap opportunity between 00h00 and 03h00 with no other breaks. Therefore, both conditions had the same amount of work time. During the shifts, subjects performed two simple, low arousal tasks (beading and packing) and completed a test battery roughly every two hours which was comprised of physiological, performance, neurophysiological and subjective measures. It was found that the inclusion of the nap opportunity significantly improved output performance and response time during a low precision, modified Fitts tapping task over the course of three night shifts, relative to no napping. Physiologically, napping resulted in higher heart rate frequency measures by the end of the shifts, which were also accompanied by significant reductions in subjective sleepiness ratings during all iii the night shifts. The nap group’s responses in this case, did not differ significantly from those of the day shift. Both simple reaction time and memory performances improved as a result of the nap inclusion, but only during the third night shift. The majority of the measures included in the research also depicted the effects of the circadian rhythm, which was indicative of the pronounced effect that this natural biological down regulation has on performance during the night. Napping reduced the severity of these effects during beading performance and measures of subjective sleepiness. With regard to habituation, the nap opportunity also resulted in positive changes in the responses of beading performance, high precision response time, simple reaction time and both subjective sleepiness measures, relative to no napping. Sleep diary responses indicated that although sleep length and quality during the day were significantly reduced for both night-time conditions, recovery sleep (length and quality) for the nap group did not differ significantly from the no nap group. The findings of this research indicate that the inclusion of a flexible napping opportunity during the night shift had positive effects on some physiological, performance and subjective responses, and that this intervention is as beneficial as scheduled napping. Specifically, napping resulted in a significantly higher output during the beading task, relative to the no nap group despite the duration of work time being the same. As such the introduction of a flexible, self-selected nap opportunity is a practical, effective and individual-specific means of alleviating the negative effects of shift work, while improving certain performance parameters. Therefore, industries should consider its inclusion in their fatigue management programs. However, contextspecific considerations must be made, with regard work scheduling, individual differences and task demands when implementing such an intervention. This will ensure that its introduction will be well received and in time, lessen the health and work-related decrements associated with shift work.

The current study therefore aimed to assess the effects of a flexible nap opportunity on the physiological, cognitive, performance, neurophysiological and subjective responses of a group of non shift workers over the course of a three-day simulated night shift regime. Additional foci were the effects of the nap condition on the extent of the circadian adaptation of the subjects to the irregular work schedule and the circadian-related influences associated with being awake during the night. 36 subjects – 18 males and 18 females – were recruited to participate in the current study. The data collection spanned twelve days, during which four, three-day long shift cycles were set up: three night shift cycles and one day shift cycle. During each night shift cycle, three separate experimental conditions were staggered, namely the nap condition, the no nap condition and a booster break condition (a collaborative study that completed the setup). The day shift served as a further comparison. Each cycle comprised of 12 subjects, which meant there were four subjects per condition during each cycle. The shifts were 8 hours in duration, with the no nap group following a standard break schedule evinced in industry. The three breaks taken during the shifts amounted to a total time of 1 hour. The nap group was afforded a 1 hour flexible nap opportunity between 00h00 and 03h00 with no other breaks. Therefore, both conditions had the same amount of work time. During the shifts, subjects performed two simple, low arousal tasks (beading and packing) and completed a test battery roughly every two hours which was comprised of physiological, performance, neurophysiological and subjective measures. It was found that the inclusion of the nap opportunity significantly improved output performance and response time during a low precision, modified Fitts tapping task over the course of three night shifts, relative to no napping. Physiologically, napping resulted in higher heart rate frequency measures by the end of the shifts, which were also accompanied by significant reductions in subjective sleepiness ratings during all iii the night shifts. The nap group’s responses in this case, did not differ significantly from those of the day shift. Both simple reaction time and memory performances improved as a result of the nap inclusion, but only during the third night shift. The majority of the measures included in the research also depicted the effects of the circadian rhythm, which was indicative of the pronounced effect that this natural biological down regulation has on performance during the night. Napping reduced the severity of these effects during beading performance and measures of subjective sleepiness. With regard to habituation, the nap opportunity also resulted in positive changes in the responses of beading performance, high precision response time, simple reaction time and both subjective sleepiness measures, relative to no napping. Sleep diary responses indicated that although sleep length and quality during the day were significantly reduced for both night-time conditions, recovery sleep (length and quality) for the nap group did not differ significantly from the no nap group. The findings of this research indicate that the inclusion of a flexible napping opportunity during the night shift had positive effects on some physiological, performance and subjective responses, and that this intervention is as beneficial as scheduled napping. Specifically, napping resulted in a significantly higher output during the beading task, relative to the no nap group despite the duration of work time being the same. As such the introduction of a flexible, self-selected nap opportunity is a practical, effective and individual-specific means of alleviating the negative effects of shift work, while improving certain performance parameters. Therefore, industries should consider its inclusion in their fatigue management programs. However, contextspecific considerations must be made, with regard work scheduling, individual differences and task demands when implementing such an intervention. This will ensure that its introduction will be well received and in time, lessen the health and work-related decrements associated with shift work.

The NMR studies with dual substituent parameters suggest that the effects of the substituents are transimitted through the ligands, via resonance effects and that the phenyl group is nonplanar with the azomethine in N, N‟-(aryl)benzyaldiimine ligands. The presence of an extra double bond in Schiff base {(N, N‟-bis(cinnamaldiimine) ligand)} altered the electron density. The UV/vis studies showed that the symmetry of the N, N‟-bis(4-R-benzyl)-1, 2-diaminoethanedihalidecopper(II) complexes were predominantly tetrahedral for both chloro and bromo complexes. The correlation studies from mid-infrared were beneficial in monitoring the effect experienced by N, N‟-(aryl)benzaldiimine ligands, the studies suggest that the inductive effect is more pronounced at the C=N.

These values indicated highly favored Hg²⁺ extractions over the two competing ions. Application of the polymer to various real water samples (tap, sea, river, pulverized coal solution, treated and untreated sewerage from the vicinity of Grahamstown in South Africa) showed high extraction efficiencies (EEs) of Hg²⁺ ions; (over 84% in all cases) as evaluated from the detected unextracted Hg²⁺ ions by inductively coupled plasma optical emission spectroscopy (ICP-OES). The limit of detection (LOD, 3ơ) of the method was evaluated to be 0.036 ng ml⁻¹ and generally the data (n=10) had percentage relative standard deviation (%RSD) of less than 4%. Microbial ecology of the Buffalo River in response to water quality changes

- Description: South Africa’s freshwater quality and quantity is declining and consequently impacting on the ecological health of these ecosystems, due to increased agricultural, urban and industrial developments. The River Health Programme (RHP) was designed for monitoring and assessing the ecological health of freshwater ecosystems in South Africa, in order to effectively manage these aquatic resources. The RHP utilises biological indicators such as in-stream biota as a structured and sensitive tool for assessing ecosystem health. Although the RHP has been widely implemented across South Africa, no attempts have been made to explore microbial ecology as a tool that could be included as one of the RHP indices. This study used selected microbial responses and water physico-chemical parameters to assess the current water quality status of the Buffalo River. This study showed that water quality impairments compounded in the urban regions of King William’s Town and Zwelitsha and also downstream of the Bridle Drift Dam. The results also showed that the lower and the upper catchments of the Buffalo River were not significantly different in terms of water physico-chemistry and microbiology, as indicated by low stress levels of an NMDS plot. Though similarities were recorded between impacted and reference sites, the results strongly showed that known impacted sites recorded the poorest water physico-chemistry, including the Yellowwoods River. However, the Laing Dam provided a buffer effect on contributions of the Yellowwoods River into the Buffalo River. Multivariate analysis showed that microbial cell counts were not influenced by water physico-chemical changes, whilst microbial activity from the water and biofilm habitats showed significant correlation levels to water physico-chemical changes. This study demonstrated that further investigations towards exploitation of microbial activity responses to water physico-chemical quality changes should be channelled towards the development of microbiological assessment index for inclusion in the RHP.

Lack of success has been ascribed to varietal differences, climate and predation of agents but these have not been tested. In this study, the effects of ant predation were tested on populations of three biological control agents for L. camara. Colonies of two species, Crematogaster sp. 1 and 2 were investigated. Crematogaster sp. 1 colonies were offered no choice between immature stages of the agents Hypena laceratalis Walker (Lepidoptera: Noctuidae), Falconia intermedia Distant (Hemiptera: Miridae) or Teleonemia scrupulosa Stål (Hemiptera: Tingidae) on lantana shoots. Density-dependent predation on F. intermedia and T. scrupulosa nymphs on lantana shoots was tested using Crematogaster sp. 2 colonies. In choice experiments Crematogaster sp. 2 colonies were offered F. intermedia or T. scrupulosa nymphs on potted lantana plants. Preliminary food trials confirmed that colonies foraged for protein, thereby validating results of no-choice experiments. Crematogaster sp.1 foragers removed 50% of F. intermedia nymphs, followed by 45% of H. laceratalis larvae and only 9% of T. scrupulosa nymphs. Foragers recruited most actively to H. laceratalis larvae and significantly more H. laceratalis biomass was removed than either F. intermedia or T. scrupulosa. A trade-off existed in prey size selection because larger larvae provided considerably more biomass but required forager cooperation and a longer time to subdue than did smaller prey. This increases both forager energy expense and mortality risk by other predators. This study showed that all Crematogaster sp. 1 colonies removed small (≤10mm) H. laceratalis larvae more frequently than larvae larger than 10mm. Thus, of these biological control agents, predators probably prefer small H. laceratalis larvae. Significantly more F. intermedia than T. scrupulosa nymphs were removed by Crematogaster sp. 1, while Crematogaster sp. 2 colonies removed comparable numbers of both agent species. Falconia intermedia nymphs' fast movement triggered a predatory response by these ant species. In contrast, the relatively immobile behaviour of T. scrupulosa nymphs was identified as a highly effective predator avoidance strategy. Since T. scrupulosa nymphs are unable to escape predators by moving, they appear to depend on the presence of alternative prey attracting predator attention. At high agent and/or forager density, T. scrupulosa nymphs attempted escape, but foragers identified them as prey once they moved and caught them. Predation on F. intermedia was also density dependent in that at high nymph and/or forager densities, escape routes were congested and nymphs were more easily caught. Survival of F. intermedia and T. scrupulosa nymphs in particular was low on ant-accessed shrubs in choice experiments and high on ant-excluded shrubs. It is likely that ants significantly depress F. intermedia populations in the field since besides predation, ant foragers probably interrupt F. intermedia feeding and ovipositioning. The combination of parasitism and predation on early instar larvae may explain why H. laceratalis occurs across lantana's range in South Africa but populations remain low. It is unlikely that T. scrupulosa nymphs are habitually preyed on by ant species unless they attract attention by being mobile. Although biological control of L. camara is influenced by climate and physiological defence mechanisms, this study has shown that predation by two ant species severely impacts leaf-feeding agents for L. camara. Thus, it is recommended that future selection of additional agents to control lantana should exclude leaf-feeding .

It is likely that ants significantly depress F. intermedia populations in the field since besides predation, ant foragers probably interrupt F. intermedia feeding and ovipositioning. The combination of parasitism and predation on early instar larvae may explain why H. laceratalis occurs across lantana's range in South Africa but populations remain low. It is unlikely that T. scrupulosa nymphs are habitually preyed on by ant species unless they attract attention by being mobile. Although biological control of L. camara is influenced by climate and physiological defence mechanisms, this study has shown that predation by two ant species severely impacts leaf-feeding agents for L. camara. Thus, it is recommended that future selection of additional agents to control lantana should exclude leaf-feeding .

Their structural features have been deduced from their elemental analytical data, IR spectral data, and electronic spectral data. - Description: The project has focused on the preparation of several series of compounds designed as potential HIV-1 integrase inhibitors. Various 2-nitrobenzaldehydes have been reacted with two activated alkenes, methyl vinyl ketone (MVK) and methyl acrylate, under Baylis-Hillman conditions to afford α-methylene-β-hydroxylalkyl derivatives in moderate to excellent yields. The reactions were conducted using the tertiary amine catalysts, 1,4-diazabicyclo[2.2.2]octane(DABCO) or 3-hydroxyquinuclidine (3-HQ) with chloroform as solvent, and yields were optimised by varying the catalyst, reagent concentrations and the reaction time. Reductive cyclization of the Baylis-Hillman adducts via catalytic hydrogenation, using 10% palladiumon-carbon catalyst in ethanol, afforded quinoline and quinoline N-oxide derivatives. In some cases “acyclic” reduction products were also isolated. Reaction of the Baylis-Hillman MVK adducts with HCl, has resulted in effective nucleophilic (SN’) displacement of the hydroxyl group to afford allylic chloride derivatives. Direct substitution of these chloro derivatives by secondary or primary amines, followed by catalytic hydrogenation gave quinoline derivatives containing a 3-aminomethyl substituent. The Baylis-Hillman ester adducts obtained from reaction with methyl acrylate were treated directly with various amines to give diastereomeric conjugate addition products. Reactions with piperazine gave N,N’-disubstituted piperazine products. The piperidine derivatives have been dehydrated to give cinnamate esters in moderate yields. The products, which have all been satisfactorily characterised by elemental (HRMS) and spectroscopic (1- and 2-D NMR) analysis, constitute a “library” of compounds for in silico and in vitro studies as potential HIV integrase inhibitors.

- Description: The sugar beet (Beta vulgaris) plant has in recent years been added to the Biofuel Industrial Strategy (Department of Minerals and Energy, 2007) by the South African government as a crop grown for the production of bio-ethanol. Sugar beet is commonly grown in Europe for the production of sucrose and has recently been cultivated in Cradock and the surrounding areas (Engineering News, 2008). The biofuel industry usually ferments the sucrose with Saccharomyces cerevisiae to yield bio-ethanol. However, researchers are presented with a critical role to increase current yields as there are concerns over the process costs from industrial biotechnologists. The beet factories produce a pulp by-product removed of all sucrose. The hemicellulose-rich pulp can be degraded by microbial enzymes to simple sugars that can be subsequently fermented to bio-ethanol. Thus, the pulp represents a potential source for second generation biofuel. The process of utilising microbial hemicellulases requires an initial chemical pre-treatment step to delignify the sugar beet pulp (SBP). An alkaline pre-treatment with ‘slake lime’ (calcium hydroxide) was investigated using a 23 factorial design and the factors examined were: lime load; temperature and time. The analysed results showed the highest release of reducing sugars at the pre-treatment conditions of: 0.4 g lime / g SBP; 40°C and 36 hours. A partial characterisation of the Clostridium cellulovorans hemicellulases was carried out to verify the optimal activity conditions stated in literature. The highest release of reducing sugars was measured at pH 6.5 – 7.0 and at 45°C for arabinofuranosidase A (ArfA); at pH 5.5 and 40°C for mannanase A (ManA) and pH 5.0 – 6.0 and 45°C for xylanase A (XynA). Temperature studies showed that a complete loss of enzymatic activity occurred after 11 hours for ManA; and 84-96 hours for ArfA. XynA was still active after 120 hours. The optimised lime pre-treated SBP was subsequently degraded using various combinations and percentages of C. cellulovorans ArfA, ManA and XynA to determine the maximal release of reducing sugars. Synergistically, the highest synergy was observed at 75% ArfA and 25% ManA, with a specific activity of 2.9 μmol/min/g protein. However, the highest release of sugars was observed at 4.2 μmol/min/g protein at 100% ArfA. This study has initiated the research within South Africa on SBP and its degradation by C. cellulovorans. Preliminary studies show that SBP has the potential to be utilised as a second generation biofuel source.

- Description: Austroglanis gilli and A. barnardi are endemic to the Clanwilliam-Olifants System in the Western Cape, South Africa. The populations of each of these species are considered to be threatened by various anthropogenic activities, which include inappropriate agricultural practice and impacts of alien invasive fish species. The purpose of this thesis was to assess the life-history of these two endangered species in order to contribute to understanding their biology. Such information is vital for the development of strategies for their conservation. Marginal zone and marginal increment analyses from sectioned lapilliar otoliths of both A. gilli and A. barnardi showed a unimodal peak, suggesting a single annulus formation. The oldest specimens of A. gilli and A. barnardi were 12+ and 14+ years, respectively. The growth of A. gilli and A. barnardi was relatively slow and was best described by the von Bertalanffy growth curve as: L, = 131.56(1 - exp(- 0.27(t - 1.18 ))) for male and L, = 113.86(1 - exp(- 0.43(1- 0.74))) for female A. gilli from the Rondegat River. In the Noordhoeks River, growth was L, = 99.67(1 - exp(- 0.53(t - 0.35))) for male and L, = 96.60(1 - exp(- 0.64(t - 0.11 ))) for female A. gilli, and L, =71.02(1-exp(-0.26(1-3.07))) and L, = 69.50(1-exp(-0.36(1 - 1.88))) for male and female A. barnardi, respectively. The average natural mortality for the combined sexes was estimated using catch curve analysis at 0.37 ± 0.12 per year for A. gilli from the Rondegat River, and at 0.71 ± 0.05 and 0.39 ± 0.04 per year for A. gilli and A. barnardi from the Noordhoeks River, respectively. For A. gilli in the Rondegat River, the first maturity was estimated at 3.3 years (97.3 mm SL) for males and 3.1 years (94.3 mm SL) for females. In the Noordhoeks River, the first maturity for A. gilli was estimated at 2.0 years (71.9 mm SL) for males and 1.7 (66.4 mm SL) for females, and for A. barnardi at 2.0 (55.0 nun SL) for males and 2.9 years (58.9 mm SL) for females. Maturity corresponded closely to the asymptotic sizes from the von Bertalanffy curves, suggesting a shift in energy use from somatic growth to gonad development. Macroscopic assessment of the state of gonads, the Ganado-somatic index and histological examinations revealed that both A. gilli and A. barnardi have a single spawning season. The presence of oocytes in different stages of development in each of the ovaries of mature females collected between November and January suggested asynchronous, iteroperous serial spawning. The resorption of yolk was observed from ovaries collected between February and March and no vitellogenic oocyte was visible from ovaries collected between April and August. A similar trend was observed for males, with spermatozoa filling the lumen between November and January and some residual spermatozoa being present in the lumens soon after the breeding season. Spermatocytes and spermatids were dominant in the testes until October. An Index of Relative Importance revealed that A. gilli feeds predominantly on the benthic macroinvertebrate larvae of Ephemeroptera (particularly Baetidae), Diptera (particularly Chironomidae and Simuliidae) and Trichoptera. Austroglanis barnardi feeds predominantly on dipteran larvae (particularly Chironornidae and Simuliidae). Chi square contingency tables showed a significant difference in the dominant prey items of A. gilli and A. barnardi in the Noordhoeks River (X₂= 53.79, d.f. = 4, p > 0.001) and A. gilli between Rondegat and Noordhoeks rivers (x₂ = 34.74, d.f. = 4, p > 0.001). The Spearman's rank correlation test showed no shifts in the diet of A. barnardi from Noordhoeks River and A. gilli from Rondegat River with a change in size and season (p>O.OS). However, there was a shift in the diet of A. gilli from the Noordhoeks River which could suggest a distinct patchiness of benthic macroinvertebrates between the riffle feeding areas used by juveniles and other biotopes used by adults. The occurrence, in stomach contents, of other prey items from a wide variety of taxa and the presence of allochthonous material from the terrestrial environment could suggest an opportunistic feeding guild for both Austroglanis species. The life-history traits of A. gilli and A. barnardi, which are charaterized by slow growth, long life span and low relative fecundity, indicate that both species are relatively precocial and K-selected. The population of a precocial species is relatively stable and if population numbers were to be greatly reduced, they would require a long time to rebuild. An urgent conservation intervention is therefore recommended for Austroglanis spp. so as to maintain the diversity of populations within these species. The creation of protected river reserves and raising public conservation awareness may minimise activities that result in altered river hydrology and the destruction of complex benthic habitats.

- Description: Austroglanis gilli and A. barnardi are endemic to the Clanwilliam-Olifants System in the Western Cape, South Africa. The populations of each of these species are considered to be threatened by various anthropogenic activities, which include inappropriate agricultural practice and impacts of alien invasive fish species. The purpose of this thesis was to assess the life-history of these two endangered species in order to contribute to understanding their biology. Such information is vital for the development of strategies for their conservation. Marginal zone and marginal increment analyses from sectioned lapilliar otoliths of both A. gilli and A. barnardi showed a unimodal peak, suggesting a single annulus formation. The oldest specimens of A. gilli and A. barnardi were 12+ and 14+ years, respectively. The growth of A. gilli and A. barnardi was relatively slow and was best described by the von Bertalanffy growth curve as: L, = 131.56(1 - exp(- 0.27(t - 1.18 ))) for male and L, = 113.86(1 - exp(- 0.43(1- 0.74))) for female A. gilli from the Rondegat River. In the Noordhoeks River, growth was L, = 99.67(1 - exp(- 0.53(t - 0.35))) for male and L, = 96.60(1 - exp(- 0.64(t - 0.11 ))) for female A. gilli, and L, =71.02(1-exp(-0.26(1-3.07))) and L, = 69.50(1-exp(-0.36(1 - 1.88))) for male and female A. barnardi, respectively. The average natural mortality for the combined sexes was estimated using catch curve analysis at 0.37 ± 0.12 per year for A. gilli from the Rondegat River, and at 0.71 ± 0.05 and 0.39 ± 0.04 per year for A. gilli and A. barnardi from the Noordhoeks River, respectively. For A. gilli in the Rondegat River, the first maturity was estimated at 3.3 years (97.3 mm SL) for males and 3.1 years (94.3 mm SL) for females. In the Noordhoeks River, the first maturity for A. gilli was estimated at 2.0 years (71.9 mm SL) for males and 1.7 (66.4 mm SL) for females, and for A. barnardi at 2.0 (55.0 nun SL) for males and 2.9 years (58.9 mm SL) for females. Maturity corresponded closely to the asymptotic sizes from the von Bertalanffy curves, suggesting a shift in energy use from somatic growth to gonad development. - Description: Epidemiological studies consistently report that muscular strains are a primary injury type in rugby union with the majority of the strains occurring to the quadricep and hamstring musculature. Recently it has been suggested that poor eccentric muscular strength is a precursor to hamstring and quadriceps strains during intermittent sports that require rapid acceleration and deceleration. Despite the high incidence of these muscle injuries in Rugby Union there has been little research into the possible mechanisms involved. Thus, the purpose of this study was to measure the physiological and perceptual responses during a simulated Rugby Union laboratory protocol and further, to identify changes in muscle recruitment patterns and muscle strength over time by comparing this protocol to a continuous, constant load protocol covering the same distance. The experimental condition (EXP) required university level players to perform 80 minutes of simulated rugby union play in a laboratory setting (on a walkway of 22m) which was compared to that of a control condition (CON) which involved subjects covering the same distance, at a constant speed of 4.2km.h-1 on a treadmill. Physiological, biophysical and perceptual responses were measured pre-, at half-time and post-protocol. Heart rate was significantly (p<0.01) greater as a result of EXP in comparison to the CON. Electromyography (EMG) of the vastus medialis was significantly (p<0.01) greater during the CON protocol. The EXP condition elicited higher iEMG activity in the hamstring musculature at all time intervals. In addition the iEMG of the semitendinosus decreased significantly (p<0.01) as a result of the EXP protocol. Peak eccentric knee extensors (EXT) (-13.19%) and flexors (FLEX) (-12.81%) torque decreased significantly during the experimental protocol. After passive half-time (236.67 + 56.27Nm (EXT) and 173.89 + 33.3NM (FLEX)) and at the end of the protocol (220.39 + 55.16Nm and 162.89 + 30.66Nm) reduced relative to pre protocol (253.89 + 54.54Nm and 186.83 + 33.3Nm). Peak eccentric knee extensors did not change during the control protocol. „Central‟ and ‟Local” Rating of Perceived Exertion values were significantly (P<0.01) greater during the EXP protocol with an increased incidence of hamstring discomfort and perceived pain (5 out of 10). The EXP protocol resulted in significantly (p<0.01) increased incidence of delayed onset muscle soreness (DOMS). In conclusion, a stop-start laboratory protocol elicited increased heart rate, negatively impacted on muscle activity of the hamstrings, decreased eccentric strength in the lower limb musculature, resulted in increased ratings of „Central‟ and „Local‟ exertion and increased pain perception and increased incidence of DOMS. Thus, a stop-start rugby specific laboratory protocol has a negative impact on performance. Due to the specificity of the protocol being designed to match the demands of competitive match play it is expected that these changes in heart rate, muscle activity and strength, particularly eccentric strength, will impact negativity on performance during rugby match play and increase the likelihood of injury

- Description: Stroke is one of the most common causes of disability and death worldwide. The most commonly experienced stroke in the clinical setting is focal ischaemia in which the middle cerebral artery (MCA) is occluded and leads to a complex series of various pathophysiological pathways that ultimately lead to neuronal cell death. Several studies have been conducted on various therapeutic agents in the search for a neuroprotective drug and various animal models have been used to carry out this research. While theanine, a component of green tea and minocycline, a tetracycline antibiotic, have been shown to possess some neuroprotective properties, the mechanisms by which these two agents carry out these effects still remains unclear. The objectives of this study were to investigate the mechanisms by which these drugs carry out these neuroprotective effects and their neuroprotective ability in a MCA occlusion model of focal ischaemia. Ischaemia leads to oxidative stress due to the imbalance of free radicals and the endogenous antioxidant defence system. An antioxidant assay using the stable 2, 2-diphenyl-1-picrylhydrazyl (DPPH●) radical was used to assess the antiradical properties of each drug. It was found that minocycline showed superior antioxidant activity in vitro when compared to theanine. Further studies on the drugs‟ ability to attenuate the Fenton reaction (in which iron catalyses the formation of reactive species) were elucidated using electrochemical analysis, UV/VIS studies, ferrozine and ferritin assays. It was found that minocycline, in contrast to theanine, was able to bind to iron ions and thus potentially prevent the participation of iron in metal catalysed radical reaction. The antioxidant activity of both drugs was further investigated by assessing their effect on cyanide-induced superoxide generation and quinolinic acid (QA)-induced lipid peroxidation (LP). Experimental evidence shows that both drugs had no significant effect on the generation of superoxide in vitro and that there was a significant decrease in LP for minocycline in vitro and theanine in vivo. The metal binding and antioxidant properties were postulated to be a possible mechanism through which these agents reduced lipid peroxidation. A study was conducted to determine the effects of the drugs on the biosynthesis of the neurotoxin, QA and it was found that minocycline increases the levels of holoenzyme activity of tryptophan-2, 3-dioxygenase (TDO) in vitro and that theanine reduces the levels of the same enzyme in vivo after treatment for 10 days. TDO is the enzyme that converts tryptophan to other products that enable enzymatic activity to change it to QA. Minocycline was thought to bring about this effect as it has been shown from preceding experimental studies that it is an effective reducing agent. Theanine on the other hand is hypothesised to bring about a reduction in holoenzyme activity by changing the binding of tryptophan to the enzyme or affecting the radicals that participate in the enzymatic degradation of tryptophan. A focal ischaemic model of stroke was induced by occluding the MCA. Histological examination of the hippocampus post -ischaemia shows a reduction in the size of the infarct after pre-treatment with minocycline only. A further study into the effects of the drugs on the generation of superoxide and on the levels of the endogenous glutathione after a stroke was carried out. Pre-treatment of the animals with either theanine or minocycline showed no significant effects on the generation of the radical species or of the endogenous antioxidant which ruled out these as a mechanism of neuroprotection of both drugs, post-ischaemia.The findings of this study provide novel information on the possible mechanisms by which both theanine and minocycline act to bring about neuroprotection. In particular in this study, pre-treatment with minocycline has shown promise in the focal ischaemic model of stroke.

Geology of the Kroonstad kimberlite cluster, South Africa

- Description: The Cretaceous (133Ma) Kroonstad Group II Kimberlite Cluster is located approximately 200km south west of Johannesburg on the Kaapvaal Craton. The cluster is made up of six kimberlite pipes and numerous other intrusive dike/sill bodies. Three of the pipes are analysed in this study, which includes the: Voorspoed, Lace (Crown) and Besterskraal North pipes. These pipes were emplaced at surface into the Karoo Supergroup, which is comprised of older sedimentary rocks (300-185Ma) overlain by flood basalts (185Ma). At depth the pipes have intruded the Transvaal (2100-2600Ma) and Ventersdorp (2700Ma) Supergroups, which are comprised dominantly of carbonates and various volcanic units respectively. The pipes have typical morphology of South African pipes with circular to sub-circular plan views and steep 82o pipe margins. The Voorspoed pipe is 12ha in size and is characterised by the presence of a large block of Karoo basalt approximately 6ha in size at the current land surface. This large basalt block extends to a maximum of 300m below the current land surface. The main Lace pipe is 2ha is size with a smaller (<0.5ha) satellite pipe approximately 50m to the west. No information is available on the morphology of the Besterskraal North pipe as it is sub-economic and no mining has occurred. Samples from the Besterskraal North pipe were collected from the De Beers archives. The Kroonstad Cluster has been subjected to approximately 1750m of erosion post-emplacement, which has been calculated by the analysis of the crustal xenoliths with the pipe infill. The hypabyssal kimberlite from the three pipes shows a gradational evolution in magma compositions, indicated by the mineralogy and geochemistry. The Lace pipe is the least evolved and has characteristics more similar to Group I kimberlites. The Voorspoed and Besterskraal North kimberlite are intermediately and highly evolved respectively. The gradational evolution is marked by an increase in SiO2 and Na2O contents. Furthermore the occurrence of abundant primary diopside, aegirine, sanidine, K-richterite and leucite indicates evolution of the magma. The root zones of the pipes are characterised by globular segregationary transitional kimberlite, which is interpreted to be hypabyssal and not the result of pyroclastic welding/agglutination. The hypabyssal transitional kimberlite (HKt) is characterised by incipient globular segregationary textures only and the typical tuffisitic transitional kimberlite (TKt) end member (Hetman et al. 2004) is not observed. The HKt contact with the overlying volcaniclastic kimberlite (VK) infill is sharp and not gradational. The presence of HKt in the satellite blind pipe at Lace further indicates that the distinct kimberlite rock type must be forming sub-volcanically. The HKt is distinctly different at the Voorspoed and Lace pipes, which is likely a result of differing compositions of the late stage magmatic liquid. Microlitic clinopyroxene is only observed at the Lace HKt and is interpreted to form as a result of both crustal xenolith contamination and CO2 degassing. Furthermore the HKt is intimately associated with contact breccias in the sidewall. The root zones of the Kroonstad pipes are interpreted to form through the development of a sub-volcanic embryonic pipe. The volcaniclastic kimberlite (VK) infill of the Kroonstad pipes is not typical of South African tuffisitic Class 1 kimberlite pipes. The VK at Voorspoed is characterised by numerous horizontally layered massive volcaniclastic kimberlite (MVK) units, which are interpreted to have formed in a deep open vent through primary pyroclastic deposition. MVK is the dominant rock type infilling the Voorspoed pipe, however numerous other minor units occur. Normally graded units are interpreted to form through gravitational collapse of the tuff ring. MVK units rich in Karoo basalt and/or Karoo sandstone are interpreted to form through gravitational sidewall failure deep within an open vent. Magmaclasts are interpreted to form in the HKt during the development of an embryonic pipe and therefore the term autolith or nucleated autolith may be applied. Debate on the validity of the term nucleated autolith is beyond this study and therefore the term nucleated magmaclast is used to refer to spherical magmaclasts in the VK. The emplacement of the Kroonstad pipes is particularly complex and is not similar to typical Class 1 tuffisitic kimberlites. However the initial stage of pipe emplacement is similar to typical South African kimberlites and is interpreted to be through the development of an embryonic pipe as described by Clement (1982). The vent clearing eruption is interpreted to be from the bottom up through the exsolution of juvenile volatiles and the pipe shape is controlled by the depth of the eruption (+/-2km) (Skinner, 2008). The initial embryonic pipe development and explosive eruption is similar to other South African kimberlites, however the vent is cleared and left open, which is typical of Class 2 Prairies type and Class 3 Lac de Gras type pipes. The latter vent infilling processes are similar to Class 3 kimberlites from Lac de Gras and are dominated at the current level by primary pyroclastic deposition.

- Description: The Cretaceous (133Ma) Kroonstad Group II Kimberlite Cluster is located approximately 200km south west of Johannesburg on the Kaapvaal Craton. The cluster is made up of six kimberlite pipes and numerous other intrusive dike/sill bodies. Three of the pipes are analysed in this study, which includes the: Voorspoed, Lace (Crown) and Besterskraal North pipes. These pipes were emplaced at surface into the Karoo Supergroup, which is comprised of older sedimentary rocks (300-185Ma) overlain by flood basalts (185Ma). At depth the pipes have intruded the Transvaal (2100-2600Ma) and Ventersdorp (2700Ma) Supergroups, which are comprised dominantly of carbonates and various volcanic units respectively. The pipes have typical morphology of South African pipes with circular to sub-circular plan views and steep 82o pipe margins. The Voorspoed pipe is 12ha in size and is characterised by the presence of a large block of Karoo basalt approximately 6ha in size at the current land surface. This large basalt block extends to a maximum of 300m below the current land surface. The main Lace pipe is 2ha is size with a smaller (<0.5ha) satellite pipe approximately 50m to the west. No information is available on the morphology of the Besterskraal North pipe as it is sub-economic and no mining has occurred. Samples from the Besterskraal North pipe were collected from the De Beers archives. The Kroonstad Cluster has been subjected to approximately 1750m of erosion post-emplacement, which has been calculated by the analysis of the crustal xenoliths with the pipe infill. The hypabyssal kimberlite from the three pipes shows a gradational evolution in magma compositions, indicated by the mineralogy and geochemistry. The Lace pipe is the least evolved and has characteristics more similar to Group I kimberlites. The Voorspoed and Besterskraal North kimberlite are intermediately and highly evolved respectively. The gradational evolution is marked by an increase in SiO2 and Na2O contents. Furthermore the occurrence of abundant primary diopside, aegirine, sanidine, K-richterite and leucite indicates evolution of the magma. The root zones of the pipes are characterised by globular segregationary transitional kimberlite, which is interpreted to be hypabyssal and not the result of pyroclastic welding/agglutination. The hypabyssal transitional kimberlite (HKt) is characterised by incipient globular segregationary textures only and the typical tuffisitic transitional kimberlite (TKt) end member (Hetman et al. 2004) is not observed. The HKt contact with the overlying volcaniclastic kimberlite (VK) infill is sharp and not gradational. The presence of HKt in the satellite blind pipe at Lace further indicates that the distinct kimberlite rock type must be forming sub-volcanically. The HKt is distinctly different at the Voorspoed and Lace pipes, which is likely a result of differing compositions of the late stage magmatic liquid. Microlitic clinopyroxene is only observed at the Lace HKt and is interpreted to form as a result of both crustal xenolith contamination and CO2 degassing. Furthermore the HKt is intimately associated with contact breccias in the sidewall. The root zones of the Kroonstad pipes are interpreted to form through the development of a sub-volcanic embryonic pipe. - Description: The invasion of Acacia mearnsii in the Kouga catchment, Eastern Cape, South Africa, has various negative impacts on the ecosystem. These impacts include: reduced species richness, increased water use, increased nutrients and increased N cycling rates. The native shrubby fynbos vegetation has adapted to the acidic nutrient poor soils and Mediterranean climate of the Kouga Mountains. Fynbos, however, is currently being out competed by the much taller Acacia mearnsii trees, due to their competitive nature and ability to fix nitrogen, thereby enriching the soil. The invaded sections of the valley bottoms and lower hill slopes are characterised by an almost complete monoculture of Acacia mearnsii, with very few fynbos species still present. The Department of Water and Environmental Affairs sponsored Working for Water programme started clearing Acacia mearnsii in 1996 in the Kouga Mountains. Cleared sites have remained bare for long periods, indicating that soil properties are not favourable for indigenous propagule re-establishment. The aim of this research was to assess how A. mearnsii invasion and clearing affect fynbos recovery through its impact on soils. This was done by characterising vegetation and soil properties on fynbos, infested and cleared slopes. Vegetation cover for various growth forms was determined and a species list was compiled for each plot. The slope angle, surface hardness, litter cover, bare ground cover and soil depth were measured in the field, whereas water repellency, particle size and the chemical composition were measured in the laboratory. Furthermore, the plant establishment capacity of soils from fynbos, infested and cleared slopes was calculated. This was done by germinating fynbos seeds and growing fynbos plants in soils from the various slopes. The effect that invasion and clearing has on soil erosion was quantified using erosion plots on fynbos, infested and cleared slopes. The invasion and clearing of Acacia mearnsii led to an increase in soil nutrients, especially nitrogen, phosphorus, potassium, carbon and manganese. Furthermore, soils became more acidic, with increased water repellency and reduced surface hardness. The vegetation changed to a tree-dominated structure, replacing the native species. Native plant germination was relatively unaffected by invasion and clearing, with an increase in germination just after clearing. Plant growth of a native grass, Themeda triandra, and herb, Helichrysum umbraculigerum, has increased on soils from cleared slopes. This study showed that soil movement increased on slopes which are invaded and cleared of Acacia mearnsii, with erosion rates doubling on invaded slopes
