- Description: This study seeks to understand land use decision-making dynamics on large residential plots in Fingo Village, Makhanda (Grahamstown). Fingo Village was selected as a study area because it is one of the poorest urban settings in South Africa where urban poverty is observed alongside access to land. A dominant economic perspective not only suggests that land use decisions are motivated by economic motives, but also implies that access to land would enable people to generate income from its use to improve their livelihoods. This study argues against an uncritical embrace of this assumption. Lefebvre’s production of space thesis provides a holistic understanding of the factors involved in the making of land use decisions. The focus of this study is on the dialectic process in the spatial triad– spatial practice, representational space and representations of space. This involved the reading of government policies and legislation together with local lived experiences to gain an understanding of the particular spatial practices seen in Fingo Village. Snowball and convenience sampling were used to select 36 household plots in Fingo Village. Primary data was gathered by means of semi-structured interviews and participatory mapping while additional information was sourced from secondary sources and desktop analysis. The findings of the study show that there is no uniform pattern of land use and success. Different land use activities found on the selected residential plots, including the main house, backyard flat or flats, spaza shops, a funeral parlour, livestock keeping, cultural use (a kraal for ancestral worship) and food gardening. These activities are motivated by residents’ perceptions and attitudes towards their spaces, as well as the value and meaning they attach to the land which is not limited to economic factors, but is also influenced by socio-cultural, political and biophysical considerations. Although some spatial practices are prohibited by the government, they are important to the residents. Other participants fail to use the land as would be expected by a conventional economic perspective, due to spatial conflict relating to different interests as a result of collective land ownership and the failure of municipalities to enforce policies and regulations. The fact that numerous factors influence households’ land use decisions means that access to land does not always directly translate into economic benefits. It is all about what people think or do about their land, as well as what the state lays out in terms of policy and legislation, that will influence whether those people with large plots of land will ‘prosper’ or not. , Thesis (PhD) -- Faculty of Science, Geography, 2021

Findings from the study suggest that social entrepreneurs contribute immensely to poverty reduction. They also contribute towards research and development, promoting gender equality and empowerment, education for all as well as access to health facilities. The segments of the population benefiting from social entrepreneurship include the poor, socially excluded, discriminated, the unemployed and disabled. The impact on poverty and hunger was achieved through microfinance initiatives, income generation activities, empowerment and capacity building. Results showed that social entrepreneurship activities solve social problems through providing food, shelter, water, education and collateral to access finance. The study concluded that social entrepreneurship is a plausible approach to promote implementation of policies to reduce extreme poverty and hunger by using readily available resources to bring sustainable solutions to problems. - Description: South Africa has experienced a high and rising burden of non-communicable diseases (NCDs) and lifestyle risk factors over the past decade. Health as a category of human capital is generally ex-pected to influence an individual’s labour supply and productivity. Despite the increasing burden of non-communicable diseases, the high rate of economically inactive population and persistent wage inequalities in South Africa, there is limited empirical research on the effect of NCDs on labour force participation, employment status and wage differentials. Given this, the main object-ive of this study was to determine the effects of NCDs on three labour market outcomes: labour force participation, employment status and wage differentials in South Africa. This was divided into three major analytical objectives. Data used was extracted from the five waves of the National Income Dynamics Study, a nationally representative survey collected by the South African Labour and Development Research Unit (SALDRU). Several econometric tests, including cross-sectional data analysis, panel data analysis and the Blinder-Oaxaca decomposition methods, were used in the study. The first analytical objective focused on estimating the effect of lifestyle risk factors on labour force participation through NCDs by gender. Endogenous multivariate probit models with a recur-sive simultaneous structure were employed as a method of analysis. The empirical findings suggested that NCDs and associated risk factors have detrimental effect on labour force participation. The analysis was further expanded to analyse the effect of gender differences, considering that the effect of NCDs may be gender-specific. The results revealed that the effect of stroke and heart diseases were significant only for men, while diabetes and high blood pressure were only significant for women. The results also emphasised the significant indirect influence of obesity, physical inactivity, and alcohol consumption on labour force participation through NCDs, especially for men. The second analytical chapter focused on investigating the effect of NCDs on employment status – that is, those employed, unemployed and economically inactive in the population of South Africa by gender. The estimation technique known as generalised linear latent and mixed methods (GLLAMM) was employed to fit the multinomial logit model with correlated random intercept. The findings suggest that NCDs affect the economically inactive population significantly relative to those employed, and the magnitude is larger for women than for men. There was no significant difference found in the effect of NCDs on the unemployed relative to the employed segment of the population. In addition, the results revealed gender differences on the effect of NCDs on employment status and that stroke had a significant influence on the employment status of both sexes, while heart diseases had significant influence only in men, whereas diabetes had significant effects only in women. The last analytical chapter focuses on estimating the effect of NCDs on wage differentials in South Africa by gender. The recentred influence function regression model and Blinder-Oaxaca de-composition with RIF were used in the chapter. The empirical results revealed that the effect of NCDs on earnings differ by gender. It was found that women with NCDs earn less than those without NCDs, while men with NCDs were found to earn more than their counterparts without NCDs. The results further revealed that women with NCDs suffer from wage discrimination in South Africa. The policy implications of this study are gender-specific. The results highlight the necessity for undertaking a massive awareness campaign regarding the prevention and control of NCDs, espe-cially among women. This can be achieved through specific female health programmes, including maternal healthcare. The findings of the study imply largely that calls for gender-responsive health approaches which take into account gender-specific needs and priorities should be promoted, compared to a blanket approach. In addition, there is a need for the government to complement education policies to promote labour market outcomes. The role of national culture characteristics and business orientations in Tanzanian small business success

- Description: Despite numerous efforts to increase the business success of Tanzanian small businesses, it is still reported that progress in this regard has stagnated. The desired success in the SMME sector, and small businesses, is not being achieved, and is one of the reasons poverty in Tanzania remains significantly high. Researchers have suggested that low-income countries (LICs) such as Tanzania often do not achieve the desired growth of business enterprises due to insufficient attention being paid to issues of national culture and business orientations. Against this background, the purpose of the current study is to contribute to small business success in Tanzania by investigating the role that national culture characteristics (egalitarianism, mastery, harmony, embeddedness, affective autonomy and intellectual autonomy) and business orientations (technology, market and entrepreneurial) play in increasing the business success of Tanzanian small businesses. In this study, business success is defined as the extent to which small businesses effectively manage marketing, operations, human resources and finances. Located in the quantitative research paradigm, and using a mail survey data collection method, a sample of 640 SMMEs was targeted from eight regional economic zones in Tanzania. Of this group, 410 SMMEs, mostly small businesses, participated in the study (response rate = 64%). Instruments based on the works of Burgess (2011) and Schwartz (2004) were used to measure the business orientations and national culture characteristics of the selected Tanzanian small business owners and managers. The instrument developed by the South African Small Enterprise Development Agency (SEDA 2004) was used to measure small business success. The data analyses in this study included tests for data normalcy and the reliability and validity of the data (using Cronbach ‘s alpha and confirmatory factor analysis, respectively). Furthermore, multiple regression analyses were conducted to investigate the hypothesised relationships and other research propositions. The results of this study provided sound empirical proof of the interactive influence of national culture characteristics and business orientations on the business success of small businesses. Based on these empirical findings, a proposed framework for small business success in Tanzania was developed. The empirical results of the study provided evidence that support several existing theories, perceptions and beliefs about the relationship between national culture characteristics and business orientations. Certain findings also provided evidence that challenged aspects of existing theories, perceptions and beliefs. The study specifically highlights the importance of the technology and entrepreneurial business orientations as key positive determinants of the effectiveness of all four business functions (marketing, operations, human resources and financial management) investigated in the study. A surprising finding was the significantly negative relationship between market orientation and human resources management. The study also highlighted the centrality of the national characteristics of egalitarianism and intellectual autonomy in the development of the three business orientations (technology, entrepreneurial and market) investigated in the study. The findings suggest that the principle of the equality of people and a commitment to the continuous search and rewarding of new ideas and unique ways of doing things through continuous and professional development are important drivers of critical business orientations among Tanzanian small business owners and managers. The empirical findings furthermore highlighted the importance of mastery as a national culture characteristic. Mastery, which emphasises competence, goal-setting, hard-work, efficiency, effectiveness and self-reliance in attaining goals, influenced the effectiveness of all four business functions and two of the three business orientations investigated in the study. Finally, the study made significant contributions to the understanding of small business success in Tanzania and, arguably, across the African continent. - Description: Variations in the essential oil yield, chemical composition and kinetics of chemical transformation, antibacterial, and antioxidant properties of the Cymbopogon citratus cultivated in Alice, Eastern Cape of South Africa, over a period of 12 months using the solvent-free microwave extraction and the modified forms of conventional hydrodistillation methods have been evaluated. The modified forms involve variation in the pH medium to effect extraction and to obtain the whole leaf extracts of Cymbopogon citratus (lemongrass). The GC-MS analyses of the essential oils revealed the presence of 10, 11, 13, and 9 principal compounds in the microwave, extraction, hydrodistillation (water-distilled), hydrodistillation (aciddistilled and hydrodistillation (base-distilled), respectively with citral being the prominent compound in all the various extraction methods, with the water-distilled method having the highest cumulative citral content under the period specified. Each of the components of the oils varied in quantity and quality of yield at different points of the year. Different extraction methods produce different compounds. Increase in the amount of citral content was observed during the maturation stage of Cymbopogon citratus in all the extraction methods. Quantitative evaluation of antibacterial activity, minimum inhibitory concentration (MIC) values were determined on the oils and extracts of Cymbopogon citratus using agar dilution method. The essential oils obtained using all the methods of extraction showed activity against all the tested bacteria at a concentration of 10 mg mL-1. The minimum inhibitory concentrations for the acid-distilled varied between 1.25 and 5.0 mg mL-1, while those obtained by both water- and base-distilled showed equal activity on the microorganisms with the MIC ranging between 2.5 and 10 mg mL-1. The microwave extraction showed the least activity with the MIC of 10 mg mL-1. This however suggested that the oils obtained through acid-distillation showed the greatest activity than all other methods of extraction. Activity of the various extracts on bacteria showed that the ethanol extract had the highest activity on the tested organisms with MIC ranging between 0.625 and 1.25 mg mL-1. - Description: Variations in the essential oil yield, chemical composition and kinetics of chemical transformation, antibacterial, and antioxidant properties of the Cymbopogon citratus cultivated in Alice, Eastern Cape of South Africa, over a period of 12 months using the solvent-free microwave extraction and the modified forms of conventional hydrodistillation methods have been evaluated. The modified forms involve variation in the pH medium to effect extraction and to obtain the whole leaf extracts of Cymbopogon citratus (lemongrass). The GC-MS analyses of the essential oils revealed the presence of 10, 11, 13, and 9 principal compounds in the microwave, extraction, hydrodistillation (water-distilled), hydrodistillation (aciddistilled and hydrodistillation (base-distilled), respectively with citral being the prominent compound in all the various extraction methods, with the water-distilled method having the highest cumulative citral content under the period specified. Mathematics learner identities feature prominently in current research on mathematics education because they affect whether and how learners engage in mathematics. They play a critical role in enhancing (or detracting from) learners’ attitudes, dispositions, emotional development, and general sense of self as they learn mathematics. Development of positive learner mathematical identity is therefore useful in making learners commit to their mathematics work. South African primary mathematics education is described as being in a state of crisis, and various programmes are being implemented to develop intervention models to improve quality and ensure the effective teaching and learning of primary mathematics. The South African Numeracy Chair Project initiative at Rhodes University provides for longitudinal research and development programmes with primary mathematics teachers and learners from previously disadvantaged schools, in order to find ways of mitigating the crisis. The after school mathematics clubs provide extra-curricular activities focused on developing a supportive learning community where learners’ active mathematical participation, engagement, enjoyment, and sense making are the focus. The clubs provide a supportive learning environment that is different to the traditional classroom and in which learners can participate actively and freely in mathematical activities. The study explores the nature of mathematics learner identities as learning trajectories that connect the past and future in negotiation of the present. It also seeks to discover how primary school club participation and experiences feature in the learners’ mathematical identities. The study employs two theoretical frameworks to analyse qualitative data that was gathered in the form of spoken and written stories, by 14 learners who participated in the after school mathematics clubs in primary school. The stories covered learners’ engagement in mathematics in different landscapes of practice that promoted the construction of different learner mathematical identities. A close analysis of the qualitative data revealed that learners’ mathematical identities are heavily influenced by the values that were foregrounded in the after school mathematics clubs. The clubs valued hard work and encouraged learners to ask for assistance when in doubt. In line with the club ethos, the learners storied resilience and hard work in their narratives. It considers protest action on a South African university campus, which at times not only resulted in significant and swift concessions from the university leadership but also sparked national political action which got an equally swift response from the South African government. It considers the social movement, the RhodesMustFall movement (RMF), as one way in which students can organise themselves to get a better hearing from the University of Cape Town (UCT) management in their attempt to make a meaningful contribution to the university’s micro democracy. This study examines whether the interaction between the UCT management and RMF could be considered ‘political listening’, and the possible role of the Cape Times newspaper within this context of participation. Using data gathered through interviews, written communications, observation and newspaper articles, the study shows that in all of the interactions between RMF and the UCT management, both groups were seldom willing to forego their power to engage in genuine listening. Instead, the two parties guessed at what power the other party might have and acted to reduce that power. It is in this context of guessing at and figuring what power the other party has that listening occurs. Furthermore, the study shows that during the RMF protest, the UCT management viewed their responsibility for the institution mainly through the lens of Private Property Law which framed protest as something to be dealt with by restoring law and order. The study also details the role of the Cape Times newspaper in the interactions between RMF and the UCT management and considers if this role could be political listening. - Description: The DSM-5 prevalence rate of anorexia nervosa is 0.4%, bulimia nervosa is 1% to 1.5%, and binge eating disorder is 1.6% (American Psychiatric Association, 2013). Although treatment approaches for eating disorders have high drop-out rates and low rates of recovery, treatment modalities that address childhood factors contributing to the eating disorder, as well as the eating disorder behaviours, have better outcomes. Schema therapy is an integrative approach that has been used for the treatment of eating disorders for more than a decade. Central features in schema therapy include the identification of early maladaptive schemas arising from unmet needs and schema modes. Schema modes, composed of schemas and coping mechanisms, are active for an individual at a particular time in response to triggers in the environment (Brown et al., 2016). Identifying an individual’s modes is a crucial aspect that reflects the underlying structure of the individual’s creation of reality. A phenomenological understanding of the modes is essential for developing a case conceptualisation and treatment plan. Differences exist in the naming and description of modes in the current schema therapy literature, which suggests the need for a phenomenological investigation of these structures. This research study used a mostly qualitative approach, in the form of clinical interviews, substantiated by questionnaires, to examine schema modes. Case presentations using the schema therapy model are provided for five women with either anorexia nervosa, bulimia nervosa or binge eating disorder. Then, through a process of interpretative phenomenological analysis, specific modes are examined as to how they are experienced by the participants and influence their behaviour. The features of schema modes in these clinical cases are compared to the existing literature to extend the understanding of schema modes in eating disorders. The participants’ experiences revealed that they had schema modes in common, regardless of the eating disorder presentation, but that the features of the individual modes varied. Modes found in the current literature such as the Detached Self-Soother and Perfectionist Overcontroller coping mode, were found in all the participants. Four of the five participants had an Eating Disordered Overcontroller mode. Features consistent with the existing descriptions of the Perfectionist Overcontroller, Eating Disordered Overcontroller and Detached Self-Soother modes were noted, and new features were identified. The Perfectionist Overcontroller and Eating Disordered Overcontroller have been presented here as complex composite modes with sub-modes that work together in a coherent way in the service of the same project (Edwards, 2020b). Twenty-three features are identified in the parent modes. Blended parent modes, with multiple features active in a situation, were described. The blended parent modes expand on the existing literature on parent modes. - Description: This study offers a comprehensive examination of the Role of the Liberation Movements’ Archives in Shaping History Writing in a Post-Apartheid South Africa. It is based on the Liberation Archives housed at the National Heritage and Cultural Studies (NAHECS) at the University of Fort Hare, near the town of Alice in the Eastern Cape Province. A new narrative, grounded in the exploration and advantages of the use of these materials in their totality, given the new environment of research created under the new dispensation, has now become possible. The first chapter begins with an overview of the nature of archives in general and proceeds from there to consider the unique circumstances under which the different liberation movements of South Africa constructed their archives, and how these circumstances affected the nature of the materials preserved therein. One key point made here is that the liberation movement, as it spearheaded the struggle against apartheid, had already positioned itself as a “Government in Waiting,” convinced of the justness of its cause, and of the ultimate defeat of apartheid leading to he political seizure of power by the people. Hence, it created struggle materials seen as vehicles for the realisation and attainment of this objective. For this purpose, the appropriate housekeeping, viewing, consultation and access is absolutely relevant. The second chapter looks at the changing environment in which the repatriation of the struggle archives was implemented, and the decision to place them specifically at the University of Fort Hare and challenges. The third chapter narrates the difficulties encountered following the location of documents at Fort Hare, contestations, and contradictions between the stakeholders and the ultimate resolutions. The fourth and longest chapter goes to the heart of this thesis which dispel the myths created by the colonists and their historians intended to obscure the oppressed into embracing a historiography of South Africa that crossly flawed and distorted, silent and ignorant of reflecting early African societal developments which were evolving undisturbed at their own rate under socio-cultural and geographical conditions prior the advent of colonialism and the consequences. Therefore, based on these circumstances one can see that the installation of the new dispensation was not the end of the struggle, but the opening of a new chapter, the struggle against the conceptual legacy of apartheid and its vestiges. An evaluation of the consultation and exploitation of the archival collections comprises this subject in the last chapter of this study. Perusing them, one learns and realizes that the objective of the liberation movements was immense, to reconnect the marginalised of their once trampled and vilified history in order to inspire and empower them to reclaim their lost sense of belonging, identity, pride and to restore their humanity. This has been vindicated by the wide range of researchers and viewers from home and afar who have turned out to consult the archives in order to unpack the complexities of the South African history and that of the struggle. The information they contain serve as guides of addressing such complexities to arrest current transitional challenges, and offer solutions for some in articulating a need of building a new cohesive nation in a post-apartheid era. This is precisely what the liberation movement had in mind when it took a decision to gather together the struggle materials and have them repatriated for preservation and access purposes. The first chapter begins with an overview of the nature of archives in general and proceeds from there to consider the unique circumstances under which the different liberation movements of South Africa constructed their archives, and how these circumstances affected the nature of the materials preserved therein. One key point made here is that the liberation movement, as it spearheaded the struggle against apartheid, had already positioned itself as a “Government in Waiting,” convinced of the justness of its cause, and of the ultimate defeat of apartheid leading to he political seizure of power by the people. Hence, it created struggle materials seen as vehicles for the realisation and attainment of this objective. For this purpose, the appropriate housekeeping, viewing, consultation and access is absolutely relevant. The second chapter looks at the changing environment in which the repatriation of the struggle archives was implemented, and the decision to place them specifically at the University of Fort Hare and challenges. The third chapter narrates the difficulties encountered following the location of documents at Fort Hare, contestations, and contradictions between the stakeholders and the ultimate resolutions. The fourth and longest chapter goes to the heart of this thesis which dispel the myths created by the colonists and their historians intended to obscure the oppressed into embracing a historiography of South Africa that crossly flawed and distorted, silent and ignorant of reflecting early African societal developments which were evolving undisturbed at their own rate under socio-cultural and geographical conditions prior the advent of colonialism and the consequences. Therefore, based on these circumstances one can see that the installation of the new dispensation was not the end of the struggle, but the opening of a new chapter, the struggle against the conceptual legacy of apartheid and its vestiges. An evaluation of the consultation and exploitation of the archival collections comprises this subject in the last chapter of this study. Perusing them, one learns and realizes that the objective of the liberation movements was immense, to reconnect the marginalised of their once trampled and vilified history in order to inspire and empower them to reclaim their lost sense of belonging, identity, pride and to restore their humanity. This has been vindicated by the wide range of researchers and viewers from home and afar who have turned out to consult the archives in order to unpack the complexities of the South African history and that of the struggle. The information they contain serve as guides of addressing such complexities to arrest current transitional challenges, and offer solutions for some in articulating a need of building a new cohesive nation in a post-apartheid era. This is precisely what the liberation movement had in mind when it took a decision to gather together the struggle materials and have them repatriated for preservation and access purposes. Insights into the drivers and impact of climate change and climate change adaptation in the Eastern Cape, South Africa: the case of Amathole District Municipality

Communal farmers rely on rain-fed agriculture and are usually too poorly resourced to cope with the frequency of climate-related events that may be expected in the future. Response and adaptation is vital to ensure the sustainability of livestock production, particularly since it is the main source of survival in communal areas. The Eastern Cape Provincial Policy on Climate Change was introduced in 2010 to facilitate a coordinated approach that assists farmers to respond, adapt and mitigate climate change. The study examines the implementation of the policy to identify farmers’ perceptions of the response rate prior to, during and after climate change disasters. In addition, the study establishes the characteristics of livestock production in the study area, seeking to ascertain how communal livestock farmers CLFs are adapting their practices to ensure sustainable livestock production in the face of climate change. In order to make informed decisions on coping strategies, farmers require access to information on climate change. The study therefore examines the communication channels used by farmers to access such information. Since food security is under threat, the study also assesses the effects of climate change on food security among the CLFs. Multi stage sampling was used to select 388 communal livestock farmers in three local municipalities in Amathole District Municipality. A cross-sectional survey was carried out in five communities randomly selected in the three local municipalities, with data collected by means of a semi-structured questionnaire. Descriptive statistics, inferential statistics, principal component analysis and regression methods were used to analyse the data. The study reveals that communal livestock farmers perceive a poor response rate from extension services before, during and after disasters, and that they have poor access to support materials. All of the respondents practised uncontrolled breeding, attributed to a lack of infrastructure such as fencing. Most farmers kept cattle for income generation. The major constraints of cattle production were diseases and pests. CLFs employed dipping, rotational grazing, water tanks, veld burning and the sale of animals as the main strategies to cope with climate change. CLFs access climate change information through multiple channels. The main sources were other farmers and media such as radio and television, although language barriers hampered full understanding of information conveyed about climate chnage. Farmer-to-farmer contact was a central aspect of the CLFs’ lives that could be better employed in the dissemination of climate change information. The results suggest a positive relationship between diversity of species kept and food security. Assets, the social safety net (mainly grants) and adaptive capacity indicators positively and significantly impacted households’ resilience to food insecurity. The study recommends that the capacity of communal livestock farmers on effective mitigating strategies be improved, making use of mass media; that more work be done by extension services to prepare farmers for adverse events and that relief materials disseminated during periods of disaster be tagged “national emergency” to speed up distribution and use. There is an urgent need for adequate and timely provision of climate change information that will help CLFs to make more effective use of their resources in the face of climate change. - Description: Malaria is a millennia-old disease with the first recorded cases dating back to 2700 BC found in Chinese medical records, and later in other civilizations. It has claimed human lives to such an extent that there are a notable associated socio-economic consequences. Currently, according to the World Health Organization (WHO), Africa holds the highest disease burden with 94% of deaths and 82% of cases with P. falciparum having ~100% prevalence. Chemotherapy, such as artemisinin combination therapy, has been and continues to be the work horse in the fight against the disease, together with seasonal malaria chemoprevention and the use of insecticides. Natural products such as quinine and artemisinin are particularly important in terms of their antimalarial activity. The emphasis in current chemotherapy research is the need for time and cost-effective workflows focussed on new mechanisms of action (MoAs) covering the target candidate profiles (TCPs). Despite a decline in cases over the past decades with, countries increasingly becoming certified malaria free, a stalling trend has been observed in the past five years resulting in missing the 2020 Global Technical Strategy (GTS) milestones. With no effective vaccine, a reduction in funding, slower drug approval than resistance emergence from resistant and invasive vectors, and threats in diagnosis with the pfhrp2/3 gene deletion, malaria remains a major health concern. Motivated by these reasons, the primary aim of this work was a contribution to the antimalarial pipeline through in silico approaches focusing on P. falciparum. We first intended an exploration of malarial targets through a proteome scale screening on 36 targets using multiple metrics to account for the multi-objective nature of drug discovery. The continuous growth of structural data offers the ideal scenario for mining new MoAs covering antimalarials TCPs. This was combined with a repurposing strategy using a set of orally available FDA approved drugs. Further, use was made of time- and cost-effective strategies combining QVina-W efficiency metrics that integrate molecular properties, GRIM rescoring for molecular interactions and a hydrogen mass repartitioning (HMR) molecular dynamics (MD) scheme for accelerated development of antimalarials in the context of resistance. This pipeline further integrates a complex ranking for better drug-target selectivity, and normalization strategies to overcome docking scoring function bias. The different metrics, ranking, normalization strategies and their combinations were first assessed using their mean ranking error (MRE). A version combining all metrics was used to select 36 unique protein-ligand complexes, assessed in MD, with the final retention of 25. From the 16 in vitro tested hits of the 25, fingolimod, abiraterone, prazosin, and terazosin showed antiplasmodial activity with IC50 2.21, 3.37, 16.67 and 34.72 μM respectively and of these, only fingolimod was found to be not safe with respect to human cell viability. These compounds were predicted active on different molecular targets, abiraterone was predicted to interact with a putative liver-stage essential target, hence promising as a transmission-blocking agent. The pipeline had a promising 25% hit rate considering the proteome-scale and use of cost-effective approaches. Secondly, we focused on Plasmodium falciparum 1-deoxy-D-xylulose-5-phosphate reductoisomerase (PfDXR) using a more extensive screening pipeline to overcome some of the current in silico screening limitations. Starting from the ZINC lead-like library of ~3M, hierarchical ligand-based virtual screening (LBVS) and structure-based virtual screening (SBVS) approaches with molecular docking and re-scoring using eleven scoring functions (SFs) were used. Later ranking with an exponential consensus strategy was included. Selected hits were further assessed through Molecular Mechanics Poisson-Boltzmann Surface Area (MM-PBSA), advanced MD sampling in a ligand pulling simulations and (Weighted Histogram Analysis Method) WHAM analysis for umbrella sampling (US) to derive binding free energies. Four leads had better predicted affinities in US than LC5, a 280 nM potent PfDXR inhibitor with ZINC000050633276 showing a promising binding of -20.43 kcal/mol. As shown with fosmidomycin, DXR inhibition offers fast acting compounds fulfilling antimalarials TCP1. Yet, fosmidomycin has a high polarity causing its short half-life and hampering its clinical use. These leads scaffolds are different from fosmidomycin and hence may offer better pharmacokinetic and pharmacodynamic properties and may also be promising for lead optimization. A combined analysis of residues’ contributions to the free energy of binding in MM-PBSA and to steered molecular dynamics (SMD) Fmax indicated GLU233, CYS268, SER270, TRP296, and HIS341 as exploitable for compound optimization. Finally, we updated the SANCDB library with new NPs and their commercially available analogs as a solution to NP availability. The library is extended to 1005 compounds from its initial 600 compounds and the database is integrated to Mcule and Molport APIs for analogs automatic update. A critical assessment of economic policies and their impact on entrepreneurship development in South Africa: a case of Khayelitsha Township in Cape Town

- Description: The complexity of the South African society due to past systematic exclusion of some communities from economic activities resulted in the triple challenge of poverty, inequality and unemployment. Although various economic policies had been developed since the advent of the democratic dispensation in 1994, these policies focused on levelling the economic environment, poverty alleviation and massive social security system. However, despite all the interventions, South Africa remains one of the most unequal societies in the world. This is profiled by the mushrooming of informal settlements around cities with job seekers and frequent protests by people who are impatient to experience improvement in their quality of life and who feel cheated out of their social contract. Advocacy from the governing party’s tripartite alliance towards a developmental state had placed more obligations on the government. This requires deliberate interventions through policies implementation to improve or at best alleviate the well-being of the citizenry. A survey conducted in the township of Khayelitsha to assess the impact of such economic policies on the development of entrepreneurship. Khayelitsha is one of South Africa’s largest townships, an informal settlement profiled by unemployment, poor infrastructure and unavailability of basic services. The study explored the entrepreneurial perception, start-up barriers, entrepreneurship climate, and general cultural factors relating to the environment of entrepreneurship development. The results reveal that despite the various economic policies, high level of unwillingness of citizens towards entrepreneurial activity and business venture is rife notwithstanding the high level of unemployment and poverty. The antidote to these challenges would be a rapid entrepreneurship drive among people most affected that would cause them to mind their own business” and become “masters of their own destinies. Given the complexity and massiveness of the challenges, the conclusion shows that a pragmatic solution is required. Therefore, the emancipation of entrepreneurship development in South Africa townships needs attention. The recommended solution is a pragmatic and innovative strategy based on the acronyms NEEDS HELP which stand for Neighbourhood Enhancement and Environmental Development Strategy and Holistic Entrepreneurial Lifestyle Programme. , Thesis (PhD) -- Faculty of Management and Commerce, 2015

A survey conducted in the township of Khayelitsha to assess the impact of such economic policies on the development of entrepreneurship. Khayelitsha is one of South Africa’s largest townships, an informal settlement profiled by unemployment, poor infrastructure and unavailability of basic services. The study explored the entrepreneurial perception, start-up barriers, entrepreneurship climate, and general cultural factors relating to the environment of entrepreneurship development. The results reveal that despite the various economic policies, high level of unwillingness of citizens towards entrepreneurial activity and business venture is rife notwithstanding the high level of unemployment and poverty. The antidote to these challenges would be a rapid entrepreneurship drive among people most affected that would cause them to mind their own business” and become “masters of their own destinies. Given the complexity and massiveness of the challenges, the conclusion shows that a pragmatic solution is required. Therefore, the emancipation of entrepreneurship development in South Africa townships needs attention. The Pcs had either H2, zinc or silicon as central metals and have pyridyloxy, benzothiozole phenoxy, and respective cationic analogues as ring substituents. The Pcs were linked to carbon based nanoparticles such as graphene quantum dots, carbon dots, and detonation nanodiamonds (DNDs) via an ester, amide bond and/or π - π stacking. The physicochemical characteristics of the Pcs were assessed when alone and when in a conjugated system. Both symmetrically and asymmetrically substituted benzothiozole Pcs when quaternised displayed higher triplet and singlet oxygen quantum yields than their unquaternised counterparts. Linkage to carbon nanoparticles (especially to detonation nanodiamonds) had an increasing effect on triplet and singlet oxygen quantum yield. However, a general decrease in singlet oxygen quantum yield on linkage to doped detonation nanodiamonds was associated with the screening effect of DNDs. Heteroatom doped DNDs-Pc nanohybrids have less singlet oxygen than Pcs alone due to molecular structural stability associated with strain that is relatively reduced upon linking Pcs. The In vitro dark cytotoxicity and photodynamic therapy of selected Pc complexes and conjugates against MCF-7 cells was tested. All studied Pc complexes and conjugates showed minimum dark toxicity making them applicable for PDT. When Pc complexes are alone, there is less phototoxicity with >22% cell viability at concentrations ≤ 50 μg/mL relative to conjugates with <22% cell viability at concentrations ≤ 50 μg/mL. There was no direct relationship between PDT and singlet oxygen quantum yields. Nonlinear optical characteristics of complexes was improved upon conjugation of DNDs. Absorbance, input energy, percentage loading, central metal, substituent of Pc and nature of interaction (covalent, noncovalent) are amongst some of the factors that influence nonlinear absorption properties of materials used in this study. - Description: This study aimed to evaluate the effect of single and extra vitamin C (VC) supplementation on water-stressed Xhosa goats during the summer by evaluating their growth performance, blood biochemical profile, nutrient balance, carcass characteristics and meat qualities. The objectives were to assess the effect of water restriction and vitamin C supplementation by evaluating: firstly, growth performance, heat tolerance responses, and blood metabolites; secondly, intake, nutrient digestibility, nitrogen and mineral balance; thirdly, stress indices, carcass characteristics and meat qualities parameters and lastly, fatty acid profile, oxidative stability of lipids and sensory attributes were evaluated. To achieve these objectives, forty-two (42) goats were divided equally into seven treatments (75-day trial); without water restriction (W0, control); water restriction of 70percent of ad libitum water intake WI (W70); water restriction of 50percent ad libitum WI (W50); water restriction of 70percent of ad libitum WI plus 3g VC daily (W70+); water restriction 50percent of ad libitum WI plus 3g VC daily (W50+); water restriction of 70percent of ad libitumWI plus 3g VC and extra 5gVC given every eight-day (70++); water restriction of 50percent of ad libitum WI plus 3g VC and extra 5gVC given every eight-day (W50++). In the first objective, growth performance parameters, body condition scores, rectal temperature, respiratory rate, heat tolerance response parameters and blood metabolites were measured.Weight loss due to water restriction was reduced by VC supplementation in treated groups. The attenuation effect of VC was significant (P<0.05) in responses to respiratory rate (RR), Na+, K+, Mg2+, Cl-, Ca2+ and urea. Supplementation of VC (either single or multiple) did not (P>0.05) improve the effect of water restriction on body condition scores (BCs), FAMACHA©, glucose, globulin, alanine aminotransferase (ALT) and high-density lipoprotein (HDL). In the second objective, intake, nutrient digestibility, nitrogen and mineral balance were evaluated. The reduced nutrient intake was similar (P>0.05) irrespective of water restrictions and VC supplementations. Water-restriction levels did not affect nutrient intake/metabolic weight. Retained nitrogen increased with water-restriction levels but not influenced by VC. NH3-N decreased with the levels of water restriction while the pH was similar across the water restricted groups. Supplementation of VC had no effect (P>0.05) on apparent digestibility and digestible nutrients induced by water restriction levels. The amount of Ca, K, Mg retained increased (P<0.05) with water-restriction levels. In the third objective, stress indices, carcass characteristics and meat quality parameters were evaluated. The level of plasma cortisol concentration due to water stress was lessened (P<0.05) by VC. Follicle stimulating hormone (FSH), Luteinizing hormone (LH) and insulin were not affected (P>0.05) by levels of water restriction. The hot and carcass weights were lower (P<0.05) in W70+ and W50+ compared to W70 and W50 respectively. Vitamin C treated goats had higher redness (a*) compared to W70, W50 and W0. The shear force and cooking loss increased (P<0.05) with water restriction levels, while thaw loss, pH45min and pH24h were not affected by water restriction levels. In the final objective, the treatments did not affect (P>0.05) the oxidative stability of lipids (TBARS). The moisture and fat content in the water restricted groups were lower (P<0.05) than W0. The consumer meat sensory appearance decreased (P<0.05) with levels of water restrictions. Regardless of VC concentration and supplementation, vaccenic, and docosahexaenoic acid increased (P<0.05) while linolenic acid decreased as the water restriction levels increased. Nonetheless, treatment did not affect (P>0.05) the saturated and unsaturated fatty acids. These studies showed that suboptimal water intake negatively affects the growth performance and nutrient intake of Xhosa goats. - Description: The production and characterization of bioflocculants produced by three bacteria belonging to Arthrobacter, Halomonas and Micrococcus genera and isolated from freshwater and marine environments were evaluated both as axenic cultures and as consortia. The influences of cultutre conditions such as carbon, nitrogen and metal ions sources, as well as initial pH on bioflocculant production by individual isolates were investigated. Both Arthrobacter sp. Raats and Halomonas sp. Okoh utilized urea as a nitrogen source of choice for optimal production of the bioflocculants with Micrococcus sp. Leo having a preference for peptone. All three strains differed in as far as the carbon source of choice was concerned with lactose, glucose and sucrose the preferred carbon sources respectively. Also, all three bacterial strains produced an extracellular bioflocculant aerobically but an intial pH 7.0 of the culture media was suitable for both Arthrobacter sp. Raats and Halomonas sp. Okoh with a slightly alkaline pH of 9.0 preferred by Micrococcus sp. Leo. The presence of Mg2+ cations stimulated bioflocculant production by both Arthrobacter sp. Raats and Micrococcus sp. Leo while Ca2+ resulted in more efficient bioflocculant production by Halomonas sp. Okoh. Chemical analyses revealed the bioflocculants produced by both Halomonas sp. Okoh and Micrococcus sp. Leo to be predominantly polysaccharides whereas Arthrobacter sp. Raats produced principally a glycoprotein composed of about 56percent protein and 25percent total carbohydrate. Response surface methodology (RSM) was used to optimize production medium for bioflocculant production by a consortium of Halomonas sp. Okoh and Micrococcus sp. Leo. Plackett-Burman experimental design showed that fructose, ammonium sulphate and MgCl2 were significant in the high yield of the bioflocculant. Furthermore, central composite design showed that optimal concentration of these critical nutritional sources were 16.14 g/L, 1.55 g/L and 1.88 g/L for fructose, ammonium sulphate and MgCl2 respectively. Quantification of the bioflocculant showed a yield of 6.43 g/L which was in close accord with the predicted value of 6.51 g/L. FTIR spectrometry of the bioflocculant indicated the presence of carboxyl, hydroxyl and amino groups, typical for heteropolysaccharide, while SEM imaging revealed a lattice-like structure. - Description: The pebble-bed reactor (PBR) is a graphite-moderated, gas-cooled high temperature nuclear reactor design and it is one of six nuclear reactor concepts included in the Generation IV initiative. Pebble-bed reactors use spherical fuel elements (called pebbles) which consist of pyrolytic graphite (which acts as the moderator) and contain thousands of micro-fuel particles called tristructural isotropic (TRISO) particles. These TRISO fuel particles consist of a fissile material (such as U235 in oxide or carbide form) surrounded by a carbon buffer layer, a pyrolytic carbon (PyC) layer, a silicon carbide (SiC) ceramic layer for structural integrity and metallic fission product containment, and an outer PyC layer. The PBR is claimed to be a passively safe design. The commercial development of the first pebble bed reactor was that of the German AVR reactor (German: Arbeitsgemeinschaft Versuchsreaktor) developed during the 1960s. The AVR design was later updated and marketed by a company called HTR. In 1999 the South African electricity company ESKOM obtained the right to access the HTR engineering database that included details of the Siemens/Interatom HTR-Module design. ESKOM worked with HTR on a new design and dubbed it the pebble bed modular reactor (PBMR). The PBMR Co. Ltd. was formed in 1999 and mandated to license and build PBMR reactors. One of the safety considerations which emerged from research is that of metal fission product release from the TRISO fuel particles. Ag110m is a radioactive metallic fission product found to have been released from intact TRISO particles. The release of this Ag isotope is of particular concern because it is highly gamma active and has a half-life of approximately 250 days, resulting in unsafe environments for maintenance workers of PBRs. During the past four decades, many different mechanisms for Ag transport in SiC and release from TRISO particles have been proposed. A promising more recent mechanism suggests that the metallic fission product palladium (Pd) plays a significant role in the transport and release of Ag from intact TRISO particles. In this mechanism Ag transport in irradiated TRISO particle fuel takes place in the presence of the fission product Pd. Functional biogeography: evaluating community assemblage patterns and ecosystem functioning in intertidal systems using trait-based approaches

- Description: Analyses of taxonomic diversity patterns within coastal systems has been critical in the development of the theory of biogeography. Increasing evidence, however, shows that the variety of functions that species perform in ecosystems (rather than their taxonomic identity) is a better predictor of the influence of the environment on the species. This information has been useful in predictive ecology leading to the development of trait-based approaches (TBA). Until the late 1970s, however, limited effort (particularly in marine systems) was channeled towards patterns in functional species traits and how they may be affected by changes in environmental gradients. Here, I mapped the functional biogeography of the South African coastline based on a suite of species' reproduction and development traits. Because species composition is one of the key tools used by zoogeographers to map species distribution patterns, I expected lower variability in trait composition within main biogeographic regions than in intervening transition zones based on the habitat templet theory and following the biomass ratio and limiting similarity hypotheses. In brief, the habitat templet theory proposes that “the habitat provides a templet upon which evolution forges species characteristics”, while the biomass ratio hypothesis assumes that the most abundant species traits determine ecosystem functioning. The limiting similarity hypothesis also sometimes referred to as the niche complementarity hypothesis, however, predicts that species can coexist if their niches complement one another. In light of the habitat being an evolutionary templet, abiotic and biotic habitat patterns were measured as nearshore SST and chlorophyll-a gradients, respectively. I expected the SST gradient to act as the stronger key filter of trait diversification because temperature is often considered the most influential environmental factor affecting species survival with seasonality of SST affecting the timing of spawning and along with food availability, possibly influencing fecundity. Functional trait data were thus compiled for macroinvertebrate species collected from fifty-two rocky shore sites from three main bioregions (east, south, and west) and two transition zones (south-west and south-east). Biological trait analysis and functional diversity indices were used to evaluate how traits related to species development and reproduction respond to temperature and chlorophyll -a (used as a proxy for food availability) gradients along the coastline. GLMM and hierarchical cluster analyses showed distinct patterns/shifts in SST and chlorophyll-a gradients across bioregions, with two main breaks in SST separating the east and south-east overlap (SEO) bioregions from the south, south-west overlap (SWO) and west bioregions. In contrast, chlorophyll-a exhibited three major breaks with the east, SEO–south–SWO, and west clustering independently of each other. The RLQ analysis (a type of co-inertia analysis) which simultaneously ordinates 3-matrix datasets [i.e., (environment × site[R]), (species × site[L]) and (species × traits [Q])] showed that the higher SST gradient on the east and SEO promoted higher abundance and biomass of simultaneous hermaphrodites while higher chlorophyll-a gradients on the SWO and west coasts strongly promoted reproductive maturity at larger-sizes. The combined fourth-corner analyses showed that the modalities within the development trait domain responding to chlorophyll-a gradients primarily included filter feeders, sessile and swimming species and also species living on the infratidal zone. In addition, the reproduction trait domain showed higher sensitivity and association to differences in chlorophyll-a and SST gradients than development traits. Overall, SST and chlorophyll-a gradients influenced the distribution of the most dominant traits as indicated by shifts in community-weighted mean trait values across bioregions. This suggests the importance of habitat filtering on coastal species reproduction. A separate study evaluating the influence of large-scale biogeographic effects vs the micro-scale biogenic habitat structure offered by coralline seaweeds across 24 sites revealed some notable effects of both factors on the diversity and abundance of macroalgal epifauna. There was a notable biogeographic influence on epifauna, with the SEO recording the highest epifaunal species richness and abundance, followed by the south coast, then the SWO and lastly the west coast. In addition, the total biomass gradient of the corallines followed a similar trend. The epifauna however, showed no host-specificity, illustrating that epifauna may not be species–centric as commonly assumed, and the higher diversity of epifaunal diversity may well be simply because those corallines are the available habitat within the sampled part of the coastline. Lastly, macroinvertebrate trait distribution on the South African coastline confirms that the habitat, particularly the biotic filter (in this case chl-a) provides a templet upon which evolution forges species traits. However, since temperature is a proxy for nutrient availability (cold upwelling brings nutrients), then temperature drives chlorophyll-a. Subsequently this means the abiotic component indirectly drives trait distribution by influencing the biotic environment (chl-a). For epifauna species, also, the coralline diversity and composition can also be regarded as a biotic filter influencing the epifaunal abundances and composition across different bioregions. Moreover, since temperature is regarded as a conservative trait in seaweeds, temperature tolerance defines the biogeographical boundaries of seaweeds, therefore temperature may be indirectly affecting epifauna abundances through coralline species diversity and biomass. In summary, considering the deterministic processes governing ecosystem functioning and community assemblage, the mass ratio and limiting similarity hypotheses showed complementary effects. Different bioregions provided variable support for these two hypotheses, but overall, the mass ratio hypothesis (weighted by species biomass) received stronger support and may be more meaningful to the interpretation of ecosystem functioning and persistence within rocky shore systems. Lastly, although, the SWO showed some of the characteristics of a subtraction zone based on the relatively low abundance, diversity, and biomass measures. Nonetheless, there was evidence of high functional redundancy across all other four bioregions. Dynamics of charge movement in ∞-Al2O3:C,Mg using thermoluminescence phototransferred and optically stimulated luminescence

- Description: The dosimetric features of ∞-Al2O3:C,Mg have been investigated for unannealed and annealed samples. The unannealed sample is referred to as sample A whereas the samples annealed at 700, 900 and 1200°C for 15 minutes each are referred to as samples B, C and D respectively. A glow curve of unannealed ∞-Al2O3:C,Mg measured at 1°C/s after irradiation to 2.0 Gy consists of peaks at 43, 73, 164, 195, 246, 284, 336 and 374°C respectively. For sample B (annealed at 700°C), a glow curve measured at 1°C/s after irradiation to 3.0 Gy has peaks at 46, 76, 100, 170, 199, 290, 330 and 375°C whereas the glow curve of sample C (annealed at 900°C) recorded under the same conditions consists of peaks at 49, 80, 100, 174, 206, 235, 290, 335 and 375°C respectively. Sample D (annealed at 1200°C) is the most sensitive of the four samples. A glow curve of sample D measured at 1°C/s after irradiation to 0.2 Gy has peaks at 52, 82, 102, 174, 234, 288 and 384°C respectively. The peaks are labelled I-VIII in order of appearance. The 100°C peak, labelled IIa, is induced by annealing at or above 700°C. The dose response of these peaks was studied for doses within 0.1-8.2 Gy. The reported peaks follow first-order kinetics irrespective of annealing temperature. Peaks I-III of each sample are reproduced under phototransfer for preheating up to 400°C. For the unannealed sample, the reproduced peaks are labelled A1-A3 whereas for the annealed samples, they are labelled B1-B3, C1-C3 and D1-D3 respectively. The annealing-induced peak at 100°C is reproduced as B2a, C2a and D2a for samples B, C and D respectively. A PTTL peak labelled C2b or D2b is also observed near 140°C in samples C and D. In addition to these PTTL peaks, a PTTL peak corresponding to peak IV is also found for sample D and for the unannealed sample. As the corresponding conventional peaks, the PTTL peaks of each sample follow first-order kinetics. Peak I and its corresponding PTTL peak for each sample are unstable and fade to a minimal level after 300 s of storage time. On the other hand, peak II of each sample and its corresponding PTTL peak could still be observed with delay up to 5000 s. Peak III of the unannealed sample remains stable with storage time up to 48 hours. Irrespective of annealing, the trap corresponding to peak III is the most sensitive to optical stimulation. Time-dependent profiles of PTTL from unannealed and annealed ∞-Al2O3:C,Mg were also studied. The mathematical analysis of the PTTL time-response profiles is based on experimental results. The role of various electron traps in PTTL was determined by using pulse annealing and by monitoring the dependence of peak intensity on duration of illumination for peaks not removed by preheating. The presence and role of deep traps were further demonstrated with thermally assisted optically stimulated luminescence. For the unannealed sample, the activation energy for thermal assistance is 0.033 ± 0.001 eV and the activation energy for thermal i quenching is 1.043 ± 0.001 eV. For sample C, the activation energy for thermal assistance is 0.044 ± 0.003 eV whereas that for thermal quenching is 1.110 ± 0.006 eV. The values for the activation energy for thermal assistance are lower than those reported in literature.
