The results from this study indicated that GeoGebra can indeed be used effectively as a teaching tool to teach for conceptual understanding in mathematics.

- Description: A previous study conducted by our group identified indolyl-3-ethanone-a-thioethers (2.1a and 2.1b) as non-toxic, nanomolar, in vitro inhibitors of Plasmodium falciparum. Since the coumarin scaffold is associated with numerous biologically active compounds including antiprotozoal, anti-viral, anti-bacterial, and anti-inflammatory agents we were prompted to investigate coumaryl-3-ethanone-a-thioethers (2.1c) inspired by the activity of 2.1a and 2.1b against P. falciparum. We proposed a three-step synthesis of our target compounds 2.1c. The first step involved the Knoevenagel synthesis of 3-acetyl coumarins (2.3.1a - e) followed by a selective a-bromination to yield 3-bromoacetyl coumarin (2.2a). The final proposed step involved the nucleophilic displacement of the bromine by appropriately substituted thiophenols in either the presence or absence of base (K2CO3). Our initial findings revealed an unexpected major reductive dehalogenation of 2.2a into 2.3.1a. Further investigation revealed a close relationship between the electron withdrawing or donating nature of the thiophenol substituents and the relative formation of nucleophilic substitution or reductive dehalogenation products. Desired thioether products were obtained in higher yields when thiophenol was substituted with electron donating groups i.e. more nucleophilic thiophenols, while conversely, electron withdrawing substituents (i.e. lowered nucleophilicity) resulted in an increase of reductive dehalogenation. Furthermore, these results were consistent when experiments were conducted using either 2 or 1.2 equivalents of thiophenols which was an important observation in the context of two previous studies, by Oki et. al. and Israel et. al. Oki proposed that dehalogenation of a-chloro carbonyls occurs via sequential nucleophilic displacement of a-thioethers, while the study of Israel concluded that the dehalogenation of a-iodo carbonyls occurred in a single discreet step. Finally, in an effort to enhance nucleophilic substitution through the addition of K2CO3, we observed a Robinson annulation resulting in previously undescribed C-8 thiophenol functionalised dibenzo[b,d]pyran-6-ones (3.4a - e). In the introduction to this thesis, we briefly summarise the utility of coumarins in medicinal chemistry and related fields. Chapter two describes the rationalisation of our original research question and a retrosynthetic analysis of our desired compounds, followed by an initial description of the unexpected reductive dehalogenation. Chapter 3, begins with a brief review of reductive dehalogenation of a-halocarbonyls, and is followed by an analysis and discussion of our results in the context of the studies by Israel et. al. and Oki et. al.

- Description: Limited research has explored the impact of working underwater on the cognitive functioning of divers and even less has focused on the effect of breathing modality (assisted breathing and apnea) underwater. Research on the effect of submersion in water and apnea on cognitive performance is also very limited. As a result, the purpose of this study was to determine the effect of submersion in water and breathing modality on different stages of the information processing chain. This was achieved by testing participants in a laboratory setting under three conditions; on land, underwater with assisted breathing and underwater in apnea. Five different tests were used to determine which aspects of cognitive functioning were impaired in which condition. The recognition task result in the assisted breathing condition was significantly faster (p=0.04) but less accurate (p=0.01) than on land. The memory task was significantly (p=0.042) worse in terms of speed in the apnea condition compared to land, however accuracy was not affected. Performance in the visual detection task was impacted on in both underwater conditions compared to land with speed and accuracy being significantly worse (p<0.01) in the underwater conditions. These results indicate that simple tasks, (reaction time and tracking task) are not affected by condition whereas more complex tasks are. For tasks where an effect was found for only one condition, the effect was attributed to a specific aspect of that condition; either the breath hold or assisted breathing component of the condition. For tasks that were found to be affected in both underwater conditions, the effect was attributed to the actual submersion in water. The effect of the different conditions and the nature of the task they impact on should be considered for underwater work places as the general equipment used may impact on the quality of observations that are made.

- Description: This study titled ‘Negotiating the line between information and panic: A case study of Vanguard’s coverage of the Ebola Outbreak in Nigeria’ investigates and evaluates Vanguard’s coverage of the 2014 Ebola Virus Disease (EVD) outbreak in Nigeria. This study was conducted to understand how Vanguard negotiated the line between the dissemination of actual EVD information and the possibility of creating panic and fear during the coverage of the outbreak. Using qualitative content analysis and interview embedded within a qualitative research design, the study was carried out against the backdrop of relevant literature that asserts that the mass media is an important carrier, prime mover and producer of tensions, anxieties, fears and panics while the print media specifically is argued to have as part of its history the business of irrational fear mongering and the creation of panic about social problems. Vanguard, rated as one of the ten largest newspapers in Nigeria, was purposively selected for this study. Vanguard did substantial reporting and dissemination of the 2014 EVD outbreak and it was possible to draw on the availability and accessibility of the electronic archives of the EVD articles. Using the theory of moral panic, the normative theories of the media and theories about essential constituents of journalism culture as a theoretical framework, the study reveals that Vanguard’s coverage of the 2014 EVD outbreak was challenging and more demanding compared to coverage of previous outbreaks of viral diseases in Nigeria by the print medium. The study shows that lack of sufficient understanding of the science that underlies EVD, the initial exclusion of the media from the national response to contain the outbreak, and the lack of sufficient proactive measures by government and the print medium were powerful factors in how the epidemic was reported. The study also shows an interesting dimension in Vanguard’s early and latter coverage of the EVD outbreak while it lasted in Nigeria. This dimension reveals a high number of EVD articles with the propensity to inspire fear and panic in the early days of the coverage compared to EVD articles with the propensity to douse fear and panic in the early and latter days of the outbreak. The study shows that while the challenges encountered stem from Ebola’s mode of transmission, lack of scientific and medically proven cure and early coverage amidst uncertainties, the inherent tensions and anxieties that characterised the outbreak coupled with Vanguard’s fire brigade approach led to the relatively high number of EVD articles with the propensity to inspire fear and panic. The study recommends the mass media’s inclusion in national responses to epidemics and ongoing training for health journalists to update their knowledge base about emerging and infectious diseases. The study also recommends for further study a reception analysis to enhance the socio-cultural understanding of how the EVD articles were received.

The ecological impacts of pollution on a river ecosystem: a community index and stable isotope approach

- Description: For decades, urbanized rivers have been modified to meet the needs of constantly expanding human populations in many countries around the world. The Bloukrans River in Grahamstown is one of the polluted and structurally modified urban rivers in South Africa, and there is no published information regarding its water quality and ecological status. Water quality is threatened by human activities including the disposal of treated and raw sewage, livestock farming, and agriculture. This study was conducted to determine the ecological status of the river by assessing its biological, chemical, and physical components in relation to man-mediated activities. Biological responses of macroinvertebrates were used to assess changes in water quality through space and time based on the South African Scoring System version 5 and the Average Score per Taxon water quality indices. The results showed poor water quality due to high nitrate and ammonium concentrations derived from sewage, and channel modifications by agricultural activities and dumping of solid waste. Channel width, water depth, dissolved oxygen, nitrate, and ammonium concentrations were the main drivers of macroinvertebrate distribution patterns and had the most influence on the variability in macroinvertebrates taxa richness, diversity and abundance. Diatoms were also used to assess water quality, specifically to indicate the trophic status of the river based on changes in the Trophic Diatom Index. The results suggested that the Bloukrans River was eutrophic during the course of this study. However, the trophic status varied with freshwater input, resulting in mesotrophic conditions during flooding and eutrophication in dry seasons. Changes in pH, phosphate concentration, water velocity (current speed), and temperature influenced the distribution of diatoms in the Bloukrans River. However, only pH was important at the community level and significantly influenced diatom abundances. Stable nitrogen isotope ratios (S15N) of autotrophs and primary and secondary consumers revealed noticeable differences between tissues of organisms exposed to treated sewage and those without any exposure. The S15N values in biota occurring above the sewage treatment discharge point were low, and those collected below the sewage point were higher. Although fertilizer derived nitrogen is generally depleted in 15N, agriculture-derived nitrogen could not be excluded as a possible source since animals at the sample site that was most affected by agricultural activities had the highest S15N values. This study provided valuable information on the ecological status of the Bloukrans River and identified the major activities associated with reduced biodiversity and water quality.

A psychosocial study of young adults’ experiences of their sibling’s mental illness

- Description: There are a number of studies that investigate the experiences of mental illness in the context of sibling relationships. However, these studies have not focused on young adulthood and limited research has been conducted in South Africa. This research uses a psychosocial framework which combines psychoanalytic theory and social constructionism to account for individual subjectivity and social influences. This method sought to answer two main research questions: how do young adults construct their experiences of having a sibling with a mental illness and with what effects, and how might we understand the emotional investments in these constructions? Six participants were interviewed and the findings suggest that participants draw on four main discourses in constructing their siblings’ mental illness: a discourse of mental illness as a sickness, a discourse of mental illness as part of the person, a discourse of mental illness as bad behaviour, and a discourse of mental illness as a spiritual issue. These discourses and the function of these discourses are discussed. Furthermore, an extract from one participant is examined in a case study format so as to explore the emotional investments in the discourse of mental illness as bad behaviour, arguing that her investments in this discourse serves to protect her by enabling her to manage feelings of not being ‘good enough’.

- Description: There are a number of studies that investigate the experiences of mental illness in the context of sibling relationships. However, these studies have not focused on young adulthood and limited research has been conducted in South Africa. This research uses a psychosocial framework which combines psychoanalytic theory and social constructionism to account for individual subjectivity and social influences. This method sought to answer two main research questions: how do young adults construct their experiences of having a sibling with a mental illness and with what effects, and how might we understand the emotional investments in these constructions? Six participants were interviewed and the findings suggest that participants draw on four main discourses in constructing their siblings’ mental illness: a discourse of mental illness as a sickness, a discourse of mental illness as part of the person, a discourse of mental illness as bad behaviour, and a discourse of mental illness as a spiritual issue. These discourses and the function of these discourses are discussed. Furthermore, an extract from one participant is examined in a case study format so as to explore the emotional investments in the discourse of mental illness as bad behaviour, arguing that her investments in this discourse serves to protect her by enabling her to manage feelings of not being ‘good enough’.

Thus, the National Curriculum of Basic Education [NCBE], 2010, p. 8) expects Grade 6 Natural Science learners to be competent in using simple scientific models, methods and skills to make scientific sense of the natural environment; and most importantly for the learners to relate the implications of this scientific understanding to their personal and social health and the sustainable use of all natural resources for future generations. Additionally, teachers are encouraged to make innovative use of various knowledge systems including indigenous knowledge. However, there is no mention of how teachers should make use of local indigenous knowledge (IK) when teaching the topic on water conservation in particular. It is against this backdrop that in this study I sought to understand how two Grade 6 Natural Science teachers use local indigenous knowledge in their classrooms to teach the topic of water conservation. This interpretive case study was carried out in collaboration with a rural community in the Erongo region. Data were collected in three stages. The first stage was the gathering of information through document analysis to strengthen my context. Documents such as the National Curriculum, Science Curriculum, Syllabi, Grade 6 Natural Science textbooks and SATs results were analysed to gain some insight and understanding of the issue before conducting interviews. In the second phase of this study I conducted semi-structured interviews with the Topnaar community members and two teachers. This was followed by classroom observations. Additionally, I used stimulated recall interviews (SRI) to provide clarity on how teachers use prior knowledge and mediate learning when teaching the topic water conservation using indigenous knowledge. The study is informed by Vygotsky’s (1978) socio-cultural theory in conjunction with Shulman’s (1986) pedagogical content knowledge (PCK). Since a classroom is a social unit where interactions occur between teachers and learners, and learners and learners, these theories provided a useful lens for observing lessons and analysing teachers’ content knowledge and how they mediate learning using local indigenous knowledge. A thematic approach to data analysis was adopted and qualitative data were analysed inductively. The findings of the study revealed that the indigenous knowledge amongst the Topnaars are governed by the ideals of Ubuntu. Findings also revealed that proper planning is needed to incorporate IK into science lessons as it may clear up misconceptions that may arise from indigenous knowledge. The study recommends that teachers and curriculum planners should explore different indigenous practices that are linked to the topics in the curriculum or syllabus to enhance learning and teaching in their science classrooms. It also recommends that curriculum developers, with the help of subject advisors, teachers and community members, should start planning on how indigenous knowledge can best be integrated into science textbooks as it has become a hindrance for the teachers to incorporate it.

The second chapter investigates the curvature of nonholonomic Riemannian manifolds and the Schouten and Wagner curvature tensors. The Schouten tensor is canonically associated to every non- holonomic Riemannian structure (in particular, we use it to define isometric invariants for structures on three-dimensional manifolds). By contrast, the Wagner tensor is not generally intrinsic, but can be used to characterise flat structures (i.e., those whose associated parallel transport is path-independent). The third chapter considers equivalence of nonholonomic Rie- mannian manifolds, particularly up to nonholonomic isometry. We also introduce the notion of a nonholonomic Riemannian submanifold, and investigate the conditions under which such a submanifold inherits its geometry from the enveloping space. This has allowed exogenous factors like sentiment and developed country monetary policy to affect developing countries capital markets and macroeconomic fundamentals. This study analyses the impact of investor sentiment alongside US monetary policy, country specific risks, inflation and domestic stock returns on the BRICS credit default spreads. To investigate this relationship, the study uses panel data and a fixed effects model. The results of the panel regressions suggest that all variables had an impact on the variation of BRICS credit default spreads however the crisis may have distorted the relationship among the variables. Sovereign ratings had an inverse relationship depicting a rise in ratings decreasing the credit default premium. This was in line with a priori expectations. Domestic company earnings also had an inverse relationship with BRCIS credit default premia, the magnitude of which is dependent on the value of the index. This is to say the higher the index, the more significant the effect on the BRICS default premium. US monetary policy was significant and in line with expectations of a linear relationship between emerging market credit default spreads when controlling for the crisis. In the crisis period however, results depicted an inverse relationship going against a priori expectations. The inflation variable was found to have a greater impact on CDS spreads during the crisis period, while the VIX index had a linear relationship with the default premia albeit the impact was not highly significant. The study concludes that the financial crisis was an important event that affected the relationship of these variables with BRICS country default spreads and had read through to market participant’s behaviour at the time.

Despite these reforms, however, the country has barely attained desirable macroeconomic performance. This study sets out to establish if the need for these policy reforms is due to the fact that the country is constrained from the simultaneous achievement of optimal levels of monetary policy independence, exchange rate stability and financial integration, as postulated by the ‘trilemma’. The trilemma is evaluated using an approach introduced by Aizenman et al. (2008), in which the Ordinary Least Squares (OLS) method is applied to a model in which a constant is regressed on indices constructed for the policy intermediate goals; the results indicate that the trilemma is a binding constraint in Malawi and that the largest trade-off is between exchange rate stability and financial integration. Given these constraints, the study also considers the combination of the trilemma intermediate policy goals that has been dominant in the country in the last three decades, using predicted values from the model and a graphical analysis to explore this objective. The analysis reveals that Malawi has, on average, prioritised exchange rate stability and monetary policy independence at the expense of financial integration. The study also assesses how the trilemma intermediate policy goals affect macroeconomic performance, specifically regarding output growth rate and inflation. The results reveal that exchange rate stability is associated with faster output growth, financial integration is associated with higher inflation, and that monetary policy independence is not a significant factor. The results emphasise the importance of consistent stability of the exchange rate if Malawi is to achieve faster and sustainable economic growth. Given this, policy makers must be cautious, as the current floating exchange rate regime, combined with financial integration, could lead to slow growth and high inflation.

- Description: This study aimed to establish the clinical utility of the Kinetic School Drawing (KSD) projective technique. It attempted to do so by using an existing self-report measure of children’s self-perceptions in the school environment (Harter’s Self-Perception Profile for Children) by which to corroborate the projections derived from the KSD’s. Following the development of this measure (in 1984) limited research has been undertaken to assess its validity, reliability and clinical utility despite promising initial findings. The KSD has therefore not received much attention and as a result, it has yet to be established as a useful psychological assessment tool. This study sought to revive interest and discussion around the KSD. Qualitative methods and specifically thematic analysis was employed to explore the usefulness of the KSD in an attempt to address the gap in the literature in a meaningful way. The sample consisted of 26 grade 5 learners from a private school in the Eastern Cape. The themes were presented under the following headings: scholastic competence, athletic competence, social competence, physical appearance and behavioural conduct.

Combined in silico approaches towards the identification of novel malarial cysteine protease inhibitors

- Description: Malaria an infectious disease caused by a group of parasitic organisms of the Plasmodium genus remains a severe public health problem in Africa, South America and parts of Asia. The leading causes for the persistence of malaria are the emergence of drug resistance to common antimalarial drugs, lack of effective vaccines and the inadequate control of mosquito vectors. Worryingly, accumulating evidence shows that the parasite has developed resistant to the current first-line treatment based on artemisinin. Hence, the identification and characterization of novel drug targets and drugs with unique mode of action remains an urgent priority. The successful sequencing and assembly of genomes from several Plasmodium species has opened an opportune window for the identification of new drug targets. Cysteine proteases are one of the major drug targets to be identified so far. The use of cysteine protease inhibitors coupled with gene manipulation studies has defined specific and putative roles of cysteine proteases which include hemoglobin degradation, erythrocyte rupture, immune evasion and erythrocyte invasion, steps which are central for the completion of the Plasmodium parasite life cycle. In an aim to discover potential novel antimalarials, this thesis focussed on falcipains (FPs), a group of four papain-like cysteine proteases from Plasmodium falciparum. Two of these enzymes, FP-2 and FP-3 are the major hemoglobinases and have been validated as drug targets. For the successful elimination of malaria, drugs must be safe and target both human and wild Plasmodium infective forms. Thus, an incipient aim was to identify protein homologs of these two proteases from other Plasmodium species and the host (human). From BLASTP analysis, up to 16 FP-2 and FP-3 homologs were identified (13 plasmodial proteases and 3 human cathepsins). Using in silico characterization approaches, the intra and inter group sequence, structural, phylogenetic and physicochemical differences were determined. To extend previous work (MSc student) involving docking studies on the identified proteins using known FP-2 and FP-3 inhibitors, a South African natural compound and its ZINC analogs, molecular dynamics and binding free energy studies were performed to determine the stabilities and quantification of the strength of interactions between the different protein-ligand complexes. From the results, key structural elements that regulate the binding and selectivity of non-peptidic compounds onto the different proteins were deciphered. Interaction fingerprints and energy decomposition analysis identified key residues and energetic terms that are central for effective ligand binding. This research presents novel insight essential for the structure-based molecular drug design of more potent antimalarial drugs.

- Description: Novobiocin (Nb) is a coumarin type antibiotic isolated from the bacterium species of Streptomyces and possesses modest anticancer and antimalarial activities. Nb and analogues have been extensively explored as potential anticancer agents through inhibition of the C- terminal domain of heat shock protein 90 (Hsp90), which plays a pivotal role in the proteinfolding machinery of cells. There has been little effort in the exploration of Nb and derivatives for antimalarial activity. Incorporation of organometallic units, such as ferrocene (Fc), into bioactive chemical scaffolds remains an attractive approach for developing new therapeutic agents for treatment of several ailments. The current study sought to investigate the anticancer and antiplasmodial effects of incorporating ferrocene (Fc) into Nb scaffold presumably through inhibition of Hsp90. The ferrocenyl Nb analogues containing simplified structural motifs such as phenyl, benzyl, and piperidine were synthesized in six to nine steps employing conventional synthetic organic protocols adapted from literature, and the compounds were accessed in reasonable yields. For comparison purposes, a selection of organic Nb analogues were also included in the study. The target compounds were characterized by spectroscopic techniques including 1-dimensional nuclear magnetic resonance (1D NMR) and high-resolution mass spectroscopy. The synthesized compounds were evaluated in vitro for potential anticancer and antiplasmodial activities using the breast cancer cell line (HCC38) and chloroquine-sensitive strain (3D7) of the malaria parasite, Plasmodium falciparum. The presence of the Fc unit was found to enhance both anticancer and antiplasmodial activities of the resultant ferrocenyl Nb compounds with IC50 values in the low to mid micromolar range. Hsp90 inhibitory studies of the ferrocenyl Nb analogues possessing superior activities (2.13a and 2.20c) were also conducted using different yeast strains expressing both human and malarial Hsp90 isoforms: hHsp90a/p and PfHsp90, respectively. The results of Hsp90 inhibitory studies suggested no direct correlation between the observed activities of the analogues and Hsp90 inhibition. - Description: A brief history of political correctness is discussed along with various definitions of it, ranging from political correctness being a benign attempt to prevent offense and avert discrimination to stronger views equating it with Communist censorship or branding it as "cultural Marxism". The aim of the research is to discover what political correctness is, how it relates to freedom of expression and what wider implications and effects it has on society. The moral foundations of rights and free speech in particular are introduced in order to set a framework to determine what authority people and governments have to censor others' expression. Different philosophical views on the limits of free speech are discussed, and arguments for and against hate speech are analysed and related to political correctness. The thesis looks at political correctness on university campuses, which involves speech codes, antidiscrimination legislation and changing the Western canon to a more multicultural syllabus. The recent South African university protests involving issues such as white privilege, university fees and rape are discussed and related to political correctness. The thesis examines the role of political correctness in the censorship of humour, it discusses the historical role of satire in challenging dogmatism and it looks at the psychology behind intolerance. Political correctness appeals to tolerance, which is sometimes elevated at the expense of truth. Truth and tolerance are therefore weighed up, along with their altered definitions in today's relativistic society. The last part of the thesis looks at South Africa's unique brand of political correctness, along with Black Economic Empowerment, colonialism and white guilt, and the research concludes that political correctness is a distinct form of censorship which has developed in modern democracies. The new forms of justice and morality seen in political correctness are distortions of left-wing liberalism, which appeal to different values to those of traditional liberalism.
