- Description: This current study of investigation reports on the synthesis of 1,4-dihydroxyanthraquinone and its derivatives on explorations of their medicinal potential. The study initially aimed to synthesize an analogue of a natural anthraquinone, 1,3,6-trihydroxy-7-((S)-1- hydroxyethyl)anthracene-9,10-dione 5 using Friedel-Crafts acylation of phthalic anhydride and a benzene derivative. Synthetic transformation of anacardic acid 63, obtained as a by- product of the cashew industry successfully afforded 4-ethoxyisobenzofuran-1,3-dione 89. However, when attempted to couple 4-ethoxyisobenzofuran-1,3-dione 89 with 2- hydroxyacetophenone 91 in a Friedel-Crafts acylation manner to form 2-acetyl-1,8- dihydroxyanthracene-9,10-dione 87 the reaction did not work efficiently. A simple derivative of benzene which is; benzene-1,4-diol 102 was reacted instead with 3-ethoxyphthalic acid 71 and isobenzofuran-1,3-dione 96 to form 1,4,5-trihydroxy anthraquinone 72 and 1,4- dihydroxyanthraquinone 42, respectively. A modified Marschalk reaction was then used to introduce the hydroxyl alkyl group to 1,4-dihydroxy anthraquinone 42, which allowed further elaboration of the hydroxyl-substituent in moderate to good yields (22-80%). A molecular docking study was performed using Schrödinger software to predict the binding affinity of the test compounds to the target protein trypanothione reductase (PDB ID: 6BU7). An in-vitro screening of 1,4-dihydroxyanthraquinone derivatives and some selected precursors for antitrypanosomal, antiplasmodial, antibacterial, and cytotoxicity activities produced encouraging results. Derivatives of anacardic acid and cardanol from CNSL were found to have moderate activity against trypanosomes with no activity against Plasmodium falciparum. Almost 63% of synthesized 1,4-dihydroxyanthraquinone derivatives displayed activity against trypanosomes. The in-vitro evaluation and the in silico molecular docking studies revealed that 1,4-dihydroxyanthraquinone derivatives can be potential drug-like candidates active against T.brucei parasites (IC50 = 0.70-1.20 μM). Only four 1,4- iv dihydroxyanthraquinone derivatives with thiosemicarbazone, chloride, pyrrole, and diethanolamine functionality displayed activity against Plasmodium falciparum (IC50 = 3.17- 14.36 μM). In-vitro evaluated of test compounds against antibacterial screen and cytotoxicity effects significantly showed that 2-hydroxy-6-pentadecylbenzoic acid 63a and 2-((2- chlorophenyl)(piperazin-1-yl) methyl)-1,4-dihydroxyanthracene-9,10-dione 78 have potency against Staphylococcus aureus and reduced the viability of the cells below 20% at an initial concentration of 50 μg/mL. Only 1,4-dihydroxyanthraquinone derivatives with thiosemicarbazone 76, piperazine 78, and diethanolamine 80 motifs were active against HeLa cells and reduced the viability of cells below 20% at a concentration of 50 μg/mL. In conclusion, this current reported study has generated useful knowlege on the applicability of the agro-waste CNSL as an agent active against trypanosomiasis but also as a low-cost starting material to synthesize hydroxy anthraquinones. The study has further given an overview to the understanding of the medicinal value 1,4-dihydroxyanthraquinone derivatives as promising candidates towards developing drugs suitable for treating neglected tropical diseases particularly trypanosomiasis. , Thesis (PhD) -- Faculty of Science, Chemistry, 2022

Older persons’ perceptions of services that support positive ageing in the Khayelitsha and Gugulethu districts of the Western cape

- Description: Globally people are growing older, and nearly every country is experiencing an increase in their ageing populace. In 1960 older people made up 5 % of the world population, while in 2018, they were about 9% of the overall population. Projections are that in 2050 people over the age of 65 will make up 16% of the overall population. To respond to an ageing population, a government must ensure that its policy framework can respond to the needs of its citizens. A society that supports a positive ageing environment improves older people’s well-being. In our complex society, each person is dependent on others in various ways, and social, emotional and practical interdependence is a feature of the human social condition. This means that older people do not function well in isolation but need support for positive ageing. This research project focused on older people’s perceptions of services that support their positive ageing in the Khayelitsha and Gugulethu districts of the Western Cape, South Africa. The theoretical framework employed in the study is Systems Theory. The research approach utilised in the study is qualitative with an explorative and descriptive research design. A non-probability purposive, criterion-based sampling technique was employed. The researcher recruited eleven older persons aged sixty years and above in the Khayelitsha and Gugulethu areas. An NGO that works with older persons’ served as gatekeepers. Semi-structured telephonic interviews were conducted. The data collected was analysed using thematic data analysis. The trustworthiness of the research process was enhanced using Guba’s model. Ethical principles as set out in the Belmont report were also adhered to. The key findings of the current study indicated that in Khayelitsha and Gugulethu areas, there are available services that support positive ageing, such as clubs, excursions and social grants. Environmental aspects challenging positive ageing included unfavourable home conditions such as financial burden and ill-treatment, poverty and unemployment, health problems, and COVID-19 effects. Older people mentioned that there are available but inadequate services such as municipal services and care facilities. Thus, they recommended that there should be poverty alleviation schemes, community development projects such as crafting, gardening, sewing, and more institutions and home-based care for older people. , Thesis (MA) -- Faculty of Health Sciences, 2022

For me, isiXhosa captures the innate musicality and deep emotions within a word or a sound that are not available in English. My poems use the music and introspection of the lyric form, as well as the emotional outpouring that prose poetry allows. As a bilingual writer, I am influenced by the transference of musicality and gravity of loss conveyed in the translated and bilingual work of Isabella Motadinyane, the Russian Marina Tsvetaeva, Stephen Watson’s Song of the Broken String, as well as the Spanish poets Antonio Machado. The structure of my thesis is inspired by the innovative mixed genre layout of Sindiswa Busuku-Mathese’s Loud and Yellow Laughter, and the new formats of isiXhosa writing presented in Mthunzikazi Mbugwana’s poetry. My work has also been shaped by the visceral imagery and briefly captured moments of loss in the work of South African poets vangile gantsho, S.S. The structure of my thesis is inspired by the innovative mixed genre layout of Sindiswa Busuku-Mathese’s Loud and Yellow Laughter, and the new formats of isiXhosa writing presented in Mthunzikazi Mbugwana’s poetry. My work has also been shaped by the visceral imagery and briefly captured moments of loss in imisebenzi ka S.S Mema, Nontsizi Mgqwetho, noPascale Petit, novangile gantsho. Imisebenzi yabo ikuzobela umfanekiso ngqondweni ophilayo. Bonke abababhali bahambe indlela endinika umdla nendifuna ukuyihamba nam njengombhali omtsha obhala ngeelwimi ezimbini. , My thesis is a collection of poems and prose that explores the disjunction of losses for women and children across the two languages I inhabit, isiXhosa and English. For me, isiXhosa captures the innate musicality and deep emotions within a word or a sound that are not available to me in English. On the other hand, English readily produces visceral images that are more difficult for me to find in isiXhosa. My poems use the music and introspection of the lyric form, as well as the emotional outpouring that prose poetry allows. As a bilingual writer, I am influenced by the transference of musicality and gravity of loss conveyed in the translated and bilingual work of Isabella Motadinyane, the Russian Marina Tsvetaeva, Stephen Watson’s Song of the Broken String, as well as the Spanish poets Antonio Machado. The researcher recommends that the CG policy be prioritised by the Mpumalanga DOH, that systems be put in place to facilitate policy implementation, and that the departmental staff establishments at all levels, prioritise healthcare professionals in key leadership positions. The Food for Us mobile application lacks a supporting sustainable social business model to sustain its continued operations, hence the need to develop one. In the study, data was generated qualitatively underpinned by an interpretive paradigm, in three workshops guided by the Delphi method. A stakeholder salience model was applied to identify key stakeholders and their salience. Three key stakeholders, namely users (farmers and buyers), Experts (App developers) and the consortium were identified. The study concluded that developing a sustainable social, innovative business model requires substantive consultation with multiple stakeholders in society. Every stakeholder is important and possesses varying salience, hence stakeholder mapping is an important exercise. The study further concluded that financial sustainability and social inclusion are critical social enterprise elements to consider in the process. The undertaking to enhance financial sustainability opens an understanding on the importance of income streams, the key activities, and value propositions offered by the mobile application. The need to remain socially inclusive brings forth questioning of value propositions, accessibility, user friendliness towards stakeholder diversity and needs. A number of studies have shown that melatonin may be of potential benefit for the treatment of insomnia, as well as neurodegenerative disorders. At present, there are numerous dosage forms of melatonin, with the oral route of administration being most popular. Presently, there is little information on the stability of melatonin over a pH range. With the changes in pH in the gastro-intestinal tract, as well as in different experimental conditions, information on the stability of melatonin would be important. We used a high-performance liquid chromatography method to determine the stability of melatonin solutions over a pH range (1.2–12) at room temperature and at 37°C over a period of 21 days. The results show that no melatonin degradation occurred in the first 2 days. From days 3 to 21, there was a gradual decline in melatonin at all pHs, with the decline not exceeding 30%. No decline in melatonin levels occurred in the first 2 days at 37°C. From days 3 to 21, melatonin levels declined gradually, with the decline not exceeding 29%.

- Description: Warburgia salutaris, locally known as “chibaha” (Ronga, Tsonga), “isiBhaha” (Zulu) and “siBhaha” (Siswati), is one of the most highly-valued medicinal plant species in southern Africa. Its bark has long been employed to treat bacterial and fungal infections, and it is often a front line herbal treatment for malaria. The species is popular in rural and urban areas due to its lower price compared to commercial pharmaceuticals and because of its long-standing spiritual and symbolic significance. Consequently, W. salutaris is threatened in Malawi, South Africa and Swaziland and is believed to be extincted in the wild in Zimbabwe. In Mozambique, where the tree is confined to the southern part of the country, it is considered vulnerable due to high local demand and cross-border export. Although there is limited research, further work is urgently needed to provide the basis for options and strategies to achieve desired outcomes. This study aimed to explore different approaches for the conservation of W. salutaris in the Lebombo Mountains (LM), the Tembe River (TR) and the Futi Corridor (FC) areas, in southern Mozambique. For this, potential distribution, socio cultural factors that influenced sustainable management of the species, demography, harvest impact, genetic diversity and population differentiation were considered. The kuenm package in R, 2 846 occurrence and 11 environmental data were used to model the potential current and future distribution in 2050 and 2070 using four general circulation models (CESM1(BGC), ACCESS1.3, FIO-ESM and IPSL-CM5A-MR) under representative concentration pathways (RCPs) 4.5 and 8.5. The extent of occurrence (EOO) and habitat suitability (HS) were also determined. Stratified random semi-structured interviews with 182 heads of households were carried out to explore cultural and socio incentives of use, compared knowledge distribution, determine local management practices and explore local ecological knowledge related to the species in the three study areas. In addition, 17 focus group discussion were conducted to clarify issues raised during household interviews. One hundred and twenty four quadrants were used to determine the abundance and population structure. The Simpson index of dominance, permutation index and quotient between consecutive classes were used to assess population stability, and an eight point scale was used to evaluate the extent of bark damages. Genomic deoxyribonucleic acid (gDNA) (n=48) was used to assessed genetic diversity and population structure through the newly developed single sequence repeats (SSR) molecular markers. Model selected was statistical significant for prediction of potential distribution of W. salutaris in southern Mozambique (partial ROC =0.00, omission rate = 0.04 and delta AICc = 0.00). The current environmental conditions mostly explained by climatic predictor, indicated in general, the potential distribution on areas of occurrence observed during the field work in southern Mozambique. Future project suggested an increase of climatic suitable areas in all the general circulation models in both conservative and pessimistic climatic scenarios (RCPs 4.5 and 8.5, respectively) in 2050 and 2070, and consequently the increase of HS and the EOO. Results of this work suggests W. salutaris would not extinct at least in the coming 50 years due to climate change but other threats such as overexploitation of the species need to be addressed. With increase of climatic suitable areas, conservation and management of this species could focus on reintroduction and cultivation for recovery of the species, with the help of time. W. salutaris was used for medicinal purposes to treat 12 health problems, with the bark being the most commonly used part (92% of informants). Knowledge of the species uses differed significantly between the three study areas but not with respondent gender or age. Informants also mentioned a considerable number of alternative species of W. salutaris (37 species in LM, 33 species in TR and 17 species in FC). Local ecological knowledge on the ecological niche of the species also varied substantial in the three areas with both types of knowledge generally higher in the Tembe River area. Local management practises were revealed mostly on harvesting through vertical bark stripping cited by 100% of informants in TR, 61.6% FC and 51.7 LM (71% of all informants). Yet most (64.3% of all informants, corresponding to 98.4% in TR, 51.7% in Fc and 41.7% LM) used non-traditional harvesting tools (machete) bark removal. For sustainable use of the species, consideration should be given to combined conservation strategies such as the use of alternative species with the same application, substitution of bark by leaves, and increases in alternative sources of plant material through cultivation. The densities of alive stems were not statistically different in the three areas but was higher in LM (mean of 4500.3±9209.8 stems/ha). Equally the mean densities of recruits (diameter ≤2 cm) were not considerable different but was higher in the FC (3296.8±9116.7 stems/ha). The regression analyses for the three study area revealed negative slopes, indicating more recruits and fewer individuals in larger classes. The Simpson indexes of dominance, permutation indexes and quotients between the consecutive size classes suggested some levels of population instability in the three areas. Harvesting took place in the eight point scale, all size classes were harvested but preference was higher on intermediate (15.1-20 cm) or larger (>20 cm) size classes in all the study areas but gatherers avoided harvesting bark from recruits. Although the mean densities of harvested stem was not significant this work indicated higher levels of stem destruction in FC. Harvested dead stems ware significant in the three areas and FC had the higher of (39.5±85.9 stems/ha) compared to 7.6±19.1 and 2.1±4.3 in TR and LM, respectively. Equally, the density of physiologically stressed stems differed substantially in the three areas with higher in FC 20.5± 75.5 stems/ha compared to 18.7±82.9 stems/ha in TR and 3.8±11.8 stems/ha in LM. This results suggests implementation of protection measures such as the inclusion of the species in the national CITES list to regulated cross-border trade and strengthening of the law enforcement. - Description: Warburgia salutaris, locally known as “chibaha” (Ronga, Tsonga), “isiBhaha” (Zulu) and “siBhaha” (Siswati), is one of the most highly-valued medicinal plant species in southern Africa. Its bark has long been employed to treat bacterial and fungal infections, and it is often a front line herbal treatment for malaria. The species is popular in rural and urban areas due to its lower price compared to commercial pharmaceuticals and because of its long-standing spiritual and symbolic significance. Consequently, W. salutaris is threatened in Malawi, South Africa and Swaziland and is believed to be extincted in the wild in Zimbabwe. In Mozambique, where the tree is confined to the southern part of the country, it is considered vulnerable due to high local demand and cross-border export. Although there is limited research, further work is urgently needed to provide the basis for options and strategies to achieve desired outcomes. This study aimed to explore different approaches for the conservation of W. salutaris in the Lebombo Mountains (LM), the Tembe River (TR) and the Futi Corridor (FC) areas, in southern Mozambique. For this, potential distribution, socio cultural factors that influenced sustainable management of the species, demography, harvest impact, genetic diversity and population differentiation were considered. The kuenm package in R, 2 846 occurrence and 11 environmental data were used to model the potential current and future distribution in 2050 and 2070 using four general circulation models (CESM1(BGC), ACCESS1.3, FIO-ESM and IPSL-CM5A-MR) under representative concentration pathways (RCPs) 4.5 and 8.5. The extent of occurrence (EOO) and habitat suitability (HS) were also determined. Stratified random semi-structured interviews with 182 heads of households were carried out to explore cultural and socio incentives of use, compared knowledge distribution, determine local management practices and explore local ecological knowledge related to the species in the three study areas. In addition, 17 focus group discussion were conducted to clarify issues raised during household interviews. One hundred and twenty four quadrants were used to determine the abundance and population structure. The Simpson index of dominance, permutation index and quotient between consecutive classes were used to assess population stability, and an eight point scale was used to evaluate the extent of bark damages. Genomic deoxyribonucleic acid (gDNA) (n=48) was used to assessed genetic diversity and population structure through the newly developed single sequence repeats (SSR) molecular markers. Model selected was statistical significant for prediction of potential distribution of W. salutaris in southern Mozambique (partial ROC =0.00, omission rate = 0.04 and delta AICc = 0.00). The current environmental conditions mostly explained by climatic predictor, indicated in general, the potential distribution on areas of occurrence observed during the field work in southern Mozambique. Future project suggested an increase of climatic suitable areas in all the general circulation models in both conservative and pessimistic climatic scenarios (RCPs 4.5 and 8.5, respectively) in 2050 and 2070, and consequently the increase of HS and the EOO. Results of this work suggests W. salutaris would not extinct at least in the coming 50 years due to climate change but other threats such as overexploitation of the species need to be addressed. With increase of climatic suitable areas, conservation and management of this species could focus on reintroduction and cultivation for recovery of the species, with the help of time. W. salutaris was used for medicinal purposes to treat 12 health problems, with the bark being the most commonly used part (92% of informants). Knowledge of the species uses differed significantly between the three study areas but not with respondent gender or age. Informants also mentioned a considerable number of alternative species of W. salutaris (37 species in LM, 33 species in TR and 17 species in FC). Local ecological knowledge on the ecological niche of the species also varied substantial in the three areas with both types of knowledge generally higher in the Tembe River area. Local management practises were revealed mostly on harvesting through vertical bark stripping cited by 100% of informants in TR, 61.6% FC and 51.7 LM (71% of all informants). Yet most (64.3% of all informants, corresponding to 98.4% in TR, 51.7% in Fc and 41.7% LM) used non-traditional harvesting tools (machete) bark removal. For sustainable use of the species, consideration should be given to combined conservation strategies such as the use of alternative species with the same application, substitution of bark by leaves, and increases in alternative sources of plant material through cultivation. The densities of alive stems were not statistically different in the three areas but was higher in LM (mean of 4500.3±9209.8 stems/ha). Equally the mean densities of recruits (diameter ≤2 cm) were not considerable different but was higher in the FC (3296.8±9116.7 stems/ha). The regression analyses for the three study area revealed negative slopes, indicating more recruits and fewer individuals in larger classes. The Simpson indexes of dominance, permutation indexes and quotients between the consecutive size classes suggested some levels of population instability in the three areas. Harvesting took place in the eight point scale, all size classes were harvested but preference was higher on intermediate (15.1-20 cm) or larger (>20 cm) size classes in all the study areas but gatherers avoided harvesting bark from recruits. Although the mean densities of harvested stem was not significant this work indicated higher levels of stem destruction in FC. Harvested dead stems ware significant in the three areas and FC had the higher of (39.5±85.9 stems/ha) compared to 7.6±19.1 and 2.1±4.3 in TR and LM, respectively. Equally, the density of physiologically stressed stems differed substantially in the three areas with higher in FC 20.5± 75.5 stems/ha compared to 18.7±82.9 stems/ha in TR and 3.8±11.8 stems/ha in LM. This results suggests implementation of protection measures such as the inclusion of the species in the national CITES list to regulated cross-border trade and strengthening of the law enforcement. The dimension of the spot obtained under these conditions was ≈ 75 μm. The electrochemical imaging by SECM of the ultra thin area locally clicked with ferrocene moieties was made thanks to the electrocatalytic properties of the ferrocene modified surface towards ferrocyanide electrooxidation. This local clicking procedure opens the gate to further controlled functionalization of restricted small substrates.

With over 60% of women in Sub–Saharan Africa involved in agriculture, there is need to explore and assess the role they play in promoting agribusiness with special focus on Kariega, in South Africa. The identified participants in the study, as self-employed farmers, participate in agribusiness, thereby creating employment opportunities whilst generating personal incomes. Several benefits to female involvement in agribusiness were identified, including the ability to sustain families, stable cash flow, and empowerment. Several challenges were identified, including adequate capital, inaccessibility to private land, droughts, water scarcity, climate change effects and stigmatisation. Through a feminist qualitative methodology, the study utilised semi- structured interviews to collect data from 10 women involved in agribusiness around Kariega, Nelson Mandela Bay. Themes from the findings are analysed to highlight the important role played by women toward agribusiness, their achievements, government and private sector role and the hinderances still faced by these women in agricultural cooperatives. Discussion from the analysis included a framework of recommendations, for policy makers, development practitioners and future researchers. The study found that all stakeholders involved in the agricultural sector should work together for sustainable agricultural development and sustainable development goals to be achieved. Methodology: This study applied the qualitative research approach and used a combination of descriptive and exploratory research design. Three clinic managers and nine professional nurses participated in the in-depth interviews. Thirteen general healthcare practitioners responsible for adolescents, sexual and reproductive health participated in key-informant interviews. Data was analysed using thematic analysis. Results: The study found that although healthcare practitioners are aware of the amendment act. However, there was inadequate understanding of the precise implications of this statutory initiative. Many of the participants were not informed of the intent of this act, whilst others suggested that they were not well trained regarding this act due to lack of funding in their respective clinics. Mixed views and perceptions on the impact of this act on their duties, on minors and on the general population were identified. Some were of the view that the Act will make it difficult to control the children while other welcomed it, because it leads to less teenage pregnancies, street abortions, and other similar experiences. Conclusion: In order to overcome these differences in perceptions, the Department of Health must create awareness among staff of the Act and its purpose. The study findings indicated that while women are part LED activities in the Makana municipality, the benefits that accrue to them are still minimal and trickling down to very few of them because of egoism and nepotism, corruption and crime, favouritism and poor communication. There is a great need for interventions requiring regular visits from LED officials and provision of educational programs in the local language, to enable the women to have a clear understanding of the benefits that accrue to them from LED projects, notwithstanding the opportunities open to them for participation. - Description: The aim of the study was to explore the role of Local economic development (LED) in empowering rural women in Makana Municipally. The researcher intended to achieve the following objectives (i) to assess the role of LED among the rural women of Makana Municipality in strengthening economic development ii) to explore the LED challenges confronted by the community and the municipality in the implementation of economic empowerment programs that will create economic opportunities for rural women iii) to identify the way forward in enhancing the LED strategy to empower rural women to enable them participate in economic programs of the local Municipality that will benefit them and the community. The study adopted a qualitative approach and purposive sampling methods which were used to select samples for the study. Open ended questions were used in face to face interviews and focus groups to collect data in Makana Municipality. The study findings indicated that while women are part LED activities in the Makana municipality, the benefits that accrue to them are still minimal and trickling down to very few of them because of egoism and nepotism, corruption and crime, favouritism and poor communication.
