Information and knowledge are key strategic resources for both social and economic development, and nowadays they most often rely on Information and Communication Technologies (ICTs). Poor communities have limited or no access to functioning e-infrastructure, which underpins ICT. The Siyakhula Living Lab (SLL) is a joint project between the universities of Rhodes and Fort Hare that has been running for over 15 years now. The SLL solution is currently implemented in schools in the Eastern Cape’s Dwesa-Mbhashe municipality as well as schools in Makhanda (formerly Grahamstown). Over the years, a number of blueprints for the meaningful connection of poor communities was developed. The research reported in this thesis sought to review and improve the Siyakhula Living Lab (SLL) blueprint regarding fixed computing infrastructure (as opposed to networking and applications). The review confirmed the viability of the GNU/Linux Terminal Server Project (LTSP) based computing infrastructure deployed in schools to serve the surrounding community. In 2019 LTSP was redesigned and rewritten to improve on the previous version. Amongst other improvements, LTSP19+ has a smaller memory footprint and supports a graphical way to prepare and maintain the client’s image using virtual machines. These improvements increase the potential life of ICT projects implementing the SLL solution, increasing the participation of members of the community (especially teachers) to the maintenance of the computing installations. The review recommends the switching from thin clients deployments to full ("thick") clients deployments, still booting from the network and mounting their file systems on a central server. The switch is motivated by reasons that go from cost-effectiveness to the ability to survive the sudden unavailability of the central server. From experience in the previous deployment, electrical power surge protection should be mandatory. Also, UPS to protect the file system of the central server should be configured to start the shutdown immediately on electrical power loss in order to protect the life of the UPS battery (and make it possible to use cheaper UPS that report only on network power loss). The research study contributed to one real-life computing infrastructure deployment in the Ntsika school in Makhanda and one re-deployment in the Ngwane school in the Dwesa-Mbhashe area. The writer's interest in this phenomenon grew out of her own experience of this situation as a clinical Masters coursework student, an experience which was of important to the writer and meaningful still as a therapist in training. In dialogue with experienced therapists the importance of this situation was again made apparant. It was the opinion of these therapists that although for some the details of this experience had become dulled by time, what remained meaningful to them was that this experience was seen as the beginning of a project which remains important to them - they identified this situation as an important moment in the history of their development as psychotherapists. In the hope that the literature pertaining to psychotherapy would throw some light on this situation, the writer turned to a number of sources in this area to discover that no literature available to her elucidated this situation in a holistic manner. This led the writer to go back to the beginning therapists themselves so that they may speak for themselves of their experience of this situation. A phenomenological method of enquiry is implemented in this study as it renders the subject matter accessible to investigation, and allows it to reveal itself as it essentially is. - Description: This work addresses the role of mirroring and mirrors encountered while working with a patient obsessed with his appearance. At the same time, however, it also attempts to provide an understanding of the process of mirroring from the developing unborn child to the adult. It examines the womb as a reflective space, the mirroring role of the mother, psychological work as mirror work, the psychopathologies of disruptive mirroring and the nature of reflection as a story, confusion and a ghostly other, and finally, the relationship between mirror reflection and psychological life. What is examined and described in this process applies not only to the specific case in study but is relevant at a universal level to all of mankind regardless of race or religion. The case-study method was used to examine the process of therapy that took place over three months. The patient’s identifying data, reasons for referral, presenting problem, family background, basic personality, interpersonal relationships, mental state, diagnosis, dynamic formulation and rationale for psychotherapy were covered in detail. The process of psychotherapy over 27 sessions was recounted and discussed in detail. The role of local level agency in a just green transition: the case of Rhodes University

- Description: The research uses a richly contextualised case study of Rhodes University to explore the role of local level agency in a just green transition. The central concept of the thesis is mainstreaming sustainability. Sustainability has become a core objective both at the macro and micro levels. The just green transition and triple bottom line are shorthand for these macro and micro concepts. At the macro level, there is increasing evidence suggesting that transitioning to a sustainable economy can be a key driver of economic development. At the micro level, the elements of the triple bottom line increasingly overlap, with sustainability no longer a separate goal, or a ‘nice to have’, but integral to organisational success. However, this potential is clearly not being realised, and sustainability often remains ‘niche’. Lack of progress at the macro-level reinforces the importance of bottom-up, local level agency. In keeping with the broader micro-level literature, the case study strongly suggests that mainstreaming sustainability would have multiple benefits. These include reducing dependence on unreliable state-provided services and enhancing Rhodes University’s standing as a genuinely transformative institution. The evidence suggests that there is a pure financial case for green investments, such as the construction of a solar farm at Rhodes University, even before accounting for the social and environmental benefits of such an initiative. This shifts the focus to why institutions like Rhodes University have not been proactive in mainstreaming sustainability. The document analysis and the interviews showed that there is an awareness of the importance and potential of mainstreaming sustainability. However, the funding squeeze is often misperceived as a binding constraint, and there is an absence of innovative thinking about how to finance projects with high returns, such as a solar farm. A theme amongst several of the interviewees was that the university should embrace a policy of enhancing small changes as a way of mainstreaming sustainability gradually. Even here, there are doubts about whether the organisational structure of the university will allow this. However, this potential is clearly not being realised, and sustainability often remains ‘niche’. Lack of progress at the macro-level reinforces the importance of bottom-up, local level agency. In keeping with the broader micro-level literature, the case study strongly suggests that mainstreaming sustainability would have multiple benefits. These include reducing dependence on unreliable state-provided services and enhancing Rhodes University’s standing as a genuinely transformative institution. The evidence suggests that there is a pure financial case for green investments, such as the construction of a solar farm at Rhodes University, even before accounting for the social and environmental benefits of such an initiative. This shifts the focus to why institutions like Rhodes University have not been proactive in mainstreaming sustainability. The document analysis and the interviews showed that there is an awareness of the importance and potential of mainstreaming sustainability. However, the funding squeeze is often misperceived as a binding constraint, and there is an absence of innovative thinking about how to finance projects with high returns, such as a solar farm. A theme amongst several of the interviewees was that the university should embrace a policy of enhancing small changes as a way of mainstreaming sustainability gradually. Even here, there are doubts about whether the organisational structure of the university will allow this. On the other hand, there are positive signs that the increasing sense of crisis means management and other key stakeholders are gradually shifting towards seeing the crucial importance of the university embracing a more proactive stance. , Thesis (MCom) -- Faculty of Commerce, Economics and Economic History, 2023

Human FN1 is regulated by the heat-shock response

- Description: Heat shock protein 90 (Hsp90) and heat shock factors (HSFs) are known to be involved in the epigenetic regulation of several fundamental oncogenic genes. Fibronectin (FN) is an extracellular matrix (ECM) glycoprotein which plays key roles in cell adhesion and migration. Hsp90 binds directly to FN and Hsp90 inhibition has been shown to regulate FN protein levels and matrix formation. Where inhibition of Hsp90 with a C-terminal inhibitor (novobiocin) induced the loss of FN matrix, treatment with an N-terminal inhibitor (geldanamycin) increased FN matrix levels. GA treatment induced a strong dose and time dependent increase in FN1 promoter activity and increased total FN mRNA respectively. By contrast, NOV showed no increase in the promoter activity and no change in the expression of FN mRNA. As GA is known to induce the stress response, we investigated the relationship between the cell stress machinery and the transcriptional regulation of FN. Three putative heat shock elements (HSEs) were identified in the FN1 promoter. The loss of two of the three identified putative HSEs resulted in a loss in the basal transcriptional activity of the FN1 promoter in our reporter model. This was in addition to the loss of the induction of transcriptional activity with GA treatment observed with the full-length promoter. Binding of HSF1 to one of the putative HSEs, which was identified as potentially functional from the truncation analysis, was confirmed using ChIP. The occupancy of this HSE by HSF1 was shown to increase with GA treatment. These data support the hypothesis that FN1 is a functional HSF1 target gene. The 5' promoter regions of seven additional ECM protein encoding genes were analysed and mRNA levels were detected by quantitative RT-PCR upon treatment with GA. Collagen 4 _2 and laminin _3 mRNA were found to increase in the presence of GA, whereas collagen 4 _3 and osteopontin showed no change. Similarly to FN1, these data indicate that a subset of ECM genes may be under the regulation of the HSF1 mediated heat-shock response. This may have implications for our understanding of ECM dynamics in cancer, where the clinical application of Hsp90 inhibitors is intended.
