Colonial displacements and African coping strategies: The experience of BaTonga of Binga, Zimbabwe, 1956-2008

- Description: This study examines the challenges faced by BaTonga in livelihood reconstruction after involuntary displacement and resettlement from the Zambezi valley to pave way for the Kariba dam reservoir. It shows how they were forcibly evicted without compensation because of the racist policies of Southern Rhodesia. Their resettlement into an arid region infested with marauding elephants, malaria and tsetse fly undermined their complex livelihoods and eroded their self-sufficiency, but the study argues that they were active and resilient agents who adopted complex coping strategies. It shows how they had to adapt to dry-land farming and counter the effects of wildlife which plundered their crops and managed to secure the harvest in the drought-prone region. The study shows that although they adopted precarious livelihoods, they managed to survive under extreme circumstances for decades without any external assistance until the coming of NGOs who began to distribute free food from 1982. It argues that prolonged distribution of emergence food aid may not have been necessary, but its coming for almost three decades largely served political interests of both the NGOs and their governments. This perpetuated poverty as BaTonga also manipulated its distribution as a coping strategy against fragile livelihoods. This caused dependency which further plunged them into chronic food insecurity because they abandoned their traditional coping strategies. The study argues that both the colonial and postcolonial government as well as NGOs failed to address the root causes of livelihood insecurity in Binga for the five decades under study. Climate for changing lenses: Reconciliation through site-specific, media arts-based environmental education on the water and climate change nexus in South Africa and Canada

- Description: This study took place in the context of a growing racialised global water crisis and increasing demands worldwide for transforming higher education at institutions of ongoing settler colonialism. It presents a conceptualisation of what education, research and activism can look like and unfolded inside a doctoral research project that expands what doctoral education can look like. Using a media arts-based praxis process, I developed a relational model of university curriculum –site-specific, media arts-based, environmental education –with potential to cultivate relations (human and nonhuman) towards reconciliation while contributing to justice at the water-climate change nexus. My aim as a settler-ally was to expand my teaching and curriculum practices, thereby also offering curriculum transformation inspiration to others. My research was rooted in my concept of reconciliation as a practice towards thriving together, where the ‘together’ was inclusive of both humans and nonhumans. The curriculum engaged students in de/re/constructing water narratives through making site-specific videos focused on local water bodies. Decolonising artistic approaches known as slow media and soundscape recording were strategically incorporated into audio/video mapping assignments where students observed water aesthetics in ways that shifted their perceptions about water and entities entangled with it. Students met with Knowledge Keepers (Indigenous and non-Indigenous people from outside the academy with existing relationships to water bodies). A photovoice methodology was used in these meetings with Knowledge Keepers to reconfigure traditional film director-subject power relations. Guest lecturers from non-traditional backgrounds contributed diverse perspectives. Ecomotricity was incorporated, whereby students were in deliberate movement in/with water bodies through canoeing together. The curriculum culminated in a public screening/education event where resulting videos, interspersed with educational games facilitated by students, surfaced emotions, knowledge co-production and new synergies amongst the event’s temporary community. Through two iterations of the curriculum, where I co-designed and taught a course called Making Waveforms, one in Vancouver, Canada and one in Cape Town, South Africa, I explored the primary research question: How can a relational site specific, media arts-based university environmental education curriculum cultivate students’ relational sensibilities and abilities oriented towards reconciliation of diverse peoples and ecosystems in South Africa and Canada? Iterating the curriculum across these two contexts allowed me to assess which aspect(s)of the curriculum may have been applicable across these and other contexts. By using mixed methods of data collection and sharing throughout the research journey, I explored the sub-questions: a) How is reconciliation understood currently by university students in South Africa and Canada? and b) How can a relational site-specific, media arts-based university environmental education curriculum and my PhD methodologies (PhD-by-publication, website, and participatory approaches to podcasting, video making, and song creation), contribute to decolonising higher education, and thereby further contribute to reconciliation of diverse peoples and ecosystems in South Africa and Canada? Integral to my praxis process, I undertook a PhD-by-publication that involved writing four academic journal articles, with each paper presenting a key stage in the process. The papers, all of which have been submitted to peer-reviewed academic journals, form part of this thesis and can be found in the Appendices. The course was originally developed around Donati’s (2011) relational sociology and Gergen’s (2009) relational education theory. Throughout my praxis process, I expanded my theoretical influences as called for by the research and teaching practice. The journey behind my first PhD paper, (Towards) Sound research practice: Podcast-building as modeling relational sensibilities at the water-climate change nexus in Cape Town, began when I officially started my doctoral studies in early 2018. The paper was co-authored with a fellow PhD scholar from Rhodes University’s Environmental Learning Research Centre (ELRC), Anna James. It presents an experimental arts-based methodology we co-developed for doing contextual profiling by building a socially-engaged podcast series, called Day One, to explore the lived experiences of the Cape Town water crisis of 2018. It includes my initial tool of analysis for exploring how the curriculum might cultivate relational sensibilities and abilities towards reconciliation. The podcast pedagogy offered opportunities to develop some relational learning processes. The analytical tool was developed from cross-referencing reconciliation and relational educational theories. This paper also incorporated theories in relational solidarity and social movement learning. The podcast episodes included personal narratives that, in turn, revealed diverse ideologies and polarisations in the water situation. Working with the audio medium highlighted possibilities for creating and shifting affective relations. Recording and editing soundscapes of waterbodies began explorations of the agential qualities of water. These were foundational dynamics to explore in building the reconciliation curriculum. The paper is published in the International Journal of New Media, Technology, and the Arts (2019, Volume14, Issue1). My second PhD paper, A media arts-based praxis process of building towards a relational model of curriculum oriented towards reconciliation through water justice, presents my methodology for and analysis of a pilot course I co-designed and taught at the Emily Carr University of Art + Design (ECUAD) in Vancouver, Canada in 2018. This course served as contextual profiling around the water situation in Vancouver. The course was offered in partnership with a science-based environmental non-profit called the David Suzuki Foundation and an Indigenous-led post-secondary school called the Native Education College. The course’s public event was hosted at the Beaty Biodiversity Museum. At this stage, I was introduced to Cree/Métis filmmaker, Gregory Coyes, and his Indigenous cinematic narrative approach known as Slow Media. Integrating slow media into video mapping assignments presented exciting possibilities for shifting views and valuing of water. This was the stage at which my concept of reconciliation expanded to explicitly include nonhumans. I applied my initial analytical tool to the curriculum here, which revealed the three most prominent relational sensibilities and abilities towards reconciliation cultivated by students through the course: (1) knowledge ecologies; (2) a hopeful social imaginary; and (3) embodied ways of knowing. I began to make connections between the curriculum and Mi’kmaq elder Albert Marshall’s concept of ‘Two-Eyed-Seeing’, and expanded the notion to ‘Three-Eyed-Seeing’ to include artistic approaches. Deeply inspired by Bekerman and Zembylas’s (2012) Teaching Contested Narratives, I began to see the growing importance of the narrative aspects of reconciliation education. The paper is published in the University of Pretoria’s Journal of Decolonising Disciplines (2021, Volume 1, Issue 2). My third PhD paper, Water as artist-collaborator: Posthumanism and reconciliation in relational media arts-based education, presents a 2019 iteration of the curriculum at ECUAD in Vancouver, and illustrates my shift to include posthuman theories in my analysis. This course was offered in affiliation with the David Suzuki Foundation, and in collaboration with the Native Education College. The culminating public event was hosted by the Beaty Biodiversity Museum. Decentring the human in this data analysis better supported my research and curricular aims. The strong technoculture of the media arts-based curriculum fits well with many posthuman concepts. This posthuman reading of the course and data enabled me to see what changes were emerging through student-water-technology intra-actions, and how these supported relations towards reconciliation as well as water justice. Most notable of these changes was the emergence of water’s agential qualities, specifically of water as becoming collaborator in artistic/knowledge co-production, where students think with water. I argued this contributes to reconciliation by decentring the human, enabling relations in which power is more equal, and where there are greater possibilities for mutual responsibility between related entities. This is where I developed the concept of audio/video as relational texts, supporting the creating and shifting of affective relations more than the monumentalised verbal/written knowledge of traditional universities. This is also where I realised that relational work towards reconciliation would require engaging with the hidden curriculum of institutions. The paper is published in the journal Reconceptualizing Educational Research Methodology (2021, Volume 12, Issue 1), as part of a special issue on Posthuman Conceptions of Change in Empirical Educational Research. My fourth PhD paper, originally entitled Making waveforms: Implicit knowledge representation through video water narratives as decolonising practice towards reconciliation in South Africa’s higher education, presents an analysis of the 2019 iteration of the curriculum in South Africa. I co-designed and led a course called Making Waveforms at the University of Cape Town’s Future Water Institute (FWI) in collaboration with Rhodes University. The course was co-designed/facilitated with FWI’s Research Fellow Amber Abrams, who also co-authored this paper. The course’s public event was hosted by a non-profit organisation called the Tshisimani Centre for Activist Education. This paper explored the ways that non-verbalisable, implicit learning –understood as part of many non-Euro/Western ways of knowing– takes place in the Making Waveforms course and how this influenced water-specific climate behaviours while contributing to decolonised reconciliation practice for higher education institutions. Drawing on theories of implicit and explicit knowledge, we first showed how implicit learning primarily took place through: 1) site-specific audio/video mapping of water bodies; 2) meetings with Knowledge Keepers; and 3) an interactive public screening event. We highlighted how this non-verbalisable learning produced feelings of empathy for diverse peoples and waterways, as well as aesthetic appreciation of water, and how this can contribute to more response-able water behaviours. This, we argued, supported the valuing of implicit knowledge within a traditional educational setting, thereby pluralising knowledge, and was key to reconciliation/decolonisation in higher education. Iterating the curriculum for the South African context emphasised the importance of context-specificity of the course overall, and also of the relational work embedded in the curriculum. This paper is under review by the University of Toronto’s journal Curriculum Inquiry (CI). Following receipt of CI's internal review process, the title of the paper has since been updated to Non-verbalisable, implicit knowledge through cellphilms as decolonised reconciliation practice towards response-able water behaviours in South Africa. Through reflective analysis of my four papers, I developed a concept for an Anatomy of Decoloniz/sed Curriculum consisting of five key parts: 1) relationality; 2) multimodality; 3) narratives/counter-narratives; 4) context-specificity; and 5) unhidden curriculum. Four meta reflections have been included in this thesis, each corresponding with one of the four papers, and presented chronologically according to the stage of the praxis process with which they correspond. In these meta reflections, I applied Kolb’s (1984) Experiential Learning Cycle model for reflective writing, based on the premise that through experiences we can expand our understanding, and included four key stages: 1) concrete experience; 2) reflective observation; 3) abstract conceptualisation; and 4) active experimentation. For the concrete experience, I provided a thick description of my process in writing the paper, as well as aspects of the phase in my praxis process that was the focus of the paper, not included in but relevant to the paper. For the reflective observation, I identified any aspects of the experience that were new to me and which therefore presented opportunities for me to learn. For the abstract conceptualisation, I critically analysed my concrete experience and reflective observation to determine which, if any, of the five key parts of the Anatomy of Decoloniz/sed Curriculum that I outline in my introduction relate to this phase of my PhD praxis process. For the active experimentation, I made conclusions about the extent to which this phase of my PhD embraced decoloniality in practice, and built on this new understanding to make recommendations for myself and others committed to the decolonial project as part of my contribution to knowledge. These meta reflections also invite readers to follow my personal narrative of becoming-with water, meaning my transformation from being water illiterate to embracing a ‘watershed mind’ (Wong,2011). Multimodality, which I propose as a key part of an Anatomy of Decoloniz/sed Curriculum, is embedded in the representational aspects of this thesis. The courses I co-designed and taught as part of this project resulted in the creation of 20 short student films. My contextual profiling involved a podcast methodology that was ongoing throughout my study, as a model of decolonised research-communication-education-action at the water-climate change nexus. This methodology resulted in the creation of four Day One podcast episodes, co-produced with a PhD colleague, Anna James. Some of these episodes are available in all three main languages of Cape Town (Xhosa, Afrikaans, and English). I evolved the podcast methodology in a later stage of my praxis process as a form of member checking with contributors involved in various stages and aspects of the research. Once the four papers were written, I created a series of four short videos called In the Flow, with each video representing a translation of one of the four papers. I invited various contributors of the research project to either watch one or more of the In the Flow videos and/or read one or more of the academic papers, and then to respond in a Zoom call with me. The responses were then shared publicly in a series of seven Climate for Changing Lenses podcast episodes. Parts of these are included in a final song/music video called Please Don’t Blow It. A Climate for Changing Lenses website was created to host all of this multimedia content that forms part of this thesis. A link to this website is provided in the Introduction section of this thesis. My research contributes to the advancement of knowledge in the areas of relational and reconciliation pedagogy, decolonising higher education, arts-based teaching, learning and research methodologies and the water-climate change nexus. This, we argued, supported the valuing of implicit knowledge within a traditional educational setting, thereby pluralising knowledge, and was key to reconciliation/decolonisation in higher education. Iterating the curriculum for the South African context emphasised the importance of context-specificity of the course overall, and also of the relational work embedded in the curriculum. This paper is under review by the University of Toronto’s journal Curriculum Inquiry (CI). Following receipt of CI's internal review process, the title of the paper has since been updated to Non-verbalisable, implicit knowledge through cellphilms as decolonised reconciliation practice towards response-able water behaviours in South Africa. Through reflective analysis of my four papers, I developed a concept for an Anatomy of Decoloniz/sed Curriculum consisting of five key parts: 1) relationality; 2) multimodality; 3) narratives/counter-narratives; 4) context-specificity; and 5) unhidden curriculum. Four meta reflections have been included in this thesis, each corresponding with one of the four papers, and presented chronologically according to the stage of the praxis process with which they correspond. In these meta reflections, I applied Kolb’s (1984) Experiential Learning Cycle model for reflective writing, based on the premise that through experiences we can expand our understanding, and included four key stages: 1) concrete experience; 2) reflective observation; 3) abstract conceptualisation; and 4) active experimentation. For the concrete experience, I provided a thick description of my process in writing the paper, as well as aspects of the phase in my praxis process that was the focus of the paper, not included in but relevant to the paper. For the reflective observation, I identified any aspects of the experience that were new to me and which therefore presented opportunities for me to learn. For the abstract conceptualisation, I critically analysed my concrete experience and reflective observation to determine which, if any, of the five key parts of the Anatomy of Decoloniz/sed Curriculum that I outline in my introduction relate to this phase of my PhD praxis process. For the active experimentation, I made conclusions about the extent to which this phase of my PhD embraced decoloniality in practice, and built on this new understanding to make recommendations for myself and others committed to the decolonial project as part of my contribution to knowledge. These meta reflections also invite readers to follow my personal narrative of becoming-with water, meaning my transformation from being water illiterate to embracing a ‘watershed mind’ (Wong,2011). Multimodality, which I propose as a key part of an Anatomy of Decoloniz/sed Curriculum, is embedded in the representational aspects of this thesis. The courses I co-designed and taught as part of this project resulted in the creation of 20 short student films. My contextual profiling involved a podcast methodology that was ongoing throughout my study, as a model of decolonised research-communication-education-action at the water-climate change nexus. This methodology resulted in the creation of four Day One podcast episodes, co-produced with a PhD colleague, Anna James. Some of these episodes are available in all three main languages of Cape Town (Xhosa, Afrikaans, and English). I evolved the podcast methodology in a later stage of my praxis process as a form of member checking with contributors involved in various stages and aspects of the research. Once the four papers were written, I created a series of four short videos called In the Flow, with each video representing a translation of one of the four papers. I invited various contributors of the research project to either watch one or more of the In the Flow videos and/or read one or more of the academic papers, and then to respond in a Zoom call with me. The responses were then shared publicly in a series of seven Climate for Changing Lenses podcast episodes. Parts of these are included in a final song/music video called Please Don’t Blow It. A Climate for Changing Lenses website was created to host all of this multimedia content that forms part of this thesis. A link to this website is provided in the Introduction section of this thesis. My research contributes to the advancement of knowledge in the areas of relational and reconciliation pedagogy, decolonising higher education, arts-based teaching, learning and research methodologies and the water-climate change nexus. A Pedagogy of Love: reflections on 25 years of informal vocational education and training practices in the commercial fishing industry in South Africa

Fourteen interventions took place during this time which involved several hundred sea-going employees who are disadvantaged by low levels of literacies and low/no Internet Computer Technology connectivity. The Production Programme was chosen as the focus of study and draws upon the influence of the other thirteen programmes. The purpose of the Production Programme was to teach technical fish processing skills to the production management teams and factory hands who work in the factories on board the vessels. The programme ran for five years and evolved through three distinct generations. At the heart of this work lies the question: ‘What made these programmes successful?’ This is an intra-programmatic study and seeks to identify the pedagogical practices which promoted or confounded the efficacy of the Production Programme; and based upon this understanding how such programmes could be improved, transferred, and taken to scale. These questions are both descriptive and explorative in nature. IVET is regarded as training which takes place outside of the formal South African National Qualifications Framework. This work is relevant because approximately 80% of sea-going staff neither finish school, nor get the opportunity of Post School Education and Training by going to a university, a technical institution, or a community college. This statistic is reflected in the general population (Department of Higher Education and Training [DHET], 2022). There is a fine line between being employed and unemployed for people working at this level in the formal economy which makes this project relevant to youth or persons who are ‘not in employment, education or training’ too. This means that for most South African adults IVET presents a significant opportunity for post-school education. Therefore, it is important to answer the questions raised above to rapidly improve inclusion of the majority of South Africans into meaningful education which improves livelihoods. The Theory of Practice Architectures (TPA), from the collection of social-material practice theories, is the conceptual framework for this thesis. The smallest unit of analysis of TPA are practices which may be ‘sayings/thinkings’, ‘doings’, or ‘relatings’. These practices bundle together into practice arrangements and form practice architectures. The reason that TPA was chosen was that practices were the only data available as we (learners, managers, facilitators, and me) knew what we had said/thought or done, and we were aware of the relationships between us over the years as the fourteen interventions played out. Under conventional research circumstances data would be collected in real time, however in this project, most of the data is historical. In addition to the fundamental building blocks of TPA, the theory is embedded in a Theory of Education. There have also been contemporary enhancements to the TPA which were significant to this study, for example, the Ecologies of Practices, a Trellis of Practices which Support Professional Learning, Middle Leadership, Travelling Practices, and moves towards transformative or transgressive education using the TPA in IVET. This is empirical, qualitative research and an ethnographic case study was chosen as the research design which is a methodology particularly suited to answering both descriptive and explorative questions. Nine methods for data collection were used, namely an historical reflective narrative; two focused-group interviews; three individual interviews; four Whatsapp videos; one WhatsApp voice note; two mobisodes; ten questionnaires; 29 documents; and 16 photographs. Because this data was collected under Covid-19 pandemic conditions, two conceptions were employed to guide the generation of data under these uncertain and constrained conditions. These were firstly, the ‘methodology of chance’ which allowed for a ‘methodological agility’; and secondly, the idea of ‘information power’ which is helpful in deciding on how much data is enough. In order to be explicit concerning a key research activity, the approach and method used to review the literature is explicated. Key practices were identified in the data set and described; and then the data was analysed using heuristics provided by TPA theorists. Seven Tables of Invention were used to synthesise the data arising from these practices. An eighth Table of Invention was used to synthesise all the practices and practice arrangements characterised; and to indicate how these evolved over time and space. The data description and analysis is supported by eight Analytic Memos, a comprehensive Data Code Table and a hyperlink to a data repository which provides access to oral and video material. The findings distinguished five key practices and practice arrangements which were: Practices of the Creation of Courseware; Practices of Teaching and Learning; Practices of Assessment; Practices of Love; Practices of Management. The thesis title is reflective of the impact which love has upon the pedagogical process of IVET. Based upon the analysis and synthesis of the corpus of data, practices which either promoted or confounded the Production Programme became visible; it is these insights which inform future improvements to similar programmes. Emanating from these findings, two overarching practice architectures (PA) were identified which restrained the Production Programme in the same manner that the banks of a river restrain a river, and yet simultaneously, are changed by the river over time. These are the PA of Methodology and Methods and the PA of Maturing Ecologies of Practices. The inferences drawn from the data were achieved through the use of deductive, inductive, and abductive reasoning. My claim to new knowledge is a lamination of a practical contribution on one side of the coin, and a theoretical contribution on the other side of the coin. The PA of Methodology and Methods provides a practical mechanism to create, deliver and assess IVET. This is done by explicating the three practice architectures which constitute the overarching PA of Methodology and Methods which are, the PA of Informality; the PA of Range, and the PA of Relationality. An IVET programme constitutes Ecologies of Practices. The theoretical conception of the overarching PA of Maturing Ecologies of Practices provides a conceptual tool which enables the transferring and scaling of IVET programmes. It does this by providing theoretical indicators to establish the ‘state’ of an IVET programme as it matures over time from a pioneer state to a settler state. An IVET educator can then work towards creating a PA which is conducive for a mature ecologies of practices to form; and the programme can then be transferred and/or taken to scale, if this is desirable in the particular context. The power of my claim to new knowledge does not lie on one side or the other of the coin, but in the lamination of the practical and theoretical contributions put to use in the service of IVET. This thesis concludes with a number of theoretical and practical recommendations which are loosely grouped according to ‘sayings/thinkings’, ‘doings’ and ‘relatings’ in deference to the value of TPA to this thesis. An urgency is conveyed in these recommendations as there is an immediate need to improve the livelihoods of ordinary South Africans. An investigation into yeast-baculovirus synergism for the improved control of Thaumatotibia leucotreta, an economically important pest of citrus

- Description: A mutualistic association between Cydia pomonella and yeasts belonging to the genus Metschnikowia has previously been demonstrated. Larval feeding galleries inoculated with M. andauensis, reduced larval mortality and enhanced larval development. Additionally, adult C. pomonella female oviposition preference was also shown to be influenced by the volatiles produced by M. andauensis. This mutualistic relationship was manipulated for biological control purposes, by combining M. pulcherrima with the baculovirus Cydia pomonella granulovirus. The combination of M. pulcherrima with brown cane sugar and CpGV in laboratory assays and field trials resulted in a significant increase in larval mortality. A similar observation was made when M. pulcherrima was substituted for Saccharomyces cerevisiae. This indicates that yeasts harbour the potential for use in biological control, especially when combined with other well-established biocontrol methods. Thaumatotibia leucotreta is a phytophagous insect endemic to southern Africa. It is highly significant to the South African citrus industry due to its classification as a phytosanitary pest by most international markets. An integrated pest management programme has been implemented to control T. leucotreta. The baculovirus Cryptophlebia leucotreta granulovirus forms one component of this programme and is highly effective. In this study, we proposed to determine which yeast species occur naturally in the gut of T. leucotreta larvae and to examine whether any of the isolated yeast species, when combined with the CrleGV-SA, enhance its effectiveness. Firstly, Navel oranges infested with T. leucotreta larvae were collected from geographically distinct citrus-producing regions across South Africa. This led to the isolation and identification of six yeast species from the gut of T. leucotreta larvae via PCR amplification and sequencing of the internal transcribed spacer region and D1/D2 domain of the large subunit. Six yeast species were identified, viz. Meyerozyma guilliermondii, Hanseniaspora uvarum, Clavispora lusitaniae, Kluyveromyces marxianus, Pichia kudriavzevii and Pichia kluyveri. Additionally, Saccharomyces cerevisiae was included as a control in all trials due to its commercial availability and use in the artificial diet used to rear T. leucotreta. Secondly, larval development and attraction assays were conducted with the isolated yeast species. Thaumatotibia leucotreta larvae that fed on Navel oranges inoculated with M. guilliermondii, P. kluyveri, H. uvarum, and S. cerevisiae had accelerated developmental periods and reduced mortality rates. Additionally, it was demonstrated that T. leucotreta neonates were attracted to YPD broth cultures inoculated with P. kluyveri, H. uvarum, P. kudriavzevii and K. marxianus for feeding. Thirdly, oviposition preference assays were conducted with adult T. leucotreta females to determine whether the isolated yeast species influence their egg-laying in two-choice and multiple-choice tests. Navel oranges were inoculated with a specific yeast isolate, and mated adult females were left to oviposit. Meyerozyma guilliermondii, P. kudriavzevii and H. uvarum were shown to influence adult T. leucotreta female oviposition preference in two-choice tests. However, multiple-choice tests using the aforementioned yeast species did not mimic these results. Lastly, a series of detached fruit bioassays were performed to determine the optimal yeast:virus ratio, test all isolated yeast species in combination with CrleGV-SA and to further enhance yeast/virus formulation through the addition of an adjuvant and surfactant. CrleGV-SA was applied at a lethal concentration that would kill 50 % of T. leucotreta larvae. The optimal yeast concentration to use alongside CrleGV-SA was determined. Pichia kluyveri, P. kudriavzevii, K. marxianus and S. cerevisiae in combination with CrleGV-SA increased larval mortality compared to CrleGV-SA alone. The inclusion of molasses and BREAK-THRU® S 240 to P. kudriavzevii and S. cerevisiae plus CrleGV-SA formulations greatly enhanced their efficacy. Additionally, semi-field trials were initiated using P. kudriavzevii and S. cerevisiae, with promising preliminary results being obtained, although more replicates need to be performed. - Description: Inasmuch as coal remains the linchpin for the generation of electricity and liquid petroleum products in South Africa, hydrocarbon waste and coal discard will continue to pose a threat to the environment. Therefore, the onus is on the associated industries to develop and implement efficient and sustainable strategies to mitigate the negative impacts of energy generating activities on the environment. Most conventional efforts in this regard, although successful for soil repair and the initiation of vegetation, have been deemed unsustainable. In an effort to find a sustainable remediation strategy a novel technology termed “FungCoal” was conceptualized and patented as a strategy for the rehabilitation of open cast coal mines, carbonaceous-rich spoils and coal wastes. This biotechnology, which exploits plant-fungal mutualism to achieve effective biodegradation of coal on discard dumps and the breakdown of the carbonaceous component in spoils, promotes revegetation to facilitate rehabilitation of mining-disturbed land. However, one limiting factor of the FungCoal bioprocess is that it requires oxidized weathered coal, a highly complex and variable resource for use as a co-substrate, for growth and proliferation of the coal degrading microorganisms. To fully exploit the potential of plant-fungal mutualism and its interaction for use in the remediation of coal contaminated soils, this study investigated the proposed relationship between plant roots, root exudate and the coal degrading fungus “Aspergillus sp.” (previously Neosartorya fischeri) strain 84 in more detail, in an effort to gain further insight into the mechanisms underpinning plant-fungal mutualism as a strategy for re-vegetation of coal discard dumps and the rehabilitation of hydrocarbon-contaminated soil using the FungCoal approach. A pot-on-beaker (PoB) method was developed for the easy cultivation and collection of extracellular polymeric substance (EPS)-containing exudates from Zea mays L. (maize) and Abelmuschus esculentus (okra). Characterisation of the EPS material from these exudates was carried out using a combination of physicochemical and biochemical methods. The results from analysis of phenolics and indoles showed that exudates contain some form of indoles and phenolic compounds, although in little proportions, which may fulfil a signalling function, responsible for attracting soil microorganisms into the rhizosphere. Spectroscopic analysis of the exudates using FT-IR revealed vibrations corresponding to functional groups of alkanes, alkenes, alkynes, and carboxylic acids. These compounds likely provide an easily accessible source of carbon to soil microorganisms and are also a better alternative to the poly-aromatics which are an inherent component locked-up in the supposed recalcitrant coal material. The results from biochemical analyses also revealed the presence of carbohydrate, proteins, lipids, and low amounts of α-amino-nitrogen in the EPS of maize and okra. These components of EPS are all essential for the stimulation of enzymatic activities in soil microorganisms and, which may in turn aid biodegradation. The action of the root EPS from maize was further tested on three coal-degrading fungal isolates identified as Aspergillus strain ECCN 84, Aspergillus strain ECCN 225 and Penicillium strain ECCN 243 for manganese peroxidase (MnP) and laccase (LAC) activities. The results revealed that the Aspergillus species, strains ECCN 84 and ECCN 225, showed with or without EPS, observable black halos surrounding each of the colonies after 7d incubation indicative of positive MnP activity, while no activity was observed for the Penicillium sp. strain ECCN 243. Analysis for LAC revealed little or no activity in any of the coal degrading fungi following addition of pulverized coal to the growth medium. Interestingly, the addition of EPS-containing exudate to the coal-containing medium resulted in increased LAC activity for all fungal isolates. This finding affirmed the positive contribution of EPS to extracellular LAC activity, purported as an important enzyme in the coal biodegradation process. Finally, the impact of plant-derived exudate on the colonisation and biodegradation of coal was investigated in situ using rhizoboxes, to simulate a coal environment, and was carried out for 16 weeks. Microscopic examination of coal samples after termination of the experiment showed fungal proliferation and attachment to coal particles. All of the rhizoboxes that contained plants had higher medium pH and EC, and the concentration of phenolics, indoles and humic acids was greater than that of control treatments. These observations indicated better rhizosphere colonisation, substrate biodegradation and humification. Therefore, root exudate appears to play a significant role in coordination of soil microorganisms within the rhizosphere and likely serves both as a scaffold for rhizospheric interactions by providing microorganisms with accessible carbon and as a likely ‘trigger’ for induction of coal-degrading enzymes such as fungal LAC for mobilisation of recalcitrant carbon. This study has shown that EPS exuded from roots of Zea mays together with coal degrading fungus Aspergillus strain ECCN 84 can alkalinise the coal substrate and facilitate introduction of oxygen, possibly as a result of increased laccase activity, and increase availability of nutrients (as indicated by higher EC) in a coal-polluted rhizosphere, to provide plants and their associated mycorrhizae and presumably other beneficial microorganisms a more mesic environment for sustained phytoremediation with enhanced rehabilitation potential. Towards a biological profile for South African perinatal remains: osteological and genetic perspectives

- Description: Forensic identification of abandoned and suspected infanticide cases admitted to the South African Forensic Pathology Services is often impossible due to decomposition of the remains. In these cases, investigation of suspected criminal activity is almost never pursued. Ancillary tests in the form of anthropological and molecular analyses can assist with the forensic identification of perinatal remains. To provide fundamental information about bone development of perinatal skeleton, osteological and genetic techniques focusing on the pars basilaris, pars lateralis, sternal rib and left femur were used. Samples were obtained from unidentified and unclaimed remains originating from the Johannesburg Forensic Pathology Service (JFPS). To provide a biological age to individuals in the collection, dental aging was used to categorize remains for comparisons with anthropological and molecular data. A molecular protocol was designed to sex individuals using the X-linked G6PD and Y-linked SRY genes. Bone development was studied using osteometric and morphological data of dry bone remains coupled with bone mineral density analysis (Micro-CT). The methylation levels of CpG rich sites within the promoter region of selected bone-associated genes were incorporated to examine silencing of genes during development. Osteological results support the use of the pars basilaris, pars lateralis and femur for age-at-death estimations as well as provide the foundation for dry bone aging criteria for South African individuals. Data compared with established skeletal aging standards indicated developmental differences between populations. Through the use of animal models and the perinatal sternal rib tissue, insights and precautions into the use of post mortem bone derived RNA for forensic applications is communicated. Taxonomic, ecological and biogeographic re-evaluation of temperate stream fishes in Southern Africa: a case study of the enteromius anoplus complex

- Description: Worldwide, cryptic species, which comprise two or more distinct species that are classified as one due to their morphological similarity, are more common than previously thought. In South Africa, the chubbyhead barb, Enteromius anoplus (Weber, 1897), is the most widespread freshwater fish species. The widespread occurrence of this species across different river basins raised questions on its taxonomic status due to the likely occurrence of hidden diversity. This further raised the need to evaluate the ecological patterns associated with likely occurrence of such hidden diversity. Phylogenetic analyses of the cytochrome b gene of E. anoplus populations across South Africa revealed the occurrence of four genetically distinct species. These findings supported the restoration Enteromius cernuus (Barnard 1943) stat. rev. from the Olifants River system; the restoration of Enteromius anoplus sensu stricto (Weber 1897) stat. rev. from the Gouritz River system; the redescription of Enteromius oraniensis (Barnard 1943) stat. rev. from the Orange River system; and the description of a new species, Enteromius mandelai sp. nov. from the Eastern Cape rivers. Analyses of morphometric and meristic data revealed that these species were differentiated based on barbel length and number of lateral line scales. Ecological studies on Enteromius mandelai were conducted within headwater streams of major river systems in the Eastern Cape region. These headwater streams had different catchment characteristics ranging from near-natural to anthropogenically-perturbed mainly due to agriculture-related disturbances and invasion of riparian zones by non-native plants. Stable isotope analysis provided evidence of trophic plasticity for E. mandelai across the different headwater streams. Specifically, E. mandelai exhibited large isotopic niches in agriculture-disturbed headwater streams, whereas small isotopic niches were observed in headwater streams invaded by non-native plants. Because of the evidence of trophic plasticity for E. mandelai, this study further hypothesized that this species would exhibit variable environmental niche patterns related to its distribution, abundance and the presence of contra-specifics due to different abiotic factors across its range. Enteromius mandelai was generally most abundant at sites with high total dissolved solids and sand substratum, which was characteristic of large pool habitats, and least abundant at sites with high turbidity. In the Great Fish River, E. mandelai occurred as the only species in most headwater sites, whereas in the Keiskamma and Buffalo River systems, it co-occurred with Amatolacypris trevelyani and Sandelia bainsii. Enteromius mandelai was, nevertheless, found to be ubiquitous together with having wide environmental niches, and exhibited no discernible patterns in its habitat associations. The taxonomic re-evaluation of the E. anoplus complex, which provided evidence of the existence of new species, has implications for the biogeographic zonation of the ichthyofauna of the Cape Fold Ecoregion (CFE), Amathole-Winterberg Highlands Ecoregion (AWHE) and the Southern Temperate Highveld Ecoregion (STHE). For example, the currently recognised southern limit of the STHE, which encompasses the AWHE, is based on the occurrence of E. anoplus, Enteromius pallidus and Labeo umbratus. However, evidence from this and other recent studies indicate the occurrence of either new species or distinct lineages in the southern-most section of the STHE, which is consistent with the patterns observed in the CFE. Comparison of taxonomic composition showed that species in the eastern part of the CFE were similar to species in the AWHE and the southern-most section of the STHE. Exploring the structural integrity of a picornavirus capsid

- Description: Picornaviruses are a diverse family of small RNA viruses that cause a broad range of human and veterinary diseases. Despite decades of research into the molecular biology of these pathogens, no antivirals and few vaccines are commercially available for the treatment and prevention of picornavirus infections. The capsids of these non-enveloped viruses are involved in many important aspects of the picornavirus lifecycle, such as cell attachment and entry, uncoating, and protection of the viral RNA. Although the structures of many picornavirus capsids have been solved, a broader understanding of the molecular determinants that are required for structural integrity and stability is imperative for an improved understanding of the basic biology of these viruses, and for designing effective control strategies. Collectively, this thesis aims to elucidate the molecular determinants of structural stability and integrity in the Theiler’s murine encephalomyelitis virus capsid (TMEV). To study the TMEV GDVII capsid using biochemical techniques, neutralising polyclonal antibodies were generated against GDVII particles. The antibodies recognised linear epitopes in the C-terminus of the VP1 protein, but not those present in VP2 or VP3. The VP1 C-terminal residues were mapped to a loop above the putative receptor binding pit on the capsid surface, which prompted an investigation into the potential binding site of the TMEV co-receptor, heparan sulphate. Molecular docking revealed that heparan interacts with residues of the receptor binding pocket, as well as residues of the adjoining VP1 C-terminal loop. These findings suggest that the antibodies neutralise virus infection by preventing attachment of the virus to the co-receptor and possibly the unknown primary receptor. Few studies have identified the specific residues and interactions at subunit interfaces that significantly contribute to picornavirus capsid stability, assembly, and function. A novel in-silico screen was developed for the prediction of hotspot residues at protein-protein interfaces of a virus capsid. This screen can be applied to elucidate the residues that contribute significantly to the intraprotomer, interprotomer and interpentamer interfaces of any picornavirus capsid, on condition that the structure of the virus is available. The screen was applied to TMEV GDVII resulting in the identification of hotspots, several of which correspond to residues that are known to be important for aspects of the virus lifecycle, such as those that contribute to pH stability or form part of receptor binding sites. This observation suggests that residues involved in specific capsid functions may also play a role in capsid stability. Many of the residues identified as hotspots in TMEV corresponded to those required for assembly, uncoating, and virus growth in representative picornaviruses from various genera, suggesting that the residues that regulate capsid stability may be somewhat conserved across the family. Hotspots identified at the interpentamer interfaces of TMEV were individually substituted to alanine to further explore their importance to the TMEV lifecycle. All the amino acid substitutions prevented completion of the virus lifecycle as no CPE was observed following transfection of susceptible cells. Immunofluorescence experiments demonstrated that virus protein synthesis and RNA replication were not inhibited by substitution of the hotspot residues, but that infectivity was severely impeded. This confirmed that the residues were required for some aspect of the virus lifecycle, such as capsid assembly, or were critical for maintaining the conformational stability of the TMEV particles. Virus capsids become unstable and are prone to dissociation under certain conditions such as extreme pH and non-physiological temperatures. The thermostability of TMEV was explored by selecting GDVII virions with improved thermal tolerance through serial passage and heat exposure. Thermostable virions that could tolerate temperatures above 57 °C had reduced infective titres compared to the wild type TMEV suggesting that the virus adapted to thermal stress at the expense of viral fitness. Sequencing the capsid encoding regions of the mutant virions revealed a pair of amino acid substitutions that were present in all mutants. Additional substitutions that were unique to viruses selected at different temperatures were also identified. Most of the substitutions were located within the intraprotomer interfaces of the virus, unlike previous studies on enteroviruses where mutations were mostly localised to the receptor binding pocket. This thesis provides the first analysis of the structural determinants of TMEV capsid stability. - Description: Picornaviruses are a diverse family of small RNA viruses that cause a broad range of human and veterinary diseases. Despite decades of research into the molecular biology of these pathogens, no antivirals and few vaccines are commercially available for the treatment and prevention of picornavirus infections. The capsids of these non-enveloped viruses are involved in many important aspects of the picornavirus lifecycle, such as cell attachment and entry, uncoating, and protection of the viral RNA. Although the structures of many picornavirus capsids have been solved, a broader understanding of the molecular determinants that are required for structural integrity and stability is imperative for an improved understanding of the basic biology of these viruses, and for designing effective control strategies. Collectively, this thesis aims to elucidate the molecular determinants of structural stability and integrity in the Theiler’s murine encephalomyelitis virus capsid (TMEV). To study the TMEV GDVII capsid using biochemical techniques, neutralising polyclonal antibodies were generated against GDVII particles. The antibodies recognised linear epitopes in the C-terminus of the VP1 protein, but not those present in VP2 or VP3. The VP1 C-terminal residues were mapped to a loop above the putative receptor binding pit on the capsid surface, which prompted an investigation into the potential binding site of the TMEV co-receptor, heparan sulphate. Molecular docking revealed that heparan interacts with residues of the receptor binding pocket, as well as residues of the adjoining VP1 C-terminal loop. These findings suggest that the antibodies neutralise virus infection by preventing attachment of the virus to the co-receptor and possibly the unknown primary receptor. Few studies have identified the specific residues and interactions at subunit interfaces that significantly contribute to picornavirus capsid stability, assembly, and function. A novel in-silico screen was developed for the prediction of hotspot residues at protein-protein interfaces of a virus capsid. This screen can be applied to elucidate the residues that contribute significantly to the intraprotomer, interprotomer and interpentamer interfaces of any picornavirus capsid, on condition that the structure of the virus is available. The screen was applied to TMEV GDVII resulting in the identification of hotspots, several of which correspond to residues that are known to be important for aspects of the virus lifecycle, such as those that contribute to pH stability or form part of receptor binding sites. This observation suggests that residues involved in specific capsid functions may also play a role in capsid stability. Many of the residues identified as hotspots in TMEV corresponded to those required for assembly, uncoating, and virus growth in representative picornaviruses from various genera, suggesting that the residues that regulate capsid stability may be somewhat conserved across the family. Hotspots identified at the interpentamer interfaces of TMEV were individually substituted to alanine to further explore their importance to the TMEV lifecycle. All the amino acid substitutions prevented completion of the virus lifecycle as no CPE was observed following transfection of susceptible cells. Deficiencies in any of these proteins causes the failure of the structure and assembly and disassembly of the nuclear envelope, which disrupts chromosome segregation and nuclear compartmentalization that are both associated with disease. Emerin also localizes in the cytoplasm where it is implicated in the structure of the cytoskeleton via interaction with tubulin and actin and thus its deficiency may equally contribute to the collapse of the cytoskeleton. The Hsp70-Hsp90 organising protein (Hop) functions as a cochaperone for entry of client proteins into the Hsp90 folding cycle. Hop is upregulated in cancer and regulates a number of cell biology processes via interactions with proteins independently of Hsp90. In a previous study using global whole cell mass spectrometry, emerin was shown to be the most significantly down regulated protein in Hop depleted cell lysates. In this current study, it was postulated that emerin interacts with Hop, and this interaction regulates the stability, and level of emerin in the nucleus which impacts on the structure of the nuclear envelope. We used HEK293T cell lines stably expressing shRNA against Hop, emerin and a non-targeting control alongside the over expression of Hop in HEK293 cells to determine the effect of Hop levels on emerin expression and vice versa via Western blotting. The effect of Hop on the localization of emerin was assessed via subcellullar fractionation and confocal microscopy, while the impact on the structure of the nucleus was determined by transmission electron microscopy (TEM). We established that the depletion of Hop using shRNA and the over expression of Hop both result in the proteasomal and lysosomal degradation of emerin. Co-immunoprecipitation assays confirmed that Hop and emerin are in a common complex, which was not dependent on the presence of Hsp90. The development, manufacture and evaluation of sustained release gastric-resistant isoniazid and gastroretentive microporous rifampicin microspheres

- Description: According to the World Health Organization Global Tuberculosis (TB) 2017 Report, there were an estimated 10.4 million new TB cases worldwide of which, in 2016, 65 % occurred in men, 28.1 % in women and 6.9 % in children. TB is the ninth leading cause of death globally and is the leading cause due to an infectious organism surpassing HIV/AIDS. Treatment is long-term and the use of a combination of medicines is required for success. The concern related to the use of fixed dose combination products for the treatment of TB is the issue of low bioavailability of rifampicin observed from a number of fixed dose combination (FDC) formulations. The hydrolysis of rifampicin, in acidic media, to form insoluble 3-formyl rifamycin SV contributes to poor bioavailability of rifampicin. The degradation of rifampicin to form this poorly absorbed compound is accelerated in the presence of isoniazid via the reversible formation of isonicotinyl hydrazone is a further factor contributing to the poor bioavailability of rifampicin. Therefore, the development of a novel drug delivery technology that prevents interactions between rifampicin and isoniazid in an acidic medium is required. A Box Behnken design was successfully used for the optimisation of a rapid and accurate stability-indicating gradient elution RP-HPLC method for the simultaneous analysis of isoniazid, pyrazinamide and rifampicin. The method was validated using ICH guidelines and the results indicate it can be used for the rapid analysis of commercially available TB FDC formulations containing the active pharmaceutical ingredients, API. The method is precise, sensitive and has the necessary selectivity for use during formulation development and optimisation studies for a combination of rifampicin, isoniazid and pyrazinamide. Initially formulation activities were undertaken with rifampicin and isoniazid for the development of an approach to enhance the effective delivery of these compounds. The characterisation of rifampicin and isoniazid was undertaken using spectroscopic, thermal and microscopic analysis. The studies revealed that the compounds are crystalline and exhibit distinct characteristic sharp peaks in X-ray diffractograms and Differential Scanning Calorimetry thermograms. The thermograms, 13C Nuclear Magnetic Resonance and Fourier Transform Infrared spectroscopy results identified that rifampicin occurs as the form II polymorph however, as there are no significant biopharmaceutic differences between the polymorphic forms of rifampicin this information was used for identification purposes only. The results were used as baseline data for comparative purposes to monitor changes that may occur when rifampicin and isoniazid are used in formulation development, dosage form manufacture and characterisation activities for a FDC technology designed to deliver both compounds simultaneously. Hydroxypropylmethylcellulose acetate succinate (HPMC-AS) and Eudragit® L100 polymers were successfully used for manufacture of isoniazid loaded gastric-resistant sustained release microspheres using an o/o solvent emulsification and evaporation approach. A Hybrid experimental design was used to investigate the influence of input variables viz., homogenisation speed and amount of HPMC-AS and Eudragit® L100 on gastric-resistance, INH release and encapsulation efficiency. The approach of using coating polymers viz., HPMC-AS and Eudragit® L100, to manufacture gastric resistant sustained release microspheres of isoniazid is unique and was efficient for preventing the release of isoniazid in an acidic environment. Only 0.523 % isoniazid was released from the optimised formulation after 2 h exposure to pH 1.2 0.1 M HCl suggesting there is also the possibility of minimising the accelerated degradation of rifampicin that occurs in the presence of isoniazid in acidic media. The microspheres also exhibited sustained release properties without burst release in pH 6.8 0.1 M phosphate buffer as < 5 % isoniazid was released at 0.5 h and only 11 % isoniazid was released at 2 h. The release of isoniazid was sustained over the entire period of dissolution testing with > 85 % isoniazid released at 24 h, implying that the majority of encapsulated isoniazid would be available for absorption. The manufacturing process resulted in the production of hard spherical particles and particle size analysis revealed that the microspheres ranged between 415.76 ± 76.93 μm and 903.35 ± 197.10 μm in diameter. The microspheres exhibited excellent flow properties attributed to the spherical nature of particles. Carr‟s index (CI) was 4.934 ± 0.775 % and the Hausner ratio (HR) was 1.148 ± 0.033 indicating good packability of the microspheres that would help in achieving weight and content uniformity of capsule dosage units. The manufacturing process however produced a low % yield suggesting that scale up difficulties may be encountered. However the high encapsulation efficiency observed may counter the challenges associated with the low yield. The DSC thermograms and FT Raman spectra of 1:1 mixtures of isoniazid, excipients and the microspheres did not reveal any potential detrimental interactions. Microporous floating sustained release microspheres for the delivery of rifampicin in the stomach have been successfully manufactured using emulsification and a diffusion/evaporation process. A novel approach using solvent mixture of acetone and dichloromethane that has not been reported for the manufacture of rifampicin microspheres was successfully used and resulted in the formation of a stable emulsion and the manufacture of rifampicin-loaded microspheres with uniform characteristics. In addition the manufacturing process was shorter than most other reported methods. A Box-Behnken experimental design was successfully used to study the influence of ethylcellulose, Eudragit® RLPO and d-glucose content on the floating properties, encapsulation efficiency and % yield of microspheres. The optimised formulation did not yield desired floating characteristics as the % buoyancy was low and floating lag times were high. The optimised formulation was modified by addition of NaHCO3 to increase the % buoyancy and reduce the floating lag time. Rifampicin release from the microspheres of the modified batch was 87.10 % at 12 h and the microspheres exhibited a % buoyancy of 87.66 ± 1.28 % (n = 6) and floating lag time of 15 ± 3.2 (n = 6) seconds. The microspheres remained buoyant for up to 12 h and an encapsulation efficiency of 88.26 ± 1.25 % was achieved. SEM images of microspheres following exposure to dissolution fluid revealed that the microspheres had numerous pores on their surface. The mean particle size distribution ranged between 423.19 ± 121.86 μm to 620.07 ± 102.67 μm. The microspheres exhibited similar flow characteristics to isoniazid microspheres with a CI of 1.422 ± 0.074 %, and HR of 1.034 ± 0.002. The excellent flow characteristics indicate that filling of the microspheres into hard gelatin capsules was unlikely to pose a challenge in respect of producing a product with uniform content. Rifampicin-excipient compatibility studies did not reveal any potential or significant interactions suggesting that the excipients used for the manufacture of the microspheres were compatible, although long term stability studies would be required to ascertain this is, indeed the case. The microporous floating sustained release microspheres manufactured in these studies has the potential to increase the bioavailability of rifampicin as they may be retained in the stomach where the solubility of rifampicin is high and from which absorption is best achieved. The degradation of rifampicin after 12 h dissolution testing in pH 1.2 0.1 M HCl in the presence of isoniazid gastric-resistant sustained release microspheres was only 4.44%. These results indicate that the degradation of rifampicin in the presence of isoniazid in acidic media can be overcome by encapsulation of both active pharmaceutical ingredients in a manner that ensure release in different segments of the gastrointestinal tract. The use of sustained release microporous gastroretentive rifampicin microspheres in combination with sustained release isoniazid gastric-resistant microspheres revealed that accelerated degradation of rifampicin in the presence of isoniazid is reduced significantly when using this approach and a FDC of rifampicin and isoniazid microspheres has the potential to improve the bioavailability of rifampicin thereby enhancing therapeutic outcomes. The excellent flow characteristics indicate that filling of the microspheres into hard gelatin capsules was unlikely to pose a challenge in respect of producing a product with uniform content. Rifampicin-excipient compatibility studies did not reveal any potential or significant interactions suggesting that the excipients used for the manufacture of the microspheres were compatible, although long term stability studies would be required to ascertain this is, indeed the case. The microporous floating sustained release microspheres manufactured in these studies has the potential to increase the bioavailability of rifampicin as they may be retained in the stomach where the solubility of rifampicin is high and from which absorption is best achieved. The degradation of rifampicin after 12 h dissolution testing in pH 1.2 0.1 M HCl in the presence of isoniazid gastric-resistant sustained release microspheres was only 4.44%. These results indicate that the degradation of rifampicin in the presence of isoniazid in acidic media can be overcome by encapsulation of both active pharmaceutical ingredients in a manner that ensure release in different segments of the gastrointestinal tract. The use of sustained release microporous gastroretentive rifampicin microspheres in combination with sustained release isoniazid gastric-resistant microspheres revealed that accelerated degradation of rifampicin in the presence of isoniazid is reduced significantly when using this approach and a FDC of rifampicin and isoniazid microspheres has the potential to improve the bioavailability of rifampicin thereby enhancing therapeutic outcomes. Although the clearing successes of WfW are well documented, the programme has focused little attention to the longer-term livelihood impacts of the temporary work and skills training provided to beneficiaries. This study suggests this could be due to a lack of the appropriate indicators to measure these outcomes. Therefore, an evaluation framework for environmental public works projects is proposed, which consists of outcome indicators to track the livelihood impact of the work experience and skills training on the beneficiaries post-participation, since the aim of these EPWP interventions is to improve beneficiaries’ labour market outcomes. The Northern Cape province’s Prosopis mesquite clearing project was used as the case study to develop and test the evaluation framework. The outcome indicators were informed by the key stakeholders’ interviews and the beneficiaries’ survey, specifically since the beneficiaries were well placed to give feedback on the benefits of the work experience and training post-participation. The combined strengths of the Sustainable Livelihoods Approach and the Capability Approach were useful for formulating the outcomes indicators, while the indicators for the inputs, activities and outputs were formulated from the key stakeholder interviews and online EPWP reports. A mixed methods approach was used and primary data were collected through key stakeholder interviews with the Prosopis mesquite clearing project managers and an online survey with some of the beneficiaries. Online EPWP reports and records obtained from WfW were used as secondary data. Data analysis used RStudio, Microsoft Excel and GraphPad Prism. - Description: Urbanisation is occurring on a massive scale globally and even more so in the less developed regions of the Global South including sub-Saharan Africa (SSA). Like other developing regions of the world, urbanisation in SSA is not occurring alongside a corresponding growth in urban economies. Resultantly, it is taking place in tandem with the rising scourge of urban poverty, including food insecurity. While urban food insecurity is a clear challenge in SSA, the challenge has however not been met with equal vigour in policy making and implementation circles and even in academia. Problematically, the urban food security literature often focuses on one element of the food system without giving due attention to other components of the system. Resultantly, broader systemic failures and the dynamics related to the different actors across the system-elements are missed. There has thus been recent calls to embrace urban governance in studying urban food systems, which this study does. The thesis examines the urban food system in Bulawayo (in Zimbabwe) with specific reference to urban governance and household food security to understand sociologically the complex multi-dimensional processes, structures, systems, and practices underpinning the urban food system. As a result of the complex nature of food systems, an eclectic analytical framework is employed encompassing Obeng Odoom’s DED framework, Clapp and Fuchs’ framework of power, Gaventa’s power cube and theories of everyday life derived from de Certeau and Lefebvre. Methodologically, the study is informed by a Critical Realism paradigm which accommodates the convergent mixed methods research design employed. The research strategy employed was that of a survey and case study. Key findings reveal that the Bulawayo food system, from production to consumption, is complex and is nested within broader national and international food systems. Although without a direct and explicit mandate on food security, the local authority is at the centre of urban governance processes as it employs a plethora of strategies to influence the nature of the food system. - Description: Consider, for instance, you and your family have lived around a place where you enjoyed the flora and fauna of the land as well as the natural environment. Fishing and farming were pleasant activities for your family, and anyone in the community not only to survive but thrive. Your neighbours and the people in the community pride themselves so much in their farming abilities and fishing techniques. Suddenly things start taking a different turn because of rising sea level and changing weather pattern. First, your land began to give way because of sweeping erosion, and later the riverbank serving the community starts drying up. Your neighbour could not hold their own in this grim condition as they started relocating. You even witnessed some of your family and friends leaving the community and you later heard that they have become climate refugees. To put it all together, you were told that what you are experiencing is one of climate change effects. You were told that the loss of the place that you once cherished is not an act of God but the result of the reckless flaring of greenhouse gasses harmful to the environment by some powerful but polluting nations. What will you do? This might sound like a fairy tale. In reality, this is the story of the people of Saint Louis Atlantic Coasts in Senegal, captured by the Global Climate Change Alliance Plus Initiative.1 Their situation has been described as ‘living on the edge’ because for centuries, Saint Louis Atlantic Coasts, home to around 230,000 inhabitants, also known as the ‘Venice of Africa’ has been protected from the pounding Atlantic waves by the ‘Langue de Barbarie,’ a narrow, 30 km peninsula at the mouth of the Senegal river. However, the low-lying sandy spit of land along this World Heritage site is itself rapidly disappearing in the present. This is the consequence of a changing climate and other man-made problems such as illegal sand-mining and over-development. Today, the southern part of the Langue de Barbarie is an island, and the village of Doune Baba Dieye is under more than a metre of water. The villagers have become climate refugees, forced to live in temporary camps on the mainland. Not only have they lost their homes, but they have also lost significant cultural heritage like the farming and fishing culture tied to this place. The ethical considerations that emerge from climate change impacts on the world’s cultural heritage are varied. However, it seems not as self-evident in the way that research on climate change ethics has been framed around economic interest and direct threats to human life and other species. Even when they mention climate impacts on heritage sites around the world, those of Africans have been side-lined. For instance, the impact of climate change on small island nations like Tuvalu, Kiribati2 have gained much traction in climate change discourse, but we do not get to see stories of Africans whose cherished cultural heritage are affected by climate despoliation. How do we respond to this intractable challenge? This is a question of justice and, to be more precise, climate justice. Many principles and proposals for climate justice have been put forward, but the insufficient attention to the vulnerabilities or loss of cultural heritage values of Africans, which is a critical aspect of their social realities, make these theories less persuasive on a global level. This thesis, then, fills this gap in the literature by suggesting that the failure to take cognizance of the injustice in neglecting cultural heritage values when dealing with the burden of climate change is the effect of three problems. One, the value of culture is less understood in this environmental age. Hence, cultural values are excluded or made to be secondary in consideration of principles of climate justice. Second, the idea of Personhood has been neglected in climate ethics and climate justice discourse. Yet, this idea of Personhood can be an enabler of climate justice in that a realization of the significance of cultural heritage to the wellness of the human persons in Africa, makes it loss morally reprehensible. Third, those whose cultural heritage is significantly affected do not get represented in the debate about sharing the burden of climate change. This dissertation thereby builds upon the general findings of the past about anthropogenic climate change, its causes and consequences. Adopting a discursive normative framework, I also address the significance of cultural heritage in this contemporary environmental age and discuss the global justice implications of cultural heritage loss to climate change. This dissertation further provides a critique of mainstream climate justice theories, especially their marginalization of the cultural dimension of climate change. In this regard, the metaphor - ‘cultural storm’ was deployed to argue that climate justice discourses have neither factored the deep socio-cultural impact of climate change nor do they draw on the cultural understanding of justice in putting forward their theories. Given the nature of the indirect, cumulative, and interconnected invisible losses to cultural heritage from climate change, it seems unlikely that they can be addressed by simple tweaks of the climate justice status quo. This dissertation proposes that the idea of personhood in African philosophy, can be conceived to ensure climate justice live up to its expectations in a world of diverse persons dealing with a complex phenomenon like climate change. It argues that a cultural dimension of climate ethics has implications for how mitigation, adaptation and compensation plans should be furthered for global climate justice. At the foundation of my argument, I suggest that what is needed in climate justice discourse is a commitment to explore new and innovative alternatives that will produce an inclusive global climate treaty that is sensitive to the cultural heritage assets that is destroyed by climate change in Africa. This will require a multi-dimensional framework that allows fundamentally different kinds of values and benefits to be given equal visibility and standing in global climate negotiations. The dissertation proceeds in six chapters. In the first chapter, I discuss how climate change denies, damage and destroys cultural heritage values in Africa and argue that it is unjust to ignore this dimension of climate change impact, particularly on the African continent. In the second chapter, I critically discuss the normative value of cultural heritage in an environmentally sustainable and morally appropriate way for this global age. I argue that what is lost when climate change affects cultural heritage is a significant cultural asset that ought to be seriously considered in climate ethics. The third chapter addresses the global justice implications of the destruction of Africa's cultural heritage by climate change that must be paid attention to. In chapter four, I evaluate the plausibility of some of the mainstream climate justice proposals. I offered a different possible critique of current approaches to climate justice to show how they have furthered cultural injustice. The critique of current climate justice theories that I offer, stems from an uninspiring approach that belies logic permitting the sacrificing of that which is connected to others wellbeing as well as the implicit assumptions and the limitation of the idea of justice that undergird these climate justice theories. I offer, in chapter five, a plausible climate ethics theory that recognizes culturally embedded ideas of justice and empower all stakeholders to build by themselves, lives that are, in the light of these ideas, deemed to be adversely impacted by climate change. This theory advances a socio-cultural perspective to climate change which could provide a nuanced basis for understanding and addressing global climate duties that will be sensitive to the loss of cultural heritage. Specifically, I apply the African conception of personhood, to provide a normative basis for a different but intuitive understanding of the cultural dimension of climate ethics. Specifically, I apply the African conception of personhood, to provide a normative basis for a different but intuitive understanding of the cultural dimension of climate ethics. The method was applied in the natural landscape of the Kruger National Park to trace sediment sources delivered to four reservoirs (Hartbeesfontein, Marheya, Nhlanganzwani, Silolweni) whose contributing catchments were underlain by a range of igneous, metamorphic, and sedimentary rocks. This research attempted to evaluate the impact of vegetation, lithology, and particle size controls on the ability of magnetic signatures to discriminate between lithology-defined potential sources. Potential source samples were collected from each lithology present in all catchments, except for the Lugmag catchment where the lithology was uniform, but the vegetation type varied significantly between woodland and grassland. One sediment core was taken in each of the four catchment reservoirs where there was more than one lithology present in order to unmix and apportion contributing sediment sources. Sampling time in the field was often restricted to short periods, dependent on anti-poaching activities and movement of free-roaming wildlife across the Park. This occasionally led to the sub-optimal collection of enough source samples to capture source signature variability. Mineral magnetic parameters were unable to discriminate between vegetation-defined sediment sources in the Lugmag catchment (homogenous underlying lithology) but were able to discriminate between lithology-defined sediment sources (to varying degrees) in the other four catchments. The contributions of each lithology-defined sediment source were estimated using a straightforward statistical protocol frequently used in published literature that included a Mann-Whitney U or Kruskal-Wallis H test, mass conservation test, discriminant function analysis, and an (un)mixing model. A contribution from each lithology source to reservoir sediment was estimated. Connectivity was a significant factor in understanding erosion in each of the catchments. Both longitudinal (e.g., drainage density) and lateral connectivity (e.g., floodplain - river) were important. Travel distance of eroded sediment to reservoirs was also an essential element in two of the four catchments. There are no defined floodplains, so channel bank soils are very similar to the catchment soils. Therefore, channel bank storage potential would be similar to the storage potential within the catchment. Vegetation played a crucial role in protecting soils, by reducing ii erosion potential as well as trapping and storing sediment, thereby interrupting lateral connectivity. Underlying geology and soils are determining factors of vegetation type and density. A published study estimated catchment area-specific sediment yields for different KNP catchments, including the Hartbeesfontein, Marheya, Nhlanganzwani and Silolweni catchments. The published data was used in combination with the (un)mixing model source contribution estimates of this thesis to determine specific sediment yields by lithology, i.e., for each catchment source. The polymodal particle size characteristics of the sample material led to an investigation into particle size controls on the ability of magnetic signatures to discriminate between potential sources. Due to time constraints, only the Hartbeesfontein and Marheya catchments were tested for grain size differences. For each catchment, one bulk sample was created for each lithology source. This bulk sample was divided into 10 subsamples. The samples were then fractionated into four particle size fraction groups: coarse (250 – 500 μm), medium (125 – 250 μm), fine (63 – 125 μm), and very fine (<63 μm). Reservoir samples were also bulked to create 10 down-core samples for each reservoir, and the samples were also fractionated into the four fraction groups. The same statistical protocol was applied to the fractionated samples and contribution estimates were obtained by lithology for each particle size fraction group. The goodness of fit and uncertainty of the (un)mixing model varied in each catchment, with the two measures of accuracy often showing an inverse relationship. The fractionated modelling estimated the same primary source in the two catchments as in the unfractionated modelling. However, additional information on the secondary and tertiary sources was obtained. Connectivity remained a significant factor in interpreting the results of the fractionated analysis. Specific sediment yields were estimated for each catchment source per particle size fraction group. These sediment yields provided a deeper understanding of sediment transport through a catchment and which particle size groups are most important in catchment erosion. An original contribution to research was made by estimating source contribution estimates for the four reservoirs, quantifying sediment yields for each catchment lithology and then for each catchment lithology by particle size. Understanding resource partitioning in three species of Gobiidae living in the warm-temperate Sundays Estuary

- Description: Estuaries represent a transition zone where saline water combines with freshwater, and this results in temporal and spatial variability in both biotic and abiotic parameters. Because the environmental gradient changes so rapidly in areas and the estuarine environment is harsh, fish need to either be able to specialise to one particular suite of abiotic contexts or have the physiological mechanisms to allow them to move throughout the gradient of the estuary. Apart from this, other factors such as climate change, habitat degradation, over exploitation of resources and the introduction of alien species also negatively influence fish species living in estuaries. These anthropogenic stressors can cause species exclusion in areas which were previously suitable for species with particular suites of traits. Measuring resource partitioning within functional groups not only helps in determining the functional structure of communities but also explains how organisms use resources in a community. Resource partitioning studies provide a useful conceptual framework to understand species interactions in a community and identify major resource dimensions along which species segregate. This study aimed to describe resource partitioning among the Gobiidae species, as the family is well represented in South African estuaries (24 species), is highly abundant and has several species which co-exist in individual estuaries. To accomplish this, different forms of resource partitioning that promote the coexistence of the three most abundant species found in warm-temperate South African estuaries; the Knysna sand goby Psammogobius knysnaensis (Smith, 1935), Prison goby Caffrogobius gilchristi (Boulenger, 1898) and River goby Glossogobius callidus (Smith, 1937) were investigated in the Sundays Estuary. This was accomplished by determining the reproductive biology, interspecific differences in feeding morphology, feeding ecology and habitat partitioning among the three species. The distribution and abundance of potential prey in the estuary (mesozooplankton and macrozoobenthic communities) were investigated, with both mesozooplankton and macrozoobenthic communities grouping into three distinct communities along the environmental gradient. Feeding morphological traits responsible for prey location, chasing, capture, mastication, ingestion and digestion were investigated. Little morphological overlap was observed between P. knysnaensis and C. gilchristi, indicating little dietary overlap between these two species. In contrast, the feeding morphology of Glossogobius callidus overlapped with both P. knysnaensis and C. gilchristi, indicating the potential to compete with both species for food. The trophic ecology of the three species was investigated using stomach content analysis, whereby prey abundance, frequency of occurrence and percentage of volume were determined. For P. knysnaensis, the incorporation of cyclopoids and ostracods decreased with fish size and that of bivalves and amphipods increased. Caffrogobius gilchristi was found to be a specialist feeder on cyclopoids but as the species increased in size the incorporation of amphipods, mysids, brachyurans and gastropods increased. Glossogobius callidus was found to be a generalist feeder, feeding on cyclopoids, amphipods and chironomids. The incorporation of cyclopoids decreased with fish size and the consumption of large sized amphipods and chironomids increased with fish size. To assess their reproductive biology, gonadosomatic index (GSI) was used to study the breeding season. In all three species, reproduction peaked in spring and summer, and this is associated with peak phytoplankton and zooplankton biomass in estuaries. Length at 50% maturity and fecundity were also determined. The three species predominated in different locations in the estuary, which explains why they were able to use similar resources and reproduce at the same time. Psammogobius knysnaensis was abundant in the sandy mouth region, with sediment type and prey (bivalves and mysids) availability affecting its distribution. Caffrogobius gilchristi was abundant in the muddy lower reaches of the estuary, with its distribution influenced by sediment composition and temperature. Glossogobius callidus was abundant in the turbid upper reaches, with its distribution influenced by prey (amphipods) availability and turbidity. Different habitat preferences and diet among the specific size ranges enabled coexistence among the species. Mediating and examining expansive learning in the context of multidimensional complexities affecting household food security activity systems in Nyanyadzi Irrigation Scheme in the Manicaland Province of Zimbabwe

- Description: The study sought to mediate and examine expansive learning in the context of multidimensional complexities affecting household food security activity systems in Nyanyadzi Irrigation Scheme in Ward 8 of Chimanimani District in the Manicaland province of Zimbabwe. Therefore, the main foci of the study were to investigate how multidimensional complexities have come to be the way they are (historicizing) and how they enable or constrain learning of household food production. The study utilized Cultural Historical Activity Theory and the Expansive Learning Cycle (Engeström & Sannino, 2010) to examine and mediate collective learning in response to the multidimensional complexities and also to contribute to transforming the farmers’ activity systems towards more sustainable practices to ensure household food security. The study used a qualitative research approach, utilizing an insider formative intervention approach in a case study design in which Nyanyadzi irrigation scheme was the case study. I have adopted the insider formative interventionist role as a 3rd generation farmer, born and bred in Nyanyadzi area, and my family has been involved in the Nyanyadzi irrigation scheme for three generations. I see this as synergistic with the need for deep cultural understanding in CHAT research. However, this role also provided challenges for me to maintain a rigorous approach to the research in which I also reflexively review my own role and influence in the research process. Following CHAT expansive learning methodological guidance, data was generated through fifteen (15) face to face interviews with three generations of farmers in the scheme (historical ethnographic data); four focus group discussions (contemporary ethnographic data) and eight (8) sessions in a three (3) day change laboratory workshop (expansive learning data). Double stimulation and ‘mirror’ data was used to surface and prioritise responses to contradictions in the Change Laboratory Workshops (CLW), which is a methodology developed in and for CHAT research (ibid). The data was analysed using both inductive and abductive approaches and were conducted in a three-phased process focusing firstly on the history of the object, followed by current perspectives on the object of activity and lastly on transformations emerging in the object of activity via the expansive learning process. Cultural Historical Activity Theoretical tools informed activity system analysis and analysis of the history of the object and emerging contradictions; and the expansive learning cycle (ELP) process framework associated with and emergent from CHAT was used to analyse the emergence of transformative agency and expansion of the object. Four levels of contradictions were used to describe and explain the emergent contradictions namely, primary, secondary, tertiary and quaternary contradictions and are presented in this sequence as catalytic opportunities for expansive learning as proposed in CHAT. In addition, the four types of discursive manifestations of contradictions namely, dilemma, conflict, critical conflict and double bind were used to describe and explain the manifestations of contradictions in this study and their role in catalysing transformative agency. The concept of linguistic cues for discursive manifestations of contradictions was adopted and employed in the preliminary data analysis phase. In addition, the transformative agency expressions and zone of proximal development (ZPD) concept was applied in and to the CLW data to examine the learning pathways and to co-develop and expand farmers’ and other stakeholders’ transformative agency and ZPD respectively. I found myself as formative insider researcher having to take on a strong role as co-engaged researcher / participant in the irrigation scheme expansive learning process. The study concluded that the farmers’ activity system is the central activity system and it interacts with other activity systems on a partially shared object “improved crop production and marketing under irrigation scheme”. Through interactions with other neighbouring activity systems, the farmers have faced multidimensional complexities that constrain their ability to fully realise their object. These multidimensional complexities manifest in three critical contradictions as a critical conflict in leadership and management crisis; a dilemma and double bind in a lack of farmer education and training; and lastly, a dilemma and double bind in poor crop marketing. These multidimensional complexities have a historical account and they have evolved in complexity over time and I argue in the thesis that, a careful cultural-historicity of the object of activity and mediation of the situated learning can help to collectively come up with solutions to these multidimensional complexities. The study further concluded that despite these multidimensional complexities in the scheme, learning has been taking place and such learning was sometimes mediated through demonstrations as “learning by doing,” “seeing is believing” and the “winners and losers” concept. Furthermore, through the CLW process the farmers and other stakeholders’ cognitive horizons were expanded by the mirror data and double stimulation processes, and the expansive learning process developed their individual and collective transformative agency pathways and expanded their collective zone of proximal development. In this study I argue that there has been little said about collective learning in irrigation schemes and given the dearth of detail on such learning, it seems that this learning is either going unnoticed or is ignored. Hence, I further argued that the multidimensional complexities in irrigation schemes are both a stimulant for learning and provide a space (object) for collective learning, as was also shown by Baloi (2016). The study also shows that the collective learning potential in these irrigation schemes can be pro-actively mediated via expansive learning formative interventions in support of improved crop production and marketing for produce developed under irrigation in irrigation schemes such as the Nyanyadzi Irrigation scheme. Lastly, being an insider formative interventionist researcher in this study, with intergenerational engagement and perspective on the history of the object and integrational engagement with the transforming object, I became part of the intergenerational transformation of the irrigation scheme’s object. The intergenerational co-construction of the history of the object, coupled with the insider formative interventionist researcher approach opened up and allowed me as a current generation agent or actor to develop an in-depth understanding of the multidimensional nature and historicity of the object. This was crucial for opening up the transformative agency pathways. It also produced a responsibility for me as an insider formative interventionist researcher to carry the summative findings of the study back into the social context to widen the engagement and mediation of the transformation needed in the community. Overall not only does the study offer an intergenerational perspective on multidimensional complexities of the object and how this can generatively be mobilised via expansive learning into emerging transformative learning agency pathways, but it also offers a new vantage point on the role of the insider formative interventionist researcher. Through this, the study offers insight into how we as third generation members of the community can be brought closer to our communities through the application of our skills, thereby also offering a new type of engaged and rigorously framed and executed research with roots in our communities. As shown in this study, not only does this expand the knowledge and experience of those we engage with, but it also expands our own knowledge and expertise in order to be more able to contribute to both the challenges of our own communities but also that of other communities and situations similar to ours, and beyond these bounded contexts. - Description: The study sought to mediate and examine expansive learning in the context of multidimensional complexities affecting household food security activity systems in Nyanyadzi Irrigation Scheme in Ward 8 of Chimanimani District in the Manicaland province of Zimbabwe. Therefore, the main foci of the study were to investigate how multidimensional complexities have come to be the way they are (historicizing) and how they enable or constrain learning of household food production. The study utilized Cultural Historical Activity Theory and the Expansive Learning Cycle (Engeström & Sannino, 2010) to examine and mediate collective learning in response to the multidimensional complexities and also to contribute to transforming the farmers’ activity systems towards more sustainable practices to ensure household food security. The study used a qualitative research approach, utilizing an insider formative intervention approach in a case study design in which Nyanyadzi irrigation scheme was the case study. I have adopted the insider formative interventionist role as a 3rd generation farmer, born and bred in Nyanyadzi area, and my family has been involved in the Nyanyadzi irrigation scheme for three generations. I see this as synergistic with the need for deep cultural understanding in CHAT research. However, this role also provided challenges for me to maintain a rigorous approach to the research in which I also reflexively review my own role and influence in the research process. Following CHAT expansive learning methodological guidance, data was generated through fifteen (15) face to face interviews with three generations of farmers in the scheme (historical ethnographic data); four focus group discussions (contemporary ethnographic data) and eight (8) sessions in a three (3) day change laboratory workshop (expansive learning data). Double stimulation and ‘mirror’ data was used to surface and prioritise responses to contradictions in the Change Laboratory Workshops (CLW), which is a methodology developed in and for CHAT research (ibid). The data was analysed using both inductive and abductive approaches and were conducted in a three-phased process focusing firstly on the history of the object, followed by current perspectives on the object of activity and lastly on transformations emerging in the object of activity via the expansive learning process. Biodiversity conservation of South Africa’s Wild Coast through the years: Exploring the tensions between western-style and local traditional conservation practices

- Description: The Transkei Wild Coast is a unique and diverse biophysical environment stretching some 280 kilometres between the Great Kei and Mtamvuna Rivers along South Africa’s eastern coastline. Its extensive indigenous forests were recognised for their biological and economic importance by the colonial government and granted protection from the late 1800s, with forced removal of indigenous people who resided within them. The state restricted and controlled the utilisation of the forest resources, particularly by Black people, through promulgation and enforcement of legislation. Similar conservation methods were employed for protection of the Wild Coast’s soils, grasslands and marine resources, usually without any consultation with local Black leadership or communities who relied upon these resources. Previous studies have focussed on the impacts of Western-style conservation legislation and the promulgation of protected areas on local people. These have suggested that local communities have been denied access to natural resources through a fences and fines approach, often accompanied by heavy-handed, strict law enforcement measures. Very little research has been conducted to document the actual reasons for and the history of conservation efforts along the Wild Coast. The aim of this study was to explore the history of natural resource management and conservation along the Wild Coast from a Western and African perspective, and to examine the conflicts that have arisen as a result thereof. The importance of utilisation of natural resources to sustain livelihoods and the conservation methods applied to protect these resources were examined, mainly by means of a literature review and face-to-face interviews with local traditional leaders and environmental managers. The successes, challenges and conflicts that resulted from conservation efforts were examined through detailed analysis of the conservation history of three case study areas: Dwesa-Cwebe, Hluleka and Mkambati nature reserves. The study concludes that traditional leaders, foresters and conservation managers have largely been ignored during previous research on management and conservation of the Wild Coast; however, they were very eager to express their views on this subject. The interviews revealed that traditional leaders and conservationists from each case study area want to protect the Wild Coast and its natural resources, albeit for different reasons. Conservationists recognise the importance of the region’s biodiversity and the need for protected areas, particularly where there is rich endemism such as at Mkambati and Dwesa-Cwebe. Traditional leaders recognise the potential for jobs for their people where protected areas preserve the aesthetic beauty of the coast and attract tourists. For them, employment opportunities are a priority, as is access to utilisation of natural resources to sustain livelihoods. The traditional customs and culture of the people living along the Wild Coast are also seen as being of critical importance. This has not been adequately considered by past conservation planners and managers, but traditional leaders express value in conserving the amaXhosa and amaMpondo way of life. Nowhere is this more evident than in relation to grazing land for cattle. Another significant finding is that the support shown by traditional leaders for conservation of the terrestrial environment is not necessarily mirrored when it comes to protection of marine resources. The Wild Coast has a rich variety of marine flora and fauna and a high percentage of endemic species. Local people have relied upon marine resources for sustenance, but conservationists acknowledge that access to these resources has historically been restricted without consultation with traditional leaders or local communities. It is concluded that these restrictions can be linked to many land claims on protected areas on the Wild Coast since 1994. While this study suggests that both traditional leaders and conservationists want to protect the Wild Coast and its natural resources, there is still mistrust based on a history of unjust forced removals from forests, restrictions on the use of natural resources through the manifestation of colonial and apartheid power, and broken promises relating to benefit sharing from proceeds of wildlife and tourism ventures, as well as those benefits linked to land claim settlement agreements in the post-apartheid era. The study concludes that, despite past conflicts relating to conservation management, the Wild Coast’s protected areas have widespread support amongst traditional leaders. There is agreement that co-management is the preferred option but the state has to assist in curbing lawlessness that threatens over-exploitation of natural resources. It is largely acknowledged by conservationists that local people should be permitted to sustainably use certain natural resources and that there have to be increased economic benefits from tourism. Although protected area managers cannot be expected to carry the responsibility of failed tourism models and economic ventures on nature reserves, this study found that they are often the only government officials available on the ground for traditional leaders and local communities to consult with. For co-management to be successful on the protected areas, regular communication about all matters pertaining to management is considered important. While it is accepted that traditional leaders no longer have the administrative powers that they historically possessed, the study found that they are still widely respected along the Wild Coast. - Description: Community engagement (CE) is now widely considered a core function of higher education worldwide. In South African higher education institutions (HEIs), there is an increasing focus on CE as a means of transforming the role of the university in society, though the forms and shapes of CE vary by institution. CE is positioned as part of the means of addressing the challenges within the South African education system, such as ensuring equity in academic access in the face of diversity and making sure higher education institutions are responsive to the needs of society. Community Engagement is increasingly being afforded the same status as teaching and learning and research in higher education. The idea that higher education should function as a public good is central to this. This study reflects on how CE can be expansively viewed as places of learning for students to achieve epistemic access with epistemic justice, particularly in increasing diverse and changing contexts. Despite the growing research on CE in HEIs, there is comparatively limited focus on the intersection between CE and language use and potential linkages with identity and epistemic access and success. Given that one of the major challenges in South African HEIs relate to difficulties experienced by students whose home language is not English, the experiences of students learning within CE contexts within those institutions warrants investigation. The main aim of this study was to explore second language English speaking students’ experiences of language in the Engaged Citizen Programme, a Community Engagement programme at Rhodes University aimed at offering students the opportunity of enhanced learning, giving students the opportunity to evaluate the theories and ideas taught in the university against the realities of the South African context. The programme is also intended to offer students the opportunity to learn with and from communities and thereby enable personal growth (ECP Handbook: 2020; p.3). Using an in-depth phenomenological approach, this study explored diverse students’ experiences of language use in both community engaged programmes and in the classroom as a basis for understanding the role language plays in such spaces and the impact of these programmes on epistemic access, justice and success for students in HEIs. The study explored the role that CE plays for second language English students as they navigate complex questions of identity and belonging in HEIs. In CE activities, such as the Engaged Citizen Programme, unlike traditional classroom learning, English is often not the medium of instruction, as learning takes place in community sites, where multiple other languages are spoken. In the traditional classrooms, English is the dominant medium of instruction which can bring challenges for students whose home language is not English. Students are faced with various challenges including failure to communicate effectively and understand content knowledge. Significantly this study found that this often related to a sense of self-worth and belonging and constrained their participation and engagement in class. It was evident from the students’ reflections on their experiences in the Engaged Citizen Programme that CE provided a more flexible space generally more comfortable to these participants; a space that promotes engaged learning without rigid rules. The students’ reflections affirmed the contribution of CE in promoting engagement of students outside the formal classes and enhancing the ways in which they use language freely. It was also evident from the students’ reflections that CE provided a space in which students can identify who they are and have a sense of belonging. In the context of diversity, the majority of the students said they come to the university feeling a level of under preparedness and cannot identity with dominant groups. The reflections from the students’ experiences therefore offer some insights into ways in which we can actively promote CE in supporting student access and addressing issues of epistemic justice in higher education. The findings suggest that many of the benefits of CE, such as higher levels of interaction and significant amounts of translanguaging, need to be brought into the formal classroom spaces because they enhanced student engagement. While CE was also seen to be challenging and there were calls for more support, the essence of the experience was as a space of personal development and awareness of social responsibility. The explicit normative value of CE was in contrast to the absence of such considerations in the formal HE curriculum and the student experiences suggest that much could be learned from this. The use of English, both on campus and in CE activities, was found to be value-laden and politically charged. The participants, black students who spoke English as an additional language, all related experiences of English being positioned as a ‘superior’ language. The students who were highly fluent in English experienced being positioned as ‘showing off’ and seen to have ‘forgotten their roots’. Students who were not highly fluent in English, on the other hand, often constrained their participation in class because they experienced concern that their mispronunciations and accents may be mocked. The essence of the experience of language use in both formal classroom settings and in CE activities is that this is tightly bound to identity and is ideologically fraught. - Description: Community engagement (CE) is now widely considered a core function of higher education worldwide. In South African higher education institutions (HEIs), there is an increasing focus on CE as a means of transforming the role of the university in society, though the forms and shapes of CE vary by institution. CE is positioned as part of the means of addressing the challenges within the South African education system, such as ensuring equity in academic access in the face of diversity and making sure higher education institutions are responsive to the needs of society. Community Engagement is increasingly being afforded the same status as teaching and learning and research in higher education. The idea that higher education should function as a public good is central to this. This study reflects on how CE can be expansively viewed as places of learning for students to achieve epistemic access with epistemic justice, particularly in increasing diverse and changing contexts. Despite the growing research on CE in HEIs, there is comparatively limited focus on the intersection between CE and language use and potential linkages with identity and epistemic access and success. Given that one of the major challenges in South African HEIs relate to difficulties experienced by students whose home language is not English, the experiences of students learning within CE contexts within those institutions warrants investigation. The main aim of this study was to explore second language English speaking students’ experiences of language in the Engaged Citizen Programme, a Community Engagement programme at Rhodes University aimed at offering students the opportunity of enhanced learning, giving students the opportunity to evaluate the theories and ideas taught in the university against the realities of the South African context. The programme is also intended to offer students the opportunity to learn with and from communities and thereby enable personal growth (ECP Handbook: 2020; p.3). Using an in-depth phenomenological approach, this study explored diverse students’ experiences of language use in both community engaged programmes and in the classroom as a basis for understanding the role language plays in such spaces and the impact of these programmes on epistemic access, justice and success for students in HEIs. The study explored the role that CE plays for second language English students as they navigate complex questions of identity and belonging in HEIs. In CE activities, such as the Engaged Citizen Programme, unlike traditional classroom learning, English is often not the medium of instruction, as learning takes place in community sites, where multiple other languages are spoken. In the traditional classrooms, English is the dominant medium of instruction which can bring challenges for students whose home language is not English. Students are faced with various challenges including failure to communicate effectively and understand content knowledge. Significantly this study found that this often related to a sense of self-worth and belonging and constrained their participation and engagement in class. It was evident from the students’ reflections on their experiences in the Engaged Citizen Programme that CE provided a more flexible space generally more comfortable to these participants; a space that promotes engaged learning without rigid rules. The students’ reflections affirmed the contribution of CE in promoting engagement of students outside the formal classes and enhancing the ways in which they use language freely. It was also evident from the students’ reflections that CE provided a space in which students can identify who they are and have a sense of belonging. In the context of diversity, the majority of the students said they come to the university feeling a level of under preparedness and cannot identity with dominant groups. The reflections from the students’ experiences therefore offer some insights into ways in which we can actively promote CE in supporting student access and addressing issues of epistemic justice in higher education. The findings suggest that many of the benefits of CE, such as higher levels of interaction and significant amounts of translanguaging, need to be brought into the formal classroom spaces because they enhanced student engagement. While CE was also seen to be challenging and there were calls for more support, the essence of the experience was as a space of personal development and awareness of social responsibility. The explicit normative value of CE was in contrast to the absence of such considerations in the formal HE curriculum and the student experiences suggest that much could be learned from this. The use of English, both on campus and in CE activities, was found to be value-laden and politically charged. The participants, black students who spoke English as an additional language, all related experiences of English being positioned as a ‘superior’ language. The students who were highly fluent in English experienced being positioned as ‘showing off’ and seen to have ‘forgotten their roots’. Students who were not highly fluent in English, on the other hand, often constrained their participation in class because they experienced concern that their mispronunciations and accents may be mocked. The essence of the experience of language use in both formal classroom settings and in CE activities is that this is tightly bound to identity and is ideologically fraught. - Description: The integration of indigenous knowledge (IK) in the science curriculum is a spreading phenomenon driven by the need to bring about relevancy and equality in science education. In South Africa, for instance, the need to integrate IK in science education is part of the global effort to build a democratic state from the debris of apartheid. Henceforth, the integration of IK is backed up by both the National Constitution of the Republic of South Africa (Act 108 of 1996) and the South African Department of Basic Education’s (2011) National Curriculum Assessment Policy Statement. However, the success of this policy seems to be hindered in part by the fact that the teachers who are the implementers of the curriculum changes seem to lack the relevant pedagogical content knowledge (PCK) to integrate IK in their science teaching repertoires. Such a trend is often blamed on their Eurocentric educational background. Interestingly, very little research has been done to explore ways of supporting teachers to develop the relevant conceptual tools and teaching strategies that will enable them to integrate IK in science teaching. It is against this background that an interventionist case study on how to support the Bachelor of Education Natural Sciences in-service teachers in particular to develop exemplar science lessons that integrate IK as easily accessible resources was conducted. The study is underpinned by three complementary paradigms, namely, the interpretive, the critical, and indigenous research paradigms. While the interpretive paradigm enabled me to understand and interpret descriptive data, the critical paradigm enabled me to take an emancipatory stance and challenge the micro-aggressive elements embedded in conventional research practices; within the indigenous research paradigm, Ubuntu was the relational perspective that informed the researcher-participant relationships in this study. Vygotsky’s sociocultural theory was used as an overarching theoretical framework, in conjunction with the cultural historical activity theory. Additionally, the topic-specific pedagogical content knowledge provided the methodological and analytical tools. Data were gathered through questionnaires, individual face-to-face interviews, focus group interview, participatory observation, and the teachers’ reflections. This study established that if teachers are given back the agency to collaboratively resolve the contradictions that confront them in their workplaces, they can generate their own ideas on how to integrate IK in science vii teaching. The teachers in this study experienced a shift in their agency from a paralysed state of resisting the integration of IK at the beginning of the intervention to an ‘I can do it’ attitude at the end of the intervention. Thus, it could be argued that this study’s major contribution to new knowledge lies in demonstrating possible ways of supporting teachers to integrate IK as easily accessible resources in their science teaching. Additionally, the study also challenged the Eurocentric approach to ethics and offered Ubuntu as a relational perspective that can be used to complement the shortcomings of Eurocentric research paradigms.
