- Description: The study reported in this thesis investigated Emirati students’ claims related to experiences of cultural bias of the reading component of the IELTS examination through a critical realist lens. Critical realism posits a layered reality which allows for the conceptualization of experiences as emerging from the interplay of events and mechanisms found in two other realms of reality – the actual and the real. Experiences, therefore, have a different ontological status than the events and the causal mechanisms to which they are attributed. Social realism was used to further explore the depth of the realm of the real through Archer’s construct of analytical dualism. This allowed for the placement of generative mechanisms into three domains: structure, culture, and agency. There were two parts to this investigation: a content analysis and a focus group study. The first part of the content analysis consisted of analyzing 60 reading passages from 20 IELTS examinations for a number of cultural categories. These included such things as cultural objects, social roles, idiomatic expressions, traditions and festivals, superstitions and beliefs, and political and historical settings. The second part of the content analysis focused on the question types and syntactical structure of the 5 different IELTS examinations that the focus groups students sat. All three components of the analysis – the cultural content, question types, and syntactical structure – were conducted at the level of the actual. Findings indicated that on average, an IELTS examination contained 14 cultural references of various kinds. Only 4% of all geographical references pertained to the Middle East with the biggest share being western locations. The most common question types were matching questions, fill in the blank questions, and yes/no questions with more than 50% of all questions requiring some form of higher order thinking due to text reinterpretation. The study also found that the question types were not consistently distributed over the examinations with each consisting of a different variety of questions and some even having repetitive question types on one reading examination. The second part of the study was the focus groups. Here, 21 Emirati students sat 5 different IELTS examinations. Upon test completion, these students underwent a semistructured interview to relate their experiences of the test. These experiences, at the level of the empirical, all shared 7 ideas: reading is hard, the questions are too difficult, the passages are too long and difficult, the topics are unfamiliar, the topics are not interesting, the vocabulary is too difficult, and there is not enough time. When the processes of retroduction and abduction were applied to both the content analysis and these common experiences, numerous structures and discourses at the level of the real were identified as having contributed to the emergence of the feeling of bias at the level of the empirical. These structures included such things as the students’ school system (eg. curriculum, assessment, instructors etc.), religion, literacy practices, and home. In the cultural domain, a number of discourses were found to contribute to the experiences at the level of the empirical. Amongst these were the ‘Unimportance of Reading’, the culture of ‘Obedience’, the rejection of the ‘un-Islamic’, and the students’ sense of ‘Entitlement’.

Shembe religion's integration of African traditional religion and Christianity : a sociological case study

This thesis documents the Shembe Church's ideas and practices; it discusses how the Shembe Church combines two ideologies that appear to be at odds with each other. In looking at Shembe ideas and practices, the thesis discusses African religion-inspired rituals like ukusina, ancestral honouring, animal sacrificing and virgin testing. The thesis also discusses the heavy Christian influence within the Shembe Church; this is done by looking at the Shembe Church's use of The Bible and Moses' Laws which play a crucial role in the Church. The challenges the Shembe Church faces are another main theme of the thesis. The thesis looks at cases of intolerance and human rights violations experienced by Shembe members. This is done in part by looking at the living conditions at eBuhleni, located at Inanda, KZN. The thesis also analyses individual Shembe member's experiences and discusses how some members of the Shembe church experience the acceptance of the Shembe religion in South African society. This thesis concludes by trying to make a distinction between intolerance and controversy. I try to highlight the idea that what many Shembe followers see as discrimination and intolerance towards them is sometimes a difference in opinion from other cultural groups. Sometimes these differences are not geared towards criticising other religious groups or perpetuating intolerance.

Investigating the English vocabulary needs, exposure, and knowledge of isiXhosa speaking learners for transition from learning to read in the Foundation Phase to reading to learn in the Intermediate Phase : a case study

- Description: The nature of learners' transition from Grade 3 to Grade 4 has serious and far reaching ramifications for their educational success in the subsequent grades. This is true of the South African education system, as it is for several other education contexts. The challenge that complicates the transition is largely a lexical one. It is with that understanding that the present study aimed to do three things namely: to determine the vocabulary needs of Grade 3 learners for transitioning to Grade 4; to establish the extent to which Grade 3 isiXhosa speaking learners are exposed to, and prepared for, the acquisition of that requisite vocabulary; and to establish the extent of Grade 4 learners' knowledge of that vocabulary at the beginning of the year. The vocabulary needs of Grade 3 learners for the transition to Grade 4 were determined from the Grade 4 subject textbook corpus. Word frequency was the criterion used to determine the usefulness and critical importance of a word for the transition. The AntConc concordance software program was used to generate word frequencies. Words with high frequency across the different subject areas, which were confirmed as high frequent in four other known word lists, were considered as constituting the vocabulary needs of learners at the verge of this significant transition. The extent of learners' preparedness for transitioning to Grade 4 in terms of their vocabulary exposure and vocabulary instruction was determined through teacher interviews, classroom observations, analysis of teacher classroom talk, analysis of the exposure and recycling of high frequent vocabulary in Grade 3 reading materials and classroom print. From these diverse sources, findings point to a paucity in both the exposure and recycling of the requisite vocabulary in these sources of classroom language input. Classroom observations and teacher interviews attest to lack of deliberate vocabulary instruction in the Grade 3 English First Additional Language lessons. The only extensive coverage of the requisite vocabulary was in the Grade 3 reading materials which included Big books, Readers and Workbooks. The Grade 4 learners' knowledge of the 60 high frequency words was tested through nine vocabulary tests, three of which tested their knowledge of word recognition, three tested passive word knowledge and the remaining three tested learners' active word knowledge. All the Grade 4 learners in the ten participating schools (297) were tested. Performance in the tests indicated that Grade 4 learners' knowledge of words requisite for reading to learn was low. That observation was consistent with an analysis of learners' performance per school, per district, per word, per test and per word bands. Tests of word recognition were done better than those of passive word knowledge and active word knowledge. Particularly problematic was test 4 which tested learners' knowledge of definition of words.

- Description: From Integrative Summary: There has been a growing interest in successful entrepreneurs. Research on entrepreneurship has focused on the identification of personality variables that would assist in the prediction of entrepreneurial success. The present study moves away from attempting to predict entrepreneurial success and instead focuses on exploring and describing the personality of a successful entrepreneur. A psychobiographical case study was adopted by the researcher to explore and describe the extent to which Steve Jobs demonstrated the entrepreneurial characteristics identified by Rauch and Frese (2007). A personality trait approach to entrepreneurship was adopted in the study. The study also attempted to explore the socio-cultural and economic context within which Jobs practised his entrepreneurial activities. Jobs was a successful entrepreneur who co-founded Apple and founded NeXT and Pixar, which were all companies that transformed various technological industries. His entrepreneurial orientation allowed him to produce innovative products that transformed society in various sectors which included personal computing, mobile phones, music, retail stores and films (Isaacson, 2011). A qualitative approach was adopted in the study. The data collection and analysis was guided by the three linked sub-processes proposed by Miles and Huberman (2002) which involved data reduction, data display and conclusion drawing and verification. The findings of this study showed that Jobs demonstrated, at varying levels, all the entrepreneurial characteristics identified by Rauch and Frese (2007) which included a need of achievement, risk-taking, innovativeness, autonomy, locus of control and self-efficacy. The researcher however noted that the desire for autonomy, risk-taking, innovativeness and self-efficacy were the most dominant characteristics driving his entrepreneurial orientation. The research thesis adopted the structure of a teaching case which can be used to explore and discuss the personality trait perspective to entrepreneurship in a classroom setting. The findings of the study can be recognised as positively contributing to the growing field of psychobiographical research on exceptional individuals, including entrepreneurs. The study can be considered as a foundation for future studies which will add to the body of knowledge relating to entrepreneurship and personality.

Much of this research and practice is influenced by constructivism, which has a relativist stance on knowledge, generally arguing, contra positivism, that knowledge is constructed in socio-historical contexts and largely inseparable from those who construct it and from issues of power. This leads to a confusion of knowledge with knowing, and knowledge is thus obscured as an object of study because it is only seen or understood as knowing or as a subject of learning and teaching. This ‘knowledge-blindness’ (Maton 2013a: 4) is problematic in higher education because knowledge and knowing are two separate parts of educational fields, and while they need to be brought together to provide a whole account of these fields, they also need to be analysed and understood separately to avoid blurring necessary boundaries and to avoid confusing knowledge itself with how it can be known. Being able to see and analyse knowledge as an object with its own properties and powers is crucial for both epistemological access and social inclusion and justice, because knowledge and knowledge practices are at the heart of academic disciplines in universities. Social realism offers an alternative to the dilemma brought about by constructivism’s tendency towards knowledge-blindness. Social realism argues that it is possible to see and analyse both actors within social fields of practice as well as knowledge as something that is produced by these actors but also about more than just these actors and their practices; thus knowledge can be understood as emergent from these practices and fields but not reducible to them (Maton & Moore 2010). Social realism, drawing from Roy Bhaskar’s critical realist philosophy (1975, 2008), is intent on looking at the real structures and mechanisms that lie beneath appearances and practices in order to understand the ways in which these practices are shaped, and change over time. Legitimation Code Theory is a realist conceptual framework that has, as its central aim, the uncovering and analysis of organising principles that shape and change intellectual and education fields of production and reproduction of knowledge. In other words, the conceptual tools Legitimation Code Theory offers can enable an analysis of both knowledge and knowers within relational social fields of practice by enabling the analysis of the ways in which these fields, such as academic disciplines, are organised and how knowledge and knowing are understood in educational practice. This study draws on social realism more broadly and Legitimation Code Theory specifically to develop a relatively novel conceptual and explanatory framework within which to analyse and answer its central question regarding how to enable cumulative knowledge building through pedagogic practice. Using qualitative data from two academic disciplines, Law and Political Science, which was analysed using a set of conceptual and analytical tools drawn from Legitimation Code Theory, this study shows that the more nuanced and layered accounts of pedagogy that have been generated are able to provide valuable insights into what lecturers are doing as they teach in terms of helping students to acquire, use and produce disciplinary and ‘powerful’ knowledge (Young 2008b). Further, the study demonstrates that the organising principles underlying academic disciplines have a profound effect on how the role of the knower and the place or purpose of knowledge is understood in pedagogy and this affects how the pedagogy is designed and enacted. This study has argued that if we can research pedagogy rigorously using tools that allow us to see the real mechanisms and principles influencing and shaping it, and if we can reclaim the role of disciplinary knowledge as a central part of the pedagogic relationship between lecturer and students, then we can begin to see how teaching both enables and constrains cumulative learning. Further, we can change pedagogy to better enable cumulative learning and greater epistemological access to disciplinary knowledge and related practices for greater numbers of students. The study concludes by suggesting that the conceptual tools offered by Legitimation Code Theory can provide academic lecturers with a set of tools that can begin to enable them to 'see' and understand their own teaching more clearly, as well as the possible gaps between what they are teaching and what their students are learning. This study argues that a social realist approach to the study of pedagogy such as the one used here can begin not only to enable changes in pedagogy aimed at filling these gaps but also begin to provide a more rigorous theoretical and practical approach to analysing, understanding and enacting pedagogic practice. This, in turn, can lead to more socially just and inclusive student learning and epistemic and social access to the powerful knowledge and ways of knowing in their disciplines.

Social realism, drawing from Roy Bhaskar’s critical realist philosophy (1975, 2008), is intent on looking at the real structures and mechanisms that lie beneath appearances and practices in order to understand the ways in which these practices are shaped, and change over time. Legitimation Code Theory is a realist conceptual framework that has, as its central aim, the uncovering and analysis of organising principles that shape and change intellectual and education fields of production and reproduction of knowledge. In other words, the conceptual tools Legitimation Code Theory offers can enable an analysis of both knowledge and knowers within relational social fields of practice by enabling the analysis of the ways in which these fields, such as academic disciplines, are organised and how knowledge and knowing are understood in educational practice. This study draws on social realism more broadly and Legitimation Code Theory specifically to develop a relatively novel conceptual and explanatory framework within which to analyse and answer its central question regarding how to enable cumulative knowledge building through pedagogic practice. Using qualitative data from two academic disciplines, Law and Political Science, which was analysed using a set of conceptual and analytical tools drawn from Legitimation Code Theory, this study shows that the more nuanced and layered accounts of pedagogy that have been generated are able to provide valuable insights into what lecturers are doing as they teach in terms of helping students to acquire, use and produce disciplinary and ‘powerful’ knowledge (Young 2008b). Further, the study demonstrates that the organising principles underlying academic disciplines have a profound effect on how the role of the knower and the place or purpose of knowledge is understood in pedagogy and this affects how the pedagogy is designed and enacted. This study has argued that if we can research pedagogy rigorously using tools that allow us to see the real mechanisms and principles influencing and shaping it, and if we can reclaim the role of disciplinary knowledge as a central part of the pedagogic relationship between lecturer and students, then we can begin to see how teaching both enables and constrains cumulative learning. Further, we can change pedagogy to better enable cumulative learning and greater epistemological access to disciplinary knowledge and related practices for greater numbers of students. The study concludes by suggesting that the conceptual tools offered by Legitimation Code Theory can provide academic lecturers with a set of tools that can begin to enable them to 'see' and understand their own teaching more clearly, as well as the possible gaps between what they are teaching and what their students are learning. This study argues that a social realist approach to the study of pedagogy such as the one used here can begin not only to enable changes in pedagogy aimed at filling these gaps but also begin to provide a more rigorous theoretical and practical approach to analysing, understanding and enacting pedagogic practice. This, in turn, can lead to more socially just and inclusive student learning and epistemic and social access to the powerful knowledge and ways of knowing in their disciplines.

This study explores situated language practices within two small multilingual businesses. The first is a bicycle rental and repair shop located in Amsterdam (the Netherlands), which is a city with a dynamic multilingual society. The second is a tyre fitment centre in Grahamstown (South Africa), which is a city characterised by a stable triglossia of English, Xhosa and Afrikaans. Using Linguistic Ethnography (Rampton 2007) as my data collection method, I spent a total of eight weeks in these businesses. For the analysis I draw on Spencer-Oatey’s (2000b; 2011) Rapport Management Framework and sociolinguistics of globalisation (Blommaert 2010). However, difficulties emerged during the analysis with applying this concept to some of the more elaborate and complex data. As a result my argument developed into two different strands. The first demonstrates how individuals turn to their multilingual repertoires to negotiate agency and power relationships in small business discourse. The analysis reveals that people at times deliberately promote and maintains discordant relations, which can be understood as a rational response to the individual’s social and economic context. The second discusses the problems that emerged during my analysis with applying rapport management orientations to my data. I propose theoretical developments, warranted by my data, to create an Enhanced Rapport Management Framework suitable for the analysis of complex small business discourse.

Studies showed that the type of substituent attached to the MPc ring influenced the overall sensitivity of the probe. Additionally, a comparative investigation using newly synthesized phthalocyanine and triaza-benzcorrole complexes was conducted when these complexes were conjugated to CdSe@ZnS QDs for analyte sensing. Results showed that the triaza-benzcorrole complex can be employed as a host-molecule sensor but displayed a lower sensitivity for analyte sensing in comparison to the phthalocyanine complex.

- Description: The increase in teenage pregnancy among school going learners is reported in the media as a crisis. Politicians and other stakeholders have also raised their views and concerns about pregnancy. In particular, these views and concerns perceive teenage pregnancy among school going learners as a cancer that needs a remedy because it has negative consequences for the learners, in particular the girl child. However, for all the sense of public crisis concerning sexuality and schooling, the voices of young people themselves regarding their own sexual subjectivity are seldom heard. This study focused on how girls in a former model C all girls high school negotiate and make sense of the meaning of the messages that they receive from the formal curriculum. The concept of student sexual cultures was employed in this study. Student sexual cultures are the informal groups that exist in the school and the girls take part in it. It is in these groups that the girls learn about sexuality and also make sense of their own gendered identities. This study employed ethnographic techniques of classroom observation coupled with in-depth interviews, focus groups and solicited narratives in order to understand how the participants experience and "take up" the messages they receive in the formal sexuality education component of the Life Orientation (LO) curriculum. The data was collected over a period of three months and was analysed using a directed content analysis. Four dominant themes emerged from the study. Firstly, the data reveals the school is a space of competing and conflicting discourses of sexuality and the learners are involved in a constant negotiation of the meanings of the messages. Secondly, the data shows the contested and confirmations of learners subjectivity. It shows that learners are regarded as sexual beings both in the formal and informal school cultures but there are limitations around one's sexual subjectivities. Thirdly, the data reveals that the school is a site in which a variety of femininities are reproduced, contested and struggled over. Femininities are constructed in the complex context of the school thus the school emerges as a site in which multiple femininities intersect with class, race and sexuality. Lastly, this study argues for the incorporation of the discourse of erotics in the formal curriculum which allows young people's voices to be heard. This approach (discourse of erotics) can be seen as a process of becoming, which focuses on possibilities of improving sexuality education as opposed to an imposed sexual model that is applied to young people and assumed to be the solution to young people's sexuality.

Located within the broad theoretical framework of cultural studies and drawing on audience reception theories, the study focuses on the ways in which Shona-speaking women bring their understandings of their social roles, derived from their lived socio-cultural experiences of patriarchy, to their decoding of the text. The study was set in Harare’s high-density suburb of Mbare and used the qualitative research methods of individual and focus group interviews. The study was conducted against the backdrop of the signing of the Global Political Agreement (GPA) of September 2008, which ended the impasse among the warring political parties, ZANU PF, MDC-T and MDC and introduced a new era of collectively tackling socio-economic development, including redressing gender disparities through women’s empowerment. This study examines the factors shaping the audiences’ readings of the programme and seeks to establish whether the mass media has determining power on its audience in the reception of messages or if the audiences (women) have interpretive freedom. Using Hall’s (1980) Encoding/ Decoding model, the study examines the factors that influence the audiences’ choice in making preferred, negotiated or oppositional readings and the arguments they advance in line with those readings. While the interviews revealed that most of the female listeners “negotiated” the dominant encoded meanings, seeking their relevance to their varied situations and contexts (O’ Sullivan et al. 1994:152; Ang 1990: 159), of interest is the manner in which the women dealt with the discourse of patriarchy within the context of promoting women empowerment. The contestation between women empowerment and addressing patriarchy reflected the subverted notions of maintaining the status quo, while applauding the women’s commitment and ability to interrogate the practicality of issues under discussion and drawing lessons relevant to their day to day lives prior to making the preferred reading. As such, the study revealed that preferred readings are not always automated, but can be a result of intense interrogation among media audiences.

Teachers and those who aspire to take up teaching are expected to conduct themselves and discharge their duties in an impeccably professional manner. However, there is concern especially among education authorities regarding unprofessional conduct of some of those employed in schools and colleges. This action research study is a response to this concern. Its goal was to explore issues of professional conduct as part of the induction process of new members entering the teaching profession, and to thereby develop a better understanding of how best to raise student teachers’ awareness around professional conduct issues. The study used a qualitative research framework located in the interpretive paradigm. Three theoretical frameworks informed the design and subsequent analysis of the findings, namely, Burn’s transformational leadership, Mezirow’s transformative learning and Kolb’s theory of experiential learning. Two cycles of workshops around issues of professional conduct were conducted over a period of four weeks with a sample of 40 pre-service student teachers: final year students enrolled in the college’s three year Diploma in Education programme, all members of a science education class. Data collection strategies used were semi-structured interviews, observation and the use of reflective journals, among others. Analysis of the data involved identification of emerging themes and patterns. Initial findings indicate that prior to the commencement of the cycles of action research, participants appeared to have a limited understanding around issues of professional conduct, but that this changed as they participated in the workshops. The data of the study suggest that further steps need to be taken to establish optimal ways of incorporating professional conduct issues into the college’s teaching curriculum.

- Description: In May 2012, a meeting was held between various stakeholder representatives of the Western Baviaanskloof to discuss the concept of a proposed landowner-enterprise. This concept was put forward in response to a need for collaboration in the midst of economic, environmental and social issues at play in the Baviaanskloof. Owing to the conservation value and rapidly decreasing economic output of the land, a radical shift to sustainable land-use practices was called for by various stakeholders. Suggested as a vehicle to generate income for the local landowners through alternative sustainable land-uses, the proposed enterprise may aid in addressing this shift by use of a bottom-up approach. During the stakeholder meeting, it was requested by the representative landowners that a feasibility assessment be conducted on the concept of the proposed enterprise prior to establishment. As an integral part of this assessment, the researcher took on the task of investigating stakeholder reception to the enterprise by means of a stakeholder analysis. It was decided to limit this to three markets: water, carbon and tourism. The purpose of this research study is twofold, namely to: investigate stakeholder influence and their reception of the proposed enterprise using a stakeholder analysis; and also to identify and advise on the opportunities and constraints relating to stakeholders, thus contributing to determining the feasibility of the proposed enterprise. In achieving the purpose of this study, a systematic stakeholder analysis framework was constructed, based on existing theory. This was necessary because, although stakeholder analysis is commonly practiced, no study was found to provide a theoretically based framework for the purpose of feasibility in the initial stages of enterprise establishment. Thus the contribution of the study is also twofold, namely the practical outcome of determining stakeholder reception for feasibility, and a secondary outcome of constructing a stakeholder analysis framework. The stakeholder analysis framework is based on an interpretation of existing stakeholder theory, with the addition of four "relational indicators" – goals, intentions, relationships, and resources. In applying the stakeholder analysis framework, ECPTA was categorised as the definitive (most influential) stakeholder to the enterprise, and DEDEAT, SANBI (through Working for Wetlands), Baviaans Tourism, GIB, and NMBM were categorised as pivotal (influential and active). In discerning stakeholder interest in the proposed enterprise, a number of emerging themes were found to affect the projected interest and behaviour of stakeholders, apart from their specified goals. Emerging themes included: tunnel visioning, internal disparity, individual/personality clashes, and misaligned interests. In addition to this, in interpreting stakeholder interest, specific intentions or agendas that might affect the interest shown towards the proposed enterprise were also taken into account. Five predominant intentions of stakeholders were identified as: implementing a stewardship programme, establishing a tourism association, establishing a water users' association, social development, and "the big vision". Findings on the final objective revealed a number of perceived opportunities and constraints relative to the feasibility of the enterprise. Three prime opportunities were identified as: partnerships with definitive and pivotal stakeholders, the possibility of tendering for implementer of the "Working for" programmes, and taking on the role of Tourism Association. The following potential constraints were also emphasised by participants: social aspects such as individuals and personalities, the incompatibility or non-existence of local market structures, and the need for external funding. With regard to stakeholder reception, most of the stakeholders, with the exception of NMBM and Saaimanshoek, responded positively to the idea of the enterprise. Overall, based on participant perceptions, the tourism market was found to be the most feasible the carbon market uncertain and a long-term possibility, and the water market the least feasible.

Beyond black and white: black solidarity in post-apartheid South Africa

- Description: Almost 20 years after the white Nationalist government was voted out, some black South Africans believe that black solidarity is still necessary in South Africa. These people argue that since post-apartheid South Africa is still marred with racial injustice, it makes sense for blacks to advocate for black solidarity. Although it is true that black solidarity played an important role in the struggle against apartheid, in this thesis I argue that the struggle against current forms of racial injustice does not necessarily require black solidarity. This is not to deny the prevailing racialized oppression in the post-apartheid era, nor to deny the importance of black solidarity in the past; rather the point I am making is that the current form of racial oppression is somewhat different from the one before 1994. Hence I argue in this thesis that the current form of racial oppression requires us to do certain things differently. Doing things differently means improving upon the strategies of the past. For this to happen, I argue that every human being who believes in and is committed to racial justice ought to be included in the struggle for justice. Change, after all, is brought about when committed human beings work together for liberation and justice.

This process is taking place with the sustained support of global agencies in terms of development cooperation, assistance and aid. Insofar as an endogenous development path which speaks to national priorities can be formulated and implemented by post-colonial states, their relationship with these agencies raises serious questions about such a path if the relationship entails dependence and indeed subordination of post-colonial states. This raises important questions about state autonomy under post-colonial conditions and the possibility of autonomy being compromised. In this light, the thesis examines European Union budget support to Mozambique and, in particular, the relationship between EU budget support and Mozambique state autonomy in pursuing national development. This is particularly pertinent given the massive dependence of the Mozambican state on foreign funding (notably EU funding) with specific regard to the national budget. Despite the broad claims existing in the prevailing literature that nation-state autonomy is seriously undermined in and through the international development system, the thesis argues against reductionist arguments that simply posit post-colonial states as mere instruments of global forces. This system, including European Union budget support, does indeed set the conditions of existence for post-colonial states such as Mozambique. But autonomy is necessarily relative and is subject to different forms and degrees. Ultimately, it is through empirical investigation that the specific form and degree of autonomy can be pinpointed and understood. The thesis contributes to this endeavour and suggests that the relationship between European budget support and Mozambican state autonomy is more complex and tension-riddled than the prevailing literature would seem to suggest.

The effects of extracellular and intracellular Hop on cell migration processes

- Description: The Hsp70/Hsp90-organising protein (Hop) is a 60 kDa co-chaperone that acts as an adaptor molecule, facilitating the transfer of client proteins between the Hsp70 and Hsp90 chaperone systems. Hop functions both intracellularly and extracellularly and has been implicated in many processes involved in cancer progression, including cell migration and invasion. Little is known about the mechanisms or domains by which extracellular Hop functions. In addition, little is known about the effects of Hop on signalling molecules involved in cell migration and invasion through regulation of actin dynamics. It was hypothesised that both extracellular and intracellular pools of Hop would regulate distinct cell migration processes by activation of cell signalling pathways or direct interactions with signalling intermediates. HS578T cells were treated with recombinant full length and truncated murine Hop proteins (overexpressed and purified in this study) to determine the effects of extracellular Hop and the independent domains on cell migration processes. Additionally, RNA interference (RNAi) techniques were used to determine the effect of Hop knockdown on cell migration related signalling intermediates and cell morphologies. A short hairpin RNA (shRNA) system for the stable knockdown of Hop was developed and used for a number of these studies. Treatment of HS578T cells with the TPR2A2B and TPR1 domains of Hop resulted in a significant decrease in cell migration and caused changes in the actin cytoskeleton and extracellular matrix proteins, gelatin and fibronectin. RhoC immunoprecipitated in a common complex with Hop and Hsp90. Hop knockdown reduced levels of actin and total RhoC, as well as active RhoC. In addition, knockdown of Hop resulted in a reduced migratory phenotype. We interpreted these data to indicate that intracellular Hop played a role in cell migration through regulation of RhoC activity, either through a direct interaction between Hop and RhoC, or an indirect interaction of RhoC with the Hsp90 multichaperone heterocomplex. Taken together, the data suggested that extracellular and intracellular Hop played distinct roles in extracellular and intracellular processes that lead to actin dynamics and cell migration. - Description: Renewable energy has become an important feature of most modern economies with clean and non-exhaustible sources of power being given a greater significance. Wind energy is one of the favoured renewable, as it is (2013) generally the cheapest and most mature technology available for commercial use. The South African government, as outlined by the Department of Energy's Integrated Resource Plan (IRP), aims to install 5 GW (Gigawatts) of wind energy by 2020. However, South Africa has had little experience in the wind energy industry which is limited to two projects, Klipheuwel (2002) and Darling (2008). Much effort has been dedicated to calculating balance sheet costs, which carries uncertainty due to the high reliance on country specific and site specific variables. An aspect which deserves more attention, and is often ignored, is the public‟s attitudes towards local wind farm developments, which have been known to "make or break" a project during the planning stages. Public backlashes have mostly been concerned with the visual "intrusiveness" of wind farms in the landscape. Detrimental effects on scenery, while seemingly innocuous, are acknowledged as being the single largest barrier to successful wind farm development. Individuals within an area become sentimentally attached to their surroundings, where significant rapid changes in the landscape are viewed as "damage". Economics recognises such declines in scenic resources as market failures, where an externalised cost is passed on to the public and is often not accounted for by private parties responsible for the liability. The primary objective therefore was to measure the magnitude of the visual impact, caused by the Waainek Wind Farm, on the Grahamstown community. Conventional NIMBY¹ (not in my back yard) reasoning, which seeks to explain local wind farm resistance, has attracted criticism with regard to its simplistic approach to wind farm problem identification. Contemporary arguments propose that NIMBY is a poor explanation for the trouble experienced on the local level because it groups problems into one all-encompassing term, leaving much of the discontent unexplained. Instead, the NIMBY explanation is really a broad set of unattended problems, largely resulting from the unsound practices present in the public participation process. Insufficient community involvement and disparities in the negotiation power structures have become the recent focus in wind farm literature. Essentially, these disparities force opposition groups to select factors which may seem more serious to developers, leading to ineffective remedial measures because the core underlying problems are not being remedied. Thus these considerations formed an additional area of investigation. No NIMBY effect was found for the Grahamstown community, as support for both the local and general level was roughly 80%. The public participation process on the other hand revealed that while many found the practices of the developer to be unfair, attitudes towards the wind farm were not adversely affected, especially for the lower income Grahamstown East areas. While the public participation process in this instance did not have any effect on people’s attitudes, careful inspection of the circumstances need to be given. Wind farms are new to South Africa, where the novelty and benefits are the focus of enthusiasm. Job opportunities as well as clean energy are positive drivers for attitudes; however given time, once the anticipation for wind farms dulls, real problems may be revealed. Thus it is crucial to implement good practice procedures during the public participation process, especially when national adoption rates of wind energy are low. Early implementation of an effective public participation process system will ensure that when major problems do arise in future projects, experience and institutional processes would have had ample opportunity to evolve appropriately over a period of time. The double bounded Contingent Valuation Method was used to value the impact of the wind farm on the Waainek scenery through a hypothetical scenario based procedure which presented pictures of the landscape before and after the wind farm had been installed. Based on the perceived impact of the wind farm, respondents were asked their Willingness to Pay to relocate the development, based solely on visual impacts. Learning design Contingent Valuation (Bateman et al., 2008) is a novel technique employed to familiarize respondents with the hypothetical market institution as well as the scenic goods being valued. - Description: Renewable energy has become an important feature of most modern economies with clean and non-exhaustible sources of power being given a greater significance. Wind energy is one of the favoured renewable, as it is (2013) generally the cheapest and most mature technology available for commercial use. The South African government, as outlined by the Department of Energy's Integrated Resource Plan (IRP), aims to install 5 GW (Gigawatts) of wind energy by 2020. However, South Africa has had little experience in the wind energy industry which is limited to two projects, Klipheuwel (2002) and Darling (2008). Much effort has been dedicated to calculating balance sheet costs, which carries uncertainty due to the high reliance on country specific and site specific variables. An aspect which deserves more attention, and is often ignored, is the public‟s attitudes towards local wind farm developments, which have been known to "make or break" a project during the planning stages. Public backlashes have mostly been concerned with the visual "intrusiveness" of wind farms in the landscape. Detrimental effects on scenery, while seemingly innocuous, are acknowledged as being the single largest barrier to successful wind farm development. Individuals within an area become sentimentally attached to their surroundings, where significant rapid changes in the landscape are viewed as "damage". Economics recognises such declines in scenic resources as market failures, where an externalised cost is passed on to the public and is often not accounted for by private parties responsible for the liability. The primary objective therefore was to measure the magnitude of the visual impact, caused by the Waainek Wind Farm, on the Grahamstown community. Conventional NIMBY¹ (not in my back yard) reasoning, which seeks to explain local wind farm resistance, has attracted criticism with regard to its simplistic approach to wind farm problem identification. Contemporary arguments propose that NIMBY is a poor explanation for the trouble experienced on the local level because it groups problems into one all-encompassing term, leaving much of the discontent unexplained. Instead, the NIMBY explanation is really a broad set of unattended problems, largely resulting from the unsound practices present in the public participation process. Insufficient community involvement and disparities in the negotiation power structures have become the recent focus in wind farm literature. Essentially, these disparities force opposition groups to select factors which may seem more serious to developers, leading to ineffective remedial measures because the core underlying problems are not being remedied. Thus these considerations formed an additional area of investigation. No NIMBY effect was found for the Grahamstown community, as support for both the local and general level was roughly 80%. The public participation process on the other hand revealed that while many found the practices of the developer to be unfair, attitudes towards the wind farm were not adversely affected, especially for the lower income Grahamstown East areas. While the public participation process in this instance did not have any effect on people’s attitudes, careful inspection of the circumstances need to be given. Wind farms are new to South Africa, where the novelty and benefits are the focus of enthusiasm. Job opportunities as well as clean energy are positive drivers for attitudes; however given time, once the anticipation for wind farms dulls, real problems may be revealed. Thus it is crucial to implement good practice procedures during the public participation process, especially when national adoption rates of wind energy are low. Early implementation of an effective public participation process system will ensure that when major problems do arise in future projects, experience and institutional processes would have had ample opportunity to evolve appropriately over a period of time. The double bounded Contingent Valuation Method was used to value the impact of the wind farm on the Waainek scenery through a hypothetical scenario based procedure which presented pictures of the landscape before and after the wind farm had been installed. Based on the perceived impact of the wind farm, respondents were asked their Willingness to Pay to relocate the development, based solely on visual impacts. Learning design Contingent Valuation (Bateman et al., 2008) is a novel technique employed to familiarize respondents with the hypothetical market institution as well as the scenic goods being valued. Both chlorpyrifos and tau-fluvalinate were effective in killing irradiated FCM for a number of days after application, after which degradation of the active ingredient rendered it harmless. This effect was found to be similar for irradiated and non-irradiated males, consequently ratios of sterile : wild male FCM should be retained regardless of whether sprays are applied or not. The modes for release of sterile FCM in an SIT programme were investigated. Efficacy of ground and aerial release platforms were tested by evaluating the recovery of released irradiated male FCM in these orchards. More irradiated FCM were recovered in orchards released from the ground compared to air. However, an economic analysis of both methods shows application of irradiated insects over a large geographical area is more cost-effective by air. Depending on the terrain and size of the target area, a combination of both methods is ideal for application of SIT for control of FCM in citrus. Development of application technology for advance of the programme is discussed and recommendations for future research and development are offered.

Refugium : an exploration through landscapes of liminality

- Description: Loosely following a narrative form and structure, this thesis begins, in chapter one, by considering people who occupy a peripheral position in - or on the fringe of - society. In so doing, it analyses the dichotomy created between centre and periphery, the line between qualities or features that are set up as different or opposed. The chapter begins by contextualising my own engagement with the themes and narratives dealt with. It then looks at the representation and significance of historical and cultural characters such as the tramp and the wanderer, the medieval wildman and the outcast, the madman and the prophet, and relates this to some extent to my own narrative. The dichotomy between culture and nature is thus examined here. The paper then moves on, in chapter two, to examine concerns around landscape, and considers how we engage or separate ourselves from it. The theme of walking is examined closely from an experiential, phenomenological standpoint, especially as an activity which allows greater connection to, and engagement with, the landscape. The relationship between landscape, place, and identity is also explored, looking at how identity is interwoven with connection to place, and, broadly, what this might mean for displaced people. These discussions and ideas are then brought to bear again on my personal narrative. In chapter three I explore again the separation between dichotomous concepts as looked at in chapter one; I examine the separations between, and within, people, as well as between people and their environment. Here, however, the line of separation is explored not as an impermeable barrier, but as a liminal space of potential and possibility. With this in mind I look at sacred or liminal places within the landscape, as well as the idea of the sublime. I also consider the camera from this perspective, and finally present my own exhibition and body of work, reviewing it in the light of the narratives and themes explored through the thesis.

This learning theory acknowledges learners’ background prior knowledge as valuable tools in the effective construction of science concepts during lessons. A corollary is that the socio-cultural circumstances of learners might negatively influence the way learners construct new knowledge in the science classroom if they are not taken into consideration. This study was thus prompted by the need to understand how trainee science teachers engage with prior everyday knowledge and experiences, so as to enhance the conceptual development of biological concepts. The study was qualitative and it was underpinned by an interpretive paradigm with some elements of action research. Science trainee teachers at Katima Mulilo, Unam Campus were used as research participants. The techniques used to gather data included document analysis, brainstorming, observation, audio-visual, microteaching and semi-structured interviews. The results of this study showed that the changes that occurred during microteaching practices of the four trainee teachers involved in the study reflected their professional development in this approach. However, it emerged that prior everyday knowledge can be both a barrier and enabler to the construction of meaningful teaching and learning; hence its oversight may lead to instructional failures. The findings also indicated that selected platforms are essential to enable trainee teachers to incorporate prior everyday knowledge and experiences into the teaching of Western science. However, larger scale study should be conducted in order to deepen the understanding of the topic.

The study focuses on Wazobia FM, a radio station in Lagos, and the first pidgin station in Nigeria. It seeks to determine whether the station’s audience engaged with the station’s programming based on its prioritisation of NigP and the linguistic identity it offers them. The study foregrounds the marginalised status of NigP within the politics of language in Nigeria. It traces the language’s evolution through popular and oppositional expressions in broadcasting and in music. It also seeks to establish the place of Pidgin English within the role that language plays in the formation of the Nigerian identity. This study thus adopts the ‘emic’ perspective, which underpins qualitative methodology, and views social life in terms of processes as opposed to static terms. The theoretical framework of this research revolves around culture, language and identity. Pertinent concepts in post-colonial studies, together with conceptual frameworks in Cultural Studies, such as popular culture, representation, hegemony and counter-culture have been used to make sense of the popularity of NigP radio stations.

A lignocellulolytic enzyme system for fruit waste degradation : commercial enzyme mixture synergy and bioreactor design

- Description: Studies into sources of alternative liquid transport fuel energy have identified agro-industrial wastes, which are lignocellulosic in nature, as a potential feedstock for biofuel production against the background of depleting nonrenewable fossil fuels. In South Africa, large quantities of apple and other fruit wastes, called pomace, are generated from fruit and juice industries. Apple pomace is a rich source of cellulose, pectin and hemicellulose, making it a potential target for utilisation as a lignocellulosic feedstock for biofuel and biorefinery chemical production. Lignocellulosic biomass is recalcitrant in nature and therefore its degradation requires the synergistic action of a number of enzymes such as cellulases, hemicellulases, pectinases and ligninases. Commercial enzyme cocktails, containing some of these enzymes, are available and can be used for apple pomace degradation. In this study, the degradation of apple pomace using commercial enzyme cocktails was investigated. The main focus was the optimisation of the release of sugar monomers that could potentially be used for biofuel and biorefinery chemical production. There is no or little information reported in literature on the enzymatic degradation of fruit waste using commercial enzyme mixtures. This study first focused on the characterisation of the substrate (apple pomace) and the commercial enzyme cocktails. Apple pomace was found to contain mainly glucose, galacturonic acid, arabinose, galactose, lignin and low amounts of xylose and fructose. Three commercial enzyme cocktails were initially selected: Biocip Membrane, Viscozyme L (from Aspergillus aculeatus) and Celluclast 1.5L (a Trichoderma reesei ATCC 26921 cellulase preparation). The selection of the enzymes was based on activities declared by the manufacturers, cost and local availability. The enzymes were screened based on their synergistic cooperation in the degradation of apple pomace and the main enzymes present in each cocktail. Viscozyme L and Celluclast 1.5L, in a 50:50 ratio, resulted in the best degree of synergy (1.6) compared to any other combination. The enzyme ratios were determined on Viscozyme L and Celluclast 1.5L based on the protein ratio. Enzyme activity was determined as glucose equivalents using the dinitrosalicylic acid (DNS) method. Sugar monomers were determined using Megazyme assay kits. There is limited information available on the enzymes present in the commercial enzyme cocktails. Therefore, the main enzymes present in Viscozyme L and Celluclast 1.5L were identified using different substrates, each targeted for a specific enzyme and activity. Characterisation of the enzyme mixtures revealed a large number of enzymes required for apple pomace degradation and these included cellulases, pectinases, xylanases, arabinases and mannanases in different proportions. Viscozyme L contained mainly pectinases and hemicellulases, while Celluclast 1.5L displayed largely cellulase and xylanase activity, hence the high degree of synergy reported. The temperature optimum was 50ºC for both enzyme mixtures and pH optima were observed at pH 5.0 and pH 3.0 for Viscozyme L and Celluclast 1.5L, respectively. At 37ºC and pH 5.0, the enzymes retained more that 90% activity after 15 days of incubation, allowing the enzymes to be used together with less energy input. The enzymes were further characterised by determining the effect of various compounds, such as alcohols, sugars, phenolic compounds and metal ions at various concentrations on the activity of the enzymes during apple pomace hydrolysis. Apart from lignin, which had almost no effect on enzyme activity, all the compounds caused inhibition of the enzymes to varying degrees. The most inhibitory compounds were some organic acids and metal ions, as well as cellobiose and xylobiose. Using the best ratio for Viscozyme L and Celluclast 1.5L (50:50) for the hydrolysis of apple pomace, it was observed that synergy was highest at the initial stages of hydrolysis and decreased over time, though the sugar concentration increased. The type of synergy for optimal apple pomace hydrolysis was found to be simultaneous. There was no synergy observed between Viscozyme L and Celluclast 1.5L with ligninases - laccase, lignin peroxidase and manganese peroxidase. Hydrolysing apple pomace with ligninases prior to addition of Viscozyme L and Celluclast 1.5L did not improve degradation of the substrate. Immobilisation of the enzyme mixtures on different supports was performed with the aim of increasing stability and enabling reuse of the enzymes. Immobilisation methods were selected based on the chemical properties of the supports, availability, cost and applicability on heterogeneous and insoluble substrate like apple pomace. These methods included crosslinked enzyme aggregates (CLEAs), immobilisation on various supports such as nylon mesh, nylon beads, sodium alginate beads, chitin and silica gel beads. The immobilisation strategies were unsuccessful, mainly due to the low percentage of immobilisation of the enzyme on the matrix and loss of activity of the immobilised enzyme. Free enzymes were therefore used for the remainder of the study. Hydrolysis conditions for apple pomace degradation were optimised using different temperatures and buffer systems in 1 L volumes mixed with compressed air. Hydrolysis at room temperature, using an unbuffered system, gave a better performance as compared to a buffered system. Reactors operated in batch mode performed better (4.2 g/L (75% yield) glucose and 16.8 g/L (75%) reducing sugar) than fed-batch reactors (3.2 g/L (66%) glucose and 14.6 g/L (72.7% yield) reducing sugar) over 100 h using Viscozyme L and Celluclast 1.5L. Supplementation of β- glucosidase activity in Viscozyme L and Celluclast 1.5L with Novozyme 188 resulted in a doubling of the amount of glucose released. The main products released from apple pomace hydrolysis were galacturonic acid, glucose and arabinose and low amounts of galactose and xylose. These products are potential raw materials for biofuel and biorefinery chemical production. An artificial neural network (ANN) model was successfully developed and used for predicting the optimum conditions for apple pomace hydrolysis using Celluclast 1.5L, Viscozyme L and Novozyme 188. Four main conditions that affect apple pomace hydrolysis were selected, namely temperature, initial pH, enzyme loading and substrate loading, which were taken as inputs. The glucose and reducing sugars released as a result of each treatment and their combinations were taken as outputs for 1–100 h. An ANN with 20, 20 and 6 neurons in the first, second and third hidden layers, respectively, was constructed. The performance and predictive ability of the ANN was good, with a R² of 0.99 and a small mean square error (MSE). New data was successfully predicted and simulated. Optimal hydrolysis conditions predicted by ANN for apple pomace hydrolysis were at 30% substrate (wet w/v) and an enzyme loading of 0.5 mg/g and 0.2 mg/mL of substrate for glucose and reducing sugar, respectively, giving sugar concentrations of 6.5 mg/mL and 28.9 mg/mL for glucose and reducing sugar, respectively. ANN showed that enzyme and substrate loadings were the most important factors for the hydrolysis of apple pomace.

- Description: Studies into sources of alternative liquid transport fuel energy have identified agro-industrial wastes, which are lignocellulosic in nature, as a potential feedstock for biofuel production against the background of depleting nonrenewable fossil fuels. In South Africa, large quantities of apple and other fruit wastes, called pomace, are generated from fruit and juice industries. Apple pomace is a rich source of cellulose, pectin and hemicellulose, making it a potential target for utilisation as a lignocellulosic feedstock for biofuel and biorefinery chemical production. Lignocellulosic biomass is recalcitrant in nature and therefore its degradation requires the synergistic action of a number of enzymes such as cellulases, hemicellulases, pectinases and ligninases. Commercial enzyme cocktails, containing some of these enzymes, are available and can be used for apple pomace degradation. In this study, the degradation of apple pomace using commercial enzyme cocktails was investigated. The main focus was the optimisation of the release of sugar monomers that could potentially be used for biofuel and biorefinery chemical production. There is no or little information reported in literature on the enzymatic degradation of fruit waste using commercial enzyme mixtures. This study first focused on the characterisation of the substrate (apple pomace) and the commercial enzyme cocktails. Apple pomace was found to contain mainly glucose, galacturonic acid, arabinose, galactose, lignin and low amounts of xylose and fructose. Three commercial enzyme cocktails were initially selected: Biocip Membrane, Viscozyme L (from Aspergillus aculeatus) and Celluclast 1.5L (a Trichoderma reesei ATCC 26921 cellulase preparation). The selection of the enzymes was based on activities declared by the manufacturers, cost and local availability. The enzymes were screened based on their synergistic cooperation in the degradation of apple pomace and the main enzymes present in each cocktail. Viscozyme L and Celluclast 1.5L, in a 50:50 ratio, resulted in the best degree of synergy (1.6) compared to any other combination. The enzyme ratios were determined on Viscozyme L and Celluclast 1.5L based on the protein ratio. Enzyme activity was determined as glucose equivalents using the dinitrosalicylic acid (DNS) method. Sugar monomers were determined using Megazyme assay kits. There is limited information available on the enzymes present in the commercial enzyme cocktails. Therefore, the main enzymes present in Viscozyme L and Celluclast 1.5L were identified using different substrates, each targeted for a specific enzyme and activity. Characterisation of the enzyme mixtures revealed a large number of enzymes required for apple pomace degradation and these included cellulases, pectinases, xylanases, arabinases and mannanases in different proportions. Viscozyme L contained mainly pectinases and hemicellulases, while Celluclast 1.5L displayed largely cellulase and xylanase activity, hence the high degree of synergy reported. The temperature optimum was 50ºC for both enzyme mixtures and pH optima were observed at pH 5.0 and pH 3.0 for Viscozyme L and Celluclast 1.5L, respectively. At 37ºC and pH 5.0, the enzymes retained more that 90% activity after 15 days of incubation, allowing the enzymes to be used together with less energy input. The enzymes were further characterised by determining the effect of various compounds, such as alcohols, sugars, phenolic compounds and metal ions at various concentrations on the activity of the enzymes during apple pomace hydrolysis. Apart from lignin, which had almost no effect on enzyme activity, all the compounds caused inhibition of the enzymes to varying degrees. The most inhibitory compounds were some organic acids and metal ions, as well as cellobiose and xylobiose. Using the best ratio for Viscozyme L and Celluclast 1.5L (50:50) for the hydrolysis of apple pomace, it was observed that synergy was highest at the initial stages of hydrolysis and decreased over time, though the sugar concentration increased. The type of synergy for optimal apple pomace hydrolysis was found to be simultaneous. There was no synergy observed between Viscozyme L and Celluclast 1.5L with ligninases - laccase, lignin peroxidase and manganese peroxidase. Hydrolysing apple pomace with ligninases prior to addition of Viscozyme L and Celluclast 1.5L did not improve degradation of the substrate.
