- Description: Various spatialisation algorithms require the knowledge of speaker locations to accurately localise sound in 3D environments. The rendering process uses speaker coordinates to feed into their algorithms so that they can render the immersive audio content as intended by an artist. The need to measure the loudspeaker coordinates becomes necessary, especially in environments where the speaker layouts change frequently. Manually measuring the coordinates, however, tends to be a laborious task that is prone to errors. This research provides an automated solution to the problem of speaker coordinates measurement. The solution system, SDIAS, is a client-server system that uses the capabilities provided by the Ethernet Audio Video Bridging standard to measure the 3D loudspeaker coordinates for immersive sound systems. SDIAS deploys commodity hardware and readily available software to implement the solution. A server sends a short tone to each speaker in the speaker configuration, at equal intervals. A microphone attached to a mobile device picks up these transmitted tones on the client side, from different locations. The transmission and reception times from both components of the system are used to measure the time of flight for each tone sent to a loudspeaker. These are then used to determine the 3D coordinates of each loudspeaker in the available layout. - Description: This work reports on the synthesis and characterization of novel acetophenone substituted phthalocyanines along with the self-assembled nanoconjugates formed via π-π stacking interaction between the synthesized unmetalated (2), zinc (3) and indium (4) phthalocyanines and graphene quantum dots (GQDs) to form 2@GQDs, 3@GQDs and 4@GQDs. The complexes and conjugates exhibited high singlet oxygen ranging from 0.20 to 0.79 in DMSO for Pcs and nanoconjugates where in all cases, the indium complexes showed the highest singlet oxygen quantum yields. The photodynamic antimicrobial chemotherapy activity of both phthalocyanines and nanoconjugates were tested against Staphylococcus aureus. 4@GQDs was found to be highly effective causing a 9.68 log reduction of the bacteria at 10 μM (based on Pc) when compared to 3.77 log reduction of 3@GQDs.

The novobiocin-induced turnover of fibronectin via low density lipoprotein receptor-related protein 1 alters matrix morphology with physiological consequences on cell growth and migration

- Description: Fibronectin (FN), an extracellular matrix protein, is secreted as a soluble dimer which is assembled into an insoluble extracellular matrix. The dynamics of FN matrix assembly and degradation play a large role in cell migration and invasion thereby contributing to the metastatic potential of cancer cells. Previous studies have shown the direct binding of Heat Shock Protein 90 kDa (Hsp90) and FN in vitro, and that inhibition of Hsp90 with novobiocin (NOV) caused internalisation of the FN matrix. Low density lipoprotein receptor-related protein 1 (LRP1) is a ubiquitous receptor known to bind both Hsp90 and FN. Using an LRP1 expressing Hs578T breast cancer cell line and an isogenic mouse embryonic fibroblast (MEF) model system of differential LRP1 expression we demonstrate that LRP1 is involved in turnover of FN in response to C-terminal Hsp90 inhibition. The first objective of this study was to identify the mechanism of NOV-induced LRP1-mediated FN turnover. Our data show that NOV-mediated FN turnover via LRP1 did not require the activity of matrix metalloproteinases (MMPs), which play an important role in processing and degradation of the extracellular matrix and FN. In addition, the levels of the main FN receptor responsible for its extracellular assembly, β1-integrin, did not change in response to NOV. LRP1 is known to undergo regulated intramembrane proteolysis (RIP) which generates smaller fragments that may translocate to the nucleus and modulate gene transcription. Using inhibitors of LRP1 cleavage and nuclear fractionation we determined that LRP1 processing was not required for the NOV-induced FN response suggesting that a mechanism unrelated to LRP1 RIP is involved. A possible mechanism may be in altered Hsp90-LRP1 cell signalling as we observed disruption of the FN-Hsp90-LRP1 complex at the cell surface in NOV treated cells. How this affects downstream eHsp90-LRP1 signalling is still to be determined but may be related to a significant increase in phospho-AKT and loss of phospho-ERK upon NOV-treatment; two key signalling proteins involved in FN matrix regulation and which are downstream of LRP1 signalling. The second objective of this study was to determine the physiological consequences associated with FN turnover in response to NOV treatment. Using migration assays we demonstrated that levels of insoluble matrix-associated FN and FN concentration are not solely responsible for migratory capacity of cells on decellularized extracellular matrices, but rather that structural composition and integrity of the matrix plays a bigger role. Using confocal and scanning electron microscopy, we identified NOV treated matrices to be flatter, less mature and contain thicker, rope-like FN fibrils to which cells adhered better but were generally less proliferative. Comparatively, cells adhered less to the more mature and 3-dimensional untreated matrices but exhibited increased spreading and cell growth, which may in part be due to the thinner fibrils and web-like matrix. In summary, this study substantiates the role of LRP1 in NOV-mediated FN turnover, and provides new insights into the possible mechanisms of the Hsp90-LRP1 mediated loss of FN matrix. Pcs were covalently linked to nanondiamonds and investigated using 532 nm laser excitation at 7 ns pulses for their optical limiting properties. Si(OH)2TBPc and Si(OH)2Pc gave larger triplet quantum yields when linked to DNDs, while the value decreased for Si(OH)2TPPc in the presence of DNDs due to aggregation. However all Pcs showed enhanced nonlinear optical properties in the presence of DNDs. DNDs-Si(OH)2TPPc and DNDs-Si (OH)2TBPc gave the highest imaginary third-order susceptibility (Im[X(3)]) and hyperpolarizability (γ) at 5.19 × 10−8 and 3.85 × 10−8 esu and 2.66 × 10−27 and 1.97 × 10−27 esu, respectively. DNDs-Si(OH)2TBPc nanoconjugates showed lowest limiting threshold (Ilim) value of 0.01 J.cm−2 relative to 0.09 for DNDs-Si (OH)2TPPc.

In this study, rare-earth sandwich-type phthalocyaninato chelates based on neodymium (1 a) and samarium (1 b) were employed as electrocatalysts to modify glassy carbon electrodes (GCE) for the first time. It was found that 1 a-modified GCE (herein referred to as 1 a-GCE) is less conductive than 1 b-modified counterpart (1 b-GCE). A larger rate constant was also obtained for 1 b-GCE. It was established that a faster oxidation rate efficiency was responsible for lower limit of detection value obtained for 1 b-GCE as compared to 1 a-GCE.

While studies undertaken in the Global North highlight that video materials in the FL classroom have the potential to influence learning and cultural knowledge acquisition, there is a lack of research on the manner in which pedagogically designed videos influence adult FL learning and cultural knowledge acquisition in a South African context. In this study, I explore the opportunities and challenges in terms of language learning and cultural knowledge acquisition that arise from three pedagogic videos in the Menschen A1 textbook which is used in teaching students registered for the German Studies 1 course at Rhodes University. I compare and contrast two sets of data to examine the relationship between pedagogic video and student knowledge acquisition: the results of a digital multimodal discourse analysis (DMDA) of these videos, and questionnaires and transcriptions collected from semi-structured group interviews with German Studies 1 students. These questionnaires and transcriptions were analysed thematically. Findings in terms of the language learning experience indicate that actor over-exaggeration and visual aids assist students when learning German at this level with this type of video. However, these visual aids can be distracting and confusing without balanced representation and contextual information. Differences between videos in terms of speech rate seem to affect students’ perceptions of their ability to comprehend the videos. Students reported forming only positive impressions of German culture(s) on the basis of the videos. In general, students find Germans portrayed as friendly and helpful in the videos. The results of this investigation provide recommendations for the optimal use of this type of teaching and learning resource, for example, teachers/ lecturers/ facilitators should allow for focus group discussions on cultural discourse to occur in order to balance stereotype formation and should consider the speech rate of videos for language learning. , Thesis (MA) -- Faculty of Humanities, Linguistics and Applied Language Studies, 2021

I compare and contrast two sets of data to examine the relationship between pedagogic video and student knowledge acquisition: the results of a digital multimodal discourse analysis (DMDA) of these videos, and questionnaires and transcriptions collected from semi-structured group interviews with German Studies 1 students. These questionnaires and transcriptions were analysed thematically. Findings in terms of the language learning experience indicate that actor over-exaggeration and visual aids assist students when learning German at this level with this type of video. However, these visual aids can be distracting and confusing without balanced representation and contextual information. Differences between videos in terms of speech rate seem to affect students’ perceptions of their ability to comprehend the videos. Students reported forming only positive impressions of German culture(s) on the basis of the videos. In general, students find Germans portrayed as friendly and helpful in the videos. The results of this investigation provide recommendations for the optimal use of this type of teaching and learning resource, for example, teachers/ lecturers/ facilitators should allow for focus group discussions on cultural discourse to occur in order to balance stereotype formation and should consider the speech rate of videos for language learning. , Thesis (MA) -- Faculty of Humanities, Linguistics and Applied Language Studies, 2021

This approach incites healthcare practitioners to achieve particular performance standards and to monitor their professional agency. Managing pregnancy risk thus entails regulating the healthcare practitioners themselves. In implementing approved pregnancy risk management strategies in an over-subscribed and under-resourced public healthcare setting, however, healthcare practitioners face potential risk to their professional reputation and integrity. In managing this risk, they resist the scientific-bureaucratic approach through: depicting themselves as victims of unfair institutional arrangements or unreasonable patients; instituting street-level bureaucracy to control access to the clinics; and controlling patients’ actions in authoritarian ways. Our research shows that without engagement with the on-the-ground realities of the antenatal clinic in resource-poor environments, a scientific-bureaucratic approach to pregnancy risk management is inevitably limited in its effectiveness.

Balancing the need for methods to be appropriate, ethical and feasible within each setting with the desire for sufficient consistency across the project is challenging. Understanding young people's experiences in urban environments is therefore important as we strive to achieve both the Sustainable Development Goals and the Paris Agreement. Two amenities identified in the urban Sustainable Development Goal 11, transport and public and green space, are specifically recognised as applying to youth. Yet, there is little analysis that explicitly considers how youth experience these amenities across the Global North and South, and no current measures for understanding progress in youth experiences of green space and transport. Results: This paper provides a comparative analysis of young people's experiences with local transport and green space in seven diverse urban communities (Christchurch, New Zealand; Dhaka, Bangladesh; Lambeth/London, UK; Makhanda, South Africa; New Delhi, India; São Paulo, Brazil; and Yokohama, Japan). Our study contributes to a growing body of literature that seeks to listen to child and youth perspectives to understand their environmental experiences. We examine the 'affordances' young residents aged 12 to 24 years currently associate with green space and transport amenities. Affordances are defined here as the inter-relationships between what a local environment offers young people and their perceptions and actions. Drawing on focus groups and interviews conducted with 332 young people, we identify five affordances young people associate in relation to transport and public space across these diverse urban settings: (1) social inclusion and belonging; (2) autonomy; (3) physical comfort and security; (4) relaxation and reflection; and (5) health and fitness. Conclusions The paper contributes to growing interdisciplinary research interest in measuring affordances as a way to advance the Sustainable Development Goals in an urban context. In providing a comparative account of young people's experiences across diverse contexts, our discussion highlights how affordances in relation to transport or public and green space can help understand the multiple interconnections between the well-being of young people and sustainability. We expand on the standard overview of the field by identifying some of the challenges associated with managing complex systems with multiple resources, multiple stakeholder groups with diverse knowledges/understandings, and multiple objectives across multiple scales, i.e., multifaceted collaborative initiatives. We then elucidate how a realist methodology, within a critical realist framing, can support efforts to compare multiple case studies of such multifaceted initiatives. The methodology we propose considers the importance and impact of context for the origins, purpose, and success of multifaceted collaborative natural resource management and conservation initiatives in social-ecological systems.

A growing recognition of the complexity of development problems and contexts calls for new framings, including a new approach to monitoring and evaluation (M and E) as one of the mechanisms by which modern societies aim to steer towards a more sustainable future. New approaches to M and E mean new skills for the M and E practitioner. Objectives: This article proposed a framing for M and E skills, comprising of technical, relational and transformational (T-R-T) competences. Method: Adapted from the literature, this competence framework was tested in a broader learning needs assessment and then applied retrospectively to author’s experience in developmental evaluations in complex social–ecological contexts in southern Africa. Results: The emerging insights were that not only technical competence is needed, but also relational competence that goes beyond interpersonal skills, to enable the production and uptake of evaluation findings. In addition, the limitations of mainstream M and E methods in the face of complexity seemed to create a need for ‘transformational’ competence, which included evaluators’ ability to develop credible M and E alternatives. Conclusion: The T-R-T framework helped to advance the notions of ‘hard’ and ‘soft’ skills and expanded on existing M and E competence frameworks. Recommendations included a call for innovative educational and professional development approaches to develop relational and transformational competencies, in addition to training for technical competence.

- Description: We reply to the article, ‘Why are black South African students less likely to consider studying biological sciences?’, authored by University of Cape Town (UCT) researcher Nicoli Nattrass1 and published in the South African Journal of Science on 27 May 2020. At the time of writing our reply the article had already received sharp criticism from the Black Academic Caucus at UCT and in a statement released on 5 June 2020, the UCT executive distanced itself from the content of the paper, inviting rigorous, respectful review of the published research. Nattrass’ article1 follows two papers published in 2019 by scholars associated with South African universities, both of which were widely denounced for the racist undertones of their content. The first article was authored by Stellenbosch University researchers who reported on low cognitive functioning of coloured women linked to education levels and lifestyles2 ; see comment by Le Grange3 . The second article was co-authored by an adjunct professor at UCT and examined the role of cognitive ability or intelligence on slave exports from Africa4 . These publications gave rise to a broader debate on enduring racism in science and the re-ascendency of race science internationally. In this reply, we focus on the methodology of attitudinal survey used for the study reported by Nattrass (and by many other scientists). There are two lines of argument that we weave together: the quality of the research as reported, and problems inherent to comparative attitudinal survey research (regardless of how well it is executed). It should be noted that the Black Academic Caucus at UCT has also critiqued the research design, identifying many of the points we make here, and they have additionally included a discourse analysis of the paper (circulated via e-mail). To demonstrate how problematic a survey design can be, particularly when it is not rigorously executed, we share data and observations that counter the apparent findings of Nattrass’ study. We then argue that the publication of this research, in this manner, is detrimental to the biodiversity sector in which the tropes being perpetuated, can cause enormous harm. In the process we hope to encourage scientists to be more reflexive about their methodology, and we encourage the South African Journal of Science to publish works that are worthy of the young people of this country - research in which they can recognise themselves, rather than being forced to look at reductionist portrayals, legitimised under the banner of science.

- Description: Landscape stewardship is increasingly understood within the framing of complex social-ecological systems. To consider the implications of this, we focus on one of the key characteristics of complex social-ecological systems: they are relationally constituted, meaning that system characteristics emerge out of dynamic relations between system components. We focus on multi-actor collaboration as a key form of relationality in landscapes, seeking a more textured understanding of the social relations between landscape actors. We draw on a set of ‘gardening tools’ to analyse the boundary-crossing work of multi-actor collaboration. These tools comprise three key concepts: relational expertise, common knowledge, and relational agency. We apply the tools to two cases of landscape stewardship in South Africa: the Langkloof Region and the Tsitsa River catchment. These landscapes are characterised by economically, socio-culturally, and politically diverse groups of actors. Our analysis reveals that history and context strongly influence relational processes, that boundary-crossing work is indeed difficult, and that doing boundary-crossing work in smaller pockets within a landscape is helpful. The tools also helped to identify three key social-relational practices which lend a new perspective on boundary-crossing work: 1. belonging while differing, 2. growing together by interacting regularly and building common knowledge, and 3. learning and adapting together with humility and empathy.

Working at landscape-level requires collaboration between diverse landscape stakeholders. In this study, we partnered with local stewardship practitioners across six cases in South Africa to investigate how they facilitate collaboration towards social-ecological sustainability outcomes. We found that practitioners facilitate collaboration among stakeholders by operating as relational hubs in the landscape. Through these hubs, they build new inter-personal relationships among stakeholders, creating social networks which enable stewardship practice. The hubs deepen human-nature relationships by creating enabling conditions for stewards to put stewardship ethics into action. Drawing on insights from these cases, we call for a relational approach to landscape stewardship which focuses on human-to-human and human-to-nature relationships. Moreover, we argue that landscape stewardship initiatives need to re-focus stewardship on stewards, recognizing them as key agents of change in addressing the conflict between agriculture and conservation inherent in many landscapes.
