The development of an in vitro system for the production of drug metabolites using microsomal enzymes from bovine liver

- Description: Drug metabolism is a specialised subset of xenobiotic metabolism, pertaining to the breakdown and elimination of pharmaceutical drugs. The enzymes involved in these pathways are the cytochrome P450 family of isozymes. Metabolism is an important factor in determining the pharmacological effects of drugs. The main aim of this study was to develop a system whereby the major metabolites of drugs can be produced in vitro. An in vitro system was developed and optimised using commercially prepared microsomes from rat liver and coumarin (by monitoring its conversion to 7-hydroxycoumarin) as a model. The optimum running conditions for the incubations were 50 μM coumarin, 50 μg protein/ml microsomes, 1 mM NADP⁺, 5 mM G6P and 1U/ml G6PDH incubated for 30 minutes at 38℃. The HPLC method for the detection of coumarin and 7-hydroxycoumarin was also validated with respect to linearity, reproducibility, precision, accuracy and lower limits of detection and quantification. The system developed was then tested using microsomes prepared from fresh bovine liver on these ten drugs of interest in doping control in horse racing: diazepam, nordiazepam, oxazepam, promazine, acepromazine, chlorpromazine, morphine, codeine, etoricoxib and lumiracoxib. The bovine liver microsomes were prepared using differential centrifugation and had activity on a par with the commercial preparations. This in vitro system metabolised the drugs and produced both phase I and II metabolites, similar to those observed in humans and horses in vivo. For example, the major metabolites of the benzodiazepine drug, diazepam, nordiazepam, temazepam and oxazepam as well as the glucuronidated phase II products were all found after incubations with the bovine liver microsomes. The metabolism of the drugs was also investigated in silico using the computational procedure, MetaSite. MetaSite was able to successfully predict known metabolites for most of the drugs studied. Differences were observed from the in vitro incubations and this is most likely due to MetaSite using only human cytochrome P450s for analysis.

It was found that faced with this dual mandate, learners chose to focus on the assessment-laden mandate as this was the ultimate agenda that would reap the reward within the traditional school environment. The study examines the gap between knowledge and practice to probe ways in which to close this gap in the context of an environmental research assignment. The outcome is a recommendation that we as the project designers review the evidence of tensions and contradictions revealed in the study to reflect on the underlying purpose of the project and reshape it in light of recent literature on the challenges of social learning and change.

- Description: The purpose of the study was to investigate the ways in which the practice of t the Flexible Firm Model (FFM) disempowered the non-citizen professional nurses at Gaborone Private Hospital (GPH) in Gaborone, Botswana. GPH is a member of the Life Health care group of private hospitals based in South Africa. This study sought to probe the manner in which the practice of FFM influenced the employment relationship for non-citizen professional nurses. It also sought to examine the impact of the nature of the employment relationship on career development and representation and participation. Both qualitative and quantitative research methodologies were applied. Questionnaires were used to collect quantitative data. Thirty-five questionnaires were issued out to professional nursing staff and thirty-three responses were received. The response rate was ninety-four percent. The qualitative research data consisted of twelve in-depth interviews with professional nurses, the human resources manager , a professional nurse agency owner and a private clinic co-owner. Document analysis in the form of labour legislation, employee guides and employment contracts was used to validate data collected from the in-depth interviews. The results from the questionnaire revealed that an overwhelming majority of the professional nurses were Non-Citizens' dependent contractors. Although just under half of the participants held two or more previous contracts with GPH, an overwhelming majority had never been promoted. In addition, none of the participants were affiliated to the Botswana Nurses Association (BNA), the local equivalency of a professional nursing trade union. The in-depth interviews disclosed a sense of helplessness at their perceived disempowerment within the employment relationship, a poor career development and weak representation and participation. This study concluded that disempowerment at GPH was manifested through worker representation and participation and in effective skill upgrade. These forms of disempowerment were made worse by the global professional nurse shortage and maladministration of avail able human resources. Effective use of human resources and continued education could be employed to circumvent the adverse results of disempowerment.

- Description: The project was initiated by Eastern Cape Parks (ECP) as a request for the construction of inventories of priority species and their population levels inside three nature reserves on the Eastern Cape Wild Coast, South Africa, and to develop a strategic management plan to manage these natural resources in each reserve. Thirty key species were identified by local communities in and around Dwesa-Cwebe, Silaka and Mkambati Nature Reserves through community workshops. For forested areas belt transects of 100 m x 6 m where used. The basal circumference of key tree species within the belt transect was measured as well as the height of saplings (height < 150 m). Tree species were categorized based on densities, size class distribution (SCD) curves and values, and spatial grain. For grassland areas straight transects of 200 m long were used, along which ten 3 m x 3 m quadrates were placed at 20 m intervals. Within each grassland transect the height of herbs or tuft diameter of grasses was recorded and percentage cover estimated. Grassland species were categorized based on density, SCD curves and percentage cover. All species were placed into harvesting categories based on analysed ecological data that was collected in the field. Category 1 species were very rare or not found in the reserve and it was recommended that species be conserved and monitored. Category 2 species had low densities in the reserve indicating declining populations and was suggested that these be monitored and not harvested. Category 3 species had high densities and have potential for harvesting with strict limitations. Category 4 species were most abundant with very high densities and can be harvested within management guidelines. These categories were grouped further using social and ecological data such as harvesting risk, frequency of collection, use value and number of uses. This highlighted which species have conservation priority within each category and a decision can be made as to how intense or limited extraction should be. By incorporating GIS the distribution of each species was looked at and harvesting and non-harvesting zones established to determine where species can be extracted. Monitoring plans must consider the quantity of plant material collected, fire regimes, optimal harvesting rates and harvesting zones, and be able to pick up changes in populations. Also, it is important that the community be involved in conserving and monitoring these species. Adaptive monitoring and management must be used to steer harvesting practices in the Wild Coast reserves. This allows for the development of harvesting practices through ‘learning by doing’, and the evolution of good questions to guide monitoring decisions

Exploring perceptions and implementation experiences of learner-centered education among history teachers : a case study in Namibia

- Description: The study sought to understand how Grade 9 History teachers perceive and implement learner-centered education (LCE) in selected schools in Caprivi educational region in the Republic of Namibia. It concentrated on three teachers in two combined and junior secondary schools. The research employed a qualitative approach and three data instruments were used: interviews, class observations and document analysis. The findings of the study show that in their interview discussions of the principles, intent and recommended key features of LCE, the three participating teachers generally correctly captured some of the essential intentions of a LCE approach. At times in the interviews they seemed to strongly grasp the essence of a key strategy and its intent, but at other times their views were sketchy. Their view of different teaching strategies at times appeared integrated but not always that strongly. When it came to their classroom practice they could and did use a number of appropriate LCE teaching approaches. The level of effectiveness in their use of many of the approaches varied from effective to far from ideal and in need of quite big improvement. In the area of resources the three classrooms were extremely limited in what they displayed, had and used. There were too few textbooks and almost no posters and wall displays on history and the geography of the world and its peoples that the students were studying. An especially interesting feature was that they all seemed to be consciously engaged in an on-going teaching experiment with the LCE approaches. The LSC [sic] practices were clearly not yet strongly imbedded as solid classroom habits or dispositions, with perhaps the exception of questioning. But this experimenting made them much more self-conscious and reflective about their experiences. They all frankly identified some tensions that they felt existed between the espoused official features of a LCE class and the demands of the covering the curriculum, size of classes etc. Overall it was an encouraging picture of teachers eager to find ways to improve their teaching and experiment with new ideas. But also a picture of people not properly exposed to good or best practice in each teaching strategy and having to reinvent and rediscover on their own even the basics of reasonable practice often making very basic mistakes, for example in questioning.

- Description: This research is premised on the understanding that media texts are discourses and that all discourses are functional, that is, they refer to things, issues and events, in meaningful and goal oriented ways. Nine articles are analysed to explicate the sorts of discourses that were promoted by The Chronicle during the Gukurahundi conflict in Zimbabwe between 1982 and 1986. It is argued that discourses in the news media are shaped by the role(s), the type(s) of journalism assumed by such media, and by the political environment in which the news media operate. The interplay between the roles, types of journalism practised, and the effect the political environment has on news discourses is assessed within the context of conflictual situations. This is done using insights from the theoretical position of peace journalism and its critique of professional or mainstream journalism as promoting war/violence journalism. Using the case of The Chronicle's reportage of the Gukurahundi conflict in Zimbabwe, it is concluded that, in performing the collaborative role, state owned/controlled media assume characteristics of war/violence journalism. On the other hand, it is concluded that The Chronicle developed practices consistent with peace journalism when it both espoused the facilitative role and journalistic objectivity. These findings undermine the conventional view among proponents of peace journalism that in times of conflict, the news media should be interventionist in favour of peace and that they should abandon the journalistic norm of objectivity which they argue, promotes war/violence journalism.

- Description: This research is premised on the understanding that media texts are discourses and that all discourses are functional, that is, they refer to things, issues and events, in meaningful and goal oriented ways. Nine articles are analysed to explicate the sorts of discourses that were promoted by The Chronicle during the Gukurahundi conflict in Zimbabwe between 1982 and 1986. It is argued that discourses in the news media are shaped by the role(s), the type(s) of journalism assumed by such media, and by the political environment in which the news media operate. The interplay between the roles, types of journalism practised, and the effect the political environment has on news discourses is assessed within the context of conflictual situations. This is done using insights from the theoretical position of peace journalism and its critique of professional or mainstream journalism as promoting war/violence journalism. Using the case of The Chronicle's reportage of the Gukurahundi conflict in Zimbabwe, it is concluded that, in performing the collaborative role, state owned/controlled media assume characteristics of war/violence journalism. On the other hand, it is concluded that The Chronicle developed practices consistent with peace journalism when it both espoused the facilitative role and journalistic objectivity. These findings undermine the conventional view among proponents of peace journalism that in times of conflict, the news media should be interventionist in favour of peace and that they should abandon the journalistic norm of objectivity which they argue, promotes war/violence journalism.

- Description: This study has focused on the design and preparation of bi– and tridentate N–Heterocyclic Carbene (NHC) ligands in order to investigate the effect of a multidentate approach to the formation, stability and catalytic activity of coordination complexes. Chapters 1 – 3 provide background information of relevant catalysis, carbene and coordination chemistry, followed by previous work performed within our research group. In Chapter 4 attention is given to the synthetic aspects of the research conducted, comprising two distinct approaches to the preparation of unsymmetrical saturated and unsaturated NHCs. Firstly, an investigation of the saturated NHC ligands yielded three novel, unsymmetrical pro–ligands, viz., two halopropyl imidazolinium salts and a bidentate hydroxypropyl imidazolinium salt. Secondly, eight imidazolium salts have been generated, including a hydroxypropyl analogue and novel decyl and tridentate malonyl derivatives. This study focuses on social learning in commercialisation of natural resource products in two case studies of commercial beekeeping in rural southern African contexts. In this study social learning entails a process of qualitative change taking place in a social context for the purpose of personal and social adaptation. This perspective is useful in this study as learning in the two cases, Hluleka in South Africa and Buhera in Zimbabwe involved the transition beekeeping.from traditional honey harvesting practices and subsistence beekeeping to commercial beekeeping. This study is informed by two related theoretical perspectives namely Cultural Historical Activity Theory (CHAT) and Social Learning Theory. CHAT was used as conceptual and methodological framework to inform the first phase of data gathering and analysis processes; as well as second phase data gathering. In the first phase, I gathered data through semistructured interviews, document reviews and observations to identify problems, challenges and critical incidents in learning commercial beekeeping, technically known as tensions and contradictions within the CHAT framework. These tensions and contradictions, surfaced through analysis of first phase data were used as "mirror data‟ in Intervention Workshops within CHAT's process of Developmental Work Research, which supports social learning in response to tensions and contradictions in workplace activity. Use of mirror data provided a basis for dialogue and the modelling of new solutions to identified contradictions. To interpret the social learning processes resulting from these interactions, I drew on Wals' (2007) analytical lenses, through which I was able to monitor social learning processes that emerged from the Intervention Workshop dialogues while beekeepers modelled new solutions to contradictions in learning commercial beekeeping. The findings of the study revealed that social learning in commercial beekeeping is internally and externally influenced by socio-cultural, political and economic complexities. Social learning in Intervention Workshops was supported by different knowledge bases of participants, in this study these are beekeepers, extension officers, trainers and development facilitators. Such knowledge bases were the source of information for learning and constructing model solutions. The study also revealed that learning in CBNRM workplaces can be observed across the development processes, and CHAT as a methodological tool and Wals‟ (2007) analytical tool are complementary and can be used in researching social learning in other CBNRM workplaces. The study contributes in-depth insight into participatory research and learning processes, especially within the context of CBRM in southern Africa. It gives some empirical and explanatory insight into how change-oriented social learning can emerge and be expanded in Education for Sustainable Development. It also provides learning and extension tools to work with contradictions that arise from socio-cultural and historical dimensions of learning commercialisation of natural resources in southern African context. Its other key contribution is that it provides further insight into the mobilisation of human agency and reflexivity in change oriented social learning processes of commercialisation of sustainable natural resources products and poverty alleviation processes that are critical for responding to socioecological issues and risks and development challenges in southern Africa.

Environment and sustainability issues require a community approach in order to bring about sustained responses to environmental challenges. As such, the study worked with social learning ideas of engaging the community in environment and sustainability learning. The data was generated mainly from face-to-face semi-structured interviews involving three community radio stations, Radio Listener Clubs and museum experts. The data generated was then presented to a strategy workshop involving the Livingstone Museum and Radio Musi-otunya staff. Arising from this workshop, recommendations were made about the possibility of the museum working in partnership with the radio to engage the community in environmental education. The study has shown that much of the museum environmental education activities have been confined to exhibitions and lectures within the museum building, which has affected the number of people being serviced by the museum. These education activities are arranged such that museum expert-led knowledge is presented to the audience with minimal community engagement on the environmental learning content. The study has also shown that community radio programming provides opportunities for community-led social learning which the Livingstone Museum could make use of to engage the community in environmental learning. Community radio programming allows community participation through Radio Listener Clubs, in identification and presentation of local environmental issues. This makes it a suitable tool to address locally relevant environmental issues, by the local community. Environmental issues are different from one place to another. Therefore environmental education approaches that bring issues into the museum may fail to address the different environmental education issues in different community context. The study concludes by recommending that Livingstone Museum should explore the use of community radio so that their expert knowledge and that of the radio producers could be used to shape environmental education programmes to go beyond awareness-raising.

- Description: A growing human population accompanied by urbanisation and industrialisation have led to over exploitation and pollution of freshwater resources and have consequently impacted on aquatic ecosystem health. The Swartkops River in the Eastern Cape of South Africa is no exception. It drains a heavily industrialised catchment which has led to deterioration of its water quality due to pollution. Integrated water resources management (IWRM) requires the concurrent sustainable use of water resources and the protection of aquatic ecosystem health. Macroinvertebrates are well known for their ability to reflect the health of the environment in which they live, thus they were used to assess anthropogenic impacts in the Swartkops River for this study. Macroinvertebrate based biomonitoring approaches, including the South African Scoring System version 5 (SASS5); a multimetric approach involving 19 metrics; Chironomidae community assessments and screening of morphological deformities in Chironomidae larvae, were applied at four selected sampling sites to assess environmental water quality in the Swartkops River. Macroinvertebrates were sampled us ing the SASS5 protocols. Chironomidae were mounted and identified as far as practically possible using available keys. Mentum, ligula, mandible, paraligula and antenna in Chironomidae larvae were screened for deformities. Physical and chemical water quality variables were measured at each of the selected sampling sites. All data were subjected to relevant statistical analyses. Of the four sites sampled during the study period, results revealed that water quality at site 1 was the least impacted with highest SASS5 scores, average score per taxa (ASPT) values, richness, diversity, equitability and Ephemeroptera –Plecoptera-Trichoptera (EPT) richness, as well as least incidences of chironomid deformities. Water quality at site 2 was considered the next least impacted with higher SASS5 scores, A SPT values, richness, diversity and equitability, and lower incidences of deformities compared to sites 3 and 4. SASS5 scores and ASPT values revealed that both sites 3 and 4 were critically modified but the multimetric analysis, Chironomidae community assessment and incidences of deformities in Chironomidae larvae indicated that site 3 is the most impacted of the four sampling sites, with least species diversity, richness, equitability and highest incidences of deformities. The study revealed the importance of multicriteria approach to environmental biomonitoring as an integrated water resources management tool, and based on the results, site 3, as the most impacted, could be prioritised for restoration intervention.

- Description: When South Africa became a democracy, many of its cultural institutions were tainted by the stigma of having been tools for the production and propagation of apartheid ideology. This thesis examines two key facets of post-apartheid museums and memorials. Firstly, how they have repositioned themselves as institutions of cultural and social standing. Secondly, their role as tools of nation building, social change, and creators of national collective memory within the new democratic South Africa. Through an analysis of cultural memory theory pertaining to museology, this study elaborates on the methods employed by museums to incorporate memory into their narratives and in turn, transfer collective memory to their viewers. This thesis provides a comparative study of the architectural, memorial and museological strategies of two post-apartheid museums; the Red Location Museum and the Apartbeid Museum. It examines the contributions of both museums to the introduction of new museological strategies for the successful creation and transmission of South African collective memory. Through this analysis, both the invaluable contributions and the drawbacks of post-apartheid museums as tools for the promotion of new democratic ideologies and philosophies are considered. This thesis does not resolve the arguments and questions which have surfaced regarding cultural institutions as tools for the promotion of reconciliation and the construction of national collective memory within South Africa. As the current climate of memorialisation is one of change and paradox, it is presently impossible to fully quantify post-apartheid museums' roles within South Africa's move toward reconciliation and social change. However, the examination of both the Red Location Museum and the Apartheid Museum reveals the extraordinary change that South African cultural institutions have undergone in addition to their potential to become institutions which facilitate active reconciliation as well as social and cultural growth.

- Description: When South Africa became a democracy, many of its cultural institutions were tainted by the stigma of having been tools for the production and propagation of apartheid ideology. This thesis examines two key facets of post-apartheid museums and memorials. Firstly, how they have repositioned themselves as institutions of cultural and social standing. Secondly, their role as tools of nation building, social change, and creators of national collective memory within the new democratic South Africa. Through an analysis of cultural memory theory pertaining to museology, this study elaborates on the methods employed by museums to incorporate memory into their narratives and in turn, transfer collective memory to their viewers. This thesis provides a comparative study of the architectural, memorial and museological strategies of two post-apartheid museums; the Red Location Museum and the Apartbeid Museum. It examines the contributions of both museums to the introduction of new museological strategies for the successful creation and transmission of South African collective memory. Through this analysis, both the invaluable contributions and the drawbacks of post-apartheid museums as tools for the promotion of new democratic ideologies and philosophies are considered. This thesis does not resolve the arguments and questions which have surfaced regarding cultural institutions as tools for the promotion of reconciliation and the construction of national collective memory within South Africa. As the current climate of memorialisation is one of change and paradox, it is presently impossible to fully quantify post-apartheid museums' roles within South Africa's move toward reconciliation and social change. However, the examination of both the Red Location Museum and the Apartheid Museum reveals the extraordinary change that South African cultural institutions have undergone in addition to their potential to become institutions which facilitate active reconciliation as well as social and cultural growth.

The first is a three-dimensional visualisation tool which allows for live, near-realtime analysis, and the second is a two-dimensional fractal based plotting scheme which allows for plots of the entire IPv4 address space to be produced, and manipulated. Using the techniques and tools developed for the analysis of this dataset, a detailed analysis of traffic recorded as destined for port 445/tcp is presented. This includes the evaluation of traffic surrounding the outbreak of the Conficker worm in November 2008. A number of metrics relating to the description and quantification of network telescope configuration and the resultant traffic captures are described, the use of which it is hoped will facilitate greater and easier collaboration among researchers utilising this network security technology. The research concludes with suggestions relating to other applications of the data and intelligence that can be extracted from network telescopes, and their use as part of an organisation’s integrated network security systems

- Description: The 28-month study assessed the impacts of five syntopic medium-sized mammalian browsers and one fire event in a woodland savanna in the Matobo Hills, Zimbabwe. Aspects of herbivory, mechanical pressures, seed dispersal and nutrient cycling were investigated for three species of small antelope (common duiker [Sylvicapra grimmia]1, klipspringer [Oreotragus oreotragus] and steenbok [Raphicerus campestris]) and two medium-sized species (bushbuck [Tragelaphus scriptus] and greater kudu [T. strepsiceros]). Focusing on Burkea africana2 woodland, in a system that does not include elephant (Loxodonta africana), effects of browsing antelope on woody and herbaceous vegetation development were investigated using exclusion plots. Browsers regulated woody plant cover (measured as basal stem area), with smaller antelope having a greater impact than larger species. This was linked to feeding height, feeding selectivity and mechanical pressures (e.g. twig breakage and trampling). Fire caused an initial reduction in above-ground standing biomass, but in the presence of fauna, pre-fire equilibria were attained within 15 months. In antelope exclosures, herbaceous biomass increased and woody biomass decreased following fire. Responses by woody vegetation to browsing varied among species, with highly palatable species typically exhibiting compensatory regrowth. Woody species richness and abundance (especially of palatable species) increased in the absence of browsers, but species richness of the herbaceous layer was promoted by moderate disturbance (trampling or fire). Faecal deposition behaviour, primarily the use of latrines by small antelope, resulted in localised soil enrichment within defended territories. Decomposition rates (and therefore return of nutrients to the soil) varied among species and seasons, due to defecation site selection, accessibility to decomposers and desiccation rates of faecal pellets. Controlled seed germination experiments indicated that ingestion by small antelope enhances germination rates of large, hard-seeded fruits such as Sclerocarya birrea. However, germination of savanna seeds may require multiple cues. This study demonstrated the critical roles of small antelope in ecosystem functioning, and highlights the importance of the less visible impacts of frequently overlooked smaller mammalian herbivores. Perturbations to the faunal community, especially small antelope, are predicted to have substantial impacts on woody plant cover.

These focus on the representation of male bodies and experience. Having begun his career as a collaborative printmaker, since 2002, his work has become more ambitious as well as critically acclaimed. In 2010, his most recent body of work, Transitions, was exhibited at the Smithsonian Museum of African Art in Washington D.C. I propose that Emmanuel represents the male body as a presence that either is not easily seen or that actively disappears or erases itself. Its subjectivity, and the viewer’s engagement with it, may be characterised as one of intimacy, exposure, loss and vulnerability. Emmanuel’s work may be said to question conventions and ideals of masculinity while, at the same time, refusing any prescriptive interpretation. To develop this proposition, I examine specifically Emmanuel’s incising drawing technique that ‘holds open’ transitions in male lives. In these liminal moments, Emmanuel represents men as ‘seen’ to change state or status, thereby exposing the ongoing process of building masculine identities. Equally elucidatory is Emmanuel’s imprinting of his own body, which, in his use of “traces” that reveal the vacillation between presence and absence, makes contingently ‘visible’ this gendering process, and has particular implications for the expression of subjectivity in a contemporary South African context.

The formation of conjugates was confirmed through the use of the following analytical techniques: UV-vis, FTIR, Raman and XRD spectroscopies, atomic force and transmission electron microscopies and voltammetry. Chemically linking SWCNT to MPcs created platforms that offered efficient transfer of electrons and this was confirmed through electrochemical impedance studies (EIS) and voltammetry as shown by lower ΔEp values observed in conjugates. Carboxy carrying MPcs have very poor electron transfer kinetics (both tetrasubstituted and low symmetry), but the presence of SWCNTs activates their catalysis. All electrochemical studies were done at pH 4. Cyclic voltammetry, rotating disk linear sweep voltammetry, chronoamperometry and EIS were used in the electrochemical characterization of 2-ME and the pesticides on poly-Ni(OH)TAPc and MPc-SWCNT modified glassy carbon electrodes (GCEs). High Tafel slopes were observed for the pesticides relative to 2-ME, an indication of the passivating nature of their oxidation products. However, conjugates showed very high resistances to passivation and were easily regenerated by shaking in methanol. Improved catalysis of the conjugates is also indicated by the high catalytic rate constants for the analytes, observed on these electrodes. Conjugates of low symmetry MPcs with SWCNTs gave the highest catalytic rate constants, confirming better catalysis on these electrode surfaces. The nature of SWCNT functionalization also affected catalysis, with amine functionalized SWCNTs inducing better catalytic properties into the MPcs than carboxylic acid terminated CNTs. The presence of amine functionalized SWCNTs activates the catalysis of non-catalytic carboxy-carrying MPcs and this is more pronounced in conjugates of tetrasubstituted MPcs relative to those of low symmetry Pcs. Ethylene amine (EA) functionalized SWCNTs reduced redox overpotentials of the MPcs more than the phenyl-amine (PA) functionalized counterparts. Poly-NiTAPc was successfully converted to poly-Ni(OH)TAPc through cyclisation in pH 4 buffer and showed very good catalytic properties towards diuron, relative to the former.

- Description: The introduction of outcomes based education (OBE) in the form of Curriculum 2005 (C2005), the National Curriculum Statement (NCS) and the Revised National Curriculum Statement (RNCS) in post-apartheid South Africa resulted in a shift from a content-centred to a learner-centred view on education. This transition took place rapidly as the new government wanted to introduce a democratic education system after the divisive Bantu education system from Apartheid. However, after the changes were implemented, education in South Africa was theoretically outcomes based but practically many educators were still teaching in a content-centred manner. The research puts forward the proposal that drama-in-education (D-i-E) is a useful means by which to align the practical and theoretical goals of OBE within the context of South Africa's current RNCS. This hypothesis drives the main research question: "Can D-i-E be an effective teaching methodology to realise the goals of the RNCS and generate OBE learning environments in a South African high school?" D-i-E is a learner-centred teaching methodology and in practise it meets many of the goals and Critical Cross-Field Outcomes (CCFOs) of OBE. Some of these include the fact that learners can: • Practice problem-solving skills; • Engage with critical and creative thinking; • Grow cultural and aesthetic sensitivity; • Work effectively in groups; and ii. • Learn in inclusive environments that cater for different learning styles and levels. The research examines the use of D-i-E as an outcomes based methodology by which the RNCS could be implemented in the classroom. This is explored through the use of qualitative research in the form of a case study investigation at a South African high school. - Description: This study aims to explore four grade 3 teachers’ understanding of teaching reading in Rukwangali, a home language in Namibia in Kavango region and to observe the strategies they use as well as the activities they set for their learners. During my School Based Studies (SBS), I noticed that learners were not reading with understanding. My interest was also stimulated by a study carried out in Namibia that found that the literacy levels for Namibian learners were poor compared to those of other African countries. This is a qualitative case study carried out within an interpretive paradigm. It seeks to understand the meaning that people attached to human actions. I selected the participants purposefully and in terms of convenience, as all three schools selected are situated in Rundu, the town in which I work as a college lecturer. The research tools I employed were semi-structured interviews, document analysis, classroom observation, and stimulated recall discussions on the lessons I observed. I used a variety of methods to triangulate data and enhance validity. This study reveals that teachers use multiple methods to teach reading. These methods include phonics and syllabification to help struggling learners to decode difficult and long (polysyllabic) words and the look-and-say method for whole word recognition and meaning making at word and sentence level. In addition, they also use the thematic approach to teaching and learning to expand learners’ vocabulary and enhance their understanding of the texts they read and to make their lessons learnercentred. However, the problems of language, lack of reading books written in Rukwangali and large class sizes constrain the teachers from teaching in a more learner-centred way. Therefore, the study gives some tentative recommendations to remedy this situation.

Transfer pricing and especially business restructuring within multinationals is a fairly new concept.Professional legal and audit firms have different views on how to approach business restructurings. This research analyses important transfer pricing aspects and the anomalies that arise through business restructurings. The research method used in this research paper is primarily qualitative, comprising the analysis of various documentary sources of data. Relevant South African and international case law, tax legislation, the OECD Transfer Pricing Guidelines for Multinational Enterprises and Tax Administrations, the Transfer Pricing Aspects of Business Restructurings Discussion Draft and other reports were consulted and analysed. Further the views of recognised legal and tax experts that have been published in technical journals and text books were also considered and examined. A hypothetical example of a business restructuring transaction was constructed in order to illustrate practical issues and different approaches to solving them. The research has argued that the arm’s length principle, which forms the bases of transfer pricing regulation, is not an exact science but theoretically it is the most suitable measure.It may not be able to incorporate all variables, such as the cost savings through synergies of multinational enterprises, but it promotes international trade and investment by ensuring that transactions are based on fair prices. Business restructurings create anomalies in applying the arm’s length principle but these anomalies can be dealt with within the regulatory structure. The business restructuring approach recommended is realistic and pragmatic, but more clarity may be needed in certain circumstances. The research has also discussed the avoidance of transfer pricing audits, including having appropriate transfer pricing policies and documentation.

Investigating the role of heat shock proteins (Hsps) 40, 70 and 90 in the life cycle of Theiler's murine encephalomyelitis virus (TMEV)

- Description: Introduction: Picornaviruses are a family of RNA viruses which are economically and clinically significant. Like many other viruses, picornaviruses utilise host cell machinery to facilitate their replication and assembly, including heat shock proteins (Hsps). The aim of this research was to investigate the role of Hsp40, Hsp70 and Hsp90 during picornavirus infection using the cardiovirus, Theiler’s murine encephalomyelitis virus (TMEV), as a study model. Methodology: Picornavirus VP1 capsid proteins were analysed by multiple sequence alignment and multiple structural comparisons. Protein domain architecture was used to analyse Hsp90 cellular and viral client proteins. Effects of Hsp90 inhibitors, novobiocin and geldanamycin, on TMEV growth in BHK-21 cells was observed over a 48hr period. Localisation of Hsp40, Hsp90 and Hsp70 in TMEV-infected BHK-21 cells was investigated by indirect immunofluorescence and confocal microscopy. Results and Discussion: VP1 proteins of picornaviruses are highly divergent within the family at the amino acid level, which might be linked to the protein’s function in determining virus tropism and antibody neutralisation. An eight-stranded anti-parallel beta-barrel structure was found conserved in the VP1 protein structures which might be linked to the highly conserved picornavirus capsid assembly process. Absence of a common protein domain between Hsp90 viral and cellular client proteins that might be functionally connected to Hsp90, suggests that Hsp90 most likely recognises surface features rather than sequence motifs/patterns. The Hsp90 inhibitors, novobiocin and geldanamycin, had a negative effect on virus growth as virus-induced cytopathic effect was not observed in treated cell after 48hrs. TMEV 2C protein was detected by Western analysis in infected cell lysates treated with geldanamycin but not novobiocin, suggesting novobiocin affects the translation or processing of TMEV 2C. Immunofluorescence analysis of TMEV-infected cells showed a relocalisation of Hsp40 into the nucleus during infection. Overlap of Hsp40 and TMEV P1 was observed in the perinuclear region, suggesting colocalisation between these proteins. Hsp70 converged around the replication complex during infection but did not overlap with TMEV 2C. Hsp90 concentrated in the region of the replication complex where it overlapped with TMEV 2C and this redistribution was found to be dependent on the stage of infection. The overlap between Hsp90 and TMEV 2C signals observed, suggested colocalisation between the two proteins. Conclusion: This study identified Hsp90, Hsp70 and Hsp40 as possible host factors required in TMEV replication.
